Engineering hierarchical Ag nanoparticles-coated periodic nanoripples for enhanced SERS performance

Duoduo Yang,Kaishen Chen,Yiping Cheng,Zhiyu Liu,Zuolian Liu,Chengyun Zhang

Surface-enhanced Raman scattering (SERS) is renowned for its ultra-sensitivity, rapidity, and non-destructive nature, making it invaluable in environmental monitoring, food safety testing, and biomedical diagnostics. Efficient fabrication of low-cost, contamination-free, reusable, and functional SERS substrates is highly desirable. This work introduces a simple and cost-effective method for fabricating SERS substrates by combining femtosecond (fs) laser-induced periodic surface structuring (LIPSS) with electrochemical deposition of silver nanoparticles (Ag NPs) on stainless steel mesh (SSM). The highly uniform and periodic nanostructures can be induced by fs laser on the surface of SSM, acting as substrate and enabling the electrochemically deposited Ag NPs to grow into three dimensional (3D) nanostructures. This significantly increases the number of "hot spots" and enhances SERS signal intensity due to the innovative hierarchical micro/nanostructures and concentration enriched effect. The performance of the substrates was evaluated using Rhodamine 6 G (R6G) and malachite green (MG) as probe molecules, achieving detection limits (LOD) of 1.35 × 10⁻¹ ² M and 1.28 × 10⁻¹ ⁰ M, with enhancement factors (EF) of 3.58 × 10⁹ and 1.88 × 10⁷, respectively. The substrates exhibit high reproducibility and uniformity, with a relative standard deviation (RSD) of 2.742 %. Moreover, stability tests demonstrated excellent mechanical and chemical robustness, as the substrate maintained stable SERS performance after ultrasonic treatment (40 kHz, 30 s) and exposure to acidic and alkaline environments (pH 3–11). Numerical simulation results demonstrate the effectiveness of the nanostructures in enhancing SERS signal. This combined method simplifies fabrication and enables scalable production, proving promising for advanced molecular detection applications.

Exploration of cucurbituril-mediated SERS plasmonic nanoarrays with sub-nanometer gaps

Teng Yuanjie,Li Xin,Chen Yingxin,Zhong Yi,Xu Pei,Shan Shengyan,Gunasekaran Sundaram

The uneven distribution of hotspots and the challenges associated with precise analyte localization within these hotspots present significant hurdles in the field of surface-enhanced Raman scattering (SERS). Here, at the water–oil interface, gold nanoparticles (AuNPs) interconnected by cucurbiturils[8] (CB[8]) with sub-nanometer gaps (AuNPs:CB[8]) were organized into plasmonic arrays. This arrangement was engineered to generate highly efficient hotspots. The CB[8] molecules, serving a dual role, not only facilitated the assembly of AuNPs with sub-nanometer (~ 1 nm) gaps to create intense plasmonic hotspots but also acted as molecular traps, enabling the precise localization of molecules within these hotspots. By comparing the enhancement effect of probe molecule on Au nanofilm, AuNPs:CB[8] colloids, and AuNPs:CB[8] nanofilm, it was found that the SERS intensity of the E1 characteristic peak in AuNPs:CB[8] nanofilm is five times higher than that on Au nanofilm, and more than 10 4 times higher than that of AuNPs:CB[8] colloids. The gaps are also accessible to different electronegativite molecules, such as estrone, p -aminoazobenzene, or methylene blue, which are captured at the plasmonic hotspots by the interaction of CB[8]. The method was employed for the practical detection of artificial antioxidant butylated hydroxyanisole (BHA), which has a weak Raman scattering cross-section, by coupling it with a reaction to enhance its SERS effect. The detection limit of BHA in soybean oil sample is 5.89 × 10 −8  mol/L, with the recovery range 85.1–115%. In conclusion, this hot-spot design and molecular capture approach will offer a highly effective method for detecting weak Raman scattering cross-section molecules and holds great promise for practical applications in the future. Fano enhancement of SERS for rapid early diagnosis of colorectal cancer

Tianxun Gong,Zhenjiang Wei,Libin Huang,Yan Hong,Yuan Li,Ke-ling Chen,Wen Huang,Xiaojing Zhong,Jinzhao He,Ming-Yi Lee,En-Chi Chang,Kien Voon Kong,Xiaosheng Zhang,Zongguang Zhou

Patients benefit greatly from early detection of colorectal cancer, but present diagnostic procedures have high costs, low sensitivity, and low specificity. However, it is still difficult to develop a strategy that can effectively detect cancer early using high-throughput blood analysis. Fano resonance-boosted SERS platform label-free serum creates an effective diagnostic system at the point of care. We obtained 220 high-quality SERS serum spectral datasets from 88 healthy volunteers and 132 patients with colorectal cancer. The biomarker detected in serum was further evaluated using 100 colorectal cancer tissues and adjacent normal intestinal tissues collected from West China Biobanks, West China Hospital, Sichuan University. The results showed that in 97 out of 100 paired samples, the biomarkers were successfully detected using the SERS platform. This demonstrates that Fano resonance-based SERS is highly effective for diagnosing colorectal cancer.

