A Decision-Oriented Calibrated Screening Workflow for Tylosin Derivatives: Closed-Loop MIC Validation Against Staphylococcus aureus and Streptococcus agalactiae

Huan Liu,Yiming Liu,Na Yu,Miao An,Yaoxin Tang,Jing Qu,Xiubo Li

Background:Antimicrobial resistance among Gram-positive veterinary pathogens has increased the need for more rational and experimentally grounded strategies to prioritize tylosin derivatives for antibacterial development. This study aimed to develop and externally a calibrated, decision-oriented screening workflow for tylosin-derived antibacterial analog prioritization againstStaphylococcus aureusandStreptococcus agalactiae. Methods:Publicly available minimum inhibitory concentration (MIC) data were used to construct organism-specific multilayer perceptron models. Model performance was evaluated using a five-fold out-of-fold framework. Calibrated activity probabilities were then combined with precision-oriented thresholds and similarity-based applicability-domain constraints to define prospective Go/No-Go decisions. To examine external transferability, six synthesized tylosin derivatives (A1–A6) were prospectively predicted and subsequently tested using in vitro MIC assays. Results:Internal validation showed stronger and more stable performance forS. aureusthan forS. agalactiae. However, closed-loop external validation revealed distinct organism-specific decision behaviors. ForS. aureus, the workflow assigned Go decisions to five compounds and included the only experimentally active analog, A6, but also generated several false Go decisions for inactive analogs. ForS. agalactiae, all six compounds were classified as No-Go under the primary decision rule, whereas MIC testing showed that four analogs were experimentally active, indicating conservative under-selection in a low-data extrapolation setting. Conclusions:Calibrated probabilities and applicability-domain analysis can improve the transparency and diagnostic value of antibacterial prioritization, but they do not guarantee robust external activity prediction when training-set coverage, threshold transferability, or chemical-space representation is limited. Overall, this study provides an early-stage, risk-aware closed-loop decision-support framework for tylosin-derived analog prioritization and highlights the need for iterative model updating, probability recalibration, and larger external validation before such workflows are used as high-confidence prospective screening tools in veterinary antibacterial discovery.

Development of a Novel Probe-Based Dual-Detection platform System for Flexible Detection of Staphylococcus aureus across Transmission Pathways

Shengyang Wang,Wenjin Hu,Yuanrong Li,Jin Qi,Yi Luo,Yaling Liu

Immunodetection plays a crucial role in the prevention and treatment of Staphylococcus aureus infections. However, existing methods often suffer from limited performance, poor quantification, and applicability due to the use of low-sensitivity, nonspecific, and nontargeted probes. To address these challenges, we identified high-affinity, specific phage-displayed nanobody probes and established a dual-detection platform that enabled accurate and flexible screening of S. aureus across diverse settings. High-affinity phage-displayed nanobodies targeting S. aureus were selected to replace the conventional probes and were randomly paired with five monoclonal antibodies to demonstrate their universality. To further enhance detection sensitivity, we synthesized and optimized a core–shell magnetic–fluorescent nanolabel (Fe3O4@PS@TQD), which outperformed traditional colorimetric and fluorescent tags. By integrating these advanced components, we established a dual-detection platform capable of qualitative detection within 10 min and quantitative analysis within 30 min, achieving superior sensitivity and applicability compared with recently reported methods and commercial kits. The system exhibited excellent specificity, reproducibility, accuracy, and stability, with a detection limit as low as 5 CFU/mL after 4 h of pre-enrichment. In summary, this study presents a novel probe-driven dual-detection platform that meets the urgent need for rapid S. aureus on-site detection in complex environments.

