Ruiqiang Wang,Hongying Xue,Jinpeng Leng,Jialiang Zhang,Zhen Yan,Xudong Liu,Hengyu Feng,Linghan Xiao,Wei Zhu

Bacteria have exerted a great threat to the human living environment. Therefore, many industries put forward higher requirements for the antibacterial properties of cellulose-based materials, especially the applications in food packaging, biomedical, and textile industry. In this paper, through using synthetic lysine grafted N-halamine organic antibacterial agent and hemp fiber as raw materials, the lysine grafted N-halamine Schiff base type hemp cellulose-based antibacterial material is prepared by simple and mild Schiff base reaction. This hemp cellulose-based antibacterial material combines various sterilization methods of hemp fiber, amino acid type Schiff base and N-halamine. The synergistic cooperation of different sterilization methods greatly improves the antibacterial performance of material and reduces the possible bacterial resistance. The antibacterial examination shows that S. aureus and E. coli could be killed totally by this material in 5 min, and the inhibition zone is increased to 18.4 mm. Meanwhile, this hemp cellulose-based antibacterial material has low cost, green preparation, excellent storage stability and regeneration ability , which provides a novel way for the development of synergistic application of different antibacterial agents and the use of cellulose-based materials in bacterial environment.

Covalent grafting of hyperbranched poly-L-lysine on Ti-based implants achieves dual functions of antibacteria and promoted osteointegration in vivo

Zhijian Yang,Yue Xi,Jun Bai,Zhiwei Jiang,Shuqin Wang,Haolan Zhang,Wei Dai,Chaozhen Chen,Zhongru Gou,Guoli Yang,Changyou Gao

The dual functional implants of antibacteria and osteointegration are highly demanded in orthopedic and dentistry, especially for patients who suffer from diabetes or osteoporosis simultaneously. However, there is lack of the facile and robust method to produce clinically applicable implants with this dual function although coatings possessing single function have been extensively developed. Herein, hyperbranched poly-L-lysine (HBPL) polymers were covalently immobilized onto the alkali-heat treated titanium (Ti) substrates and implants by using 3-glycidyloxypropyltrimethoxysilane (GPTMS) as the coupling agent, which displayed excellent antibacterial activity against S. aureus and E. coli with an efficiency as high as 89.4% and 92.2% in vitro , respectively. The HBPL coating also significantly promoted the adhesion, spreading, proliferation and osteogenic differentiation of MC3T3-E1 cells in vitro . Furthermore, the results of a S. aureus infection rat model in vivo ulteriorly verified that the HBPL-modified screws had good antibacterial and anti-inflammatory abilities at an early stage of implantation and better osteointegration compared with the control Ti screws.

Curcumin-loaded high internal phase emulsions stabilized with lysine modified lignin: a biological agent with high photothermal protection and antibacterial properties

Kai Chen,Shengrong Yuan,Dan Wang,Dongming Qi,Fengfeng Chen,Xueqing Qiu

Bacterial infections and multidrug resistance can seriously endanger the health and lives of humans, therefore the development of novel and efficient antibacterial strategies and drugs is urgently needed. Herein, a series of highly biocompatible lysine modified enzymatic hydrolysis lignins (EHL-Lys-x) were synthesized using the Mannich reaction. The sterilizing efficiency of EHL-Lys-2.0 against S. aureus and E. coli at 20 mg mL−1 is 93% and 50%, respectively, which is 26% higher than pure EHL. Quartz crystal microbalance with dissipation monitoring (QCM-D) and atomic force microscopy (AFM) analysis showed that the adsorption and adhesive force between EHL-Lys-x and bacteria increase with the increased amount of grafting of Lys on EHL owing to the increase of the electrostatic interaction between the EHL-Lys-x and bacteria, which results in an improvement in the antimicrobial activity of EHL-Lys-x. Subsequently, EHL-Lys-x combined with alkyl polyglucoside (APG) was used to stabilize the high internal phase emulsion containing curcumin (HIPEs-cur). The dispersed phase fraction of HIPE-cur is 87 vol%, which is the highest internal phase reported to date in the medical research area. The highest residual levels of curcumin in HIPEs are 60-fold, 3-fold and 5-fold compared to that in bulk oil after treatment with UV radiation, thermal emittance and after storage, respectively. The minimum inhibitory concentrations of HIPEs-cur against S. aureus and E. coli were found to be 1.56 and 6.25 mg mL−1, respectively, which are far higher than that of pure EHL-Lys-x. This strategy not only increases the chemical stability and bioavailability of curcumin, but also provides a novel method for the application of lignin in biomedical science.

