Bacterial diseases pose a serious threat to human health. Continued development of precise diagnostic methods and synergistic therapy techniques for combating bacteria are needed. Herein a hybrid nanosystem (Ag2S@ZIF-Van NS) was constructed by one-step self-assembly of Zn2+, vancomycin (Van) and Ag2S quantum dots (QDs). The nanosystem possesses excellent second near-infrared transparency window (NIR-II) fluorescence properties (∼1200 nm emission wavelength), good photothermal conversion properties, and biocompatibility. The material system enables precise, targeted NIR-II fluorescent imaging of bacterial inflammation in vivo as well as promoting anti-bacterial and wound healing effects.

It is essential to develop novel multifunctional and easily synthesized stable NIR-II fluorescent probes to guide photothermal therapy for tumors. Here, we propose a new strategy to construct boron dipyrromethene (BODIPY) J-aggregates by intermolecular hydrogen bonding (H-bond) and π-π stacking interactions to achieve fluorescence emission in the second near-infrared window (NIR-II, 1000–1700 nm). A novel meso -benzamide galactose hexanoate-BODIPY (Gal-OH-BDP) amphiphilic small molecular dye was synthesized and it formed nanoparticles spontaneously in aqueous solution with a maximum emission wavelength near 1060 nm, which works as a smart nanomedicine for targeting NIR-II imaging-guided photothermal therapy (PTT) of hepatocellular carcinoma. Galactose not only provided hydrogen bonds to regulate the aggregation pattern of the molecules but also effectively targeted hepatocellular carcinoma cells and promoted the formation of well-dispersed nanoparticles of dye molecules due to their hydrophilicity . Moreover, due to high photothermal conversion efficiency (PCE = 55%), Gal-OH-BDP NPs achieve galactose-targeted NIR-II imaging and PTT, which is important for the precise diagnosis and treatment of tumors ( Scheme 1 ). In the present research work, H-bond was introduced for the first time into BODIPY for building J-aggregates to achieve the NIR-II fluorescence.

Aqueous electrochemiluminescence (ECL) in the second near-infrared biowindow (NIR-II, 900–1700 nm) was anticipated for ECL evolution and spectral multiplexing. Herein, aqueous and monochromatic ECL with a single emission peak beyond 900 nm was achieved by employing methionine (Met)-capped Au–Ag bimetallic nanoclusters (BNCs) as luminophores and triethanolamine (TEOA) as a coreactant. The Met-capped Au–Ag BNCs with surface-defect-induced PL around 756 nm were water-soluble and synthesized via doping Met-capped Au NCs with Ag in a doping-in-growth way. By extensively exploiting the red-shifting nature of surface-defect-induced ECL to PL and the synergetic-effect-enhanced ECL of BNCs, physically surface-confined Au–Ag BNCs exhibited efficient NIR-II ECL around 906 nm in aqueous medium. A spectrum-based NIR-II ECL immunoassay around 915 nm was also achieved by immobilizing the Au–Ag BNCs onto an electrode surface via forming a sandwich immunocomplex, which could selectively determine CA125 from 5 × 10–4 to 1 U/mL with a detection limit of 5 × 10–5 U/mL (S/N = 3). The combined strategy of surface-defect-induced ECL and synergetic-effect-enhanced ECL would enable promising biorelated application of NIR-II ECL.

Local administration of therapeutic agents with long-term retention capabilities efficiently avoids nonspecific distribution in normal organs with an increased drug concentration in pathological tissue. Herein, we developed an injectable and degradable alginate-calcium (Ca2+) hydrogel for the local administration of corn-like Au/Ag nanorods (NRs) and doxorubicin hydrochloride (DOX·HCl). The immobilized Au/Ag NRs with strong absorbance in the near-infrared II (NIR-II) window efficiently ablated the majority of tumor cells after 1064 nm laser irradiation and triggered the release of DOX to kill residual tumor cells. As a result, injectable hydrogel-mediated NIR-II photothermal therapy (PTT) and chemotherapy efficiently inhibited tumor growth, resulting in the complete eradication of tumors in most of the treated mice. Furthermore, owing to the confinement of the Au/Ag NRs and DOX·HCl within the hydrogel, such treatment exhibited excellent biocompatibility.

Developing new strategies to enhance drug accumulation in the tumor and therapeutic efficacy is of great importance in the field of tumor therapy. Herein, a peanut-like multifunctional nanomedicine (CuS-PGH NMs) made of CuS nanoparticles encapsulated in poly(L-lysine)(PLL)/glucose oxidase (GOx)-hyaluronic acid (HA) shells has been constructed via layer-by-layer (LbL) assembly, and shows good biocompatibility and effective multi-gradient therapy. Because of the enhanced permeability and retention (EPR) effect, the CuS-PGH NMs could significantly enhance the cellular uptake by tumors overexpressing CD44 receptors, which respond to hyaluronidase (HAase)-triggered surface charge conversion. Once internalized by the tumor, GOx was the first to be exposed and could effectively deplete endogenous glucose for starvation therapy, and the excess H2O2 was then converted into highly toxic hydroxyl radicals (˙OH) via a Cu+-mediated Fenton-like reaction for chemodynamic therapy (CDT). Meanwhile, the as-obtained Cu+ ions accompanied the regenerated less-active Cu2+ ions. Interestingly, the high content of H2O2 could, in turn, accelerate Cu2+/Cu+ conversion to promote the Cu+–H2O2 reaction for enhanced chemodynamic therapy (CDT), thereby achieving efficient tumor growth suppression via synergistic starvation/CDT therapy. Subsequently, owing to the strong NIR-II absorption capability of CuS-PGH NMs, effective photothermal tumor ablation of the weakened tumor cells could be realized with the precise guidance of NIR-II PAI. This multi-gradient therapeutic strategy has been demonstrated to have excellent antitumor activity with minimal nonspecific damages, and offers a new avenue to precise tumor therapy.

