Oral spatial-to-point cascade targeting “sugar-coated bullets” for precise and safe chemotherapy by intervention Warburg effect

Xiudan Wang,Wen Guo,Jianan Han,Jia Li,Qinfu Zhao,Yuling Mao,Siling Wang

Glycolysis plays a vital role in the development and progression of tumors. Inhibiting glycolysis via smart and safe methods serves as a promising target for cancer therapy. Here, an oral “sugar-coated bullet” aiming at intervening Warburg effect is designed by coating colloidal mesoporous silica nanoparticles (CMS) encapsulating glycolysis inhibitor shikonin (SHK) with dextran , namely DCMS/SHK. The solubility and drug-loading capacity of SHK were enhanced by the special structure of CMS. Besides, the tempting bullets possess the spatial-to-point cascade targeting ability in delivering SHK from the colonic lumen to colon cancer cells and finally to PKM2 . After DCMS/SHK reaches the colon, the dextran is hydrolyzed by dextranase especially existing in the colon site to glucose and the carriers become glucose-coated nanoparticles. The glucose-cloak nanoparticles would be largely endocytosed by tumor cells and complete the efficient delivery of SHK. The encapsulated SHK can prevent the glycolysis of cancer cells and thus inhibit tumor growth effectively. This work presents an ingenious cascade colon-targeting strategy to treat colon cancer by destroying cell energy metabolism.

Long-Term Exposure to Phenanthrene Induced Gene Expressions and Enzyme Activities of Cyprinus carpio below the Safe Concentration

Xin Kang,Dongpeng Li,Xiaoxiang Zhao,Yanfeng Lv,Xi Chen,Xinshan Song,Xiangyu Liu,Chengrong Chen,Xin Cao

Phenanthrene (PHE) is a typical compound biomagnified in the food chain which endangers human health and generally accumulates from marine life. It has been listed as one of the 16 priority PAHs evaluated in toxicology. In order to evaluate the changes ofCYP1A GSTmRNA expression and EROD GST enzyme activity in carp exposed to lower than safe concentrations of PHE. Long-term exposure of carp to PHE at lower than safe concentrations for up to 25 days. The mRNA expression level and cytochrome P450 (CYP1A/EROD (7-Ethoxylesorufin O-deethylase)) and glutathione S-transferase (GST) activity were measured in carp liver and brain tissue. The results showed that PHE stress induced low-concentration induction and high-concentration inhibition ofCYP1Aexpression and EROD enzyme activity in the liver and brain of carp. In both two organs, GST enzyme activity was also induced. However, the expression ofGSTmRNA was first induced and then inhibited, after the 15th day. These results indicate that long-term exposure to PHE at lower than safe concentrations still poses a potential threat to carp’s oxidase system and gene expression. Keywords:Cyprinus carpio;phenanthrene ecotoxicology;cytochrome P4501A;7-ethoxylesorufin O-deethylase;glutathione S-transferase

Biodegradable MoOx@MB incorporated hydrogel as light-activated dressing for rapid and safe bacteria eradication and wound healing

Yifan Wang,Huiqin Yao,Yan Zu,Wenyan Yin

Wounds infected with drug-resistant bacteria are hard to treat, which remains a serious problem in clinical practice. An innovative strategy for treating wound infections is thus imperative. Herein, we describe the construction of a nanocomposite from biocompatible poly(vinyl alcohol) (PVA)/polyethylene glycol (PEG) hydrogel loaded biodegradable MoOx nanoparticles (NPs) and photosensitizer methylene blue (MB), denoted as MoOx@MB-hy. By incorporating MoOx@MB NPs, the nanocomposite hydrogel can act as a photoactivated wound dressing for near-infrared-II 1064 nm and 660 nm laser synergetic photothermal–photodynamic therapy (PTT–PDT). The key to PTT-induced heat becomes the most controllable release of MB from MoOx@MB-hy to produce more 1O2 under 660 nm irradiation. Importantly, MoOx@MB-hy can consume glutathione (GSH) and trap bacteria nearer to the distance limit of ROS damage to achieve a self-migration-enhanced accumulation of reactive oxygen species (ROS), thereby conquering the intrinsic shortcomings of short diffusion distance and lifetime of ROS. Consequently, MoOx@MB-hy has high antibacterial efficiencies of 99.28% and 99.16% against Ampr E. coli and B. subtilis within 15 min. Moreover, the light-activated strategy can rapidly promote healing in wounds infected by drug-resistant bacteria. This work paves a way to design a novel nanocomposite hydrogel dressing for safe and highly-efficient antibacterial therapy.

