In this study, we designed a highly integrated microfluidic chip for nucleic acid extraction, amplification, and detection. Magnetic beads, which are used to capture nucleic acids on the chip, are trapped in the microwell arrays in a one-well-one-bead manner after local surface modification of the inner faces of the microwells. On-chip liquid introduction, delivery, and mixing are all carried out manually with one syringe and no other equipment. A hand-held device with precise temperature control and high-quality imaging is developed, which is only 2.3 cubic decimeters in volume and 1.2 kg in weight. Via the use of the Internet for wireless communication, the experiment and data analysis after inserting the chip into the device can be conducted by a smartphone anywhere there is an Internet connection. We carried out reverse transcription loop-mediated isothermal amplification (RT-LAMP) on the chip with the hand-held device. SARS-CoV-2 pseudoviruses are extracted, reverse transcribed, amplified, and detected on the chip with the hand-held device with satisfactory results. Thus, a highly integrated, easy-to-operate, and rapid nucleic acid detection microfluidic chip with a hand-held smartphone-controlled device is proposed, and this new platform for nucleic acid detection shows great potential for mobile point-of-care testing (POCT).

Early diagnosis of Alzheimer’s disease (AD) is critical for preventing disease progression, however, the diagnosis of AD remains challenging for most patients due to limitations of current sensing technologies. A common pathological feature found in AD-affected brains is the accumulation of Amyloid-β (Aβ) polypeptides, which lead to neurofibrillary tangles and neuroinflammatory plaques. Here, we developed a portable ultrasensitive FET biosensor chip based on a self-assembled nanoporous membrane for ultrasensitive detection of Aβ protein in complex environments. The microscale semiconductor channel was covered with a self-assembled organic nanoporous membrane modified by antibody molecules to pick up and amplify the Aβ protein signal. The nanoporous structure helps protect the sensitive channel from non-target proteins and improves its stability since no chemical functionalization process involved, largely reduces background noise of the sensing platform. When a bio-gated target is captured, the doping state of the polymer bulk could be tuned and amplified the strength of the weak signal, achieving ultrasensitive detecting performance (enabling the device to detect target protein less than 1 fg/ml in 1 µl sample). Moreover, the device simplifies the circuit connection by integrating all the connections on a 2 cm × 2 cm chip, avoiding expensive and complex manufacturing processes, and makes it usable for portable prognosis. We believe that this ultrasensitive, portable, low-cost Aβ sensor chip shows the great potential in the early diagnosis of AD and large-scale population screening applications.

Noninvasive glucose detection for diabetes monitoring, in comparison with the conventional finger-prick test that causes inevitable pain, is on demand. Salivary glucose level that possesses a well-established correlation with blood glucose level can be a promising alternative for noninvasive detection, however, the sensitivity and user-friendliness remained to be improved. Herein, an “all-in-one” paper chip was fabricated by one-pot method for sensitive and user-friendly detection of salivary glucose level. Glucose oxidase (GOx) and the chromogenic reagent, luminol, were both in situ encapsulated in a metal–organic framework, ZIF-67, to form the GOx& [email protected] @Paper (G& [email protected] @Paper). Owing to the specificity of GOx and the peroxidase activity of ZIF-67, the paper chips are highly sensitive and specific for glucose with a linear range from 0.2 to 2 mM, and a limit of detection of 0.12 mM (S/N = 3). Moreover, ascribing to the protection and “all-in-one” structure by ZIF-67, the chip is highly stable that maintains the detection performance after storage at room temperature in air for 4 weeks, and this strategy also facilitates the one-pot detection without multiple steps of sampling and incubation. With the assistance of a smartphone application, the glucose level in saliva can be accurately detected with “sample-in-result-out” manner, and the results were highly consistent with those measured by commercial kit. This paper chip is a promising device for daily glucose monitoring, and the highly-integrated strategy provides an innovative resolution for the construction of colorimetric paper chips.

Rapid and sensitive pathogen detection methods are critical for disease diagnosis and treatment. RPA-CRISPR/Cas12 systems have displayed remarkable potential in pathogen detection. A self-priming digital PCR chip is a powerful and attractive tool for nucleic detection. However, the application of the RPA-CRISPR/Cas12 system to the self-priming chip still has great challenges due to the problems of protein adsorption and two-step detection mode of RPA-CRISPR/Cas12. In this study, an adsorption-free self-priming digital chip was developed and a direct digital dual-crRNAs (3D) assay was established based on the chip for ultrasensitive detection of pathogens. This 3D assay combined the advantages of rapid amplification of RPA, specific cleavage of Cas12a, accurate quantification of digital PCR, and point-of-care testing (POCT) of microfluidics, enabling accurate and reliable digital absolute quantification of Salmonella in POCT. Our method can provide a good linear relationship of Salmonella detection in the range from 2.58 × 101 to 2.58 × 104 cells/mL with a limit of detection ∼0.2 cells/mL within 30 min in a digital chip by targeting the invA gene of Salmonella. Moreover, the assay could directly detect Salmonella in milk without nucleic acid extraction. Therefore, the 3D assay has the significant potential to provide accurate and rapid pathogen detection in POCT. This study provides a powerful nucleic detection platform and facilitates the application of CRISPR/Cas-assisted detection and microfluidic chips.

