Synthesis and antimicrobial activities of chitosan/polypropylene carbonate-based nanoparticles

Zhilong Quan,Chunyang Luo,Bitong Zhu,Chungui Zhao,Mingyi Yang,Magnar Bjørås,Kaizheng Zhu,Anna-Lena Kjøniksen

Antibiotic resistance is an emerging threat to public health. The development of a new generation of antimicrobial compounds is therefore currently required. Here we report a novel antimicrobial polymer of chitosan/polypropylene carbonate nanoparticles (CS/PPC NPs). These were designed and synthesized from readily available chitosan and a reactive oligomer polypropylene carbonate (PPC)-derived epoxy intermediate. By employing a simple and efficient functionalized strategy, a series of micelle-like chitosan-graft-polypropylene carbonate (CS-g-PPC) polymers and chitosan–polypropylene carbonate (CS–PPC) microgels were prepared by reacting mono-/bis-epoxy capped PPC with chitosan. The chemical structure, particle size, and surface charge of the newly synthesized polymers were characterized by infrared (IR) spectroscopy, nuclear magnetic resonance (NMR) spectroscopy, dynamic light scattering (DLS), and zeta potential measurements. The antimicrobial activities of these nanoparticles were determined in both Gram-positive bacteria (S. aureus) and Gram-negative bacteria (E. coli). Minimum inhibitory concentration (MIC), the nanoparticle concentration needed to completely inhibit the bacterial growth, was found at 128 μg mL−1 to 1024 μg mL−1, strongly depending both on the nature of the epoxy-imine network formed from the functional groups (mono- or bis-capped epoxy groups reacting with amine groups) and the feed ratio of the functional groups (-epoxy/-NH2) between the functionalized PPC and chitosan. No hemolysis was observed at concentrations well in excess of the effective bacteria-inhibiting concentrations. These findings provide a novel strategy to fabricate a new type of nanoantibiotic for antimicrobial applications.

Polysaccharides-modified chitosan as improved and rapid hemostasis foam sponges

Xiaohui Wei,Sheng Ding,Shisheng Liu,Kun Yang,Junjie Cai,Fan Li,Chunlai Wang,Song Lin,Feng Tian

Serial hemostatic sponges consisting of polysaccharides-modified chitosan foam sponges were prepared by Schiff base crosslinking reaction between the deacetylated chitosan and oxidized dialdehyde cellulose. Such composite foam sponges were characterized by scanning electron microscopy and Fourier-transform infrared spectroscopy to confirm their morphology and compositions. Then the coagulation process was evaluated in vitro by thrombus elasticity meters. Furthermore, the hemostasis experiments on mouse tail vein and rabbit femoral artery were also performed in vivo . The results strongly indicated that such synergistic cellulose-modified chitosan foam sponges showed comprehensively excellent water-absorbing quality, improved mechanical performance, low hemolysis rates, benign cytotoxicity, good resilience ability after repeated compression, and superior hemostasis capability both in vitro and in vivo . Furthermore, the hemostatic mechanism is via adhering/activating the red blood cell/platelet to form robust blood clots through the endogenous coagulation pathway, which serves as a good candidate for emergency trauma treatment in daily civilian and military hemostasis.

Preparation and antimicrobial activity of pectin-chitosan embedding nisin microcapsules

Junqing Qian,Yan Chen,Qian Wang,Xiaohua Zhao,Haiyan Yang,Feng Gong,Hui Guo

Nisin is food preservative with good food safety and is widely used around the world. However, nisin degrades during food processing and storage, seriously affecting its antibacterial function. In order to improve the stability of nisin, nisin microcapsules were prepared by complex embedding nisin with pectin and chitosan. By selecting the mass ratio of pectin to nisin and chitosan, the microcapsules of nisin loading 200 mg with 81.15% encapsulation efficiency were prepared in aqueous solution of pH 4.0. The results of zeta potential of pectin, chitosan, nisin and microcapsules, infrared spectra and X-Ray diffraction spectra confirmed the embedding of nisin by pectin-chitosan. The adsorption isotherm of pectin and chitosan showed that chitosan and pectin mainly adsorbed by monolayer to form microcapsules. Compared with the same amount of free nisin, bactericidal efficiency decreased within 24 h, which showed there is a release process in microcapsules. However, after treated with 121 °C for 10 min, the sterilization activity loss of nisin was significantly lower than that of free nisin, which showed the thermal stability of nisin pectin microcapsules was greatly increased. The nisin microcapsules with good thermal stability and sustained-release effect can be better applied in future.

