Enzymatic depolymerization is a potentially efficient and environmentally friendly solution for poly(butylene adipate-co-terephthalate) (PBAT) waste recycling and utilization. However, the depolymerization capacity of reported depolymerases against diverse, real-world commercial PBAT films remains unclear, hindering accurate evaluation of their utility. In this study, a mutant PBAT depolymerase (Tfcut-B3) with enhanced thermal stability and improved activity was obtained through directed evolution of Tfcut-DM. Tfcut-B3 completely depolymerized PBAT white film into terephthalic acid (TPA) within 48 h, yielding 2.0 and 1.3 times more TPA than Tfcut-DM and Tfcut-DM(Q132Y), respectively. Tfcut-B3 completely degraded 8/10 commercial PBAT-based films into TPA within 48 h, with the remaining two (PB-6 and PM-4) being broken down into small fragments; Tfcut-DM was unable to completely degrade any of the films. Tfcut-B3 overexpression and extracellular secretion in Bacillus subtilis was achieved using chromosomal integration; in flask fermentation, a Tfcut-B3 expression level of 274 mg/L was achieved through promoter and host optimization. Tfcut-B3 fermentation supernatant completely degraded up to 64.9 mg of PBAT white film within a 20 mL system at 72 h. These findings provide a theoretical basis for the large-scale application of PBAT depolymerases.

An assay enables high-throughput screening of phosphorylation activity. • Umbelliferone enables activity readout via phosphorylation-induced fluorescence quenching. • The assay was validated with high accuracy, precision, and reproducibility. • Directed evolution using the HTS platform identified a thermostable Bs PPS variant (G145D). To date, enzymatic systems capable of phosphorylating phenolic compounds are rare, and phenolic phosphate synthetase from Bacillus subtilis ( Bs PPS) is distinguished by its ability to regioselectively phosphorylate a broad range of structurally diverse natural polyphenols, a transformation valuable for improving the aqueous solubility and bioavailability of phytochemicals. However, the lack of an efficient high-throughput screening (HTS) platform has limited its engineering and application. Here, we developed a fluorescence-based HTS assay for rapid estimation of Bs PPS activity using umbelliferone as a fluorogenic probe. Phosphorylation of umbelliferone causes pronounced fluorescence quenching, providing an inverse correlation between fluorescence intensity and catalytic activity. The assay was systematically optimized and validated, demonstrating high accuracy, precision, and reproducibility. Application to an error-prone PCR library identified Bs PPS variants with improved thermostability while maintaining catalytic activity. Overall, this assay fills a critical methodological gap and provides a practical foundation for future exploration of related phenolic compound–phosphorylating enzymes. This study investigates the effects of atmospheric room temperature plasma (ARTP) mutagenesis on the morphological and genetic modifications of the lipase-producing strain Bacillus licheniformis NCU CS-5. While traditional strain improvement methods often suffer from low mutation rates and uneven mutation distribution, ARTP mutagenesis offers a powerful and efficient alternative. The significance of this study lies in its potential to enhance lipase production, a key enzyme in various industrial applications, through the generation of high-performance mutant strains. The gap in existing literature pertains to the limited understanding of ARTP-induced mutations in B. licheniformis and their impact on enzyme production. The novelty of this study is to explore the morphological and genetic changes induced by ARTP and to identify key genetic determinants contributing to these changes. Key findings include significant morphological alterations, higher intracellular Ca2+ levels, and numerous single-nucleotide variants (SNVs) and insertions/deletions (InDels) in the mutant strain. These findings imply that ARTP mutagenesis can effectively enhance lipase production, paving the way for optimized industrial microbial strains.

In this study, we screened a new aerobic bacterium ( Bacillus megaterium Y-4) that can efficiently reduce Pd(II) with different electron donors. The best electron donor was sodium formate and the best reduction of Pd(II) were by log growth phase cells. The high removal capacity of Pd(II) (1658.3 mg/g) was obtained with 30 mg/L dry cell weight and 50 mg/L Pd (II) in the presence of 5 mM sodium formate. The removal amount of Pd(II) increased with initial Pd(II) concentrations ranging from 50 to 200 mg/L with 100 mg/L Pd(II) being completely removed by 148 mg/L dry cell weight in 6 h. The cell wall, periplasmic space and intracellular contents of B . megaterium Y-4 contains different kinds of enzymes for reducing Pd(II). In addition, the activity of extracellular and periplasmic enzymes was more sensitive to temperature than intracellular enzymes . XRD and XPS analysis revealed that the enzyme for reducing Pd(II) in B. megaterium Y-4 can tolerate a broad range of temperatures (20–60 °C) and pH (2.0–7.0) but was sensitive to oxygen. TEM analysis showed that biogenic palladium nanoparticles (Pd-NPs) were mainly distributed evenly in the periplasmic space of the live cells and were released from cells into aqueous solution, which reduced the toxicity of Pd(II), allowing Pd-NP recovery without cell destruction. B. megaterium Y-4 is a potential bacterium for efficient treatment and reclamation of Pd(II) pollution and formation of Pd-NPs.

