Development of copper vacancy defects in a silver-doped CuS nanoplatform for high-efficiency photothermal–chemodynamic synergistic antitumor therapy

Zizhen Qin,Mengjun Qiu,Quanyuan Zhang,Shenli Yang,Guangfu Liao,Zhifan Xiong,Zushun Xu

The combination of chemodynamic and photothermal materials can not only improve the therapeutic effect of chemodynamic therapy (CDT) by thermal stimulation, but also play a synergistic therapeutic role. Benefitting from the strong near-infrared absorption ability, copper sulfide (CuS) nanomaterials are widely used in photothermal therapy. However, due to the harsh preparation conditions, low photothermal efficiency and poor biocompatibility, further biomedical application is limited. In this work, silver-doped copper sulfide nanoparticles (BSA-Ag:CuS) were synthesized using a biomineralization strategy using bovine serum albumin (BSA) as a template and stabilizer. Silver doping greatly improved the near-infrared absorption and photothermal efficiency of CuS nanoparticles, which can be used for 1064 nm laser-guided photothermal therapy (PTT). Meanwhile, BSA-Ag:CuS nanoparticles had a synergistic therapeutic effect with CDT and thus showed excellent antitumor performance. In vivo and in vitro biological experiments have shown that BSA-Ag:CuS nanoparticles have good stability, low toxicity, good biocompatibility and strong antitumor ability, and are promising as antitumor agents for future clinical cancer treatment.

Iron Doped in the Subsurface of CuS Nanosheets by Interionic Redox: Highly Efficient Electrocatalysts toward the Oxygen Evolution Reaction

Jing Chen,Mingzheng Gu,Shoujie Liu,Tian Sheng,Xiaojun Zhang

Modifying the electronic structure of electrocatalysts by metal doping is favorable to their electrocatalytic activity. Herein, by a facile one-pot redox process of Fe(III) and Cu(I), Fe(II) was successfully doped into the subsurface of CuS nanosheets (NSs) for the first time to obtain a novel electrocatalyst (Fesub-CuS NSs) that possesses not only subtle lattice defects but also an atomic-level coupled nanointerface, greatly enhancing the oxygen evolution reaction (OER) performances. Meanwhile, Fe(II) and Fe(III) coexisting in Fesub-CuS nanosheets are favorable to OER through valence regulation. As expected, by simultaneously controlling the abovementioned three factors to optimize Fesub-CuS nanosheets, they display a lower overpotential of 252 mV at a current density of 20 mA cm–2 for OER, better than 389 mV for pristine CuS nanosheets. This discovery furnishes low-cost and efficient Cu-based electrocatalysts by metal doping. Density functional theory (DFT) calculations further verify that Fesub-CuS(100) is thermodynamically stable and is more active for OER. This research provides a strategy for the atomic-scale engineering of nanocatalysts and also sheds light on the design of novel and efficient electrocatalysts.

Insight into CuX (CuO, Cu2O, and CuS) for enhanced performance of CuX/g-C3N4 nanocomposites-based acetaminophen electrochemical sensors

Yao Chen,Yanyan Zhu,Yuhui Zhao,Jing Wang,Meng Li

The complexation of metals and metal compounds with organic skeletons into nanohybrid composites attracts attention in electrochemical applications due to the improved charge transfer pathways, which enhance the electrocatalytic activity. In this study, CuO, Cu 2 O, and CuS were anchored on g-C 3 N 4 sheets to construct electrochemical sensors for the detection of acetaminophen (AP). The CuX/g-C 3 N 4 nanohybrid composites were analyzed via transmission electron microscopy, infrared spectroscopy, X-ray diffraction, and X-ray photoelectron spectroscopy. Among the CuX/g-C 3 N 4 nanocomposites, CuS/g-C 3 N 4 displays a higher electrochemical activity. The electrochemical CuS/g-C 3 N 4 , CuO/g-C 3 N 4 , and Cu 2 O/g-C 3 N 4 sensors have wide linear ranges of 5 to 500 µM with a low detection limit (LOD) of 0.26 µM, 5 to 300 µM with a LOD of 0.32 µM, and 5 to 250 µM with a LOD of 0.47 µM, respectively. The better performance of the CuS/g-C 3 N 4 sensor is probably because of its significantly higher catalytic rate constant with respect to the oxidation of AP in electrochemical kinetics compared with those of the two other sensors, which leads to the enhanced electrocatalytic oxidation of AP. The CuX/g-C 3 N 4 -based electrochemical sensors display an excellent selectivity, repeatability, reproducibility, and stability. These features suggest that CuX/g-C 3 N 4 based electrochemical sensors can be sensitively detected of AP.