Safe disposal of hazardous waste incineration fly ash: Stabilization/solidification of heavy metals and removal of soluble salts

Xuankun Wei,Feng Xie,Chunling Dong,Pengju Wang,Jiyun Xu,Feng Yan,Zuotai Zhang

Hazardous waste incineration fly ash (HFA) is considered a hazardous waste owing to the high associated concentrations of heavy metals and soluble salts . Hence, cost effective methods are urgently needed to properly dispose HFA. In this study, geopolymers were prepared by alkali-activation technology to stabilize and solidify heavy metals in HFA. In addition, the effects of three different aluminosilicates (metakaolin, fly ash, and glass powder) on the heavy metal immobilization efficiency were investigated. Because the soluble salt content of HFA is too high for their direct placement in flexible landfill sites and water washing can lead to heavy metal leaching, water-washing experiments were conducted after alkali-activation treatment to remove soluble salts. The results suggest that the concentrations of heavy metals leached from geopolymers can satisfy the Chinese Standard limits (GB18598-2019) when the addition of aluminosilicates exceeds 20 wt%. More than 77% of Cl − and >64% of SO 4 2− in geopolymers could be removed via water-washing treatment. The Zn leaching concentration was maintained below approximately 0.52 ppm. After alkali-activation treatment, the water-washing process could efficiently remove soluble salts while inhibiting heavy metal leaching. Sodium-aluminosilicate-hydrate (N-A-S-H) gel, a product of the geopolymerization process in this study, was demonstrated to act as a protective shell that inhibited heavy metal leaching. Hence, HFA-based geopolymers are considered suitable for disposal in flexible landfills.

Self-targeting platinum(IV) amphiphilic prodrug nano-assembly as radiosensitizer for synergistic and safe chemoradiotherapy of hepatocellular carcinoma

Xiaohui Xiao,Yupeng Wang,Jieyao Chen,Peng Qin,Peiyao Chen,Dongfang Zhou,Yue Pan

Chemoradiotherapy is a widely used treatment for patients with malignancies such as hepatocellular carcinoma (HCC). However, it remains challenging to realize safe and synergistic chemotherapy and radiation sensitization . Herein, we design a self-targeting nano-assembly (STNA) based on platinum(IV)-lactose amphiphilic prodrug for synergistic and safe chemoradiotherapy of HCC. The Pt STNA would improve the tumor accumulation due to the targeting ability of lactose to HCC cells. After receptor-mediated endocytosis , Pt STNA would release cisplatin(II) in cancer cells to form DNA-binding, thus inducing DNA damage and cell apoptosis. Meanwhile, the DNA-binding also causes cell cycle arrest in the radiation-sensitive G2/M phase by the up-regulation of phosphorylated checkpoint kinase 1 (p-Chk1) expression. Furthermore, under X-ray irradiation, Pt STNA as radiosensitizer possesses a strong X-ray attenuation ability to deposit more energy, thus elevating the level of reactive oxygen species (ROS) to amplify the cell-killing effect of radiotherapy in the G2/M phase with increased DNA damage. As a result, Pt STNA exhibits significant synergistic therapeutic effects in chemoradiotherapy with no adverse effects in vitro and in vivo . Overall, we present a novel self-targeting nano-assembly strategy based on widely used Pt drugs for synergistic chemotherapy and radiation sensitization of HCC treatment.

A P/Si-containing polyethylenimine curing agent towards transparent, durable fire-safe, mechanically-robust and tough epoxy resins

Cheng Wang,Siqi Huo,Guofeng Ye,Pingan Song,Hao Wang,Zhitian Liu

Epoxy resin (EP) features superior comprehensive performances, but inherent flammability seriously limits its industrial applications. Current flame-retardant strategies often bring about enhanced flame retardancy but deteriorated mechanical, thermal and optical properties and neglect flame retardant durability. Herein, a phosphorus/silicon-containing polyethylenimine (PES) is designed as a multifunctional curing agent for EP. Replacing polyethylenimine (PEI) with PES maintains high visible light transmittance of EPs and improves the UV-blocking properties. The EP cured by 30 wt% PES (EP/30PES) exhibits enhanced mechanical properties compared with the EP cured by 15 wt% PEI (EP/15PEI). EP/30PES achieves a limiting oxygen index (LOI) of 35.0% and a UL-94 V-0 rating, and its peak smoke release rate (PSPR) reduces by 63.8% relative to that of EP/15PEI. Hence, the great flame retardancy, smoke suppression and mechanical performances of EP/30PES enable it to outperform previous flame-retardant EP/aliphatic amine counterparts. Meanwhile, EP/30PES shows durable flame retardancy, which still achieves a UL-94 V-0 classification after acid/alkali/aging resistance tests. This work provides an advanced design of multifunctional curing agents to create durable fire-safe EPs with great UV-shielding and mechanical performances.

