The morphology of catalysts is a key factor influencing their photocatalytic performance. Herein, we prepared three W 18 O 49 photocatalysts with different morphologies of sea urchin (W 18 O 49 SU), nanorod (W 18 O 49 NR) and hollow sphere (W 18 O 49 HS), and evaluate their performances in photocatalytic hydrolysis of ammonia borane (AB). Remarkably, W 18 O 49 SU exhibited an impressive photocatalytic H 2 production rate with the highest TOF of 53.1 min −1 under visible light, which is 10.4 and 7.5 times higher than those of W 18 O 49 HS and W 18 O 49 NR, respectively. The comprehensive characterizations and control experiments revealed that the unique sea urchin-like morphology enabled the strong light harvesting by multiple light scattering, fast separation of photogenerated charges and directional electron enrichment by nanotip effect, which collaboratively boost the photocatalytic activity . Moreover, the enhancement effect of sea urchin-like morphology is again approved well on Nb 2 O 5 photocatalysts. This work opens a paradigm to synergistically improve light harvesting and charge separation efficiency through rational design of photocatalyst nanostructure .

As a typical 3d transition metal, Cu exhibits competitive potential in the catalytic transfer hydrogenation of nitriles due to its low price and appropriate catalytic activity . Unfortunately, the Cu-based catalyst usually requires additional metals to realize good catalytic activity, this makes it difficult to understand the role of Cu. Herein, an amino-modified hollow mesoporous double-shell SiO 2 @C (AmHMSiO 2 @C) microsphere was applied as a nanoreactor, which achieves the high selective hydrogenation of nitriles over the Cu-only catalyst. The conversion and selectivity to primary amines were 100% and 96% respectively at 60 °C using amino borane as the hydrogen source, and the TOF reaches 199.75 h −1 for Cu. The unique structure of the Cu@AmHMSiO 2 @C enhances the activity and stability of the Cu active sites. Comparative experiments demonstrate that the synergy between the nanoreactor and Cu nanoparticles enables the desirable catalytic performance toward the hydrogenation of nitriles to primary amines under mild reaction conditions. This work offers a distinct case for constructing an efficient Cu-based catalyst for heterogeneous hydrogenation reactions.

Cu single-atom catalysts (Cu SACs) have been considered as promising catalysts for efficient electrocatalytic CO2 reduction reactions (ECRRs). However, the reports on Cu SACs with an asymmetric atomic interface to obtain CO are few. Herein, we rationally designed two Cu SACs with different asymmetric atomic interfaces to explore their catalytic performance. The catalyst of CuN3O/C delivers high ECRR selectivity with an FECO value of above 90% in a wide potential window from −0.5 to −0.9 V vs RHE (in particular, 96% at −0.8 V), while CuCO3/C delivers poor selectivity for CO production with a maximum FECO value of only 20.0% at −0.5 V vs RHE. Besides, CuN3O/C exhibited a large turnover frequency (TOF) up to 2782.6 h–1 at −0.9 V vs RHE, which is much better than the maximum 4.8 h–1 of CuCO3/C. Density functional theory (DFT) results demonstrate that the CuN3O site needs a lower Gibbs free energy than CuCO3 in the rate-determining step of CO desorption, leading to the outstanding performance of CuN3O/C on the process of ECRR-to-CO. This work provides an efficient strategy to improve the selectivity and activity of the ECRR via regulating asymmetric atomic interfaces of SACs by adjusting the coordination atoms.

In the present study, a comprehensive strategy integrating affinity ultrafiltration high-performance liquid chromatography quadrupole-time-of-flight mass spectrometry (UF-HPLC-Q-TOF-MS), in silico molecular docking and bioassays was established to rapidly screen natural SOD activators from traditional Chinese medicines. As illustrative case studies, Schisandra chinensis, Fructus cnidii and Radix ophiopogonis were chosen to develop and verify the strategy. The HPLC-Q-TOF-MS was used to identify the compounds in comparison with reference standards and literature data. A total of eight compounds, including four biphenyl-cyclooctene ligands from Schisandra chinensis and four coumarins from Fructus cnidii, were found to potentially increase SOD activities. No ligands were found in the extract of Radix ophiopogonis. Then, in silico molecular docking was performed to investigate the binding site and binding affinity of the candidates on SOD. Compared to the nonspecific ligands screened from the extract, the specific ligands presented stronger binding affinities. In addition, the activity and kinetic parameters of the SOD-ligand were investigated through an improved pyrogallol autoxidation method. Gomisin J and xanthotoxin showed a stronger ability to increase SOD activities. The present study indicated that combining UF-HPLC-Q-TOF-MS and in silico molecular docking offers a powerful and meaningful tool to rapidly screen SOD activators from traditional Chinese medicines.