Catalytic degradation of ciprofloxacin by magnetic CuS/Fe2O3/Mn2O3 nanocomposite activated peroxymonosulfate: Influence factors, degradation pathways and reaction mechanism

Yan Huang,Li-chao Nengzi,Xinyi Zhang,Jianfeng Gou,Yingjie Gao,Guixian Zhu,Qingfeng Cheng,Xiuwen Cheng

In the study, CuS/Fe 2 O 3 /Mn 2 O 3 magnetic nanocomposite was successfully synthesized and the properties were investigated via a series of characterization. Subsequently, the magnetic nanocomposite was applied to activate peroxymonosulfate (PMS) for ciprofloxacin (CIP) degradation. The results showed that the CuS/Fe 2 O 3 /Mn 2 O 3 magnetic nanocomposite possessed higher catalytic performance for ciprofloxacin degradation than bare CuS and Fe 2 O 3 /Mn 2 O 3 composite. The degradation process of CIP was suitable for the pseudo-first-order kinetic model and the highest rate was reached 0.10083 min −1 . And under the optimized conditions (catalyst = 0.6 g·L −1 , PMS = 0.6 g·L −1 , pH = 5.84, C 0  = 0.2 g·L −1 and t = 120 min), the efficiency of 88% and 48.6% were corresponding to degradation and mineralization of ciprofloxacin, respectively. In addition, the key factors of degradation process such as catalyst dosage, PMS concentration, initial pH value and common anions were investigated. And the results indicated that the degradation efficiency of ciprofloxacin was reduced, which affected by the common anions in order of Cl −  > SO 4 2−  > NO 3 −  > HCO 3 −  > Humic acid. Besides, several intermediates of ciprofloxacin were detected and the plausible degradation pathways were proposed. In addition, the underlying reaction mechanism was proposed. Meanwhile, the results of free radical quenching experiment and electron paramagnetic resonance (EPR) spectra indicated that both OH and SO 4 − radicals were existed in the reaction system and the OH radicals had a conquer role in the degradation progress. Moreover, the stability and reusability of CuS/Fe 2 O 3 /Mn 2 O 3 magnetic nanocomposite were evaluated, which the degradation efficiency still reached 72% after five times used. Ultimately, the as-constructed CuS/Fe 2 O 3 /Mn 2 O 3 magnetic nanocomposite might be a candidate to the removal of refractory pollutants in environmental remediation.

Construction of immobilized 0D/1D heterostructure photocatalyst Au/CuS/CdS/TiO2 NBs with enhanced photocatalytic activity towards moxifloxacin degradation

Qinghua Chen,Mengmeng Zhang,Jingying Li,Guodong Zhang,Yanjun Xin,Chao Chai

Strong absorption in a wide wavelength range and effective charge separation are main factors for improving photocatalytic activity of semiconductor photocatalysts. Herein, immobilized zero-dimensional/one-dimensional (0D/1D) Au/CuS/CdS/TiO 2 nanobelts (Au/CuS/CdS/TiO 2 NBs) heterostructure photocatalyst with innovative morphology and appropriate band-gap configuration was synthesized for the first time. For this construction, TiO 2 NBs provide the 1D structure for a large superficial area and fast electron mobility, 0D CdS and CuS nanoparticles are the visible-light responsive nanomaterials, and Au nanoparticles serve as the surface plasmon resonance effect and electron transfer. The Au/CuS/CdS/TiO 2 NBs showed improved photocatalytic performance towards moxifloxacin (MOX) degradation, which can be attributed to the promoted photoabsorption contribution and enhanced separation and transfer of photoexcited electrons and holes. The morphology, constitution, optical and electrochemical properties of the prepared photocatalyst were investigated comprehensively. Moreover, the photocatalytic degradation mechanism and degradation by-products of moxifloxacin were studied. Results showed that the OH, O 2 − and h + are the main chemical oxidation species responsible for the degradation of MOX. Eight different photocatalytic degradation products were detected. The stability assessment showed that photodissolution plays the primary role in the photocatalytic performance decrease of Au/CuS/CdS/TiO 2 NBs. This work gives a promising strategy to rational design efficient hybrid metal-semiconductor photocatalysts for solar energy conversion.