Electrochemical cell respiration strategy by Cu(II)-chitosan functionalized carbon paper for rapid and sensitive detection of staphylococcus aureus

Feng Hu,Mengyi Liu,Jubo Zhang,Fangjiu He,Xiaowen Shi

This study presents an innovative electrochemical biosensor for the rapid and sensitive detection of Staphylococcus aureus ( S. aureus ), a significant foodborne pathogen. The biosensor is based on carbon paper modified with a Cu(Ⅱ)-crosslinked chitosan hydrogel film. Unlike conventional methods that rely on biomolecular recognition, this paper sensor detects S. aureus by measuring its unique respiration cycle, specifically its ability to reduce ferricyanide to ferrocyanide in the presence of mannitol. The chitosan hydrogel enhances the surface concentration of ferrocyanide and exhibits anti-fouling properties, while Cu(Ⅱ) crosslinking improves conductivity and diffusion. The biosensor demonstrates high sensitivity (0.082 μA/μM in I-t tests and 5.31 μA/μM in DPV tests) and low limits of detection (3.47   μM in I-t tests and 0.64   μM in DPV tests). It achieves a detection linear range of 10 0 to 10 5 CFU/mL with an LOD of 3.4 CFU/mL as well as excellent repeatability (RSD of 4.2%) and stability (retaining 83.4% of its initial signal after 15 days). Its performance in real food and wound samples confirms its high sensitivity and reasonable recovery rates, even in complex matrices. This research provides a strong foundation for the application of electrochemical biosensors in food safety monitoring and pathogen detection.

Inactivation Effects of Plasma-Activated Water on Fungal-Bacterial Polymicrobial Biofilms of Candida albicans and Methicillin-Resistant Staphylococcus aureus

Sihong Ma,Tianyi Song,Pengyu Zhao,Haoqian Chen,Tianshuo Yan,Li Guo,Miaojing Li,Zifeng Wang,Dingxin Liu,Xiaohua Wang,Mingzhe Rong

Mixed infections caused by fungal-bacterial polymicrobial biofilms pose major clinical challenges, and plasma-activated water (PAW) has potential for the treatment of infections. In this study, Candida albicans and methicillin-resistant Staphylococcus aureus (MRSA) were used to establish fungal-bacterial biofilms, and the inactivation effects of PAWs on the two microorganisms in biofilms were evaluated. The 30-min PAW treatment reduced the viable C. albicans and MRSA within biofilms on silica films to below 2.00-log 10 CFU/mL. The two consecutive 30-min PAW treatments achieved significant reductions of approximately 3.32-log 10 CFU/mL for C. albicans and 4.08-log 10 CFU/mL for MRSA in biofilms on wound-mimicking porcine skins. Therefore, owing to its non-selective inactivation ability, PAW provides an alternative therapy for the mixed infections caused by diverse microorganisms.

An osmium-modified Prussian blue heterostructure for biofilm disruption and oxidative stress to combat methicillin-resistant Staphylococcus aureus infections

Xia Chen,Xinlin Jia,Chaohong Yu,Xilong Li,Jinglin Zou,Yuanqing Mao,Yongsheng Li

OsCoPBA possesses high-affinity binding to bacterial biofilm and can disrupt biofilms through its photothermal effect. • OsCoPBA with responsive catalysis selectively generated bactericidal ROS via the POD-like activity in acidic wounds. • Interfacial modification enhanced the CAT- and SOD-like activities of OsCoPBA to scavenge excess ROS, accelerating healing. One major contributor to the therapeutic recalcitrance of methicillin-resistant Staphylococcus aureus (MRSA) infections is its robust biofilm formation, which impedes therapeutics penetration, diminishes bacterial susceptibility to exogenous stimulus and exacerbates host inflammation, collectively leading to suboptimal therapeutic outcomes. To address these interrelated challenges, we developed an osmium-modified cobalt‑iron Prussian blue analogue (OsCoPBA) heterostructure with high-affinity binding to MRSA biofilm and exceptional photothermal conversion efficiency under near-infrared (NIR) irradiation, thereby enabling effective biofilm disruption. The OsCoPBA heterostructure was characterized by its redox property, imparting both pro-oxidant and anti-oxidant capabilities under different conditions. In acidic wounds, OsCoPBA exhibited a peroxidase (POD)-like activity, generating bactericidal reactive oxygen species (ROS) to induce oxidative stress within MRSA. Conversely, during the wound healing phase, OsCoPBA exhibited robust superoxide dismutase (SOD)-like activity (a peak ·O₂ − scavenging efficiency of 95% at the optimal concentration of 250 μg mL −1 ) and enhanced catalase (CAT)-like activity, effectively scavenging excess ROS to suppress inflammation. This heterostructure achieved on-demand regulation of ROS for meeting the antibacterial and anti-inflammatory requirements, representing a rational design strategy for managing MRSA-infected wounds. A therapeutic strategy to treat methicillin-resistant Staphylococcus aureus (MRSA)- infected wounds is presented. The OsCoPBA heterostructure exhibits microenvironment-adaptive catalytic performance, imparting both pro-oxidant and anti-oxidant capabilities under different conditions. The photothermal nanozyme (OsCoPBA) can inhibit biofilm formation and regulate ROS on demand to achieve antibacterial and anti-inflammatory effects. A crRNA-optimized CRISPR/Cas12a-based aggregation - dispersion colorimetric biosensor using gold nanoparticles for ultrasensitive detection of Staphylococcus aureus

