Thermally controlled Er3+-Ho3+ energy transfer in NaYGeO4 olivine phosphor for enhanced NIR-II luminescence and sensitive thermometry

Hui Zeng,Xiaolang Fan,Yangbo Wang,Huaiyong Li

Lanthanide NIR-II luminescence free from thermal quenching is attractive for various applications at elevated temperatures. Here, we present a thermal enhancement of Er 3+ NIR-II luminescence based on the synergy of phonon assisted Er 3+ -Er 3+ cross relaxations and controllable Er 3+ -Ho 3+ energy transfer. The lattice expansion of NaYGeO 4 olivine phosphor upon heating (298–823 K) suppresses the Er 3+ -Ho 3+ energy transfer and leads up to 46.1 times enhancement of Er 3+ NIR-II luminescence under 379 nm excitation. The NIR-II luminescence evolution as increasing temperature allows dual-mode ratiometric thermometry, i.e., luminescence intensity ratios (LIRs) based on Er 3+ 4 I 13/2 Stark sublevels and on Er 3+ 4 I 13/2 and Ho 3+ 5 I 6 levels. Obtained relative sensitivities above 0.46% K ‒1 over a broad temperature range of 298–673 K with a maximum of 2.32% K -1 suggest NaYGeO 4 :Er 3+ /Ho 3+ phosphor as a promising NIR-II luminescence thermometer. Moreover, this research offers an available perspective for designing thermal-enhanced NIR luminescence in ordinary thermal expansion materials.

NIR-II conjugated polymer/emodin nanoplatform for mild photothermal therapy/pyroptosis of head and neck squamous cell carcinoma

Yehui Kang,Yahui Zhang,Weijiao Fan,Liya Yu,Mengdie Yu,Yuejia Ding,Yu Cai,Aihong Jiao,Jiajie Xu

Head and neck squamous cell carcinoma (HNSCC) remains a clinically challenging malignancy due to limited therapeutic options and considerable treatment-related toxicity. In this study, we present an innovative nanotherapeutic platform—monocarboxylate transporter 1 (MCT1) antibody-targeted nanoparticles co-loaded with a second near-infrared window (NIR-II, 1000–1700 nm) absorbing conjugated polymer (PDTP-TBZ-OD, denoted as PTO) and emodin (MCT@PTO-Em NPs) —that synergistically integrates emodin-induced pyroptosis with NIR-II mediated mild photothermal therapy (MPTT) to address critical limitations of current HNSCC treatments strategies. The synthesized NIR-II-absorbing conjugated polymer PTO exhibits strong absorption at 1064 nm and a high photothermal conversion efficiency (39 %), enabling deeptissue penetration and precise hyperthermia control. Accordingly, MCT@PTO-Em NPs implement a dual-modal therapeutic strategy that triggers pyroptosis through NLRP3 inflammasome activation while simultaneously enhancing tumor cell death via mild hyperthermia. Moreover, emodin encapsulated within the nanoparticles (NPs) inhibits heat-shock protein 70 (HSP70), thereby disrupting cellular thermoresistance mechanisms and sensitizing cancer cells to MPTT. Additionally, surface functionalization of the nanoparticles with MCT1-targeting antibodies enhances tumor-specific delivery and intracellular accumulation, thereby minimizing off-target effects. Both in vitro and in vivo studies demonstrate that MCT@PTO-Em NPs achieve superior tumor suppression through the synergistic interplay between pyroptosis and MPTT, while exhibiting excellent biocompatibility. This work presents a precision nanomedicine strategy that integrates molecular targeting, pyroptosis-mediated immune activation, and MPTT, offering a transformative therapeutic paradigm for HNSCC and other solid tumors.