Hypoxic tumor microenvironment leads to resistance or failure of radiotherapy (RT). As a non-invasive therapy, photothermal therapy (PTT) can improve the tumor hypoxic microenvironment in addition to directly killing tumor cells. PTT combined with RT (thermoradiotherapy) becomes an emerging treatment. Multi-functional nanoparticles used for hypoxia-resistant thermoradiotherapy in the second near-infrared (NIR-II) biological windows (1000–1700 nm) are urgently needed to be developed. Here, a facil method synthesis of ultra-small cysteamine (Cys)-coated FePd bimetallic nanodots (NDs) is reported. These NDs can not only produce effective hyperthermia (35.4%) when irradiated in the NIR-II region (1064 nm) but also have an enhanced radiation effect due to i) Hypoxic improvement in tumor tissues by photothermal treatment in the NIR-II Biological Windows can greatly enhance the sensitivity of tumor cells to radiotherapy ii) The ability of NDs to deposit radiation energy in tumors has further enhanced the sensitivity of tumor cells to radiotherapy. Meanwhile, NDs was a contrast agent for tri-modal imaging including computed tomography (CT)/magnetic resonance imaging (MRI)/photoacoustic imaging (PAI) in vitro and in vivo . Both in vitro and in vivo tests demonstrated good biocompatibility and excellent stability of NDs, indicating great potential for clinical applications.

Second near-infrared (NIR-II) fluorescence imaging in the range of 1000–1700 nm has great prospects for in vivo imaging and theranostics monitoring. At present, few NIR-II probes with theranostics properties have been developed, especially the high-performance organic theranostics material remains underexploited. Herein, we demonstrate a selenium (Se)-tailoring method to develop high-efficient NIR-II imaging-guided material for in vivo cancer phototheranostics. Via Se-tailoring strategy, conjugated oligomer TPSe-based nanoparticles (TPSe NPs) achieve bright NIR-II emission up to 1400 nm and exhibit a relatively high photothermal conversion efficiency of 60% with good stability. Moreover, the TPSe NPs demonstrate their photothermal ablation of cancer cells in vitro and tumor in vivo with the guidance of NIR-II imaging. It is worth noting that the TPSe NPs have good biocompatibility without obvious side effects. Thus, this work provides new insight into the development of NIR-II theranostics agents.

Near-infrared (NIR) phosphor-converted light-emitting diodes (pc-LEDs) lighting sources are applied in non-destructive examination and illumination supplements for night monitoring. As a critical component of NIR pc-LED, NIR phosphors are required to possess a high external quantum efficiency (EQE), super thermal stability, and broadband emission to meet applications. Here, Na 2 SrGe 6 O 14 :Cr 3+ (NSGO:Cr) glass ceramics (GC) bulk via highly crystallization from glass precursor is fabricated and demonstrates an ultrabroad emission band centered at 815 nm with a full width at half-maximum (FWHM) of 214 nm and a high EQE of 49.2%. Moreover, Yb 3+ co-doping significantly enhances Cr 3+ → Yb 3+ energy transfer (ET) due to the stronger Yb 3+ : 2 F 7/2 - 2 F 5/2 absorption. Benefiting from the ET process, the Cr/Yb co-doped GC exhibits NIR-I/II emission with improved thermal stability from 30.1% to 65.0%. Furthermore, an encapsulant-free NIR GC-converted LED (GCc-LED) is prepared and exhibits 52.2 mW NIR output power with a high photoelectric conversion efficiency (PCE) of 19.4%. Under a large current of 600 mA, the NIR device exhibits a watt-level output power of 922.4 mW with a PCE of 9.1%. Compared to conventional NIR powder phosphors, NSGO:Cr/Yb GC with high performance can open up an avenue for the fabrication of high power NIR lighting sources.