Pepper southern blight, caused by Agroathelia rolfsii (Sacc.) Redhead & Mullineux (syn. Sclerotium rolfsii Sacc.), is a serious soil-borne fungal disease. The overuse of chemical pesticides to control this disease has led to pathogen resistance and environmental pollution, making biological control methods a more sustainable alternative. In this study, a strain SEC-482 with significant antagonistic effect against A. rolfsii was isolated from the rhizosphere soil of peppers. It was identified as Bacillus velezensis through morphological, physiological, biochemical and molecular analyses. The strain showed a high inhibition rate of 76.44% ± 0.37% against A. rolfsii in vitro and a control effect of 72.73% on pepper southern blight in pot experiments. Furthermore, it was observed to have a favourable impact on the growth of pepper plants. The genome sequencing and analysis revealed many genes related to antibiosis and growth promotion, as well as 14 secondary metabolite synthesis gene clusters. The strain's volatile organic compounds (VOCs), such as 2,6-diethylpyrazine, isobutyric acid and 3,4-diaminophenol, 3,5-dimethyl-1-prop-2-ynyl-pyrazole, were identified as the main antimicrobial substances. This study demonstrates that B. velezensis SEC-482 has potential for the biological control of pepper southern blight, laying a foundation for subsequent field trials to confirm its practical application value. The identification of VOCs and the exploration of the strain’s fermentation conditions provide valuable insights into its potential applications in sustainable agricultural practices.

Bacillus cereus is one of the most common foodborne pathogens and is widely distributed in foods, soil, water, plants and fodder. As an aerobic Gram-positive bacteria, B. cereus also plays an important role in liquor production, generating flavor enhancers and antibacterial substances. Therefore, a rapid and precise analysis strategy for this microbe is highly desired in food safety and sustainable food production. In this study, we propose an effective approach for B. cereus quantification using a dual recognition biosensor. B. cereus was specifically enriched with aptamer-functionalized magnetic beads, and then detected using the broad-spectrum fluorescent material, fragmented crystalline mannose-conjugated lectin-modified nano-MIL-53(Al)-NH 2 ( [email protected] ). The dual recognition method was applied for specific quantification of B. cereus in the range of 20–2 × 10 8 CFU/mL with a limit of detection of 4 CFU/mL within 60 min. Recoveries ranged from 93.8%−100.8% in milk, rice, chicken meat, eggs, and distilled yeast as real samples were also obtained.

Penicillium expansum is the main causal organism of apple rot and causes huge economic losses during apple storage. Patulin (PAT) produced by P. expansum , occurs widely in apples and their products because of its water-solubility and thermal stability . In the present study, we isolated bacterial strains with antagonistic activity against P. expansum to effectively control the growth of P. expansum and production of PAT. We isolated a bacterium named YL-RY0310, which inhibited the growth of P. expansum and production of PAT in apples by 77.4% and 48.4%, respectively. Strain YL-RY0310 was 96% homologous to Bacillus mojavensis using morphological and 16S rRNA gene sequencing analyses. In addition, DNA amplification analysis using Bacillus -specific primers showed that the genes responsible for synthesis of lipopeptides of surfactin, iturins and fengycin are expressed. Oxford cup agar diffusion test showed that its crude lipopeptides could effectively inhibit the growth of P. expansum . Furthermore, strain YL-RY0310 could significantly degrade PAT; the degradation efficiencies of the intracellular and extracellular metabolites of the strain were 62.6% and 56.9%, respectively. Therefore, B. mojavensis YL-RY0310 could be used as a biocontrol agent to inhibit the growth of P. expansum and control PAT contamination in apples. This study provides a theoretical basis for a novel biological control strategy and degradation of mycotoxin.

l -syn-3-[4-(methylsulfonyl)] phenylserine is a key industrial intermediate for the synthesis of florfenicol, thiamphenicol , and other products. l -Threonine aldolase can synthesize l -syn-3-[4-(methylsulfonyl)] phenylserine from glycine and 4-methylsulphonyl benzaldehyde directly. However, the selectivity of free l -threonine aldolase was difficult to meet the requirement of industrialization. Here, we selected the suitable site to insert His tag as immobilized site to improve the selectivity of l -threonine aldolase based on the molecular dynamics simulation and prior protein engineering. The phenanthroline was coupled to the surface of magnetic nanoparticles coated with silicon and thus enabled the selective and oriented immobilization of His-tagged l -threonine aldolase. Due to magnetic nanoparticles coated with silicon as supports, it was easy to separate the residual reactant and impurities from the immobilized enzyme by magnetic attraction. Moreover, the immobilization was combined with the purification of free enzymes through directional immobilization, which avoided heterozyme linking supports and improved the utilization efficiency of supports. Our study demonstrates an effective approach: how to analyze and select enzyme-targeted ligation sites in conjunction with previous modifications of engineered bacteria, and the immobilized enzyme obtained also provided a good candidate for the industrial production of l -syn-3-[4-(methylsulfonyl)] phenylserine.

