Two-pronged strategy: A mitochondria targeting AIE photosensitizer for hydrogen sulfide detection and type I and type II photodynamic therapy

Jizhong Shi,Manshan Liang,Yiting Qiu,Jin Zhang,Shihua Wang,Heng Fang,Yongsheng Jiang,Xiaoxia Ye,Yanshu Luo,Zu-Sheng Huang,Yun-Yun Quan

Multifunctional type-I photosensitizers (PSs) for hydrogen sulfide (H 2 S) detection and photodynamic therapy (PDT) of hypoxia tumors exhibits attractive curative effect but remains a challenging task. Herein, a mitochondria targeted aggregation-induced emission (AIE) photosensitizer TSPy-SS-P was designed and synthesized, which could be used for H 2 S detection and simultaneously type I and type II PDT. TSPy-SS-P had excellent selectivity and anti-interference abilities for endogenous and exogenous H 2 S detection in tumor cells. TSPy-SS-P was able to distinguish tumor cells with high level of H 2 S from normal cells by fluorescence “turn off” response to H 2 S. In addition, TSPy-SS-P showed type Ⅰ and type Ⅱ reactive oxygen species (ROS) generation ability to effectively ablate hypoxic tumor cells. TSPy-SS-P showed mitochondria targeting capacity which could produce ROS in situ to disrupt mitochondria and promote cell apoptosis. In vivo PDT experiments showcased that TSPy-SS-P had excellent tumor retention capability, effective tumor ablation ability and good biocompatibility. This work provided a two-pronged strategy to design organelles targeted photosensitizers for H 2 S detection and effective PDT of tumors.

J-aggregate fluorescence probe constructed based on AIE units for analysis of Zn2+, Cu2+ ions in food and biological systems

Taiyi Chen,Xiaoping Lin,Peisong Li,Wei Liu,Lu Jiang,Leyuan Huang,Zhongliang Li,Ruirui Zhu,Zhi-Liang Chen,Rongying Zeng,Mengqin Liu,Wen Chen

The precise detection of metal ions is crucial in food safety, biomedical diagnostics, and environmental monitoring. Although traditional “on-off” fluorescence probes are frequently utilized, their anti-interference capability remains a limitation. This study synthesized three new copper-based fluorescent probes: PTA, PTP, and PTD. Among them, PTD exhibited unique aggregation-induced emission (AIE) properties. PTD's remarkable selectivity for Zn 2+ was due to the formation of Zn 2+ -induced J-aggregates, which resulted in a concentration-dependent fluorescence redshift from green to yellow and then to red. Additionally, PTD demonstrated a rapid and sensitive response to Cu 2+ , and it was successfully integrated into a smartphone-based platform for the semi-quantitative detection of Zn 2+ and Cu 2+ in real food samples. The effectiveness in biological applications was also validated through live-cell imaging tests. This work contributes to the creation of highly selective and interference-resistant ratiometric fluorescence sensors by leveraging the special process of target metal ion-induced J-aggregate production in AIE-based probes.

High circularly polarized luminescence based on hydrogen-bonding columnar mesophase of an AIE-active mesogen with stronger D-A structure

Hongyu Guo,Yanling Huang,Liangbin Lin,Shibing Chen,Fafu Yang

A high luminescence asymmetry factor (g lum ) and an excellent luminescence quantum yield in aggregated state are crucial factors for evaluating the application potential of circularly polarized luminescence (CPL) materials, yet achieving both simultaneously remains challenging. In this work, two AIE-active fluorogens based on two conjugated cyano-distyrylbenzenes attaching one or two cholesterol moieties with amide groups as spacers ( BiCDB-1Ch and BiCDB-2Ch ) were designed and synthesized. Both BiCDB-1Ch and BiCDB-2Ch showed the excellent CPL properties in aggregated states based on their hydrogen-bonding columnar liquid crystalline self-assembly. The absolute fluorescence quantum yield of BiCDB-1Ch with stronger D-A structure was 0.43 and the g lum value was as high as 0.30, which was the highest one among the reported columnar CPL liquid crystals. The detailed liquid crystalline self-assembly behaviour and the mechanism of efficient chirality transfer and amplification were elucidated, providing a strategic approach for developing the novel liquid crystalline materials with superior CPL performance.

