The infections of chicken coccidiosis impact the welfare of chickens and the economical production of poultry. Eimeria mitis is ubiquitous in chicken coccidiosis, and E. mitis infection can significantly affect the productivity of birds. Up to now, few efficient vaccines against E. mitis have been reported, whereas the recombinant subunit vaccines delivered by nanomaterials may elicit an encouraging outcome. Thus, in this study, we chose E. mitis 1a (Em1a) protein as the candidate antigen to generate Em1a preparations. The recombinant Em1a (rEm1a) protein was encapsulated with poly lactic-co-glycolic acid (PLGA) and chitosan (CS) nanospheres. The physical characterization of the rEm1a-PLGA and rEm1a-CS nanospheres was investigated, and the resulting nanospheres were proven to be nontoxic. The protective efficacy of rEm1a-PLGA and rEm1a-CS preparations was evaluated in E. mitis-challenged birds in comparison with two preparations containing rEm1a antigen emulsified in commercially available adjuvants. ELISA assay, flow cytometry analysis, and quantitative real-time PCR (qPCR) analysis indicated that vaccination with rEm1a-loaded nanospheres significantly upregulated the secretions of antibodies and cytokines and proportions of CD4+ and CD8+ T lymphocytes. Compared with the other three preparations, rEm1a-PLGA nanosphere was more effective in improving growth performance and inhibiting oocyst output in feces, indicating that the PLGA nanosphere was associated with optimal protection against E. mitis. Collectively, our results highlighted the advantages of nanovaccine in eliciting protective immunity and may provide a new perspective for developing effective vaccines against chicken coccidiosis.

Covalent immobilization of bioactive compounds onto modified poly lactic-co-glycolic acid (PLGA) surfaces is being rapidly developed in tissue engineering, but the compounds and the grafting procedure require optimization. Here, PLGA membranes were grafted with various ratios of collagen/chitosan (COL/CHI) composites after modification by polydopamine and then analyzed using attenuated total reflectance Fourier transform infrared spectroscopy, field emission scanning electron microscopy, and a contact angle meter. Mechanical properties of the membranes were examined by tensile testing. Proliferation of osteoblastic cell line MC3T3-E1 cultured on the membranes was examined by MTT (3-(4, 5-dimethylthiozole-2-yl)-2, 5-diphenyltetrazolium bromide) and flow cytometric analysis. Effects on cell behavior, including cytotaxis, adhesion, and migration, were further investigated by continuous time-lapse imaging for 8 h. The COL/CHI composites were successfully immobilized onto PLGA surfaces. PLGA mainly contributed the mechanical properties, while COL/CHI played a major role in bioactivity. COL facilitated cell adhesion and spread, but the addition of CHI decreased both. A critical ratio of COL/CHI (2:1) above which the addition of CHI only slightly impacted cell proliferation was found. The results should be useful for combining versatile materials from different origins to construct guided bone regeneration membranes and to further optimize the ratio of COL/CHI composites. © 2012 Wiley Periodicals, Inc. Maxillofacial bone defects present a significant clinical challenge. Although guided bone regeneration (GBR) is widely employed, the therapeutic efficacy of conventional barrier membranes remains limited due to their lack of multifunctional bioactivity. To address this, a novel coaxial electrospun barrier membrane was fabricated, featuring a berberine hydrochloride (BBR)-loaded polyvinyl alcohol (PVA) core and a poly (lactic-co-glycolic acid) (PLGA) shell (denoted as BBR/PVA@PLGA). The membrane exhibited a well-defined nanostructure, suitable hydrophilicity, controllable degradation, favorable mechanical properties, and sustained drug release. In vitro studies confirmed its excellent cytocompatibility with MC3T3-E1, human umbilical vein endothelial cells, and L929 cells and its ability to promote cell migration. Functionally, the membrane significantly enhanced osteogenic differentiation and mineralization in pre-osteoblasts, accompanied by the upregulation of osteogenic gene expression. It also promoted angiogenic activity in endothelial cells, supported by increased expression of pro-angiogenic genes and improved tube formation. Moreover, the membrane served as an effective physical barrier, preventing cellular infiltration. In summary, this multifunctional BBR/PVA@PLGA coaxial electrospun membrane concurrently supports osteogenesis, angiogenesis, and barrier functions in vitro, demonstrating promising potential as a candidate biomaterial for future application in maxillofacial bone defect regeneration.