Background Rapid diagnosis of harmful microorganisms demonstrated its great importance for social health. Continuous flow PCR (CF-PCR) can realize rapid amplification of target genes by placing the microfluidic chip on heaters with different temperature. However, bubbles and evaporation always arise from heating, which makes the amplification not stable. Water-in-oil droplets running in CF-PCR microfluidic chip with uniform height takes long time because of the high resistance induced by long meandering microchannel . To overcome those drawbacks, we proposed a double-layer droplet CF-PCR microfluidic chip to reduce the fluidic resistance, and meanwhile nanoliter droplets were generated to minimize the bubbles and evaporation. Results Experiments showed that (1) fluidic resistance could be reduced with the increase of the height of the serpentine microchannel if the height of the T-junction part was certain. (2) Running speed, the size and the number of generated droplets were positively correlated with the cross-sectional area of the T-junction and water pressure. (3) Droplet fusion happened at higher water pressure if other experimental conditions were the same. (4) 0.032 nL droplet was created if the cross-sectional area of T-junction and water pressure were 1600 μm 2 (40 × 40 μm) and 7 kPa, respectively. Finally, we successfully amplified the target genes of Porphyromonas gingivalis within 11′16″ and observed the fluorescence from droplets. Significance and novelty Such a microfluidic chip can effectively reduce the high resistance induced by long meandering microchannel, and greatly save time required for droplets CF-PCR. It offers a new way for the rapid detection of bacterial.

Abnormally expressed miRNAs have been employed as crucial indicators for the diagnosis and prognosis of disease. It is essential to develop quick, affordable, accurate, and multiplexed methods for miRNA detection. In this study, polyethylene glycol (PEG) hydrogel microparticles were used as solid-phase carriers for the hybridization chain reaction (HCR) in combination with cation-exchange fluorescence amplification, thus enabling the development of a new method for miRNA determination. The assay was sensitive with the detection limit for miRNA let7a equal to 46 pM. The assay was specific, the base-mismatched sequences of miRNA let7a were distinguished, and the serum matrix did not cause any interference. Moreover, by integration with a microfluidic chip, the throughput was significantly enhanced and the process was noticeably simplified. With further development, the developed method may be suitable for practical clinical analysis.

Flocculation is a key purification step in cell-based processes for the food and pharmaceutical industry where the removal of cells and cellular debris is aided by adding flocculating agents. However, finding the best suited flocculating agent and optimal conditions to achieve rapid and effective flocculation is a non-trivial task. In conventional analytical systems, turbulent mixing creates a dynamic equilibrium between floc growth and breakage, constraining the determination of floc formation rates. Furthermore, these systems typically rely on end-point measurements only. We have successfully developed for the first time a microfluidic system for the study of flocculation under well controlled conditions. In our microfluidic device (μFLOC), floc sizes and growth rates were monitored in real time using high-speed imaging and computational image analysis. The on-line and in situ detection allowed quantification of floc sizes and their growth kinetics. This eliminated the issues of sample handling, sample dispersion, and end-point measurements. We demonstrated the power of this approach by quantifying the growth rates of floc formation under forty different growth conditions by varying industrially relevant flocculating agents (pDADMAC, PEI, PEG), their concentration and dosage. Growth rates between 12.2 μm s−1 for a strongly cationic flocculant (pDADMAC) and 0.6 μm s−1 for a non-ionic flocculant (PEG) were observed, demonstrating the potential to rank flocculating conditions in a quantitative way. We have therefore created a screening tool to efficiently compare flocculating agents and rapidly find the best flocculating condition, which will significantly accelerate early bioprocess development.

A surface-enhanced Raman scattering (SERS) platform for the selective trace analysis of Hg2+ ions was reported, based on poly-thymine (T) aptamer/2-naphthalenethiol (2-NT)-modified gold nanoparticles (AuNPs), which was an oligonucleotide-functionalized nanosensor and SERS chip. 2-NT was used as a Raman reporter, and T aptamer could form a T-Hg2+-T structure with Hg2+ ions making an SERS nanosensor absorbed to the SERS chip. The optimum concentrations of DNA and 2-NT were obtained. An average of 960 DNA molecules attached to each AuNP were measured. The limit of detection (LOD) was 1.0 ppt (1.0 × 10−12 g/mL), which is far below the limit of 10.0 ppb for drinking water, stipulated by the World Health Organization. The sensor has the advantages of low detection cost, a simple sample pretreatment, a green solution and reducing false positives. Furthermore, the nanosensor was used for the determination of trace Hg2+ in the water of a lake; a reliable result was obtained accurately.

