Synthesis of N-isonicotinic sulfonate chitosan and its antibiofilm activity against E. coli and S. aureus

Lin Wang,Yu Pang,Zhongwen Su,Meihua Xin,Mingchun Li,Yangfan Mao

N -(sodium 2-hydroxypropylsulfonate) chitosan (HSCS), N -sulfonate chitosan (SCS) and N -isonicotinic sulfonate chitosan (ISCS) were prepared. The structures of the prepared chitosan derivatives were characterized by nuclear magnetic resonance ( 1 H NMR) spectroscopy, Fourier transform infrared (FTIR) spectroscopy, ultraviolet–visible (UV–vis) spectroscopy and elemental analysis (EA). Antibacterial and antibiofilm activities of these chitosan derivatives were evaluated in vitro . The minimum inhibitory concentration (MIC) of HSCS and SCS against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) were 0.625 mg/mL and 0.156 mg/mL, respectively. ISCS exhibited MIC values of 0.313 mg/mL and 0.078 mg/mL against E. coli and S. aureus , respectively. ISCS demonstrated superior antibacterial and antibiofilm properties compared to SCS and HSCS. These findings suggest that the incorporation of a pyridine structure into sulfonate chitosan enhances its antibacterial and antibiofilm activities, and the prepared ISCS has a promising application prospect for controlling the reproduction of microorganisms in the field of food packaging.

The Antimicrobial Peptide D-CONGA-Q7 Eradicates Drug-Resistant E. coli by Disrupting Bacterial Cell Membranes

Zonghan Jiang,Leisheng Sun,Yuanyuan Li,Haoyu Li,Yu Fu,Jiyun Li,Zhiliang Sun

Simple SummaryEscherichia coliis a ubiquitous pathogen, with certain strains causing severe diarrhea and intestinal illnesses, which pose significant health risks to both children and animals. As antimicrobial resistance continues to rise, traditional antibiotics are increasingly ineffective, highlighting the urgent need for novel therapeutic strategies. In this study, we investigated the antimicrobial peptide D-CONGA-Q7, aiming to evaluate its efficacy and underlying mechanism against drug-resistantE. colistrains. Our results demonstrate that D-CONGA-Q7 effectively disrupts bacterial cell membranes, leading to bacterial death, and exhibits comparable activity against polymyxin- and meropenem-resistant strains. In vivo experiments revealed that D-CONGA-Q7 significantly alleviated intestinal inflammation, reduced inflammatory cytokine levels, and promoted the repair of damaged intestinal tissues. Furthermore, D-CONGA-Q7 displayed good stability. These findings suggest that D-CONGA-Q7 not only represents a promising new approach to combat drug-resistant bacterial infections but also holds potential for clinical application.Escherichia coli(E. coli) is a zoonotic bacterium widespread in the environment, highly transmissible, and responsible for significant economic losses and millions of cases of illness annually. The rise of multidrug-resistant (MDR) strains has rendered last-line antibiotics such as polymyxin and meropenem ineffective, making the development of new antibiotics urgent. Although D-CONGA-Q7 has broad-spectrum bactericidal activity, its underlying mechanism remains poorly understood. In this study, we used in vitro and in vivo experiments to demonstrate that D-CONGA-Q7 effectively kills both antibiotic-sensitive and multidrug-resistant strains ofE. coli. D-CONGA-Q7 disrupts the cell membranes of Gram-negative bacteria, and the treatment ofE. colistrainLN175with D-CONGA-Q7 resulted in a significant up-regulation of theMlacgene, suggesting that D-CONGA-Q7 may interact with phospholipids in the cell membrane. Furthermore, in treatingK88-induced bacterial enteritis in the small intestine, D-CONGA-Q7 significantly reduced intestinal inflammation. In conclusion, this study provides a novel approach to combat drug-resistantE. coli.

Flexible and Robust Pressure Sensor Based on MXene-Incorporated Polyamide 6 Membrane for E-Skin Applications

Salah Naji Sufyan,Yusof A. Y. A. Mohammed,Chenglong Mu,Kashan Bashir,Ning Wen,Muhammad Mamoor,Xiuling Jiao,Dairong Chen,Ting Wang

Flexible piezoresistive pressure sensors are crucial for next-generation wearable electronics, yet achieving simultaneous high sensitivity, durability, and mechanical robustness remains a challenge. Herein, we report a high-performance piezoresistive sensor fabricated through the integration of two-dimensional MXene nanosheets with a hierarchically structured modified polyamide 6 nanofiber net (D-PA6 NFN) using scalable vacuum filtration. Benefiting from the synergistic combination of MXene’s excellent electrical properties, web-like architecture, the composite nanofiber tensile strength (52 MPa), and flexibility (40%), the resulting sensor exhibited good sensitivity (0.189 kPa–1), a rapid response/recovery time (14/13 ms), and an ultralow detection limit (4.2 Pa). Moreover, the device maintained stability over 12,000 compression cycles. The sensor precisely captured both subtle physiological signals (pulse waveforms) and large-scale dynamic motions (joint flexion). This work establishes a scalable strategy for engineering durable, high-sensitivity electronic skins, with immediate applications in advanced human-machine interfaces and real-time physiological monitoring.

