Remendable Cross-Linked Alginate/Gelatin Hydrogels Incorporating Nanofibers for Wound Repair and Regeneration

Changgi Hong,Haeun Chung,Gyubok Lee,Dongwoo Kim,Zhuomin Jiang,Sang-Heon Kim,Kangwon Lee

Wound dressings made from natural-derived polymers are highly valued for their biocompatibility, biodegradability, and biofunctionality. However, natural polymer-based hydrogels can come with their own set of limitations, such as low mechanical strength, limited cell affinity, and the potential cytotoxicity of cross-linkers, which delineate the boundaries of their usage and hamper their practical application. To overcome the limitation of natural-derived polymers, this study utilized a mixture of oxidized alginate and gelatin with 5 mg/mL polycaprolactone (PCL):gelatin nanofiber fragments at a ratio of 7:3 (OGN-7) to develop a hydrogel composite wound dressing that can be injected and has the ability to be remended. The in situ formation of the remendable hydrogel is facilitated by dual cross-linking of oxidized alginate chains with gelatin and PCL/gelatin nanofibers through Schiff-base mechanisms, supported by the physical integration of nanofibers, thereby obviating the need for additional cross-linking agents. Furthermore, OGN-7 exhibits increased stiffness (γ = 79.4–316.3%), reduced gelation time (543 ± 5 to 475 ± 5 s), improved remendability of the hydrogel, and excellent biocompatibility. Notably, OGN-7 achieves full fusion within 1 h of incubation and maintains structural integrity under external stress, effectively overcoming the inherent mechanical weaknesses of natural polymer-based dressings and enhancing biofunctionality. The therapeutic efficacy of OGN-7 was validated through a full-thickness in vivo wound healing analysis, which demonstrated that OGN-7 significantly accelerates wound closure compared to alginate-based dressings and control groups. Histological analysis further revealed that re-epithelialization and collagen deposition were markedly enhanced in the regenerating skin of the OGN-7 group, confirming the superior therapeutic performance of OGN-7. In summary, OGN-7 optimized the synergistic effects of natural polymers, which enhances their collective functionality as a wound dressing and expands their utility across diverse biomedical applications.

Electrospun nanofiber regulates assembly of keratin and vimentin intermediate filaments of PANC-1 pancreatic carcinoma cells

Yiqun Wang,Yi Lu,Jinkang Gong,Yuan Yao

Intermediate filaments, together with actin microfilaments and microtubules constituent the cytoskeleton of mammalian cells, involving in various cellular activities. The roles of intermediate filaments in cell skeleton reorganization when responding with extracellular matrix (ECM) nanostructure are poorly understood yet. To unveil the effects of fibrous composition and orientation on cells, we developed electrospun nanofibers of varying topology and components, and the effects on assembly of intermediate filaments as keratin and vimentin were investigated in detail. We found that aligned nanofibers enhanced expression of E-cadherin and promoted assembly of keratin intermediate filaments. Meanwhile, the compositional variation show different preference on up-regulation of the two intermediate filaments. Compared to keratin, the assembly of vimentin intermediate filaments were promoted by incorporating bovine serum albumin (BSA) functionalized graphene oxide (BSA-GO) into polycaprolactone (PCL) nanofibers. Thus, our findings elucidate how the different physical factors of fibrous extracellular matrix affect the reorganization of cytoskeleton by assembly of keratin and vimentin intermediate filaments.

Sustained release of collagen VI potentiates sciatic nerve regeneration by modulating macrophage phenotype

Dan Lv,Lijuan Zhou,Xianyu Zheng,Yongcheng Hu

Although the peripheral nervous system has an intrinsic ability for repair and regeneration after injury, bridging long peripheral nerve defects remains a challenge. Functional nerve regeneration depends on interactions among axons, Schwann cells, fibroblasts and immune cells. Macrophages, as immune cells recruited early in this process, show polarization toward phenotypes that are detrimental or beneficial to tissue remodeling depending on the microenvironment of the scaffolds. In this study, we investigated the effects of macrophage phenotypes modulated by collagen VI on axonal regeneration and functional recovery by bridging a 15-mm-long sciatic nerve defect in rats. Our results showed that local delivery of collagen VI within a polycaprolactone (PCL) electrospun conduit increased the recruitment of macrophages and their polarization toward the pro-healing (M2) phenotype. In addition, the axonal regeneration and neurologic functional recovery in the PCL/collagen VI conduit group are equivalent to that of an autograft. In conclusion, the present study confirmed that PCL/collagen VI conduits with sustained release of collagen VI in the local microenvironment may, through triggering macrophage M2 polarization to enhance the nerve regeneration, suggest that our combined biomaterial-immunomodulatory system may be an attractive candidate for peripheral nerve regeneration.