Di-2-ethylhexyl phthalate (DEHP) widely exists in the environment and has raised an increasing global concern. A bacterial strain capable of degrading DEHP was isolated from the activated sludge at a local wastewater treatment plant and identified as Bacillus sp. MY156. A nearly complete degradation of DEHP at 500 mg L −1 was reached within 5 days when MY156 was grown at pH 8 and 30 °C. The biodegradation of DEHP (50–600 mg L −1 ) followed the first-order kinetics. Mass balance analysis showed the efficient degradation of DEHP with the ratio of biomass to DEHP at 0.72. The dehydrogenase activity was positively related to the biodegradation of DEHP, while the extracellular polymeric substance (EPS) might have played a key role in overcoming the potential inhibition caused by DEHP. The microelements (Fe 3+ , Zn 2+ , and Mn 2+ ) supplements stimulated the DEHP degradation while copper (Cu 2+ ) showed inhibitory. The proposed intermediate metabolites of the DEHP degradation included mono-ethylhexyl phthalate, diethyl phthalate, dimethyl phthalate, phthalic acid, and protocatechuate. Together with the micro-morphological observation, biodegradation experiments in saline and different real water environments suggested the isolate possessing strong halo-tolerance and potential applicable ability to adapt to various environmental conditions. The isolate could be used as a potential and efficient phthalic acid esters (PAEs) degrader for the bioremediation of contaminated sites.

Previous studies have shown that the combination disinfectant, Ortho -phthalaldehyde and benzyldimethyldodecylammonium chloride (ODB), can effectively kill a variety of microorganisms, such as Escherichia coli , Staphylococcus aureus , and Candida albicans . To observe the sporicidal ability and mechanism of ODB for spores, Bacillus subtilis spores were used as the research object in this experiment. TEM images revealed that ODB destroyed the integrity of the coat, cortex, and inner membrane of the spores after 0.5-h treatment, and the nuclear material was also broken and exuded after 4-h treatment. The broken structure led to the release of dipicolinic acid (DPA) in large amount. The results show that B. subtilis spores can be effetely killed by ODB through destroying the structure of the spores.

Contamination of chlortetracycline (CTC) in water is a global problem that needs to be addressed urgently. Biochar immobilization of microorganisms is an effective way to remove CTC contamination from sewage. In this study, the CTC removal effect of free bacteria LZ01 and erding medicine residues biochar immobilized LZ01 was compared by shake flask batch experiments. The results showed that immobilized LZ01 significantly improved the CTC removal rate by 15.27% compared to free LZ01. The immobilization of LZ01 also significantly improved the environmental tolerance and the reusability and storage stability of strain LZ01. The optimal conditions for CTC removal (83.83%) by immobilized LZ01 were determined by response surface methodology: temperature of 33.98 °C, pH of 7.84, initial CTC concentration of 73.75 mg/L, and inoculation dose of 0.51% (w/v). The experimental results of treating CTC from simulated sewage showed that the immobilized LZ01 could remove more than 64% of CTC from the sewage. The ion of Cu 2+ accelerated the removal of CTC by immobilized LZ01, and the removal rates could reach 81.07%, 85.07%, and 87.50% in three sewages, i.e., river sewage, pond water, and farming wastewater, respectively. This study provides a feasible, effective, and rapid biological removal technique for the treatment of CTC pollution in sewage.

Mango juice (MJ) has high nutritional value and decent flavor. However, its aroma could be easily changed by heat treatment during processing, which affects its acceptability and quality. In this study, Bacillus subtilis -HNU-B3 that could transfer carotenoid, was obtained from orchard soil to improve the aroma of fermented mango juice (FMJ). Compared with MJ, the organic acid content of FMJ by B. subtilis -HNU-B3 was significantly changed, and the contents of succinic acid and malic acid were increased to 3.68 mg/mL and 2.04 mg/mL, respectively. Moreover, volatile compounds identified by headspace solid-phase micro-extraction gas-phase mass spectrometry in FMJ were norisoprenoid and acetoin, accounting for 51.52 % of the total volatile substances. In addition, B. subtilis -HNU-B3 fermentation significantly increased the antioxidant capacity and total phenol contents of MJ, and the activity of carotenoid cleavage dioxygenase was 188 U/L. In conclusion, the aroma of FMJ could be improved by B. subtilis -HNU-B3 through increasing the content of norisoprenoid and acetoin. Our results provide theatrical support for Bacillus subtilis to improve the aroma quality of FMJ.
