Light availability modulates the responses of the microalgae Desmodesmus sp. to micron-sized polyvinyl chloride microplastics

Chun Wang,Lijuan Jiang,Wenxi Huang,Changhai Wang,Meilin He

The vertical movement of large-size and high-density MPs in the water column is usually along with dynamic changes in light intensity. However, whether the change in light availability affects the bioeffects of MPs on surrounding microalgae is currently unknown. This study investigated the effects of micron-sized polyvinyl chloride (mPVC, 143.5 μm) microplastics, alone and in combination with light intensity (from 7.5 to 162.5 μmol·m −2 ·s −1 ) on the growth and physiology of Desmodesmus sp. Although mPVC did not impact microalgal growth under optimal light (40 and 93.8 μmol·m −2 ·s −1 ), it could induce a no-contact shading effect, thereby significantly affecting the physiology of Desmodesmus sp. The growth of Desmodesmus sp. exposed to mPVC was enhanced under a high light intensity of 162.5 μmol·m −2 ·s −1 which can induce growth inhibition but was retarded when under a light inadequacy condition (20 μmol·m −2 ·s −1 ), along with a dose-dependent effect. Significantly, the photosynthesis of Desmodesmus sp. was a highly sensitive metabolic pathway to mPVC stress and largely influenced by the plastic particles under different light conditions. Additionally, mPVC modulated the energy metabolism strategy of Desmodesmus sp., depending on exposure dose and external light availability. Our findings provided a critical basis for the risk assessment of MPs in fluctuating light conditions.

Simultaneously degradation of various phthalate esters by Rhodococcus sp. AH-ZY2: Strain, omics and enzymatic study

Zhengyu Hou,Hejuan Pan,Mengjie Gu,Xiaowang Chen,Tongtong Ying,Pei Qiao,Junwei Cao,Haixia Wang,Tong Hu,Lianbao Zheng,Weihong Zhong

Phthalate esters (PAEs) are widely used as plasticizers and cause serious complex pollution problem in environment. Thus, strains with efficient ability to simultaneously degrade various PAEs are required. In this study, a newly isolated strain Rhodococcus sp. AH-ZY2 can degrade 500 mg/L Di-n-octyl phthalate completely within 16 h and other 500 mg/L PAEs almost completely within 48 h at 37 °C, 180 rpm, and 2 % (v/v) inoculum size of cultures with a OD 600 of 0.8. OD 600 = 0.8, 2 % (v/v). Twenty genes in its genome were annotated as potential esterase and four of them ( 3963 , 4547 , 5294 and 5359 ) were heterogeneously expressed and characterized. Esterase 3963 and 4547 is a type I PAEs esterase that hydrolyzes PAEs to phthalate monoesters. Esterase 5294 is a type II PAEs esterase that hydrolyzes phthalate monoesters to phthalate acid (PA). Esterase 5359 is a type III PAEs esterase that simultaneously degrades various PAEs to PA. Molecular docking results of 5359 suggested that the size and indiscriminate binding feature of spacious substrate binding pocket may contribute to its substrate versatility. AH-ZY2 is a potential strain for efficient remediation of PAEs complex pollution in environment. It is first to report an esterase that can efficiently degrade mixed various PAEs.

Study on oxidative stress and nitric oxide response mechanisms in Gymnodinium sp. under polystyrene microplastic stress

Ren Zhen,Hu Zheng–Xin,Deng Xue,Li Pei–Feng,Liu Chun–Ying,Duan Shan–Shan,Kong Xiang–Feng,Yang Gui–Peng

The toxic effects of microplastic pollution on marine microalgae have attracted considerable attention, yet the underlying mechanisms remain unclear. This study used the dinoflagellate Gymnodinium sp. as a model organism to investigate the effects of polystyrene (PS) microplastics at different particle sizes (0.1 and 10 μm) and concentrations (0–50 mg L –1 ) on its growth, photosynthesis, oxidative stress, and nitric oxide (NO) metabolism. Results indicated that PS microplastics significantly inhibited Gymnodinium sp. growth, chlorophyll a (Chl a ) concentration, and photochemical efficiency by 16.4%, 40.3%, and 72.0% respectively, with smaller particles (0.1 μm) exhibiting stronger inhibitory effects. Low concentrations (10–20 mg L –1 ) of microplastics might temporarily enhance algal density and Chl a content, yet exhibited long–term inhibitory effects. Microplastic exposure induced significant reactive oxygen species (ROS) accumulation by 1.96 times, with ROS levels positively correlated to microplastic concentration and exposure duration, indicating oxidative damage as a key mechanism of microplastic toxicity. Microplastic stress markedly elevated nitrate reductase activity (NRA) by 2.86 times and altered NO release patterns–exhibiting NO uptake at low concentrations, shifting to net release at high concentrations. The dynamic changes in NO revealed its dual role in microalgal stress resistance: initially acting as an antioxidant to alleviate stress, but at higher concentrations potentially exacerbating cellular damage. This study elucidates the mechanisms by which microplastics inhibit microalgal growth through physical shading and oxidative stress, and proposes the key regulatory role of NO metabolism in microplastic stress responses, providing a theoretical basis for assessing the ecological risks of marine microplastics.

