A metal mediated specific poly(cyclotriphosphazene-co-curcumin) fluorescent micro-probe for tetracycline sensing

Xiqi Su,Le Wang,Xiao Zhang,Jin-Hua Xie,Yi Qu,Di Zhang,Li-June Ming

The extensive use of tetracycline antibiotics (TCs) renders their widespread in the environment that may cause bacterial resistance. Thus, detection of residual TCs and understanding of their broader roles and impacts in the environment and soil ecology are is of critical significance. This paper describes the investigation of a Mg (II)-specific fluorescent micro-probe (PCC–Mg) based on cyclotriphosphazene (Cpz) and curcumin for classical TC•HCl sensing with a low detection limit of 29 nM and a fast response time of less than 10 s. The sensing mechanism is presumably attributed to specific Mg 2+ binding with TCs since the fluorescence enhancement is not observed in the absence of Mg 2+ and not influenced by the presence of several metal ions and anions. The binding of Mg 2+ to curcumin/TC and the formation of TC-metal-curcumin ternary complex can be concluded with NMR and mass spectrometry . Moreover, for the purpose of easy applications, test strips are made to render visual detection of TC with noticeable and selective fluorescence enhancement. The formation of the ternary complex between Cur from the popular herb Curcuma longa and TC from Streptomyces spp. mediated by metal ions also hints a potential role of microbiolochemical-phytochemical interactions in maintaining biodiversity and equilibria in soil microecology.

Protective effects and mechanisms of curcumin on podophyllotoxin toxicity in vitro and in vivo

Juan Li,Cai-Xia Dai,Hua Sun,Lu Jin,Chong-Yi Guo,Wei Cao,Jie Wu,Hai-Yan Tian,Cheng Luo,Wen-Cai Ye,Ren-Wang Jiang

Podophyllotoxin (POD) is a naturally occurring lignan with pronounced antineoplastic and antiviral properties. POD binds to tubulin and prevents the formation of mitotic spindle . Although cases of overdose or accidental ingestion are quite often, no specific therapy is currently available to treat the POD intoxication. In the current investigation, the protective effects and mechanisms of curcumin (CUR) on podophyllotoxin toxicity were evaluated in vitro and in vivo . The results showed that CUR could protect POD-induced cytotoxicity by recovering the G2/M arrest and decrease the changes of membrane potential and microtubule structure in Vero cells. A significant decrease of mortality rates was observed in Swiss mice treated by intragastrical administration of POD   +   CUR as compared with POD alone. The POD   +   CUR group also exhibited decreases in plasma transaminases , alkaline phosphatase , lactate dehydrogenase , plasma urea , creatinine and malondialdehyde level but elevated superoxide dismutase and glutathione levels as compared to the POD group. Histological examination of the liver and kidney demonstrated less morphological changes in the treatment of POD   +   CUR as compared with POD alone. The mechanism of the protective effects might be due to the competitive binding of CUR with POD in the same colchicines binding site as revealed by the tubulin polymerization assay and the molecular docking analysis, and the antioxidant activity against the oxidative stress induced by POD. In summary, both in vitro and in vivo data indicated the promising role of CUR as a protective agent against the POD poisoning.

Solubility improvement of curcumin by crystallization inhibition from polymeric surfactants in amorphous solid dispersions

Ziyue Xi,Yali Fei,Yuxin Wang,Qing Lin,Qidong Ke,Guotai Feng,Lu Xu

To improve the solubility and maintain amorphous state of curcumin (CUR), solid dispersions of CUR with Kollidon CLSF and surfactant (TPGS or HPMC) were prepared by solvent evaporated method. The solid dispersions were characterized by scanning electron microscope, powder X-ray diffraction, differential scanning calorimetry and fourier transform infrared spectroscopy . The solubilization mechanism of surfactants in vitro and the pharmaceutical properties of optimal formulations in vivo were studied. The results showed that TPGS as a carrier provided superior solubility and stability for SDs due to its stable hydrogen bonding strength and related crystal nuclei growth mechanism. At different pH conditions (pH 1.2 and 6.8), TPGS significantly improved the solubility and release rate of CUR in solid dispersions. Thus, solid dispersion with high solubility and stability significantly enhanced the oral bioavailability and gastric ulcer healing rate. Therefore, solid dispersion with novel applied surfactants is an effect strategy to improve pharmaceutical properties of CUR.

