Raman spectroscopy in the second near-infrared (NIR-II, 1000–1700 nm) window has received ever-increasing attention in intraoperative tumor imaging due to its high specificity and penetration depth. However, the overwhelming majority of existing NIR-II Raman probes are fabricated by SERS with substrate enhancement, which raises serious biosafety concerns. Herein, we present van der Waals (vdW) enhanced Raman scattering (vdW-ERS) effect based on intermolecular charge transfer of dithiolene to develop ultrabright metal-free NIR-II Raman probes. The intermolecular charge transfer within dithiolene through vdW stacking is well illustrated by independent gradient model and ultrafast spectroscopy, which significantly boosts the intrinsic Raman properties with ultrahigh scattering cross-section (1.59 × 10 −19 cm 2 ) as well as the photothermal conversion efficacy (60.4%) through an additional femtosecond-scale non-radiative decay channel. Moreover, the fluorescence-free property of dithiolene greatly inhibits the spectral backgrounds and improves the photostability. In the tumor models of triple-negative breast cancer, vdW-ERS probes enable intraoperative CXCR4-targeted Raman-guided surgical resection and postoperative hyperthermia ablation of the residual microscopic tumors, resulting in approximately 100% survival rate. Importantly, vdW-ERS probes are hepatically excreted (ca. 92% excretion within two months), in stark contrast to accumulation in the liver for typical gold substrate-enhanced counterparts. Free from fluorescence background and photobleaching, intermolecular vdW-ERS effect presents a new mechanism for fabricating charge transfer Raman probes with the potential for clinical translation.

Triple-negative breast cancer (TNBC) is an aggressive and often fatal disease, especially since the brain metastasis of TNBC has been a particularly severe manifestation. However, brain metastasis in TNBC at early stages often lacks noticeable symptoms, making it challenging to detect. Near-infrared II (NIR-II) fluorescence microscopic imaging obtains long wavelength, which enables reduced scattering, high spatial resolution and minimal autofluorescence, it is also a favorable imaging method for tumor diagnosis. PbS@CdS quantum dots (QDs) are one of the popular NIR-II fluorescence nanoprobes for well brightness. In this study, NIR-II emissive PbS@CdS QDs were utilized and further encapsulated with thiol-terminated poly(ethylene oxide) (SH-PEG, MW = 5000) to form PbS@CdS@PEG QDs nanoparticles (NPs). The obtained PbS@CdS@PEG QDs NPs were then characterized and further studied in detail. The PbS@CdS@PEG QDs NPs had large absorption spectra, exhibited strong NIR-II fluorescence emission at approximately 1300 nm, and possessed good NIR-II fluorescence properties. Then, the mice model of early-stage brain metastases of TNBC was established, and the PbS@CdS@PEG QDs NPs were injected into the tumor-bearing mice for NIR-II fluorescence microscopic bioimaging. The brain vessels and tumors of the living mice were detected with high spatial resolution under the NIR-II fluorescence microscopic imaging system with irradiation of 808 nm laser. The tumor tissues were further restricted and prepared as thin slices. The NIR-II fluorescence signals were collected from the tumor slices with high spatial resolution and signal-to-background ratio (SBR). Thus, the PbS@CdS@PEG QDs NPs-assisted NIR-II fluorescence microscopic system can effectively achieve targeting brain metastases of TNBC imaging, offering a novel and promising approach for TNBC-specific diagnosis.

Ferroptosis, characterized by iron-dependent lipid peroxidation, represents a promising therapeutic target for cancer treatment. Strategies that disrupt intracellular antioxidant systems to induce ferroptosis in cancer cells have been extensively explored. Herein, we developed a pH-responsive phototheranostic agent (designed as FSB 4 Ca NPs) by encapsulating conjugated boron dipyrromethene tetramers (B4) within ferric ion-sulfasalazine metallo-network polymercoated calcium carbonate hollow nanoparticles. Sulfasalazine, a known ferroptosis inducer that inhibits System X c --mediated cysteine influx, synergizes with ferric ion-driven glutathione (GSH) depletion to collectively amplify intracellular lipid peroxidation. In addition to serving as a second near-infrared (NIR-II) fluorophore for tracking the in vivo distribution of FSB 4 Ca NPs, B4 mediates a photothermal effect that significantly enhances lipid peroxidation induction by boosting the Fenton catalytic activity of ferrous ions. Combined with localized 915-nm laser irradiation, intravenously administrated FSB 4 Ca NPs achieved substantial tumor suppression in mouse models, with a complete remission rate of 80%. This study establishes a facile strategy for developing long-circulating NIR-II phototheranostic agents with self-amplified lipid peroxidation induction capacity, enabling photothermally augmented ferroptosis for cancer therapy.

