HA greatly suppresses ·OH and halves 1 O 2 formation compared with plastic-derived DOM. • PS-DOM shows over threefold stronger ·O 2 − inhibition than PE-DOM. • Aromatic ketones in plastic-derived DOM prolong 3 DOM ⁎ lifetime while HA moieties quench ROS. • Energy and electron transfer pathways define DOM-controlled ROS dynamics. Porous carbon-based photocatalysts offer a promising platform for visible light-driven water treatment, yet dissolved organic matter (DOM) from natural and anthropogenic sources can substantially modulate reactive oxygen species (ROS) dynamics, affecting pollutant degradation. A systematic understanding of how DOM composition and molecular structure influence ROS generation remains limited. Here, a boron-doped covalent triazine framework (B-CTF) was employed as a model photocatalyst, with humic acid (HA) and microplastic-derived DOM (PE-DOM, PS-DOM) representing natural and anthropogenic coexisting components. Adsorption experiments revealed that B-CTF exhibited enhanced maximum adsorption capacities ( q m ) for HA, PE-DOM, and PS-DOM and accelerated intraparticle diffusion, demonstrating efficient DOM-catalyst interfacial interactions. ROS analysis identified 1 O 2 and ·OH as dominant species, strongly modulated by DOM. HA suppressed ·OH generation by 2.00 × 10 −2 h −1 , ∼1.0 order of magnitude higher than PE-DOM and PS-DOM, and reduced 1 O 2 formation nearly twofold relative to plastic-derived DOM. PS-DOM caused the strongest ·O 2 − inhibition (8.34 × 10 −2 h −1 , 3.3-fold higher than PE-DOM), while PE- and PS-DOM moderately suppressed overall ROS. Fluorescence component analysis indicated that aromatic ketones in plastic-derived DOM facilitate ROS persistence, while humic-like substances in HA promote ROS quenching, highlighting the decisive role of DOM composition in interfacial ROS regulation. These findings elucidate the mechanistic pathways by which distinct DOM types regulate energy and electron transfer at the B-CTF interface, thereby controlling ROS yields, lifetimes, and spatial distribution. They also provide quantitative guidance for designing DOM-resilient photocatalysts and developing targeted water treatment strategies. Natural and anthropogenic DOM (HA, PE-DOM, PS-DOM) critically regulate ROS formation, reactivity, and persistence in B-CTF photocatalytic systems via distinct energy- and electron-transfer pathways. Specific structural components within DOM quench or sustain ROS, thereby reshaping degradation routes and pollutant removal efficiency. These mechanistic insights highlight the structure-activity relationship governing DOM + B-CTF interactions and provide guidance for developing photocatalysts with resilience to DOM interferences in complex aquatic environments. Diabetic bone defects suffer from impaired self-repair due to pathological microenvironments characterized by excessive reactive oxygen species (ROS) accumulation and immune dysregulation. To address this challenge, we developed a multifunctional composite hydrogel (Gel/ZR) by integrating resveratrol (RSV)-loaded ZIF-8 nanoparticles into an oxidized alginate-gelatin self-healing matrix. This system leverages the pH-responsive degradation of ZIF-8 to enable targeted release of RSV and Zn 2+ in acidic bone defect sites. Comprehensive characterization confirmed the hydrogel's injectability, self-healing properties, and sustained drug release kinetics. In vitro studies demonstrated that Gel/ZR effectively scavenged ROS, restored mitochondrial membrane potential in bone marrow mesenchymal stem cells (BMSCs), and enhanced osteogenic differentiation. It concurrently promoted pro-regenerative M2 macrophage polarization. In diabetic rat cranial defects, Gel/ZR accelerated bone regeneration, while modulating local immune responses toward tissue-reparative phenotypes. These results validate Gel/ZR as a synergistic therapeutic platform that remodels diabetic bone microenvironments through coordinated ROS elimination, immunomodulation, and osteoinduction.

Bone defects resulting from trauma, malignant tumors, or infections are common clinical conditions. Current clinical treatments for bone defects, however, are associated with secondary injury, poor morphological matching, and immune rejection, falling short of clinical needs. Multifunctional bioscaffolds with osteogenic induction capability have emerged as a highly promising therapeutic strategy. In this study, 3D printing technology was utilized to fabricate scaffolds integrating ROS scavenging and osteogenic differentiation dual functions, and their physicochemical properties and biocompatibility were systematically investigated. Polycaprolactone (PCL) and strontium-magnesium-doped calcium silicate (SMCS) were selected to prepare PCL/SMCS scaffolds with varying SMCS ratios. SMCS incorporation effectively enhanced scaffold hydrophilicity and accelerated degradation. Cell culture experiments confirmed good biocompatibility of the PCL/SMCS scaffolds. Subsequently, cerium-doped SMCS bioceramics were prepared via a post-impregnation process. Notably, the PCL/0.1M Ce-SMCS scaffold exhibited optimal compressive performance, achieving a strength of 22.78 MPa—a 26.42% increase over the PCL/4SMCS scaffold. Compared with PCL/4SMCS, Ce doping promoted cell proliferation and adhesion, conferred ROS scavenging ability, and in vitro osteogenic assays indicated that low-content Ce-SMCS enhanced osteogenic differentiation.