A simple molecular design towards the conversion of a MCL backbone to a multifunctional emitter exhibiting polymorphism, AIE, TADF and MCL

Bin Huang,Wenbing Yu,Li Yang,Yan Li,Ning Gu

Compared with the large number of single-function materials such as aggregation-induced emission (AIE), mechanochromic luminescence (MCL), or thermally activated delayed fluorescence (TADF) emitters, multifunctional emitting materials offer more opportunities in practical applications. In this report, we provide a simple molecular design strategy towards the conversion of a MCL building block to a multifunctional emitter. Through altering the substituent sites and increasing the number of electron donors and steric hindrance on a normal MCL backbone benzo[ d , e ]benzo[4,5]imidazo[2,1- a ]isoquinolin-7-one, a novel multifunctional material 10,11-bis-(4-diphenylamino-phenyl)-benzo[ d , e ]benzo[4,5]imidazo[2,1- a ]isoquinolin-7-one (10,11-2TPA-BBI) is designed and synthesized. 10,11-2TPA-BBI exhibits simultaneous polymorphism, AIE, MCL and TADF properties. It can form four different aggregate species: yellow solid (YS) and orange solid (OS), orange flake-shaped crystal (OC), and red prism-like crystal (RC). Among them, because of the small energy gaps (Δ E ST s < 0.3 eV) between the singlet and triplet excited states, OS, OC and RC exhibit TADF properties, while YS show normal fluorescence characteristics with a large Δ E ST of 0.33 eV. OS can be reversibly transformed into YS upon external stimuli, which can be attributed to the emission switch between local excited state and charge transfer state. Crystallographic study indicates that the bulky structure and weak intermolecular interactions account for polymorphism and AIE properties. This work will provide a simple molecular design strategy for multifunctional materials.

Ziyu Huo,Luoxin Zhuang,Ge Shi,Yanjie He,Zhe Cui,Peng Fu,Minying Liu,Xiaoguang Qiao,Xinchang Pang

Multi-stimuli responsive fluorescence probe could pave the way for monitoring more complex environmental changes. Here we prepared multifunctional nanoparticle Fe 3 O 4 @SiO 2 @P(DMAEMA- co -TPEE), which displayed yolk-shell morphology with well-defined polymer brush. With superparamagnetic Fe 3 O 4 component and pH/temperature dual sensitive PDMAEMA polymer brush, the as prepared nanoparticles (YS-NPs) exhibited as multi-stimuli responsive fluorescence probe for real-time visual monitoring of environmental changes such as magnetic field, temperature and pH. Such YS-NPs could also be applied as a sensitive detector for CO 2 in aqueous solution. Notably, the solution of YS-NPs showed high colloidal stability during the environmental changes, and surface aggregation-induced emission (S-AIE) was proposed for the aggregation of TPE residue on the surface of YS-NPs.

Yu Gao,Linan Zhou,Qingqing Peng,Tingting Cui,Ziyi Guan,Fang Zhang,Li Wang,Huiyi Yang,Fang Chai

Multiple trace heavy metal pollutants present a serious threat to both the environment and ecosystems, and their coexistence also affected the detection efficiency. Therefore, multi-mode responsive optical probes with versatile application prospects for optical sensing are highly desired. Here, yellow-emissive carbon dots (Y-CDs) with aggregation-induced emission (AIE) characteristics were fabricated via a hydrothermal approach, boasting a absolute quantum yield of 13.88% and a fluorescence lifetime of 8.55 μs. Leveraging the AIE behavior and quenching effect, Y-CDs were successfully achieved the multivariate detection of Ag + , Hg 2+ separately, and further realized cascade sensing BSA, with detection limits of 5.59 μM, 7.7 nM, and 0.9 μM respectively. Subsequently, a portable smartphone-based sensing platform was developed to facilitate rapid on-site analysis, and a logic gate was designed to expedite integration into intelligent devices. Furthermore, Y-CDs demonstrated remarkable antibacterial action via in vitro antibacterial assays, displaying significant inhibitory effects against both E. coli and S. aureus , providing the possibilities for testing in the food and medical fields with high standards of hygiene permits. With their exceptional multifunctional sensing capabilities, Y-CDs were expected holding extensive promising application prospects in the realm of real-time monitoring of environmental and food safety.