Fabrication of complexed nanostructure using AAO template for ultrasensitive SERS detection

Jun Dong,Chenlu Li,Yan Wang,Yimeng Fan,Qingyan Han,Wei Gao,Yongkai Wang,Kaili Ren,Jianxia Qi,Enjie He

In the study of surface-enhanced Raman scattering (SERS) processes, a simple and fast approach is needed to ensure the large-scale preparation of SERS substrates. This article uses anodic aluminum oxide (AAO) as a template to assemble gold nanoparticles (Au NPs) into an ordered array. By changing the pore size of AAO and silanizing the pores, the number and density of Au NPs entering the pores through liquid–liquid two-phase self-assembly (LLSA) can be effectively regulated. Using Rh6G (Rhodamine 6G) and CV (Crystal Violet) molecules as probe molecules, substrate sensitivity was evaluated with an enhancement factor of up to 6.34 × 10 7 . In addition, the uniformity of the substrate is good, with a relative standard deviation (RSD) of 9.94%, and the logarithmic concentration and the Raman signal presented significant linear correlations R 2 was 0.997 and 0.985, respectively. The detection limit of the substrate for APM (aspartame) as a solvent is as low as 0.0078 g/L. Finally, the substrate was subjected to high sensitivity testing on two types of beverages containing APM sold, proving the practicality of the substrate. It is expected to achieve simple and rapid detection in food additive trace detection in the future.

Hydrogel-Coated SERS Microneedles for Drug Monitoring in Dermal Interstitial Fluid

Yan Li,Yunqing Wang,Rongchao Mei,Bingqian Lv,Xizhen Zhao,Liyan Bi,Hui Xu,Lingxin Chen

In vivo drug monitoring is crucial for evaluating the effectiveness and safety of drug treatment. Blood sampling and analysis is the current gold standard but needs professional skills and cannot meet the requirements of point-of-care testing. Dermal interstitial fluid (ISF) showed great potential to replace blood for in vivo drug monitoring; however, the detection was challenging, and the drug distribution behavior in ISF was still unclear until now. In this study, we proposed surface-enhanced Raman spectroscopy (SERS) microneedles (MNs) for the painless and real-time analysis of drugs in ISF after intravenous injection. Using methylene blue (MB) and mitoxantrone (MTO) as model drugs, the innovative core–satellite structured Au@Ag SERS substrate, hydrogel coating over the MNs, rendered sensitive and quantitative drug detection in ISF of mice within 10 min. Based on this technique, the pharmacokinetics of the two drugs in ISF was investigated and compared with those in blood, where the drugs were analyzed via liquid chromatography–mass spectrometry. It was found that the MB concentration in ISF and blood was comparable, whereas the concentration of MTO in ISF was 2–3 orders of magnitude lower than in blood. This work proposed an efficient tool for ISF drug monitoring. More importantly, it experimentally proved that the penetration ratio of blood to ISF was drug-dependent, providing insightful information into the potential of ISF as a blood alternative for in vivo drug detection.

Hypersensitive detection of CYFRA21-1 by SERS dual antibody sandwich method

Song Ge,Mingxin Wang,Shunhua Zhu,Han Wu,Jing Li,Dongsheng Liu,Qingli Huang,Shibao Li,Xiaojing Sun

