Structural Diversity of Nucleotide Coordination Polymers of Cytidine Mono-, Di-, and Tri-Phosphates and Their Selective Recognition of Tryptophan and Tyrosine

Yaqoot khan,Yunyun Du,Li Yan,Niu Zhang,Menglei Zhang,Hongwei Ma,Hui Li

Understanding the coordination geometry of nucleotide mono-, di-, and triphosphates is pivotal for unraveling the intricate relationships between molecular structure and biological function, particularly in metal–ligand interactions and their role in biomolecular recognition. This study investigates the structure of nucleotide-metal polymers and their selective interactions with amino acids, specifically Tryptophan (Trp) and Tyrosine (Tyr). We synthesized and comprehensively analyzed five coordination polymers of cytidine-nucleotides: cytidine monophosphate (CMP), deoxycytidine monophosphate (dCMP), cytidine diphosphate (CDP), and cytidine triphosphate (CTP): {[Cu(CMP)(bpa)(H₂O)₃](CMP)·3H₂O}n (1), {[Cu₂(dCMP)(4,4′-bipy)₂(H₂O)₂]·4H₂O}n (2), {[Cu₂(CDP)₂(azpy)(H₂O)]·3H₂O}n (3), {[Cd₂(CDP)₂(bpa)₂(H₂O)₂]·8H₂O}n (4), and {[Cu(CTP)(2,2′-bipy)]·2H₂O}n (5), where (3), (4) and (5) mark the first report of CDP and CTP coordination complexes. Single crystal X-ray diffraction unveiled the structure of the polymers to be one-dimensional (1, 5) or two-dimensional (2, 3, 4). Circular dichroism (CD) spectroscopy in both the solid and solution states elucidates the chirality-driven assembly in these nucleotide-metal polymers. The selective interactions of coordination polymers with Tryptophan (Trp) and Tyrosine (Tyr) were studied using spectroscopic titrations. Experimental and computational analyses reveal distinct interactions between all five coordination polymers and the amino acids, highlighting their biosensing potential. Binding affinity variations across the polymers offer insights into nucleotide-metal coordination chemistry and suggest applications in molecular recognition and functional materials.

l-Tryptophan Schiff base cadmium(II) complexes as a new class of proteasome inhibitors and apoptosis inducers in human breast cancer cells

Nan Zhang,Yuhua Fan,Guimei Huang,Daniela Buac,Caifeng Bi,Yanfei Ma,Xinping Wang,Zhen Zhang,Xia Zhang,Q. Ping Dou

Although cadmium is a widespread environmental contaminant and human carcinogen, our studies indicate an organic cadmium complex to be a potent inhibitor of proteasomal chymotrypsin-like (CT-like) activity, further capable of inducing apoptosis in a cancer cell-specific manner. Many clinical studies suggest the use of proteasome inhibitors as potential novel anticancer agents and in the present study, we have synthesized three novel l -tryptophan-containing cadmium complexes: Cd(C 17 H 15 N 4 O 2 ) 2 ·2CH 3 OH ( 1 ) (C 17 H 15 N 4 O 2   =   2-acetylpyrazine- l -tryptophan), Cd(C 17 H 15 N 2 O 3 ) 2 ·2CH 3 OH ( 2 ) (C 17 H 15 N 2 O 3   =   5-methylfurfural- l -tryptophan) and Cd(C 16 H 12 N 2 O 2 SBr) 2 ·2CH 3 OH ( 3 ) (C 16 H 12 N 2 O 2 SBr   =   5-bromo-2-thiophenecarbaldehyde- l -tryptophan) and found that under comparable conditions cadmium complexes 1 and 2 , but not 3 , have proteasome-inhibitory activity in human breast cancer MDA-MB-231 cells. Our results suggest that l -tryptophan Schiff base cadmium complexes are potent proteasome inhibitors.

