Porous, environment-friendly chitosan-ZIF composite fibers for dye adsorption

Xuebin Hou,Wei Chen,Meihong Fan,Chen Shi

Chitosan (CS) is regarded as a promising sorbent due to its biodegradability, wide availability, easy processing, and multiple active groups. In this work, CS-based porous fibers (ZIF-n@CS) consisting of CS and zeolitic imidazolate frameworks (ZIFs) were fabricated through wet spinning and self-assembly methods. This combination effectively utilizes the dual advantages of CS and ZIFs, which effectively remove dyes from water and exhibit excellent recyclability. In addition, the adsorption mechanism of the adsorbent was explored through the analysis of adsorption kinetics, thermodynamics, isotherms, and chemical characterization after adsorption. This work provides an operable and universal strategy for the efficient adsorption of dyes, promoting the further promotion and application of MOFs and CS materials.

Preparation and characterization of chitosan/polyvinyl alcohol antibacterial sponge materials

Xue Chen,Jing Xu,Alidha Gafur,Baoyu Chen,Yibo Han,Liyuan Zhang,Lingwen Kong,Guixue Wang,Zhiyi Ye

This study utilized the freeze-drying method to create a chitosan (CS) and polyvinyl alcohol (PVA) sponge. To enhance its antibacterial properties, curcumin and nano silver (Cur@Ag) were added for synergistic antibacterial. After adding curcumin and nano silver, the mechanical properties of the composite sponge dressing (CS-PVA-Cur@Ag) were improved. The porosity of the composite sponge dressing was closed to 80%, which was helpful for drug release, and it had good water absorption and water retention rate. The nano silver diameter was 50–80 nm, which was optimal for killing bacteria. Antibacterial tests used Escherichia coli and Staphylococcus aureus demonstrated that little nano silver was required to eliminate bacteria. Finally, in the rat full-thickness skin wound model, the composite sponge dressing can promote wound healing in a short time. In summary, CS-PVA-Cur@Ag wound dressing could protect from bacterial infection and accelerate wound healing. Thus, it had high potential application value for wound dressing.

Self-Oxidated Hydrophilic Chitosan Fibrous Mats for Fatal Hemorrhage Control

Jinzhi Shi,Kai Shi,Qi Dong,Junfeng Yang,Yingshan Zhou,Peng Ma,Sha She,Fan Yang,Zuojiong Gong

Enriching erythrocytes and platelets in seconds and providing a fast seal in bleeding sites is vital to fatal hemorrhage control. Herein, hydrophilic chitosan fibrous mats (CECS-D mats) are fabricated by introducing hydrophilic carboxyethyl groups and subsequent catechol groups onto chitosan fibers. Due to strong hydrophilicity, CECS-D mats exhibit rapid liquid-absorption capacity, especially instantaneous absorptivity to the rabbit blood, which can achieve erythrocyte and platelet aggregations quickly by concentrating blood, thus promoting the formation of blood clots. Furthermore, the mats are self-oxidated to form quinone-amine adducts or quinone multimers by adjusting pH conditions, which not only provides tissue adhesion but also induces erythrocyte aggregation and platelet adhesion, further enhancing the seal and triggering quick closure to achieve fast hemostasis. Therefore, the mats reveal superior hemostatic performance in rabbit liver and spleen models over CECS mats and gauze. Especially in the fatal femoral artery injury model of rabbits, the mats reduce the blood loss by ∼75% and shortened the bleeding time by ∼50% compared with CECS mats, which have been reported to have the same hemostatic effect as commercialized Celox products in a swine femoral artery injury model. Besides, the mats are cytocompatible and degradable as well as antibacterial. This chitosan mat is a promising hemostatic material for fatal hemorrhage control.