Overcoming thermoresistance and achieving precise, efficient tumor therapy remain major challenges. Heat shock protein 70 (HSP70) expression and autophagy activation are two major cellular defense mechanisms associated with thermal resistance. However, strategies that target both mechanisms to improve tumor treatment efficacy remain limited. Herein, this study presents a novel type of near-infrared II (NIR-II)-responsive theranostic nanoplatform (IR-820@γ-PGA/Arg NPs, PIRA NPs) that integrates photothermal therapy (PTT), photodynamic therapy (PDT), nitric oxide (NO)-mediated gas therapy (GT), and dual-pathway inhibition mechanisms to overcome thermal resistance. The theranostic nanoplatform utilizes a γ-polyglutamic acid (γ-PGA) to encapsulate the NIR-II dye IR-820, which forms stable J-aggregates to improve NIR-II fluorescence emission and photothermal conversion efficiency. The nanoplatform adsorbs L-arginine (Arg) and releases NO under 808 nm laser excitation, exhibiting excellent photothermal properties and NIR-II fluorescence emission for precise tumor treatment. PIRA NPs overcome photothermal resistance via two pathways: suppressing HSP70-mediated thermoresistance and disrupting the autophagy-lysosomal degradation pathway (LC3B/p62 axis) through Arg-derived NO generation. This dual-pathway blockade effectively neutralizes tumor defenses, achieving a 92.7% tumor inhibition rate with minimal systemic toxicity. The γ-PGA framework enhances the stability of J-aggregates and their biocompatibility, further improving therapeutic efficacy. This work provides a versatile nanotherapeutic platform based on a promising strategy for reversing thermoresistance and achieving highly efficient cancer therapy.

Near-infrared II (NIR-II) emissions are unique and selective for non-destructive detection owing to their specific response to temperature changes and the ability to cover the characteristic overtone absorption bands of molecular functional groups. However, cross-interference in the spectral responses occurs for simultaneous temperature sensing and chemical reagent identification, therefore it is necessary to realize reliable decoding. Herein, the Nd 3+ / Er 3+ co-doped Y 2 MgTiO 6 phosphors with NIR-II emissions under 808 nm excitation have been designed and prepared, and the temperature sensing properties have been evaluated, in which the distinctive thermally enhanced luminescence is attributed to the efficient energy transfer process from Nd 3+ to Er 3+ ions. Besides, NIR-II-emitting composite film based on the rationally designed phosphors has been fabricated as the sensing platform, and the concentration recognition of water and methanol is enabled by the overtone absorption of C─H and O─H bonds. Finally, a Large Scale Convolutional Neural Network model is constructed to synchronously decouple, which can accurately extract temperature and chemical concentration from complex spectral data. This work paves the way for constructing a feasible and compact platform for dual-function intelligent sensors.

Bacterial infection is now a significant obstacle to the effective repair and regeneration of both soft tissues and bone in clinical settings. Managing infected sites is challenging due to multiple antimicrobial resistance, abnormal inflammatory responses, and an excessive inflammatory microenvironment. To meet the complex healing needs following soft tissue or bone, an immunomodulatory thermosensitive hydrogel was designed and synthesized. The composite hydrogel (CQDs@UPDA@gel) combines carbon quantum dots (CQDs) as the second near-infrared window (NIR-II) photothermal agents (PTAs) and ultrasmall polydopamine nanoparticles (UPDA NPs) as reactive oxygen species (ROS) scavengers and immunomodulators, embedded within a chitosan matrix coordinated with Cu2+ that enhances the good swelling performance and stable rheological properties of the hydrogel. This synergistic approach simultaneously inhibits bacterial infection and alters the immunosuppressive microenvironment, thereby promoting coordinated soft tissue or bone regeneration. In vivo studies have shown that the hydrogel can encourage the regeneration of soft tissue or bone under NIR-II laser irradiation for the treatment of infectious wounds, subcutaneous abscesses, and infectious bone defects in the maxillofacial region. Overall, this project offers a general strategy for creating multifunctional hydrogels that both eradicate pathogens and modulate the immune microenvironment to support the repair of complex soft tissue or bone infections.

