Early detection and control of foodborne diseases are crucial for ensuring food safety and consumer health. Therefore, there is a need for faster and more sensitive detection methods. We developed a rapid, portable and visible lateral flow biosensor (LFB) for foodborne pathogenic bacteria detection powered by a Pyrococcus furiosus Argonaute nuclease ( Pf Ago), named as Pf Ago-LFB. LAMP (Loop-mediated isothermal amplification) was used to amplify the target nucleic acid . By using Pf Ago as the “lighting up” technology, we can overcome the limitations of false-positive results and lack of effective endpoint monitoring in LAMP, greatly enhancing its applicability. It turned out that Pf Ago-LFB was able to provide yes-or-no (positive or negative) results with a limit of detection (LOD) as low as 1 CFU/mL for S. typhimurium and the detection range was between 10 0 to 10 8 CFU/mL. It was able to secure a sample-to-answer test for contaminated food samples in less than 45 min and demonstrated satisfactory selectivity. In summary, we designed a Pf Ago-LFB that rendered a novel, rapid, accurate, easy-to-deploy, on-site and yes or no detection of S. typhimurium , which could be generalized for building a “one-for-all” biosensing platform.

Dental implants face challenges such as poor osseointegration, delayed angiogenesis, and chronic inflammation. Endogenous gaseous signaling molecules hydrogen sulfide (H 2 S) and nitric oxide (NO) have emerged as key gasotransmitters that regulate angiogenesis, osteogenic differentiation and immune responses, making them attractive candidates for microenvironment-targeted therapies. Here, we engineer a dual gasotransmitter-releasing coating by co-electrospinning hydrogen sulfide (H 2 S) and nitric oxide (NO) donors, JK-2 and S-Nitroso- N -acetyl-DL-penicillamine (SNAP), into gelatin/polycaprolactone (Gel/PCL) nanofibers (NFs). These NFs were applied to laser-microgrooved titanium (Ti) surfaces, further modified with polyepigallocatechin gallate (pEGCG) to ensure stable attachment. The resulting coating exhibited sustained, pH-responsive release of H 2 S and NO, which acted synergistically to enhance cellular responses. In vitro , the dual gasotransmitter-releasing NFs coating enhanced endothelial cell migration, tube formation, and upregulated angiogenic markers (VEGF, TGF-β1, eNOS). Additionally, MC3T3 preosteoblasts showed increased alkaline phosphatase (ALP) activity, mineralization, and upregulation of osteogenic genes (Runx2, COL1, OPN, OPG). RNA-sequencing identified enriched pathways related to histone methylation and oxidative phosphorylation, crucial for osteogenesis and cellular metabolism. Macrophages cultured with these NFs demonstrated suppressed M1 pro-inflammatory markers and enhanced M2 polarization, indicating a shift toward a more regenerative immune response. In vivo , the NFs coating significantly improved neovascularization, bone volume, collagen deposition, and peri-implant tissue thickness. This synergistic release of H 2 S and NO provides a promising strategy to enhance osseointegration and bone regeneration by modulating angiogenesis and immune responses.

Diabetic wound infection remains a devastating threat to human health, largely due to bacterial colonization and increased antibiotic resistance during conventional treatments, and alternative therapeutic strategies are thus urgent to improve diabetic wound healing. Herein, we developed a multifaceted nanoplatform (CuO@SiO 2 @NO@Au, CSNA NPs) consisting of a cupric oxide (CuO) core, a mesoporous silicon nanoshell loaded with nitric oxide (NO), and in situ grown ultrasmall Au nanoparticles (NPs) for improved diabetic wound treatment. The results showed that the prepared CSNA NPs exhibited remarkable dual-enzyme mimic activity of glucose oxidase (GOx) and peroxidase (POD), effectively oxidizing glucose to generate gluconic acid, thereby reducing the glucose levels and reversing the acidic wound microenvironment. In addition, the fabricated nanoplatform generated abundant H 2 O 2 , which was converted into highly toxic hydroxyl radical ( · OH), leading to efficient bacterial eradication that was subsequently. Under near-infrared (NIR) light irradiation, the CSNA nanozyme also triggered the release of NO gas and aided in the removal of bacterial biofilms, collectively improving the wound microenvironment. By integrating chemodynamic therapy (CDT), photothermal therapy, and NO gas therapy, this self-activatable NIR- augmented nanozyme provides a promising antimicrobial strategy for diabetic wound treatment.