Rhein-loaded chitosan nanoparticles for treatment of MRSA-infected wound

Shuang Cai,Linlin Wang,Xilong Cui,Xinshu Zou,Sidi Zheng,Fanhui Liu,Chun Shi,Yanhua Li,Zhiyun Zhang

The multi-drug resistance of methicillin-resistant Staphylococcus aureus (MRSA) and complex wound microenvironment challenge the repair of MRSA infected wound. Herein, in this study, α-tocopherol modified glycol chitosan (TG) nanoparticles encapsulated with phytochemical rhein (Rhein@TG NPs) were prepared for comprehensive anti-infection and promotion of MRSA infected wound healing. Rhein@TG NPs could not only specifically release rhein in the infection site in response to low pH and lipase of infectious microenvironment, but also up-regulated M1 macrophage polarization in the infection stage, thus achieving synergistically bacterial elimination with low possibility of developing resistance. Additionally, the NPs reduced the levels of pro-inflammatory factors in the post-infection stage, scavenged the ROS, promoted cell migration and angiogenesis, which significantly improved the microenvironment of infected wound healing. Therefore, this antibiotic-free NPs enabling anti-infection and promotion of wound healing provides a new and long-term strategy for the treatment of MRSA infected wound.

Temperature-sensitive injectable chitosan-based hydrogel for endoscopic submucosal dissection

Shourui Liu,Ruibao Ju,Zhenguo Zhang,Zhen Jiang,Jingzhao Cui,Wanshun Liu,Baoqin Han,Shuo Wang

Endoscopic submucosal dissection (ESD) is an effective treatment for polyps and early gastrointestinal cancers, but requires a high level of operator skill. Injecting submucosal materials (SIM) helps create a fluid cushion between the mucosal and muscular layers, making the procedure easier and reducing associated risks. However, SIMs commonly used in current clinical practice tend to spread quickly and fail to provide long-lasting submucosal fluid cushions (SFC). Thus, there is a critical need for a material that is easy to inject while also maintaining a durable barrier. We prepared succinylated hydroxybutyl chitosan (HBC-SA) by adding succinic anhydride (SA) to hydroxybutyl chitosan (HBC). The hydrogel had excellent temperature-sensitive properties and was able to be injected via an endoscopic injection needle even after gel formation. In vitro and in vivo studies showed that it has satisfactory biocompatibility. Functional experiments showed that the submucosal lifting properties of this hydrogel were significantly better than that of normal saline (NS) and sodium hyaluronate (SH), two commonly used clinical materials. In addition, the hydrogel possessed excellent hemostatic properties. Based on these results, HBC-SA is a promising candidate for submucosal injection during ESD.

Protonated-chitosan sponge with procoagulation activity for hemostasis in coagulopathy

Zhenhua Huang,Dong Zhang,Laiqiang Tong,Fan Gao,Shaozan Zhang,Xinqing Wang,Yina Xie,Fangping Chen,Changsheng Liu

Hemostatic materials are essential for managing acute bleeding in medical settings. Chitosan (CS) shows promise in hemostasis but its underlying mechanism remains incompletely understood. We unexpectedly discovered that certain protonated-chitosan (PCS) rapidly assembled plasma proteins to form protein membrane (PM) upon contact with platelet-poor plasma (PPP). We hypothesized that the novel observation was intricately related to the procoagulant effect of chitosan. Herein, the study aimed to elucidate the conditions necessary and mechanism for PM formation, identify the proteins within the PM and PCS's procoagulant action at the molecule levels. We confirmed that the amount of –NH 3 + groups (>4.9 mmol/g) on PCS molecules played a crucial role in promoting coagulation. The –NH 3 + group interacted with blood's multiple active components to exert hemostatic effects: assembling plasma proteins including coagulation factors such as FII, FV, FX, activating blood cells and promoting the secretion of coagulation-related substances (FV, ADP, etc) by platelets. Notably, the hemostatic mechanism can be extended to protonated-chitosan derivatives like quaternized, alkylated, and catechol-chitosan. In the blood clotting index (BCI) experiment, compared to other groups, PCS95 achieved the lowest BCI value (∼6 %) within 30 s. Protonated-chitosan exhibited excellent biocompatibility and antibacterial properties, with PCS95 demonstrating inhibition effectiveness of over 95 % against Escherichia coli ( E.coil ) and Staphylococcus aureus ( S. aureus ). Moreover, PCS performed enhanced hemostatic effectiveness over chitosan-based commercially agents (Celox™ and ChitoGauze®XR) in diverse bleeding models. In particular, PCS95 reduced bleeding time by 70 % in rabbit models of coagulopathy. Overall, this study investigated the coagulation mechanism of materials at the molecular level, paving the way for innovative approaches in designing new hemostatic materials.