Turn on fluorescence detection of curcumin in food matrices by the novel fluorescence sensitizer

Ya-Qian Wang,Li Li,Jin Yin,Xu Yu,Xiaowei Wu,Li Xu

In this study, silane reagents were for the first time explored as the fluorescence sensitizer. They were demonstrated to have fluorescence sensitization effect on curcumin and 3-glycidoxypropyltrimethoxysilane (GPTMS) possessed the strongest effect. Thus, GPTMS was adopted as the novel fluorescence sensitizer to turn on the fluorescence of curcumin by more than two orders of magnitude for detection. In this way, curcumin could be determined with a linear range of 0.2–2000 ng/mL and an LOD of 0.067 ng/mL. The method was applicable to determine curcumin in several actual food samples, which had the good consistency with the high performance liquid chromatographic method, demonstrating the high accuracy of the proposed method. In addition, the curcumins sensitized by GPTMS could be cured under certain conditions and held the potential for solid fluorescence application. This study expanded the scope of fluorescence sensitizer to silane reagents, and provided the novel approach for fluorescence detection of curcumin and further to generate new solid fluorescence system.

Impact of Solid-State Properties on the Aerosolization Performance of Spray-Dried Curcumin Powders

Zhang Qian,Li Yongquan,Li Linghui,Cheng Yushan,Yu Fangkun,Li Rui,Hou Shuguang

Amorphous and crystalline active pharmaceutical ingredients (APIs) are both widely studied for pulmonary delivery. The past research mainly studied the impact of solid-state properties on pharmacokinetic attributes; however, the influence of solid-state properties on aerosolization performance was much less studied. This study aimed to investigate the different aerosolization performances of amorphous and crystalline curcumin (Cur) stabilized with L-leucine. Cur was spray-dried with different concentrations of L-leucine (0, 5, 20, 35, and 50%, w/w) as both solution-based and suspension-based formulations to acquire amorphous and crystalline Cur powders. The physicochemical properties of the spray-dried powders, including particle size, morphology, and solid-state characteristics, were studied. The aerosolization performance as well as dissolution properties were evaluated. It was found that 35% (w/w) L-leucine or above led to the formation of amorphous Cur in the spray-dried powders, and the amorphous Cur powders exhibited higher FPF (70.8%, with 50% L-leucine, w/w) than the crystalline Cur formulations with an FPF at 56.3% (with 50% L-leucine, w/w). In conclusion, with a high concentration of L-leucine (35% or above) in the formulations, amorphous Cur would exhibit higher aerosolization efficiency than crystalline Cur. However, with a low concentration of L-leucine (20% or less) in the formulations, crystalline Cur would be preferred for more enhanced consideration. Graphical Abstract

Development and characterization of chitosan/bacterial cellulose/pullulan bilayer film with sustained release curcumin

Miaoyu Gan,Caoyu Guo,Wenying Liao,Xiaoli Liu,Qi Wang

A natural biopolymer bilayer film based on chitosan and bacterial cellulose with a protective layer of pullulan was developed by a two-step solution casting method. Curcumin was incorporated as an active antioxidant and antibacterial agent into the inner layer. The films with different curcumin concentrations were systematically characterized. Fourier transform infrared spectroscopy and X-ray diffraction analyses showed high compatibility between curcumin and the polysaccharide matrix through intermolecular interactions, which was verified by enhanced mechanical and barrier properties. The curcumin incorporation improved the thermal stability by >35.4 %, along with lower visible and ultraviolet light transmittance (< 8.6 %) and water solubility (< 25.1 %). The film had both antibacterial and antioxidant properties, and the sustained release of curcumin was largest (> 58.8 %) in the fatty food simulant lasting for over 155 h. The results suggested that the film containing 0.2 % curcumin had ideal physical and functional properties, suggesting its potential as a novel packaging material for the preservation of high-fat food.