Multifunctional MXene/Chitosan-Coated Cotton Fabric for Intelligent Fire Protection

Binglin Wang,Xuejun Lai,Hongqiang Li,Changcheng Jiang,Jiefeng Gao,Xingrong Zeng

Multifunctional intelligent fireproof cotton fabrics are urgently demanded in the era of the Internet of Things. Herein, a novel high fire safety cotton fabric (denoted as MXene/CCS@CF) with temperature sensing, fire-warning, piezoresistivity, and Joule heating performance was developed by coating MXene nanosheet and carboxymethyl chitosan (CCS) via an eco-friendly layer-by-layer assembly method. Benefiting from the thermoelectric characteristic and high conductivity of MXene nanosheet, MXene/CCS@CF exhibited accurate wide-range temperature sensing performance. When being burned, it could repeatedly trigger the fire-warning system in less than 10 s. More importantly, MXene/CCS@CF showed outstanding flame retardancy because of the synergistic carbonization between MXene and CCS. The limiting oxygen index of MXene/CCS@CF was as high as 45.5%, and the char length was only 33 mm after the vertical burning test. Meanwhile, its peak heat release rate reduced more than 66%. Besides, the obtained fabric could detect a variety of human motions. Moreover, the controllable Joule heating performance enabled the fabric to be used in extreme cold weather. This work provides a facile approach to fabricating a next-generation high fire safety cotton fabric, showing promising applications in firefighting, home automation, and smart transportation.

Biocompatible Gradient Chitosan Fibers with Controllable Swelling and Antibacterial Properties

Liu Shangpeng,Yu Yi,Jiang Shasha,Li Jiwei,Wang Shuang,Chen Shaojuan,Ma Jianwei

This study developed a partially water-soluble carboxymethyl chitosan (PWCS) fibers by ethanol-water exhaustion method. Sodium carboxylate was successfully introduced into chitosan fibers, as evidenced by FTIR, which enhanced natural chitosan water solubility. SEM-EDS revealed that the sodium carboxylate groups presented in a gradient distribution from the exterior to the interior of the PWCS fibers, which leads to a controllable swelling performance. Moreover, the PWCS fibers had excellent antibacterial ability against S. aureus and E. coli bacteria comparing with origin CS fibers. The cytotoxicity assay (CCK-8) and fluorescence staining confirmed that all developed samples showed non-toxicity towards MRC-5 and exhibited a surprising promoting effect on cell proliferation, corroborating their biocompatibility. Besides, due to its excellent controllable water selling ability and protonated amine groups, the PWCS fibers possessed an efficient hemostatic performance. In conclusion, PWCS fibers could become a promising material in hemostatic and wound healing applications.

Stealth PEGylated chitosan polyelectrolyte complex nanoparticles as drug delivery carrier

Xingyue Deng,Jing Zhao,Kaiwen Liu,Chao Wu,Fei Liang

PEGylated stealth nanoparticles have emerged as promising drug delivery carrier for cancer therapy. In this study, natural polycationic chitosan was grafted with poly(ethylene glycol) (PEG) to improve the water-solubility and long-circulation. Then PEGylated chitosan nanoparticles were formed by electrostatic interaction between sulfonic acid group of anionic functional polymer and protic amino group of PEGylated chitosan, using polyelectrolyte complex method. Effects of various factors on particle size and distribution, and the stability, biocompatibility, long-circulation ability were investigated. The results showed that when the concentration of PEGylated chitosan and anionic polymer was 0.20 mg/mL, pH of PEGylated chitosan was 5.0, pH of polymer was 5.5 and molar ratio (S/N) was 0.83, particle size of the prepared nanoparticles was 261.2 ± 5.5 nm with pdI of 0.070. Nanoparticles were relatively stable for more than 4 days under pH < 7.0 and normal saline conditions. The results of cytotoxicity experiments showed that the toxicity of PEGylated chitosan nanoparticles was greatly reduced, which met the basic requirements of biomedical materials. The cellular uptake efficiency of PEGylated chitosan nanoparticles was about 4 times lower than that of conventional chitosan nanoparticles, which indicated long circulation time of PEGylated chitosan nanoparticles in the blood. It was expected that this kind of stealth nanoparticles would have a broad application prospect in the field of drug delivery system.

Molecular level study of cadmium adsorption on dithiocarbamate modified chitosan

Zheng Yin,Dong Qiu,Meiyi Zhang

It has been shown that chemical modification of chitosan with sulfur (S) functional groups could significantly enhance its chelating capability with heavy metals included Cd(II). However, a molecular level understanding has been lacking. Here, we carried out X-ray absorption fine structure (XAFS) and Fourier transformed infrared (FTIR) spectra studies to bridge this knowledge gap. The results indicate that both Cd–O/N and Cd–S bonds exist in the complex of Cd(II) with dithiocarbamate chitosan (DTC-CTS). S functional groups (dithiocarbamate) in DTC-CTS play the major role in complexation with Cd(II) and S content affects the adsorption mechanism. At low S content, Cd(II) is mainly adsorbed on DTC-CTS as an outer-sphere complex with two monodentate amino groups and two water molecules in tetrahedral configuration. At high S content, Cd adsorption dominantly occurs by formation of an inner-sphere complex with two bidentate mononuclear S ligands in tetrahedral configuration. This investigation provides information on the effectiveness and mechanisms of Cd(II) removal that is critical for evaluating modified chitosan applications for stabilization of Cd(II) in surface water, groundwater, soils and sediments.