A novel polydopamine-loaded copper sulfide (CuS@PDA) for activating H2O2 to eliminate tetracycline via 1O2 dominated oxidation pathway

Zhou Shi,Chenxi He,Hao Huang,Xile Huang,Tong Hu,Yijia He,Dazhi Yang,Simeng Xia,Haojie Zhang,Lin Deng

The frequently detected antibiotics in aquatic environments can induce antibiotic-resistance genes, thereby posing significant risks to both ecosystems and human health. Thus, it is imperative to remove antibiotics from water environments. We constructed a novel polydopamine-loaded copper sulfide (CuS@PDA) through a simple hydrothermal method to activate H 2 O 2 to degrade tetracycline (TC). Compared to CuS/H 2 O 2 , the CuS@PDA/H 2 O 2 system not only achieved efficient TC removal with kinetic rate constant of 0.20 min −1 , but also showed much lower Cu 2+ ions leaching (3.81 mg/L from CuS vs. 0.21 mg/L from CuS@PDA). Besides, CuS@PDA exhibited remarkable recyclability with 93 % removal in the fifth consecutive cycle. Mechanisms analysis revealed that Cu and S contributed to the H 2 O 2 activation and S promoted the conversion of Cu(II) to Cu(I), beneficial for the production of reactive oxygen species. 1 O 2 was found to play the dominant role in the degradation of TC on the basis of quenching tests and electron paramagnetic resonance (EPR) analysis. PDA in CuS@PDA composites facilitated easier complexation with H 2 O 2 and conferred stronger oxidation capability. Lastly, the TC degradation pathway by CuS@PDA/H 2 O 2 was proposed, and the ecotoxicity of its degradation intermediates was estimated. In conclusion, this work demonstrates that the use of PDA can not only improve the catalytic activity and recyclability of CuS towards H 2 O 2 activation for organic pollutants degradation, but also greatly mitigate copper ions leaching.

Fe3O4@mSiO2-FA-CuS-PEG nanocomposites for magnetic resonance imaging and targeted chemo-photothermal synergistic therapy of cancer cells

Zhifang Gao,Xijian Liu,Guoying Deng,Feng Zhou,Lijuan Zhang,Qian Wang,Jie Lu

In this work, a new multifunctional nanoplatform (Fe3O4@mSiO2-FA-CuS-PEG nanocomposite) for magnetic resonance imaging (MRI) and targeted chemo-photothermal therapy, was firstly fabricated on the basis of magnetic mesoporous silica nanoparticles (Fe3O4@mSiO2), on which folic acid (FA) was grafted as the targeting reagent, CuS nanocrystals were attached as the photothermal agent, and polyethylene glycol (PEG) was coupled to improve biocompatibility. The characterization results demonstrated that the fabricated Fe3O4@mSiO2-FA-CuS-PEG nanocomposites not only showed strong magnetism and excellent MRI performance, but also had a high doxorubicin (DOX, an anticancer drug) loading capacity (22.1%). The loaded DOX can be sustainably released, which was apt to be controlled by pH adjustment and near infrared (NIR) laser irradiation. More importantly, targeted delivery of the DOX-loaded Fe3O4@mSiO2-FA-CuS-PEG nanocomposites could be accomplished in HeLa cells via the receptor-mediated endocytosis pathway, and this exhibited synergistic effect of chemotherapy and photothermal therapy against HeLa cells under irradiation with a 915 nm laser. Therefore, the fabricated multifunctional Fe3O4@mSiO2-FA-CuS-PEG nanocomposite has a great potential in image-guided therapy of cancers.

