BACKGROUND Neonicotinoids are among the most essential chemical insecticides worldwide because of their high activity against many important pests and wide application. However, their application is limited by their toxicity to honeybees. Therefore, the development of a facile route to fabricate efficient and eco-friendly pesticide formulations is of great significance. RESULTS In this study, clothianidin-loaded zeolitic imidazolate framework-8 (CLO@ZIF-8) nanoparticles were fabricated by a facile one-pot route using zinc nitrate as a Zn 2+ source and characterized by scanning electron microscopy, transmission electron microscopy, X-ray diffraction, thermogravimetric analysis, energy-dispersive spectrometry and Fourier transform infrared spectroscopy. Based on the pH response of ZIF-8, a ‘burst release effect’ was observed for CLO@ZIF-8 at pH 3 and 5 within 12 h, in contrast to the slow and sustainable release at pH 8. CLO@ZIF-8 improved the retention ability of the pesticide liquid and remained 70% control efficacy on Nilaparvata lugens after water rinsed of sprayed CLO@ZIF-8. The pH response of CLO@ZIF-8 allowed it to maintain 43% control efficacy against N. lugens after 10 days of application, which was twice the efficacy of clothianidin solution (SCA). Moreover, CLO@ZIF-8 reduced the acute toxicity to honeybees ( Apis mellifera ) by ≥120-fold compared with SCA. CONCLUSION This study provides new insights into the application of ZIF-8 to neonicotinoids and suggests the need for the development of a biocompatible and eco-friendly pesticide formulation. © 2023 Society of Chemical Industry.

BACKGROUND The complex preparation process and storage instability of nanoformulations hinders their development and commercialization. In this study, nanocapsules loaded with abamectin were prepared by interfacial polymerization at room temperature and ordinary pressure using the monomers of epoxy resin (ER) and diamine. The potential mechanisms of primary amine and tertiary amine in influencing the shell strength of the nanocapsules and the dynamic stability of abamectin nanocapsules (Aba@ER) in the suspension system were systematically researched. RESULTS The tertiary amine catalyzed the self-polymerization of epoxy resin into linear macromolecules with unstable structures. The structural stability of the diamine curing agent with a primary amine group played a key role in enhancing the structural stability of the polymers. The intramolecular structure of the nanocapsule shell formed by isophorondiamine (IPDA) crosslinked epoxy resin has multiple spatial conformations and a rigid saturated six-membered ring. Its structure was stable, and the shell strength was strong. The formulation had stable dynamic changes during storage and maintained excellent biological activity. Compared with emulsifiable concentrate (EC), Aba@ER/IPDA had superior biological activity, and the field efficacy on tomato root-knot nematode was enhanced by approximately 31.28% at 150 days after transplanting. CONCLUSION Aba@ER/IPDA, which has excellent storage stability and simple preparation technology, can provide a nanoplatform with industrial prospects for efficient pesticide delivery. © 2023 Society of Chemical Industry.

BACKGROUND Echinochloa glabrescens Munro ex Hook. f. is one of the main Echinochloa spp. seriously invading Chinese rice fields and has evolved resistance to commonly used herbicides. Previously, an E. glabrescens population (LJ-02) with suspected resistance to the acetyl-CoA carboxylase (ACCase)-inhibiting herbicide metamifop was collected. This study aimed to determine its resistance status to metamifop and investigate the internal molecular mechanisms of resistance. RESULTS Single-dose testing confirmed that the LJ-02 population had evolved resistance to metamifop. Gene sequencing and a relative expression assay of ACCase ruled out target-site based resistance to metamifop in LJ-02. Whole-plant bioassays revealed that, compared with the susceptible population XZ-01, LJ-02 was highly resistant to metamifop and exhibited cross-resistance to fenoxaprop- P -ethyl. Pretreatment with the known glutathione S -transferase (GST) inhibitor, 4-chloro-7-nitrobenzoxadiazole (NBD-Cl), largely reversed the resistance to metamifop by approximately 81%. Liquid chromatography–tandem mass spectrometry analysis indicated that the metabolic rates of one of the major metabolites of metamifop, N -(2-fluorophenyl)-2-hydroxy- N -methylpropionamide (HPFMA), were up to 383-fold faster in LJ-02 plants than in XZ-01 plants. There were higher basal and metamifop-inducible GST activities toward 1-chloro-2,4-dinitrobenzene (CDNB) in LJ-02 than in XZ-01. Six GST genes were metamifop-induced and overexpressed in the resistant LJ-02 population. CONCLUSION This study reports, for the first time, the occurrence of metabolic metamifop resistance in E. glabrescens worldwide. The high-level metamifop resistance in the LJ-02 population may mainly involve specific isoforms of GSTs that endow high catalytic activity and strong substrate specificity. © 2023 Society of Chemical Industry.