Purpose As an important reference index for the early diagnosis of non-small cell lung cancer (NSCLC), CYFRA21-1 still lacks the detection of low equipment cost, wide linear range and high sensitivity. Surface-enhanced Raman scattering (SERS) is a vibration spectrum technology based on the surface plasma of precious metal nanoparticles, which has been effectively applied to the detection of tumor markers. The combination of SERS technology and sensors has great potential in the ultrasensitive detection of tumor markers. The purpose of this study was to develop a sensitive method using SERS to quantitatively detect CYFRA21-1 for early diagnosis of NSCLC. Methods A double-antibody sandwich immunoassay based on SERS was designed and tested to implement the ultrasensitive detection of CYFRA21-1 in the serum of NSCLC patients. Results Gold @Ag nanorods (Au @Ag NRs) with higher Raman signals were prepared and used as probes, while magnetic graphene oxide was used as a magnetic substrate. The immunized probe, immune substrate and CYFRA21-1 standard substance in the buffer system formed a double-antibody sandwich structure. The standard curve displayed a liner range from 1pg mL −1 to 10 ng mL −1 , and the detection limit (LOD) is 0.8943pg mL −1 . The Raman intensity exhibited a wide linear relationship with the logarithm of CYFRA21-1 concentration. Conclusion Our study successfully established a double-antibody sandwich immunoassay based on SERS. This method demonstrated high specificity and sensitivity for detecting CYFRA21-1 protein content in serum. It has the potential to be applied for early detection of lung cancer biomarkers.

A Single-Cell Metabolic Profiling Characterizes Human Aging via SlipChip-SERS

Fugang Liu,Jiaqing Liu,Yang Luo,Siyi Wu,Xu Liu,Haoran Chen,Zhewen Luo,Haitao Yuan,Feng Shen,Fangfang Zhu,Jian Ye

Metabolic dysregulation is a key driver of cellular senescence, contributing to the progression of systemic aging. The heterogeneity of senescent cells and their metabolic shifts are complex and unexplored. A microfluidic SlipChip integrated with surface-enhanced Raman spectroscopy (SERS), termed SlipChip-SERS, is developed for single-cell metabolism analysis. This SlipChip-SERS enables compartmentalization of single cells, parallel delivery of saponin and nanoparticles to release intracellular metabolites and to realize SERS detection with simple slipping operations. Analysis of different cancer cell lines using SlipChip-SERS demonstrated its capability for sensitive and multiplexed metabolic profiling of individual cells. When applied to human primary fibroblasts of different ages, it identified 12 differential metabolites, with spermine validated as a potent inducer of cellular senescence. Prolonged exposure to spermine can induce a classic senescence phenotype, such as increased senescence-associated β-glactosidase activity, elevated expression of senescence-related genes and reduced LMNB1 levels. Additionally, the senescence-inducing capacity of spermine in HUVECs and WRL-68 cells is confirmed, and exogenous spermine treatment increased the accumulation and release of H 2 O 2 . Overall, a novel SlipChip-SERS system is developed for single-cell metabolic analysis, revealing spermine as a potential inducer of senescence across multiple cell types, which may offer new strategies for addressing ageing and ageing-related diseases.

Size-Resolved SERS Detection of Trace Polystyrene Nanoplastics via Selective Electrosorption

Tong Wang,Shu Li,Run Mu,Zhengwei Lu,Jie Su,Jing Chen,Jinhua Zhan

Microplastics and nanoplastics are emerging contaminants that pose a threat to the environment and human. Spectroscopic technologies are advantageous in analyzing nanoplastics, but it is challenging to selectively detect nanoplastics with different size thresholds. In this work, the hyphenated method of electrosorption and surface-enhanced Raman spectroscopy (ES-SERS) was developed for the simple, rapid, and size-resolved analysis of trace polystyrene (PS) nanoplastics from 20 to 300 nm. A rough silver was used as both the working electrode for electrosorption and the substrate for the SERS response. By applying a positive electric potential to the rough silver, the PS nanoplastics accelerated toward the silver surface and were adsorbed tightly at the SERS “hot spot” inside the rough silver nanostructure. The proposed ES-SERS method achieved a detection limit of 100 ng/L for 300 and 100 nm PS, 50 ng/L for 50 nm PS, and 30 ng/L for 20 nm PS nanoplastics. It is worth noting that smaller nanoplastics typically exhibit larger analytical enhancement factor values in ES-SERS. According to the difference in electromigration behavior of PS in various sizes, PS nanoplastics under a certain size can be selectively enriched and detected by controlling the electrosorption time. The ES-SERS method was successfully demonstrated for detecting nanoplastics released from the lids of disposable beverage cups. This work opens up new possibilities for size-resolved analysis of nanoplastics.