Hexagonal boron nitride (hBN) was used as a CuOX/TiO2 catalyst carrier and its effect on NO reduction with NH3 was studied. After hBN was treated with concentrated HNO3, CuOX/TiO2 nanoparticles were dispersed well onto hBN, and the addition of hBN was found to promote NO oxidation, while at the same time suppress NH3 oxidation to NO, and thus promoted the selective catalytic reduction of NO at reaction temperatures between 150 to 350 °C, and a high de-NOX efficiency of 90.6% was achieved at 275 °C. Our study indicates that hBN is a promising catalyst promoter and carrier with excellent stability compared to carbonaceous materials.

Diabetic wounds represent a common and challenging complication of diabetes mellitus, characterized by a complex pathological microenvironment that includes excessive reactive oxygen species (ROS), chronic inflammation, hypoxia, and impaired nitric oxide (NO) synthesis. These factors collectively contribute to delayed wound healing, increased infection risk, and potential progression to chronic non-healing ulcers. Herein, we constructed a multifunctional injectable hydrogel (Prussian blue nanozyme and S-nitrosothiol-loaded hydrogel, PBE&SNO@HG) co-loaded with multi-enzyme-mimicking Prussian blue nanozymes (PBE) and an ultrasound-responsive nitric oxide (NO) donor (SNO). This composite hydrogel exhibited favorable injectability and tissue adhesiveness. The incorporated PBE nanozymes simultaneously mimicked superoxide dismutase (SOD) and catalase (CAT), enabling the scavenging of superoxide anions (⋅O 2 - ) and hydrogen peroxide (H 2 O 2 ), thereby reducing oxidative stress levels at the wound site. Additionally, the ultrasound (US)-responsive release of NO promoted angiogenesis and alleviates inflammatory responses in the wound area. Animal experiments demonstrated that this hydrogel system significantly enhances collagen deposition, accelerates re-epithelialization, promotes hair follicle regeneration, and accelerates wound healing. This study demonstrated the synergistic effect of combining antioxidant strategies with gas therapy in diabetic wound treatment, while also indicating that US-responsive hydrogel materials can achieve on-demand release of therapeutic molecules via external field stimulation, thereby providing a novel strategy for chronic wound management.

Rationale: Chronic diabetic wounds present significant therapeutic challenges due to biofilm resistance and dysregulated metabolism of glucose, reactive oxygen species (ROS), and nitric oxide (NO), which collectively exacerbate immunosuppression and impair tissue repair. Methods: This study developed a glucose-driven cascade nanozyme-loaded dissolvable microneedle system (PPLG@MN) that enables spatiotemporal regulation of the wound microenvironment through a closed-loop mechanism involving nutrient deprivation, gas modulation, and immune reprogramming. The system consists of porous Prussian blue nanozymes (PPB) loaded with glucose oxidase (GOx) and L-arginine (L-arg), which is precisely delivered into the tissue via microneedles. Results: PPLG@MN initiates a self-sustaining therapeutic cycle within the biofilm microenvironment, in which GOx catalyzes the oxidation of glucose to generate hydrogen peroxide (H 2 O 2 ), inducing bacterial starvation. Subsequently, H 2 O 2 reacts with PPB and L-arg to release oxygen (O 2 ) and biofilm-disrupting NO, thereby alleviating local hypoxia and enhancing antibacterial efficacy. Furthermore, the synergistic action of O 2 and NO reprograms macrophages toward an anti-inflammatory M2 phenotype (approximately 30-fold increase compared to the model group), effectively resolving inflammation and promoting angiogenesis. In vivo studies confirmed that the system achieved > 99.9% biofilm eradication efficiency and accelerated wound healing by 25.6% compared to the model group. Conclusion: This nanoplatform offers a clinically translatable therapeutic strategy for biofilm-associated diabetic wounds by synergistically combining gas therapy and immune reprogramming.