Background Thiamethoxam (TMX) is insecticidal, but also can trigger physiological and metabolic reactions of plant cycles. The objective of this work was to evaluate the physiological and metabolic effect of TMX on tea plants and its potential benefits. Results In this study, dose of TMX (0.09, 0.135 and 0.18 kg a.i./ha) were tested. Except for peroxidase (POD) and glutathione S-transferase (GST), chlorophyll, carotenoid, catalase (CAT) and malondialdehyde (MDA) were significantly affected compared with the controls. The CAT activity was increased by 3.38, 1.71, 2.91 times, respectively, under three doses of TMX treatment. The metabolic response between TMX treatment and control groups on the third day was compared using a widely targeted metabolomics. A total of 97 different metabolites were identified, including benzenoids, flavonoids, lipids and lipid-like molecules, organic acids and derivatives, organic nitrogen compounds, organic oxygen compounds, organoheterocyclic compounds, phenylpropanoids and polyketides, and others. Those metabolites were mapped on the perturbed metabolic pathways. The results demonstrated that the most perturbation occurred in flavone and flavonol biosynthesis. The beneficial secondary metabolites luteolin and kaempferol were upregulated 1.46 and 1.31 times respectively, which protect plants from biotic and abiotic stresses. Molecular docking models suggest interactions between TMX and flavonoid 3-O-glucosyltransferase. Conclusion Thiamethoxam spray positively promoted the physiological and metabolic response of tea plants. And this work also provided the useful information of TMX metabolism in tea plants as well as rational application of insecticides. © 2023 Society of Chemical Industry.

Background Pesticides are irreplaceable inputs for protecting crops from pests and improving crop yield and quality. Self-assembly nanotechnology is a promising strategy by which to develop novel nano-formulations for pesticides. Nano-formulations improve the effective utilization of pesticides and reduce risks to the environment because of their eco-friendly preparation, high drug loading, and desirable physicochemical properties. Here, to enhance the utilization efficiency of myclobutanil (MYC) and develop a novel nano-formulation, carrier-free co-assembled nanoparticles (MT NPs) based on MYC and tannic acid (TA) were prepared by noncovalent molecular interactions using a green preparation process without any additives. Results The results showed that the prepared spherical nanoparticles had good stability in neutral and acidic aqueous solutions, low surface tension (40.53 mN m −1 ), high rainfastness, and good maximum retention values on plant leaves. Release of active ingredients from MT NPs could be regulated by altering the molar ratio of subassemblies in the co-assembly and the pH of the environment. Antifungal experiments demonstrated that MT NPs had better activities against Alternaria alternata and Fusarium graminearum [half-maximal effective concentration (EC 50 ) = 6.40 and 77.08 mg/L] compared with free MYC (EC 50 = 11.46 and 124.82 mg/L), TA (EC 50 = 251.19 and 503.81 mg/L), and an MYC + TA mixture (EC 50 = 9.62 and 136.21 mg/L). These results suggested that MYC and TA incorporated in the co-assembled nanoparticles had a synergistic antifungal activity. The results of a genotoxicity assessment indicated that MT NPs could reduce the genotoxicity of MYC to plant cells. Conclusion Co-assembled MT NPs with synergistic antifungal activity have outstanding potential for the management of plant diseases. © 2023 Society of Chemical Industry.

BACKGROUND Bioherbicides are becoming more attractive as safe weed control tools towards sustainable agriculture. Natural products constitute an important source chemicals and chemical leads for discovery and development of novel pesticide target sites. Citrinin is a bioactive compound produced by fungi of the genera Penicillium and Aspergillus . However, its physiological-biochemical mechanism as a phytotoxin remains unclear. RESULTS Citrinin causes visible leaf lesions on Ageratina adenophora similar to those produced by the commercial herbicide bromoxynil. Phytotoxicity bioassay tests using 24 plant species confirmed that citrinin has a broad activity spectrum and therefore has potential as a bioherbicide. Based on chlorophyll fluorescence studies, citrinin mainly blocks PSII electron flow beyond plastoquinone Q A at the acceptor side, resulting in the inactivation of PSII reaction centers. Furthermore, molecular modeling of citrinin docking to the A. adenophora D1 protein suggests that it binds to the plastoquinone Q B site by a hydrogen bond between the O1 hydroxy oxygen atom of citrinin and the histidine 215 of the D1 protein, the same way as classical phenolic PSII herbicides do. Finally, 32 new citrinin derivatives were designed and sorted according to free energies on the basis of the molecular model of an interaction between the citrinin molecule and the D1 protein. Five of the modeled compounds had much higher ligand binding affinity within the D1 protein compared with lead compound citrinin. CONCLUSION Citrinin is a novel natural PSII inhibitor that has the potential to be developed into a bioherbicide or utilized as a lead compound for discovery of new derivatives with high herbicidal potency. © 2023 Society of Chemical Industry.

