The purpose of this study was to investigate the effects of Lactobacillus rhamnosus GG (LGG) fermentation on the composition, structure, and functional properties of dietary fiber (DF) in bamboo shoot. Then, we added it to bread to evaluate the texture properties, digestive properties, and functionality of bread. After LGG fermentation, the DF was decomposed into pieces, which had stronger water-swelling capacity and nitrite adsorption capacity. The ability of producing short-chain fatty acids was significantly improved and the digestive resistance was remarkable enhanced as well. Except the bread hardness was increased, there was no significant difference in other texture properties when adding 3% FTDF-LGG to bread. It had good adsorption capacity of cholesterol and more than 25% reduced the release of reducing sugar. Overall, the technic of LGG fermentation had improved functional properties of DF in bamboo shoot, which could be applied to bread production for exerting its effects in the future. Practical applications Bamboo shoots are immature and tender stems of bamboo, rich in nutritional value, and rich in DF. Bamboo shoot DF has been proven to have a variety of biological activities, and is the main material for bamboo shoot to exert functional activities. In this study, bamboo shoot DF was modified by LGG fermentation, which showed stronger functional activity, and was successfully applied to bread. This study lays the foundation for the fermented modified bamboo shoot DF and its application in food.

Simple SummaryThe leguminous forageChamaecrista rotundifoliacontains a high crude protein content and can be an important feed source for livestock. Forage is preserved mainly as silage, and microbial fermentation is key to high-quality silage. The fermentation quality of silage depends on the microbial community structure and the metabolites produced during silage fermentation. The current study provides evidence that the addition of malic or citric acid and/orLactobacillus acidophilus(L) improves the quality of silage and inhibits the growth of harmful microorganisms.AbstractThis study aimed to investigate the effects of citric acid, malic acid, andLactobacillus acidophilus(L) on fermentation parameters and the microbial community of leguminousChamaecrista rotundifoliasilage. FreshC. rotundifoliawas treated without any additive (CK), or with L (106CFU/g fresh weight), different levels (0.1, 0.3, 0.5, and 1% fresh weight) of organic acid (malic or citric acid), and the combinations of L and the different levels of organic acids for 30, 45, and 60 days of ensiling. The effects of malic acid and citric acid were similar during the ensiling process. Treatment with either citric or malic acid and also when combined with L inhibited crude protein degradation, lowered pH and ammonia nitrogen, and increased lactic acid concentration and dry matter content (p< 0.05). The neutral detergent fiber and acid detergent fiber increased initially and then decreased with fermentation time in all treatments (p< 0.05). Increasing the level of organic acid positively affected the chemical composition ofC. rotundifoliasilage. In addition, the addition of 1% organic acid increased the relative abundance ofLactobacillus, while the relative abundances ofClostridiumandEnterobacterdecreased at 60 days (p< 0.05). Moreover, both organic acids and combined additives increased (p< 0.05) the relative abundance ofCyanobacteriaat 60 days of fermentation. We concluded that adding malic acid, citric acid, and L combined with an organic acid could improve the quality ofC. rotundifoliasilage and increase the relative abundance of beneficial bacteria. The addition of organic acid at a level of 1% was the most effective. Keywords:citric acid;Chamaecrista rotundifoliasilage;Lactobacillus acidophilus;malic acid;microbial diversity

This research sought to assess the anticancer effects of Lactobacillus acidophilus CICC 6074 and its underlying mechanism of action. L. acidophilus CICC 6074 inhibited HT-29 cell activity and induced its apoptosis in a dose- and time-dependent manner. HT-29 cells exposed to L. acidophilus CICC 6074 showed mitochondrial membrane potential (ΔΨm) loss, as well as Cyt c is released from the mitochondria into the cytoplasm, causing Bax, Caspase-9 and Caspase-3 to be up-regulated and Bcl-2 to be down-regulated. To further explore the mechanism by which L. acidophilus CICC 6074 achieves its anti-cancer effect, a HT-29 cell-induced colon cancer mouse model was used. Compared with a colon cancer model control group, the high-dose L. acidophilus CICC 6074 group showed obvious colon cancer cell apoptosis as measured by histological morphology, electron microscopy, and terminal dexynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) assay. An in vivo study showed that L. acidophilus CICC 6074 induced colon cancer apoptosis by up-regulating Bax, down-regulating Bcl-2, releasing Cyt c from the mitochondria into the cytoplasm, and activating Caspase-3 and Caspase-9. These results demonstrate that L. acidophilus CICC 6074 exerts anticancer effects via the activation of mitochondrial pathways which then induce apoptosis. The research could provide new ideas and theoretical basis for development of probiotics functional food market.

