In the present study, nanocomplexes composed of perilla seed protein isolate (PSPI) and pectin (PEC) were assembled for the delivery of curcumin (Cur) based on the pH-driven self-assembly method. The encapsulation efficiency of Cur was 87.77%, and the PSPI-Cur-PEC exhibited controlled release of Cur during the in vitro digestion. Moreover, the PSPI-Cur-PEC possessed efficient antioxidant activity and provided greater defense for Cur against thermal treatment and ultraviolet irradiation. Fourier transform infrared spectroscopy , fluorescence spectroscopy and X-ray diffraction results indicated that entrapped Cur was existed in the form of amorphous state in PSPI-PEC, and the process was driven by hydrophobic effects, hydrogen bonding and electrostatic interactions. Scanning electron microscopy images revealed substantial morphological changes of the nanocomplexes throughout the process, while transmission electron microscopy images observed the core-shell structure of PSPI-Cur-PEC nanocomplexes. Furthermore, PSPI-PEC exhibited excellent biocompatibility by the in vitro cytotoxicity assay on Caco-2 cells, and the intermolecular interactions and possible formation mechanism of PSPI-Cur-PEC were further determined by molecular docking . This work elucidated that PSPI-PEC nanocomplexes assembled by a green and energy-efficient pH-driven method was feasible to protect sensitive hydrophobic bioactive components and provided a possible utilization of perilla seed protein in delivery system for nutraceuticals.

Quercetin (Que) is a hydrophobic flavanol that has the potential to prevent colon diseases. This study aimed to design hordein/pectin nanoparticle as a colon-specific delivery system for quercetin. The encapsulation efficiency, physicochemical stability and release properties of the nanoparticles were estimated. The FTIR and secondary structure analysis indicated that hydrogen bonds, hydrophobic interactions and electrostatic attractions were formed in the quercetin-loaded hordein/pectin nanoparticles (Que-hordein/pectin NPs). In comparison to Que-hordein NPs, Que-hordein/pectin NPs exhibited better colloidal stability (physical, UV light, heating and salt). Furthermore, the release properties studies showed that pectin coating restrained the premature release of Que from hordein NPs in gastric fluid and intestinal fluid. In-vitro release, when the Que-hordein/pectin NPs were exposed to simulated colonic fluid (SCF) for 6 h, quercetin was greatly released from the hordein/pectin NPs (15.29 ± 1.17% – 80.60 ± 1.78%). In-vivo release, the concentration of Que (μg/g) in Que-hordein/pectin NPs was 2.18 times higher than Que-hordein NPs in colon tissue after 6 h of oral administration. This study suggests that Que-hordein/pectin NPs have promising applications in the specific delivery and release of quercetin to the colon.

Photoactivated pesticides have many advantages, such as high activity, low toxicity, and no drug resistance. However, poor photostability and a low utilization rate limit their practical application. Herein, the photosensitizer hematoporphyrin (HP) was used as a photoactivated pesticide, covalently linked with pectin (PEC) via ester bonds, to prepare an amphiphilic polymer pro-bactericide, and subsequently self-assembled in aqueous solutions to obtain an esterase-triggered nanobactericide delivery system. The fluorescence quenching effect due to the aggregation of HP in nanoparticles (NPs) enabled the inhibition of photodegradation of HP in this system. Esterase stimulation could trigger HP release and increase its photodynamic activity. Antibacterial assays have shown that the NPs had potent antibacterial capacity, almost completely inactivating bacteria after 60 min of exposure to light. The NPs had good adherence to the leaves. Safety assessment indicated that the NPs have no obvious toxic effects on plants. Antibacterial studies on plants have shown that the NPs have excellent antibacterial effects on infected plants. These results provide a new strategy for obtaining a photoactivated bactericide nanosystem with a high utilization rate and good photostability and targeting ability.

The functional modifications of debranched starch (DBS) has been attracting the interest of researchers. This study marks the first time that DBS was modified by cationization through the use of (3-chloro-2-hydroxypropyl) trimethylammonium chloride with the introduction of cationic functional groups. The physicochemical properties and structural characteristics of cationized debranched starch (CDBS) were systematically assessed. The results demonstrate that the maximum degree of substitution (DS) value obtained was as high as 1.14, and the corresponding CDBS exhibited significantly higher zeta potential values: approximately +35 mV. The minimal inhibitory concentration values of the CDBS of DS 1.14 against Escherichia coli and Staphylococcus aureus were 6 and 8 mg mL–1, respectively. In addition, nanoparticles were successfully prepared with a combination of CDBS and low methoxyl pectin (LMP) and a combination of CDBS and κ-carrageenan (CRG). The maximum encapsulation efficiency of nanoparticles for (−)-epigallocatechingallate can reach 87.8%.

