Teicoplanin-functionalized magnetic beads for detection of Staphylococcus aureus via inhibition of the luminol chemiluminescence by intracellular catalase

Wu Yue,Wang Mengyao,Ouyang Hui,He Yong,Zhao Xiaoyan,Fu Zhifeng

An affinity-based protocol is described for the detection of Staphylococcus aureus ( S. aureus ). It is utilizing teicoplanin-functionalized magnetic beads as carriers. Teicoplanin, which binds to the walls of cells of S. aureus via five hydrogen bonds, acts as the recognition agent. Captured S. aureus is magnetically separated from the sample matrix and then specifically lysed by lysostaphin which cleaves the cross-linking pentaglycine bridges of peptidoglycan in the cell wall. Lastly, S. aureus is quantified via the inhibitory effect of released intracellular catalase on a chemiluminescent (CL) system composed of peroxidase, luminol, H 2 O 2 and p -iodophenol because catalase decomposes H 2 O 2 . S. aureus can be detected with CL response in the 140 to 1.4 × 10 7  CFU·mL −1 concentration range and a detection limit as low as 47 CFU·mL −1 at a signal-to-noise ratio of 3. The method was evaluated by analyzing spiked samples including milk, human urine and saline injection solutions. The reliability was demonstrated by a recovery test and by comparison with a conventional plate counting method. An antibiotic-affinity protocol is developed to detect Staphylococcus aureus ( S. aureus ) by utilizing teicoplanin-functionalized magnetic beads (Teic-MBs) as carriers. S. aureus can be quantified by measuring the inhibition of luminol chemiluminescence (CL) signal by intracellular catalase.

Polyprodrug Antimicrobials: Remarkable Membrane Damage and Concurrent Drug Release to Combat Antibiotic Resistance of Methicillin-Resistant Staphylococcus aureus

Bing Cao,Fengfeng Xiao,Da Xing,Xianglong Hu

The increased threat of antibiotic resistance has created an urgent need for new strategies. Herein, polyprodrug antimicrobials are proposed to mimic antimicrobial peptides appended with a concurrent drug release property, exhibiting broad-spectrum antibacterial activity and especially high potency to inhibit methicillin-resistant Staphylococcus aureus (MRSA) without inducing resistance. Two series of polyprodrug antimicrobials are fabricated by facile polymerization of triclosan prodrug monomer (TMA) and subsequent quaternization of hydrophilic poly( N , N -dimethylaminoethyl methacrylate) (PDMAEMA), affording PDMAEMA- b -PTMA and PQDMA- b -PTMA, respectively. Optimized samples with proper hydrophobic ratio are screened out, which exhibit remarkable bacterial inhibition and low hemolysis toward red blood cells. Furthermore, synergistic antibacterial mechanisms contribute to the bacteria killing, including serious membrane damage, increased out-diffusion of cytosolic milieu across the membrane, and intracellular reductive milieu-mediated triclosan release. No detectable resistance is observed for polyprodrug antimicrobials against MRSA, which is demonstrated to be better than commercial triclosan and vancomycin against in vivo MRSA-infected burn models and a promising approach to the hurdle of antibiotic resistance in biomedicine.

Menthone Exerts its Antimicrobial Activity Against Methicillin Resistant Staphylococcus aureus by Affecting Cell Membrane Properties and Lipid Profile

Wenming Zhao,Chengwei Yang,Ning Zhang,Yuanyuan Peng,Ying Ma,Keru Gu,Xia Liu,Xiaohui Liu,Xijian Liu,Yumin Liu,Songkai Li,Linjing Zhao

Objective The characteristic constituents of essential oils from aromatic plants have been widely applied as antimicrobial agents in the last decades. However, their mechanisms of action remain obscure, especially from the metabolic perspective. The aim of the study was to explore the antimicrobial effect and mechanism of menthone, a main component of peppermint oil, against methicillin resistant Staphylococcus aureus (MRSA).Methods An integrated approach including the microbiology and the high-coverage lipidomics was applied. The changes of membrane properties were studies by the fluorescence and electron microscopical observations. The lipid profile was analyzed by ultra-high performance liquid chromatography coupled with quadruple Exactive mass spectrometry (UHPLC-QE-MS). The lipid-related key targets which were associated with the inhibitory effect of menthone against MRSA, were studied by network analysis and molecular docking. Results Menthone exhibited antibacterial activities against MRSA, with minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of 3,540 and 7,080 μg/mL, respectively. The membrane potential and membrane integrity upon menthone treatment were observed to change strikingly. Further, lipids fingerprinting identified 136 significantly differential lipid species in MRSA cells exposed to menthone at subinhibitory level of 0.1× MIC. These metabolites span 30 important lipid classes belonging to glycerophospholipids, glycolipids, and sphingolipids. Lastly, the correlations of these altered lipids, as well as the potential metabolic pathways and targets associated with menthone treatment were deciphered preliminarily. Conclusion Menthone had potent antibacterial effect on MRSA, and the mechanism of action involved the alteration of membrane structural components and corresponding properties. The interactions of identified key lipid species and their biological functions need to be further determined and verified, for the development of novel antimicrobial strategies against MRSA.