In this study, a new strain of Lactobacillus plantarum (CY.6) was identified and its L-arabinose isomerase (L-AI) encoding gene ( ara A) was overexpressed in Escherichia coli BL21 for the biosynthesis of D-tagatose from milk whey powders (WP). Whole-cell biotransformation of lactose in WP into D-tagatose was done by three technological approaches, including 100%, 50% and 0% hydrolysis of lactose in WP before biotransformation, where simultaneous saccharification and biotransformation (SSB, 0% prior hydrolysis of lactose) produced maximum amounts of D-tagatose. Two-stage SSB provided 73.6% conversion efficiency (based on D-galactose) and 36.8% (in term of lactose), with 51.5 g/L of D-tagatose after 96 h, while concentration of D-tagatose produced after first stage was 34.4 g/L. Yield and volumetric productivity of D-tagatose after two-stage SSB were found to be 0.26 g/g of WP (0.37 g/g of lactose, 0.74 g/g of D-galactose produced from lactose) and 0.54 g/L/h, respectively.

Lactic acid bacteria strain (LAB) NCU116 fermented Asparagus officinalis polysaccharides (FAOP) have been proven to cause substantial changes in physicochemical properties such as monosaccharide composition and molecular weight, accounting for their enhanced immune activity than unprocessed Asparagus officinalis polysaccharides (AOP). In the current study, the hepatoprotective effects of FAOP in mice with cyclophosphamide (CTX)-induced hepatotoxicity were investigated. FAOP were more effective than AOP in alleviating CTX-induced hepatic damage, including inhibition of hepatic biochemical markers (ALT, AST, AKP and LDH) and pro-inflammatory cytokines (TNF-α and IL-1β) as well as reinforcement of antioxidant systems (T-AOC, SOD, CAT, and MDA). In particular, compared with AOP, FAOP showed superior performance by promoting GSH biosynthesis, and normalizing the expression level of bile acid receptors (FXR and SHP) and key enzymes in bile acid synthesis (CYP7A1, CYP8B1 and CYP27A1). Modulation of disordered homeostasis of bile acids by FAOP can be attributed to the upregulation of hepatic short chain fatty acid (SCFA) receptors GPR41 and GPR109A as well as intestinal SCFA production. Furthermore, serum metabolomics study validated the hepatoprotective superiority of FAOP than AOP with evidence from variations in bile acid compositions and the construction of related metabolic pathways. Therefore, LAB NCU116 fermentation of Asparagus officinalis was practical and effective to obtain promising hepatoprotective polysaccharides, which might arise from enhanced SCFA production than unprocessed AOP.

The enhancement of cellulose degradation is important for improving the quality of corn-stalk silage. However, the rapid drop in pH caused by the propagation of lactic acid bacteria (LAB) can influence the degradation of cellulose by cellulose-degrading microorganisms (CDMs) during the mixed fermentation process of ensilage. In this study, a CDM (Bacillus methylotrophic 2–4, BM 2–4) was isolated, and its lyophilization condition was studied. Moreover, a slow-release strategy was developed to delay the release of LAB (Lactobacillus plantarum S-1, LP S-1) by embedding technology to provide time for BM 2–4 to degrade cellulose during the corn-stalk ensilage process. The results showed that BM 2–4 had a higher survival rate (89.53%) under the following conditions: cell collection (5,600 r/min in 4.4°C for 9.5-min centrifugation) and lyophilization using cryoprotectants [skim milk (10.4%), peptone (10.4%), and glucose (5.2%)] at −50°C with a vacuum pressure of <10 Pa. Based on the results of the previous study on embedded LP S-1, simultaneous inoculation of embedded LP S-1 and lyophilized BM2-4 at a 2:1 ratio, with an inoculum size of 6% and moisture content of 85%, significantly degraded CF by 3.8% and increased CP by 3.7% (p < 0.05). This treatment did not significantly influence the final pH of corn-stalk silage (p > 0.05) after 7 days of fermentation.