8-Epidiosbulbin E acetate triggers apoptosis via metabolic activation-mediated crosstalk between mitochondria and endoplasmic reticulum

Shiyu Zhang,Bowen Gong,Rong Tan,Yufen Liao,Xin Wang,Xu Wang,Ying Liu,Ting Liu,Jun Luo,Weiwei Li,Jia Yu,Ying Peng,Jiang Zheng

8-Epidiosbulbin E acetate (EEA), a furan-containing diterpenoid lactone and key component of Dioscorea bulbifera L., has been found to induce notable liver injury in mice through CYP3A-mediated metabolic activation to a cis -enedial reactive metabolite. EEA exposure at 25, 50, 100, or 200 µM triggered apoptosis in cultured mouse primary hepatocytes, evidenced by alterations in mitochondrial and endoplasmic reticulum (ER) pathway protein levels. Pretreatment with ketoconazole, vitamin C, glutathione ethyl ester, or 4-phenylbutyric acid effectively reduced the apoptotic ratio and restored the balance of mitochondrial and ER pathway protein levels impacted by EEA in primary hepatocytes. In contrast, pretreatment with L-buthionine sulfoximine exacerbated the apoptotic ratio and altered protein levels associated with the mitochondrial and ER pathways induced by EEA. In summary, metabolic activation, GSH depletion, and ROS are implicated in the induction of apoptosis. EEA triggered apoptosis in cultured primary hepatocytes via disruption of the crosstalk between the mitochondrial and ER pathways.

Salicylic acid regulates biosynthesis of floral fragrance (E)-β-farnesene via NPR3-WRKY1 module in chrysanthemum

Wang Zhiling,Yuan Yixin,Dong Rui,Zeng Ruihong,Zhao Xin,Xu Yanjie,Gao Junping,Hong Bo,Gu Zhaoyu

Floral fragrance is essential for the attraction of pollinators and responses to biotic and abiotic stresses. It also enhances the quality and economic value of plants. Phytohormones, acting as key signaling molecules, are crucial roles in regulating plant growth and development. However, the molecular mechanisms underlying the biosynthesis of fragrance-related volatiles and their crosstalk with other endogenous signals within plants remain largely unknown. Here, we identified a sesquiterpene synthase gene, CmEβFS , in chrysanthemum, which encodes a catalytic enzyme responsible for synthesizing the key fragrance-related compound ( E )-β-farnesene. We demonstrated that CmEβFS is negatively regulated by CmWRKY1, thereby inhibiting ( E )-β-farnesene biosynthesis. Furthermore, CmWRKY1 interacts with the salicylic acid (SA) receptor CmNPR3 to regulate SA-mediated CmEβFS transcription. Our findings reveal that SA regulates ( E )-β-farnesene biosynthesis by controlling CmEβFS expression via the CmWRKY1-CmNPR3 module during floral development. These findings enhanced our understanding of the mechanisms underlying SA-mediated regulation of volatile organic compounds (VOCs) biosynthesis and provided insights into improving floral fragrance.

Quality identification of Amomi fructus using E-nose, HS-GC-IMS, and intelligent data fusion methods

Pan-Pan Zhang,Xin-Jing Gui,Xue-Hua Fan,Han-Li,Hai-Yang Li,Xiao-Peng Li,Feng-Yu Dong,Yan-Li Wang,Jing-Yao,Jun-Han Shi,Rui-Xin Liu

