Application of umbilical cord mesenchymal stem cell lysate in dry eye syndrome and retinal injury

Hang Zhongci,Li Yingxian,Ren Weijie,Du Hongwu

Purpose Human umbilical cord mesenchymal stem cell lysates (UCML) have crucial bioactivities, including tissue regeneration, wound healing, and immune regulation, showing great potential for treating eye diseases. This study investigates the therapeutic efficacy of UCML loaded in artificial tears (AT@UCML) on dry eye syndrome (DES) and retinal injury. Methods This study first evaluated the safety, antioxidant properties, and tissue-repairing effects of AT@UCML in disease-relevant ocular cells. Then, we treated DES and ischemia/reperfusion (I/R) retinal injury mouse models by AT@UCML. To assess the effects in DES mice, we measured tear secretion, corneal damage, and goblet cell numbers. For I/R mice, we examined retinal ganglion cell numbers and retinal thickness. Results Experiments showed that AT@UCML reduced oxidative stress and protected ocular cells. In DES mice, it increased tear secretion, repaired corneal damage, and restored goblet cell counts. In I/R mice, it reduced retinal ganglion cell loss and restored retinal thickness. Conclusions AT@UCML effectively treated DES and I/R injuries by protecting key ocular cells. This study expands the application of stem cell therapy in ophthalmology and provides a basis for developing related treatments.

In situ cancer stem cell vaccination and immune activation enabled by CSC-specific ligand screening

Yujia Li,Sijia Liu,Zimai Liu,Zixian Wu,Jiao Lu,Yuanyuan Wei,Qianxi Lu,Xiaoman Guo,Xinmeng Zhang,Yaxin Yang,Yilin Wang,Yingying Zhao,Yiyuan Guo,Yongchao Wang,Zhenzhen Chen

Cancer stem cells (CSCs) are pivotal drivers of therapeutic resistance, tumor recurrence, and metastasis due to their self-renewal capacity and localization in poorly vascularized, hypoxic tumor niches. These characteristics limit the efficacy of conventional therapies and underscore the need for CSC-targeted strategies. In this study, we propose a novel in situ cancer vaccine strategy specifically targeting CSCs to overcome these limitations. Using phage display screening, we identified a CD133-targeting peptide and constructed a CD133-functionalized PLGA nanoparticle (CD133-PLGA@DOX) for the selective delivery of doxorubicin to CD133 + CSCs. This nanoplatform achieved precise CSC targeting and induced effective cytotoxicity, particularly against CSCs enriched post-radiotherapy. Importantly, targeted CSC destruction led to the local release of CSC antigens, which served as an endogenous antigen reservoir and triggered a potent in situ vaccine-like immune response. Treatment with CD133-PLGA@DOX significantly promoted dendritic cell maturation, enhanced antigen-specific cytotoxic T cell infiltration, and established long-term immune memory. This strategy—simultaneous CSC eradication and in situ CSC antigen release—demonstrates strong therapeutic efficacy and holds promise for preventing tumor recurrence and overcoming resistance in solid tumors.

Physical and Combustion Properties of Binder-Assisted Hydrochar Pellets from Hydrothermal Carbonization of Tobacco Stem

Liang Miao,Lu Wenjie,Lei Ping,Wang Le,Wang Bing,Li Bin,Shen Yi,Zhang Ke

Abstract Hydrothermal carbonization (HTC) combined with pelletization is an efficient upgrading process to convert biomass into energy-dense pellets. Binders usually serve an important role in the quality of biofuel pellets. Here, hydrochar was produced from tobacco stem through HTC, and was pelletized in the presence of various binders. The effect of four kinds of binders (K 2 CO 3 , CaCO 3 , xanthan gum and carboxymethyl cellulose) on the basic fuel properties, mechanical strength, moisture adsorption and combustion characteristics was investigated. The results showed that K 2 CO 3 assisted pellet had decreased compressive strength, increased hygroscopicity and high combustion residues. The compressive strength of pellets increased with increasing amount CaCO 3 binder probably due to the hardening effect of enhanced adhesion attraction forces between adjacent particles. Meanwhile, the CaCO 3 assisted pellet exhibited the lowest moisture uptake behavior. In addition, the introduction of organic binders led to improved mechanical strength, especially for xanthan binder, which could be ascribed to the formation of solid bridges and enhancement of adhesion forces between particles. TGA results suggested that the binder-assisted pellets exhibited desirable combustion characteristics such as decreased ignition temperature and increased comprehensive combustibility index. Kinetic parameters of the combustion process of binder-assisted pellets were also determined by employing the model free isoconversional Kissinger–Akahira–Sunose method. This study demonstrated that HTC of tobacco stem combined with pelletization in the presence of appropriate binders provide an alternative for solid biofuel pellets production. Graphic abstract Hydrothermal carbonization (HTC) of tobacco stem combined with pelletization in the presence of appropriate binders provide an alternative for solid biofuel pellets production.

