Gelatin-based nanofiber membranes loaded with curcumin and borneol as a sustainable wound dressing

Yarong Lv,Zhongxun Yu,Chenxi Li,Jianwei Zhou,Xujin Lv,Jia Chen,Ming Wei,Jun Liu,Xiaomin Yu,Ce Wang,Ping Hu,Yong Liu

Infection is a huge obstacle to wound healing. Thus, to enhance the healing of infected wounds, wound dressings that permit the dual delivery of antimicrobials and antioxidants are highly desirable. In this study, a series of gelatin-based nanofiber membranes with different curcumin contents were fabricated via solution electrospinning. The obtained membranes were characterized in terms of their morphologies, in addition to their physical, mechanical, and in vitro properties. The results showed that the membranes maintained an integrated morphology, excellent water absorption capability, satisfactory mechanical properties, and a high dissolution rate of curcumin. The addition of curcumin and borneol conferred the membranes the ability to inhibit Staphylococcus aureus and eliminate free radicals. Furthermore, cytocompatibility testing using the L929 cell line confirmed the excellent biocompatibility of the membranes. These gelatin-based nanofiber membranes loaded with curcumin and borneol can therefore be considered as promising materials for dressing wounds. Moreover, the use of biodegradable polymers and environmentally sustainable production techniques in this system render it suitable for the commercial manufacture of composite membranes.

Application of Curcumin Emulsion Carrier from Ultrasonic-Assisted Prepared Octenyl Succinic Anhydride Rice Starch

Yuxue Zheng,Huiling Zhang,Xiaobo Wei,Haitian Fang,Jinhu Tian

The emulsification of ultrasonic-assisted prepared octenyl succinic anhydride (OSA) rice starch on curcumin was investigated in the present study. The results indicated that the encapsulation efficiency of curcumin in emulsions stabilized by OSA-ultrasonic treatment rice starch was improved, from 81.65 ± 0.14% to 89.03 ± 0.09%. During the in vitro oral digestion, the particle size and Zeta potential of the curcumin emulsion did not change significantly (p> 0.05). During the in vitro digestive stage of the stomach and small intestine, the particle size of the curcumin emulsion continued to increase, and the absolute potential continued to decrease. Our work showed that OSA-pre-treatment ultrasonic rice starch could improve curcumin bioavailability by increasing the encapsulation efficiency with stronger stability to avoid the attack of enzymes and high intensity ion, providing a way to develop new emulsion-based delivery systems for bioactive lipophilic compounds using OSA starch. Keywords:rice starch;octenyl succinic anhydride;curcumin;emulsion

Protective effect of curcumin on hepatolenticular degeneration through copper excretion and inhibition of ferroptosis

Xun Sun,Xinyu Zhang,Hui Yan,Hongfei Wu,Shijian Cao,Wenchen Zhao,Ting Dong,An Zhou

Background Hepatolenticular degeneration (HLD) is an autosomal recessive disorder concerning copper metabolism . Copper overload is also accompanied by iron overload in HLD patients , which can lead to ferroptosis . Curcumin , the active component in turmeric, has the potential to inhibit ferroptosis. Purpose The current study proposed a systematic investigation of the protective effects of curcumin against HLD and the underlying mechanisms. Methods The protective effect of curcumin on toxic milk (TX) mice was studied. Liver tissue was observed via hematoxylin-eosin (H&E) staining and the ultrastructure of the liver tissue was observed through transmission electron microscopy . Copper levels in the tissues, serum, and metabolites were measured by atomic absorption spectrometry (AAS). In addition, serum and liver indicators were evaluated. In cellular experiments, the effect of curcumin on the viability of rat normal liver cells (BRL-3A) was determined via the 3-[4,5-dimethylthiazol-2-yl)]-2,5-diphenyltetrazolium bromide (MTT) assay. Cell and mitochondrial morphology were observed in curcumin-mediated HLD model cells. The intracellular copper ion fluorescence intensity was observed via fluorescence microscopy , and intracellular copper iron content was detected using AAS. Further, oxidative stress indicators were evaluated. Cellular reactive oxygen species (ROS) and cellular mitochondrial membrane potential were examined via flow cytometry. Furthermore, the expression levels of nuclear factor erythroid-2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), and glutathione peroxidase 4 (GPX4) were determined via western blotting (WB). Results The histopathology of the liver confirmed the hepatoprotective effects of curcumin. Curcumin improved copper metabolism in TX mice. Both serum liver enzyme markers and antioxidant enzyme levels indicated the protective effect of curcumin against HLD-related liver injury. The MTT assay results showed that curcumin was protective against excess copper-induced injury. Curcumin improved the morphology of HLD model cells and their mitochondrial morphology. The Cu 2+ fluorescent probe and the AAS results indicated that curcumin reduced Cu 2+ content in HLD hepatocytes. In addition, curcumin improved oxidative stress levels and prevented the decline of mitochondrial membrane potential in HLD model cells. The ferroptosis inducer Erastin reversed these effects of curcumin. WB revealed that curcumin promoted Nrf2, HO-1, and GPX4 protein expression in HLD model cells, and the Nrf2 inhibitor ML385 reversed the effects of curcumin. Conclusion Curcumin demonstrates a protective role by expelling copper and inhibiting ferroptosis, activating the Nrf2/HO-1/GPX4 signaling pathway in HLD.