Polysaccharides of vinegar-baked Radix Bupleuri (VBCP) have been reported to exhibit liver-targeting and immunomodulatory activities through oral administration, but the absorption behavior and mechanism of VBCPs have not been extensively studied. In this study, a novel HG type pectin polysaccharide, VBCP1–4, with a high molecular weight of 2.94 × 10 6 Da, was separated from VBCP. VBCP1–4 backbone was contained 1,4-α-D-Gal p A, 1,4-α-D-Gal p A6OMe, 1,3,4-α-D-Gal p A and 1,2,4-α-D-Rhap. The branches were mainly contained 1,5-α-L-Araf, 1,3,5-α-L-Araf, t-α-L-Araf and t-α-D-Gal p , which linked to the 3 position of 1,3,4-α-D-Gal p A and the 4 position of 1,2,4-α-D-Rha p . VBCP1–4 could self-assemble to nanoparticles in water, with CMC values of 106.41 μg/mL, particle sizes of 178.20 ± 2.82 nm and zeta potentials of −23.19 ± 1.44 mV. The pharmacokinetic study of VBCP1–4, which detected by marking with FITC, revealed that it could be partially absorbed into the body through Peyer's patches of the ileum. In vitro absorption study demonstrated that VBCP1–4 was difficult to be absorbed by Caco-2 cell monolayer, but could be absorbed by M cells in a time and concentration dependent manner. The absorption mechanism was elucidated that VBCP1–4 entered M cells through clathrin-mediated endocytosis in the form of nanoparticles. These findings provide valuable insights into the absorption behavior of VBCP and contribute to its further development.

How to solve the contradiction between the efficiency and adsorption rate of porous materials in adsorbing pollutants has always been one of the focus issues. In this study, the small landscape cypress trees structure like biomimetic of a hierarchical and dual morphology 3D porous HA-based aerogel was designed and synthesized to use humic acid (HA), pectin (PE) and chitosan (CTS) as raw materials, which it was formed by the disorderly overlapping of lamella composed of fiber networks in 3D space. Due to its special microstructure, it can be used like separation membrane, which allowing for rapid adsorption of pollutants in the water while the water flow passes through quick. In general, this work provides a new concept for owning fast adsorption rate and efficient adsorption of porous materials of preparation to use green method.

Choline leucine ionic liquid solution was chosen to simultaneously extract flavonoids and pectin from ponkan peels, which were subsequently isolated by the corresponding K3PO4-based aqueous biphasic systems (ABS). Under optimized conditions of 54% solution for 100 min extraction at liquid–solid ratio 17, the predicted yields of flavonoids and pectin achieved 1.33% and 13.42%, respectively, the flavonoids determined directly after extraction and the pectin after ABS purification, with the two corresponding measured yields of 1.26% ± 0.04% and 12.85% ± 0.51%. Under optimal extraction, crude flavonoids contained maximal total flavonoid content of about 20% ± 1% after purification of the ABS involving 25% K3PO4 and dilution precipitation. What was directly isolated from K3PO4-rich phase was presumed to be a modified pectin.

Background In this study, soy protein isolate/sugar beet pectin (SPI/SBP) emulsion gels were prepared through an enzymatic gelation process. The effects of emulsifier (SBP, SPI or SPI/SBP complex) and emulsification process on the microstructure, texture, breakdown properties and aroma release behavior of resulting emulsion gels were investigated. Results Oil emulsification by SBP/SPI complex resulted in a higher amount of emulsifier absorbing on the oil–water interface than by SBP and SPI alone, indicating that a more compact interfacial network was formed. Flocculation of oil droplets was observed and corresponding emulsion gels exhibited lower fracture force and strain when the oil was emulsified by SPI and SBP/SPI complex. Moreover, emulsion gels with small droplets produced a greater quantity of small fragments after mastication. However, microstructure did not have a significant effect on breakdown properties of emulsion gels. Headspace gas chromatography analysis showed that the release rate of ethyl butyrate before and after mastication was significantly lower in emulsion gel with more compact network, but the release of aroma compounds with higher hydrophobicity did not show a significant influence of the microstructure and texture of emulsion gel. Conclusion This finding provides a useful application for designing semi-solid foods with desirable flavor perception. © 2016 Society of Chemical Industry