Fluorinated Hypoxia-Responsive Aza-BODIPY for NIR-II FL/19F MR/PA Imaging and Phototherapy of Lung Cancer

Anfeng Li,Fang Wang,Mou Jiang,Yu Li,Xin Zhou,Zhong-Xing Jiang

Tumor hypoxia limits the efficacy of photodynamic therapy (PDT), necessitating photosensitizers with hypoxia-adaptive therapy and imaging. Here, we present a fluorinated N -oxide aza-BODIPY ( OFBD ) nanoemulsion ( OFBD-NP ) for hypoxia-responsive, multimodal imaging-guided phototherapy. OFBD generates robust singlet oxygen under normoxia for PDT, but is reduced by CYP450 enzymes in hypoxic cells to photothermal-potent FBD , enabling switchable PDT/PTT. Co-assembly with fluorinated oil enhances oxygen delivery, boosts 19 F MRI sensitivity, and promotes J -aggregation, shifting fluorescence into the NIR-II window for deep-tissue imaging. The redox conversion also activates photoacoustic signals, enabling responsive tri-modal imaging (NIR-II FLI/ 19 F MRI/PAI). OFBD-NP shows potent cytotoxicity in vitro under both normoxic and hypoxic conditions via apoptosis. In vivo, it selectively accumulates in tumors, offers high-contrast imaging, and leads to complete tumor regression in subcutaneous A549 models after laser irradiation, without systemic toxicity. This work demonstrates a smart nanoplatform that integrates deep-tissue imaging and hypoxia-triggered therapeutic switching, addressing major limitations of conventional photosensitizers in cancer imaging and phototherapy.

Engineering of 2D Ultrathin CuS-PMA Heterostructures for Enhanced NIR-II Photothermal Therapy Facilitating Oxidative Stress and Immunotherapy

Dong Wang,Feng Yuan,Xuliang Deng,Haoyang Li,Siyang Nie,Qingda Liu,Xun Wang

Therapy based on heat and reactive oxygen species (ROS) generation have attracted increasing attention, while the weak photothermal ability and low ROS production have severely limited the efficiency of the tumor treatment. Here, 2D ultrathin CuS-phosphomolybdic acid (PMA) nanosheets are synthesized at sub-nanoscale. Typically, the improved second near-infrared light (NIR-II) light absorption, small bandgap, fast hot carrier response, abundant oxygen vacancies, and the electron delocalization between closely connected PMA and CuS units in sub-nanoscale result in the notably enhanced NIR-II photothermal therapy (PTT) performance of CuS-PMA. The reversible redox of Mo active sites in PMA provides basis for glutathione (GSH) depletion while triggering chemodynamic therapy (CDT) in the tumor microenvironment (TME), and the heat produced by the PTT greatly promotes the CDT and GSH depletion, where CuS-PMA presents sevenfold more ROS generation and fourfold more GSH depletion under PTT. In addition, the ROS generation/GSH depletion can downregulate heat shock proteins, weakening the heat resistance of tumor cells and thereby amplifying the killing effect of PTT. Moreover, the combination of antiprogrammed death-ligand 1 (αPD-L1) with the treatment of CuS-PMA promote the immunotherapy, resulting in the suppression of untreated abscopal tumors.

Gold nanobipyramid@copper sulfide nanotheranostics for image-guided NIR-II photo/chemodynamic cancer therapy with enhanced immune response

Yiyu Chen,Ping Liu,Chunze Zhou,Tao Zhang,Tianxing Zhou,Dandan Men,Guihua Jiang,Lifeng Hang

Photothermal therapy (PTT), photodynamic therapy (PDT), and chemodynamic therapy (CDT) can cause cancer cell death through an immunogenic process. However, the study of second near-infrared window (NIR-II)-triggered PTT and PDT combined with CDT to induce an immune response has not been recently reported. Here, we integrated gold nanobipyramids and copper sulfide in a core/shell architecture (AuNBP@CuS). The material displays both photodynamic and photothermal properties under irradiation with a NIR-II laser. The released Cu 2+ from CuS under an acidic tumor microenvironment can be converted to Cu + by glutathione following a Fenton-like reaction with hydrogen peroxide to generate highly toxic hydroxyl radicals in the tumor region. Both in vitro and in vivo results demonstrated that such multifunctional nanoplatforms could achieve enhanced efficiency for image-guided tumor suppression based on the NIR-II photo/chemodynamic therapy. We found that damage-associated molecular pattern molecules such as adenosine triphosphate, pre-apoptotic calreticulin , and high mobility group box-1 in dying cells induced by the NIR-II photo/chemodynamic therapy could simultaneously trigger adaptive immune responses. This is the first report revealing that NIR-II photo/chemodynamic therapy based on AuNBP@CuS had promising performance on tumor suppressor with an effective immunogenic cell death process. Statement of significance 1. AuNBP@CuS displays both NIR-II photodynamic and photothermal properties. 2. Cu + following a Fenton-like reaction to generate highly toxic hydroxyl radicals. 3. The NIR-II photo/chemodynamic therapy can trigger adaptive immune responses. 4. Such multifunctional nanoplatforms could achieve enhanced efficiency for tumor suppression.

