Molecularly imprinted polymers for extraction of malachite green from fish samples prior to its determination by HPLC

Li Lu,Lin Zheng-zhong,Chen Xiao-mei,Zhang Hong-yuan,Lin Yi-dong,Lai Zhu-zhi,Huang Zhi-yong

Molecularly imprinted polymer (MIP) particles for malachite green (MG) were prepared by emulsion polymerization using methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker, and a combination of Span-80 and Tween-80 as an emulsifier. The MIP particles were characterized by SEM micrographs and FT-IR spectra. Their binding capacity for MG was evaluated based on kinetic and isothermal adsorption experiments and compared to non-imprinted polymer particles. Analytical figures of merit include an adsorption equilibrium time of 15 min, an adsorption capacity of 1.9 mg∙g‾ 1 in acetonitrile-water (20:80), and an imprinting factor of 1.85. The MIP particles were successfully applied to the extraction of MG from fish samples spiked with MG and the other interfering substances prior to its determination of MG by HPLC. Spiked samples gave recoveries of MG that ranged from 86 to 104 %, much higher than that of the other interfering substance. Molecularly imprinted polymer particles for malachite green (MG) were prepared by emulsion polymerization. They show adsorption capacity of 1.9 mg∙g‾¹ with imprinting factor of 1.85, and were successfully applied to the extraction of MG from spiked fish samples.

Bioinspired Polydopamine Sheathed Nanofibers for High-Efficient in Vivo Solid-Phase Microextraction of Pharmaceuticals in Fish Muscle

Jianqiao Xu,Shuyao Huang,Rongben Wu,Ruifen Jiang,Fang Zhu,Jing Wang,Gangfeng Ouyang

In this study, electrospun nanofibers were used as solid-phase microextraction (SPME) fiber coatings after substituting the water-soluble sheath of the emulsion electrospun polystyrene (PS)@Plurinic F-127 core–sheath nanofibers with biocompatible and water-stable polydopamine (PDA) and subsequently being appropriately cross-linked with glutaraldehyde (GA) to enhance the strength of the electrospun architecture. The novel custom-made PS@PDA-GA coating was wettable in aqueous solutions and thus exhibited much higher extraction efficiency than the nonsheathed PS nanofiber coating and the thicker polydimethylsiloxane (PDMS) coating. The novel coating also possessed excellent stability (relative standard deviations (RSDs) less than 7.3% for six sampling-desorption cycles), interfiber reproducibility (RSDs less than 14.3%), and antibiofouling ability, which were beneficial for in vivo sampling. The PS@PDA-GA fiber was used to monitor pharmaceuticals in dorsal-epaxial muscle of living fish, and satisfactory sensitivities with the limits of detection in the range of 1.1 (mefenamic acid) to 8.9 (fluoxetine) ng·g–1 and comparable accuracies to liquid extraction were achieved. In general, this study explored a convenient and effective method to sheath nanofibers for high-efficient in vivo SPME of analytes of interest in semisolid tissues.

Development of a lateral flow immunoassay for the detection of total malachite green residues in fish tissues

Naifeng Xu,Liqiang Li,Shanshan Song,Liguang Xu,Hua Kuang,Chuanlai Xu

We developed a sensitive lateral flow immunoassay (LFI) for the detection of total malachite green (MG) residues in fish tissues. LFI was based on a colloidal gold nanoparticle-labeled monoclonal antibody against MG , which was generated by fast immunization of carboxymalachite green-bovine serum albumin. The half-maximum inhibition concentration (IC50) of MG monoclonal antibody (MG mAb) was 0.057 ng/ml with a linear range of 0.020–0.162 ng/ml. Following the optimization of LFI, a qualitatively visual limit of detection of 1 ng/ml and a semi-quantitatively limit of detection of 0.418 ng/ml (IC50 of the calibration curve) were obtained for total MG residues in 8 min. Cross-reactivity with most MG analogs was negligible, expect with crystal violet (CV; 78.6% cross-reactivity). LFI is a novel and sensitive assay based on MG mAb that has applications for the rapid detection of total MG and CV in fish products.