Wenji Bao,Ailing Su,Gerile Aodeng,Lu Ga,Lan Yu,Jun Ai

A highly sensitive colorimetric sensing platform was developed based on the trans-cleavage activity of CRISPR/Cas12a and the controllable aggregation of gold nanoparticles (AuNPs) for rapid and visual detection of the foodborne pathogen Staphylococcus aureus. Following PCR or RPA, the target DNA was specifically recognized by the Cas12a/crRNA complex, thereby activating its trans-cleavage activity. This process led to the degradation of linker single-stranded DNA (ssDNA) responsible for crosslinking AuNPs probes, resulting in significant changes in both UV–vis absorption signals and solution color. Three different crRNA sequences were systematically evaluated, revealing distinct efficiencies in target recognition and signal amplification. Among them, the crRNA-3 mediated system exhibited superior analytical performance, achieving an ultralow detection limit of 1.14 fM and a good linear response over a low concentration range. Furthermore, the proposed platform demonstrated excellent anti-interference capability in complex matrices, including serum, milk, bovine serum albumin (BSA), and phosphate-buffered saline (PBS), with recovery rates ranging from 83.2% to 125.9%. This work provides a robust and visual tool for the on-site detection of S. aureus and offers new insights into the design of portable CRISPR-based diagnostic platforms.

A highly selective and sensitive electrochemical CS–MWCNTs/Au-NPs composite DNA biosensor for Staphylococcus aureus gene sequence detection

Yange Sun,Xingxing He,Jian Ji,Min Jia,Zhouping Wang,Xiulan Sun

This paper presents a new electrochemical DNA biosensor constructed using a substrate electrode composed of a novel nanocomposite material prepared using gold nanoparticles (Au-NPs) and multiwalled carbon nanotubes (MWCNTs) and further modified with an Au electrode (AuE), which was used as the substrate electrode. A single-stranded DNA (ssDNA) probe was immobilized on the Au-NPs/CS–MWCNTs/AuE electrode by means of facile gold–thiol affinity, which resulted in hybridization with the target ssDNA sequence. Hybridization reactions were assessed by using the reduction peak current of methylene blue (MB) as an electrochemical indicator. The advantages of the nanomaterials were found to include high surface area, favorable electronic properties, and strong electrocatalytic activity . The amount of ssDNA adsorbed on the electrode surface was increased and the electrochemical response of MB accelerated. The differential pulse voltammetric responses of MB were in line with the specific target ssDNA sequence within the concentration range 1.0×10 −15 –1.0×10 −8   M with the detection limit 3.3×10 −16   M (3 σ ). In the colony forming unit (CFU) we were able to detect 10   CFU   mL −1 of Staphylococcus aureus in the tap water, achieving good discrimination ability between one- and three-base mismatched ssDNA sequences. The polymerase chain reaction (PCR) amplification products of S. aureus nuc gene sequence were also detected with satisfactory results.