A multicolor time-resolved fluorescence aptasensor for the simultaneous detection of multiplex Staphylococcus aureus enterotoxins in the milk

Yukun Huang,Hui Zhang,Xiujuan Chen,Xiaole Wang,Nuo Duan,Shijia Wu,Baocai Xu,Zhouping Wang

Food safety is one of the most important public health issues worldwide. Foodborne illnesses caused by Staphylococcus aureus enterotoxins (SEs) commonly occur, affecting both developing and developed countries. In this study, multicolor lanthanide-doped time-resolved fluorescence nanoparticles labeled with aptamers were used as bioprobes, and graphene oxide (GO) was employed as a resonance energy acceptor. Based on the “turn down” strategy, the simultaneous detection of multiplex SEs was realized in a homogeneous solution. Under the optimal conditions, the developed method exhibited high sensitivity and selectivity to three serological types of enterotoxins, including type A, B, C1, with limits of detection below 1   ng   mL −1 . The application of this bioassay in milk analysis with no sample dilution was also investigated, and the results of recovery rates covered from 92.76% to 114.58%, revealing that the developed method was accurate. Therefore, this detection aptasnesor can be a good candidate for multiplex analysis and screening with simple and effective operations.

Nitrogen-phosphorous co-doped carbonized chitosan nanoparticles for chemotherapy and ROS-mediated immunotherapy of intracellular Staphylococcus aureus infection

Xiaomei Dai,Xiaojun Liu,Yu Li,Qingqing Xu,Lele Yang,Feng Gao

Staphylococcus aureus ( S. aureus ) residing in host macrophages is hard to clear because intracellular S. aureus has evolved mechanisms to hijack and subvert the immune response to favor intracellular infection. To overcome this challenge, nitrogen-phosphorous co-doped carbonized chitosan nanoparticles (NPCNs), which possess the polymer/carbon hybrid structures, were fabricated to clear intracellular S. aureus infection through chemotherapy and immunotherapy. Multi-heteroatom NPCNs were fabricated through the hydrothermal method , where chitosan and imidazole were used as the C and N sources and phosphoric acid as the P source. NPCNs can not only be used as a fluorescent probe for bacteria imaging but also kill extracellular and intracellular bacteria with low cytotoxicity. NPCNs could generate ROS and polarize macrophages into classically activated (M1) phenotypes to increase antibacterial immunity. Furthermore, NPCNs could accelerate intracellular S. aureus -infected wound healing in vivo. We envision that these carbonized chitosan nanoparticles may provide a new platform for clearing intracellular bacterial infection through chemotherapy and ROS-mediated immunotherapy.

Antimicrobial activity and mechanism of Magnolia officinalis root extract against methicillin-resistant Staphylococcus aureus based on mannose transporter

Jingwen Sun,Yu Xie,Zhiyuan Chen,Yipin Fan,Yunfeng Liu,Qi Gao,Jianqiang Li,Jingwen Bai,Yu Yang

Magnolia officinalis root extract was obtained by high-pressure-assisted extraction, and the yield of honokiol (HNK) was 86.97 ± 1.05 mg/g dry power. The purified extracts and HNK inhibited the growth of methicillin-resistant Staphylococcus aureus (MRSA). Assays for alkaline phosphatases , proteins, nucleic acids leakage, and electrical conductivity, along with scanning and transmission electron microscopy measurements, confirmed the damaging effects of HNK on the cell wall and membrane of MRSA. Intake of sugar, protein production, molecular docking , and dynamic simulations revealed that HNK interfered with the transport of mannose-based carbohydrates by binding to VAL196, ILE291, ALA293, PRO294, and MET214 on the mannose transporter (ManP) in the phosphotransferase system, and thereby inhibited the carbohydrate metabolism of MRSA. These results indicated that HNK prevented MRSA from using carbohydrates by cutting off its nutritional process, and ultimately caused bacterial rupture and lysis. Therefore, ManP may be a potential target for HNK to combat MRSA infections.

