Reduced In vivo burst release of ginkgolide B microcrystals achieved by polymeric H+ depot

Jingxin Gou,Silin Wang,Xu Li,Tian Yin,Haibing He,Yu Zhang,Xing Tang,Wei Xiao,Zhenzhong Wang

Drug microcrystal (MC) is an important category of long acting injections (LAIs), whose release profile is controlled mainly by particle size, crystal form, drug solubility , and other parameters. In this study, a polymeric H + depot composed of alginate was combined with drug MC using ginkgolide B (GB) as model drug to reduce the initial burst release of the drugs with elevated solubility in body fluid. The parameters of GB MC preparation by precipitation, including stabilizer type and amount, phase volume ratio, and stirring rate were investigated. And the particle size/distribution, particle morphology, crystallinity, in vitro release of optimized GB MC were characterized. The in vivo dissolution retardation effect of acidified alginate (SAA) was first confirmed by live imaging using fluorescein as probe. And the pharmacokinetic profiles of different GB MC-polymer blends were investigated by UPLC-MS/MS. The prepared GB MC was spherical-like particles with a mean particle size of 5.863 μm and a span value of 1.634. The crystallinity of GB MC was lowered by the precipitation process compared with that of the crude drug , which resulted in a faster in vitro dissolution with more than 90% of drugs dissolved in 20 min. When administered with SAA, an alginate amount-dependent reduction in C max (1243.5 ± 281.4 ng/mL of GB-SAA, 1:5, 820.9 ± 84.0 ng/mL of GB-SAA, 1:10, 2850.9 ± 1059.3 ng/mL of GB alone and 2476.9 ± 396.4 ng/mL of GB-HPMC E5, 1:10) and prolongation in T max (3.2 ± 1.1 h of GB-SAA, 1:5, 6.4 ± 2.2 h of GB-SAA, 1:10, 1.2 ± 0.45 h of GB alone and 1.4 ± 0.55 h of GB-HPMC E5, 1:10) was noticed, indicating the capability of polymeric H + depot in controlling GB release in vivo . In conclusion, the polymeric H + depot strategy influenced drug dissolution and outward drug diffusion sequentially to retard the initial burst release of drugs with elevated solubility in body fluid.

A hollow in hollow nanoreactor of H-PtCu@SiO2 for the selective transfer hydrogenation

Rongxiu Guo,Xinyue Zhang,Ze-Nan Hu,Hong Li,Jianyi Gao,Jiaping Wang,Qionglin Liang,Xiaodong Li,Dun Niu,Hong-bin Sun

Hollow metal nanoparticles have attracted great interest in heterogeneous catalysis research due to their unique structures as nanoreactors. Here, we report hollow PtCu nanoparticles encapsulated in microporous silica shells (H-PtCu@SiO 2 ), which can serve as the nanoreactor with high catalytic activity and high stability. The H-PtCu@SiO 2 nanoreactor shows the state-of-the-art catalytic performance for the transfer hydrogenation of benzonitrile to dibenzylamine, which achieves up to 100% conversion and 99% selectivity in only 0.5 h. The void-confinement effect of H-PtCu@SiO 2 is crucial for the outstanding catalytic performance by encaging the reactants in the nanoreactor. The computation demonstrates the limiting diffusion effect of the silica shell toward the ammonia borane, thus reduces the concentration of reductant species and prolongs the lifetime of the intermediate imine to improve the selectivity of secondary amine. Moreover, the silica shells can effectively enhance the catalytic stability of the H-PtCu@SiO 2 nanoreactor.

Highly efficient cataluminescence gas sensor based on nanosized h-BN for trace acetylacetone detection

Qianchun Zhang,Yan Wang,Qin Zhou,Yafei Chen,Yuguo Zheng,Shan Tang,Yuxian Zhao,Li Jiang

Nonmetallic nanocatalysts have demonstrated powerful potential in cataluminescence (CTL) sensing fields. In this study, an original method for the fabrication of a CTL gas sensor was developed to detect acetylacetone based on nanosized hexagonal boron nitride (h-BN). The luminescence characteristics and optimum operating temperature, wavelength, and carrier gas flow rate parameters for the h-BN sensor were studied in detail. Compared with other common gases (such as cyclohexanone, acetaldehyde, and acetone), the fabricated sensor demonstrated a high selectivity and sensitivity toward acetylacetone. This was verified by density functional theory calculations, which were consistent with the experimental results. The CTL intensity and concentration of acetylacetone had a linear relationship in the range of 0.050–20 μg ml−1 (r = 0.9989) with a low detection limit of 16 ng ml−1 (S/N = 3). The developed sensor was applied to analyze spiked samples, and satisfactory recoveries for acetylacetone of 82.2%–118% were obtained with excellent reproducibility (2.8%–11%). These results indicated that the system has excellent sensing properties including a high selectivity, sensitivity, and stability, proving its broad application prospects for detecting acetylacetone.