H2O2 is a vital atmospheric oxidant, while its photolysis in the troposphere is negligible in conventional view. For the first time, we find that gas-phase H2O2 can undergo heterogeneous photolysis to produce OH radicals on the surfaces of particles such as SiO2, Fe2O3, and Na2SO4 under solar irradiation, which further convert NO to produce HONO and NO2. It is demonstrated that acidity can inhibit the formation of HONO and promote the formation of NO2 significantly. Moreover, the heterogeneous photolysis of H2O2 is dependent on the reactivity of particles, with inert surfaces being more conducive to the generation of OH radicals. A MCM box model indicates that the HONO formation rate via the H2O2 heterogeneous reaction can reach 0.10 ppb·h–1 at 9 a.m., and the relative contribution to the total daytime HONO production can account for approximately 5% at 1 p.m. Given the ubiquitous presence of SiO2 in mineral dust particles, glass curtain walls, and terrestrial soils, these findings indicate that heterogeneous photolysis of H2O2 on the particle surface could be a potential source of daytime HONO. Our research also underscores the importance of atmospheric interfacial chemistry and its contribution to unknown sources of atmospheric oxidizing capacity.

A metal-free PEI/Nv-C 3 N 4 photocatalyst enables efficient gas-phase NO oxidation via an in situ photo-self-Fenton process. • Nitrogen vacancies and PEI cooperatively regulate interfacial electronic structure, O 2 activation, and proton-coupled electron transfer. • PEI functionalization switches ROS evolution from a ·O 2 ⁻ pathway to an H 2 O 2 /·OH-dominated oxidation pathway. • PEI/Nv-C 3 N 4 achieves 85% NO removal, 98.4% nitrate selectivity, and stable operation over 800 min under visible light. Photo-Fenton processes are highly effective for pollutant degradation, yet their application to gas-phase NO oxidation remains limited by metal leaching, external H 2 O 2 addition, and inefficient oxidant utilization. Herein, we develop a metal-free photocatalyst based on polyethyleneimine-functionalized nitrogen-vacancy-rich graphitic carbon nitride (PEI/N v -C 3 N 4 ) for efficient NO oxidation via an in situ photo-self-Fenton process under visible light irradiation. Nitrogen vacancies serve as the primary sites for O 2 adsorption and activation, while PEI functionalization reconstructs the interfacial electronic structure by introducing defect-molecule coupled mid-gap states and a proton-rich microenvironment. This molecular-defect synergy alters the oxygen reduction pathway from a conventional one-electron route toward a selective two-electron process (O 2 → ⁎OOH → H 2 O 2 ), followed by efficient in situ H 2 O 2 activation into ·OH radicals. Simultaneously, PEI enhances NO adsorption and stabilizes NO-derived intermediates, facilitating their complete oxidation into nitrate. As a result, PEI/N v -C 3 N 4 achieves 85% NO removal efficiency, 98.4% nitrate selectivity, and stable operation over 800 min without introducing metal components. Combined experimental and theoretical analyses reveal a mechanistic transition from a ·O 2 - -dominated pathway to an H 2 O 2 /·OH-mediated photo-self-Fenton process. This work provides a general molecular-defect co-engineering strategy for regulating ROS evolution in metal-free photocatalytic systems toward sustainable air purification. Enzyme-mediated bacterial active endocytosis enhanced the permeability of NG/NO to biofilms. • The chemical structures of Schiff base and NONOates provided dual proton-responsive of gas release. • NO induces S-nitrosylation of PDEs in S. mutans 。, thereby reducing lactate production. Dental caries in permanent teeth have remained one of the most prevalent oral diseases over the past three decades, with their global incidence exhibiting a steady upward trajectory. Streptococcus mutans ( S. mutans ) colonization on dental surfaces, coupled with its acidogenic metabolic activity, constitutes a primary etiological factor in caries pathogenesis. In this work, a NO-loaded polysaccharide nanogel, NG/NO was developed to address this issue. Dextran component of the NG/NO demonstrated selective targeting capabilities, promoting active internalization by S. mutans through enzyme-mediated endocytosis. This mechanism significantly enhanced biofilm penetration of the NG/NO (>70% biofilm penetration depth) within a 4-h exposure. The Schiff base bonds and N-diazeniumdiolates conferred the dual-acid-responsive NO release kinetics, achieving mM concentration of NO liberation within several minutes under cariogenic conditions (pH 4.5–5.5), thereby triggering rapid biofilm disrupting (>80% biomass reduction). Moreover, lower concentration of NO was shown to inhibit glycolysis and synthesis of lactic acid in S. mutans , thus improving acidic environment and demonstrating the potential to prevent enamel demineralization and dental caries. In conclusion, the NG/NO has promising applications in daily oral hygiene maintenance and caries prevention.