Common materials, extraordinary behavior: An ultrasensitive and enantioselective strategy for D-Tryptophan recognition based on electrochemical Au@p-L-cysteine chiral interface

Yanan Deng,Zihao Zhang,Yuanhao Pang,Xueying Zhou,Yu Wang,Yun Zhang,Yali Yuan

The poly-L-cysteine modified Au nanoparticles (Au@p-L-Cys) were constructed on electrode surface as a highly efficient chiral interface for tryptophan (Trp) enantiomers recognition via one step electropolymerization. With the aid of Cu 2+ , L-Cys residues and D-Trp target formed a sandwich complex D-Trp-Cu 2+ -L-Cys, while L-Trp was unable to form such complex due to the steric hindrance provided by the chiral interface, which was confirmed by the electrochemical and SEM results. With the introduction of ferricyanide probe, D-Trp produced significant current decrease while L-Trp produced a slight current increase, which implied the successful enantioselective recognition of Trp enantiomers (specifically D-Trp) in the true sense. This novel sensor showed a surprisingly wide linear range toward D-Trp of 6 × 10 −7  M to 1 × 10 −2  M, with a detection limit as low as 75 nM (S/N = 3). Moreover, the exclusive enantioselectivity toward D-Trp was discovered since other amino acids showed negligible interference to detection of D-Trp. The recovery of D-Trp in human serum was between 91.30 and 109.3%, which further verified the satisfying specificity and practicality of the proposed strategy. The coordination thermodynamics by UV–Vis spectroscopy and DFT simulation were also used to investigate the enantioselective mechanism. These results highlight the great potential of using Au@p-L-Cys to construct chiral interface for enantiomers recognition and hold the promise of practical application of electrochemical chiral sensors in fields like pharmaceutics and bioanalysis .

Modulation of chronic obstructive pulmonary disease progression by antioxidant metabolites from Pediococcus pentosaceus: enhancing gut probiotics abundance and the tryptophan-melatonin pathway

Yiting Liu,Longjie Li,Jing Feng,Bing Wan,Qiang Tu,Wei Cai,Fa Jin,Guiying Tang,Lígia R. Rodrigues,Xiuwei Zhang,Jia Yin,Yunlei Zhang

Chronic obstructive pulmonary disease (COPD), a condition primarily linked to oxidative stress, poses significant health burdens worldwide. Recent evidence has shed light on the association between the dysbiosis of gut microbiota and COPD, and their metabolites have emerged as potential modulators of disease progression through the intricate gut-lung axis. Here, we demonstrate the efficacy of oral administration of the probiotic Pediococcus pentosaceus SMM914 (SMM914) in delaying the progression of COPD by attenuating pulmonary oxidative stress. Specially, SMM914 induces a notable shift in the gut microbiota toward a community structure characterized by an augmented abundance of probiotics producing short-chain fatty acids and antioxidant metabolisms. Concurrently, SMM914 synthesizes L-tryptophanamide, 5-hydroxy-L-tryptophan, and 3-sulfino-L-alanine, thereby enhancing the tryptophan-melatonin pathway and elevating 6-hydroxymelatonin and hypotaurine in the lung environment. This modulation amplifies the secretion of endogenous anti-inflammatory factors, diminishes macrophage polarization toward the M1 phenotype, and ultimately mitigates the oxidative stress in mice with COPD. The demonstrated efficacy of the probiotic intervention, specifically with SMM914, not only highlights the modulation of intestine microbiota but also emphasizes the consequential impact on the intricate interplay between the gastrointestinal system and respiratory health.