Poly-l-lysine-modified reduced graphene oxide stabilizes the copper nanoparticles with higher water-solubility and long-term additively antibacterial activity

Yu Ouyang,Xiang Cai,QingShan Shi,Lili Liu,Dongliang Wan,Shaozao Tan,Yousheng Ouyang

In order to improve the water-solubility and long-term antibacterial activity of copper nanoparticles (CuNPs), a poly- l -lysine-modified reduced graphene oxide (PLL–rGO) was used as the carrier of CuNPs, and a poly- l -lysine/reduced graphene oxide/copper nanoparticles (PLL–rGO–CuNPs) hybrid was prepared by anchoring the CuNPs on the reduced graphene oxide surface. The novel PLL–rGO–CuNPs hybrid was characterized and the antibacterial activity of it on gram-negative Escherichia coli and Gram-positive Staphylococcus aureus was tested. Such a hybrid showed additively antibacterial activity, and the CuNPs on PLL–rGO were more stable than those on polyvinyl pyrrolidone, resulting in long-term additively antibacterial effect. Meanwhile, this hybrid showed excellent water-solubility, suggesting great potential application in microbial control.

Selective recognition of d-tryptophan from d/l-tryptophan mixtures in the presence of Cu(II) by electropolymerized l-lysine film

Fang Wang,Wencheng Gong,Lili Wang,Zilin Chen

Selective recognition of d -tryptophan ( d -Trp) in the presence of Cu(II) was investigated at poly- l -lysine (p-l-Lys) film using electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV). p-l-Lys film was immobilized on a glassy carbon electrode (GCE) by cyclic voltammetry between 0.0 and 1.9 V in 20 mM phosphate buffer solution (pH 8.6). After the p-l-Lys/GCE electrode was incubated with d -Trp solution containing Cu(II) ions, obvious enhancement of electron transfer resistance and decrease of voltammetric current could be observed. If d -Trp was replaced by l -tryptophan (l-Trp), there was no apparent resistance and current changes. Moreover, no resistance and current changes could be found in the absence of Cu(II). It may be due to the formation of Cu complex with l -lysine and d -tryptophan. Finally, this method was successfully applied to monitoring enantiomeric composition of the d -Trp and l-Trp mixtures.

Aspirin, lysine, mifepristone and doxycycline combined can effectively and safely prevent and treat cancer metastasis: prevent seeds from gemmating on soil

Liyuan Wan,Haiyan Dong,Huo Xu,Ji Ma,Yewei Zhu,Yusheng Lu,Jichuang Wang,Ting Zhang,Tao Li,Jingjing Xie,Bo Xu,Fangwei Xie,Yu Gao,Jingwei Shao,Xiaohuang Tu,Lee Jia