Modeling and optimization of tea polyphenol-alginate/chitosan magnetic microcapsules

Chao Li,Baoguo Li,Chuanhui Zhu,Xiang Meng

New preparation condition of magnetic microcapsule was designed and optimized by response surface methodology(RSM) in this study. The optimal formula was as follows: the concentration of chitosan(CTS), magnetofluid(MHD), calcium chloride(CaCl 2 ) and sodium alginate solution(ALG) were 10.4(g/L),4.83(g/L), 5.625(g/L), and 28(g/L),respectively. The optimized magnetic microcapsules had good slow release effect, and the release time of magnetic microcapsule was up to 100 h. The chemical structure of magnetic microcapsules was determined by Fourier transform infrared(FT-IR) spectrometer. Meanwhile, the chemical composition of the magnetic microcapsule was determined by X-ray diffraction(XRD) instrument.Thermogravimetric analysis(TGA) was used to detect the thermal properties . The properties of anti-oxidation and drug release of the magnetic microcapsules were further analyzed. Most importantly, magnetic microcapsules had superparamagnetism with extremely low magnetic retention and coercivity , which could be used as a good microcapsule embolization agent for tumor endovascular embolization therapy.

Antibacterial and Hemostatic Thiol-Modified Chitosan-Immobilized AgNPs Composite Sponges

Zhengguo Wu,Wei Zhou,Weijie Deng,Changliang Xu,Yun Cai,Xiaoying Wang

Wound bleeding and infection are two of the major threats to patients’ lives, but developing safe materials with high hemostasis efficiency and antibacterial activity remains a major challenge. Silver nanoparticles (AgNPs) are suitable as antibacterial agents in the hemostatic process, but the application is hampered because of easy accumulation of toxicity. Herein, thiol-modified chitosan (TMC) was prepared by modifying with mercaptosuccinic acid and then was used to immobilize AgNPs to obtain composite sponges (TMC/AgNPs) for stemming the bleeding and preventing infection. TMC/AgNPs sponges had complex interlaced tubular porous structure with high porosity (99.42%), indicating high absorption. TMC had high immobilization efficiency for AgNPs—the release rate of AgNPs was 14.35% after 14 days—but the TMC/AgNPs sponge still had excellent antibacterial activity against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa. In vitro and in vivo experiments confirm that the TMC/AgNPs sponge had fast and efficient hemostatic performance in comparison with the PVF sponge, and its possible mechanism was the synergistic effect of high blood absorption capacity and the interaction between amino, sulfydryl, and blood cells. Furthermore, the TMC/AgNPs sponge can promote wound healing by preventing wound infection, while the PVF sponge cannot. More importantly, the sponges had good safety due to the immobilization of TMC for AgNPs.

Robust and nanostructured chitosan–silica hybrids for bone repair application

Jin-Ning Liang,Le-Ping Yan,Yi-Fan Dong,Xiao Liu,Gang Wu,Na-Ru Zhao

In this study, chitosan–silica hybrids (CSHs) with superior mechanical strength and homogeneous dispersion of nano-sized silica particles were synthesized via a facile sol–gel method aiming for bone regeneration. The effects of varied acidic conditions of sol–gel reaction and inorganic/organic ratios on the performance of the hybrid were investigated. CSHs synthesized under weak acidic conditions (acetic acid, pH 4.0) showed a homogeneous nanostructure and robust strength (maximum compressive strength: 42.6 ± 3.3 MPa and 271 ± 31 MPa in wet and dry forms, respectively). However, those developed under the strong acidic condition (HCl, pH 4.0) and the strong acid condition plus lower pH (HCl, pH 2.8) tended to aggregate and exhibited inferior mechanical properties (compressive strength: 6.3 ± 0.3 MPa in wet form at pH 2.8). Under the latter conditions, the interactions between silica and chitosan were weak. Moreover, the mechanical properties of the CSHs could be tuned in a wide range by conveniently varying the inorganic/organic composition ratio between 50% and 70%. In vitro cytocompatibility study indicated that CSHs were non-cytotoxic. These results suggested that the weak acidic sol–gel process were essential for fabricating chitosan–silica hybrids with high mechanical strength, which had potential to be applied as a bone substitute.

Gentamicin decorated phosphatidylcholine-chitosan nanoparticles against biofilms and intracellular bacteria

Yuanhao Qiu,Dan Xu,Guoqing Sui,Dongdong Wang,Ming Wu,Lipeng Han,Haibo Mu,Jinyou Duan

Biofilms and intracellular bacteria often cause a series of overwhelming public health issues due to their strong drug resistance. Hence, chitosan nanoparticles (CS NPs), phosphatidylcholine and gentamicin were used to synthesize a novel nanodrug delivery system (GPC NPs). Dynamic light scattering (DLS) demonstrated that the surface zeta-potential of GPC NPs was −19.5 mV. The morphology of GPC NPs was observed by scanning electron microscopy (SEM). The gentamicin adsorption and release behaviors of GPC NPs were also investigated. The GPC NPs could effectively damage and remove the biofilm formed by pathogens through permeation of the antibiotic into the biofilm. In addition, the nanoparticles were readily engulfed by macrophages which facilitated the killing of intracellular bacteria and had neglectable cytotoxicity. Our study indicated that GPC NPs could be used as a promising nanoantibacterial agent against biofilms and intracellular bacteria.