A Simple Strategy for the Simultaneous Determination of Dopamine, Uric Acid, L-Tryptophan and Theophylline Based on a Carbon Dots Modified Electrode

Ziwei Wang,Rui An,Yuxuan Dai,Hongxia Luo

In this work, carbon dots (CDs) were produced by one-step ultrasound technology with glucose as the precursor. The effective synthesis of CDs was characterized by Raman spectroscopy and UV-visible spectra. The as-prepared CDs were fixed on a glassy carbon electrode (GCE) via electrochemical deposition to fabricate CDs/GCE, which simultaneously detected dopamine (DA), uric acid (UA), L-tryptophan (Trp) and theophylline (TP). With the differential pulse voltammetry technique, the oxidation peak currents of these four biomolecules were significantly enhanced on CDs/GCE compared to those on bare GCE. The potential differences of DA-UA, UA-Trp, and Trp-TP were computed as 145 mV, 381 mV, and 323 mV, respectively. The broad linear ranges were 0.5-50 μM for DA, 3-75 μM for UA, 1-65 μM for Trp and 10-200 μM for TP with limits of detection (LODs) of 0.007 μM, 0.011 μM, 0.11 μM and 0.33 μM (S/N = 3), respectively. The CD/GCE sensor had good stability and strong anti-interference ability and was applicable to detecting actual samples.

Nitric Oxide-Releasing Tryptophan-Based Poly(ester urea)s Electrospun Composite Nanofiber Mats with Antibacterial and Antibiofilm Activities for Infected Wound Healing

Mengna Li,Weiwang Qiu,Qian Wang,Na Li,Lu Liu,Xueli Wang,Jianyong Yu,Xiaoran Li,Faxue Li,Dequn Wu

Bacterial biofilms on wounds can lead to ongoing inflammation and delayed reepithelialization, which brings a heavy burden to the medical systems. Nitric oxide based treatment has attracted attention because it is a promising strategy to eliminate biofilms and heal infected wounds. Herein, a series of tryptophan-based poly(ester urea)s with good biodegradation and biocompatibility were developed for the preparation of composite mats by electrospinning. Furthermore, the mats were grafted with a nitric oxide donor (nitrosoglutathione, GSNO) to provide one type of NO loading cargo. The mats were found to have a prolonged NO release profile for 408 h with a maximum release of 1.0 μmol/L, which had a significant effect on killing bacteria and destructing biofilms. The designed mats were demonstrated to promote the growth of cells, regulate inflammatory factors, and significantly improve collagen deposition in the wound, eventually accelerating wound-size reduction. Thus, the studies presented herein provide insights into the production of NO-releasing wound dressings and support the application of full-thickness wound healing.

Preparation and evaluation of a tryptophan based hypercrosslinked porous polymer as an efficient adsorbent for pipette tip solid-phase extraction of sulfonamides

Kai Hu,Lixin Li,Pengzhao Han,Weixia Zhu,Zhenqiang Zhang,Wenjie Zhao,Shusheng Zhang

A novel tryptophan-based porous polymer is designed and synthesized via a facile one-step hypercrosslinking polymerization process, and applied as sorbent for extraction of trace sulfonamides in foodstuffs. The developed polymer has high surface area, large conjugate system, and abundant functional groups (e.g., π-π stacking, hydrogen bonding, hydrophobic and electrostatic attraction interactions), which endow it with superior affinity and high adsorption capacity for sulfonamides (16.16–59.29 mg g −1 ). The optimized SPE method is coupled with HPLC-DAD to create a sensitive and efficient protocol that provides good linearity (R 2  ≥ 0.9979), low limits of detection, satisfactory recoveries (92.5–109.5 %) and high precisions (RSDs < 8.24). In addition, the newly proposed method greatly reduces the amount of adsorbent (2.0 mg) and organic solvent (2.0 mL) used. Adsorption kinetics, isotherms, and simulation calculations studies further reveal the presence of monolayer adsorption, chemical adsorption process, and multiple interactions. Thus, this work presents a polymer capable of multiple interactions for the pretreatment of trace sulfonamides in foodstuffs.