Recent scientific advances have increased our understanding of the cancer metastatic complexities and provided further impetus for new combination therapies to prevent cancer metastasis. Here, we demonstrated that a combination (HAMPT) of aspirin, lysine, mifepristone and doxycycline can effectively and safely prevent cancer metastasis. The pharmaceutically-formulated HAMPT inhibited adhesion of cancer cells to either endothelial cells or extracellular matrix via down-regulating cell adhesion molecules ICAM-1 and α 4 -integrin. HAMPT inhibited the cloak effect by activated platelets on cancer cells, thereby interfering adhesion and invasion of cancer cells to the underlying stroma. At the effective concentration, HAMPT induced cancer cells into dormancy with minor inhibition on cell viability. Four-day pretreatment followed by 30-day oral administration of HAMPT (33.5-134 mg/kg) to the mice inoculated with cancer cells produced significant inhibition on cancer metastasis dose-dependently without marked side effects. Fifty-day rat toxicity study with HAMPT at doses (335-1340 mg/kg) 20-fold higher than its therapeutic dose produced no significant toxicity. Interestingly, the acute toxic death could not be reached at the maximum administrable dose (5 g/kg). This proof-of-concept study provides the first conceptual evidence that cancer metastasis can be controlled by using affordable old drugs to restrain circulating tumor cells from gemmating on the metastatic soil without the need for cytotoxicity.

Poly(amino acid-hydroxyethyl methacrylate)s with chiral lysine and/or leucine side moieties and their antibacterial abilities for biomedical applications

Suli Hu,Wenhong Hu,Zhenguang Li,Lixia Ren,Yunhui Zhao,Xiaoyan Yuan

Biomedical applications of the synthetic polymers with amino acid side moieties have been studied in light of their biocompatibility. In this work, poly(amino acid-hydroxyethyl methacrylate)s with lysine (Lys)/leucine side moieties were synthesized via reversible addition-fragmentation chain transfer polymerization, and were verified with intrinsic chirality. For biomedical uses, their antibacterial abilities as well as hemolysis and cytotoxicity were investigated. The results indicated that there were similar inhibitions for d - and l -enantiomeric polymers against Staphylococcus aureus when the polymer concentrations were lower than ~   300   μg/mL, while the polymer containing d -Lys moiety was more effective against Escherichia coli . The d -enantiomers at concentrations <   500   μg/mL showed relatively lower hemolysis, and all the polymers showed good in vitro cell viability with concentrations lower than 200   μg/mL. It was indicated that the antibacterial amino acid-related synthetic polymers could be potentially applied for biomedical applications.

Hemostatic Dressing Immobilized with ε-poly-L-lysine and Alginate Coated Mesoporous Bioactive Glass Prevents Blood Permeation by Pseudo-Dewetting Behavior

Zilin Wu,Yilin Ding,Zhihao Qin,Zhipeng Sun,Zetao Wang,Xiaodong Cao

The integration of hemostats with cotton fabrics is recognized as an effective approach to improve the hemostatic performance of dressings. However, concerns regarding the uncontrollable absorption of blood by hydrophilic dressings and the risk of distal thrombosis from shed hemostatic agents are increasingly scrutinized. To address these issues, this work develops an advanced dressing (AQG) with immobilized nano-scale mesoporous bioactive glass (MBG) to safely and durably augment hemostasis. The doubly immobilized MBGs, pre-coated with ε-poly-L-lysine and alginate, demonstrate less than 1% detachment after ultrasonic washing. Notably, this MBG layer significantly promotes the adhesion, aggregation, and activation of red blood cells and platelets, adhered five times more red blood cells and 29 times more platelets than raw dressing, respectively. Specially, with the rapid formation of protein corona and amplification of thrombin, dense fibrin network is built on MBG layer and then blocked blood permeation transversely and longitudinally, showing an autophobic pseudo-dewetting behavior and allowing AQG to concentrate blood in situ and culminate in faster hemostasis with lower blood loss. Furthermore, the potent antibacterial properties of AQG extend its potential for broader application in daily care and clinical setting.