Multifunctional chitosan-lignin-containing nanocellulose composite films for food packing

Xiao-Ping Zheng,Pei-Ru Zhou,Jia-Tong Chen,Yu Chai,Yu-Cang Zhang,Yan-Zhen Zheng

Maximizing the valorization of renewable resources to develop bio-composite materials is crucial for advancing environmental sustainability and ensuring food chain security. In this study, lignin-containing nanocelluloses (LCNs), derived from passion fruit husk using a combination of deep eutectic solvent (DES) and ultra-high-pressure homogenization (UHPH), were incorporated as nanocellulose into a chitosan matrix to produce innovative bioactive packaging films. The results indicated that the inclusion of LCNs improved the water resistance performance and the antioxidant and antimicrobial properties of the films. The tensile strength of the composite films significantly increased, while the elongation at break decreased. Moreover, the presence of LCNs notably enhanced UV shielding properties of the composite films, particularly in the short-wavelength UVC (200–280 nm) and medium-wavelength UVB (280–315 nm) regions. The banana preservation test further demonstrated that the chitosan-LCN composite films effectively inhibited bacterial growth and extended the shelf life of bananas at room temperature. Thus, these composite films offer a promising and sustainable solution for the high-value commercial use of agricultural by-products and chitosan in food packaging applications.

Design, synthesis and antibacterial evaluation of aldehyde chitosan-antibiotic conjugates

Yun Liu,Xuefang Shang,Ashraf H. Abadi,Mingpan Cheng

To address the challenge of antibiotic resistance and enhance the functionality of pharmaceutical materials, aldehyde chitosan-based dynamic polymers (ACs) incorporating amikacin and cefotaxime, denoted as ACs-amikacin and ACs-cefotaxime, respectively, were synthesized. These polymer's chemical structure was characterized using 1H-NMR and FT-IR analysis. Subsequently, methylthiazolyldiphenyl-tetrazolium bromide (MTT) and lactate dehydrogenase (LDH) assays were employed to evaluate their safety. Furthermore, it was demonstrated that these polymers retained the antibacterial activity of the antibiotics. This study introduces a novel strategy for dynamic natural materials capable of exerting a synergistic effect in pharmaceutical applications through the conjugation of natural materials with active pharmaceutical agents.

Quercetin-crosslinked chitosan nanoparticles: a potential treatment for allergic rhinitis

Mu Dehong,Zhou Li,Shi Lingyu,Liu Ting,Guo Ying,Chen Hao,Luo Hongping,Ma Junhao,Zhang Hui,Xiong Peizheng,Tian Li

Allergic rhinitis (AR) remains a major health problem worldwide. Compared with traditional oral drugs, nasal administration avoids first-pass metabolism and achieve faster and more effective efficacy. In this study, we used the ion crosslinking method to prepare quercetin–chitosan nasal adaptive nanomedicine (QCS) delivery system and evaluated in the treatment of allergic rhinitis mice models. The obtained positively charged nanoparticles with a particle size of 229.2 ± 0.2 nm have excellent characteristics in encapsulation efficiency (79.604%), drug loading rate (14.068%), drug release (673.068 μg) and stability(> 7 days). Excitingly, QCS treatment significantly reduced the number of sneezing and nasal rubbing events in AR mice, while reducing the levels of inflammatory factors such as immunoglobulin E (IgE), interleukin (IL)-17, tumor necrosis factor (TNF)-α, and (IL)-6 to alleviate AR symptoms. Hematoxylin–eosin (HE) staining also showed the damaged nasal mucosa was improved. These experimental results suggest that QCS can effectively suppress allergic inflammation in a mouse model and hold promise as a therapeutic option for allergic rhinitis.