Cr-activated phosphor-converted light sources have attracted widespread attention in various applications based on near-infrared (NIR) spectroscopy. Unfortunately, Cr ions usually face the issue of multivalence and typically exist in the valence of +3, which gives the emission mainly at NIR-I (700–1000 nm) and greatly limits its potential application. In this study, we propose the control strategies of site occupancy confinement and sintering atmosphere to synthesize Cr4+-activated Li2SrGeO4 (LSGO:Cr4+) phosphors. Interestingly, the as-optimized LSGO:Cr4+ achieves a NIR broadband emission peak at 1170 nm with an FWHM of 208 nm upon excitation at 680 nm and exhibits excellent thermal stability and high efficiency with the external quantum efficiency of 30.0%. Furthermore, the as-fabricated LSGO:Cr NIR phosphor-converted light emitting diode (NIR pc-LED) device shows a high power of up to 69.1 mW at 550 mA. Furthermore, as NIR-II (1000–1700 nm) light source, the phosphor successfully distinguishes deuterium and non-deuterium reagents. For example, the quantitative detection of H2O in heavy water (D2O) is demonstrated by NIR spectroscopy with a limit of detection (LOD) of 0.47 vol%. Our findings will open up a new avenue to develop novel Cr4+-activated NIR-II phosphors and NIR-II light sources.

The deep-seated locations and hypoxic microenvironments of tumors critically hinder conventional phototherapies. To overcome these limitations, we engineered a novel Au@Cu 2 − x Se nanoprobe via precise aspect ratio control of gold nanorods for NIR-II plasmonic resonance and site-specific growth of Cu 2 − x Se domains, resulting in the formation of a unique “lollipop” nanostructure. This design enables single-wavelength NIR-II laser-activated photothermal therapy and dynamic dual-modal therapy with intrinsic hypoxia tolerance: the nanoprobes achieve exceptional photothermal stability while simultaneously driving photothermally enhanced chemodynamic activity through accelerated Fenton-like catalytic cycles. Crucially, this strategy not only eradicates deep tumors but also activates systemic antitumor immunity. Combined with real-time photoacoustic imaging guidance, the platform establishes an oxygen-independent paradigm for treating hypoxic solid tumors. Graphical

Vanadium carbide (V 2 C) MXene shows great potential for addressing challenging implant-associated infections in bone regeneration due to its strong photothermal conversion efficiency. However, its photothermal efficacy is restricted to the near-infrared I (NIR-I) region due to a limited absorption range. To address this, we designed platinum nanoparticle-decorated V 2 C heterostructures (Pt@V 2 C) via an in situ growth method, leveraging Pt’s plasmonic and catalytic properties to extend the photoresponse to the NIR-II window. Subsequently, Pt@V 2 C was integrated into poly-L-lactic acid (PLLA) to fabricate PLLA-Pt@V 2 C scaffolds with photothermal antibacterial function by selective laser sintering. The optimized PLLA-Pt@V 2 C scaffold achieves a record photothermal conversion efficiency (56.03% at 1064 nm), triggering simultaneous hyperthermia (>52 °C) and catalytic ·OH radical generation. In vitro studies demonstrate exceptional antibacterial efficacy against Staphylococcus aureus and Escherichia coli, achieving over 99% killing rates upon 1064 nm near-infrared irradiation. Furthermore, the scaffold demonstrated significant inhibition of biofilm formation, achieving over 90% reduction in biofilm biomass. Moreover, the scaffold demonstrated high cell viability, confirming its dual functionality of potent bactericidal activity and biocompatibility that supports tissue regeneration. This work provides a feasible strategy for combating implant-associated infections.

