Photothermal and Acid-Responsive Fucoidan-CuS Bubble Pump Microneedles for Combined CDT/PTT/CT Treatment of Melanoma

Jiaojiao Tao,Bingjie Wang,Yu Dong,XiangYan Chen,Shuang Li,Tianze Jiang,Xia Zhao

Transdermal cancer therapy faces great challenges in clinical practice due to the low drug transdermal efficiency and the unsatisfactory effect of monotherapy. Herein, we develop a novel bubble pump microneedle system (BPMN-CuS/DOX) by embedding sodium bicarbonate (NaHCO3) into hyaluronic acid microneedles (MNs) loaded with fucoidan-based copper sulfide nanoparticles (Fuc-CuS NPs) and doxorubicin (DOX). BPMN-CuS/DOX can generate CO2 bubbles triggered by an acidic tumor microenvironment for deep and rapid intradermal drug delivery. Fuc-CuS NPs exhibit excellent photothermal effect and Fenton-like catalytic activity, producing more reactive oxygen species (ROS) by photothermal therapy (PTT) and chemodynamic therapy (CDT), which enhances the antitumor efficacy of DOX and reduces the dosage of its chemotherapy (CT). Simultaneously, DOX increases intracellular hydrogen peroxide (H2O2) supplementation and promotes the sustained production of ROS. BPMN-CuS/DOX significantly inhibits melanoma both in vitro and in vivo by the combination of CDT, PTT, and CT. In short, our study significantly enhances the effectiveness of transdermal drug delivery by constructing BPMNs and provides a promising novel strategy for transdermal cancer treatment with multiple therapies.

S-Nitrosylated CuS Hybrid Hydrogel Patches with Robust Antibacterial and Repair-Promoting Activity for Infected Wound Healing

Pengpeng Jia,Yu Zou,Jiang Jiang

Development of wound dressing with robust antibacterial and repair-promoting activity has always been an urgent biomedical task during the past years. The therapeutic effect of current hydrogel dressings containing single bioactive agent like nanoparticle or gas is still unsatisfactory for the treatment of infected wound. Herein, a CuS/NO co-loaded hydrogel (CuS/NO Gel) is proposed, which is constructed by sequential polymerization, reduction, and S-nitrosylation of CuS hybrid hydrogel with disulfide bonds. These CuS/NO Gel patches show good mechanical stability, high photothermal activity and excellent biocompatibility. When being applied to treat infected wound, CuS/NO Gel can not only eliminate infection effectively by the synergistic effect of mild photothermal heating and boosted NO release in infection phase, but also promote vascularization and collagen deposition due to the synchronous supply of Cu ion nutrients and low concentration NO signaling molecules in wound repair phase. Compared to hydrogel dressings with individual active agent (CuS Gel or NO Gel), CuS/NO Gel exhibits better antibacterial and repair-promoting activity both in vitro and in vivo. Therefore, this CuS/NO Gel holds great promise in the future clinical treatment against infected wound.

Yiyang Zhao,Yonghui Shao,Hao Chen,Xinwen Luo,Xiaodi Liu

Herein, well-defined hollow CuS microspheres assembled from nanosheets were successfully synthesized through a facile solvothermal method. Hollow CuS microspheres have an average diameter of 1.5 μm; moreover, the primary CuS nanosheets have an ultrathin thickness of about 10 nm and are bound by {0001} polar facets. When used as anodes for lithium-ion batteries (LIBs), hollow CuS microspheres exhibit excellent electrochemical properties, including a large discharge capacity (610.1 mAh g−1at 0.5 C), an excellent rate capability (207.6 and 143.4 mAh g−1at 1 and 5 C), and a superior cyclic stability (196.3 mAh g−1at 1 C after 500 cycles). When used as photocatalysts for Rhodamine B (RhB), hollow CuS microspheres can degrade more than 99% of the initial RhB within 21 min. These excellent Li-ion storage properties and photocatalytical performances are attributed to their unique hierarchical hollow structure. Keywords:CuS;hollow microsphere;nanosheet;lithium-ion batteries;photocatalyst

Construction of novel CdS@CuS/g-C3N4 heterojunctions for efficient visible light-driven photo-Fenton degradation performance

Rongping Xu,Jinlong Li,Guozhe Sui,Yan Zhuang,Dongxuan Guo,Ze Luo,Shuang Liang,Hong Yao,Chao Wang