The therapeutic mechanism of oleanolic acid (OA) in breast cancer has been widely reported, but little has been known about the combined effects of transcriptome and gut microbiome. In this study, the phenotypic effect of oleanolic acid on mice was tested at the end of the administration cycle, and RNA sequencing on murine tumor tissue and 16S-rRNA sequencing on intestinal contents were conducted to analyze gene expression profiles and microbial diversity between the control group and OA treated group using 4T1-induced mice breast cancer model. As a result, it has been confirmed that oleanolic acid would play a significant inhibitory effect on the development of breast tumors in mice. Based on the integrative analysis of the transcriptomic and metagenomic data, it was found that the abundance of Lactobacillus in the intestinal flora of mice significantly increased in the OA group. Moreover, the up-regulation of Il10 had a significant effect on inhibiting the tumor progression, which played a role through cytokine-cytokine receptor interaction pathway.

This study aimed to determine the optimal fermentation process for enhancing the content of small peptides (C SP ) and the degradation efficiency of free gossypol (D FG ), improving nutritional value. The results indicated that after fermentation, the D FG was reduced to 85.63 % (below both safety standards for human consumption (<200 mg/kg) and animal feed requirements (100–500 mg/kg)), and the C SP content was elevated to 46.25 %. The proportion of soluble protein to total protein and the total free AA were increased by 281.25 % and 1015.68 % than that of the raw feedstock, respectively ( P < 0.05). Furthermore, the enrichment of harmful bacteria in CSM was inhibited by the fermentation process, and the ability to improve in vitro fermentation characteristics of cows and in vitro dry matter and crude protein digestibility of pigs was also demonstrated by fermented CSM. Overall, the synergistic fermentation process of CSM with LLK-XR1 and acid protease enhanced its nutritional value and bioavailability, offering a practical solution for utilizing CSM.

Stress is a well-established disruptor of reproductive function in poultry. Gamma-aminobutyric acid (GABA), a notable postbiotic derived from Lactobacillus plantarum ( L. plantarum ), has attracted increasing attention as a dietary supplement. While existing research predominantly focuses on GABA applications in broilers and other species, its effects on laying hens remain underexplored. This study investigated whether GABA and L. plantarum 1-2-3 supplementation mitigated corticosterone (CORT)-induced chronic stress in laying hens. Results demonstrated that both GABA and L. plantarum 1-2-3 effectively mitigated CORT-induced declines in laying performance and ovarian damage. They reduced ovarian oxidative stress, inflammation, and atretic follicles by enhancing antioxidant capacity and normalizing expression of inflammatory factors ( IL18 , LITAF , IL6 , IL1B) , and apoptosis-related factors ( BCL2 , BAX , CASP9 , CASP3 ). Furthermore, GABA and L. plantarum 1-2-3 markedly increased the serum reproductive hormone levels, as well as the transcription level of ovarian steroidogenic genes ( LHCGR, FSHR, StAR, CYP11A1 ), while modulated ovarian cholesterol metabolism via PPARα-mediated pathways. These findings elucidate the mechanisms by which GABA and L. plantarum 1-2-3 alleviate stress in laying hens, providing a theoretical and practical basis for their use as functional feed additives in stress-exposed flocks.