Accidents and surgical procedures inevitably lead to wounds, presenting clinical challenges such as inflammation and microbial infections that impede the wound-healing process. This study aimed to address these challenges by developing a series of novel wound dressings known as electrospun biomimetic nanofiber membranes. These membranes were prepared using electrostatic spinning technique, incorporating hydroxypropyl-β-cyclodextrin/dihydromyricetin inclusion complexes. The prepared electrospun biomimetic nanofiber membranes exhibited randomly arranged fiber morphology with average fiber diameters ranging from 200 to 400 nm, resembling the collagen fibers in the native skin. These membranes demonstrated excellent biocompatibility, hemocompatibility, surface hydrophilicity, and wettability, while also releasing dihydromyricetin in a sustained manner. In vitro testing revealed that these membranes, loaded with hydroxypropyl-β-cyclodextrin/dihydromyricetin inclusion complexes, displayed higher antioxidant potential and inhibitory effects against Staphylococcus aureus and Escherichia coli . Furthermore, these membranes significantly reduced the M1 phenotypic transition in RAW264.7 cells, even when stimulated by lipopolysaccharides, effectively restoring M2 polarization, thereby shortening the inflammatory period. Additionally, the in vivo wound healing effects of these membranes were validated. In conclusion, this study introduces a promising nanofiber membrane with diverse biological properties that holds promise for addressing various crucial aspects of the wound-healing process.

Pectin is a structurally complex heteropolysaccharide that affects intestinal microorganisms and mucin O -glycans. The present study employed an in vitro model to investigate dynamic changes in microbiota during pectin fermentation. Residual pectin fragments arising from its fermentation were applied to HT29-MTX cells to study the effect of pectin structure on mucin O -glycosylation. Prevotella , Bacteroides , and Parabacteroides were found to preferentially degrade galactose, arabinose, and on the rhamnogalacturonan RG-I side chain region and methyl esterification groups of pectin. Bifidobacterium , Enterococcus , Megamonas , and Dorea metabolized the galacturonic HG region on pectin to produce butyrate. All pectin fragments were found to up-regulate total O -glycans (1.55–2.73 fold) and neutral O -glycans (1.11–1.49 fold) on HT29-MTX mucins. The large HG fragment (81.04 kDa) increased significantly the amount of non-fucosylated glycans (by 2.46-fold); whereas the small HG fragment (16.02 kDa) promoted fucosylated (by 9.25 fold), and especially di-fucosylated O -glycans. Collectively, these results demonstrate that gut microorganisms degrade pectin fragments in the following order of utilization: RG-I, RG-II, and HG. The small fragment of HG improves the expression of fucosylated O -glycans in HT29-MTX cells, mainly owing to an increase in di-fucosylated O -glycans.

In this study, changes in the basic composition and structural characterisation of water-soluble pectin (WSP), homogalacturonan (HG) and rhamnogalacturonan-I (RG-I) from pomegranate peel were investigated after high-pressure-homogenisation (HPH) at 50 MPa and 300 MPa. The interactions between three pectin and anthocyanin (ACN) complex were also studied. The three pectin fractions were mainly composed of galacturonic acid (34.95%–87.69%), all with low degrees of methyl-esterification ≤41.20%. HPH at 300 MPa increased the binding ratios of ACN to three pectin fractions by 34.22%–34.59%. Changes in the structural characterisation results of pectin confirmed that the depolymerisation and breakdown of the side chains of pectin after HPH promoted electrostatic interactions, hydrogen bond and hydrophobic interaction between pectin and ACN. Correspondingly, the thermal and storage stabilities of ACN in the complex was boosted after HPH at 300 MPa. This study provides insights into the interaction between pectin and ACN under HPH.

Traditional drug-delivery methods are limited by low bioavailability and nonspecific drug distribution, resulting in poor therapeutic efficacy and potential risks of toxicity. Mesoporous silica nanoparticles (MSNs) have attracted wide attention as drug-delivery carriers due to their large specific surface area, adjustable pore size, good mechanical strength, good biocompatibility, and rich hydroxyl groups on their surface. In this paper, MSNs were synthesized by a template method, and the morphology and pore structure were regulated. The obtained particles possessed a narrow particle size distribution and good sphericity with a diameter of 878 nm, and their growth process was consistent with the La-Mer growth mechanism. Furthermore, MSNs were modified with amino acids and loaded with rosmarinic acid (RosA), accompanied by the “outer packaging” of pectin to obtain RosA@MSNs-Pec. RosA was added at 200, 150, and 100 mg within 72 h, with a drug loading of 193, 175.5, and 156 mg/g and an encapsulation rate of 19.3, 17.6, and 15.6, respectively. Interestingly, RosA@MSNs-Pec demonstrated the dual pH and pectinase response property, and its release curve conformed to the Higuchi model, indicating that its drug-controlled release was based on Fick’s law.