Nanozyme-catalyzed reactive oxygen species (ROS) generation
has
emerged as a promising strategy for advanced ROS-based therapeutics.
However, achieving efficient ROS production with favorable catalytic
kinetics in a confined microenvironment remains challenging. Herein,
a cancer-targeting heterogeneous Janus nanozyme (JN, Cu 2– x Se–Au-TLS11a) was rationally fabricated via
a facile modular synthesis and assembly. Upon near-infrared II (NIR-II)
light irradiation, the well-designed JN realized multimodal synergistic
anticancer effects via three coordinated mechanisms: (i) efficient
photothermal conversion with a high efficiency of 61.9% for photothermal
therapy (PTT); (ii) NIR-II-triggered hot-electron transfer from the
Au domain to Cu 2– x Se promoted the
generation of Cu + , which accelerated Fenton-like reactions
to produce hydroxyl radicals (•OH); and (iii) NIR-II photoexcitation
induced the formation of electron–hole (e – -h + ) pairs on the Janus nanostructure, enabling additional
photocatalytic •OH generation. Under NIR-II light activation,
the JNs, integrating photothermal, photoenhanced chemodynamic, and
photocatalytic effects, demonstrated highly specific and potent in
vitro anticancer efficacy, achieving an IC 50 value of approximately
12.5 μg mL –1 against HepG2 cells and reducing
cell viability to 13.8%, thereby providing an innovative paradigm
for designing spatiotemporally controlled, ROS-amplifying nanoplatforms
toward precision cancer therapy.

Photodynamic therapy (PDT) is a minimally invasive anti-tumor treatment modality that relies on the cytotoxicity of reactive oxygen species (ROS) produced by light-activated photosensitizers. However, traditional photosensitizers only produce ROS upon photoexcitation, resulting in limited treatment duration of ROS, which largely restricts the therapeutic efficacy of PDT. To address such limitation, here we engineered a copper doped gold nanoclusters with holo-transferrin (Tf) as ligand (Tf@ACNCs), realizing the generation of superoxide anions (O 2 − ) under irradiation and the sustained production of hydroxyl radicals ( OH) without any limitations on photoexcitation periods. Specifically, benefiting from copper doping, Tf@ACNCs exhibit a lower highest occupied molecular orbital (HOMO)-lowest unoccupied molecular orbital (LUMO) energy gap, which is favorable for easier photoexcitation, and a lower Gibbs free energy barrier, which is conducive to stronger peroxidase (POD)-like catalysis compared to undoped gold nanoclusters (Tf@AuNCs). Therefore, Tf@ACNCs can significantly prolong the ROS generation for up to 6 h following irradiation through its multi-source ROS generation capability. Moreover, the prolongation of ROS effectively promoted ferroptosis and apoptosis, achieving a 41.38 % improvement in the anti-tumor efficacy of Tf@ACNCs compared with Tf@AuNCs. Overall, this ROS prolongation strategy affords a novel reference for enhancing PDT.

Malignant tumors are one of the most dangerous types of diseases seriously threatening global health, and how to better treat them is a difficult issue for the medical community. Podophyllotoxin and its derivatives were known to have good antitumor efficacy. But their poor water solubility, toxicity to normal tissues, susceptibility to drug resistance, and low absorption by cancer cells have limited their clinical application. A striking feature of cancer cells compared to normal cells is the existence of intracellular overproduction of reactive oxygen species (ROS), frequently used as stimuli for drug release from nanocarriers. In this study, we developed a ROS responsive twin prodrug PPT-S-PPT, in which two podophyllotoxins (PPTs) were coupled by 3,3′-thiodipropionic acid via a thioether bond. PPT-S-PPT can form spherical nanoparticles with a mean diameter of about 200 nm in water. These self-assembled nanoparticles maintained good storage stability for up to 3 weeks. In vitro release study showed that H 2 O 2 can accelerate the drug release of PPT-S-PPT NPs in PBS. Both HPLC assay and fluorescence study showed that the intracellular uptake of PPT-S-PPT NPs was significantly higher than free drug or free fluorescence probe. The PPT-S-PPT NPs were up-taken by Hela cells via multiple endocytic pathways, and lipid rafts may play a greater role in the endocytosis of PPT-S-PPT NPs. In addition, the PPT-S-PPT NPs showed higher inhibitory effect (almost 14-fold) than free PPT against Hela cells after 24h treatment. The potent cellular inhibition effect of NPs might be due to the increased cellular uptake as well as the enhanced intracellular drug release resulting from the ROS responsiveness. Moreover, PPT-S-PPT NPs revealed favorable cytocompatibility and good hemocompatiblity. This carrier-free ROS-sensitive twin drug nanoassebly provides possibilities for the development of ROS-responsive prodrugs or dimer nanoparticles for efficient cancer therapy.