Breaking the activity-stability trade-off in oxygen reduction via sp-hybridized nitrogen/nanoparticle dual-regulation

Shizhe Liu,Yang Liu,Yi Ren,Menghao Kong,Tianfang Yang,Shilong Li,Pinghao Cui,Xinxin Jia,Liyuan Wang,Shuyan Gao

Dual-regulation via sp-N and Co nanoparticles optimizes the Co d-band center. • Operando SERS reveals rapid interfacial *OOH conversion for efficient 4e − ORR. • CoNP/Co@sp-N breaks the inherent ORR activity-stability trade-off effectively. • Catalyst yields 0.89 V E 1/2 and enables Zn-air batteries with >240 h stability. Single-atom catalysts have drawn massive interest for their ability to electrocatalyze the oxygen reduction reaction (ORR), primarily attributed to their ultimate atom economy and structurally distinct active centers. Nevertheless, simultaneously achieving robust durability and exceptional catalytic efficiency persists as a critical bottleneck. Herein we develop a graphyne-based composite material (CoNP/Co@sp-N) composite wherein sp-hybridized nitrogen (sp-N) anchored Co single atoms are synergistically coupled with adjacent cobalt nanoparticles. Density functional theory (DFT) reveals that while strong sp-N coordination provides robust atomic anchoring, the sp-N/nanoparticle electronic synergy precisely optimizes the Co d-band center to achieve near-ideal OOH binding, effectively decoupling the inherent ORR activity-stability trade-off. The unique coordination environment provides formidable orbital hybridization for unyielding atomic anchoring while the nanoparticle-induced electronic modulation optimizes the adsorption free energy of OOH intermediates. This synergistic integration bypasses the traditional compromise by simultaneously enhancing electrocatalytic efficiency and preserving operational stability. Notably, the CoNP/Co@sp-N composite delivers outstanding oxygen reduction performance, achieving a half-wave potential of 0.89 V, and exhibits zero efficiency loss following 5000 continuous cycles. Moreover, when incorporated into a functional zinc-air battery, this catalyst attained a peak power density of 187.33 mW cm −2 and exhibited remarkable cycling stability surpassing 240 h, thereby underscoring its significant potential for extensive energy conversion applications. A dual-regulation strategy breaks the oxygen reduction activity-stability trade-off via a graphyne-based CoNP/Co@sp-N composite. Strong sp-hybridized nitrogen coordination ensures robust atomic anchoring, while adjacent cobalt nanoparticles electronically modulate the Co single atoms to optimize OOH binding. This synergistic architecture drives highly efficient and durable O 2 -to-H 2 O conversion for advanced zinc-air batteries. Elucidation of the substrate specificity of Type III phthalate esters esterases from Acinetobacter sp. DY-1

Pei Qiao,Yi Guo,Yanan Liu,Chenghua Zhu,Di Cheng,Youshan Zhou,Panpan Song

Phthalate esters (PAEs) are pervasive environmental pollutants that require efficient biodegradation. The substrate specificity of PAEs-degrading microorganisms hinges critically on the catalytic selectivity of microbial esterases. To elucidate the molecular basis of the specificity of Type III PAEs esterases, we developed a chromogenic assay and further integrated machine learning-driven enzyme mining with functional characterization and molecular dynamics analysis. A novel microorganism, Acinetobacter sp. DY-1, was isolated with diverse PAEs-degrading capacities. The esterase 1952 from strain DY-1 exhibited broad substrate specificity and high catalytic efficiency, with a marked preference for long-chain PAEs, such as DEHP. Molecular dynamics simulations revealed stable hydrophobic interactions underpinning 1952s affinity for DEHP. Through targeted mutagenesis, we identified key residues (Leu64, Leu68, and His229) that are essential for substrate binding and catalysis. Their substitution with polar residues drastically reduced DEHP degradation, while the G178Y mutant enhanced activity via π-stacking interactions. This study deciphers the structural determinants of PAE esterase specificity, enabling precise enzyme engineering.