Spatially Confined/Ligand-Directed Ultrabright NIR-AIE Nanoprobe for In Vitro Rapid Detection and In Situ Imaging of Bacterial Septic Arthritis

Yujian Li,Jia Shu,Sijia Lv,Leilei Qin,Xuefeng Luo,PengCheng Xiao,Huan Cai,Xin Gao,Yan Zhao,Haotian Zhou,Yaji Yang,Zhenglin Zhu,Shijia Ding,Juan Li,Rui Chen,Wei Huang

Bacterial infection is a major threat to joint surgery and can lead to amputation or death. However, advanced tools for early, rapid, and accurate identification of joint-infecting bacterial species remain limited. Here, we report a universal strategy for fabricating ultrabright near-infrared (NIR) AIEgen fluorescent probes (PPANPs) that enable lateral flow assay (LFA)-based rapid bacterial identification in joint fluid before surgery, as well as in situ imaging diagnosis and photothermal antibacterial intervention in the joint cavity. Structurally, PPANPs display a core–shell–shell architecture, in which aggregated NIR AIEgens form the fluorescent core, NIR lead sulfide (OA-PbS) forms the inner shell, and poly(maleic anhydride-alt-1-octadecene) (PMAO) forms the outer shell. This spatially confined design combines physical confinement, ligand-mediated molecular organization, and cooperative luminescence within the PbS–AIE hybrid nanoassembly, promoting compact and ordered AIE packing and thereby enhancing NIR fluorescence intensity, quantum yield, and photothermal performance compared with conventional embedding methods. s a diagnostic platform, PPANP-labeled LFA achieves 100% sensitivity for Staphylococcus aureus ( S. aureus ) identification within 10 min, outperforming standard plate culture (>96 h). Moreover, PPANPs enable rapid in situ imaging of S. aureus infection in mouse joints within 30 min and exhibit potent photothermal antibacterial activity in vitro under low-power-density laser irradiation.

Dual-Stimuli-Responsive Fluorescent Supramolecular Polymer Based on a Diselenium-Bridged Pillar[5]arene Dimer and an AIE-Active Tetraphenylethylene Guest

Yan Wang,Ming-Zhe Lv,Nan Song,Zeng-Jie Liu,Chunyu Wang,Ying-Wei Yang

We report a new strategy to construct a multi-stimuli-responsive fluorescent supramolecular polymer by the strong host–guest interactions between a diselenium bond-bridged pillar[5]arene dimer and an AIE-active tetraphenylethylene (TPE)-containing neutral guest bearing two imidazole terminal binding sites. The resulting supramolecular polymer shows a remarkable fluorescence emission decrease at low concentration. Significantly, the diselenium bonds introduced into the supramolecular polymer serve as redox-responsive building blocks. Upon addition of reductants, the supramolecular polymer depolymerized owing to the cleavage of the covalent diselenium bonds in the system. On the other hand, competitive guests such as adiponitrile, which bind strongly with pillar[5]arenes, could lead the disassembly of the polymer to oligomers without breaking any covalent bonds in the system. These two types of depolymerization approaches can both result in the fluorescence intensity recovery of the system to a certain extent, which will hopefully enrich the methodology toward the construction of smart supramolecular polymeric materials with different properties.

Enhanced photo-hypoxia-activated combination therapy traced by AIE photosensitizer and targeted by hyaluronic acid: Disulphide bond interference of detoxification barrier

Feng Ouyang,Xiaoli Zhang,Li Zhang,Yu Liu,Qi Shuai

The treatment efficacy of anticancer drugs in complex physiological environments is still restricted by multi-drug resistance. To overcome this issue, a nanodrug system of HA-SS@CuS@ZIF-8@TPZ&TBMACN (HSCZTT) that breaks through the detoxification barrier for tirapazamine (TPZ) delivery was developed in this manuscript. In addition to the photothermal effect aroused by CuS in HSCZTT, which can damage tumour cells, TBMACN with photostable fluorescence in the aggregate state can also generate sufficient reactive oxygen species (ROS) to destroy tumour cells. The continuous consumption of oxygen in PDT aggravates the hypoxic environment of tumours, which further activates the TPZ released in the acidic microenvironment of the tumour to achieve apoptosis of the tumour cells. The HSCZTT can not only target the CD44 receptor overexpressed on the surface of the cancer cell , but can also effectively consume a large amount of glutathione (GSH) through the disulphide bond-modified hyaluronic acid, which serves as a targeted disulphide bond, interfering with the detoxification barrier. Our finding presents a rational strategy to overcome multidrug resistance for the improved efficacy of anticancer drugs by the targeting of Hyaluronic acid (HA), release of the drug by the acid response of ZIF-8, breakthrough of the detoxification barrier, precise positioning of the drug release and combined treatment with phototherapy and hypoxia-activated chemotherapy.