Introduction: Facial nerve injury significantly impacts both the physical and psychological] wellbeing of patients. Despite advancements, there are still limitations associated with autografts transplantation. Consequently, there is an urgent need for effective artificial grafts to address these limitations and repair injuries. Recent years have witnessed the recognition of the beneficial effects of chitosan (CS) and graphene in the realm of nerve repair. Dental pulp stem cells (DPSCs) hold great promise due to their high proliferative and multi-directional differentiation capabilities. Methods: In this study, Graphene/CS (G/CST) composite tubes were synthesized and their physical, chemical and biological properties were evaluated, then DPSCs were employed as seed cells and G/CST as a scaffold to investigate their combined effect on promoting facial nerve injury repair. Results and Disscussion: The experimental results indicate that G/CST possesses favorable physical and chemical properties, along with good cyto-compatibility. making it suitable for repairing facial nerve transection injuries. Furthermore, the synergistic application of G/CST and DPSCs significantly enhanced the repair process for a 10 mm facial nerve defect in rabbits, highlighting the efficacy of graphene as a reinforcement material and DPSCs as a functional material in facial nerve injury repair. This approach offers an effective treatment strategy and introduces a novel concept for clinically managing facial nerve injuries.

Jiaying Zhai,Xiaohuan Peng,Yuanyuan Hu,Zhiyuan Li,Wenxiu Li,Chengguang Zhu,Li Zheng,Jin Chen,Yajing Wang

Object The aim of this study was to investigate the effects of aligned poly (lactic-co-glycolic acid)/nano-hydroxyapatite (PLGA/nHA) fiber scaffolds with micro-/nanostructures on the osteogenic differentiation potential of human periodontal ligament stem cells (hPDLSCs). Methods Aligned/random PLGA and PLGA/nHA fiber scaffolds with micro-/nanostructures were created by electrospinning. Comprehensive characterization of the scaffolds with scanning electron microscopy (SEM), X-ray diffraction, Fourier transform infrared spectroscopy, mechanical testing, thermogravimetric analysis and hydrophilicity was performed. In addition, they were observed if they could promote the osteogenic differentiation of hPDLSCs by RT-qPCR, SEM, alkaline phosphatase staining and alizarin red staining. The subcellular distribution of YAP protein between the nucleus and cytoplasm was detected by YAP immunofluorescence staining. Results Aligned PLGA/nHA fiber scaffolds had a better mechanical strength, thermal stability, hydrophilicity than random fiber scaffolds. Furthermore, aligned PLGA/nHA scaffolds also promoted cell adhesion, obvious cytoskeletal elongation and enhanced osteogenic differentiation of hPDLSCs was observed by the upregulation of the key genes (OCN, RUNX2, OPN) expression, increased the activity of alkaline phosphatase, and positive for alizarin red. Meanwhile, the a-PLGA/nHA scaffolds showed the strongest nuclear YAP fluorescence intensity. Conclusion The synergistic effect of fiber arrangement and nHA can be used to increase the osteogenic differentiation of hPDLSCs and the composite scaffold developed may have translational potential for alveolar bone regeneration.