Hepatocellular carcinoma (HCC) poses a severe threat to public health, with clinical therapy limited by narrow surgical indications, chemotherapeutic side effects, and targeted drug resistance. Herein, we developed Fe 3+ coordination Polydopamine@Poly (lactic-co-glycolic acid)-Curcumin (Fe 3+ -PDA@PLGA-Cur, FePPC) nanoparticles integrating photothermal therapy (PTT), chemodynamic therapy (CDT), sonodynamic therapy (SDT), and curcumin (Cur)'s intrinsic anticancer activity for synergistic HCC treatment. FePPC was fabricated via emulsion solvent evaporation, in-situ dopamine polymerization, and Fe 3+ coordination chelation, with its structure and physicochemical properties characterized by FT-IR, UV-vis, SEM, XPS, and particle size analysis. FePPC exhibited uniform spherical morphology (198.58 nm), favorable stability, 73.60 ± 0.51% encapsulation efficiency, and NIR-triggered temperature-dependent Cur release. Under 808 nm NIR light (2 W/cm 2 , 4 cm), FePPC achieved a 24.4 °C temperature elevation with robust photothermal stability. Catalytic assays confirmed FePPC generated ·OH via Fe 3+ -mediated Fenton-like reactions, 1 O 2 under NIR irradiation, and ·O₂⁻ upon ultrasound stimulation, realizing synergistic reactive oxygen species (ROS) production. In vitro experiments on HepG2 cells showed time-dependent FePPC internalization; the FePPC + PTT + CDT + SDT group reduced cell viability to 53.45%, induced 47.55% apoptosis, elevated intracellular ROS, disrupted mitochondrial membrane potential, downregulated LDH, modulated Bax/Bcl-2 expression, and inhibited cell migration. This study highlights FePPC's potential as a multifunctional nanoplatform for HCC therapy via coordinated chemical-biological interactions.

Sulforaphane (SFN) is a natural isothiocyanate compound with multiple bioactive effects, abundantly found in cruciferous vegetables. SFN and cadmium (Cd) were limited in their application as chemotherapeutic agents due to insufficient cellular uptake, low bioavailability, and high systemic toxicity, respectively. In this study, mPEG-PLGA nanoparticles were used as a carrier to load Cd-γ-PGA conjugates and SFN, enabling favorable drug release under acidic microenvironments with excellent pH responsiveness. The NP-Cd-SFN nanoparticles exhibited a particle size of 102.1 nm, a zeta potential of -14.48 mV, and a PDI value of 0.257. These characteristics contribute to the nanoparticles’ prolonged circulation in the bloodstream and their ability to passively target tumors. Compared to the single-dose groups and the combined Cd + SFN group, the NP-Cd-SFN group significantly reduced the viability of HepG2 cells and increased their apoptosis rate by inducing mitochondrial oxidative stress and promoting cell apoptosis. Overall, the addition of SFN and the encapsulation of mPEG-PLGA enhanced the therapeutic effects of Cd on HepG2 cells.

Since diabetes lead to alterations in bone metabolism with reductions in bone mineral content and delayed bone formation, the most effective method for bone regeneration in diabetes remains to be determined. In this study, type 2 diabetes were successfully induced via a high-fat diet and low-dose streptozotocin intraperitoneal injection. Excess reactive oxygen species (ROS) has been implicated in diabetes mellitus. Overexpression of ROS can lead to oxidative stress and subsequently to H 2 O 2 -mediated impaired proliferation and delayed cellular differentiation. As a result, antioxidant alpha-lipoic acid (ALA)-loaded poly (lactic-co-glycolic acid) (PLGA) microspheres were fabricated using the emulsion solvent evaporation method, and a sustained and controlled release of ALA was observed up to 27 days. It was demonstrated that biodegradable PLGA microspheres loaded with ALA acted as ROS scavengers and partially recover the mesenchymal stem cell proliferation and differentiation. The bone formation of ALA loaded scaffolds in rat cranial bone defects were greater than the prime three-dimensional collagen scaffold. These results suggest the application of ALA loaded PLGA microsphere exhibit good bioactivity and bone forming ability in diabetes.