The construction of heterojunctions is considered as an effective strategy to boost catalytic degradation activity of photo-Fenton catalyst. Herein, a series of CdS@CuS/g-C 3 N 4 (CC-x/CN) heterojunctions have been successfully prepared by hydrothermal and cation exchange process, which have been applied for the removal of methyl orange (MO) via photo-Fenton process. The results demonstrate that the exchange of Cu 2+ for Cd 2+ on g-C 3 N 4 could inhibit the aggregation of CdS@CuS and increase the light absorption range and the active sites. Under visible light illumination, CC-10/CN (molar ratio of CuS to CdS is 10%) exhibits the optimal photo-Fenton-like catalytic performance with the degradation efficiency of 96% towards MO (40 mg L −1 ) and the total organic carbon removal rate of 75% within 35 min illumination. The apparent reaction rate constant of CC-10/CN (0.088 min −1 ) is approximately 22 and 6.29 times higher than those of pure g-C 3 N 4 and CdS@CuS (molar ratio of CuS to CdS is 10%), respectively. The enhanced photo-Fenton catalytic activity of CC-x/CN is mainly ascribed to the formation of the photo-Fenton effect between CC-10 and g-C 3 N 4 , which promotes the conversion and recycling of Cu 2+ and Cu + . Meanwhile, the successful formation of the heterojunctions reduces the recombination rate of carriers. In addition, the cycling experiments demonstrate that CC-10/CN possesses favorable stability and recoverability. Overall, the well-designed CC-x/CN heterojunctions reveal desirable potential application in the effective treatment of sewage via photo-Fenton process.

Evaluating the therapeutic efficacy of radiolabeled BSA@CuS nanoparticle-induced radio-photothermal therapy against anaplastic thyroid cancer

Chunmei Zhang,Jinyan Chai,Qiang Jia,Jian Tan,Zhaowei Meng,Ning Li,Menghui Yuan

Bovine serum albumin (BSA) has been employed as a mild biological template in nanoscale particles. Copper sulfide (CuS) has been used for photothermal therapy (PTT) in several studies. In this study, we aimed to synthesize the 131 I-labeled BSA-modified CuS nanoparticles ( 131 I-BSA@CuS), with attributes of both radiotherapy and PTT, as a therapeutic agent against anaplastic thyroid carcinoma (ATC). BSA@CuS nanoparticles were prepared using the solvothermal reaction and then labeled with Na 131 I by the chloramine-T method. The products were characterized and their cytotoxicity was investigated in vitro and in vivo. The therapeutic efficacy of 131 I-BSA@CuS was evaluated in ARO cell (an ATC cell line) subcutaneous tumors. The nanoparticles showed good biocompatibility and low toxicity in vitro and in vivo. BSA@CuS rapidly and effectively converted the light energy from an 808 nm laser into thermal energy with a conversion efficiency of 28.07%. SPECT/CT imaging demonstrated that the accumulation of radioactivity peaked within 24 hr and resided in the tumors for 5 days post intratumoral injection. In vivo assays indicated that, compared to monotherapy, the synthesized nanoparticles employing both PTT and radiotherapy possess better therapeutic efficacy against tumors. The synthesized nanomaterial showed uniform dispersion, good stability and aqueous solubility, excellent photothermal properties, and long-term retention in ATC. Hence, combined radiotherapy and PTT can significantly inhibit tumor growth compared to monotherapy, and can be applied in clinical settings.

Fabrication of CuS-modified inverse opal g-C3N4 photocatalyst with enhanced performance of photocatalytic reduction of CO2

Xiaoyue Huang,Yan Hu,Liang Zhou,Juying Lei,Lingzhi Wang,Jinlong Zhang

As a greenhouse gas, CO 2 is also a rich carbon resource. Using sunlight to reduce CO 2 to usable substances through photocatalysis can alleviate the greenhouse effect and realize energy recycling. Herein, CuS nanoparticles are grown in situ on the surface of inverse opal g-C 3 N 4 by a one-step hydrothermal method. The inverse opal structure can increase the specific surface area of the material, and CuS can be used as the cocatalyst to accept photo-generated electrons on the conduction band of g-C 3 N 4 , thereby promoting the separation and transfer of photo-generated charges. And the interfacial charge transfer mechanism of CuS further improves the separation efficiency of photogenerated electrons and holes. The photocatalytic reduction of CO 2 activity of the prepared catalyst is greatly improved. The CO yield reaches 13.24 μmol g −1 h −1 . A new strategy is provided to develop artificial photosynthesis for reduction of CO 2 .