Feruloyl esterase (FAE)-producing lactobacillus strains were selected from the citrus whole-fruit pulp after 48 h of fermentation. Among them, Lactiplantibacillus plantarum K33 showed the highest ferulic acid (FA) producing ability. Using citrus whole-fruit pulp as the fermentation matrix, physicochemical, compositional, and sensory outcomes were compared between pulps fermented by K33 and by the commercial strain L. plantarum N13 (Lp-N13). After fermentation, K33 increased total acidity, lowered pH, and markedly raised FA concentration and total phenolic content (TPC). Fermentation also altered other organic acids and volatile profiles: L-malic acid decreased significantly, whereas lactic and succinic acids increased. Terpenes remained predominant, with a significant rise in the relative proportion of alcohols. KEGG analysis showed that the differential of organic acids was mainly associated with arginine biosynthesis and other pathways. Sensory evaluation indicated enhanced lactic acid flavor and reduced citrus-peel aroma. Overall, K33 shows promise for fermentation of functional plant-based beverages.

β-LG-EPS-EGCG complex could effectively stabilize W 1 /O/W 2 emulsion during long-time storage. • W 1 /O/W 2 emulsion stabilized by β-LG-EPS-EGCG complex was used to encapsulate Lactobacillus plantarum. • Lactobacillus plantarum encapsulated in W 1 /O/W 2 emulsion was clearly observed under CLSM. • Flow cytometry was used to analyze the survival rate of strains under different treatments. The efficacy of probiotics is limited by their low survival capacity in human gastrointestinal tract. In this study, a water-in-oil-in-water (W 1 /O/W 2 ) emulsion delivery system stabilized by a ternary complex composed of β-lactoglobulin (β-LG), exopolysaccharide (EPS) and epigallocatechin gallate (EGCG) was constructed to encapsulate L. plantarum A81. Compared with native β-LG and binary β-LG-EPS complexes, the β-LG-EPS-EGCG ternary complex significantly decreased emulsion droplet size, improved interfacial protein adsorption efficiency, and formed a denser, elastic interfacial structure. Confocal laser scanning microscopy confirmed more uniform droplet distribution and favorable bacterial encapsulation in ternary complex-stabilized emulsions. Furthermore, the emulsion stabilized by β-LG-EPS-EGCG complex significantly improved the survival rate of strains during 30-day storage (from 27.62% ± 1.35% to 47.33% ± 1.23%), after pasteurization (from 10.55% ± 1.24% to 18.87% ± 1.56%), and after in vitro simulated gastrointestinal digestion (from 3.01% ± 0.81% to 8.97% ± 1.37%). These results demonstrate that β-LG-EPS-EGCG ternary complex is a good stabilizer for W1/O/W2 emulsion. Download: Download high-res image (295KB) Download: Download full-size image

BACKGROUND Cyclophosphamide (CTX), a widely used alkylating chemotherapeutic agent, causes severe systemic immunosuppression and gut microbiota dysbiosis through non-specific cytotoxicity, increasing infection risk and impairing chemotherapy outcomes. Safe and effective adjuvant therapies for chemotherapy-related immune injury remain limited. This study evaluated the protective effect of compound AB probiotics ( Lactobacillus acidophilus LA15 combined with Bifidobacterium animalis subsp. lactis Bi66) against CTX-induced immunosuppression in mice and explored correlations associated with the gut-immune axis. RESULTS CTX challenge induced immune organ atrophy, cytokine imbalance, impaired innate and adaptive immunity, intestinal barrier disruption, reduced short-chain fatty acid (SCFA) synthesis and gut dysbiosis. AB probiotic intervention alleviated these injuries dose-dependently: it restored thymus and spleen indexes, rebalanced serum cytokines, improved core immune functions, and repaired ileal mucosal pathological damage. Furthermore, probiotic treatment was accompanied by elevated fecal SCFA concentrations, upregulated tight junction gene expression, recovered aryl hydrocarbon receptor (AhR)/ERK/STAT3 transcription, enriched beneficial bacterial genera, and suppressed opportunistic pathogens. CONCLUSION Compound AB probiotics alleviate CTX-induced immunosuppression in mice, and this effect correlates with restored gut microecology, enhanced SCFA metabolism, repaired intestinal barrier, and activated AhR signaling. These findings provide evidence for developing probiotic preparations as adjuvant interventions against chemotherapy-related immunosuppression. © 2026 Society of Chemical Industry.