This study investigated the therapeutic efficiency of monomethoxy polyethylene glycol-poly(lactic-co-glycolic acid) (mPEG-PLGA) co-loaded with syringopicroside and hydroxytyrosol as a drug with effective targeting and loading capacity as well as persistent circulation in vivo. The nanoparticles were prepared using a nanoprecipitation method with mPEG-PLGA as nano-carrier co-loaded with syringopicroside and hydroxytyrosol (SH-NPs). The parameters like in vivo pharmacokinetics, biodistribution in vivo, fluorescence in vivo endomicroscopy, and cellular uptake of SH-NPs were investigated. Results showed that the total encapsulation efficiency was 32.38 ± 2.76 %. Total drug loading was 12.01 ± 0.42 %, particle size was 91.70 ± 2.11 nm, polydispersity index was 0.22 ± 0.01, and zeta potential was −24.5 ± 1.16 mV for the optimized SH-NPs. The nanoparticle morphology was characterized using transmission electron microscopy, which indicated that the particles of SH-NPs were in uniformity within the nanosize range and of spherical core shell morphology. Drug release followed Higuchi kinetics. Compared with syringopicroside and hydroxytyrosol mixture (SH), SH-NPs produced drug concentrations that persisted for a significantly longer time in plasma following second-order kinetics. The nanoparticles moved gradually into the cell, thereby increasing the quantity. ALT, AST, and MDA levels were significantly lower on exposure to SH-NPs than in controls. SH-NPs could inhibit the proliferation of HepG2.2.15 cells and could be taken up by HepG2.2.15 cells. The results confirmed that syringopicroside and hydroxytyrosol can be loaded simultaneously into mPEG-PLGA nanoparticles. Using mPEG-PLGA as nano-carrier, sustained release, high distribution in the liver, and protective effects against hepatic injury were observed in comparison to SH.

Bioactive synthetic scaffolds with 3D porous structure are the most attractive materials among various guided bone regeneration membranes. However, its osteogenic potential is still insufficient. This study was aimed to develop a Calcium surface-anchored Collagen I-PLGA/PCL scaffolds (PP/COL I-pDA-Ca) with enhanced osteogenicity. PP/COL I-pDA-Ca was electrospun from a collagen I-blended PLGA/PCL matrix and then modified by the chelation of Ca-ions via mussel-inspired polydopamine coating. PLGA/PCL, PLGA/PCL-polydopamine, PLGA/PCL-pDA chelated by Ca-ions were used as controls. Osteogenic effects of the scaffolds were examined using MC3T3-E1 cell culture. PP/COL I-pDA-Ca maintained 3D porous architecture with interconnected pores formed by randomly-oriented filamentous fibers and MC3T3-E1 cells cultured on it for 12 h or 24 h were more stretched and spread than those on the controls. PP/COL I-pDA-Ca significantly upregulated α10, α11 and β1 integrin expression after 48 h culture. ALP activity, OCN, OSX, BMP2 and RUNX2 expression of MC3T3-E1 cells cultured on PP/COL I-pDA-Ca scaffolds for 7 and 14 days were substantially enhanced when compared to controls ( p < 0.05). The pDA-based Ca chelation, COL I incorporation and 3D bionic structure promoted cell adhesion and osteogenic differentiation of MC3T3-E1 cells. This novel PP/COL I-pDA-Ca scaffolds is an attractive alternative for guided bone regeneration.

Bone regeneration in compromised environments, such as infected or inflammatory defects, remains a significant clinical challenge. This study addresses the limitations of curcumin (Cur), including poor solubility, low bioavailability, and limited antibacterial activity, by developing a series of curcumin-metal complexes through chelation reactions. These complexes were encapsulated into nanoliposomes and subsequently incorporated into poly(lactic-co-glycolic acid)/polycaprolactone (PLGA/PCL) electrospun scaffolds to construct multifunctional bone regeneration platforms. The fabricated scaffolds exhibited uniform nanofibrous morphology, enhanced mechanical properties, and improved surface hydrophilicity, which collectively promoted cell adhesion and proliferation. Among the complexes, Cur-Cu demonstrated the highest antioxidant activity and superior osteogenic performance, significantly enhancing alkaline phosphatase (ALP) activity and upregulating integrin β1/α1 gene expression in MC3T3-E1 cells. In contrast, Cur-Zn exhibited broad-spectrum antibacterial efficacy against both Escherichia coli and Staphylococcus aureus , while Cur-Ca showed limited functional benefits due to rapid ion release and potential cellular interference. These findings highlight the tailored functionality of Cur-metal complexes. Cur-Cu scaffolds are promising for repairing bone defects under oxidative stress, whereas Cur-Zn scaffolds are effective in preventing implant-associated infections. This work provides a rational strategy for designing microenvironment-responsive biomaterials with integrated antioxidant, osteogenic, and antibacterial properties for advanced bone tissue engineering.