Synthesis and thermo-responsive self-assembly behavior of amphiphilic copolymer β-CD–(PCL–P(MEO2MA-co-PEGMA))21 for the controlled intracellular delivery of doxorubicin

Beibei Lu,Lei Li,Lulu Wei,Xuhong Guo,Jun Hou,Zhiyong Liu

Well-defined amphiphilic β-cyclodextrin (β-CD) star-shaped copolymers with poly(ε-caprolactone)–poly(2-(2-methoxyethoxy)ethyl methacrylate)-co-poly(ethylene glycol)methacrylate) (β-CD–(PCL–P(MEO2MA-co-PEGMA))21 were synthesized via ring opening polymerization (ROP) and atom transfer radical polymerization (ATRP). These thermo-responsive copolymers exhibited a lower critical solution temperature (LCST) in water, which could be finely tuned by changing the feed ratio of PEGMA and MEO2MA. The LCST of star-shaped β-CD–(PCL–P(MEO2MA-co-PEGMA))21 increased from 26 to 62 °C with the increasing PEGMA content, the copolymers were characterized using Fourier transform infrared spectroscopy (FTIR), proton nuclear magnetic resonance (1H NMR) and gel permeation chromatography (GPC). The star-shaped copolymers could self-assemble into micelles in aqueous solution due to their amphiphilic properties resulting from the hydrophobic β-CD and PCL core and the hydrophilic P(MEO2MA-co-PEGMA) segments, which was investigated by dynamic light scattering (DLS) and transmission electron microscopy (TEM). The in vitro DOX release studies showed that DOX released from drug-loaded micelles in a thermo-sensitive manner. CCK-8 assays demonstrated that these star-shaped copolymers could possess low cytotoxicity against HeLa cells, and the DOX-loaded micelles exhibited a higher inhibition of the proliferation of HeLa cells in comparison with free DOX. Moreover, the results from confocal laser scanning microscopy (CLSM) revealed that these polymeric micelles could efficiently deliver and release DOX into the nuclei of HeLa cells. This kind of biodegradable, biocompatible and stimuli-responsive copolymer could serve as a promising material for drug delivery.

Fabrication of polydopamine-altered PCL/PLGA nanofibrous scaffolds on cerium to enhance cytocompatibility, microbial inhibition, and improved osteogenic properties for tissue regeneration in orofacial treatment

Chang Luo,Cheng Kang,Shi Qiu,Weiming Wang,Yiyi Lou,Xiaofei Wang,Ting Zhu

Biodegradable scaffolds have significant potential for directed tissue regeneration in craniofacial and oral applications and bone regeneration. We designed an innovative cerium-altered/collagen-clacked polycaprolactone/poly-lactic-co-glycolic acid (PC/PL) electrospun scaffolds (PC/PL-PD-Ce-CL) to address the deficiencies of existing scaffold materials regarding osteogenic and antimicrobial characteristics. Novel scaffolds were developed by electrospinning a fundamental PCL/PLGA matrix, impregnating in situ reduction with cerium nanoparticles (CeNPs), applying a polydopamine coating, and clacking with collagen. The mechanical tests and scanning electron microscope demonstrated that the distinctive NSs preserved their tensile strength and elasticity modulus post-modification. The cell proliferation CCK-8 assay indicated that the PC/PL-PD-Ce-CL NSs exhibited a superior MG63 cell proliferation and cell adhesion ratio compared to the rest of the NSs. The alkaline phosphatase (ALP) activity and the expression levels of BMP2 and RUNX2 in MG63 cells cultivated on PC/PL-PD-Ce-CL NSs for 7 and 14 days were considerably elevated compared to the controls. The PC/PL-PD-Ce-CL and PC/PL-PD-Ce NSs exhibited a broader antibacterial zone compared to the control NSs, and bacterial fluorescence diminished on the Ce-altered NSs of incubation with S. mutans and S. aureus . Our innovative PC/PL-PD-Ce-CL NSs improved cytocompatibility, osteogenesis, and antibacterial capabilities, demonstrating their promising therapeutic applications for orofacial treatment.