Triclosan (TCS) is a compound in pharmaceutical and personal care products (PPCPs) that preferentially accumulates in soil and may threaten human health. Iron-based persulfate activation processes (Fe-PAPs) are a promising technique in soil remediation but are limited by their narrow pH application range and resultant iron precipitation. In this study, oxalic acid (OA) was specifically introduced to improve the efficiency of Fe 2+ -activated persulfate (PS) processes for TCS degradation in soil. The complexation of OA and Fe 2+ improved the total iron concentration in the Fe 2+ /OA/PS system and prevented iron precipitation. Fe-minerals in soil were also dissolved by OA, which participated in PS activation. The most appropriate proportion of Fe 2+ /OA/PS is 1:1:1, and approximately 85% of the TCS was degraded within 240 min compared with 45% in the Fe 2+ /PS system. SO 4 • - and 1 O 2 played important roles on TCS oxidation, while O 2 • - and CO 2 • - mainly participated in the iron cycle. The weak stretch of Fe-O in OA containing treatments in ATR-FTIR measurements provided evidence that OA dissolved the Fe-minerals. The introduction of OA in the Fe 2+ /PS system in four different soils reduced the negative effect of soil organic matter on TCS degradation. Three types of intermediates including hydroxylation, cleavage of ether bonds and cyclization products, were identified by LC-Q-TOF-MS. Column experiments were used to study the feasibility of in situ chemical oxidation (ISCO) remediation of TCS in the Fe 2+ /OA/PS system. Our findings indicated the ability of OA to be used as a cheating agent in Fe-PAPs for TCS soil remediation.

To systematically characterize the chemical constituents and serum metabolites of Qijingmingmu decoction (QJMM) and to explore its potential anti-aging mechanisms using an integrated strategy combining UPLC-Q-TOF-MS and network pharmacology. The chemical components of QJMM were identified using UPLC-Q-TOF-MS. Rats were administered QJMM at different doses, and serum samples were collected for metabolite profiling. The absorbed prototype components and metabolites were analyzed. Potential targets of QJMM were predicted using public databases, and protein–protein interaction (PPI) analysis, Gene Ontology (GO), and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed. A total of 66 compounds were identified in QJMM, and 38 metabolites were detected in rat serum, including both prototype compounds and their transformed products. Several representative metabolites, such as dihydroferulic acid 4-O-glucuronide, ferulic acid sulfate, and wedelolactone glucuronide, were identified as key in vivo components. Network pharmacology analysis suggested that QJMM may exert anti-aging effects by regulating multiple targets, including RELA, ESR1, TNF, and HSP90AB1, and modulating signaling pathways such as the NF-κB, TNF, and HIF-1 pathways. This study provides a preliminary characterization of the chemical and metabolic profiles of QJMM and offers insights into its potential multi-target mechanisms underlying anti-aging effects.

Clanis Bilineata larva (CBL) is a unique Chinese insect delicacy in Jiangsu Province, which is known for its umami taste and high protein nutrition. To develop its desired flavor, the quantitative descriptive analysis (QDA) combined with high throughput instruments were applied to analyze the sensory attributes and compounds of raw, boiled and roasted CBLs. Through QDA, cooked CBLs showed higher meaty odor intensities than Raw CBL significantly (p < 0.05). Compared with boiled CBL, the roasting process could generate the roasted meaty/beany odors and umami/savory taste while reducing the negative effects of grassy/boiled vegetable/earthy odors and bitter taste. Pearson correlation analyses results showed that meaty odor was highly correlated with compounds produced by fatty acid degradation such as hexanal and 1-octen-3-ol. Meanwhile, histidine was highly correlated with bitter and adenosine monophosphate was highly correlated with umami. The taste and odor of cooked CBLs are mainly formed from the lipid degradation, protein degradation and nucleotide degradation. This study provides insights to use a proper cooking process to develop consumer-friendly CBL products.

Developing candidates from medicinal plants for snakebite treatment is quite promising but still remains challenging. The widespread snake venom phospholipase A 2 (svPLA 2 ) is considered as an important therapeutic target for treating snakebite envenomation. In this study, a ligand fishing based platform was established following the immobilization of svPLA 2 onto Fe 3 O 4 magnetic nanoparticles for the first time. After investigating the enzymatic properties and verifying the ligand fishing efficacy, the developed platform was applied to screen svPLA 2 inhibitors from Hedyotis diffusa Willd . and Paris polyphylla . Three compounds (chlorogenic acid, caffeic acid, 2-hydroxycinnamic acid) from Hedyotis diffusa Willd . aqueous extracts and four compounds (rutin, cynaroside, kaempferol, 2-hydroxycinnamic acid) from Paris polyphylla aqueous extracts were screened and identified as potential inhibitors by coupling with UPLC-Q-TOF/MS. Their inhibitory activity was verified through an enzymatic inhibition assay, and their interaction with svPLA 2 enzyme was confirmed by molecular docking. The results demonstrated the developed platform was feasible for the rapid and accurate screening of svPLA 2 inhibitors from medicinal plant extracts. This study provides a strategy for the future development of next-generation antivenom treatments.