Bright, Magnetic NIR-II Quantum Dot Probe for Sensitive Dual-Modality Imaging and Intensive Combination Therapy of Cancer

Yingying Li,Peisen Zhang,Wen Tang,Kevin J. McHugh,Stephen V. Kershaw,Mingxia Jiao,Xiaodan Huang,Sergii Kalytchuk,Collin F. Perkinson,Saisai Yue,Yuanyuan Qiao,Lichong Zhu,Lihong Jing,Mingyuan Gao,Buxing Han

Improving the effectiveness of cancer therapy will require tools that enable more specific cancer targeting and improved tumor visualization. Theranostics have the potential for improving cancer care because of their ability to serve as both diagnostics and therapeutics; however, their diagnostic potential is often limited by tissue-associated light absorption and scattering. Herein, we develop CuInSe2@ZnS:Mn quantum dots (QDs) with intrinsic multifunctionality that both enable the accurate localization of small metastases and act as potent tumor ablation agents. By leveraging the growth kinetics of a ZnS shell on a biocompatible CuInSe2 core, Mn doping, and folic acid functionalization, we produce biocompatible QDs with high near-infrared (NIR)-II fluorescence efficiency up to 31.2%, high contrast on magnetic resonance imaging (MRI), and preferential distribution in 4T1 breast cancer tumors. MRI-enabled contrast of these nanoprobes is sufficient to timely identify small metastases in the lungs, which is critically important for preventing cancer spreading and recurrence. Further, exciting tumor-resident QDs with NIR light produces both fluorescence for tumor visualization through radiative recombination pathways as well as heat and radicals through nonradiative recombination pathways that kill cancer cells and initiate an anticancer immune response, which eliminates tumor and prevents tumor regrowth in 80% of mice.

Iron-based and BRD4-downregulated strategy for amplified ferroptosis based on pH-sensitive/NIR-II-boosted nano-matchbox

Lujing Geng,Tong Lu,Huaqing Jing,Yue Zhou,Xiaoyang Liang,Jiao Li,Nan Li

Ferroptosis (FPT), a novel form of programmed cell death, is characterized by overwhelming iron/reactive oxygen species (ROS)-dependent accumulation of lipid peroxidation (LPO). However, the insufficiency of endogenous iron and ROS level limited the FPT therapeutic efficacy to a large extent. To overcome this obstacle, the bromodomain-containing protein 4 (BRD 4 )-inhibitor (+)-JQ1 (JQ1) and iron-supplement ferric ammonium citrate (FAC)-loaded gold nanorods (GNRs) are encapsulated into the zeolitic imidazolate framework-8 (ZIF-8) to form matchbox-like [email protected] /ZIF-8 for the amplified FPT therapy. The existence of matchbox (ZIF-8) is stable in physiologically neutral conditions but degradable in acidic environment, which could prevent the loaded agents from prematurely reacting. Moreover, GNRs as the drug-carriers induce the photothermal therapy (PTT) effect under the irradiation of near-infrared II (NIR-II) light owing to the absorption by localized surface plasmon resonance (LSPR), while the hyperthermia also boosts the JQ1 and FAC releasing in the tumor microenvironment (TME). On one hand, the FAC-induced Fenton/Fenton-like reactions in TME can simultaneously generate iron (Fe 3+ /Fe 2+ ) and ROS to initiate the FPT treatment by LPO elevation. On the other hand, JQ1 as a small molecule inhibitor of BRD 4 protein can amplify FPT through downregulating the expression of glutathione peroxidase 4 (GPX4), thus inhibiting the ROS elimination and leading to the LPO accumulation. Both in vitro and in vivo studies reveal that this pH-sensitive nano-matchbox achieves obvious suppression of tumor growth with good biosafety and biocompatibility. As a result, our study points out a PTT combined iron-based/BRD 4 -downregulated strategy for amplified ferrotherapy which also opens the door of future exploitation of ferrotherapy systems.