Light-Switched Mesenchymal Stem Cells for In Situ Exosome Amplification in Craniofacial Bone Defect Reconstruction

Tingting Wu,Yajing Liu,Shuman Wang,Xiaoming Bai,Luyun Zhang,Yuwei Liu,Zhiwen Fu,Chen Shi

Mesenchymal stem cell (MSC)-based therapies hold great promise for tissue regeneration, yet precise spatiotemporal regulation of their bioactivity remains challenging. Here, we report a light-switchable MSC system (MSC-UCNPs) enabled by intracellular upconversion nanoparticles (UCNPs), which allowed remote control of exosome biogenesis and regenerative function. Upon 980 nm near-infrared irradiation, intracellular UCNPs emitted localized 365 nm ultraviolet light without compromising MSC viability. The generated UVA stimulus activated the ROS/HEXB/LAMP1 signaling cascade, suppressing lysosome–multivesicular body fusion and thereby markedly enhancing exosome production (increased to 2.7-fold). The MSC-derived exosomes exerted autocrine effects to promote MSC proliferation and osteogenic differentiation, while also facilitating osteoblast maturation via activating the Wnt/β-catenin pathway. To facilitate in vivo application, an injectable hydrogel composed of sodium alginate, calcium alginate, and hyaluronic acid was constructed through electrostatic interactions for the localized delivery of MSC-UCNPs. Positron emission tomography–computed tomography (PET-CT) imaging confirmed the in vivo light-switchable behavior of MSC-UCNPs, allowing on-demand enhancement of in situ exosome release. Benefiting from the synergistic regenerative effects of MSCs and their exosomes, this light-responsive MSC platform achieved robust cranial bone regeneration with a 3.2-fold greater bone volume fraction compared to the control group.

Deep Learning-Assisted Nanomechanical Evaluation of Replicative Senescence in Human Umbilical Cord Mesenchymal Stem Cells

Jinhong Fu,Ruiyu Wang,Qiuyang Deng,Xiyao Yin,Jianjun Dong,Yujuan Chen,Zuobin Wang

The replicative senescence during in vitro expansion severely limits the clinical application of human umbilical cord mesenchymal stem cells (hUC-MSCs). Existing senescence assessment methods still face significant limitations in terms of noninvasive and real-time quantification. In this study, using atomic force microscopy (AFM), we systematically characterized the nanomorphology and mechanical properties of naive and senescent hUC-MSCs. The results revealed that senescent cells exhibited significantly increased height, surface roughness, adhesion, and elastic modulus compared to naive cells, along with enhanced bundling and formation of F-actin stress fibers. These findings show a new “senescence-associated mechanical phenotype” unique to hUC-MSCs. Notably, the hypoxic intervention effectively reversed these senescence-related mechanical changes, demonstrating the high sensitivity of AFM in detecting senescence. To achieve precise quantitative assessment of cell senescence, we also developed a deep learning model based on a variational autoencoder (VAE), which successfully established continuous low-dimensional representations of the age-related mechanical phenotype. This work exhibited excellent predictive performance and generalization ability under different culture conditions, enabling accurate prediction and early identification of hUC-MSCs senescence.

Artificial human stem cell niche created in cartilage-inspired hydrogel for enhanced articular cartilage regeneration

Zheyuan Zhou,Yixuan Hou,Ziyang Yao,Qian Han,Siqi Wang,Hua Mei,Haobin Chen,Bo Tian,Ming Li,Lamei Cheng,Chao Zeng,Zhuxin Dong

Stem cell-based therapies and liposome-integrated hydrogel matrix strategies are promising for articular cartilage regeneration. However, challenges persist due to the loss of stemness during in vitro expansion and the lack of control over stem cell behavior in vivo . Herein, we design a cartilage-inspired hydrogel scaffold similar to the native stem cell niche and expect to enhance chondrogenesis for cartilage repair. Human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) are encapsulated within a composite hydrogel composed of methacrylate hyaluronic acid and collagen networks, mimicking the extracellular matrix of native cartilage. Additionally, embedded liposomes participate to form a lubrication mechanism, the boundary layer in the synovial joints, which lowers the coefficient of friction close to the native level. In vitro , our hydrogel niche system enhances cell-cell and cell-matrix interactions, induces hUC-MSCs mesenchymal condensation, establishes the hypoxic microenvironment formation, and therefore improves the chondrogenic differentiation. Moreover, SOX9, COL2, and HIF-1α expression are all upregulated, whereas MMP13 is downregulated. Meanwhile, in vivo implantation in an osteochondral defect model demonstrates our repair strategy is superior to the conventional hydrogel scaffolds. The advantages include the restoration of PRG4, the suppression of MMP13 and the increasement of ICRS II score to the native levels at 6 weeks post-implantation. These findings are important biophysical cues in regulating stem cell fate in a three-dimensional hydrogel niche and conducive to developing biomimetic scaffolds for cartilage repair.

