Pickering emulsion stabilized by zein/Adzuki bean seed coat polyphenol nanoparticles to enhance the stability and bioaccessibility of astaxanthin

Sitong Ge,Rui Jia,Qi Li,Wei Liu,Meihong Liu,Dan Cai,Mingzhu Zheng,Huimin Liu,Jingsheng Liu

The objective of this study was to explore the physicochemical stability and in vitro digestibility of astaxanthin (AST)-loaded Pickering emulsions stabilized by zein and Adzuki bean seed coat polyphenol (ABSCP) covalent nanoparticles. Compared to zein-alone stabilized Pickering emulsion (ZE), ABSCP fabrication improved the Pickering emulsion (ZAE) against the ionic strength and high-temperature treatment. Furthermore, the ratio of AST remaining in ZAE after exposure to high levels of UV light irradiation was higher than 90%. Due to the change in electrostatic repulsion during in vitro digestion, droplet size, zeta potential, and microstructure of ZAE changed. More importantly, the bioaccessibility of AST reached up to 58.56% ± 0.89% and 55.66% ± 1.48% for ZE and ZAE, respectively. The results indicate that ZAE may be an effective delivery system for AST or other hydrophobic bioactive compounds. This study may contribute to the protection and delivery of AST and the development of novel functional foods.

Preparation and characterization of zein/gelatin electrospun film loaded with ε-polylysine and gallic acid as tuna packaging system

Qiuying Li,Wenxuan Zhou,Xinrui Yu,Fangchao Cui,Xiqian Tan,Tong Sun,Jianrong Li

BACKGROUND In order to develop effective active packaging films for tuna preservation, zein/gelatin (ZG) composite nanofiber films loaded with ε-polylysine (PL) and gallic acid (GA) were first prepared by electrospun method. The morphology, structure, thermal stability, hydrophobicity, antibacterial and antioxidant properties of the films and application in tuna during 4°C storages were investigated. RESULTS PL reduced the average diameter of ZG fibers, while GA increased it. PL/GA/ZG film possessed well-distributed fiber morphology with an average diameter of 810 ± 150 nm. Fourier transform infrared spectroscopy and X-ray diffraction results showed the physical loading of PL and GA in ZG film with unchanged main chemical bonds and crystal structure. Both the addition of PL and GA reduced hydrophobicity of the ZG film, while the PL/GA/ZG film is still hydrophobic. GA enhanced thermal stability and contributed to antioxidant activity of PL/GA/ZG film. PL and GA synergetic enhanced that antibacterial activity of ZG film against Shewanella putrefaciens . PL combined with GA is more suitable for modifying ZG film than GA alone. Moreover, PL/GA/ZG film effectively inhibited total viable count, total volatile base nitrogen, fat oxidation and texture deterioration of tuna fillets at 4°C storage, and could extend the shelf life by 3 d. CONCLUSIONS PL/GA/ZG nanofiber film have shown promising potential application in the preservation of aquatic products as a new antibacterial and antioxidant food packaging. Electrospun antibacterial and antiviral poly(ε-caprolactone)/zein/Ag bead-on-string membranes and its application in air filtration

Yaowen Liu,Siying Li,Wenting Lan,Md Alomgir Hossen,Wen Qin,Kangju Lee

In this study, poly (ε-caprolactone) (PCL)/zein/Ag nanoparticle (AgNP) fibrous membranes were prepared by ultrasonication-assisted electrospinning. The effects of the AgNP content and ultrasonication time on the membrane morphology, chemical structure, biodegradability, mechanical properties, antibacterial/antiviral activity, and air filtration performance were studied. The best-performing membrane (AgNP content = 1% (w/v), sonication time = 30 min) exhibited a bead-on-string morphology, an air permeability of 93.6 ± 1.5 mm s −1 , and the Escherichia coli and Staphylococcus aureus removal efficiencies of 98.1 ± 1.2% and 97.4 ± 2.5%, respectively. In an experiment with E. coli phage, the above-mentioned membrane exhibited a 97.1 ± 2.1% virus removal efficiency after 60 min. The high performance of this bead-on-string membrane for air filtration was ascribed to the low packing density due to the presence of open interconnected beaded airflow channels and small beads.

