Quercetin-crosslinked chitosan nanoparticles: a potential treatment for allergic rhinitis

Mu Dehong,Zhou Li,Shi Lingyu,Liu Ting,Guo Ying,Chen Hao,Luo Hongping,Ma Junhao,Zhang Hui,Xiong Peizheng,Tian Li

Allergic rhinitis (AR) remains a major health problem worldwide. Compared with traditional oral drugs, nasal administration avoids first-pass metabolism and achieve faster and more effective efficacy. In this study, we used the ion crosslinking method to prepare quercetin–chitosan nasal adaptive nanomedicine (QCS) delivery system and evaluated in the treatment of allergic rhinitis mice models. The obtained positively charged nanoparticles with a particle size of 229.2 ± 0.2 nm have excellent characteristics in encapsulation efficiency (79.604%), drug loading rate (14.068%), drug release (673.068 μg) and stability(> 7 days). Excitingly, QCS treatment significantly reduced the number of sneezing and nasal rubbing events in AR mice, while reducing the levels of inflammatory factors such as immunoglobulin E (IgE), interleukin (IL)-17, tumor necrosis factor (TNF)-α, and (IL)-6 to alleviate AR symptoms. Hematoxylin–eosin (HE) staining also showed the damaged nasal mucosa was improved. These experimental results suggest that QCS can effectively suppress allergic inflammation in a mouse model and hold promise as a therapeutic option for allergic rhinitis.

Rhein-loaded chitosan nanoparticles for treatment of MRSA-infected wound

Shuang Cai,Linlin Wang,Xilong Cui,Xinshu Zou,Sidi Zheng,Fanhui Liu,Chun Shi,Yanhua Li,Zhiyun Zhang

The multi-drug resistance of methicillin-resistant Staphylococcus aureus (MRSA) and complex wound microenvironment challenge the repair of MRSA infected wound. Herein, in this study, α-tocopherol modified glycol chitosan (TG) nanoparticles encapsulated with phytochemical rhein (Rhein@TG NPs) were prepared for comprehensive anti-infection and promotion of MRSA infected wound healing. Rhein@TG NPs could not only specifically release rhein in the infection site in response to low pH and lipase of infectious microenvironment, but also up-regulated M1 macrophage polarization in the infection stage, thus achieving synergistically bacterial elimination with low possibility of developing resistance. Additionally, the NPs reduced the levels of pro-inflammatory factors in the post-infection stage, scavenged the ROS, promoted cell migration and angiogenesis, which significantly improved the microenvironment of infected wound healing. Therefore, this antibiotic-free NPs enabling anti-infection and promotion of wound healing provides a new and long-term strategy for the treatment of MRSA infected wound.

Protonated-chitosan sponge with procoagulation activity for hemostasis in coagulopathy

Zhenhua Huang,Dong Zhang,Laiqiang Tong,Fan Gao,Shaozan Zhang,Xinqing Wang,Yina Xie,Fangping Chen,Changsheng Liu

Hemostatic materials are essential for managing acute bleeding in medical settings. Chitosan (CS) shows promise in hemostasis but its underlying mechanism remains incompletely understood. We unexpectedly discovered that certain protonated-chitosan (PCS) rapidly assembled plasma proteins to form protein membrane (PM) upon contact with platelet-poor plasma (PPP). We hypothesized that the novel observation was intricately related to the procoagulant effect of chitosan. Herein, the study aimed to elucidate the conditions necessary and mechanism for PM formation, identify the proteins within the PM and PCS's procoagulant action at the molecule levels. We confirmed that the amount of –NH 3 + groups (>4.9 mmol/g) on PCS molecules played a crucial role in promoting coagulation. The –NH 3 + group interacted with blood's multiple active components to exert hemostatic effects: assembling plasma proteins including coagulation factors such as FII, FV, FX, activating blood cells and promoting the secretion of coagulation-related substances (FV, ADP, etc) by platelets. Notably, the hemostatic mechanism can be extended to protonated-chitosan derivatives like quaternized, alkylated, and catechol-chitosan. In the blood clotting index (BCI) experiment, compared to other groups, PCS95 achieved the lowest BCI value (∼6 %) within 30 s. Protonated-chitosan exhibited excellent biocompatibility and antibacterial properties, with PCS95 demonstrating inhibition effectiveness of over 95 % against Escherichia coli ( E.coil ) and Staphylococcus aureus ( S. aureus ). Moreover, PCS performed enhanced hemostatic effectiveness over chitosan-based commercially agents (Celox™ and ChitoGauze®XR) in diverse bleeding models. In particular, PCS95 reduced bleeding time by 70 % in rabbit models of coagulopathy. Overall, this study investigated the coagulation mechanism of materials at the molecular level, paving the way for innovative approaches in designing new hemostatic materials.