Osteoporosis is characterized by increased osteoclast activity, which is strongly associated with increased levels of reactive oxygen species (ROS). Fraxin, a natural coumarin glycoside, has shown anti-inflammatory and antioxidant properties, but its effects on bone homeostasis are obscure. The effects of fraxin on osteoclast formation and activation were measured via an in vitro osteoclastogenesis assay. Mitochondrial and total ROS production were evaluated with the aid of MitoSOX Red and DCFH-DA, respectively. Osteoclast-related gene expression analysis was performed via qPCR. Key proteins related to osteoclast formation, ROS scavenging, and ROS-regulated signaling, such as mitogen-activated protein kinases (MAPKs), NF-κB pathways, and nuclear factor of activated T cells 1 (NFATc1) signaling, were detected via western blotting. An ovariectomized mouse model was used to evaluate the therapeutic effects of fraxin in vivo. Fraxin inhibited osteoclastogenesis and osteoclast-related gene expression. Mechanistically, fraxin restored the levels of ROS-scavenging enzymes to inhibit ROS accumulation, eventually downregulating ROS-regulated signaling. The measurement of Micro-CT and histological analyses revealed that fraxin treatment significantly reduced OVX-induced bone loss by decreasing the number of osteoclasts. Fraxin shows promise as a novel therapeutic agent for osteoclast-related bone diseases, especially osteoporosis.

Objective The incidence of metabolic dysfunction-associated steatohepatitis (MASH) is increasing because people’s dietary habits are dominated by high caloric intake and sedentary lifestyles, leading to the accumulation of lipid, reactive oxygen species (ROS) and inflammation. However, treating MASH remains a challenge. Methods Two-dimensional (2D) niobium carbide (Nb4C3) MXene nanoenzymes (MXenzymes) possess both antioxidant and anti-inflammatory properties and have attracted considerable attention in the tumor and engineering fields. The Nb4C3 MXenzyme was developed for MASH therapy and exhibited biosafety and antilipid peroxidation activity. Results Nb4C3 reduced excessive ROS and proinflammatory cytokine levels through its antilipid peroxidation activities, resulting in the inhibition of hepatocyte lipid accumulation and inflammation in a methionine–choline-deficient diet (MCD)-induced murine MASH model. Mechanistically, Nb4C3 not only inhibited lipid accumulation and disrupted lipid metabolism in hepatocytes but also attenuated fatty acid-induced cell death by reducing intracellular ROS levels, which significantly promoted the polarization of M1 macrophages to M2 macrophages by alleviating oxidative stress and suppressing inflammatory factor expression. Conclusion The Nb4C3 MXenzyme can be used as a multifunctional bioactive material to alleviate hepatic steatosis and inflammation in MASH mice through its robust antioxidant and anti-inflammatory activities.

Antibacterial sonodynamic therapy (aSDT) has emerged as a promising non-invasive antimicrobial modality utilizing ultrasound activation. This study employed curcumin (CUR) as a sonosensitizer to systematically investigate the mechanisms underlying aSDT. We first examined ultrasound (US)-induced intracellular reactive oxygen species (ROS) generation and subsequently evaluated the bactericidal enhancement mediated by microbubble-augmented cavitation, including membrane disruption and apoptotic pathways. A comprehensive experimental approach was implemented, including: (1) bacterial viability assays on solid media coupled with biomass quantification, (2) flow cytometric apoptosis detection, (3) intracellular ROS measurement, (4) gene expression changes through RNA-seq and qRT-PCR, and (5) scanning electron microscopy (SEM) analysis of Pseudomonas aeruginosa morphology. Results demonstrated a dose-dependent relationship between cavitation intensity and antibacterial efficacy. Although US can stimulate intracellular ROS elevation, experimental results indicated that aSDT operates primarily through two mechanisms: ultrasound-induced mechanical disruption of cell membranes, and apoptosis triggered by extracellular ROS generated via US/sonosensitizer synergy that enter cells. Genomic analysis further elucidated that aSDT-induced bacterial apoptosis originated from the synergistic effect of ultrasonic cavitation and sonosensitizer. It differentially regulated the expression of key genes in P. aeruginosa (downregulating PA4211, PA0876, PA3361 and upregulating PA3570, PA2433, PA4880), damaged the biofilm structure and inhibited its remodeling. Meanwhile, it disrupted the redox homeostasis and metabolic balance, utimately leading to irreversible damage to the cell membrane and a massive accumulation of intracellular ROS.