BACKGROUND Pesticides are indispensable in agriculture and can effectively improve the yields and quality of crops. Due to their weak water solubility, most pesticides need to be dissolved by adding solubilizing adjuvants. In this work, based on molecular recognition of the macrocyclic host, we developed a novel supramolecular adjuvant, called sulfonated azocalix[4]arene (SAC4A), which significantly improves the water solubility of pesticides. RESULTS SAC4A presents multiple advantages, including high water solubility, strong binding affinity, universality, and simple preparation. SAC4A showed an average binding constant value of 1.66 × 10 5 M −1 for 25 pesticides. Phase solubility results indicated that SAC4A increased the water solubility of pesticides by 80–1310 times. The herbicidal, fungicidal, and insecticidal activities of supramolecular formulations were found to be superior to those of technical pesticides, and the herbicidal effects were even better than those of commercial formulations. CONCLUSION Overall results revealed the potential of SAC4A to improve the solubility and effectiveness of pesticides, providing a new development idea for the application of adjuvants in agriculture. © 2023 Society of Chemical Industry.

Background Efficient deposition of high-speed droplets on superhydrophobic leaf surfaces remains an important challenge. For anisotropic wired superhydrophobic leaf surfaces, the splashing phenomenon is especially serious because it leads to the low effective utilization of pesticides by biological targets. The lost pesticides cause serious ecological environment pollution, therefore there is an urgent need to develop a green and sustainable cost-effective strategy to achieve efficient deposition of high-speed droplets on anisotropic superhydrophobic leaf surfaces at low dosage. Results One type of green pseudogemini surfactant is constructed based on fatty acids and hexamethylenediamine by electrostatic interaction to control the splashing and spreading of high-speed droplets on superhydrophobic surfaces. The formed surfactant can not only achieve complete inhibition of the bouncing of droplets, but also promote rapid spreading on superhydrophobic leaf surfaces at very low usage. The efficient deposition and superspreading phenomenon are attributed to the rapid migration and adsorption of the surfactant from the dynamic spherical micelles at the newly formed solid–liquid interface, the network-like aggregated spherical micelles, and the Marangoni effect caused by the surface tension gradient. Moreover, the surfactant shows an excellent synergistic effect with herbicides to control weeds by inhibiting droplet splashing. Conclusion This work provides a simpler, more effective and sustainable approach to utilize aggregated spherical micelles rather than conventional vesicles or wormlike micelles to improve the droplet deposition on superhydrophobic leaf surfaces and reduce the impact of surfactants and pesticides on the ecological environment. © 2023 Society of Chemical Industry.

BACKGROUND Mythimna separata is a notorious pest causing crop damages at the larval stage. Gaining insight into larval olfaction mechanisms would provide knowledge for olfaction-based management of M. separata larvae. RESULTS In the present research, ( Z )-11-hexadecenal ( Z 11-16: Ald), a major component of M. separata sex pheromone, was found to attract early-instar larvae of M. separata in a food context. Using a fluorescent binding assay, we found that M. separata general odorant binding protein 2 (MsepGOBP2) exhibited high binding affinity to Z 11-16: Ald. Further, silencing of MsepGOBP2 resulted in a sharp reduction of the response to Z 11-16: Ald, which could not be mitigated by increasing the concentration of Z 11-16: Ald. Additionally, we employed molecular dynamics-based approaches to unravel the interaction details between MsepGOBP2 and Z 11-16: Ald, specifically the binding of Z 11-16: Ald to MsepGOBP2. CONCLUSION Z 11-16: Ald is attractive to early-instar larvae of M. separata , and MsepGOBP2 is identified to be indispensable in the larval detection of Z 11-16: Ald. These results could aid in the development of olfaction-based methods for controlling M. separata in the larval stage. © 2023 Society of Chemical Industry.