Organic solvent-free starch-based green electrospun nanofiber mats for curcumin encapsulation and delivery

Zhen Du,Haowei Lv,Chenxi Wang,Deyun He,Enbo Xu,Zhengyu Jin,Chao Yuan,Li Guo,Zhengzong Wu,Pengfei Liu,Bo Cui

Developing green and efficient methods for the delivery of active food substances is a sustained demand for food scientists and industries. In this work, for the first time, we prepared a curcumin (CUR)-loaded starch-based fast-dissolving nanofiber by electrospinning technology. This green nanofiber was obtained by incorporating CUR with octenyl succinic anhydride starch (OSA) and pullulan (PUL) matrix using pure water as the solvent. To overcome the poor water-solubility and bioavailability of CUR, hydroxypropyl-beta-cyclodextrin (HPβCD) was used to form inclusion complexes. Phase solubility test results showed that by introducing HPβCD, the water-solubility of CUR was obviously improved. The prepared electrospun nanofibers were systematically characterized through scanning electron microscopy (SEM), X-ray diffraction (XRD), proton nuclear magnetic resonance spectroscopy ( 1 H NMR), Fourier transform infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), encapsulation efficiency testing, solubility testing and antioxidant activity testing. The results demonstrated that CUR was well encapsulated into HPβCD and OSA/PUL/CUR-HPβCD electrospun nanofibers with fine morphology and fast-dissolving character were successfully prepared. It is worth noting that the whole process and raw materials were green, suggesting that the prepared fast-dissolving nanofiber has great application potential in the food and pharmaceutical fields.

Multiple response behaviors of curcumin-loaded ammonium alginate/polyvinyl alcohol hydrogel and its application

Cai Jinyun,Zhong Haiyi,Tang Weihao,Wen Fangzhou,Lv Yingbin,Huang Xinghai,Luo Jianwei,Li Peiyuan

Safety, non-toxic, and degradable ammonium alginate with polyvinyl alcohol were selected as substrates for the preparation of AA/PVA hydrogel by simple co-blending and cyclic freeze-thaw methods. Curcumin as the model drug was encapsulated to obtain AA/PVA/Cur hydrogel. The resulting hydrogels were characterized using various techniques to confirm the success of preparation, including FTIR, SEM, XRD, TGA, and rheometer. In addition, multiple stimuli-responsive behaviors of hydrogels were studied in depth. For one thing, AA/PVA hydrogel showed dual response shape memory properties which could be fixed in a temporary shape in Ca 2+ or low temperature and followed recovery to the original shape with more than 90% recovery rate in Na + (Na 2 CO 3 and EDTA.2Na) or high temperature respectively, confirming that the hydrogels have ion- and temperature-responsive behaviors. For another, the drug-loaded hydrogel AA/PVA/Cur exhibited different drug release properties under different pH stimulation conditions, where curcumin could reach nearly 90% release under alkaline conditions after 6 h, whereas the release rate was only nearly 20% under acidic conditions. AA/PVA/Cur hydrogel had antibacterial and antioxidant effects, especially under alkaline conditions. Thereinto, the antibacterial rate of Escherichia coli and Staphylococcus aureus reached over 90%, respectively. Among the four antioxidant systems, up to 98.5%, ABTS radical scavenging rate was prominent. In conclusion, multiple stimulus-response properties of AA/PVA hydrogel exhibit a unique three-dimensional network structure capable of drug loading, combined with various properties such as good hydrophilicity, biocompatibility, biodegradability, non-cytotoxicity, and hemocompatibility, which have potential applications in medical and pharmaceutical fields.

Enhancement of the chemical stability of nanoemulsions loaded with curcumin by unsaturated mannuronate oligosaccharide

Decheng Bi,Meiting Li,Lijun Yao,Nanting Zhu,Weishan Fang,Wushuang Guo,Yan Wu,Hong Xu,Zhangli Hu,Xu Xu

Unsaturated mannuronate oligosaccharide (MOS) is an acidic oligosaccharide prepared from alginate-derived polymannuronate by enzymatic depolymerization, followed by double bond formation between C-4 and C-5 at the nonreducing end. In this study, MOS was used as a stabilizer to fabricate O/W nanoemulsions loaded with curcumin (MOS-CUR) for the first time. The results revealed that the MOS-CUR showed small droplet sizes and narrow size distributions and was slightly more stable than normal oil-in-water (O/W) curcumin nanoemulsions (water-CUR). Additionally, MOS can improve the superoxide anion scavenging ability and iron ion reducing ability of the curcumin nanoemulsion system. Although the digestion behaviour of MOS-CUR and water-CUR was similar, the bioavailability of curcumin in MOS-CUR was significantly higher than that in water-CUR. All these results indicated that MOS could be used as a stabilizer for preparing nanoemulsions to easily encapsulate labile nutrients and to enhance the bioavailability and antioxidant capacity of these nutrients.