Radiotherapy (RT) is dominantly used in breast cancer therapy but is facing fierce side effects because of the limited difference between tumor and normal tissues in response to ionizing radiation. Herein, we construct a core-shell nanoparticle of UiO-66-NH2@AuNS. Then the solid gold shell was etched into hollow AuNS (HAuNS) and further modified with biotin-PEG-SH (PEG-bio) to obtain HAuNS@PEG-bio. HAuNS@PEG-bio demonstrates effective near infrared II (NIR-II) region photothermal therapy (PTT) performance, and the increase of temperature at the tumor site promotes the blood circulation to alleviate the hypoxia in the tumor microenvironment (TME). Meanwhile, HAuNS exhibits strong X-ray absorption and deposition ability due to the high atomic coefficient of elemental Au (Z = 79) and hollowed-out structure. Through the dual radiosensitization of the high atomic coefficient of Au and the hypoxia alleviation from PTT of HAuNS, the breast cancer cells could undergo immunogenic cell death (ICD) to activate the immune response. The outstanding anticancer effects of HAuNS@PEG-bio by cell targeting to exert IR thermal and CT imaging-guided NIR-II photothermal, radiosensitizing therapy, and ICD in vivo. This work highlights that the constructed biotin-decorated hollow gold nanoshell has a promising potential as a diagnostic and treatment integration reagents for the breast cancer.

Orthotopic advanced hepatic tumor resection without precise location and preoperative downstaging may cause clinical postoperative recurrence and metastasis. Early accurate monitoring and tumor size reduction based on the multifunctional diagnostic-therapeutic integration platform could improve real-time imaging-guided resection efficacy. Here, a Near-Infrared II/Photoacoustic Imaging/Magnetic Resonance Imaging (NIR-II/PAI/MRI) organic nanoplatform IRFEP-FA-DOTA-Gd (IFDG) is developed for integrated diagnosis and treatment of orthotopic hepatic tumor. The IFDG is designed rationally based on the core “S-D-A-D-S” NIR-II probe IRFEP modified with folic acid (FA) for active tumor targeting and Gd-DOTA agent for MR imaging. The IFDG exhibits several advantages, including efficient tumor tissue accumulation, good tumor margin imaging effect, and excellent photothermal conversion effect. Therefore, the IFDG could realize accurate long-term monitoring and photothermal therapy non-invasively of the hepatic tumor to reduce its size. Next, the complete resection of the hepatic tumor in situ lesions could be realized by the intraoperative real-time NIR-II imaging guidance. Notably, the preoperative downstaging strategy is confirmed to lower the postoperative recurrence rate of the liver cancer patients under middle and advanced stage effectively with fewer side effects. Overall, the designed nanoplatform demonstrates great potential as a diagnostic-therapeutic integration platform for precise imaging-guided surgical navigation of orthotopic hepatic tumors with a low recurrence rate after surgery, providing a paradigm for diagnosing and treating the advanced tumors in the future clinical translation application. Cuproptosis is a newly recognized copper-dependent nonapoptotic form of cell death, which stimulate studies exploring copper-based nanomaterials to treat cancer through distinct mechanistic action. However, it remains a challenge to completely eradicate tumors via monotherapy. Herein, a copper-doped BiSe x (CBS) nanozyme was developed to boost αPD-L1-mediated immune checkpoint blocking (ICB) via synergetic apoptosis/cuproptosis-induced immunogenic cell death (ICD). The defect-engineered CBS nanozyme exhibits strong peroxidase-mimicking activities and generates abundant reactive oxygen species (ROS) production causing cell apoptosis, which could be further augmented by NIR photoirradiation. Meanwhile, the CBS could cause mitochondrial lipoylated protein aggregation, leading to cell cuproptosis. The photothermal/catalytic/cuproprosis synergistic therapy triggered by CBS nanozyme combined with αPD-L1 antibody effectively inhibits the growth of primary tumors and distant tumors in prostate cancer model. Furthermore, CBS nanozyme exhibited significant germicidal effect in wound infection. Collectively, this work provides a new insight into cancer treatment of copper-based nanozyme based on cuproptosis/apoptosis-related combined therapy.

Background Photothermal therapy (PTT) in the second near-infrared (NIR-II) window has attracted extensive attention due to the benefits in high maximum permissible exposure and penetration depth. Current photothermal agents generally show a broadband absorption accompanied by a gradual attenuation of absorption in the NIR-II window, leading to poor effect of PTT. It remains a great challenge to gain photothermal agents with strong and characteristic absorption in NIR-II regions. To overcome this problem, based on carbon dots (CDs)-mediated growth strategy, we proposed a simple and feasible approach to prepare plasmonic gold nanodendrites (AuNDs) with NIR-II absorption to enhance the therapeutic effect of PTT. Results By rationally regulating the size and branch length of AuNDs, the AuNDs exhibited a broadband absorption from 300 to 1350 nm, with two characteristic absorption peaks located at 1077 and 1265 nm. The AuNDs demonstrated desired optical photothermal conversion efficiency (38.0%), which was further applied in NIR-II photoacoustic imaging (PAI) and PTT in human colon cancer cells (HCT 116)-tumor-bearing mice model. The tumor cells could be effectively eliminated in vivo under 1064 nm laser irradiation by the guidance of PAI. Conclusions We reported a simple but powerful synthetic method to obtain the unique AuNDs with strong and characteristic absorption peaks in the NIR-II window. This study provides a promising solution to tuning the growth of nanoparticles for bioimaging and phototherapy in the NIR-II window. Graphical Abstract