Alveolar bone defects from periodontitis are common clinical challenges in dentistry. Guided bone regeneration (GBR) proves effective, yet current membranes like Bio-Gide, suffer from inadequate mechanical strength, rapid degradation, and no antibacterial activity, limiting clinical performance. Asymmetric bilayer nanofibrous membranes with functional zoning offer a promising solution. Herein, we fabricated an asymmetric bilayer nanofibrous membrane via coaxial electrospinning, consisting of a dense PLGA-GO/PVA-MgCl2 outer layer and a highly porous PLGA-GO/PVA-MgCl2 inner layer. The dense layer prevents rapid soft-tissue cells invasion into the defect area while providing antibacterial effects. The porous layer facilitates osteoblast adhesion and proliferation. Graphene oxide (GO) and Mg2+ exert synergistic action: GO facilitates initial bacterial attachment, protein adsorption, and cell adhesion, while released Mg2+ disrupts bacterial membrane integrity and metabolic pathways. This results in durable bacterial killing and enhanced osteogenic effects compared to either component alone. The GO-enhanced membrane exhibited a Young’s modulus of 84.8 MPa and a matched degradation rate (23% over 2 months), providing mechanical support while maintaining structural integrity. This property combination addresses the fragility issue of Bio-Gide membranes. In vitro studies demonstrate that the dense layer effectively blocked soft-tissue invasion while permitting cell proliferation. The porous layer, with high porosity and sustained Mg2+ release, promoted MC3T3-E1 migration and differentiation. Furthermore, the membrane exhibited excellent antibacterial activity, cytocompatibility, and osteogenic capacity, as evidenced by significantly enhanced bone regeneration in a rat cranial defect model. These findings indicate the great potential of this multifunctional asymmetric bilayer membrane for clinical applications in alveolar bone defect repair.

Poly (lactic-co-glycolic acid) (PLGA) nanoparticles are an ideal paclitaxel (PTX)-carrying system due to its biocompatibility and biodegradability. But it possessed disadvantage of drug burst release. In this research, a layer-by-layer deposition of chitosan (CS) and sodium alginate (ALG) was applied to modify the PLGA nanoparticles. The surface charges and morphology of the PLGA, PLGA/CS and PLGA/CS/ALG particles was measured by capillary electrophoresis and SEM and TEM, respectively. The drug encapsulation and loading efficiency were confirmed by ultraviolet spectrophotometer. The nanoparticles were stable and exhibited controlled drug release performance, with good cytotoxicity to human lung carcinoma cells (HepG 2). Cumulatively, our research suggests that this kind of three-layer nanoparticle with LbL-coated shield has great properties to act as a novel drug-loaded system.

The aim of this study was to fabricate different risperidone (RIS)–loaded poly(lactic-co-glycolic acid) microspheres (PLGA MS) with various viscosity of PLGA polymers and investigate the RIS release profiles. Risperidone-loaded PLGA MS were fabricated via an oil/water–type emulsion solvent evaporation method, using ethyl acetate and benzil alcohol as the dispersed solvents. The PLGA (75:25) with the viscosity of 0.82, 0.93, and 1.10 dL/g was used in the fabrication. The morphology and the degradation of the RIS-PLGA MS were investigated with a scanning electron microscope. The distribution states of RIS in the PLGA MS were studied with differential scanning calorimetry. The residual of ethyl acetate and benzil alcohol in the resulting MS was monitored with gas chromatography. The in vitro release profiles of RIS from PLGA MS were also studied. Scanning electron microscope photographs illustrated that the obtained PLGA MS were monodisperse spheres with an average diameter of 100 μm. Gas chromatography results suggested that no residual ethyl acetate was left in the final RIS-PLGA MS, and the residue amount of benzil alcohol was about 1%. In vitro drug release profiles from the microsphere showed a substantially sigmoidal pattern of negligible initial burst up to 24 hours and minimal release (time lag) for 14 days. After the lag phase, slow release took a place up to 30 days, and then rapid release occurred sharply for about 2 weeks. The RIS release reached equilibrium around day 50. All release profiles showed the similar trend, and no significant difference was observed among these release profiles ( P > .05). This phenomenon indicated that RIS-loaded PLGA MS with the viscosity of 0.82 to 0.93 dL/g showed the optimal release behavior. However, no obvious differences were found among the MS obtained from the viscosity of 0.82 to 1.10 dL/g. These studies provided basis for the quality control in the industrial production of PLGA MS.