Porous, environment-friendly chitosan-ZIF composite fibers for dye adsorption

Xuebin Hou,Wei Chen,Meihong Fan,Chen Shi

Chitosan (CS) is regarded as a promising sorbent due to its biodegradability, wide availability, easy processing, and multiple active groups. In this work, CS-based porous fibers (ZIF-n@CS) consisting of CS and zeolitic imidazolate frameworks (ZIFs) were fabricated through wet spinning and self-assembly methods. This combination effectively utilizes the dual advantages of CS and ZIFs, which effectively remove dyes from water and exhibit excellent recyclability. In addition, the adsorption mechanism of the adsorbent was explored through the analysis of adsorption kinetics, thermodynamics, isotherms, and chemical characterization after adsorption. This work provides an operable and universal strategy for the efficient adsorption of dyes, promoting the further promotion and application of MOFs and CS materials.

Glycosylation-Engineered Chitosan Bioelectronic Interfaces for P. aeruginosa Gradient Analysis

Kun Yan,Zongrun Zhang,Xianzhu Yang,Chenguang Yang,Zhentan Lu,Dong Wang

Point-of-care diagnostics for pathogen detection are crucial for informed clinical pretreatment strategies, rendering the emergence of electro-fluidic systems increasingly attractive. However, both optical and electrical sensing strategies still face challenges in achieving high sensitivity and specificity. Herein, we present a facile and general approach for effectively integrating an electro-fluidic device that enables optical-electrical dual-mode gradient analysis of bacterial populations, mediated by a 2-lactobionamidoethyl methacrylate (SLM)-functionalized chitosan film (CS). Notably, this rationally synthesized CS@SLM exhibits robust interfacial monolayer adsorption performance, serving as a versatile sensing platform for specific bacterial recognition, uniform capture, and rapid accumulation of Pseudomonas aeruginosa. Furthermore, the biointerface demonstrates distinct concentration-dependent adsorption behavior, particularly in diluted bacterial solutions, achieving remarkably high removal efficiencies (78.3–100%) while facilitating bacterial detection at ultralow concentrations (≤101 CFU/mL, 0–10 min). These characteristics enable the creation of a pronounced concentration gradient through subsequent diffusion processes in microfluidic channels, allowing for differential analysis of bacterial populations with thousands of fold enhancement in signal variation. Importantly, the surface-immobilized bacteria retain their bioactivities and can be effectively integrated with electrodes via the engineered biointerface, highlighting significant potential for electro-fluidic multimodal sensing systems. Collectively, this general strategy and versatile biointerface provide new insights into designing advanced electro-fluidic devices for precise bacterial density control and effective infection management.

3D-printable polyacrylamide/quaternary chitosan-based multifunctional hydrogel flexible sensors

Dingxin Wang,Lin Mei,Qian Liu,Xinhao Zou,Shaoshan Pan,Tianyu Xu,Panfeng Zhao,Tianzhi Luo

Hydrogel-based wearable flexible pressure sensors show great potential for applications in human health and motion monitoring. However, fabricating hydrogel sensors with excellent mechanical properties, sensitivity, and stability remains challenging. In this study, we developed a poloxamer-quaternized chitosan-polyacrylamide (P407/QCS-PAAm) hydrogel with multifunctional capabilities. These hydrogels exhibit high strength, impact resistance, excellent electrical conductivity, hydrophilicity, and outstanding antimicrobial properties. Due to the dynamic hydrogen bonding provided by QCS, the mechanical properties of the hydrogel can be adjusted by regulating the QCS content, achieving tensile strengths ranging from 0.14 to 0.47 MPa and compressive strengths from 2.07 to 5.49 MPa. The quaternary ammonium groups carried by QCS enhance the gel's conductivity (0.29 S/m) and its bacteriostatic effects. Furthermore, the incorporation of P407 improves the hydrogel's hydrophilicity. These properties make the gel an excellent biosafety material for wearable devices. Notably, the hydrogel precursor exhibits rate-tunable sol-gel transition properties, enabling 3D printing. The 3D-printed P407/QCS-PAAm hydrogel sensors demonstrate high sensitivity (GF < −0.5) across a wide strain range (−40 % to 0 % strain). Considering these combined properties, the gel demonstrates significant potential for use in flexible wearable and 3D-printed electronic devices.