Glucose promotes iron bi-directional redox transformation mediated by a single strain Comamonas sp. HJ-2

Rong Peng,Rui Yang,Yuhang Liang,Haichuan Sun,Hui Liu

In anaerobic underground environments, the iron redox cycle is fundamentally driven by microbial activities, wherein the metabolic capacity of individual strains plays a pivotal role. This study investigated the regulatory mechanisms of three carbon sources — acetate, glucose, and lactate — on the iron redox cycle mediated by Comamonas sp. HJ-2 under neutral pH anaerobic conditions in Fe(III)–Fe(II) coexisting and Fe(III)-free systems. The results demonstrated that glucose enhanced Fe(II) bio-oxidation and Fe(III) bio-reduction by promoting energy metabolism. Glucose promoted sustained FMN accumulation, which is consistent with a potential contribution of the glycolysis-pentose phosphate pathway-riboflavin synthesis axis, which accumulated to a final concentration of approximately 0.40 μM. The accumulated FMN may function as an electron shuttle and thereby contribute to Fe(III) reduction. Mineralogical analyses further showed system-dependent effects of glucose. In ferrihydrite-containing systems, glucose inhibited the transformation of ferrihydrite toward more crystalline iron phases, whereas in ferrihydrite-free systems it shifted the product distribution toward a higher proportion of ferrihydrite and a lower proportion of lepidocrocite. Glucose also increased the content of surface-bound Fe(II) on minerals. In conclusion, glucose emerged as the most effective carbon source for optimizing the HJ-2-driven iron redox coupling process, likely due to its association with enhanced energy metabolism, FMN accumulation, and altered mineral phase transformation. This study elucidates the carbon source-dependent mechanisms governing microbial iron cycling in anaerobic environments and highlights the critical role of carbon source selection in enhancing iron biotransformation efficiency.

Biochar-assisted biotransformation of 2,4,6-trinitrotoluene by Pseudomonas sp. CMR5c T22: synergistic mechanisms and metabolic reprogramming

Bin Dong,Chengxu Lai,Mengwei Han,Sanping Zhao,Songhao Luo,Yongbing Zhu

2,4,6-Trinitrotoluene (TNT) contamination presents serious threats to ecological safety and human health due to its high toxicity, carcinogenicity, and environmental persistence. Although microbial bioremediation is eco-friendly and cost-effective, its efficiency is often limited by low removal rates and weak microbial tolerance under TNT stress. This study utilized biochar to facilitate microbial biotransformation of TNT and systematically elucidated its synergistic mechanisms. Four types of biochar from different waste sources were screened, among which the wood biochar (MBC) exhibited the optimal performance. With MBC amendment, the TNT removal efficiency increased from 46.2% (strain-only control) to 88.7% within 48 h at an initial TNT concentration of 100 mg/L. Meanwhile, the first-order kinetic rate constant rose from 0.01323 h −1 (strain-only) to 0.03987 h −1 , revealing that MBC greatly accelerated the TNT removal rate. Biochar accelerated removal kinetics, alleviated TNT-induced growth inhibition of strain T22 via lowering aqueous TNT concentration, and stimulated extracellular polymeric substance secretion. Untargeted metabolomics revealed 330 up-regulated differential metabolites in the biochar-microbe system, indicating intensive metabolic reprogramming. Mechanistically, MBC directly enhanced TNT transformation via activation of the nitrotoluene degradation and cofactor biosynthesis pathways. The synergistic mechanisms were proposed to include: (1) weakened TNT exposure; (2) enhanced antioxidant metabolism; (3) activated core transformation pathways; and (4) supplemented basal metabolic supply. Although complete mineralization and soil-scale validation are beyond the present scope, our findings establish a mechanistic foundation for designing biochar-microbe systems for enhanced nitroaromatic remediation.

Characterization of a Purine Nucleoside Phosphorylase From Colwellia sp. WH041 for Multienzyme Synthesis of Nucleoside Analogs