Antiproliferative Activity and Potential Mechanism of Marine-Sourced Streptoglutarimide H against Lung Cancer Cells

Hengju Ge,Di Zhang,Muran Shi,Xiaoyuan Lian,Zhizhen Zhang

In 2019, streptoglutarimide H (SGH) was characterized as a new glutarimide from the secondary metabolites produced by a marine-derived actinomyceteStreptomycessp. ZZ741 and shown to have in vitro antiglioma activity. However, the antiproliferative activity and potential mechanism of SGH against lung cancer cells have not yet been characterized. This study demonstrated that SGH significantly inhibited the proliferation of different lung cancer cells. In terms of mechanism of action, SGH downregulated cell cycle- and nucleotide synthesis-related proteins to block cell cycle at G0/G1 phase, reduced the expression levels of glycolytic metabolic enzymes to inhibit glycolysis, and downregulated the important cancer transcription factor c-Myc and the therapeutic target deubiquitinase USP28. Potent anticancer activity and multiple mechanisms indicated SGH to be a novel antitumor compound against lung cancer cells. Keywords:Streptoglutarimide H;lung cancer cells;antiproliferative activity;cell cycle block;glycolysis;USP28;c-Myc

Ultrathin h-BN/Bi2MoO6 heterojunction with synergetic effect for visible-light photocatalytic tetracycline degradation

Zhao Du,Lang Feng,Zhonglu Guo,Tongqi Yan,Qi Hu,Jing Lin,Yang Huang,Chengchun Tang,Yi Fang

Herein, a series of ultrathin h-BN/Bi 2 MoO 6 heterojunction with excellent photocatalytic activity has been firstly prepared via a feasible solvothermal method . Our results suggest that the optimized photocatalyst possesses a high degradation ratio of tetracycline (TC), oxytetracycline (OTC) and doxycycline (DC) up to 99.19%, 95.28% and 91.04% under visible-light irradiation, respectively. We highlight that the outstanding photocatalytic activity is mainly attributed to the prominent performance of tetracycline adsorption on h-BN and the dominant visible-light absorption by Bi 2 MoO 6 as well as the effective photogenerated carrier separation induced by the synergetic effect between h-BN and Bi 2 MoO 6 . Moreover, DFT calculations reveal that the built-in electric field formed between h-BN and Bi 2 MoO 6 gives rise to the separation of carriers and the polarization of tetracycline molecules. Last but not the least, the specific process and micro-mechanism of the photocatalytic tetracycline degradation under visible-light driven have also been illuminated. The present work pioneers the application of ultrathin h-BN/Bi 2 MoO 6 heterojunction as a novel tetracycline degradation photocatalyst and further guides the design of more visible-light photocatalysts.

Heterogeneous Ni–Ru/H-ZSM-5 one-pot catalytic conversion of lignin into monophenols

Feng Lin,Yulong Ma,Yonggang Sun,Kanghe Zhao,Tingting Gao,Yingbo Zhu

Due to the inertness and structural complexity of lignin, upgrading lignin to biofuels and fine chemicals still faces great challenges. In this paper, a one-pot method was used to catalyze straw lignin with H-ZSM-5 supported by Ni and Ru bimetals, and the lignin conversion rate reached 88.1%. The yield of monophenol was 19.5%, of which the yield of 4-ethylphenol and 4-ethylguaiacol was 4.9% and 3.8%, respectively. The 2D HSQC NMR analysis confirmed that the Ni–Ru/H-ZSM-5 catalyst had an obvious activity of breaking C β -O bonds before and after the reaction. The result showed that after loading metal Ni and Ru, the Lewis acid of the catalytic system was greatly increased, which caused electron transfer between Ni and Ru. 3Ni-1.5Ru/H-ZSM-5 had the strongest activity due to the largest synergy among Ni, Ru and H-ZSM-5. The synergy was the key factor leading to the cleavage of C–C and C–O bonds in straw lignin and the high-performance catalytic degradation of lignin. Besides, the effect of reaction solvent and hydrogen resource on upgrading lignin to fine chemicals over 3Ni-1.5Ru/HZSM-5 catalyst was also intensively investigated. This research developed a method for the direct conversion of lignin into valuable monophenols.