Nitrogen-doped carbon dots/curcumin nanocomposite for combined Photodynamic/photothermal dual-mode antibacterial therapy

Fangzhou Wen,Peiyuan Li,Hongrong Meng,Hongjun Yan,Xiaodong Huang,Hao Cui,Wei Su

Due to their excellent photophysical properties, carbon quantum dots have great potential in biomedical and drug delivery fields. In this study, nitrogen-doped carbon quantum dots with good water solubility were prepared using citric acid and ethylenediamine as precursors, and compounded with curcumin, a photosensitive component, to produce composite nanomaterials with photodynamic therapy and synergistic photothermal therapy. The formation of nitrogen-doped carbon quantum dots and composite nanomaterials was verified using physical and optical means. In addition, the composite nanomaterials produced single-linear oxygen and exacerbated the increase of solution temperature under blue (405 nm) and near-infrared (808 nm) light irradiation, respectively. The plate counting method showed that the composite nanomaterials exhibited good photodynamic synergistic photothermal antibacterial properties against both Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus under dual light source (405+808 nm) irradiation, which improved the photoinactivation of curcumin against bacteria. In addition, the composite nanomaterials exhibited low toxicity and good hemocompatibility. These experimental results suggested that the composite nanomaterials showed great potential in a multimodal photodynamic therapy synergistic photothermal treatment platform.

pH-responsive curcumin-based nanoscale ZIF-8 combining chemophotodynamic therapy for excellent antibacterial activity

Xiaofeng Meng,Jingwei Guan,Shanshan Lai,Liming Fang,Jianyu Su

Antimicrobial photodynamic therapy (aPDT) is a highly attractive therapy due to its advantages of being a non-antibiotic procedure for reducing drug-resistant microbes. Curcumin (CCM) has been considered as a natural photosensitizer for PDT with prominent antibacterial, antifungal, and anti-proliferative activity. However, its excellent biological and pharmacological activities are limited because of its low solubility, rapid metabolization and instability. Herein, we reported a promising agent based on CCM-incorporated into zeolitic imidazolate framework-8 (ZIF@CCM). The as-prepared nanoparticle exhibited high drug loading capability (11.57%) and drug loading encapsulation (82.76%). Additionally, ZIF@CCM displayed a pH-responsive drug release behavior and chemophotodynamic therapy for excellent antibacterial activity. The underlying mechanism elucidated that Zn2+ released from ZIF-8 increased the permeability of the bacterial cell membrane with leakages of K+. The overproduction of extracellular ROS further resulted in the disrupted bacterial cell membrane and distorted bacterial morphology. Thus, ZIF@CCM-mediated photodynamic activation might be a promising treatment strategy for microbial inactivation.

Curcumin-Encapsulated Chitosan-Coated Nanoformulation as an Improved Otoprotective Strategy for Ototoxic Hearing Loss

Xiaozhu Chen,Hong Zhang,Chu Wang,Yue Su,Min Xiong,Xiaohua Feng,Daishi Chen,Zhaoyang Ke,Lu Wen,Gang Chen