One Step Fabrication and Application of Antibacterial Electrospun Zein/Cinnamon Oil Membrane Wound Dressing via In situ Electrospinning Process

Qin Mei,Liu Daqing,Dai Zhang,Meng Xin,Liu Guosai,Liu Hao,Huang Xiaowei,Yan Xu,Chen Shaojuan

A ntibacterial wound dressing can benefit the wound healing by preventing bacterial infection, especially for the electrospun ones due to their porous structures and easily loading antibacterial drugs. However, it is challenging to apply the antibacterial electrospun wound dressing to covering the wound conveniently and safely. Here, we presented one step fabrication and application of antibacterial electrospun zein/cinnamon oil wound dressing via a handheld electrospinning setup. The prepared zein/cinnamon oil wound dressing showed gas permeability of (76.1–5.45) mm/s, hydrophilicity with zero body fluid contact angle, swelling stability after 24 h as well as antibacterial zones over 5 cm against both E. coli and S. aureus bacteria. Moreover, in situ electrospinning process can deposit the electrospun zein/cinnamon oil fibers directly onto the wound, meantime forming a wound dressing. The mice cut-wound model experiment demonstrated that the one step in situ fabrication and application of zein/cinnamon oil wound dressing could nearly heal the wound within 11 d.

In vitro and in vivo assessment of glucose cross-linked gelatin/zein nanofibrous scaffolds for cranial bone defects regeneration

Lingli Deng,Yang Li,Hui Zhang

The purpose of this study was to evaluate the glucose cross-linked gelatin/zein scaffolds for bone regeneration in vitro and in vivo. The nanofibrous scaffolds exhibited fast mineralization in the concentrated simulated body fluid with the deposited octacalcium phosphate and dicalcium phosphate dehydrate. The nanofibrous scaffolds exhibited no cytotoxic effect on MC3T3e1 cells in a CCK-8 test. Additionally, scanning electron microscope and confocal laser scanning microscopy images revealed that all the scaffolds were biocompatible and showed excellent support for MC3T3e1 cells. In the osteogenesis characterizations, Alizarin Red staining experiments indicated the improved calcium deposits on the cross-linked scaffolds, while the alkaline phosphatase activity showed no difference. Furthermore, the in vivo cranial bone regeneration results suggested that the cross-linked gelatin/zein scaffolds presented a strong positive effect on the cranial bone regeneration with the increased new bone volume and connective tissue formation, but the incorporation of zein in the gelatin scaffolds did not favor the bone regeneration. Moreover, the cross-linked gelatin scaffold retarded the bone resorption as indicated by the higher levels of IFN-γ and lower levels of IL-6, which restricted the differentiation of osteoclasts.

Poly(L-lysine) modified zein nanofibrous membranes as efficient scaffold for adhesion, proliferation, and differentiation of neural stem cells

Yingling Miao,Ruirui Yang,David Y. B. Deng,Li-Ming Zhang

Excellent biocompatibility and bioactivity are necessary requirements for a scaffold for nerve repair and regeneration. Natural plant protein zein was chosen as the primary material and poly(L-lysine), which is composed of common amino acids in the human body, was used to modify it. Poly(L-lysine) modified zein (ZPLL) with different PLL contents of 1.46%, 3.57%, and 6.18% was synthesized and nanofibrous membranes were prepared by electrospinning. The hydrophilicity of the membranes improved with an increase of PLL content. The biodegradability of the membranes was proved by in vitro experiments. Compared with pure zein membranes, ZPLL membranes can efficiently improve cell viability, adhesion, proliferation, and differentiation of neural stem cells (NSCs) and the effect of PLL content was further investigated. The results show that when the PLL content was 3.57%, cell adhesion and proliferation proved to be the best and most differentiated toward mature neurons with extensive neurite formation and astrocytes rather than oligodendrocytes. The ZPLL was made into nerve conduits for future study and they may be a promising biomaterial for nerve repair and regeneration.