1, 8-Cineole (Cin), a cyclic monoterpenoid derived from tea trees and eucalyptus species, exhibits diverse pharmacological properties. Yet, its therapeutic impact and underlying mechanism against Staphylococcus aureus (S. aureus) pneumonia remain to be elucidated. In this study, metabolomics based on UPLC-MS/MS was integrated with network pharmacology, molecular biology, and molecular docking to investigate the effects of Cin. The findings demonstrated that Cin markedly reduced mortality and lung bacterial load, lessened pulmonary damage while suppressing the levels of proinflammatory factors, including tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in the bronchoalveolar lavage fluid (BALF) of infected mice. Additionally, 19 metabolites, primarily involved in tryptophan metabolism and arginine biosynthesis, were notably modified by Cin via suppressing the enzymatic activity of indoleamine 2, 3-dioxygenase 1 (IDO1) and inducible nitric oxide synthase (iNOS), thereby attenuating the inflammatory response. Notably, knockdown of IDO1 or iNOS significantly diminished the anti-inflammation effect of Cin. In conclusion, our study validates the therapeutic potential of Cin against S. aureus pneumonia via anti-inflammation by downregulating IDO1 and iNOS. Our results provide a theoretical basis of natural substances applied in bacterial pneumonia treatment.

Early aging is critical for colour formation in tobacco leaves, yet the underlying metabolic mechanisms remain elusive. This study integrated image processing with untargeted and targeted LC-MS to construct a metabolic network characterising colour evolution in flue-cured tobacco (cv. Cuibi-1, CB-1 ) during early aging (0–120 days). Results indicated that leaf colour significantly brightened during the first 30 days, followed by deepening and reddening. Specifically, L* and b* values showed an initial increase followed by a decrease, whereas the a* value exhibited a sustained upward trend. The constructed metabolic network was anchored in saccharide metabolism, connecting downstream phenolic, flavonoid, sphingolipid, and terpenoid pathways. Sixteen differentially accumulated metabolites (DAMs) were identified across categories including alcohols, flavonoids, lignans, nucleotides, phenolic acids, saccharides, and xanthophylls. These DAMs exhibited distinct change patterns and significant correlations with colour parameters (L* and a*, r ranging from −0.88 to −0.58 and 0.56–0.74, respectively; P < 0.05). For the first time, this study elucidated the metabolic mechanisms governing colour changes in CB-1 tobacco leaves during early aging for the first time, providing novel insights for assessing aging progression and maintaining high-value raw material quality.

The accurate determination of NO 2 concentration in exhaled gases is imperative for discriminating between patients with respiratory/cardiovascular diseases and healthy individuals. Therefore, it is important to employ a streamlined and expeditious methodology for the detection of NO 2 . Semiconducting single-walled carbon nanotubes (sc-SWCNTs) exhibit immense potential as next-generation gas sensors due to their high specific surface area, distinctive atomic structure, and remarkable electrical responsiveness to the surrounding environment. In this work, two conjugated polymers (fluorene-alt-C-benzene (PFCN) and fluorene-alt-N-benzene (PFNC)) were synthesized by the Suzuki coupling, featuring positional isomerism of imine groups (-CH=N- vs. -N = CH-). The introduction of a phenyl side chain via imine bonds (-CH=N-) not only enhances the polymer but also regulates the alignment of the imine groups via steric and electronic effects, optimizing their interaction with sc-SWCNTs. The resulting polymer/sc-SWCNTs complexes were utilized for sensor fabrication. In PFCN, more electronegative N atom is positioned away from the skeleton-forming main chain, increasing its environmental exposure and enabling effective attraction of electron-deficient NO 2 . Consequently, the PFCN/sc-SWCNTs sensor showed a significantly higher response (27.6 %) to 1 ppm NO 2 , which was 1.7-fold higher than the PFNC/sc-SWCNTs sensor (16.52 %). This study presents novel strategies for designing high-performance sensors by integrating sc-SWCNTs with imine-functionalized polymers, advancing sensitive NO 2 detection for applications such as non-invasive medical diagnostics.