Magnetic particle imaging (MPI) has emerged as a versatile biomedical imaging modality, yet a significant challenge persists in the absence of activatable MPI probes for targeted imaging of disease biomarkers. In this study, a reactive oxygen species (ROS)-responsive ordered assembly of magnetic nanoparticles (MNPs) is reported, engineered through the meticulous design of magnetic nanoparticle building blocks and ROS-responsive polymeric ligands, enabling precise control over the assembly structure. This ordered configuration amplifies magnetic dipole-dipole interactions, raising the energy barrier during nonequilibrium dynamic magnetization and effectively quenching the MPI signal. By modulating the assembly and disassembly of these ordered structures in response to ROS, this nanoprobe achieves ROS-dependent off/on MPI signal regulation. Consequently, the probe enables the monitoring of early pathological ROS change in a mice model of early acute liver injury, facilitating highly sensitive monitoring of the ROS level-dependent severity of the disease. This work represents the inaugural application of microenvironment-responsive MPI probes for the early diagnosis of ROS-associated diseases. The introduction of ordered assembly structures for MPI signal tuning offers a promising translational approach in the development of next-generation activatable MPI probes.

To synthesize an effective and versatile nano-platform serving as a promising carrier for controlled drug delivery, visible-light-induced diselenide-crosslinked polyurethane micelles were designed and prepared for ROS-triggered on-demand doxorubicin (DOX) release. A rationally designed amphiphilic block copolymer, poly(ethylene glycol)-b-poly(diselenolane diol-co-isophorone diisocyanate)-b-poly(ethylene glycol) (PEG-b-PUSe-b-PEG), which incorporates dangling diselenolane groups within the hydrophobic PU segments, was initially synthesized through the polycondensation reaction. In aqueous media, this type of amphiphilic block copolymer can self-assemble into micellar aggregates and encapsulate DOX within the micellar core, forming DOX-loaded micelles that are subsequently in situ core-crosslinked by diselenides via a visible-light-triggered metathesis reaction of Se-Se bonds. Compared with the non-crosslinked micelles (NCLMs), the as-prepared diselenide-crosslinked micelles (CLMs) exhibited a smaller particle size and improved colloidal stability. In vitro release studies have demonstrated suppressed drug release behavior for CLMs in physiological conditions, as compared to the NCLMs, whereas a burst release of DOX occurred upon exposure to an oxidation environment. Moreover, MTT assay results have revealed that the crosslinked polyurethane micelles displayed no significant cytotoxicity towards HeLa cells. Cellular uptake analyses have suggested the effective internalization of DOX-loaded crosslinked micelles and DOX release within cancer cells. These findings suggest that this kind of ROS-triggered reversibly crosslinked polyurethane micelles hold significant potential as a ROS-responsive drug delivery system.

Diabetic wound healing remains a significant challenge due to complex pathological mechanisms, including prolonged inflammation, excessive reactive oxygen species (ROS) accumulation, angiogenesis dysfunction, and increased susceptibility to bacterial infection. In this study, we developed a dual ROS/glucose-responsive quercetin-loaded supramolecular hydrogel (GPQ hydrogel) for treating diabetic wounds. The hydrogel was fabricated by incorporating quercetin (QUE) into a guanosine–phenylboronic acid (GP) hydrogel network through dynamic borate ester bonds. Structural characterization revealed the formation of a typical G-quadruplex structure in the GPQ hydrogel. The dual responsiveness to ROS and glucose enabled the controlled release of QUE, effectively addressing the abnormal wound microenvironment in diabetes. In vitro studies demonstrated the excellent antibacterial, antioxidant, anti-inflammatory, and pro-angiogenic properties of the GPQ hydrogel. Furthermore, the in vivo diabetic wound healing study using a full-thickness wound model in streptozotocin-induced diabetic rats showed that the GPQ hydrogel significantly accelerated wound closure, enhanced re-epithelialization and collagen deposition, and promoted angiogenesis compared to the control and GP hydrogel groups. Immunofluorescence analysis confirmed the superior antioxidant and pro-angiogenic effects of the GPQ hydrogel in the wound microenvironment. This study presents a promising multifunctional biomaterial for effectively managing diabetic wounds.