Amomi fructus (AF) has been used for both medicinal and food purposes for centuries. However, issues such as source mixing, substandard quality, and product adulteration often affect its efficacy. This study used E-nose (EN) and headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) to determine and analyze the volatile organic compounds (VOCs) in AF and its counterfeit products. A total of 111 VOCs were detected by HS-GC-IMS, with 101 tentatively identified. Orthogonal Partial Least Squares-Discriminant Analysis (OPLS-DA) identified 47 VOCs as differential markers for distinguishing authentic AF from counterfeits (VIP value >1 and P < 0.05). Based on the E-nose sensor response value and the peak volumes of the 111 VOCs, the unguided Principal Component Analysis (PCA), guided Principal Component Analysis-Discriminant Analysis (PCA-DA), and Partial Least Squares-Discriminant Analysis (PLS-DA) models were established to differentiate AF by authenticity, origin, and provenance. The authenticity identification model achieved 100.00% accuracy after PCA analysis, while the origin identification model and the provenance identification model were 95.65% (HS-GC-IMS: PLS-DA) and 98.18% (HS-GC-IMS: PCA-DA/PLS-DA), respectively. Further data-level fusion of E-nose and HS-GC-IMS significantly improved the accuracy of the origin identification model to 97.96% (PLS-DA), outperforming single-source data modeling. In conclusion, the intelligent data fusion algorithm based on E-nose and HS-GC-IMS data effectively identifies the authenticity, origin, and provenance of AF, providing a rapid and accurate method for quality evaluation.

Molecularly engineering cellulose to functional cellulose-based aerogel adsorbent for precious metals recovery from e-waste

Yumei Chen,Chunhui Xie,Yang You,Tonghui Xu,Yunqi Li,Jili Yuan,Haibo Xie,Yuanlong Guo

The demand for the recovery of precious metals has become increasingly prominent due to their scarcity and wide application, while the facile preparation of sustainable absorption and separation materials via molecular engineering still face great challenge by using renewable polymer resources. Herein, a water-soluble cellulose levulinate ester (CLE) with ketocarbonyl groups was utilized as 3D network scaffold to fabricate a cellulose-based aerogel adsorbent (CLE@PEI) by introducing polyethyleneimine (PEI) with multiple amine groups. The abundant functional groups and porous architecture allowed CLE@PEI to efficiently capture Au(III), Pt(IV) and Pd(II). The maximum adsorption capacity of CLE@50PEI reached a staggering 1752.0 mg/g for Au(III) at 298 K which was superior than that of Pt(IV) (1420.5 mg/g) and Pd(II) (495.1 mg/g). The adsorption process followed pseudo-second-order kinetic model and Langmuir isotherm behavior, respectively. Additionally, the positively charged CLE@50PEI exhibited excellent selectivity for precious metals in multi-metal mixtures. The outstanding adsorption performance of the CLE@50PEI may be due to the new “adsorption-in situ reduction” mechanism, in which the process primarily involved the ion exchange coupled with electrostatic interaction and complexation, and the in situ reduction of absorbed Au(III) to single crystalline flakes. Moreover, quantitative desorption of the CLE@50PEI-loaded metals was achieved in acidic thiourea solution.

Instant and secure fixation of e-PTFE medical implants in surgery by designing a bioadhesive interface

Yang Gao,Daming Liu,Jing Zhou,Wen Zhang,Danqi Sun,Shuyang Wang,Yipeng Yang,Yuchen Xiang,Yang Zhang,Wei Lei,Tongqing Lu

Expanded polytetrafluoroethylene (e-PTFE) is extensively used in medical implants for its excellent bioinertness. Existing methods to fix e-PTFE implants on host tissues mainly use invasive anchorage such as sutures, spiral tacks, or metal skeletons, which has limitations such as being time-consuming and causing leakage and tissue damage. To overcome these limitations, we introduce a bioadhesive interface to realize the adhering fixation of e-PTFE implants. We integrate a hydrophilic and bioadhesive hydrogel layer on the hydrophobic and bioinert e-PTFE by designing a facile approach of freezing-induced micromechanical interlocking. The integration is robust enough as pulling hydrogel out of the interlocked pores on e-PTFE requires large energy dissipation. This bioadhesive interface enables instant (operating time < 10 seconds) and secure (adhesion energy >200 joules per square meter) adhering fixation of e-PTFE implants to wet tissue. The advantages for reducing inflammatory response, fibrogenesis, and the resultant postoperative adhesion are further demonstrated in a reconstructive surgery of an abdominal wall defect in rabbits.