A label-free AuNP bioprobe-assisted CRISPR/Cas12a colorimetric platform for high-throughput detection of Staphylococcus aureus ST398

Baoqing Zhou,Qinghua Ye,Moutong Chen,Chufang Wang,Xinran Xiang,Ying Li,Jumei Zhang,Youxiong Zhang,Juan Wang,Shi Wu,Qihui Gu,Yu Ding,Qingping Wu

Rapid and accurate screening of foodborne pathogens in contaminated food is with great significance for food safety. AuNPs-based colorimetric assay is an attractive strategy in the field of food safety inspection, but is limited by tedious probe preparation, low color rendering resolution, and narrow color gamut. Herein, we proposed an e nhanced c olorimetric p latform based on the C RISPR/Cas12a system and label-free D NA-AuNP p robe (CDP-ECP) for high-throughput detection of Staphylococcus aureus ST398. In this strategy, a novel and specific gene targeting S. aureus ST398 was identified and efficient AuNP-bioprobe was successfully prepared. Through a PCR/RAA amplification, the trans -ssDNA cleavage ability induced by target DNA product triggered an aggregation performance for the prepared AuNP-bioprobe pairs, which could realize target colorimetric detection within 50 min. Under optimal conditions, the colorimetric assay could detect as low as 6.4 × 10 2  cfu/mL of S. aureus ST398 in pure culture. Specific gene mining and target-induced trans -cleavage character gave this method a superior specificity and anti-interference. More importantly, this colorimetric method can be extended to a high-throughput platform for rapid detection of spiked and natural sample by using a common microplate reader. Since it is simple to operate, low cost, high sensitivity and specificity, this new platform is fully qualified for clinical diagnosis, environmental monitoring onsite testing requirements.

A Vehicle-Free Antimicrobial Polymer Hybrid Gold Nanoparticle as Synergistically Therapeutic Platforms for Staphylococcus aureus Infected Wound Healing

Xiaojun He,Lixiong Dai,Lisong Ye,Xiaoshuai Sun,Obeng Enoch,Rongdang Hu,Xingjie Zan,Feng Lin,Jianliang Shen

Pathogenic bacteria infection is a serious threat to human public health due to the high morbidity and mortality rates. Nano delivery system for delivering antibiotics provides an alternative option to improve the efficiency compared to conventional therapeutic agents. In addition to the drug loading capacity of nanocarriers, which is typically around 10%, further lowers the drug dose that pathological bacteria are exposed to. Moreover, nanocarriers that are not eliminated from the body may cause side effects. These limitations have motivated the development of self-delivery systems that are formed by the self-assembly of different therapeutic agents. In this study, a vehicle-free antimicrobial polymer polyhexamethylene biguanide (PHMB, with bactericidal and anti-biofilm functions) hybrid gold nanoparticle (Au NPs, with photothermal therapy (PTT)) platform (PHMB@Au NPs) is developed. This platform exhibits an excellent synergistic effect to enhance the photothermal bactericidal effect for Staphylococcus aureus under near-infrared irradiation. Furthermore, the results showed that PHMB@Au NPs inhibit the formation of biofilms, quickly remove bacteria to promote wound healing through PTT in infection model in vivo, and even mediate the transition of macrophages from M1 to M2 type, and accelerate tissue angiogenesis. PHMB@Au NPs will have promising value as highly effective antimicrobial agents for patient management.

Characterization of the Inclusion Complexes of Isothiocyanates with γ-Cyclodextrin for Improvement of Antibacterial Activities against Staphylococcus aureus

Jianan Liu,Hongyan Wu,Xinying Ao,Hongshun Hao,Jingran Bi,Hongman Hou,Gongliang Zhang