Ultrasensitive and Highly Selective Detection of Bioaccumulation of Methyl-Mercury in Fish Samples via Ag0/Hg0 Amalgamation

Li Deng,Yan Li,Xiuping Yan,Jun Xiao,Cheng Ma,Jing Zheng,Shaojun Liu,Ronghua Yang

Methylmercury (CH3Hg+), the common organic source of mercury, is well-known as one of the most toxic compounds that is more toxic than inorganic or elemental mercury. In seabeds, the deposited Hg2+ ions are converted into CH3Hg+ by bacteria, where they are subsequently consumed and bioaccumulated in the tissue of fish, and finally, to enter the human diet, causing severe health problems. Therefore, sensitive and selective detection of bioaccumulation of CH3Hg+ in fish samples is desirable. However, selective assay of CH3Hg+ in the mercury-containing samples has been seriously hampered by the difficulty to distinguish CH3Hg+ from ionic mercury. We report here that metal amalgamation, a natural phenomenon occurring between mercury atoms and certain metal atoms, combining with DNA-protected silver nanoparticles, can be used to detect CH3Hg+ with high sensitivity and superior selectivity over Hg2+ and other heavy metals. In our proposed approach, discrimination between CH3Hg+ and Hg2+ ions was realized by forming Ag/Hg amalgam with a CH3Hg+-specific scaffold. We have found that Ag/Hg amalgam can be formed on a CH3Hg+-specific DNA template between silver atoms and mercury atoms but cannot between silver atoms and CH3Hg+. With a dye-labeled DNA strand, the sensor can detect CH3Hg+ down to the picomolar level, which is >125-fold sensitive over Hg2+. Moreover, the presence of 50-fold Hg2+ and 106-fold other metal ions do not interfere with the CH3Hg+ detection. The results shown herein have important implications for the fast, easy, and selective detection and monitoring of CH3Hg+ in environmental and biological samples.

Rapid detection of malachite green in fish based on CdTe quantum dots coated with molecularly imprinted silica

Le Wu,Zheng-zhong Lin,Hui-ping Zhong,Ai-hong Peng,Xiao-mei Chen,Zhi-yong Huang

A sensitive fluorescence sensor for the detection of malachite green (MG) was fabricated by grafting molecularly imprinted polymers (MIPs) onto the surface of CdTe quantum dots (QDs). The MIP-coated QDs were synthesized via a reverse microemulsion method using (3-aminopropyl)triethoxysilane (APTES) and tetraethyl orthosilicate (TEOS) as functional monomer and cross-linker, respectively. The optimum molar ratio of MG, functional monomer and cross-linker was 1:3:10. The MIP-coated QDs exhibited uniform spheres with diameter around 49   nm and excellent fluorescence emission at λ ex 370   nm. A linear relationship with two segments between the relative fluorescence intensities and the MG concentrations ranging from 0.08 to 20   μmol·L −1 could be obtained with a detection limit of 12   μg·kg −1 . The fluorescent probe was successfully applied to the determination of MG in fish samples with the spiked recoveries ranging from 94.3% to 109.5% which were in accordance with those of the measurement by HPLC-UV.

Effect of the Introduction of Fish Collagen on the Thermal and Mechanical Properties of Poly(lactic acid)

Hui Ma,Jiajia Shen,Qun Yang,Jie Zhou,Shuangshuang Xia,Jianda Cao

Collagen/poly(lactic acid) composite was developed by covalent combination of collagen and poly(lactic acid) and the thermal and mechanical properties of the composites were also investigated. Owing to the triple helix structure, collagen exhibited important advances in the improvement of thermal and mechanical properties of poly(lactic acid) compared with other polymers. In our work, collagen molecules were covalently grafted on the poly(lactic acid) chains through the condensation of amino- and carboxyl- groups with the catalyzation of 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride. Moreover, the enthalpy of collagen/poly(lactic acid) materials was 30.09 J/g at the highest, compared to the enthalpy of 1.52 J/g for pure PLA. In addition, 88.60% improvement in tensile strength and 176.88% improvement in elongation at break were observed. The strategy for improving the properties of poly(lactic acid) by protein can be useful to develop high impact resistance of PLA-based composites and expand the application fields of PLA materials.