Fabrication of metronidazole loaded poly (ε-caprolactone)/zein core/shell nanofiber membranes via coaxial electrospinning for guided tissue regeneration

Min He,Huiyi Jiang,Rui Wang,Yi Xie,Changsheng Zhao

To develop a biologically mimetic guided tissue regeneration (GTR) membrane with localized sustained drug release function to prevent infection, coaxial electrospinning technique was conducted to fabricate metronidazole (MNA)-loaded poly (ε-caprolactone) (PCL)/zein core/shell nanofibers . The nanofibers displayed a uniform bead-free round morphology as observed by scanning electron microscopy (SEM), and a core/shell structure as confirmed by transmission electron microscopy (TEM). X-ray diffraction (XRD) and differential scanning calorimetry (DSC) characterizations demonstrated that the MNA was well dispersed in the nanofibers matrix. Due to the encapsulation of the hydrophobic zein , the MNA was released in a controlled, sustained manner over 4   days, and the released MNA showed high antibacterial activity towards anaerobic bacteria. In addition, the encapsulation of natural zein resulted in enhanced cell adhesion and proliferation, and the loading of MNA did not show any cytotoxicity. Thus, these results demonstrated that the MNA-loaded core/shell nanofibers had the potential to be used as GTR membranes with antibacterial function for extensive biomedical applications.

Characterization of sodium alginate film containing zein-Arabic gum nanoparticles encapsulated with oregano essential oil for chilled pork packaging

Yuhang Cao,Lingyu Yin,Fei Li,Yi Deng,Baohua Kong,Qian Liu,Hao Wang,Hui Wang

Chilled pork retains most of its nutrients but is prone to deterioration during the production-to-consumption process. To address this issue this study aimed to develop zein-Arabic gum composite nanoparticles loaded with oregano essential oil (ZAG-OEO) and incorporate them into sodium alginate films to enhance the freshness and shelf life of chilled pork. Sodium alginate, known for its excellent film-forming properties, was selected as the matrix to prepare ZAG-OEO-containing sodium alginate films (SA-ZAG-OEO). The results revealed that the tensile strength and elongation at break of the prepared films were 47.73 ± 2.15 MPa and 6.27 ± 0.21 %, respectively, at a 2.5 % nanoparticle concentration. The water contact angle of the films incorporating nanoparticles reached 81.5 ± 1.95°. The incorporation of nanoparticles enhanced the thermal stability and antibacterial activity of the films. The prepared films were utilized for the storage of chilled pork, and the quality changes were analyzed. The results demonstrate that SA-ZAG-OEO films inhibit microbial growth and lipid oxidation, thereby delaying pork spoilage. This study offers new insights into extending the shelf life of chilled pork and developing advanced meat preservation methods for the future development of the meat industry.

Gastrointestinal pH-sensitive Pickering emulsions stabilized by glycosylated zein conjugates ferulic acid nanoparticles: Improving oral bioaccessibility of Coenzyme Q10

Tongliang Yang,Yaqing Bian,Zihan Qu,Yifu Zhang,Shuhong Li,Guiyun Chen,Ye Chen

Pickering emulsion stabilized by food grade nanoparticles with stimulus response as a targeted delivery system for lipophilic bioactive compounds has attracted people's attention. In this study, ferulic acid was used to modify saccharified zein to prepare pH-sensitive nanoparticles for stabilizing Pickering emulsion. The structure, interface behavior, stability of Pickering emulsion and gastrointestinal digestion characteristics of nanoparticles in vitro were studied. The results showed that covalent embedding of ferulic acid (ZGF-con) effectively improved the surface properties of zein nanoparticles based on glycosylation modification of zein, further regulating their behavior at the oil-water interface. In addition, the particle size of ZGF-con was small (92.93 nm), the wettability was moderate (89.85 °), and it was spherical, with orderly transition of secondary structure, which was conducive to the formation of stable emulsion at the oil-water interface. The stable Pickering emulsion formed by ZGF-con showed ideal emulsification performance, and the electrostatic repulsion between droplets and the formation of a robust spatial network structure promoted the stability of the emulsion. In addition, the encapsulation efficiency of CoQ10 in ZGF-con stabilized Pickering emulsion reached 96.11 %. In vitro simulated digestion, ZGF-con stabilized Pickering emulsion was relatively stable in the gastric acid environment, and slowly released in the small intestine, realizing the small intestine targeted release of CoQ10, which increased its bioaccessibility from 10.57 % to 56.42 %. This study provides an effective strategy for the preparation of pH-sensitive Pickering emulsion to improve the bioaccessibility of hydrophobic active ingredients.