An electrochemical microfluidic chip is described for the determination of the insecticide carbofuran. It is making use of a molecularly imprinted film (MIP) and a DNA aptamer as dual recognition units. The analyte (carbofuran) is transported to the MIP and captured at the identification site in the channel. Then, carbofuran is eluted with carbinol-acetic acid and transported to the DNA aptamer on the testing position of the chip. It is captured again, this time by the aptamer, and detected by differential pulse voltammetry (DPV). The dual recognition (by aptamer and MIP) results in outstanding selectivity. Additionally, graphene oxide-supported gold nanoparticles (GO-AuNPs) were used to improve the sensitivity of electrochemical detector. DPV response is linear in the 0.2 to 50 nM carbofuran concentration range at a potential of −1.2 V, with a 67 pM detection limit. The method has attractive features such as its potential for high throughput, high degree of automation, and high integration. Conceivably, the method may be extended to other analytes for which appropriate MIPs and aptamers are available.

A facile and controllable method for one-step synthesis of small-sized monodisperse fluorescent–magnetic bifunctional microparticles based on droplet splitting and photopolymerization was established. Using this method, magnetic nanoparticles and acryloyl rhodamine B (acryloyl-RhB) molecules were incorporated into the dispersed phase, which allowed the direct generation of functional microparticles. Functional microparticles with diameters in the range of 2.8 to 19.2 μm were obtained by controlling the fluidic characteristics. The fluorescence intensity of the microparticles could be regulated by adjusting the concentration of the acryloyl-RhB molecule in the dispersed phase. Furthermore, the fluorescent–magnetic bifunctional microparticles were successfully used to detect proteins specifically by ratiometric fluorescence analysis, showing their potential in biological sample detection.

Early diagnosis of chronic heart failure (CHF) relies on the ultra-sensitive detection assay of its main biomarker, Brain natriuretic peptide (BNP). To meet such demand, we designed a near-infrared immunoassay biosensor, utilizing a fiber-embedded microfluidic chip for surface-enhanced Raman scattering (SERS) detection. A SERS probe, consisting of metal–organic framework and Au-HEPES coupling nanoparticles (AuHPs), was used to enhance the Raman signal of the toluidine blue (TB) dye. The probe anchored with TB is used as a SERS tag, which is linked to BNP through sandwich method. To enhance the Raman signal further, we embedded a SERS substrate at the bottom of the microfluidic channel, which serves an enhancer when the SERS tagged BNP passes through the microfluidic chip. The amplified Raman signal was collected by an optical fiber probe of a simple and portable Raman detection system with an all-fiber connected configuration. Such design resulted in the lowest detection limit of pg/mL, indicating that the proposed platform can be used for rapid and highly sensitive detection of BNP. In addition, our platform also has the advantages of low sample consumption and high throughput rate, providing an effective scheme for the early diagnosis of CHF.

Thermal interface materials that possess high thermal conductivity and can reach low thermal impedance during service, are highly desirable for chip heat dissipation systems, especially when cooling high-power chips. Herein, a novel thermal pad was developed by combining low melting point alloy having metal-level thermal conductivity and liquid-like rheological property with expanded graphite possessing large pore volume and high thermal conductivity. Specifically, the alloy 62.5Ga21.5In16Sn was modified by the trace-oxidation method to obtain oxidized alloy, followed by adsorbing into expanded graphite and then molding and pressing to fabricate the thermal pad. It is found that the oxidation treatment resulted in the generation of refractory metal oxides Ga 2 O 3 and SnO 2 within the alloy, thereby preventing the leakage of the alloy from the composite thermal pad. Impressively, the optimal low melting point alloy/expanded graphite composite thermal pad, prepared at the oxidation duration of 24 min, expanded graphite mass fraction of 4 % and compact density of 3.125 g/cm 3 , reached a thermal conductivity of as high as 26.94 W/m·K and a thermal impedance of as low as 0.42 K·cm 2 /W, along with exhibiting a good shape-stability. More significantly, when employed into a simulative desktop CPU cooling system, this thermal pad exhibited 36.78 % higher cooling efficiency under the forced air convection, extended ten times in the time for the chip to reach the upper limit temperature (100 ℃) under the natural air cooling, and benefited to the chip to fight with the pulse-type instantaneous thermal shock, as compared to the commercial thermal pad. These excellent characteristics and good heat dissipation performance make the low melting point alloy/expanded graphite composite thermal pad show great potential for use in chip thermal management systems.