Multifunctional CuTax nanozyme-based chitosan edible coatings for fruit preservation

Fengxian Luo,Xinchuang Wang,Wei Tian,Beiwei Zhu,Jiangning Hu

The preservation of fresh fruits is critically challenged by oxidative degradation and microbial contamination, which lead to quality deterioration and reduced shelf life. In this study, we took advantage of the multiple benefits of Taxfolin (Tax) and copper to develop CuTax nanozymes, and which were showed to have excellent free radical scavenging ability, as evidenced by 90.3 % ± 0.2 % DPPH and 85.61 % ± 0.08 % ABTS free radical scavenging rates. Additionally, the CuTax exhibited peroxidase-like (POD-like) activity and effective glutathione (GSH) depletion. Moreover, the CuTax were found to effectively suppress the colony formation of E. coli and S. aureus , reduce bacterial viability, and disrupt bacterial structures. Ultimately, a CuTax/CS composite coating/films for food preservation was successfully developed using chitosan (CS) as a carrier and the protective efficacy against food spoilage was evaluated using bananas and apples as representative fruits. These findings suggest that CuTax/CS composite coatings offer a multifunctional approach to active food packaging that effectively extend the shelf life and preserve the quality of fresh fruits.

Enhancing triboelectric nanogenerator performance using chitosan-modified multiwalled carbon nanotubes

Irfan Khan,Saad Ahmed,Zeeshan Haider,Kashan Memon,Azam Fareed

The rising demand for a sustainable and immaculate energy source on a global scale is fascinating regarding the usage and potential effects of green energy. Triboelectric nanogenerators have emerged as some of the major green energy sources, converting mechanical energy into electrical energy by using different electropositive and tribonegative materials. We used surface-modified multiwalled carbon nanotubes incorporated with a chitosan membrane as a tribopositive material in triboelectric nanogenerators for improved efficiency. We demonstrate that the chitosan-modified multiwalled carbon nanotube-based tribopositive material with PDMS as the tribonegative layer with a constant device size of 2 × 2 cm2 produced a short circuit current density (Jsc) of 42.004 mA m−2, an open circuit voltage (Voc) of 244.90 volts, and a peak power density of 4.22 W m−2, which can light up more than hundreds of LED lights. The high electrical performance of chitosan-modified multiwalled carbon nanotubes as a tribopositive material is attributed to the increased surface roughness and charge separation without causing significant charge leakage, which is produced by carbon nanotubes. Henceforth, the present work successfully introduces the use of chitosan-modified multiwalled carbon nanotubes as a tribopositive material for enhancing the TENG's output performance.

Multi-factor protective chitosan/polyvinyl alcohol aerogel antibacterial buffer packaging

Jing Jin,Yu Ren,Hua Zhou,Yujie Hou,Peng Jin,Yonghua Zheng,Zhengguo Wu

Berries, being juicy and thin-skinned, are prone to mechanical damage and microbial contamination during transportation. Therefore, developing functional cushioning packaging becomes crucial to minimize product losses. Herein, modified MXene was used to immobilize silver nanoparticles (AgNPs) and graft trans -2-hexenal (E2H), synthesizing MXene-NH 2 /AgNPs/E2H preservative. Subsequently, the CPMAE aerogel buffer packaging was obtained by MXene-NH 2 /AgNPs/E2H, chitosan and polyvinyl alcohol, and then using to the strawberry preservation. The physical entangling, electrostatic and hydrogen bonding between chitosan and polyvinyl alcohol, as well as the interaction with MXene-NH 2 /AgNPs/E2H endow CPMAE aerogel excellent buffering properties. Besides, only about 67.40 % of E2H and 5.80 % of AgNPs were released from the CPMAE aerogel after 7 days. Although the immobilization of MXene-NH 2 significantly reduced the release rate of AgNPs and E2H, the CPMAE aerogel still had excellent antioxidant (36.5 μg FeSO 4 /5 mg) and antimicrobial (reaches 99 %) capabilities. Moreover, these features enable the CPMAE aerogel buffer packaging to have an efficient fresh-keeping effect on strawberries and extend the storage time at 4 ℃ to 12 days. This study offers a novel approach for applying chitosan aerogel in buffer and antimicrobial packaging for berries.