Overexpression of apoptotic factors in the inner ear is generally proven to induce ototoxicity. This has aroused research interest in various antiapoptotic drugs, the most representative of which is curcumin (CUR). In this study, two nanoformulations of CUR were developed with sustained-release behavior to improve their protective effects against ototoxic hearing loss (HL), which were the nanoparticles of CUR-loaded poly(lactic acid–glycolic acid) (CUR-PLGA NPs) and CUR-loaded chitosan-coated PLGA NPs (CUR-CS/PLGA NPs). The obtained results revealed that both CUR-NPs provided otoprotection in vitro and in vivo, and their effective doses in guinea pigs were much less than that of dexamethasone, which was clinically used to treat HL. Moreover, relative to CUR and CUR-PLGA NPs, CUR-CS/PLGA NPs exhibited the highest accumulation in HEI-OC1 cells and guinea pigs’ cochlea. In pharmacodynamic experiments, the optimal administration timing was investigated, and CUR-CS/PLGA NPs showed sustained efficacy and the best hearing improvement at all tested sound frequencies. Lastly, the protective effect of CUR nanoformulations was further validated via inhibition of Caspase-3 and Bax activation, thereby reducing the concentration of reactive oxygen species and protecting mitochondrial integrity in hair cells. Collectively, CUR-CS/PLGA NPs demonstrated potent and lasting effects against ototoxic HL, making our novel formulation a promising candidate for the alleviation of sensorineural HL.

Construction and evaluation of curcumin upconversion nanocarriers decorated with MnO2 for tumor photodynamic therapy

Chen Xinru,Li Quandong,Huang Zipeng,Lin Wen,Ma Yan

The limited tissue penetration depth and tumor hypoxic microenvironment have become the two pivotal obstacles that alleviate the antineoplastic efficacy in tumor photodynamic therapy (PDT). In the research, MnO 2 -decorated upconversion nanoparticles (UCSMn) have been designed to generate certain oxygen within the solid tumor, and also increase the light penetrating depth due to the optical conversion ability derived from upconversion nanoparticles. Furthermore, upconversion nanoparticles as the inner core are coated by mesoporous silica for the loading of curcumin as photosensitizer and chemotherapeutics, and then a MnO 2 shell is proceeding to grow via redox method. When reaching the tumor tissue, the MnO 2 nanoshells of UCSMn could be rapidly degraded into manganese ions (Mn 2+ ) owing to the reaction with H 2 O 2 in acidic tumor microenvironment, meanwhile producing oxygen and facilitating curcumin release. Once the tumor is illuminated by 980 nm light, the upconversion nanoparticles can transform the infrared light to visible light of 450 nm and 475.5 nm, which can be efficiently absorbed by curcumin, and then produce singlet oxygen to induce tumor cell apoptosis. Curcumin played a dual role which can not only be acted as a photosensitizer, but also a chemotherapeutic agent to further reinforce the antitumor activity. In short, the intelligent nanostructure has the potential to overcome the above-mentioned shortcomings existed in PDT and eventually do work well in the hypoxia tumors. Oleogel-based Pickering emulsions stabilized by ovotransferrin–carboxymethyl chitosan nanoparticles for delivery of curcumin

Yue Dong,Zihao Wei,Yuming Wang,Qingjuan Tang,Changhu Xue,Qingrong Huang

This work prepared food-grade oleogel-based Pickering emulsion stabilized by ovotransferrin (OVT)–carboxymethyl chitosan (CMCS) nanoparticles (OPEOC). The analysis of emulsified phase volumes and stability index exhibited that OPEOC with different volumes of oil fraction remained stable for 2 months of storage. The prepared Pickering emulsions were used as delivery systems for curcumin. In vitro digestive studies showed that OPEOC significantly improved the degree of lipolysis and the bioaccessibility of curcumin compared to the sunflower seed oil-based oleogel. The oleogel-based Pickering emulsions fabricated in the present study had noticeable storage stability, high loading and improved bioaccessibility of nutraceuticals, which might provide new insights into the development of novel food-grade Pickering emulsions with excellent properties.

Rheology and texture analysis of gelatin/dialdehyde starch hydrogel carriers for curcumin controlled release

Tianqi Cui,Yue Wu,Chunlei Ni,Yuxue Sun,Jianjun Cheng

A hydrogel carrier was prepared by synergistic cross-linking of dynamic imine bonds and hydrogen bonds between gelatin and dialdehyde starch (DS). The dynamic gelation process and rheological and mechanical properties of hydrogels with different concentrations of DS were studied by dynamic rheology and texture analysis. By changing the content of DS, the gelation kinetics of the hydrogel were adjusted to determine the different gelation times and ensure good injectability. Hydrogels have a stable and firm network structure, and the network can regenerate rapidly after dissociation and collapse. The hydrogel had self-recovery and fatigue resistance, and its hardness, elasticity, deformation capacity, compressive strength and modulus increased with increasing DS content. The hydrogel had good pH-dependent controlled-release ability for curcumin . Injectable hydrogels with biocompatibility, tunable mechanical properties and self-healing properties were used as drug or nutrient delivery carriers for more controlled placement and release.