CuS@mSiO2-PEG core–shell nanoparticles as a NIR light responsive drug delivery nanoplatform for efficient chemo-photothermal therapy

Xijian Liu,Qilong Ren,Fanfan Fu,Rujia Zou,Qian Wang,Guobing Xin,Zhiyin Xiao,Xiaojuan Huang,Qian Liu,Junqing Hu

We report a facile and low-cost approach to design a difunctional nanoplatform (CuS@mSiO2-PEG) as a near-infrared (NIR) light responsive drug delivery system for efficient chemo-photothermal therapy. The nanoplatform demonstrated good biocompatibility and colloidal stability, as well as high loading capacity for the anticancer drug (26.5 wt% for doxorubicin (DOX)). The CuS nanocrystals (core) within these CuS@mSiO2-PEG core–shell nanoparticles can effectively absorb and convert NIR light to fatal heat under NIR light irradiation for photothermal therapy, and the release of DOX from the mesoporous silica (shell) can be triggered by pH and NIR light for chemotherapy. When the CuS@mSiO2-PEG/DOX nanocomposites were irradiated by 980 nm light, both chemotherapy and photothermal therapy were simultaneously driven, resulting in a synergistic effect for killing cancer cells. Importantly, compared with chemotherapy or photothermal treatment alone, the combined therapy significantly improved the therapeutic efficacy.

Green synthesis of Bio-CuS quantum dots by sulfate reducing bacteria for solid-phase photocatalytic degradation of polyethylene film

Jian Cheng,Xu Wang,Gongchu Shi,Shihao Zhang,Yanyu Zhou,Yanchao Cui,Jia Wang,Baoping Xin

In this study, CuS quantum dots (QDs) synthesized through biological method were used as photocatalysts for the photocatalytic degradation of polyethylene (PE) plastics. Bio-CuS QDs were synthesized through a green process using sulfate reducing bacteria, while Che-CuS QDs were prepared by chemical precipitation for comparison. The crystal structure, surface morphology, microstructure and optical properties of the samples were detected by XRD, SEM, TEM and UV–Vis DRS. The green synthesis mechanism of Bio-CuS was inferred by analyzing changes in amino acid concentration and the three-dimensional fluorescence spectra of extracellular polymeric substances before and after synthesis. The photocatalytic degradation of PE films by Che-CuS and Bio-CuS was investigated. The weight loss rates of PE+Che-CuS and PE+Bio-CuS were 9.1 % and 12.8 % after 14 d irradiation, respectively, while pure PE was only 1.7 %. Furthermore, SEM, UV–Vis DRS, contact angle, TGA, GPC and FTIR were adopted to characterize the PE film. The results revealed that PE+Che-CuS and PE+Bio-CuS underwent major changes in morphology, transparency, hydrophilicity, thermal stability, molecular weight and functional groups compared with PE, with more pronounced changes observed for PE+Bio-CuS. The quenching experiments of active species showed that h + and·OH were critical to the PE degradation. The intermediate products of PE degradation were analyzed by GC–MS, and a possible degradation pathway was proposed. In addition, the possible mechanism of photocatalytic degradation of PE films by CuS QDs was proposed. This work provides a green remediation approach to addressing the growing plastic pollution in the environment.

Green hydrogen and acetaldehyde production via photo-induced thermal dehydrogenation of bioethanol over a highly dispersed CuS/ TiO2 catalyst

Shuai Liu,Yang Meng,Chunlin Luo,Huiwen Zhu,Yunshen Song,Honglei Zhang,Tao Wu

The emergence of global warming issues has triggered the interest in producing carbon-neutral energy sources and fossil-based chemicals from affordable and renewable feedstocks (e.g., bio-ethanol). Herein, the synthesis of highly dispersed CuS/TiO 2 photothermal catalysts is reported for simultaneously converting ethanol into hydrogen and acetaldehyde under simulated sunlight. The catalyst achieved a hydrogen evolution rate of 51.61 mmol g −1  h −1 and an acetaldehyde production rate of 48.63 mmol g −1  h −1 , with a maximal acetaldehyde selectivity of 97% among carbon-containing products. Compared to pristine TiO 2 , the introduction of plasmonic CuS largely extended the light adsorption range and enhanced the photothermal performance, whilst the formation of heterojunction structure improved the charge transfer and separation efficiency. The density functional theory calculation results demonstrate that CuS/TiO 2 composite interfaces could significantly lower the activation energy of ethanol dehydrogenation reaction and improve hydrogen evolution reaction ability. This study provides deep insight into understanding and optimizing hot carrier dynamics in photothermal catalytic systems.