Selective SERS Sensing of R6G Molecules Using MoS2 Nanoflowers under Pressure

Hongyan Yu,Yongxue Chen,Zhenyu Wen,Rensheng Wang,Sisi Jia,Wenjie Zhu,Yanping Song,Huanhuan Sun,Bingbing Liu

Pressure-induced surface-enhanced Raman spectroscopy (PI-SERS) has garnered significant attention as a subfield of SERS detection due to its capacity to regulate the band gap between molecules and substrates through pressure modulation. Currently, SERS detection primarily focuses on single molecules at atmospheric pressure with limited investigations conducted under high pressure conditions. Herein, we employed rose-shaped MoS2 nanoflowers as the SERS substrate and realized selective PI-SERS enhancement of R6G molecules in the binary (MV+R6G) and ternary (MV+R6G+RhB) systems. The MoS2 demonstrated an exceptionally low SERS detection limit of 5 × 10–6 M in binary and ternary systems with equimolar amounts of molecules. High-pressure experimental results indicate that MoS2 displays selective enhancement for R6G molecules, as evidenced by the comparison of the PI-SERS peak intensity ratio between MoS2 and the probe molecules. The proposed enhancement mechanism in binary and ternary SERS systems under high pressure involves pressure-induced changes in both the band structures of the MoS2 substrate and molecules, thereby influencing their charge transfer dynamics. Consequently, this approach holds great promise for practical applications in complex SERS systems operating under extreme conditions.

Sensitive SERS assay for L-cysteine based on functionalized silver nanoparticles

Yaxian Chen,Huiting Wang,Jie Zhou,Dongxue Lin,Ling Zhang,Zhiqiang Xing,Qian Zhang,Lixin Xia

L-cysteine, an indispensable amino acid present in natural proteins, plays pivotal roles in various biological processes. Consequently, precise and selective monitoring of its concentrations is imperative. Herein, we propose a Surface-enhanced Raman Scattering (SERS) sensor for detecting L-cysteine based on the anti-aggregation of 4-mercaptobenzoic acid (4-MBA) and histidine (His) functionalized silver nanoparticles (Ag NPs). The presence of Hg 2+ ions can induce the aggregation of Ag NPs@His@4-MBA due to the unique nanostructures of Ag NPs@His@4-MBA, resulting in a robust SERS intensity of 4-MBA. However, in the presence of L-cysteine, the stronger affinity between L-cysteine and Hg 2+ reduces the concentration of free Hg 2+ , causing the dispersion of the aggregated functionalized Ag NPs and the reduction of the SERS signal intensity of 4-MBA. The developed SERS platform demonstrates excellent performance with a low detection limit of 5 nM (S/N = 3) and linear detection capabilities within the range of 0.01–100 μM for L-cysteine. Additionally, the method was successfully employed for the determination of L-cysteine in spiked serum samples, yielding recoveries ranging from 95.0 % to 108.1 % with relative standard deviations of less than 3.3 %. This study not only presents a novel approach for fabricating highly sensitive and specific SERS biosensors for biomolecule detection but also offers a significant strategy for the development and construction of SERS substrates using anti-aggregation design.

SERS detection of uranyl based on MOF-coated gold nanooctahedron hybrid

Yuan Chao,Ge Hongwei,Cao Benmei,Wang Suhua

The ability to sensitively and quantitatively detect uranyl in complex samples plays a vital role in environmental monitoring. In this work, an MOF-coated gold (Au) nanohybrid was synthesized for uranyl detection by surface-enhanced Raman scattering (SERS) technology. The MOF shell not only prevents the Au nanoparticles from rapid aggregation, but also effectively enhances the Raman signal of uranyl. A detection limit of as low as 0.5 μM could be achieved in solution, which could be comparable to the previously reported ones from SERS-based approaches. Moreover, the prepared SERS-active substrate was also applied to uranyl detection in real samples. Graphical Abstract

Chen Kun,Ma Chaoqun,Chen Guoqing,Yang Taiqun,Gao Hui,Li Lei,Hu Anqi,Cao Jun,Zheng Chenkai,Ma Longyao,Chen Zehao

The detection of pefloxacin (PEF) in milk is of significant importance for food safety. Traditional methods for detecting PEF often rely on techniques such as high-performance liquid chromatography (HPLC), which are complex and time-consuming. This article introduces a surface-enhanced Raman scattering (SERS) substrate based on a composite of covalent organic frameworks (COFs) and silver nanoparticles (AgNPs) for the rapid detection of PEF in milk. After testing PEF in both water and milk, the detection limits were calculated to be 6.31 μg/L for water and 82 μg/L for milk, with correlation coefficients of 0.991 and 0.998, respectively. Furthermore, the average recovery rate of PEF in milk ranged from 96.25% to 106.17%, with a relative standard deviation (RSD) of 4.32% to 8.88%. This work offers a simple and rapid method for detecting PEF in milk. Graphical