BACKGROUND The evolution of insect resistance to pesticides poses a continuing threat to sustainable pest management. While much is known about the molecular mechanisms that confer resistance in model insects and few agricultural pests, far less is known about fruit pests. Field-evolved resistance to synthetic insecticides such as lambda -cyhalothrin has been widely documented in Cydia pomonella , a major invasive pest of pome fruit worldwide, and the increased production of cytochrome P450 monooxygenases (P450s) has been linked to resistance in field-evolved resistant populations. However, the underlying molecular mechanisms of P450-mediated insecticide resistance remain largely unknown . RESULTS Here we found that functional redundancy and preference of metabolism by P450s genes in the CYP9A subfamily confer resistance to lambda -cyhalothrin in Cydia pomonella . A total of four CYP9A genes, including CYP9A61 , CYP9A120 , CYP9A121 , and CYP9A122 , were identified from Cydia pomonella . Among these, CYP9A120 , CYP9A121 , and CYP9A122 were predominantly expressed in the midgut of larvae. The expression levels of these P450 genes were significantly induced by a lethal dose that would kill 10% (LD 10 ) of lambda -cyhalothrin and were overexpressed in a field-evolved lambda -cyhalothrin resistant population. Knockdown of CYP9A120 and CYP9A121 by RNA-mediated interference (RNAi) increased the susceptibility of larvae to lambda -cyhalothrin. In vitro assays demonstrated that recombinant P450s expressed in Sf9 cells can metabolize lambda -cyhalothrin, but with functional redundancy and divergence through regioselectivity of metabolism. CYP9A121 preferred to convert lambda -cyhalothrin to 2′-hydroxy- lambda -cyhalothrin, whereas CYP9A122 only generated 4′-hydroxy metabolite of lambda -cyhalothrin. Although possesses a relatively low metabolic capability, CYP9A120 balanced catalytic competence to generate both 2′- and 4′-metabolites. CONCLUSION Collectively, these results reveal that metabolic functional redundancy of three members of the CYP9A subfamily leads to P450-mediated lambda -cyhalothrin resistance in Cydia pomonella , thus representing a potential adaptive evolutionary strategy during its worldwide expansion. © 2022 Society of Chemical Industry.

BACKGROUND Succinate dehydrogenase inhibitor (SDHI) fungicides are an important class of agricultural fungicides with the advantages of high efficiency and a broad bactericidal spectrum. To pursue novel SDHIs, a series of N -substituted dithiin tetracarboximide derivatives were designed, synthesized, and characterized by 1 H NMR, 13 C NMR, and high resolution mass spectrum (HRMS). RESULTS These engineered compounds displayed potent fungicidal activity against phytopathogens, including Sclerotinia sclerotiorum , Botrytis cinerea , and Rhizoctonia solani , comparable with that of the commercial SDHI fungicide boscalid . In particular, compound 18 stood out with prominent activity against S. sclerotiorum with a half-maximal effective concentration (EC 50 ) value of 1.37 μg ml −1 . Compound 1 exhibited the most potent antifungal activity against B. cinerea with EC 50 values of 5.02 μg ml −1 . As for R. solani , 12 and 13 exhibited remarkably inhibitory activity with EC 50 values of 4.26 and 5.76 μg ml −1 , respectively. In the succinate dehydrogenase (SDH) inhibition assay, 13 presented significant inhibitory activity with a half-maximal inhibitory concentration (IC 50 ) value of 15.3 μ m , which was approximately equivalent to that of boscalid (14.2 μ m ). Furthermore, molecular docking studies revealed that 13 could anchor in the binding site of SDH. CONCLUSION Taken together, results suggested that the dithiin tetracarboximide scaffold possessed a huge potential to be developed as novel fungicides and SDHIs. © 2023 Society of Chemical Industry.

BACKGROUND Insect RDL (resistant to dieldrin) receptors are essential pentameric ligand-gated chloride channels that mediate the neuroinhibitory effect of GABA, the chief inhibitory neurotransmitter in the central nervous system. These receptors serve as primary targets for various insecticides, including noncompetitive antagonists (NCAs) such as cyclodiene organochlorines and phenylpyrazoles, as well as allosteric modulators like meta-diamides and isoxazolines. This study focuses on a newly discovered A2′N mutation within the RDL receptors, identified in fipronil-resistant planthoppers. Despite in vitro electrophysiological studies have proposed its role in conferring target-site resistance, in vivo genetic functional validation of this mutation remains unexplored. RESULTS Our research employed toxicity bioassays, assessing various Rdl genotypes against a spectrum of insecticides, including fipronil, α-endosulfan, broflanilide, and isocycloseram. Results revealed distinct resistance profiles for A2′N and A2′S mutants, indicating different binding interactions of RDL receptors with NCAs. Significantly, the A2′N heterozygote showed substantial resistance to fipronil, despite its homozygous lethality. Molecular modeling and docking simulations further supported these findings, highlighting unique binding poses for fipronil and α-endosulfan. CONCLUSION This study confirmed that A2′N mutation of the RDL GABA receptor confer high resistance to fipronil in vivo . The observed resistance in A2′N mutants is likely attributable to a steric hindrance mechanism, wherein the introduction of larger side chains hampers fipronil binding, even in a heterozygous state. © 2023 Society of Chemical Industry.