Effect of oligosaccharides as lyoprotectants on the stability of curcumin-loaded nanoliposomes during lyophilization

Hanyun Jiang,Yanping Wang,Xiankang Xu,Leiyu Deng,Lifang Feng,Jianzhong Han,Weilin Liu

Nanoliposome is a promising delivery system, whereas its commercial application is limited by the structural instability, cargo leakage and particles aggregation during the processing such as freeze-drying. In this study, the effect of four oligosaccharides, fructo-oligosaccharides, lactose, inulin and sucrose (control), on the physicochemical properties, structural stability, and in vitro semi-dynamic digestion behavior of curcumin-loaded nanoliposomes were investigated before and after lyophilization. The results showed that the addition of the oligosaccharides inhibited the changes in particle size and reduced curcumin leakage from lyophilized nanoliposomes. Oligosaccharides significantly improved the physical stability of lyophilized nanoliposomes and delayed curcumin release during in vitro digestion. In addition, oligosaccharides could decrease the hydrophobicity of liposomal membrane and the tightness of phospholipid molecule arrangement, with the increase in micropolarity and fluidity of the bilayer membranes. These results suggested that fructo-oligosaccharides, lactose and inulin could be effective lyoprotectants for lyophilized nanoliposomes.

Gelatin-based nanofiber membranes loaded with curcumin and borneol as a sustainable wound dressing

Yarong Lv,Zhongxun Yu,Chenxi Li,Jianwei Zhou,Xujin Lv,Jia Chen,Ming Wei,Jun Liu,Xiaomin Yu,Ce Wang,Ping Hu,Yong Liu

Infection is a huge obstacle to wound healing. Thus, to enhance the healing of infected wounds, wound dressings that permit the dual delivery of antimicrobials and antioxidants are highly desirable. In this study, a series of gelatin-based nanofiber membranes with different curcumin contents were fabricated via solution electrospinning. The obtained membranes were characterized in terms of their morphologies, in addition to their physical, mechanical, and in vitro properties. The results showed that the membranes maintained an integrated morphology, excellent water absorption capability, satisfactory mechanical properties, and a high dissolution rate of curcumin. The addition of curcumin and borneol conferred the membranes the ability to inhibit Staphylococcus aureus and eliminate free radicals. Furthermore, cytocompatibility testing using the L929 cell line confirmed the excellent biocompatibility of the membranes. These gelatin-based nanofiber membranes loaded with curcumin and borneol can therefore be considered as promising materials for dressing wounds. Moreover, the use of biodegradable polymers and environmentally sustainable production techniques in this system render it suitable for the commercial manufacture of composite membranes.

Application of Curcumin Emulsion Carrier from Ultrasonic-Assisted Prepared Octenyl Succinic Anhydride Rice Starch

Yuxue Zheng,Huiling Zhang,Xiaobo Wei,Haitian Fang,Jinhu Tian

The emulsification of ultrasonic-assisted prepared octenyl succinic anhydride (OSA) rice starch on curcumin was investigated in the present study. The results indicated that the encapsulation efficiency of curcumin in emulsions stabilized by OSA-ultrasonic treatment rice starch was improved, from 81.65 ± 0.14% to 89.03 ± 0.09%. During the in vitro oral digestion, the particle size and Zeta potential of the curcumin emulsion did not change significantly (p> 0.05). During the in vitro digestive stage of the stomach and small intestine, the particle size of the curcumin emulsion continued to increase, and the absolute potential continued to decrease. Our work showed that OSA-pre-treatment ultrasonic rice starch could improve curcumin bioavailability by increasing the encapsulation efficiency with stronger stability to avoid the attack of enzymes and high intensity ion, providing a way to develop new emulsion-based delivery systems for bioactive lipophilic compounds using OSA starch. Keywords:rice starch;octenyl succinic anhydride;curcumin;emulsion