Kynurenine, a derivative of tryptophan, inhibits progesterone biosynthesis in porcine granulosa luteal cells through AHR-mediated downregulation of GATA4, GATA6, and CEBPB

Liao Shiying,Cheng Jinhua,Zhao Weimin,Dai Chaohui,Fu Yanfeng,Li Bixia,Deng Yanfei,Li Hui

Kynurenine (KYN) is a primary tryptophan derivative found in the human body and fermented foods. Previous studies have shown that KYN is an Aryl hydrocarbon receptor (AHR) agonist and is important in regulating various physiological activities, including female reproduction. Progesterone is a vital steroid hormone that facilitates embryo implantation and maintains pregnancy. However, whether KYN affects its biosynthesis remains unclear. To gain understanding, in vitro luteinized porcine granulosa luteal (pGL) cells were treated with KYN. The results showed that KYN disrupted progesterone biosynthesis by decreasing the expression of STAR and HSD3B in pGL cells. In addition, the expression of three transcription factors of STAR and HSD3B (GATA4, GATA6, and CEBPB) decreased after KYN treatment. Furthermore, the AHR blockade results showed comparable to those of KYN treatment, and subsequent knockdown experiments confirmed these results. These findings suggest that KYN inhibits progesterone biosynthesis in pGL cells by downregulating GATA4, GATA6, and CEBPB expression through AHR. Thus, our results showed for the first time, a previously unknown connection between KYN and progesterone biosynthesis.

Limosillactobacillus reuteri C501 modulates tryptophan metabolism and alleviates obesity-induced gut-adipose tissue axis metabolic disorders through AhR and Bmal1/Pparγ signaling

Shufeng Wu,Xiang Kuang,Haizhao Song,Zhigang Liu,Ling Xiong,Bo Ren,Fang Wang,Xinchun Shen,Luanfeng Wang

Limosillactobacillus reuteri ( L. reuteri ) is known to play a beneficial role in various aspects of human health, including diabetes, inflammatory bowel disease, and neurological disorders. However, its role in regulating obesity-induced metabolic disorders remains poorly understood. We demonstrate that administering L. reuteri C501 to male mice for 10 weeks significantly alleviates high-fat diet (HFD)-induced obesity and related metabolic disorders. Treatment with L. reuteri C501 led to reduced body weight gain, improved insulin sensitivity, and attenuated inflammatory responses. Moreover, the treatment activated the aryl hydrocarbon receptor (AhR) and the circadian rhythm regulator brain and muscle ARNT-like protein 1 (Bmal1)-two key players in immune function and energy metabolism-thereby influencing lipid metabolism in HFD-fed mice. In addition, L. reuteri C501 modulated gut microbiota composition and enhanced the production of indole derivatives. Supplementation with indole-3-lactic acid (ILA), one such derivative, also improved obesity outcomes in HFD-fed mice. Notably, administering L. reuteri C501 at zeitgeber time 0 (ZT0, the beginning of the light cycle) resulted in stronger therapeutic effects, suggesting that timing of intake may critically influence its efficacy. These findings highlight L. reuteri C501 as a promising probiotic intervention for obesity, with chronobiological factors playing a key role in optimizing its metabolic benefits.

Orally-deliverable liposome-microgel complexes dynamically remodel intestinal environment to enhance probiotic ulcerative colitis therapy via TLR4 inhibition and tryptophan metabolic crosstalk

Yuanyuan Fu,Ting Wang,Xinyue Ge,Hong Wen,Yang Fei,Menghuan Li,Zhong Luo

Probiotics emerges as a promising option for ulcerative colitis (UC) treatment, but its application remains challenging due to insufficient colon-targeted delivery efficiency and survival against the inflammation-associated intestinal oxidative stress. To address these issues, here we report a supramolecular liposome-microgel complex (SLMC) incorporated with Bacillus subtilis spores (BSSs) and dexamethasone (DEX) for orally-deliverable probiotic UC therapy. Specifically, BSSs and cholesterols were conjugated with gelatin via diselenide ligation to prepare microgels, followed by supramolecular complexation with UC-targeted DEX-loaded liposome via microfluidic engineering. The orally-administered SLMC efficiently accumulated in UC-affected colonic sites to release BSSs and DEX. DEX elicited rapid anti-inflammatory effect to reduce ROS generation, which cooperated with the ROS consumption by spore germination and diselenide cleavage to orchestrate an anaerobic intestinal microenvironment, thus promoting Bacillus subtilis colonization to restore gut homeostasis and initiate anti-inflammatory microbiota-macrophage metabolic crosstalk. Indeed, in vivo analysis showed that the SLMC treatment markedly inhibited pro-inflammatory TLR4-NF-κB signaling activities in mucosal macrophages through localized DEX delivery and boosting tryptophan metabolite production, leading to robust and durable UC abolishment. This study offers a practical approach for improving UC treatment in the clinic.