Cascade targeting tumor mitochondria with CuS nanoparticles for enhanced photothermal therapy in the second near-infrared window

Haiyan Wu,Pengpeng Jia,Yu Zou,Jiang Jiang

Photothermal therapy, assisted by local heat generation using photothermal nanoparticles (NPs), is an emerging strategy to treat tumors noninvasively. To improve treatment outcomes and to alleviate potential side effects on normal tissue cells, utilizing the optically transparent second near-infrared (NIR-II) window and actively targeting tumors are critical. Considering that mitochondria are heat sensitive and play an important role in the up-regulation of metabolic activity in tumor cells, herein we report a cascade targeting scheme that enables active photothermal ablation of tumor mitochondria. First, NIR-II absorbing CuS NPs were surface modified with the mitochondria targeting moiety (3-carboxypropyl) triphenylphosphonium bromide (TPP) and then shielded with CD44 targeting hyaluronic acid, which will only expose TPP upon reaching the tumor sites. This allowed over 90% CuS NP enrichment at tumor mitochondria, and as a result, significantly improved tumor cell photothermal ablation was observed at the cellular level. An in vivo study demonstrated enhanced tumor uptake and improved tumor growth suppression by using these cascade targeting CuS NPs as NIR-II photothermal agents.

Injectable Self-Healing Hydrogels Containing CuS Nanoparticles with Abilities of Hemostasis, Antibacterial activity, and Promoting Wound Healing

Yue Kong,Zishuo Hou,Liangqin Zhou,Panfeng Zhang,Yaowen Ouyang,Peiwen Wang,Yuanwei Chen,Xianglin Luo

Injectable self-healing hydrogels containing functional nanoparticles (NPs) have attracted much attention in many fields of biomedicine. A series of injectable self-healing hydrogels containing PEGylation CuS NPs based on N-carboxyethyl chitosan (CEC) and oxidized sodium alginate (OA) were developed by taking advantages of the unique functions of CuS NPs and chitosan, referred to as CuS NP hydrogels or CEC–OAm–CuSn, where “m” stands for the concentration percentage of the added OA solution (w/v) and “n” represents the molar concentration of CuS NPs in the hydrogels. The physical properties of CuS NP hydrogels, syringeability, rapid self-repair ability, and photothermal performance were systematically investigated. The multiple functions for CuS NP hydrogels requested in the skin healing process were explored. The results showed that CuS NP hydrogels had not only adjustable physical properties and good injectable self-healing characteristics but also excellent functionalities, concurrently including hemostatic ability, bacteria killing capability, and cell migration and proliferation promotion. In vivo wound healing and histomorphological examinations of immunofluorescence staining in a mouse full-thickness wound model demonstrated good acceleration effects of these hydrogels for infected wound healing. Therefore, these injectable self-healing CuS NP hydrogels which possess the abilities of hemostasis, antibacterial activity, and infected-wound healing promotion exhibit great potential as in situ wound dressings.

Fabrication of Folic Acid-Decorated Hollow ZIF-8/Au/CuS Nanocomposites for Enhanced and Selective Anticancer Therapy

Le Gao,Zhitao Wu,Abdul-Rauf Ibrahim,Shu-Feng Zhou,Guowu Zhan

Hollow nanomaterials have been used as an attractive platform for the integration of multiple bioactive components for effective anticancer therapy. Herein, we report a novel and facile strategy for the fabrication of hollow and monodispersed zeolitic imidazolate framework-8 (ZIF-8) by the self-template method with folic acid (FA) as a bioetcher. Gold nanocluster and folic acid were critical for the formation of the hollow ZIF-8 (thickness of 38 nm) during solvothermal synthesis. By integrating CuS nanoparticles (size of 4.9 nm), the resultant quadruple ZIF-8/Au/CuS/FA nanocomposites (denoted as FACZ) exhibited effective anticancer activities on FA receptor-positive MCF-7 and HepG-2 tumor cells but a weak killing effect on HCMEC/D3 cells. Folic acid molecules were conjugated to the external surface of FACZ, which simultaneously offered an excellent tumor-targeting ability and fluorescence imaging property. Although the photothermal therapy caused by CuS was not so obvious due to partial reduction, the nanosized FACZ after cellular uptake was able to release Cu(I) to enable chemodynamic therapy. This catalytically decomposed H2O2 to produce highly reactive oxygen species via the Fenton-like reaction as determined by the extracellular and intracellular hydroxyl radical. Our work offers a simple route for the fabrication of hollow ZIF-8 nanocomposite with active and selective anticancer activity. This is envisaged to have great potentials in biomedical applications.