A facile AIE fluorescent probe with large Stokes shift for the detection of Cd2+ in real water samples and living cells

Huie Jiang,Lijuan Chen,Zhijian Li,Junwei Li,Huaiqin Ma,Lulu Ning,Nihao Li,Xinhua Liu

A facile AIE fluorescent probe which exhibits a large stokes shift of 202 nm has been successfully developed. The probe can recognize Cd 2+ with high selectivity by inhibiting ESIPT process, leading to observably blue-shift and significantly enhanced fluorescence with the colour changed from green to cyan. Excitedly, the recognition of Cd 2+ is completely free from the interference of Zn 2+ and Cd 2+ could be quantitatively detected in aqueous solution with the detection limit of 1.5 × 10 −7  M. Moreover, the probe has been successfully applied to detect Cd 2+ in real water samples and image Cd 2+ in living cells.

The novel damage-reporting and self-healing coatings prepared by electrospinning technology based on AIE agents and photothermal shape memory polymer

Xue-qi Zhang,Rui Ding,Jing Lv,Yu-chen Zhang,An-lan Ji,Jie Fu,Xiao Lv,Sheng-ying Yang,Lan Yao,Xuan Liang,Jie Liu,Hai-bin Yu

The self-healing coatings for metals aroused researchers' interest due to their spontaneous repair of coating damage. In this study, intelligent damage-reporting and self-healing anti-corrosion coatings were developed using electrospun fiber as the core functional materials. The fibers exhibited core–shell structures, with the core materials being the solutions of the composite fluorescence probe RTPE. The shell materials consisted of the photothermal shape memory polymer, namely, phosphotungstic acid modified polyaniline-polyurethane composite polymer. Rhodamine Tetraphenylethylene (RTPE) demonstrated water-triggered aggregation-induced emission (AIE) effects and also exhibited fluorescence response to corrosive factors such as hydrogen ions and iron ions at the damaged area of the coatings. Under the trigger of the infrared laser, the modified polyaniline converted light energy into heat, causing the softening of epoxy and the shape memory effect of polyurethane . These processes facilitated the closure and repair of the damage, thus restoring their anti-corrosion functionality. Phosphotungstic acid played a role in enhancing the photothermal efficiency. The self-healing mechanism reinforced the corrosion resistance of the coatings. The self-indicating feature for damaged areas provided guidance for artificial repair in regions that did not self-heal satisfactorily. The synergistic and intelligent functionalities of the prepared coatings foreshadowed the significant potential applications in marine and aerospace corrosion protection fields.

Fabrication of a Luminescent Supramolecular Hydrogel Based on the AIE Strategy of Gold Nanoclusters and their Application as a Luminescence Switch

Junxiao Liu,Jin Feng,You Yu,Liandi Xu,Qi Liu,Hao Zhang,Jinglin Shen,Wei Qi

Gold nanoclusters (AuNCs) have been recognized as an emerging category of inorganic–organic hybrid luminescent materials owing to their ultrasmall size, good biocompatibility, and especially the aggregation-induced emission (AIE) property. Some works have been reported about the AIE property of AuNCs; however, the mechanism is still indistinct. In the present work, we studied the coassembly behavior of thiobarbituric acid-modified AuNCs (TBA-AuNCs) with Zn2+. The coassembly system could not only enhance the emission of AuNCs but also induce the gelation along with the morphology transformation from nanofibers to micron-sized bowknot-like aggregates. Various methods were used to characterize the properties of a hydrogel such as photoluminescent (PL) spectra, morphology, lifetime, and intermolecular interaction. Significantly, time-/temperature-dependent PL spectra as well as morphological evolution monitoring were implemented to analyze the structure–emission property relationship of TBA-AuNCs assembly systems. It is verified that the π–π stacking of ligands, ligand-to-metal charge transition (LMCT), and the orderliness degree of aggregates contribute uniquely to the emission energy/intensity of TBA-AuNCs, which is significant for the comprehension of the AIE nature of nanoclusters. Besides, the gelation and photoluminescence of TBA-AuNCs are sensitive to heating and ethylenediaminetetraacetic acid disodium salt (EDTA-2Na), making the hydrogels promising candidates as smart luminescent switches. This work is of interest for not only providing a versatile methodology for lighting up AuNCs through gelation but also pointing out a way to understand and regulate the emission of AuNCs.