Enhancement in photocatalytic performance of Ag–AgCl decorated with h-WO3 and mechanism insight

Chai Chen,Liu Jianxin,Wang Yawen,Zhang Xiaochao,Duan Donghong,Fan Caimei,Wang Yunfang

A h-WO 3 decorated Ag–AgCl composite was prepared through facile precipitation and photoreduction methods and demonstrated to be a highly efficient and stable photocatalyst for the degradation of Rhodamine B (RhB) under visible light. Ag–AgCl/h-WO 3 exhibited much improved photocatalytic performance was elucidated from the physical–chemical properties of AgCl and h-WO 3 and the surface plasmon resonance effect of the Ag particles. The techniques of X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), X-ray photoelectron spectrometry (XPS) and photocurrent were used to characterize crystalline phases, morphology, composition and separation efficiency between electrons (e − ) and holes (h + ) of the synthesized composite photocatalyst. XPS results confirmed the existence of W 5+ and W 6+ on the surface of h-WO 3 sample, which favored electrons transfer between AgCl and Ag, and generated superoxide radical for the degradation of RhB. The active species trapping experiments results further demonstrated that the formation of superoxide radical during the system. Finally, the underlying photocatalytic mechanism for the removal of RhB by Ag–AgCl/h-WO 3 composite was examined.

Fabrication of h-BN-rGO@PDA nanohybrids for composite coatings with enhanced anticorrosion performance

Haowei Huang,Xiaofeng Huang,Yuhui Xie,Yuqin Tian,Xiang Jiang,Xinya Zhang

Inspired by mussels’ universal adhesive ability to various substrates, polydopamine (PDA) was used to bind graphene oxide (GO) and hexagonal boron nitride (h-BN) together to fabricate h-BN-rGO@PDA nanohybrids. The as-prepared h-BN-rGO@PDA nanohybrids were served as anticorrosive nanofillers for polyvinylbutyral (PVB) coatings on mild steel. The structure of h-BN-rGO@PDA nanohybrids were characterized by SEM, XRD, FT-IR, UV–vis and Raman spectroscopies. Electrochemical corrosion tests were conducted to examine the anticorrosion properties of h-BN-rGO@PDA/PVB composite coatings. The results show that the stacked h-BN nanosheets are exfoliated by GO. And PDA is successfully grafted onto the surface of h-BN-rGO@PDA and simultaneously reduced GO into rGO. The h-BN-rGO@PDA nanohybrids are homogeneously distributed in the PVB matrix. When the mass ratio of GO to h-BN in h-BN-rGO@PDA is 1:1, the as-prepared h-BN-rGO@PDA possesses effective aspect ratio and thus its PVB composite coating obtains the optimal anticorrosion performance with 2 orders of magnitude higher than pure PVB.

Superior adsorption of methyl orange by h-MoS2 microspheres: Isotherm, kinetics, and thermodynamic studies

Yanhong Wu,Minhua Su,Jiawei Chen,Zuopeng Xu,Jiafeng Tang,Xiangyang Chang,Diyun Chen

A batch equilibrium system was used in this study to investigate the adsorption of methyl orange (MO) on hollow molybdenum disulfide (h-MoS 2 ) microspheres . The effects of experimental conditions, such as the initial concentration, agitation time, solution pH and temperature, on the adsorption were examined. The adsorption of MO on h-MoS 2 microspheres was unexpectedly ultrafast. Adsorption equilibrium was rapidly achieved in 10 seconds, and the maximum adsorption capacity for MO was 41.52 mg/g. The Langmuir and Freundlich models were used for determining the adsorption parameters. The results reveal that the Freundlich model fits well with the experimental adsorption data. Pseudo-first-order, pseudo-second-order, and intraparticle diffusion models were applied to determine the kinetics of MO ultrafast adsorption. The obtained results indicate that the experimental kinetics data of h-MoS 2 microspheres are well explained by the second-order model. The obtained values of the thermodynamic parameters (Δ H 0 , Δ S 0 , and Δ G 0 ) indicated that MO adsorption on h-MoS 2 microspheres was an exothermic and spontaneous process. The thermodynamic parameters also confirmed that the MO adsorption is a chemical and physical adsorption process on the surfaces of h-MoS 2 microspheres. The adsorption mechanism was further advanced by FT-IR. The findings of this study open an avenue for applying h-MoS 2 microspheres for organic dyes removal from industrial wastewater streams.