Shirong Wang,Yu Zhang,Fan Yang,Fuxue Chen,Xingcheng Shao,Gaofei Liu,Bin Wu

Multienzyme cascades provide an efficient biocatalytic strategy for synthesizing nucleoside analogs from simple sugar donors and nucleobases. In pentose‐to‐nucleoside cascades, purine nucleoside phosphorylase (PNP) catalyzes the terminal N‐glycosylation step and strongly influences product formation and substrate scope. Here, seven putative PNP homologs from microorganisms isolated from low‐temperature environments were evaluated under Ec RK‐ Ec PPM‐PNP cascade conditions. A PNP from Colwellia sp. WH041, designated Col PNP, showed the highest arabinofuranosylation activity and was selected for further characterization. Col PNP displayed optimal activity at 45 °C and exhibited a 12.7‐fold higher catalytic efficiency than Escherichia coli PNP at the same temperature. Molecular dynamics simulations supported the favorable catalytic performance of Col PNP under moderate‐temperature conditions. After optimization of the Ec RK‐ Ec PPM‐ Col PNP cascade, vidarabine conversion reached 58.2% within 12 h. The optimized system was further applied to synthesize diverse nucleoside analogs with conversions of 45.8%–89.8%, and guanosine analogs were obtained through sequential Ec ADA‐mediated deamination. This study expands the PNP toolbox and demonstrates Col PNP as an efficient module for enzymatic nucleoside analog synthesis. A purine nucleoside phosphorylase from Colwellia sp. WH041, Col PNP, is identified as an efficient module for pentose‐to‐purine nucleoside synthesis. Integrated with Ec RK and Ec PPM, Col PNP enables direct conversion of unprotected D‐pentoses and purine bases into diverse nucleoside analogs with up to 89.9% conversion.

Enhanced biodegradation of Baolixile coal by Bacillus sp. XK1 using combined surfactants to produce chain alkanes

Chen Shi,Ya-Kun Wang,Long Yan,Yu-Hong Kang,Guang-Hui Liu,Yong Gao,Ya-Nan Zhu,Xian-Yong Wei,Zhang-Feng Zhou

The biodegradation of coal is an effective way of clean and efficient utilization of coal. However, how to improve the degradation rate has been the focus of attention. The effect of combined surfactants Tr-Rh (Tr-octyl phenoxy poly ethoxy, Rh-rhamnolipid) on the degradation of Baolixile coal by Bacillus sp. XK1 was studied. The degradation experiment showed that compared with non-surfactant, the degradation rate of Tr-Rh increased by 25.4 % and the degradation time was shortened by 2 days. The bacterial growth characteristics experiment found that in the presence of Tr-Rh, the highest OD 600 of Bacillus sp. XK1 reached 1.311 and the alkaline protease activity was 93.09 U/mL, indicating that Tr-Rh promoted the growth of Bacillus sp. XK1 , secreting more alkaline proteases, and increasing the contact with coal. The surface property experiments of coal showed that, the contact angle and adsorption rate of coal sample were 16.2 ° and 68.3 % in the presence of Tr-Rh respectively, which indicated that Tr-Rh improved the hydrophilicity and charge of coal surface, and increased the adsorption of bacteria on the coal surface. Furthermore, the degradation products were characterized by GC–MS and FTIR. Compared with non-surfactant, the chain alkanes of the liquid bioproducts increased from 48.22 % to 66.10 % in the presence of Tr-Rh. FITR analysis indicated that Bacillus sp. XK1 , in the presence of Tr-Rh, degraded certain aromatic rings and side-chain groups in coal samples. This study provides a new method and theoretical basis for the efficient biodegradation of coal by microorganisms to produce chain alkanes.

Cysteine-induced sulfide bioprecipitation enables simultaneous efficient dechlorination and cadmium removal by Pseudomonas sp. CP-1

Yunxia Zu,Xueqi Chen,Zimeng Zhang,Aijie Wang,Zhiling Li

Organohalides and heavy metals often co-contaminate groundwater, however, the biotoxicity of heavy metal strongly inhibits microbial reductive dechlorination activity and existing bioremediation strategies seldom achieve simultaneous removal of both types of pollutants. Here, we demonstrated that cysteine amendment enabled synchronous reductive dechlorination and cadmium (Cd(II)) removal in an organohalide-respiring bacterium Pseudomonas sp. CP-1 through cysteine-mediated sulfide bioprecipitation. Batch experiments revealed that cysteine supplementation at a Cd(II):Cys molar ratio of 1:2 achieved an enhanced 2,4,6-trichlorophenol dechlorination kinetics ( k d  = 0.28 d -1 ) and 91.20% recovery of Cd(II) as nanocrystalline CdS deposited on the cell surface. In simulated continuous-flow biobarriers, the addition of cysteine sustained high removal efficiencies for both contaminants despite hydraulic fluctuations, with a highest dechlorination rate reaching 120 µM·(L·d) -1 , 9.6 times higher than the pure dechlorination process. Multi-omics analysis revealed the potential coupled metabolic mechanism of dechlorination and Cd(II) removal in which the amendment of cysteine not only activated cysteine desulfurase to produce H 2 S for extracellular precipitation of CdS, but also generated alanine and pyruvate, which likely assimilated into the TCA cycle to augment NADH production and electron supply for reductive dechlorination. This work establishes a unique single-strain based strategy for synergistic remediation of organohalide and heavy metal ions in groundwater, highlighting the potential of cysteine-driven metabolism as an effective bioaugmentation tool for sustainable remediation of organohalide and heavy metal co-contaminated groundwater.