Construction of hollow TiO2/CuS nanoboxes for boosting full-spectrum driven photocatalytic hydrogen evolution and environmental remediation

Bo Yu,Fanming Meng,Ting Zhou,Aoyu Fan,Muhammad Wasim Khan,Hao Wu,Xingbin Liu

The plasma semiconductor Cu 2-x S has disclosed enormous potential in the field of broad-spectrum driven photocatalyst design due to its unique LSPR effect in the NIR region. Nevertheless, ineffective carrier separation and severe photocorrosion hinder the further functional applications of this promising material. Here, the TiO 2 @CuS double-shell nanoboxes were successfully constructed via a multistep control strategy involving template participation, and were firstly employed to broad-spectrum photocatalytic hydrogen evolution and pollutant elimination. The hollow core-shell nanocubes with LSPR effect have prominent advantages in broad-spectrum light response, improved photocharge separation and effective photocorrosion inhibition. Besides, the heterogeneous cavity structure also provides a larger specific surface area and ameliorates the light efficiency. Consequently, the optimized TiO 2 @CuS double-shell nanoboxes exhibit superior activity for H 2 production under full-spectrum/NIR light irradiation (2467 and 173 μmol g −1  h −1 , respectively), and maintain high activity in the photodegradation of stubborn pollutants. This work will motivate hollow photocatalysts designed to effectively utilize the spectrum and protect the sulfide core.

Ag+ ions-mediated fluorescence “off-on” sensing platform for the detection of glutathione based on CuS quantum dots

Jinqiu Xu,Haoyu Chen,Jing Liu,Jianru Tang,Xiaohua Zhu,Sheng Yang,Shu Huang,Youyu Zhang,Meiling Liu

The development of analytical methods capable of simultaneously detecting multiple components in biological samples holds significant potential for advancing disease diagnosis. However, constructing optical platforms for continuous detection remains challenging due to limitations such as insufficient selectivity, specificity, response speed, and sensitivity. Herein, we developed a facile and efficient microwave-assisted method to rapidly synthesize CuS quantum dots (QDs) and constructed a fluorescence-based sensing platform for detecting Ag + and glutathione (GSH). The fluorescence emission of CuS QDs at 500 nm was selectively quenched upon interaction with Ag + , exhibiting a linear correlation with Ag + concentrations ranging from 0.3 to 300 μM. Notably, the quenched fluorescence could be restored by introducing GSH, accompanied by a slight redshift in the emission wavelength. This Ag + -mediated “off-on” fluorescence sensing mechanism significantly enhanced the sensitivity for GSH detection. This platform demonstrated high selectivity towards both Ag + and GSH, enabling their quantitative determination in complex matrices, including tap water, human serum, and cell lysate. This work not only presents a novel synthetic approach for CuS QDs but also achieves successive detection of Ag + and GSH without requiring complex probe modifications or the integration of multiple nanomaterials, representing a simplified yet highly effective design compared to more intricate systems. The proposed platform serves as a versatile measurement tool applicable across various fields, from environmental contaminant monitoring to the detection of key biological thiols, demonstrating a generalizable strategy for diverse analytical scenarios in both environmental and clinical contexts.

Engineering of 2D Ultrathin CuS-PMA Heterostructures for Enhanced NIR-II Photothermal Therapy Facilitating Oxidative Stress and Immunotherapy

Dong Wang,Feng Yuan,Xuliang Deng,Haoyang Li,Siyang Nie,Qingda Liu,Xun Wang

Therapy based on heat and reactive oxygen species (ROS) generation have attracted increasing attention, while the weak photothermal ability and low ROS production have severely limited the efficiency of the tumor treatment. Here, 2D ultrathin CuS-phosphomolybdic acid (PMA) nanosheets are synthesized at sub-nanoscale. Typically, the improved second near-infrared light (NIR-II) light absorption, small bandgap, fast hot carrier response, abundant oxygen vacancies, and the electron delocalization between closely connected PMA and CuS units in sub-nanoscale result in the notably enhanced NIR-II photothermal therapy (PTT) performance of CuS-PMA. The reversible redox of Mo active sites in PMA provides basis for glutathione (GSH) depletion while triggering chemodynamic therapy (CDT) in the tumor microenvironment (TME), and the heat produced by the PTT greatly promotes the CDT and GSH depletion, where CuS-PMA presents sevenfold more ROS generation and fourfold more GSH depletion under PTT. In addition, the ROS generation/GSH depletion can downregulate heat shock proteins, weakening the heat resistance of tumor cells and thereby amplifying the killing effect of PTT. Moreover, the combination of antiprogrammed death-ligand 1 (αPD-L1) with the treatment of CuS-PMA promote the immunotherapy, resulting in the suppression of untreated abscopal tumors.