Large artificial bone from 3D printed polycaprolactone/β-tricalcium phosphate (3D PCL/β-TCP) effectively promoting MC3T3-E1 cell adhesion, proliferation, and new bone formation

Yuxin Niu,Fuxiang Song,Rong Shan,Qianjuan Zhao,Zhuang Chen,Na Wang,Weibo Xie

The use of 3D printing technology has advanced the bone tissue engineering, and constructing large artificial bones for repairing large-scale bone defects is highly significant. This study aimed to construct large artificial bones in a precise and controllable manner, focusing on repairing critical-sized bone defects. The researchers used 3D printing technology to synthesize 3D PCL/β-TCP, and then evaluated its ability to promote MC3T3-E1 cell adhesion, proliferation, and new bone formation through a series of characterizations. The results confirmed that 3D PCL/β-TCP, presented as a lattice structure similar to natural bone, could be used to prepare personalized artificial bone blocks based on the actual range of bone defects. The researchers also experimented with dipping 3D PCL/β-TCP bone blocks in different proportions of β-TCP and found that PT2 showed better mechanical and hydrophilic properties. Further experiments showed that PT2 presented excellent biocompatibility and outstanding capacity to promote MC3T3-E1 cell proliferation and adhesion, and that 3D PCL/β-TCP bone blocks possessed good osteogenic activity. Finally, it was suggested that PT2 exhibited the property of promoting new bone formation. These findings provide experimental data support for the repair of critical-sized bone defects.

Surface-fragmenting hyperbranched copolymers with hydrolysis-generating zwitterions for antifouling coatings

Liqin Mei,Xiaoqing Ai,Chunfeng Ma,Guangzhao Zhang

Zwitterionic polymers have attracted increasing attention due to their excellent fouling resistance ability and eco-friendliness. Yet, their non-degradability and hydrophilic nature limit their applications. In this study, we have prepared a novel surface-fragmenting hyperbranched copolymer with tertiary carboxybetaine ester (TCB) primary chains and poly(ε-caprolactone) (PCL) bridged chains, where the former and the latter can hydrolyze and degrade in marine environments, continuously generating zwitterions, so the polymer coating has a fouling resistant and renewable surface. Our study demonstrates that the degradation rate of the polymer is well controlled by the content of PCL bridges. Protein resistance and antibacterial assays show that the coating can inhibit the adhesion of protein and marine bacteria (Pseudomonas sp.). This new surface-fragmenting, self-regenerating hyperbranched zwitterionic copolymer has multiple applications in antifouling coatings.

Magnetic scaffold constructing by micro-injection for bone tissue engineering under static magnetic field

Zhengyi Xu,Yujie Zhang,Lang Zheng,He Cai,Xiangjun Yang,Yiyuan Xue,Qianbing Wan,Junyu Chen,Yijun Li

The utilization of static magnetic fields in conjunction with tissue engineering scaffolds has demonstrated significant potential in the restoration of bone defects. However, previous techniques used for fabricating magnetic tissue engineering scaffolds have shown certain shortcomings that require further investigation. In this study, a series of scaffolds made from a combination of polylactic acid (PLA), polycaprolactone (PCL), and iron tetraoxide (Fe 3 O 4 ) nanoparticles were designed and prepared using the microinjection molding technique. The composite scaffolds, which were doped with nanoparticles and fabricated through microinjection, exhibited exceptional physicochemical properties and biocompatibility. Additionally, the PLA/PCL/Fe 3 O 4 scaffolds were found to promote the proliferation and adhesion of bone marrow cells (BMSCs) under relatively low magnetic fields (25–30 mT). These findings suggest that PLA/PCL/Fe 3 O 4 scaffolds hold great potential for use in bone tissue engineering applications.

Implantable composite fibres with Self-supplied H2O2 for localized chemodynamic therapy

Gang Wang,Jiayu Gao,Yike Fu,Zhaohui Ren,Jie Huang,Xiang Li,Gaorong Han

The anticancer efficacy of chemodynamic therapy (CDT) is considerably restrained by the inadequate content of H 2 O 2 within tumor microenvironment (TME), unexpected oxidation of Fenton agents and low targeting efficiency of particulate therapeutic platforms. Herein for the first time, an implantable CDT platform was designed and synthesized by combining Fe 0 nanocrystal embedded mesoporous silica nanoparticles (FeMSN) and PCL-gelatin fibres (PG fibres), denoted as [email protected] fibres. FeMSN was fabricated via in situ reduction of ferrous ions within the mesopores of MSNs, and assembled at the surface of electrospun PG fibres by electrostatic interaction to form a fibrous mesh. The findings reflected that, in an acid condition, FeMSN nanoparticles can liberate from the composite fibres, and be effectively uptaken by 4T1 cancer cells. The FeMSN released may react with oxygen to generate H 2 O 2 in the acidic microenvironment, and subsequently transform H 2 O 2 to intracellular hydroxyl radicals, inducing enhanced cell-killing effect. By implanting the composite fibres locally at the tumor site of mice, promoted anticancer efficacy was observed with comparison of the intratumoral-injected FeMSN nanoparticles. This study suggests that [email protected] composite fibres can serve as a potential localized PDT platform for effective antitumor purposes, which may spark a series of follow-on investigations for localized cancer therapeutic technology.