Acute sinusitis, common across populations, worsens seasonally due to pollution and improper medication, increasing the risk of chronic disease and impairing quality of life. To improve treatment and limit antimicrobial resistance in acute sinusitis, we developed a natural injectable QACS/SA@Py@Ta 4 C 3 hydrogel. It integrates the traditional Chinese medicine phillyrin with Ta 4 C 3 quantum dots and natural polymers—quaternary chitosan and sodium alginate. The hydrogel is designed for NIR II–mediated treatment of acute sinusitis via combined photothermal and photodynamic therapy. It exhibited a photothermal conversion efficiency of 22.6% and generated transient ROS, as demonstrated in vitro. Bacteriostatic experiments revealed that the hydrogel inhibited S. aureus with an efficiency of 99.7%. Furthermore, cell studies showed that the hydrogel was minimally cytotoxic, modulated macrophage cytokine expression, and displayed anti-inflammatory and antioxidant activities. Finally, in a rabbit sinusitis model, hydrogel implantation at the treatment site significantly reduced bacterial burden and decreased inflammation after NIR II laser treatment, demonstrating promising therapeutic potential.

Reactive oxygen species (ROS)-mediated cell cycle arrest mechanism shows distinguished potential for the inhibiting of cancer cells. However, poly ADP-ribose polymerase (PARP), an enzyme which can contribute to the repair of single-strand DNA nicks, largely limited the efficiency in cell cycle arrest. Here, polydopamine (PDA)/hyaluronic acid (HA) decorated and protein translation inhibitor homoharringtonine (HHT) loaded tellurium (Te)-prussian blue (PB) nanorods ( [email protected] @PDA-HA/HHT NRs) were synthesized for boosted G2 cell cycle arrest to realize tumor ablation. Under NIR-II irradiation, Te and Fe elements can collectively generate ROS and deplete GSH due to the fenton-like reaction and metal-reducing reaction under tumor microenvironment (TME), thus disturbing the DNA structure to induce cell cycle arrest. More importantly, HHT suppresses PARP production to induce DNA repair dysfunction, thereby boosting DNA oxidative damage. Remarkably, this nanoplatform degrades into ionic form under acid condition, illustrating inherently TME-responsive biodegradability. The anti-tumor evaluation illustrated that [email protected] @PDA-HA/HHT NRs could notably destroy tumor cells in both cellular experiments and mouse models. This study made a successful attempt in boosted cell cycle arrest and encouraged more biological explorations on biodegradation nanomaterials, especially in combating cancers.

IR820, an analog of FDA-approved indocyanine green, is a promising second near-infrared window (NIR-II) fluorescence probe with better NIR-II fluorescence stability and great clinical transformation potential. Moreover, its fluorescence can be further remarkably enhanced by the interaction with albumin. Therefore, it is significant to flexibly design IR820–albumin complex using endogenous or exogenetic albumin to meet the requirements of different biological applications. Herein, we show the rational synthesis of IR820–albumin complex for NIR-II fluorescence imaging-guided surgical treatment of tumors and gastrointestinal obstruction. We compared the NIR-II fluorescence imaging ability of IR820 pre-incubated with albumin or not to visualize tumors and the gastrointestinal tract in vivo and found that the formation of IR820–albumin was essential for the intense NIR-II fluorescence. For imaging-guided tumor treatment, after intravenous injection of free IR820, IR820–albumin complex can be formed in vivo due to the presence of plenty of albumin in the blood. For imaging-guided gastrointestinal obstruction removal, IR820–albumin complex should be synthesized in vitro before oral administration. NIR-II fluorescence imaging-guided surgeries were successfully realized in both tumor resection and gastrointestinal obstruction removal. Besides, toxicity assessments in vitro and in vivo confirmed the good biocompatibility of IR820. Our study provides a flexible paradigm for IR820-based NIR-II fluorescence imaging and surgical navigation towards different diseases.