The aim of this study was to develop inclusions formed by γ-cyclodextrin (γ-CD) and three isothiocyanates (ITCs), including benzyl isothiocyanate (BITC), phenethyl isothiocyanate (PEITC), and 3-methylthiopropyl isothiocyanate (MTPITC) to improve their controlled release for the inhibition ofStaphylococcus aureus(S. aureus). These inclusion complexes were characterized using X-ray diffraction, Fourier-transform infrared, thermogravimetry, and scanning electron microscopy (SEM), providing appropriate evidence to confirm the formation of inclusion complexes. Preliminary evaluation of the antimicrobial activity of the different inclusion complexes, carried out in vitro by agar diffusion, showed that such activity lasted 5–7 days longer in γ-CD-BITC, in comparison with γ-CD-PEITC and γ-CD-MTPITC. The biofilm formation was less inS. aureustreated with γ-CD-BITC than that of BITC by using crystal violet quantification assay and SEM. The expression of virulence genes, includingsarA,agr,cp5D,cp8F,clf,nuc, andspa, showed sustained downregulation inS. aureustreated with γ-CD-BITC for 24 h by quantitative real-time polymerase chain reaction (qRT-PCR). Moreover, the growth ofS. aureusin cooked chicken breast treated with γ-CD-BITC and BITC was predicted by the Gompertz model. The lag time of γ-CD-BITC was 1.3–2.4 times longer than that of BITC, and correlation coefficient (R2) of the secondary models was 0.94–0.99, respectively. These results suggest that BITC has a more durable antibacterial effect againstS. aureusafter encapsulation by γ-CD.Keywords:cyclodextrin;inclusion;isothiocyanates;Staphylococcus aureus;controlled release;antibacterial activities

Fluorescence detection of Staphylococcus aureus using vancomycin functionalized magnetic beads combined with rolling circle amplification in fruit juice

Yutong Wang,Zhengzheng Wang,Zhongxu Zhan,Ju Liu,Tingting Deng,Hengyi Xu

Staphylococcus aureus is a common foodborne pathogen that can cause a suppurative infection after eating contaminated food. Detection of S. aureus plays an important role in the food industry. In this study, a strategy for the detection of S. aureus using magnetic separation (MS) technology combined with rolling circle amplification (MS-RCA) was proposed. The strategy used antibiotics to capture bacteria and employed RCA products as signal output probes. Vancomycin (Van), as a commonly used antibiotic, can recognize peptidoglycan on the cell wall of Gram-positive bacteria and can effectively identify target bacteria. Therefore, we prepared BSAylated-Van functionalized magnetic beads (Van-MBs) for the pre-enrichment of S. aureus . To ensure the selectivity of this method, we used biotin-pig IgG to bind S. aureus . In addition, to amplify the output signal of the MS-RCA strategy, we introduced streptavidin (SA) and successfully obtained the [email protected] S. aureus @biotin-pig [email protected] @biotin-RCA probe complex and used the biotin-avidin-system (BAS) by combining magnetic separation technology and RCA technology to realize the enrichment and specific detection of S. aureus . Furthermore, by optimizing the experimental conditions such as the magnetic separation time and the amount of Van-MBs, the detection performance of this method was improved. Under the optimal conditions, the detection limit of this method for S. aureus was 3.3 × 10 2  CFU/mL in fruit juice, and it was less affected by other bacteria.

Self-Assemblies Based on Traditional Medicine Berberine and Cinnamic Acid for Adhesion-Induced Inhibition Multidrug-Resistant Staphylococcus aureus

Xuemei Huang,Penglong Wang,Tong Li,Xuehao Tian,Wenbo Guo,Bing Xu,Guangrui Huang,Desheng Cai,Fei Zhou,Hao Zhang,Haimin Lei

S. aureus is resistant to various first-line antibiotics, and seeking multifarious strategies aimed at effective control of antibiotic-resistant behavior is urgently needed. Here, we report a two-component directed self-assembly mode: the phytochemicals berberine and cinnamic acid can directly self-assemble into nanoparticles (NPs) displaying good bacteriostastic activity. Compared with several first-line antibiotics, the obtained nanostructures have a better inhibitory effect on multidrug-resistant S. aureus (MRSA) and stronger ability for biofilm removal. These qualities are attributed to the fact that organic assemblies can first spontaneously adhere to the surface of the bacteria, infiltrate into the cell, and then lead to converging attack against MRSA; thereafter, multipath bactericidal mechanisms of NPs on MRSA are found by both transcriptomic analysis and quantitative Polymerase Chain Reaction analysis. Moreover, when combined with spectral data and single crystal X-ray diffraction, the NPs’ self-assembly mechanism governed by hydrogen bonds and π–π stacking interactions is clearly elucidated. These non-covalent interactions induce the NPs’ formation of butterfly-like one-dimensional self-assembled units and finally layered three-dimensional spatial configuration. In addition, biocompatibility tests show that the NPs are nonhemolytic with little toxicity in vitro and in vivo. This directed self-assembly mode can offer a new perspective toward the design of biocompatible antimicrobial nanomedicines for clinical translation.