New approach to overcoming antimicrobial resistance of Staphylococcus aureus by combining Guanghuoxiang (Herba Pogostemonis) and Penicillin G Sodium treatments

Wenguang JING,Zhixia WANG,Wenmin PI,Haonan WU,Minghua LI,Penglong WANG,Xianlong CHENG,Feng WEI

OBJECTIVE: Antibiotics are the main drugs used to treat bacterial infections, which have been extensively utilized across various fields. However, the problem of antimicrobial resistance (AMR) has greatly limited its use, leading to the creation of various superbugs such as Methicillin-resistant Staphylococcus aureus (MRSA), making antibiotics less effective, become a major global public health challenge. MRSA, which is resistant to most β-lactam and cephalosporin antibiotics, poses a significant threat to public health. Combination therapy has shown promise as a strategy to combat multi-drug-resistant bacteria. Chinese medicine also has potential applications in this field. Thus, we tried to find a new approach to overcoming MRSA by combining Chinese herb and penicillin G sodium (PGS). METHODS: The antibacterial activity of Guanghuoxiang ( Herba Pogostemonis ) samples was investigated by turbidimetry. Then using the checkerboard assay, live/ dead bacterial staining and scanning electron microscopy (SEM) to investigate whether the combination of Guanghuoxiang ( Herba Pogostemonis ) samples and PGS could improve the sensitivity of MRSA, and metabolomics was used to investigate the underlying mechanisms. RESULTS: In this study, we find that the Guanghuoxiang ( Herba Pogostemonis ) samples had good inhibitory effects on MRSA, and showed a synergistic effect when combined with PGS, enhancing the sensitivity of MRSA to PGS. Metabolomics data further revealed that this combination exerts a broader impact on the energy and material metabolism of the bacteria, resulting in improved antibacterial efficacy. CONCLUSION: Combining Guanghuoxiang ( Herba Pogostemonis ) with antibiotics could improve the sensitivity of drug-resistant bacteria, provided a new direction for solving the problem of AMR, and offered a valuable strategy for clinical response to MRSA.

Synergistic antibacterial activity of benzyl isothiocyanate and resveratrol against Staphylococcus aureus: Multi-omics perspective insights on cell wall disruption

Jianan Liu,Jinle Ma,Siyang Pei,Siyi Bao,Hongshun Hao,Jingran Bi,Hongman Hou,Gongliang Zhang

Staphylococcus aureus ( S. aureus ) is a typical foodborne pathogen, and the development of novel synergistic antimicrobial strategies is urgently needed. Therefore, the aim of this study was to investigate the mechanism underlying the synergistic inhibition of S. aureus by benzyl isothiocyanate (BITC) and resveratrol (RES). The synergistic effect of BITC and RES on S. aureus was verified by a time-elimination curve, cell hemolysis, intracellular reactive oxygen species accumulation and transmission electron microscopy. The results of the transcriptomic and proteomic analyses revealed that the combination of BITC and RES against S. aureus synergistically downregulated the expression of the cell wall anchoring protein immunoglobulin G-binding protein (Sbi) compared with that in the individual treatments, a finding further supported by qRT-PCR and molecular docking. In addition, synergistic effects significantly disrupted cell wall integrity, alkaline phosphatase leakage increased by 1.1–1.9 times, the zeta potential increased by 3.0–6.3 mV, and the hydrophobicity decreased by 19.2 %–25.0 %. These findings uncover a novel mechanism of synergistic antimicrobial action and provide valuable clues for developing strategies against S. aureus .

A Sustained Oxygen- and Nitric Oxide-Producing Hydrogel Accelerates Methicillin-Resistant Staphylococcus aureus-Infected Diabetic Wound Healing under Hypoxia

Que Bai,Fangfang Hu,Qian Gao,Caiyun Zheng,Yanni Zhang,S. Ramakrishna,Xianglong Wu,Tingli Lu

Chronic inflammation and hypoxia in the diabetic hyperglycemic microenvironment can lead to persistent vascular impairment, hindering tissue regeneration and exacerbating adverse outcomes. We propose a multifunctional regeneration strategy capable of simultaneously releasing oxygen (O2) and nitric oxide (NO) to enhance immune regulation and angiogenesis, significantly accelerating wound healing. Here, a dual-dynamic-network hydrogel was designed for in situ depletion of excess reactive oxygen species (ROS) and immunomodulation to accelerate the remodeling of MRSA-infected diabetic wounds. The hydrogel was constructed using Schiff base and phenylborate ester bonds, with l-arginine-modified manganese dioxide (ACM) nanoparticles integrated into its framework. This hydrogel could respond to ROS in the diabetic microenvironment and subsequently generate oxygen and nitric oxide. Studies have shown that PDM hydrogel exhibited good biocompatibility, promoted cell migration, and facilitated endothelial cell tubular formation. Mechanistically, in vitro and in vivo analyses revealed that PDM could relieve excessive ROS levels, restore mitochondrial membrane potential, down-regulate pro-inflammatory marker CD86, up-regulate anti-inflammatory marker CD206, and promote macrophage polarization toward a reparative phenotype. In MRSA-infected full-thickness skin wounds, the PDM hydrogel alleviated hypoxia by decomposing endogenous H2O2 into O2 (thereby reducing HIF-1α expression), modulated inflammatory responses (downregulating TNF-α and upregulating CD206), enhanced the expression of angiogenic factors VEGF and CD31, and increased collagen I deposition. These findings suggest that the multifunctional PDM hydrogel holds clinical potential for reshaping the damaged tissue microenvironment and offers an effective therapeutic strategy for chronic skin wound regeneration.