Resuscitation-promoting factor stimulates PCB dechlorination and mineralization by Stenotrophomonas sp. FS3 under alternating redox conditions

Sijia Fei,Wenjie Yu,Faqian Sun,Xiao Xiao,Chaofeng Shen,Xiaomei Su

Bioremediation of highly chlorinated polychlorinated biphenyls (PCBs) typically requires sequential anaerobic-aerobic treatment, highlighting the need for robust facultative anaerobic degraders. In this study, a novel facultative anaerobic strain, Stenotrophomonas sp. FS3, was resuscitated with resuscitation-promoting factor (Rpf) under alternating anaerobic-aerobic conditions. Whole-genome sequencing revealed a comprehensive set of genes for biphenyl, benzoate, catechol, and protocatechuate catabolism, indicating strong intrinsic potential for PCB biodegradation. FS3 achieved 61.2 % removal of Aroclor 1260 at 10 mg/L after 30 days, and Rpf supplementation significantly enhanced both anaerobic dechlorination and aerobic degradation, resulting in a 1.66-fold increase in removal efficiency and greater chloride ion release relative to controls. Viability and morphophysiological analyses revealed that Rpf preserved cell integrity and metabolic activity, preventing the transition into a viable but non-culturable (VBNC) state. Transcriptomic analysis revealed that Rpf supplementation significantly upregulated bph cluster genes and downstream catabolic genes (e.g., pobA , ligB , pcaGH ) without altering the overall degradation pathway, which proceeded via the HOPDA-benzoate pathway followed by catechol- and protocatechuate-mediated mineralization. Collectively, this work establishes FS3 as a robust degrader suitable for PCB bioremediation under fluctuating redox conditions and underscore Rpf supplementation as a viable strategy to enhance the resilience and metabolic capacity of PCB-degrading bacteria.

Pseudomonas sp. ST4 produces variety of active compounds to interfere fungal sexual mating and hyphal growth

Shiyin Liu,Fei He,Nuoqiao Lin,Yumei Chen,Zhibin Liang,Lisheng Liao,Mingfa Lv,Yufan Chen,Shaohua Chen,Jianuan Zhou,Lian-Hui Zhang

Summary Sexual mating of compatible sporida is essential for Sporisorium scitamineum to form dikaryotic mycelia and then cause infection on sugarcane. Our previous work identified a Pseudomonas sp. ST4 from a soil sample, which showed a promising biocontrol potential by inhibiting the mating of S. scitamineum sporida and hyphal growth. In this study, we set to isolate the active compounds from Pseudomonas sp. ST4 through solid fermentation. High-performance liquid chromatography (HPLC) separation coupling with bioassay showed that Pseudomonas sp. ST4 produced a range of antimicrobial compounds. Two of the major components were purified following acetate extraction, silica gel and HPLC separation. Nuclear magnetic resonance (NMR) and liquid chromatography–mass spectrometry (LC-MS) analysis identified these active compounds are 4-hydroxybenzaldehyde and indole-3-carbaldehyde respectively. Further analysis showed that the former compound only inhibited the hyphal growth of the fungus at a concentration of 3 mM, while the latter interfered the fungal sexual mating at a concentration of 0.6 mM and affected hyphal growth at a concentration of 2 mM. Treatment of corn plants with 3 mM indole-3-carbaldehyde significantly inhibited corn smut infection, with a control rate up to 94%. Further analysis of the structure and activity relationship revealed that indole has a much stronger inhibitory activity against the fungal sexual mating than indole-3-carbaldehyde. The results from this study provide new agents for control and prevention of the sugarcane smut disease, and the active compounds could also be used to probe the molecular mechanisms of fungal sexual mating. Graphical 4-hydroxybenzaldehyde and indole-3-carbaldehyde from Pseudomonas sp. ST4 were isolated and identified, the former of which inhibited hyphal growth of Sporisorium scitamineum at a concentration of 3 mM, and the latter of which interfered the fungal sexual mating at a concentration of 0.6 mM, and affected hyphal growth at a concentration of 2 mM and has a good control efficacy against sugarcane smut.