Sustained TGF-β3 delivery and stem cell loading via injectable microspheres enhance cartilage defect repair

Wei Zhan,Hui Deng,Yifeng Xia,Xiao Liu,Jinping You,Fanrong Ai,Jinjun Huang

Microspheres can efficiently load and sustainably deliver growth factors and stem cells. • Microspheres provide a favorable microenvironment that supports chondrogenic differentiation of stem cells. • Injectable microspheres can be applied to irregular and complex cartilage defects in a minimally invasive manner. • Microspheres loaded with stem cells and growth factors offer a new strategy for cartilage defect repair. Cartilage defects severely compromise patients’ quality of life, as the unique structural characteristics of native cartilage endow it with extremely limited self-repair capacity. Mesenchymal stem cells (MSCs) possess tremendous therapeutic potential for cartilage tissue engineering due to their multilineage differentiation capability; however, technical bottlenecks associated with current cell delivery systems greatly restrict their clinical translation. Herein, we fabricated an injectable, multifunctional hydrogel microsphere system (PGCT-MS). Specifically, arginine-glycine-aspartate (RGD) sequences conjugated on gelatin molecules facilitate MSC adhesion, while chondroitin sulfate—an essential component of the chondrocyte extracellular matrix—constructs a favorable microenvironment for MSC spreading. Moreover, polydopamine modification on the microsphere surface enables efficient immobilization and sustained release of transforming growth factor-β3 (TGF-β3), which effectively induces the chondrogenic differentiation of bone marrow MSCs (BMSCs) at cartilage defect sites after in situ injection. In vitro experimental results demonstrated that PGCT-MS significantly enhances BMSC adhesion and chondrogenic differentiation, as verified by the upregulated expression of chondrogenic marker genes, and exhibits excellent adhesion affinity for native cartilage matrices. In vivo animal studies further validated that BMSC-loaded PGCT-MS markedly accelerates high-quality cartilage regeneration. Collectively, this multifunctional therapeutic delivery system provides a promising and feasible strategy for clinical cartilage defect repair.

Continuous OFT-Based Behavioral Phenotyping Reveals Neural Stem Cell-Associated Shifts in Depression-Like Mice

Han Wang,Zhaoming Liu,Xianjie Li,Lin Fu,Tingxin Lu,Dantong Wang,Guanzhen Lin,Caixia Wu

Background:Depression-like phenotypes in animal models often show graded behavioral variation rather than uniform group-wise changes, yet treatment effects are still commonly evaluated using categorical comparisons. In particular, it remains difficult to determine whether treated animals retain model-like behavioral features or shift toward a more control-like state. Here, we developed a continuous behavioral phenotyping framework based on multidimensional open-field-test (OFT) features to quantify model-like behavioral states and treatment-associated behavioral shifts. Methods:In the present dataset, untreated CUMS- and LPS-induced mice jointly defined a shared model-like behavioral state characterized by reduced locomotor activity and altered spatial exploration. By integrating principal component analysis with k-nearest-neighbor-based behavioral inference, we derived two complementary indices: Δd, representing the relative distance of an animal from the model-like versus control-like behavioral centroids, and p_depression, a kNN-derived score representing similarity to the untreated model-like reference state. These indices were used to position individual animals along a spectrum between control-like and model-like behavioral states. Results:Higher p_depression and lower Δd indicated OFT phenotypes more similar to untreated model mice. Using this framework, NSC-treated animals showed treatment-associated shifts away from model-like behavioral patterns and toward more control-like states, while preserving substantial inter-individual variability. Behavioral scores also showed group-level correspondence with neuroinflammatory markers, suggesting preliminary biological relevance. Because the primary model-specific biological comparisons were based on a limited subgroup size (n= 4 per group), the observed NSC-associated treatment effects should be regarded as exploratory and require confirmation in larger, prospectively powered cohorts. Conclusions:Together, these findings support continuous OFT-based behavioral phenotyping as a promising exploratory approach for evaluating treatment-associated behavioral change and phenotypic heterogeneity in preclinical depression research.