Efficient luminescence emission in both visible and NIR-II regions by Er3+ partitioning doping and interfacial energy transfer

Jiajia Meng,Yanyan Cui,Yaling Wang

Lanthanide-doped nanoparticles (LDNPs) are widely used in biophotonics due to their narrow emission band, long lifetime and broad-spectrum tunable luminescence properties. Usually, photosensitizers for biotherapeutic applications need to be triggered by visible light, while bioimaging requires near-infrared (NIR) light with better penetration. However, it is still challenging to achieve efficient luminescence emission in the visible to second near-infrared (NIR-II, 1000–1700 nm) range simultaneously for multifunctional nanoparticles excited at a single NIR wavelength. Here, a simple NaYF 4 :Er/Ce@NaYbF 4 :Er@NaYF 4 :Nd core-shell-shell nanoparticle with interfacial energy transfer by the Yb-sensitized shell and partition doping of Er 3+ under 808 nm excitation, which allows the system to achieve visible and NIR-II luminescence emission from different partitions of Er 3+ . When Ce 3+ is introduced into the core, the Er 3+ in the Yb-sensitized shell is responsible for the emission in the visible region, while the core-doped Er 3+ is mainly responsible for the emission at 1550 nm in the NIR-II. In addition, this core-shell-shell nanostructure has a long luminescence emission lifetime in the visible and NIR-II regions, especially the luminescence lifetime of Er 3+ at 1550 nm is extended to 6.4 ms. More importantly, efficient 1 O 2 generation can be achieved by coupling core-shell-shell nanoparticles with photosensitizer. These results will provide previously unattainable opportunities for LDNPs in biotherapeutic and imaging applications.

Persistent luminescence resonance energy transfer (PLRET)-engineered ZGO@IR-820 with NIR-II afterglow emission and phototherapeutic properties

Houlin Li,Jiaxin Sun,Peng Yan,Aikelaimu Aihemaiti,Abdukader Abdukayum

Near-infrared II (NIR-II) luminescence imaging, with its significant advantages of deep tissue penetration and high spatial resolution, has become a promising technology for cancer diagnosis. The integration of NIR-II imaging with multifunctional therapeutic modalities has been widely explored in biomedicine, offering valuable development opportunities. Persistent luminescent nanoparticles (PLNPs) are of great interest in biomedical applications due to their ability to emit afterglow without the need for in situ excitation. However, single PLNPs exhibit limited performance in processes related to photothermal therapy (PTT) and photodynamic therapy (PDT), and there remains a lack of PLNPs-based multifunctional nanocomposites that simultaneously possess NIR-II afterglow luminescence, as well as PDT, PTT and photocatalytic therapy (PCT) properties. Herein, we report a novel multifunctional nanocomposite, ZGO@IR-820, which is composed of PLNPs (ZnGa 1.97 O 4 :Cr 0.01 ,Bi 0.02 , ZGO) and the near-infrared fluorescent dye IR-820. This nanocomposite not only exhibits NIR-II afterglow emission, but also integrates PDT, PTT and PCT properties. The NIR-II afterglow performance is achieved through the persistent luminescence resonance energy transfer between the PLNPs and IR-820. This multifunctional design expands the photophysical and photochemical properties of PLNPs-based nanocomposites, providing an innovative material design strategy for the development of precise disease diagnosis and efficient therapy.

Metal–Organic Framework (MOF)-Based Single-Atom Iron Catalysts for NIR-II Enhanced Catalytic-Photothermal Synergistic Antibacterial Therapy

Changrui Ying,Xiaolong Zhu,Shuqing Xiang,Mingqian Wang,Xuefeng Jiang,Jian Shen

Single-atom catalysts based on carbon-based materials have a high catalytic activity and excellent photothermal performance. In this article, iron single-atom catalysts loaded on C-MOF-5 carriers have been prepared for the application of photothermal catalytic antimicrobial therapy. Iron single-atom catalysts (Fe SACs) exhibit significant peroxidase-like catalytic activity and catalyze the production of large amounts of hydroxyl radicals (·OH) when hydrogen peroxide is present, which have a killing effect on bacteria. In addition, the excellent photothermal properties of the carbon material allow Fe SACs to rapidly generate a localized high temperature under NIR-II laser light (1064 nm) to kill bacteria and enhance peroxidase-like catalytic activity to produce a large number of hydroxyl radicals. In vitro bacterial assays demonstrated that the synergistic antimicrobial strategy of chemodynamic therapy (CDT) and photothermal therapy (PTT) is nearly 100% effective against E. coli and S. aureus, demonstrating greater antimicrobial efficiency than chemodynamic therapy and photothermal therapy alone. In vivo bacterial assay shows better wound healing in mice treated with CDT, PTT synergistic antimicrobial therapy. In summary, these carbon-based single-atom catalysts could be used as antimicrobial agents through the CDT and PTT synergistic antimicrobial effect.