A multifunctional and environmentally safe superhydrophobic membrane with superior oil/water separation, photocatalytic degradation and anti-biofouling performance

Zuozhu Yin,Feng Yuan,Mingshan Xue,Yahui Xue,Yu Xie,Junfei Ou,Yidan Luo,Zhen Hong,Chan Xie

Wastewater is typically complicated with spilled oil, water soluble toxic dyes and microorganisms, making it hard to be processed and causing a significant threat to the environmental safety and human health. In this paper, we demonstrate a simple solution immersion method to obtain a multifunctional cellulose-based membrane (CBM) that possesses both superhydrophobicity with a water contact angle of 163° and superior functionalities including self-cleaning, oil–water separation, anti-biofouling, and photocatalytic degradation capabilities. The achievement of separation efficiency (96%), comparatively high flux (141 L·m −2 ·h −1 ) and recyclable (7 times) oil/water separation performance is attributed to the robust superhydrophobicity enabled by the synergy of metal oxide (i.e., CuO) nanostructure coating and stearic acid (SA) modification. The superhydrophobic CBM also preferentially adsorbs organic dyes in aqueous solution, e.g., methylene blue (MB), promoting their efficient decomposition (about 70.3% of MB decomposed in 3 h) with high recyclability under UV irradiation. Most remarkably, the CBM exhibits superior anti-biofouling capability and persistently resists the algae adhesion in long duration (over 20 days), as a result of the self-cleaning ability as well as the antimicrobial property of CuO nanoparticles. Our finding here paves the way to use simple, cost-effective, environmentally safe, and reliable method to fabricate multifunctional materials for wastewater treatment in complex environments.

Interfacial engineering of polypropylene separator with outstanding high-temperature stability for highly safe and stable lithium-sulfur batteries

Mao Yang,Jue Nan,Wei Chen,Anjun Hu,He Sun,Yuanfu Chen,Chunyang Wu

The thermal shrinkage of commercial polypropylene (PP) separator at high temperature seriously restricts the development of safe and stable lithium-sulfur (Li-S) batteries. To address such issue, herein, we present an interfacial engineering strategy by coating conductive polypyrrole (PPY) modified lithium-montmorillonite (PPY/Li-MMT) on the PP separator. The PPY/Li-MMT can service as an effective heat release layer to enhance the high-temperature stability of Li-S batteries. The engineered PP separator exhibits superior performance: it shows nearly no thermal shrinkage even up to 150 °C. Most importantly, the interface engineered separator has not be punctured during the hot piercing testing and the Li-S batteries deliver outstanding thermal stability even at 80 °C. In addition, the PPY/Li-MMT modified PP separator owns very high electrolyte uptake of 348.6%, large ionic conductivity of 3.63 mS cm −1 and stable performance over 600 cycles. This work provides an effective interfacial engineering strategy by coating PPY/Li-MMT interlayer to significantly improve the electrochemical performance and particularly the high-temperature stability of Li-S batteries.

Synergistic antibacterial activity of physical-chemical multi-mechanism by TiO2 nanorod arrays for safe biofilm eradication on implant