Improved methyl supply for 5-methyltetrahydrofolate production in E. coli through a novel C1 transfer pathway

Liu Wen,Guo Jing,Lu Wei,Cheng Tao,Li Yiting,Xian Mo,Zhang Rubing

Background L -5-Methyltetrahydrofolate (5-MTHF) is the sole biologically active form of folate present in human blood and serves as an essential nutritional supplement. While microbial biosynthesis of 5-MTHF offers a sustainable alternative to chemical synthesis, its low yield limits industrial potential. Results In this study, strategies for improving the methyl supply combined with engineering the tetrahydrofolate (THF) synthetic pathway were employed in E. coli to increase 5-MTHF production. First, a new exogenous C1 pathway was introduced to improve the intracellular methyl supply through acetyl-CoA breakdown. High expression of key rate-limiting genes folE , folP and purU enhanced metabolic flux of THF pathway, resulting in a 5-MTHF titer of 1.075 mg/L during shake-flask fermentation. A subsequent increase in 5-MTHF production was achieved by knocking out the metE gene, which is involved in the consumption of 5-MTHF. The best engineered strain, M3012, produced 8.2 mg/L 5-MTHF in a 5 L bioreactor via fed-batch fermentation, which presented the highest 5-MTHF titer to date. Conclusion We successfully engineered E. coli by introducing a novel exogenous C1 metabolic pathway to augment the methyl donor pool essential for the biosynthesis of 5-MTHF. Further metabolic optimizations, including the enhancement of the THF precursor flux and the elimination of competing degradation pathways, developed a recombinant strain with significantly increased yield, which paves the way for industrial production of 5-MTHF.

Extracellular production of Ulp1403-621 in leaky E. coli and its application in antimicrobial peptide production

Fu Linglong,Sun Mengning,Wen Weizhang,Dong Na,Li Defa

Abstract Small ubiquitin-like modifier (SUMO) tag is widely used to promote soluble expression of exogenous proteins, which can then be cleaved by ubiquitin-like protease 1 (Ulp1) to obtain interested protein. But the application of Ulp1 in large-scale recombinant protein production is limited by complicated purification procedures and high cost. In this study, we describe an efficient and simple method of extracellular production of Ulp1 403-621 using a leaky Escherichia coli BL21(DE3), engineered by deleting the peptidoglycan-associated outer membrane lipoprotein ( pal ) gene. Ulp1 403-621 was successfully leaked into extracellular supernatant by the BL21(DE3)-Δ pal strain after IPTG induction. The addition of 1% glycine increased the extracellular production of Ulp1 403-621 approximately four fold. Moreover, extracellular Ulp1 403-621 without purification had high activities for cleaving SUMO fusion proteins, and antimicrobial peptide pBD2 obtained after cleavage can inhibit the growth of Staphylococcus aureus . The specific activity of extracellular Ulp1 403-621 containing 1 mM EDTA and 8 mM DTT reached 2.0 × 10 6 U/L. Another commonly used protease, human rhinovirus 3C protease, was also successfully secreted by leaky E. coli strains. In conclusion, extracellular production of tool enzymes is an attractive way for producing large-scale active recombinant proteins at a lower cost for pharmaceutical, industrial, and biotechnological applications. Key points • First report of extracellular production of Ulp1 403-621 in leaky Escherichia coli BL21(DE3) strain. • One percent glycine addition into cultivation medium increased the extracellular production of Ulp1 403-621 approximately four fold. • The specific activity of extracellular Ulp1 403-621 produced in this study reached 2.0 × 10 6 U/L.

Freestanding Transparent Organic–Inorganic Mesh E-Tattoo for Breathable Bioelectrical Membranes with Enhanced Capillary-Driven Adhesion

Xiang Li,Junyi Zhang,Bo Shi,Yawen Li,Yanan Wang,Kexiang Shuai,Yue Li,Gege Ming,Tao Song,Weihua Pei,Baoquan Sun

The electronic tattoo (e-tattoo), a cutting-edge wearable sensor technology adhered to human skin, has garnered significant attention for its potential in brain–computer interfaces (BCIs) and routine health monitoring. Conventionally, flexible substrates with adhesion force on dewy surfaces pursue seamless contact with skin, employing compact airtight substrates, hindering air circulation between skin and the surrounding environment, and compromising long-term wearing comfort. To address these challenges, we have developed a freestanding transparent e-tattoo featuring flexible serpentine mesh bridges with a unique full-breathable multilayer structure. The mesh e-tattoo demonstrates remarkable ductility and air permeability while maintaining robust electronic properties, even after significant mechanical deformation. Furthermore, it exhibits an impressive visible-light transmittance of up to 95%, coupled with a low sheet resistance of 0.268 Ω sq–1, ensuring both optical clarity and electrical efficiency. By increasing the number of menisci between the mesh e-tattoo and the skin, the total adhesion force increases due to the cumulative capillary-driven effect. We also successfully demonstrated high-quality bioelectric signal collections. In particular, the controlling virtual reality (VR) objects using electrooculogram (EOG) signals collected by mesh e-tattoos were achieved to demonstrate their potential for human–computer interactions (HCIs). This freestanding transparent e-tattoo with a fully breathable mesh structure represents a significant advancement in flexible electrodes for bioelectrical signal monitoring applications.