Protein-enriched fish “biowaste” converted to three-dimensional porous carbon nano-network for advanced oxygen reduction electrocatalysis

Chaozhong Guo,Rong Hu,Wenli Liao,Zhongbin Li,Lingtao Sun,Dongping Shi,Yanrong Li,Changguo Chen

Recycling and utilizing organic biowastes will effectively help to decrease the damage to the natural environment and synchronously facilitate the development of new carbon materials for energy applications. In this study, we directly convert protein-rich fish-scale biowaste to hierarchically porous three-dimentional (3D)-network nanocarbons via two-step pyrolysis process combined with ZnCl 2 activation and acidic-treatment. It is interestingly found that this material exhibits more excellent oxygen reduction electrocatalytic activity and stability compared to the commercial 20   wt% Pt/C catalyst in both alkaline and acidic solutions, which can be closely correlated to its chemical state of nitrogen atoms, BET surface area and inner porous structure. The addition of ZnCl 2 activator during pyrolysis process can help to produce the 3D network nanostructure and then to enhance the mesopore surface area, making for the improvement of oxygen reduction performance. More remarkably, the ORR onset potential on our material is about 60   mV higher than that on the Pt/C catalyst in alkaline electrolyte. In addition, we also propose that pyridinic- and graphitic-nitrogen species may be key factors to be responsible for the electrocatalytic activity. This study can encourage the exploration of high porosity nanocarbons from widely-existed biowastes, functioning as highly active and stable oxygen reduction electrocatalysts.

Engineered nickel-based metal − organic framework nanofiber membrane with fish-scale microstructure for enhanced photocatalytic CO2 conversion

Pengfei Lin,Yanglei Huang,Qingyuan Yang,Hao Wu,Xinping He,Chunhai Yi,Muhammad Usman Farid,Alicia Kyoungjin An,Jiaxin Guo

Metal-organic frameworks (MOFs) have attracted considerable interest due to their wide-ranging applications in heterogeneous catalysis. However, their limited separation efficiency and challenging recovery from reaction mixtures pose practical challenges. Membrane-based catalysts offer a solution by preventing light scattering and aggregation while enabling easier recovery compared to powdered catalysts. While MOF membranes have been extensively studied for photocatalytic pollutant decomposition, their application in photocatalytic CO 2 reduction remains underexplored. Hence, in this study, we present the successful immobilization of defective Ni-MOF-74 (NMD) nanofragments containing more unsaturated Ni active sites onto electrospun polyacrylonitrile (PAN) nanofiber membranes (NFMs). The resulting composite catalyst, referred to as NMD/PAN NFMs (NPNs) with a uniform fish-scale microstructure, was evaluated for photocatalytic CO 2 reduction under mild conditions. The results demonstrate exceptional photocatalytic performance and stability of NPNs. The membranes exhibited a CO production rate of 1,641 μmol g -1 h −1 with a selectivity of 91 %, surpassing even that of powdered NMD. Moreover, the NPNs demonstrated advantages such as ease of recycling and reusability, further enhancing their practical applicability. This innovative approach to constructing high-performance MOF nanofiber membranes opens up new possibilities for a broader range of applications in CO 2 photocatalytic conversion.

Development of Sensitive R6G/AuNCs Ratiometric Fluorescent Probes for the Detection of Biogenic Amines in Fish Products

Yutong Huang,Simiao Zhang,Mei Zhou,Xiaokang Xu,Weiqing Sun,Jing Ma,Long Wu

Biogenic amines (BAs), produced in fish and seafood due to microbial contamination, pose significant health risks. This study introduces a novel ratiometric fluorescent probe, synthesized by integrating rhodamine 6G(R6G) and gold nanoparticles (AuNCs), for the sensitive and specific detection of BAs. The probe operates on the principle of BAs hydrolysis, catalyzed by diamine oxidase, to produce hydrogen peroxide (H2O2), which selectively quenches the fluorescence of AuNCs at 620 nm, while the fluorescence of R6Gat 533 nm remains unaffected. The ratio of I620/I553demonstrated excellent linearity with a wide dynamic range (1–1000 μM) and a low detection limit of 0.1 μM. Validation using grass carp samples showed high recovery rates (97.57% to 104.29%), confirming the probe’s efficacy and potential for practical application in food safety monitoring.

High strength and water tolerance fish gelatin-xanthan gum acid-induced electrostatic film by melt extrusion method

Xiaohan Chen,Zhuolin Liu,Wenya Ma,Hao Wang,Qingfeng Dong,Li Li

Gelatin is a biodegradable natural polymer that can be utilized in preparing edible films , and its films tend to have defects in mechanical properties and water tolerance, limiting their application. In this study, we combined the acid-induced electrostatic system to prepare the gelatin extruded film, and the relevant mechanisms were explored. The films' thermal stability, microstructure, mechanical properties, water tolerance, and degradability were also evaluated. Films were prepared by adding different levels of xanthan gum and malic acid to the film substrate. Compared to FG-M-0%, FG-MX-0.5% showed a 120.61% increase in tensile strength, a 9.79 °C and a 16.72 °C increase in melting temperature and glass transition temperature, respectively, the acidified FG-MX group as a whole had a higher water contact angle and lower swelling rate. FTIR and XRD confirmed that acid induction enhanced the electrostatic interactions and hydrogen bonding between gelatin-xanthan gum. The results showed that the melt-extruded gelatin film could also maintain the acid-induced electrostatic effect well, with stronger physical properties and better water tolerance, and were conducive to the efficient preparation of gelatin films with desirable properties and the expansion of their range of applications.