Preparation and Characterization of pH-Responsive Microspheres Based on Sodium Alginate and Polyether Amine: Loading and Release of Tryptophan-Derived Cationic Antimicrobials

Yufeng Shi,Yingying Li,Nan Wu,Yudan Wang,Lijia Liu,Yawen Liu,Xuan Yuan,Junqing Li

In weakly alkaline environments (such as the human intestinal tract), the effectiveness of conventional antimicrobials may be limited; therefore, a series of cationic tryptophan fatty alcohol esterifications were designed and synthesized, and these compounds were found to possess excellent broad-spectrum antimicrobial properties and good biosafety. Among the agents, tryptophan dodecyl ester (L-Trp-Z12) exhibited the highest antimicrobial efficacy, achieving minimal inhibitory concentrations of merely 0.30 μg/mL against Staphylococcus aureus and 0.60 μg/mL against Escherichia coli, and it caused 50% hemolysis at a concentration of 167 μg/mL. Moreover, based on these cationic antimicrobial agents, a pH-responsive controlled release microsphere was designed and fabricated using SA and T403 as critical constituents. The composition, morphology, controllable release properties, and in vitro antimicrobial properties of the microspheres were systematically investigated. Microsphere SA/T403-2, which features a distinct surface topology with grooves and folds, achieved an encapsulation efficiency of 91.38% and a drug loading capacity of 4.02%. Additionally, the cumulative release rate in the first 10 h was 62% at pH 7.4 (simulated intestinal fluid), which was twice as high as at pH 1.2 (simulated gastric fluid), indicating the microspheres’ pH-responsive behavior. The SA/T403-L-Trp-Z12 microspheres demonstrated excellent antibacterial effects against S. aureus. Therefore, the SA/T403-L-Trp-Z12 microspheres prepared in this study have promising applications in the field of drug carriers and antimicrobials for targeted drug delivery to the colon, providing valuable insights into the design of pH-responsive microspheres.

Green and Controlled Synthesis of Copper-Based Tryptophan Flower-like Chiral Metal-Organic Framework for Enantioselective Recognition and Volatile Organic Compound Separation

Jinhua Xu,Minghui Tang,Jinyu Zhang,Wenmin Zhang,Shiye Xie,Lan Zhang

This study reports the synthesis and application of a flower-like chiral metal-organic framework (CMOF) based on copper ions and tryptophan (Cu(Trp)-CMOF). The Cu(Trp)-CMOF was successfully synthesized at room temperature through a facile method with hexadecyl trimethyl ammonium bromide (CTAB) as a structure-directing agent. The material exhibits a flower-like morphology with hierarchical porosity and well-defined crystallinity in the monoclinic P2 1 /c space group. Comprehensive characterization revealed that different optical configurations ((R)-type, (S)-type, and (RS)-type) could be selectively obtained by controlling the chirality of tryptophan ligands. The synthesized CMOFs demonstrated excellent performance in two key applications: colorimetric recognition of amino acid enantiomers and separation of volatile organic compounds (VOCs). When used as a chromatographic stationary phase, the CMOF significantly enhanced the separation of challenging structural isomers, particularly improving the resolution (R S ) of xylene isomers (R S from 1.03 to 2.62) and ethyltoluene isomers (R S from 4.83 to 9.23) compared to conventional columns. This work provides new insights into the design and application of CMOFs for chiral separation and recognition.