Preparation of high-strength, colored and multifunctional chitosan fibers via a novel strategy of enzyme-catalyzed phenolic-amine crosslinking within chitosan fibers in a swollen state

Zhichao Song,Shujie Xu,Bo Xu,Man Zhou,Ying Liu,Ying Zhang,Li Cui,Ping Wang,Qiang Wang,Yuanyuan Yu

This study prepared high-strength, colored and multifunctional polyphenol-chitosan fibers via a novel strategy of laccase-mediated phenolic-amine crosslinking within chitosan fibers in a swollen state. A dense covalent network within the fiber was formed through Michael addition and Schiff base reactions. Compared to unmodified chitosan fibers, the enzymatically cross-linked fibers exhibited significantly enhanced mechanical properties, with an increase of about 270% in dry tensile strength (up to 156.8 MPa) and excellent wet stability. In addition, the polyphenol-chitosan fibers were given color and different properties by the covalent cross-linking of polyphenols into the fiber. The polyphenol-chitosan fibers had stable colors and did not fade in the environment. They also had an exceptional antibacterial rate of 99.99% and excellent UV protection. This laccase-mediated phenolic-amine crosslinking strategy provides an effective, sustainable method for producing high-performance, bio-based functional fibers.

Removal of quinclorac herbicide from aqueous solution by chitosan/montmorillonite bionanocomposite

Chunxia Ding,Daoxin Gong,Peng Yu,Jihai Shao,Mei-E Zhong

Chitosan–montmorillonite bionanocomposite (CTS–MMT) was prepared for removing the herbicide quinclorac (QC) from aqueous solution. The resulting nanocomposites were characterized by X-ray powder diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), Brunauer–Emmett–Teller (BET) surface area and Barrett–Joyner–Halenda (BJH) pore size distribution analysis. The adsorption of QC onto CTS–MMT as a function of adsorbent dosage, pH, temperature, ionic strength, contact time, and initial QC concentration was investigated by conducting batch adsorption experiments. The results showed that CTS–MMT exhibited higher adsorption affinity and higher adsorption capacity towards QC than the unmodified montmorillonite. The adsorption kinetics data of QC onto CTS–MMT were well described by the pseudo-second-order kinetics model. Adsorption isotherms fitted well with the Freundlich isotherm model in the heterogeneous adsorption process. Adsorption thermodynamics study indicated that the adsorption of QC onto CTS–MMT was a spontaneous and endothermic process. The QC molecules on CTS–MMT could hardly be desorbed by water, but could be desorbed by 0.1 mol/L NaOH solution. QC adsorption on CTS–MMT may involve electrostatic attraction, cation–dipole, hydrogen bonding, and van der Waals interactions. The findings of this study suggest that CTS–MMT is a potential adsorbent for quinclorac pollution remediation.

Cytotoxicity and Antibacterial Activity of Chitosan-organic Rectorite Intercalated Nanofibrous Mats

Du Jing,Li Xueyong,Yang Chen,Li Wei,Huang Weijuan,Huang Rong,Zhou Xue,Deng Hongbing

The chitosan (CS)-organic rectorite (OREC) composites are prepared with the method of solution intercalation. Electrospun nanofibrous mats are made from intercalated CS-OREC composites at different mass ratios. The properties and structure of intercalated CS-OREC can be verified by small angle X-ray diffraction (SAXRD) and transmission electron microscopy (TEM) micrographs. Energy dispersive X-ray (EDX) spectroscopy and fourier transform infrared (FT-IR) spectra reveal that OREC presents in the composite nanofibrous mats. The MTT assay indicates that the cytotoxicity of the intercalated CS-OREC nanofibrous mats is low. Besides, the content of OREC in nanofibrous mats has a significant influence on the viability of cells. Antibacterial test confirms that OREC can increase the inhibition rate of the nanofibrous mats against Escherichia coli.