Design and characterization of edible chitooligosaccharide/fish skin gelatin nanofiber-based hydrogel with antibacterial and antioxidant characteristics

Yanjing Liu,Xiaodong Xia,Xiyue Li,Fuming Wang,Yaping Huang,Botian Zhu,Xuyang Feng,Ying Wang

Antibacterial and active packaging materials have gained significant research attention in response to the growing interest in food packaging. In this investigation, we developed hydrogel packaging materials with antibacterial and antioxidant properties by incorporating chitooligosaccharide (COS) and fish skin gelatin (FSG) nanofiber membranes, which readily absorbed water and exhibited swelling characteristics. The nanofiber membranes were fabricated by electrospinning technology, embedding COS within FSG, and subsequently crosslinked through the Maillard reaction facilitated by the addition of glucose. The behavior of conductivity, viscosity, and surface tension in the spinning solutions was analyzed to understand their variation patterns. Scanning electron microscopy (SEM) results revealed that the crosslinked COS/FSG nanofiber membranes possessed a uniform yet disordered fiber structure, with the diameter of the nanofibers increasing as the COS content increased. Remarkably, when the COS content reached 25 %, the COS/FSG nanofiber membranes (CF-C-25) exhibited a suitable fiber diameter of 437.16 ± 63.20 nm. Furthermore, the thermal crosslinking process involving glucose supplementation enhanced the hydrophobicity of CF-C-25. Upon hydration, the CF-H-25 hydrogel displayed a distinctive porous structure, exhibiting a remarkable swelling rate of 954 %. Notably, the inclusion of COS significantly augmented the antibacterial and antioxidant properties of the hydrogel-based nanofiber membranes. CF-H-25 demonstrated an impressive growth inhibition of 90.56 ± 5.91 % against E. coli , coupled with excellent antioxidant capabilities. In continuation, we performed a comprehensive analysis of the total colony count, pH, TVB-N, and TBA of crucian carp. The CF-H-25 hydrogel proved highly effective in extending the shelf life of crucian carp by 2–4 days, suggesting its potential application as an edible membrane for aquatic product packaging.

Phylogeny-metabolism dual-directed single-cell genomics for dissecting and mining ecosystem function by FISH-scRACS-seq

Xiaoyan Jing,Yanhai Gong,Zhidian Diao,Yan Ma,Yu Meng,Jie Chen,Yishang Ren,Yuting Liang,Yinchao Li,Weihan Sun,Jia Zhang,Yuetong Ji,Zhiqi Cong,Shengying Li,Bo Ma,Zhisong Cui,Li Ma,Jian Xu

Microbiome-wide association studies (MWASs) have uncovered microbial markers linked to ecosystem traits, but the mechanisms underlying their functions can remain elusive. This is largely due to challenges in validating their in situ metabolic activities and tracing such activities to individual genomes. Here, we introduced a phylogeny-metabolism dual-directed single-cell genomics approach called fluorescence- in situ -hybridization-guided single-cell Raman-activated sorting and sequencing (FISH-scRACS-seq). It directly localizes individual cells from target taxon via an FISH probe for marker organism, profiles their in situ metabolic functions via single-cell Raman spectra, sorts cells of target taxonomy and target metabolism, and produces indexed, high-coverage, and precisely-one-cell genomes. From cyclohexane-contaminated seawater, cells representing the MWAS-derived marker taxon of γ-Proteobacteria and that are actively degrading cyclohexane in situ were directly identified via FISH and Raman, respectively, then sorted and sequenced for one-cell full genomes. In such a Pseudoalteromonas fuliginea cell, we discovered a three-component cytochrome P450 system that can convert cyclohexane to cyclohexanol in vitro , representing a previously unknown group of cyclohexane-degrading enzymes and organisms. Therefore, by unveiling enzymes, pathways, genomes, and their in situ cellular functions specifically for those organisms with ecological relevance at one-cell resolution, FISH-scRACS-seq is a rational and generally applicable approach to dissecting and mining microbiota functions.