Biomimetic Bamboo-Inspired Bone Repair Scaffold: Reversing Degraded States and Promoting Mesenchymal Stem Cell Differentiation

HaoRan Jiang,Yang Qu,Qi Meng,Chenyujun Hu,Haoran Zhang,Jinqiu Tian,Bohan Xing,Bojiang Wang,Zhihao Lin,Meng Zhang,Peixun Zhang

Cellular senescence of bone marrow mesenchymal stem cells (BMSCs) represents the fundamental pathological barrier to healing in osteoporotic patients. This study reports a multifunctional bone scaffold specifically designed to reverse this senescent phenotype by upregulating Slc25a33 and Crabp1, key regulators of mitochondrial biogenesis and retinoic acid signaling. The scaffold features interconnected spindle-shaped pores (long axis ∼100 μm) inspired by trabecular bone canalicular lacunae, combining polylactic acid nano-oriented fibers and Type I collagen to simulate rod-like and plate-like trabeculae. This biomimetic design enhances mechanical performance (Young's modulus: 51.478 ± 0.993 MPa, porosity: 65.341 ± 0.863%), facilitates cell migration and substance exchange, and continuously provides Type I collagen to the microenvironment. In vitro scaffold extracts reversed senescence in aged BMSCs, enhancing proliferation (wound closure: 42.46 ± 8.47% vs 39.51 ± 4.35%, p < 0.05), migration, and osteogenic differentiation through Slc25a33/Crabp1 upregulation. In vivo 15-month-old osteoporotic rats showed superior bone regeneration at 12 weeks versus allogeneic bone controls, with significantly improved cancellous bone parameters (BV/TV, Tb.N, and Tb.Th). Single-cell sequencing confirmed expansion of a rejuvenated BMSC subpopulation (cluster 2: 27.9% vs 0.0%, p < 0.05) with activated oxidative phosphorylation and osteogenic pathways. This biomimetic scaffold reverses BMSC senescence to restore bone regeneration capacity in osteoporotic defects.

Mesenchymal Stem Cell Protection and Chondrogenic Differentiation in Tracheal Fistula Therapy via Bioactive Platinum Nanozymes

Liyong Shi,Wei Ji,Qionghua Zheng,Xiaoyang Chen,Huanran Shen,Guoying Zhang,Shaobin He,Wei Chen,Yi-Ming Zeng

Stem cells are highly regarded in regenerative medicine for their ability to promote wound healing and tissue regeneration, exhibiting great potential for tracheal fistula (TF) repair. However, excessive reactive oxygen species (ROS) at damaged TF sites compromise stem cell viability, differentiation, and functional capacity. This challenge is particularly pronounced due to the limited intrinsic healing potential of the tracheal cartilage, which further complicates effective TF repair. Herein, we report the use of a sodium hyaluronate-platinum nanoparticle composite (SHA-PtNPs) to promote fistula healing while supporting the protection and chondrogenic differentiation of adipose-derived stem cells (ADSCs). In vitro experiments demonstrated that SHA-PtNPs significantly enhanced ADSCs’ viability, migration, ROS scavenging, and anti-inflammatory activity. Under oxidative stress conditions, SHA-PtNPs promoted chondrogenic differentiation, as evidenced by upregulated expression of SRY-box transcription factor 9 (SOX-9), collagen type II alpha 1 chain (COL2A1), and aggrecan (ACAN). Strikingly, the combination of SHA-PtNPs and ADSCs achieved 100% healing by day 14 and facilitated cartilage regeneration, as confirmed by green fluorescent protein (GFP) labeling, verifying ADSC integration. Additionally, SHA-PtNPs-ADSCs reduced ROS levels and inhibited the NF-κB/IκBα/IL-1β signaling pathway, highlighting their potent antioxidative and anti-inflammatory effects. These findings underscore the potential of bioactive platinum (Pt) nanozymes to enhance mesenchymal stem cell protection and chondrogenic differentiation in TF therapy.