Preparation of a high-capacity three-dimensional CuS/Cu2S/rGO composite thin film electrode and its application in supercapacitors

Junhua Lu,Hedong Jiang,Pingchun Guo,Jiake Li,Hua Zhu,Xueyun Fan,Liqun Huang,Jian Sun,Yanxiang Wang

In recent years, significant progress has been made in the development of supercapacitors (SCs) for high-power applications, including electric vehicles, consumer electronics, military, and industrial uses. One crucial research area focuses on enhancing the electrochemical properties of SCs through the development of superior electrode materials. A composite electrode material (CuS/Cu 2 S/rGO) that combines Cu-sulfur compounds and graphene has been synthesized using the continuous ion layer adsorption (SILAR) method on reduced graphene thin films (rGO). The CuS/Cu 2 S/rGO film SC demonstrates exceptional electrochemical properties, attributed to the synergistic effect of CuS, Cu 2 S, and rGO. At a current density of 0.5 A g −1 , the mass-specific capacitance reaches 355.70 F g −1 , with a high energy density of 49.40 Wh kg −1 and a peak power density of 8511.49 W kg −1 . Even at an increased current density of 5 A g −1 , the mass-specific capacitance remains impressive at 239.62 F g −1 , indicating excellent rate performance. Furthermore, after 1000 constant current charge and discharge cycles, the material maintains a capacity retention rate of 87.13 %, underscoring its robust cycling stability.

pH-Responsive CuS/DSF/EL/PVP Nanoplatform Alleviates Inflammatory Bowel Disease in Mice via Regulating Gut Immunity and Microbiota

Jinpeng Yao,Yu Chen,Liang Zhang,Yuancun Cheng,Zheng Chen,Yanhui Zhang,Xiaoyi Zheng,Yanwei Lv,Shige Wang,Zhaoshen Li,Jiulong Zhao

The clinical treatment of inflammatory bowel disease (IBD) is challenging. We developed copper sulfate (CuS)/disulfiram (DSF)/methacrylic acid-ethyl acrylate copolymer (EL)/polyvinylpyrrolidone (PVP) nanoplatform (CuS/DSF/EL/PVP) and evaluated its efficiency for treating IBD. After oral administration, the pH-sensitive EL protected the CuS/DSF/EL/PVP against degradation by acidic gastric juices. Once the colon was reached, EL was dissolved, releasing DSF and Cu 2+ . Further, the main in vivo metabolite of DSF can bind to Cu 2+ and form copper (II) N, N-diethyldithiocarbamate (CuET), which significantly alleviated acute colitis in mice. Notably, CuS/DSF/EL/PVP outperformed CuS/EL/PVP and DSF/EL/PVP nanoplatforms in reducing colonic pathology and improving the secretion of inflammation-related cytokines (such as IL-4 and IL-10) in the colonic mucosa. RNA-seq analysis revealed that the nanoplatform reduced colonic inflammation and promoted intestinal mucosal repair by upregulating C-type lectin receptor (CLR)-related genes and signaling pathways. Furthermore, CuS/DSF/EL/PVP showed potential for improving colitis Th1/Th17 cells through innate immunity stimulation, down-regulation of inflammatory cytokines, and upregulation of anti-inflammatory cytokines. Additionally, the intervention with CuS/DSF/EL/PVP led to increased intestinal flora diversity, decreased Escherichia–Shigella abundance, and elevated levels of short-chain fatty acid (SCFA)-producing bacteria Prevotella, Lactobacillus , and Bifidobacterium , indicating their potential to modulate the dysregulated intestinal flora and suppress inflammation. Statement of Significance Our study introduces the CuS/DSF/EL/PVP nanoplatform as a therapeutic strategy for treating inflammatory bowel disease (IBD). This approach demonstrates significant efficacy in targeting the colon and alleviating acute colitis in mice. It uniquely modulates gut immunity and microbiota, exhibiting a notable impact on inflammation-related cytokines and promoting intestinal mucosal repair. The nanoplatform's ability to regulate gut flora diversity, combined with its cost-effective and scalable production, positions it as a potentially transformative treatment for IBD, offering new avenues for personalized medical interventions.