Fabrication, characterization and mechanistic exploration of zein/pectin binary nanocomplex by a one-step and organic solvent free technique

Lingyuan Wang,Diandian Huang,Keting Li,Yan Dai,Angxin Song,Laping He,Xingyi Chen,Lei Wang

Previous studies have shown that the organic solvent-free method has considerable potential for delivery systems in food applications. In this study, Nanocomplex of zein and pectin from apple (AP) or citrus (CP) was systemically fabricated and characterized by adjusting pectin pH (2.0–11.0) and mass ratios of zein:pectin (1:0, 1:0.5, 1:1, 1:2 and 1:3) via one-step pH-shifted process. Results showed that the size of zein/pectin nanocomplex remarkably decreased from >500 nm to ∼150 nm with the increasing pH level of pectin solution and proportion of pectin addition. Spherical shape was observed for nanocomplex formed by AP and CP in transmission electron microscopy images. Compared with zein/AP nanocomplex , zein/CP nanocomplex exhibited the smaller size, smoother surface and lower surface hydrophobicity, prompting us to conclude that CP, as compared to AP, had better performance. To further explore the mechanism, the behavior difference could be interpreted by the altered hydrophobic interaction between zein and pectin as revealed by fluorescence spectroscopy and Fourier transform infrared spectroscopy (FTIR). Findings in this study would facilitate the understanding of interaction between zein and pectin and provide new perspectives of pH-shift induced zein nanocomplex.

Fabricating cellulose nanocrystals from passion fruit peel to enhance the properties of electrospun zein/poly(ethylene oxide) nanofibrous films

Baoying Fan,Lei Zhou,Lujuan Xing,Wangang Zhang

Passion fruit peel is a food by-product containing a large amount of cellulose. This study investigated the effect of extracted cellulose nanocrystals (CNC) from passion fruit peel on the properties of electrospun zein/poly (ethylene oxide) (OZP) nanofibrous films loaded with oregano essential oil (OEO). These nanofibrous films were active packaging for pork preservation, in which OEO was the antimicrobial substance, while zein and PEO were film matrix with electrospinnability and film-forming property. The newly sourced CNC had a rigid rod-like structure with a high aspect ratio (27.9), which indicated that CNC from passion fruit peel could reinforce the better properties of materials than the near-spherical CNC from other sources. CNC had the higher crystallinity (64.87 %) compared with that of passion fruit peel powder (PP, 27.31 %) and cellulose powder from peel (PPC, 48.83 %). After the addition of CNC to OZP films, the average diameter of the electrospun fibers increased, which also led to a decrease in porosity. Furthermore, CNC addition improved the embedding ratio of OEO and retarded its release from the electrospun nanofibrous films. In addition, OZP films incorporating CNC (C-OZP) exhibited a higher tensile strength and water vapor barrier because hydrogen bonding formed by CNC and films matrix provided nanofibrous films with a higher crystallinity. The 3 % C-OZP suppressed the increasing trend of TVB-N and TVC of chilled pork during storage and prolonged the shelf-life of pork from 5 to 7 d. In conclusion, this study demonstrated that CNC from passion fruit peel can enhance the properties of OZP electrospun nanofibrous films, which had the significant potential as an active food packaging material.

Co-delivery of resveratrol and curcumin based on Mesona chinensis polysaccharides/zein nanoparticle for targeted alleviation of ulcerative colitis

Jun Yang,Xianxiang Chen,Jieqiong Lin,Mingyue Shen,Yuanxing Wang,Anwesha Sarkar,Huiliang Wen,Jianhua Xie

Herein, a novel oral nano-delivery system (CR-NPs) was designed for co-delivering curcumin and resveratrol to treat ulcerative colitis (UC) using Mesona chinensis polysaccharides and zein via antisolvent precipitation and continuous water evaporation crosslinking. It showed high encapsulation rates for both bioactives due to hydrogen bonding and hydrophobic forces. CR-NPs have favorable in vitro gastrointestinal stability and low pre-colonic release rates, resulting in increased accumulation in the colon. In vivo therapeutic effects of CR-NPs were confirmed through alleviation of UC symptoms and inflammation of UC mice. CR-NPs maintained the balance of intestinal flora and promoted the production of short-chain fatty acids in UC mice. Furthermore, CR-NPs intervention can alleviate the development of UC by modulating arachidonic acid, linoleic acid, and tryptophan metabolism. This study provides some theoretical basis using a novel dietary protocol option that co-delivering natural bioactives to the colon for alleviation of UC.