Layer-by-layer assembled film of TiO2/poly(l-lysine)/graphene quantum dots for simultaneous electrochemical determination of guanine and adenine

Raril Chenthattil,Sijian Luo,Hang Ao,Wencheng Xiao,Huangxian Ju

This work proposed an electrochemical biosensing method for simultaneous detection of guanine and adenine. This electrochemical biosensor was constructed by layer-by-layer assembling TiO 2 nanoparticles, poly( l -lysine) and graphene quantum dots (GQDs) on a glassy carbon electrode. The assembled film of TiO 2 /poly( l -lysine)/GQDs showed better conductivity and significantly increased effective surface area for the electrochemical oxidation of guanine and adenine along with a quasi-reversible electrode process. Under optimal preparation and square wave voltammetric detection conditions, the proposed electrochemical methods could simultaneously detect guanine and adenine ranging from 1.0 to 35.0 µM with detection limits of 0.56 µM for GA and 0.81 µM for AD. The limits of quantification for sample analysis were 1.90 and 2.70 µM, respectively. The developed electrochemical biosensor exhibited good reproducibility, anti-interfering properties, and stability, and could be practically applied in the simultaneous determination of guanine and adenine in calf thymus DNA, cow milk, and urine samples with acceptable precision.

Lysine-assisted ultrasonic treatment of fish myofibrillar proteins conformational rearrangement: A synergistic strategy for enhanced volatile compounds adsorption and emulsion stabilization

Yamei Wu,Zhenxing Li,Yanyan Wu,Yongqiang Zhao,Huan Xiang,Di Wang,Zhiming Hao,Yueqi Wang

This study elucidated the synergistic mechanism of lysine-assisted ultrasonic treatment in mediating sea bass myofibrillar proteins (MPs) conformational rearrangement and optimizing their flavor retention and emulsification dual functionalities. The Lysine-assisted ultrasonic treatment reduced α-helix content to 24.1 %, exposing hydrophobic groups and enhancing volatile compound adsorption. Molecular docking confirmed hexanal, octanal, and heptanal bound to MPs via hydrophobic interactions, van der Waals forces, and hydrogen bonds. Additionally, collaborative treatment decreased the particle size of MPs to 150 nm and increased the absolute value of zeta potential by 23.8 %. These changes not only inhibited MPs aggregation but also promoted covalent cross-linking and interfacial adsorption of MPs. Low-field nuclear magnetic resonance showed treated emulsion transverse relaxation time (T 2 ) rose to 727.29 ms, improving oil droplet fluidity and uniform distribution, thus boosting long-term stability. This work demonstrates potential for developing stable, flavor-controllable emulsified products.

Vancomycin-modified poly-l-lysine magnetic separation combined with multiplex polymerase chain reaction assay for efficient detection of Bacillus cereus in milk

Qianying Li,Guoyang Xie,Yutong Wang,Zoraida P. Aguilar,Hengyi Xu

ABSTRACT In this study, a new vancomycin (Van)-modified poly- l -lysine (PLL) magnetic bead (MB) technique was developed for isolation of gram-positive bacteria. The method combines magnetic separation with a multiplex PCR (mPCR) assay and gel electrophoresis for easy and rapid detection of Bacillus cereus . Vancomycin was used as a molecular ligand between the MB and the d -alanyl- d -alanine moieties on the cell wall surface of B. cereus . The PLL served as a flexible molecular tether between the MB and Van that reduced steric hindrance maintaining the biological activity of Van. The MB-PLL-Van capture nanoprobes exhibited excellent capture and isolation efficiency for B. cereus in spiked milk matrix samples without interference from the complex food matrix. The subsequent mPCR assay showed high specificity for the 4 target genes in B. cereus , the entFM , cesB , cer , and 16S rRNA genes, that were used to achieve efficient genotyping and detection. Under optimum conditions, the limit of detection reached 10 3 cfu/mL, with a dynamic range of detection at 10 3 to 10 7 cfu/mL in pure culture. Application of the MB-PLL-Van mediated mPCR assay for B. cereus in milk matrix samples achieved results similar to those of the pure culture. In addition, with a 6-h pre-enrichment of B. cereus that was spiked in milk matrix samples, the limit of detection reached 10 1 cfu/mL. The MB-PLL-Van mediated mPCR assay developed in this study could be used as a universal technology platform for the efficient enrichment and genotyping of gram-positive bacteria.