LBL deposition of chitosan/heparin bilayers for improving biological ability and reducing infection of nanofibers

Long Li,Xianguo Wang,Dan Li,Jinfa Qin,Ming Zhang,Kaijie Wang,Jinping Zhao,Li Zhang

The complexity of cardiovascular disease requires that the materials for preparing vascular grafts possess good biocompatibility, high mechanical property, and even some excellent additional properties. In this study, polycaprolactone (PCL) with good mechanical property and natural source silk fibroin (SF) were electrospun into PCL/SF nanofibers to obtain the nanofibrous substrate. With the addition of SF, the mechanical property of PCL/SF nanofibrous mats was maintained to a certain extent. While, the hydrophilicity of PCL/SF nanofibrous mats was greatly improved which is more suitable for immersive layer-by-layer assembly (LBL). The oppositely charged heparin (Hep) and chitosan (CS) were alternatively deposited on the surface of PCL/SF nanofibers via LBL. After implanting human umbilical vein endothelial cells (HUVEC) on the LBL-structured nanofibrous mats for 48 h, it was confirmed that the CS/Hep bilayers enhanced the biocompatibility of the nanofibers. Furthermore, the results of the antibacterial test showed that the antibacterial effects of the LBL-structured nanofibrous mats for Escherichia coli and Staphylococcus aureus were both achieved 95% when the number of Hep/CS bilayer was 10. It can be demonstrated that the LBL-structured nanofibrous mats with improved biocompatibility and reduced infectivity had been prepared successfully, and can be potentially used in vascular grafts.

Electrospun vein grafts with high cell infiltration for vascular tissue engineering

Zhikai Tan,Xiangkai Gao,Tong Liu,Yikun Yang,Juchang Zhong,Chunyi Tong,Yongjun Tan

Demand is increasing for functional small-diameter vascular grafts (diameter   <   6   mm) for clinical arterial replacement. In the present study, we develop a bilayer poly(ε-caprolactone, PCL) fibrous vascular graft consisting of a thin internal layer made of longitudinally aligned fibers and a relatively thick highly porous external layer. The internal layer provides a scaffold with the necessary mechanical strength and enhances the growth of endothelial cells, whereas the external layer enhances cell motility through the scaffold bulk. The biocompatibility and biological performance of bilayer fibrous scaffolds are evaluated by in vivo experiments, molecular biology, and histology studies. Our bilayer scaffolds demonstrate much better fiber alignment and higher porosity than do normal electrospun vascular grafts with randomly distributed fibers. The results suggest that the proposed grafts can overcome limitations owing to the inadequate porosity, small pores, and poor cell infiltration of scaffolds fabricated by conventional electrospinning. The unique structure of bilayer scaffolds is satisfactory and promotes cell proliferation, collagen–fiber deposition, and ingrowth of smooth muscle cells and endothelial cells in vivo . The results of this study illustrate the strong potential of such bilayer fibrous scaffolds for vascular tissue engineering and regeneration.

Development and fabrication of co-axially electrospun biomimetic periosteum with a decellularized periosteal ECM shell/PCL core structure to promote the repair of critical-sized bone defects

Shuyi Li,Rongli Deng,Xuenong Zou,Qiong Rong,Jiali Shou,Zilong Rao,Wanqiu Wu,Gang Wu,Daping Quan,Miao Zhou,Tim Forouzanfar

Periosteum is crucial to the initial healing of bone defects because it provides a stable periosteum-specific microenvironment and abundant osteogenic cells towards bone repair. However, the usage of autologous periosteum is restricted for limited availability. Xenogeneic decellularized periosteum (DP) preserves periosteum-specific cues but the hardly-controlled physicochemical properties render it inconvenient to use widely. Our study aimed to fabricate a tissue-engineered periosteum (TEP) with a continuous periosteum-specifically bioactive surface and tailored physicochemical properties by co-axial electrospinning of poly(ε-caprolactone) (PCL)/periosteal decellularized extracellular matrix (dECM) to promote bone defects healing. Before use, DP was confirmed for effective removal of residual DNA and well preservation of biological components, e.g., collagen and glycosaminoglycans . After optimization of fabrication processes, PCL/dECM co-axially electrospun membrane (PEC) bore an exquisite core-shell structure, which was effectively equipped with composite advantages of dECM and PCL. Notably, PEC stood out in terms of tensile strength and long-term durability within a physiological environment compared to that of the dECM uniaxial electrospun membrane (ECM). Besides, PEC exhibited remarkably better cell proliferation , migration, bio-mineralization, and osteogenic properties as compared to that of the electrospun PCL membrane. Moreover, PEC-TEP could significantly enhance the recovery of critical-sized bone defects in rats than that of PCL-TEP. To the best of our knowledge, biomimetic PEC-TEP was fabricated and used for the first time to repair bone defects. This novel strategy is promising to fabricate a dECM-based advanced membrane for tissue engineering .