Dye-sensitized rare-earth-doped nanoprobe for simultaneously enhanced NIR-II imaging and precise treatment of bacterial infection

Yingxin Zhu,Xianzhu Luo,Zihang Yu,Shihui Wen,Guochen Bao,Le Zhang,Cuiling Zhang,Yuezhong Xian

Methicillin-resistant Staphylococcus aureus (MRSA) is responsible for causing life-threatening infections that result in high morbidity and mortality rates. The development of advanced imaging and therapeutic methods for in vivo diagnosis and treatment of MRSA infections remains challenging. Here, we develop a hybrid nanoplatform based on rare-earth-doped nanoparticles (RENPs) sensitized by a moiety-engineered near-infrared (NIR) TPEO-820 dye and with a ZIF-8 layer that incorporates CysNO, a photochemically triggered nitric oxide donor. We then use the hybrid for both NIR-II bioimaging and photoactivatable treatment of MRSA-infected wounds. We show that the NIR dye sensitization leads to an 8.5-fold enhancement of the downshifting emission and facilitates deep-tissue NIR-II imaging of bacterial infections. Moreover, the sensitization strategy enhances the UV emission of RENPs by two orders of magnitude, leading to the efficiently controllable release of nitric oxide for effective disinfection of MRSA in vitro and in vivo. The hybrid nanoplatform thus offers promising opportunities for simultaneous localization and controllable treatment of MRSA. Statement of Significance Early detection and treatment of MRSA infections are crucial for reducing public health risks. It is a significant challenge that develops sensitive in vivo diagnosis and complete elimination of drug-resistant bacterial infections. Herein, a nanoplatform has been developed for photoactivatable therapy of MRSA infections and deep tissue NIR-II imaging. This platform utilizes lanthanide-doped rare earth nanoparticles (RENPs) that are sensitized by a moiety-engineered near-infrared (NIR) dye TPEO-820. The TPEO-820 sensitized RENPs exhibit 5 times increase in the release of NO concentration for MRSA treatment compared to unsensitized RENPs, enabling precise therapy of MRSA infection both in vitro and in vivo. Moreover, the platform demonstrates NIR-II luminescence in vivo, allowing for sensitive imaging in deep tissue for MRSA infection.

A full-spectrum responsive B-TiO2@SiO2–HA nanotheranostic system for NIR-II photoacoustic imaging-guided cancer phototherapy

Xiaolu Guo,Changchun Wen,Qianxin Xu,Changping Ruan,Xing-Can Shen,Hong Liang

The second near-infrared (NIR-II) window (1000–1350 nm) usually offers further improved light penetration, a higher maximum permissible exposure (MPE), and a lower background signal. Development of NIR-II optical diagnosis and phototherapy technologies is of great significance for precise, efficient tumor therapy. In this work, a new type of Ti-based targeting agent (B-TiO2@SiO2–HA) nanotheranostic system with strong NIR-II absorption was designed and fabricated for the first time. Oxygen vacancies were formed in B-TiO2 and its band gap was narrowed, resulting in nanotheranostic systems with full-spectrum responses to stimulation with light. The experimental results showed that B-TiO2@SiO2–HA not only can enable high NIR-II photothermal conversion and provide excellent reactive oxygen species (ROS) production capacity, but also can enable high-resolution photoacoustic imaging (PAI) under NIR-II laser irradiation. Moreover, HA modification gives the nanotheranostic systems the useful ability to target high-CD44-expression tumor cells and tissues. In vitro and in vivo experiments demonstrated that B-TiO2@SiO2–HA exhibited a targeted photothermal/photodynamic (PTT/PDT) effect that produced tumor-cell ablation and apoptosis under the guidance of real-time NIR-II PA imaging. B-TiO2@SiO2–HA exhibits precise nanotheranostic potential for PAI-guided tumor-targeting phototherapy.