A rapid and naked-eye methicillin resistant Staphylococcus aureus screening method based on CRISPR/Cas12a and hybridization chain reaction

Yayun Jiang,Zongyao Chen,Xiao Liu,Qi Xin,Dengchao Wang,Caixia Ji,Youwei Li,Gang Mai

Methicillin-resistant Staphylococcus aureus (MRSA), a widely drug-resistant bacterium, poses a significant threat to global health. Current culturing and nucleic acid detection methods are time-consuming and require complex instruments, which do not meet the detection needs. Herein, we developed a rapid and visual MRSA detection method (MCFHCR) using ssDNA-functionalized magnetic beads as a trigger chain combined with the trans-cleavage activity of the Cas12a protein and fluorescence signal amplification of the hybridization chain reaction (HCR). MCFHCR is a signal-off platform for the detection of MRSA. In the absence of DNA targets, the trans-cleavage activity of Cas12a is inactivated, allowing HCR to proceed and form long double-stranded DNA, resulting in an increased fluorescent signal. In the presence of the DNA targets, the trans-cleavage activity of CRISPR/Cas12a is activated to cleave the trigger strand, failing HCR and leading to a decrease in the fluorescence signal. Combined with RPA, MCFHCR was completed within 35 min, achieving a limit of detection (LOD) of five copies/μL for mecA DNA and 8 CFU/mL for MRSA. In detecting clinical strains, MCFHCR demonstrated comparable performance to qPCR and drug sensitivity testing. Therefore, with its simple, rapid operation and convenient signal acquisition, MCFHCR shows significant practical applicability in detecting MRSA.

Chitosan-based double-network hydrogel with synergistic photothermal/nitric oxide therapy for methicillin-resistance Staphylococcus aureus infected wound healing

Guangzheng Zhang,Yi Liu,Jinmei Wu,Jiahui Xu,Wenqing Wei,Ziyan Yuan,Heyou Han,Linlin Bu,Zhiyong Song

The development of advanced wound dressings that incorporate synergistic antibacterial strategies is essential for addressing antibiotic-resistant infections and facilitating tissue regeneration. This study presents a near-infrared (NIR)-responsive double-network hydrogel (Gel@HS) that combines photothermal therapy (PTT) and nitric oxide (NO) gas therapy for the treatment of methicillin-resistant Staphylococcus aureus (MRSA)-infected wounds. The hydrogel was engineered with acrylamide as the primary network and chitosan, a carbohydrate polymer, as the secondary network, capitalizing on chitosan's natural tissue adhesion and antibacterial properties. Hollow copper sulfide nanoparticles (HCuS) and sodium nitroprusside (SNP) were incorporated to enable photothermal conversion and controlled NO release upon NIR irradiation. The hydrogel demonstrated remarkable mechanical stability, adhesion, and targeted antibacterial activity, facilitating bacterial eradication through PTT-induced membrane disruption and NO-mediated protein inactivation. In vitro experiments confirmed its broad-spectrum antibacterial efficacy (>99 % MRSA elimination) and anti-inflammatory effects via macrophage modulation. In vivo testing using an MRSA-infected murine wound model showed accelerated healing (93 % wound closure within 9 days), reduced bacterial load, and diminished inflammatory cytokine levels. This research highlights the potential of carbohydrate polymer-based hydrogels as multifunctional platforms for combating resistant infections and promoting wound healing through physicochemical synergy.