Xiangyu Zhang,Guannan Zhang,Maozhou Chai,Xiaohong Yao,Weiyi Chen,Paul K. Chu

Treatment of implant-associated infection is becoming more challenging, especially when bacterial biofilms form on the surface of the implants. Developing multi-mechanism antibacterial methods to combat bacterial biofilm infections by the synergistic effects are superior to those based on single modality due to avoiding the adverse effects arising from the latter. In this work, TiO 2 nanorod arrays in combination with irradiation with 808 near-infrared (NIR) light are proven to eradicate single specie biofilms by combining photothermal therapy, photodynamic therapy, and physical killing of bacteria. The TiO 2 nanorod arrays possess efficient photothermal conversion ability and produce a small amount of reactive oxygen species (ROS). Physiologically, the combined actions of hyperthermia, ROS, and puncturing by nanorods give rise to excellent antibacterial properties on titanium requiring irradiation for only 15 min as demonstrated by our experiments conducted in vitro and in vivo . More importantly, bone biofilm infection is successfully treated efficiently by the synergistic antibacterial effects and at the same time, the TiO 2 nanorod arrays improve the new bone formation around implants. In this protocol, besides the biocompatible TiO 2 nanorod arrays, an extra photosensitizer is not needed and no other ions would be released. Our findings reveal a rapid bacteria-killing method based on the multiple synergetic antibacterial modalities with high biosafety that can be implemented in vivo and obviate the need for a second operation. The concept and antibacterial system described here have large clinical potential in orthopedic and dental applications.

Urease immobilized GO core@shell heparin-mimicking polymer beads with safe and effective urea removal for blood purification

Jue Zhang,Zhenqiang Shi,Chao He,Xin Song,Ye Yang,Shudong Sun,Weifeng Zhao,Changsheng Zhao

Adsorbents are usually used to remove uremic toxins for blood purification. However, the removal of urea is still an intractable problem, since no effective material has been found for urea removal by physical adsorption. Here, urease immobilized graphene oxide core@shell heparin-mimicking polymer (U-GO-HMP) beads were designed, which exhibited good urea removal ability with a removal amount of about 635 mg/g and a removal ratio of about 80% from urea solution. In addition, urea could be removed from collected dialysate and the removal ratio could reach 60% within 480 min. Beyond that, the U-GO-HMP beads also showed good reusability with sustainable relative activity after 5 cycles. Furthermore, the U-GO-HMP beads exhibited good blood compatibility with low hemolysis ratio, suppressed complement activation and contact activation, as well as increased clotting times. It is worthy believing that the U-GO-HMP beads may have great potential in the field of blood purification for urea removal.

Functionalized Nano-MoS2 with Peroxidase Catalytic and Near-Infrared Photothermal Activities for Safe and Synergetic Wound Antibacterial Applications

Wenyan Yin,Jie Yu,Fengting Lv,Liang Yan,Li Rong Zheng,Zhanjun Gu,Yuliang Zhao

We have developed a biocompatible antibacterial system based on polyethylene glycol functionalized molybdenum disulfide nanoflowers (PEG-MoS2 NFs). The PEG-MoS2 NFs have high near-infrared (NIR) absorption and peroxidase-like activity, which can efficiently catalyze decomposition of low concentration of H2O2 to generate hydroxyl radicals (·OH). The conversion of H2O2 into ·OH can avoid the toxicity of high concentration of H2O2 and the ·OH has higher antibacterial activity, making resistant bacteria more vulnerable and wounds more easily cured. The PEG-MoS2 NFs combine the catalysis with NIR photothermal effect, providing a rapid and effective killing outcome in vitro for Gram-negative ampicillin resistant Escherichia coli (AmprE. coli) and Gram-positive endospore-forming Bacillus subtilis (B. subtilis) as compared to catalytic treatment or photothermal therapy (PTT) alone. Wound healing results indicate that the synergy antibacterial system could be conveniently used for wound disinfection in vivo. Interestingly, glutathione (GSH) oxidation can be accelerated due to the 808 nm irradiation induced hyperthermia at the presence of PEG-MoS2 NFs proved by X-ray near-edge absorption spectra and X-ray spectroscopy. The accelerated GSH oxidation can result in bacterial death more easily. A mechanism based on ·OH-enhanced PTT is proposed to explain the antibacterial process.

Highly Stretchable and Conductive Core–Sheath Chemical Vapor Deposition Graphene Fibers and Their Applications in Safe Strain Sensors

Xiaona Wang,Yunfeng Qiu,Wenwu Cao,PingAn Hu

Highly stretchable and conductive core–sheath nanofibers are significant for flexible and wearable microelectronics. Core–sheath fibers were massively fabricated from ultralong chemical vapor deposition (CVD)-grown graphene bundles. They exhibited superior conductivity and excellent mechanical properties that exceeded those of the reduced graphene oxide fibers. The intrinsic dynamic fracture procedure and mechanism of the core–sheath nanofibers were investigated. Furthermore, safe strain sensors based on as-prepared core–sheath CVD graphene fibers have been demonstrated as a proof-of-concept application. The performance of strain sensors has been greatly improved by using CVD graphene fibers.