Esterase-triggered rapid release of succinic anhydride conjugated curcumin co-prodrug for osteosarcoma therapy

Yuxin Wan,Zhenze Xie,Mingjie Wang,Yuelian Liu,Mubin Zheng,Dong Xu,Chang Du

Osteosarcoma (OS) is one of the most common primary bone malignancies in children and adolescents worldwide. The serious side effects and multi-drug resistance of high-dose chemotherapy have become the major cause of the failure in OS treatment. Curcumin (Cur) is reported to selectively kill OS cells and has a low cytotoxicity on healthy osteoblasts, but its delivery remains a great challenge. Herein, a co-polymer-Cur prodrug (co-prodrug) was developed by polymerizing Cur with hydrophilic polymer modified by succinic anhydride (SA) to explore the physicochemical properties, drug release and potential biological effects for OS treatment. This design endowed the co-prodrug with excellent solubility and stability under different solution conditions (pH = 4.5, 7.4 and 8.5), and improved the release efficiency of reactive oxygen species. Interestingly, co-prodrug exhibited an esterase-triggered rapid drug release in a concentration-dependent manner rather than the well-known acid response. Moreover, co-prodrug could be internalized into cells through endocytosis and significantly inhibit the proliferation of OS cells. Additionally, co-prodrug had no obvious systemic toxicity, and could suppress the OS growth by promoting apoptosis, inhibiting proliferation activity and vascularization in vivo. Our research furnishes a fresh perspective for the formulation design of controlled release of insoluble and instable anticancer agents within tumor cells.

Redox-Responsive Lipidic Prodrug Nano-Delivery System Improves Antitumor Effect of Curcumin Derivative C210

Xin Guo,Min Wu,Yanping Deng,Yan Liu,Yanpeng Liu,Jianhua Xu

The poor bioavailability of curcumin and its derivatives limits their antitumor efficacy and clinical translation. Although curcumin derivative C210 has more potent antitumor activity than curcumin, it has a similar deficiency to curcumin. In order to improve its bioavailability and accordingly enhance its antitumor activity in vivo, we developed a redox-responsive lipidic prodrug nano-delivery system of C210. Briefly, we synthesized three conjugates of C210 and oleyl alcohol (OA) via different linkages containing single sulfur/disulfide/carbon bonds and prepared their nanoparticles using a nanoprecipitation method. The prodrugs required only a very small amount of DSPE-PEG2000 as a stabilizer to self-assemble in aqueous solution to form nanoparticles (NPs) with a high drug loading capacity (~50%). Among them, the prodrug (single sulfur bond) nanoparticles (C210-S-OA NPs) were the most sensitive to the intracellular redox level of cancer cells; therefore, they could rapidly release C210 in cancer cells and thus had the strongest cytotoxicity to cancer cells. Furthermore, C210-S-OA NPs exerted a dramatic improvement in its pharmacokinetic behavior; that is, the area under the curve (AUC), mean retention time and accumulation in tumor tissue were 10, 7 and 3 folds that of free C210, respectively. Thus, C210-S-OA NPs exhibited the strongest antitumor activity in vivo than C210 or other prodrug NPs in mouse models of breast cancer and liver cancer. The results demonstrated that the novel prodrug self-assembled redox-responsive nano-delivery platform was able to improve the bioavailability and antitumor activity of curcumin derivative C210, which provides a basis for further clinical applications of curcumin and its derivatives. Keywords:curcumin derivatives;redox-responsive;self-assemble;prodrug;bioavailability;antitumor

Binding affinity of curcumin to bovine serum albumin enhanced by pulsed electric field pretreatment

Rui Wang,Qin-Hui Wen,Xin-An Zeng,Jia-Wei Lin,Jian Li,Fei-Yue Xu

The present study investigated the effect of pulsed electric field (PEF) pretreatment on the interaction between bovine serum albumin (BSA) and curcumin. Fluorescence quenching results showed that proper PEF pretreatment significantly increased the binding affinity of curcumin and BSA, the binding constant increased by 6.77 times under the conditions of 15 kV/cm for 0.51 ms. However, at higher PEF strength (≥25 kV/cm) and longer processing time (≥0.68 ms), the binding affinity was weakened. PEF pretreatment made the protein structure more disordered and induced partial unfolding of BSA, exposing more hydrophobic regions, thereby increasing the binding affinity to curcumin. PEF-treated BSA (PBSA) possessed better encapsulation efficiency (95.19%) and loading capacity (5.25 mg/g) for curcumin, and the storage stability of curcumin were enhanced by the formation of a complex with PBSA. This study provides new insights into the design of BSA-based delivery systems for curcumin and other hydrophobic nutrients.