Comparison and investigation of H-bond assisted reusable PU adhesives with high shear strength

Biru Shi,Xiaoling Xu,Jiahui Zhong,Zhenyu Zhang,Haosen Ma,Yu He,Xinxin Tian,Yan Zhou,Xiangdong Liu,Dekun Sheng,Yuming Yang

Hot-melt adhesive (HMA) is commonly used in industrial manufacturing due to its ease of use. However, the direct factors that affect shear strength have been unclear in recent years, making it difficult to prepare hot melt adhesives with high shear strength, which are mostly lower than 6 MPa. In this study, three types of polyurethane HMAs, differing in the position of H-bond motifs, ureidopyrimidinone (UPy), were synthesized and characterized, the most central factor for shear strength, Young's modulus , was firstly found out and proved. Side-chain UPy provides better flexibility and more chain entanglement . The lap shear strength of SPU was about 13.48 MPa, which was the highest shear strength of PU HMA, and remained 73.4 % even after 4 cycles. Compared to the other two samples, the side-chain motifs improved the possibility of forming hydrogen bonds with adherends and increased chain mobility. This led to a more efficient wetting process and strengthened Young's modulus directly, enhancing adhesive property incomparably. This work provides a satisfactory method for achieving high shear strength in HMA.

Detection of H. pylori VacA protein using a graphene-Au nanoparticle electrochemical sensing platform

Hao Deng,Shangyao Mo,Yong Xie,Rong Chen

This study presents a novel electrochemical biosensor for the detection of Helicobacter pylori VacA protein using a graphene-Au nanocomposite platform. The nanocomposite was synthesized via a one-pot reduction method and characterized using SEM, XRD, and FTIR. The biosensor fabrication process was monitored through cyclic voltammetry and electrochemical impedance spectroscopy. Optimization studies revealed ideal conditions of pH 7.4, 30-minute incubation time, and 100 μg/mL antibody concentration. The biosensor exhibited a linear response range of 0.1–100 ng/mL with a detection limit of 0.03 ng/mL. Selectivity tests against common interfering substances showed minimal cross-reactivity (<5 % signal change). The sensor demonstrated good reproducibility (3.8 % RSD) and stability (95 % signal retention after 30 days). In spiked serum samples, recoveries ranged from 96.5 % to 103.2 % with RSDs below 4.5 %. Compared to traditional methods, this biosensor offers improved sensitivity and shorter analysis time for VacA detection, potentially enhancing early diagnosis of H. pylori infections and monitoring treatment efficacy.

H*ads dynamics engineering via bimetallic Pd–Cu@MXene catalyst for enhanced electrocatalytic hydrodechlorination

Lan-Ying Liu,Min-Hua Cui,John Justo Ambuchi,Shi-Ming Niu,Xin-Hui Li,Wo-Long Wang,He Liu,Guo-Shuai Liu,Ai-Jie Wang

Electrocatalytic hydrodechlorination (EHDC) is a promising approach to safely remove halogenated emerging contaminants (HECs) pollutants. However, sluggish production dynamics of adsorbed atomic H (H* ads ) limit the applicability of this green process. In this study, bimetallic Pd–Cu@MXene catalysts were synthesized to achieve highly efficient removal of HECs. The alloy electrode (Pd–Cu@MX/CC) exhibited better EHDC performance in comparison to Pd@MX/CC electrode, resulting in diclofenac degradation efficiency of 93.3 ± 0.1%. The characterization analysis revealed that the Pd 0 /Pd II ratio decreased by forming bimetallic Pd–Cu alloy. Density functional theory calculations further demonstrated the electronic configuration modulation of the Pd–Cu@MXene catalysts, optimizing binging energies for H* and thereby facilitating H* ads production and tuning the reduction capability of H* ads . Noteably, the amounts and reduction potential of H* ads for Pd–Cu@MXene catalysts were 1.5 times higher and 0.37 eV lower than those observed for the mono Pd electrode. Hence, the introduction of Cu into the Pd catalyst optimized the dynamics of H* ads production, thereby conferring significant advantages to EHDC reactions. This augmentation was underscored by the successful application of the alloy catalysts supported by MXene in EHDC experiments involving other HECs, which represented a new paradigm for EHDC for efficient recalcitrant pollutant removal by H* ads .