Zwitterionic functionalized chitosan with dual-antifouling for selective uranium extraction

Luyan Li,Hao Li,Mingzhang Lin,Sheng Hu,Jun Wen

Uranium adsorbents deployed in natural water environments rich in microorganisms for a long time are inevitably affected. Biofouling severely reduces the uranium adsorption performance and damages the structure of the adsorbents. Therefore, antifouling performance is necessary for adsorbents that extract uranium from seawater and uranium-containing groundwater. Previously reported antifouling uranium adsorbents mainly have two types: single antibacterial or anti-adhesion, and few studies have developed dual-antifouling adsorbents that combine the two antifouling pathways. In this study, a dual-antifouling adsorbent CS-MPC with both anti-adhesion and antibacterial properties was constructed. The synergistic effect of the superhydrophilic 2-methylacryloxyethyl phosphocholine (MPC) and the natural bactericidal chitosan (CS) enabled the inhibition rate of dual-antifouling CS-MPC against various bacteria to exceed 99 %. Furthermore, the uranium adsorption capacity of CS-MPC was 579 mg/g in simulated uranium-containing groundwater and 8.28 mg/g after immersion for 42 days in real seawater. The SA/CS-MPC composite beads prepared to solve powder defects also exhibited good dual-antifouling activity and uranium adsorption properties, making them potential candidates for uranium extraction. The construction of dual-antifouling adsorbents provides a new strategy for effective biofouling control.

Preparation of high-strength, colored and multifunctional chitosan fibers via a novel strategy of enzyme-catalyzed phenolic-amine crosslinking within chitosan fibers in a swollen state

Zhichao Song,Shujie Xu,Bo Xu,Man Zhou,Ying Liu,Ying Zhang,Li Cui,Ping Wang,Qiang Wang,Yuanyuan Yu

This study prepared high-strength, colored and multifunctional polyphenol-chitosan fibers via a novel strategy of laccase-mediated phenolic-amine crosslinking within chitosan fibers in a swollen state. A dense covalent network within the fiber was formed through Michael addition and Schiff base reactions. Compared to unmodified chitosan fibers, the enzymatically cross-linked fibers exhibited significantly enhanced mechanical properties, with an increase of about 270% in dry tensile strength (up to 156.8 MPa) and excellent wet stability. In addition, the polyphenol-chitosan fibers were given color and different properties by the covalent cross-linking of polyphenols into the fiber. The polyphenol-chitosan fibers had stable colors and did not fade in the environment. They also had an exceptional antibacterial rate of 99.99% and excellent UV protection. This laccase-mediated phenolic-amine crosslinking strategy provides an effective, sustainable method for producing high-performance, bio-based functional fibers.

pH-sensitive chitosan-deoxycholic acid/alginate nanoparticles for oral insulin delivery

Ya-Wen Zhang,Ling-Lan Tu,Zhan Tang,Qiao Wang,Gao-Li Zheng,Li-Na Yin

Oral absorption of peptides/proteins is usually compromised by various gastrointestinal tract barriers. To improve delivery efficiency, chitosan-conjugated deoxycholic acid (CS-DCA) coupled with sodium alginate (ALG) was prepared to load insulin into pH-sensitive nanoparticles. The insulin-loaded chitosan-deoxycholic acid/alginate nanoparticles (CDA NPs) were characterized by size (143.3 ± 10.8 nm), zeta potential (19.5 ± 1.6 mV), entrapment efficiency (61.14 ± 1.67%), and insulin drug loading (3.36 ± 0.09%). The CDA NPs exhibited pH-triggered release characteristics in vitro and protected the wrapped insulin from gastric degradation. Stability of the CDA NPs in enzyme-containing simulated gastrointestinal fluids suggested that the NPs could partially protect the wrapped insulin from enzymatic degradation. Additionally, CS-DCA-modified NPs promoted the permeability of Caco-2 cells and enhanced intracellular absorption of FITC-labeled insulin by 9.4 and 1.2-folds, when compared to insulin solution and unmodified NPs, respectively. The positively charged NPs increased intestinal villi adhesion and enhanced insulin absorption in the intestines of diabetic rat models. Furthermore, the hypoglycemic test showed that CDA NPs prolonged insulin release in vivo and exerted a remarkable hypoglycemic effect on diabetic rats with an oral bioavailability of 15%. In conclusion, CDA NPs is a potential oral insulin delivery system.