(1) Background: The identification of microorganisms includes traditional biochemical methods, molecular biology methods evaluating the conserved regions of rRNA, and the molecular biology of proteins (proteomics), such as MALDI-TOF MS mass spectrometry. This work aimed to identify the biodiversity of yeasts associated with stingless bee species’ honey and pollen,Melipona scutellaris,Nannotrigona testaceicornes, andTetragonisca angustula, from the region of São Gonçalo dos Campos-Bahia (BA) state, Brazil. (2) Methods: Cellular proteins were extracted from 2837 microbial isolates (pollen and honey) and identified via MALDI-TOF MS. The identified yeast species were also compared to the mass spectra of taxonomically well-characterized reference strains, available from the National Center of Biotechnology Information (NCBI) database. (3) Results: Nine yeast species were identified:Candida maltosa,Candida norvegica,Kazachstania telluris,Schizosaccharomyces pombe,Scheffersomyces insectosus,Meyerozyma guilliermondii,Brettanomyces bruxellensis,Kazachstania exigua, andStarmerella lactis-condensi. Nannotrigona testaceicornespollen had the highest number of yeast colonies. The yeastsBrettanomyces bruxellensisandKazachstania tellurisshowed high populations in the samples ofNannotrigona testaceicornesandMelipona scutellaris, respectively. This work shows that there is some sharing of the same species of yeast between honey and pollen from the same beehive. (4) Conclusions: A total of 71.84% of the identified species present a high level of confidence at the species level. Eight yeast species (Candida maltosa,Candida norvegica,Kazachstania telluris,Schizosaccharomyces pombe,Scheffersomyces insectosus,Meyerozyma guilliermondii,Kazachstania exigua, andStarmerella lactis-condensi) were found for the first time in the samples that the authors inspected. This contributes to the construction of new knowledge about the diversity of yeasts associated with stingless bee products, as well as to the possibility of the biotechnological application of some yeast species.

Background The metabolism study of active components for traditional Chinese medicine (TCM) in target organs is conducive to clarify the authentic active ingredients. Notoginsenoside R1 (NG-R1), one of the bioactive components of Panax notoginsenoside (PNS), is commonly acknowledged as the characteristic marker of PNS. However, the metabolism of NG-R1 in target organs has not been clarified yet due to the lack of robust technique and approach. Purpose The present study aimed to develop a reliable and efficient strategy and technology for revealing the qualitative and quantitative metabolism of active components of TCMs in target organs, and to clarify the biotransformation of NG-R1 in liver-brain-intestinal axis. Methods The metabolic transformation of NG-R1 in the brain gut axis was investigated in the in vitro incubation system of fresh rat brain, liver homogenate , and intestinal flora . To quickly lock the target metabolites, we set the mass defect filter (MDF) in different ranges to screen metabolites with different molecular weight (MW). This strategy was defined as multi-stage MDF (mMDF). In addition, we performed relative quantitative analysis on all metabolites according to the peak area acquired by LC-IT-TOF/MS to overcome the challenge that metabolites are difficult to be quantified due to the lack of standards. Results When MDF was set at 0.50 to 0.65 to screen metabolites with MW of 900 to 1200 Da, 6 novel metabolites were quickly found, and then identified as glucuronic acid binding, oxidation , dehydrogenation , methylation and hydrogenation products according to their LC and MS characteristics. When setting MDF at 0.42 - 0.52, 6 metabolites with MW of 600 to 900 Da were effectively screened and identified as Rg1, NG-R2, Rh1, Rg1+CH 2 +2H and Rg1+CH 2 . To screen the metabolites with MW of 300 to 600 Da, MDF was set at 0.25 - 0.42, and 4 novel metabolites were screened rapidly. The results of quantitative metabolism suggested that intestinal flora was the main metabolic site of NG-R1 in rat, and more than 60% of NG-R1 was converted to Rg1 by deglycosylation in the intestinal flora. Conclusion The mMDF strategy can significantly improve the research efficiency of qualitative metabolism of saponins . Although NG-R1 could be transformed into a variety of metabolites in rat liver and brain homogenate , it still existed mainly in prototype form. In the rat flora, NG-R1 mainly existed in the form of deglycosylated metabolite Rg1.

γ-Glutamyl dipeptides (γ-GGDPs) have been implicated be generated by enzymatic action on γ‑glutamyl groups, modulate kokumi sensation of fermented foods via activating calcium-sensing receptors (CaSR). However, their variation and potential formation mechanism in Jinhua ham remain unclear. 10 γ-GGDPs (γ-EA, γ-ED, γ-EE, γ-EL, γ-EM, γ-EG, γ-EF, γ-EY, γ-EI, γ-EW) in Jinhua ham were quantified by UHPLC-Q-TOF-MS and accumulating significantly during processing ( P < 0.05), with final contents ranging from 0.78 ± 0.01 μg/g (γ-EW) to 24.75 ± 0.33 μg/g (γ-EA). Sensory evaluation confirmed these γ-GGDPs enhanced the kokumi, saltiness, and umami of Jinhua ham during the post-ripening periods. Mechanistic investigation indicated that γ-glutamyl transpeptidase (GGT) transfer and hydrolytic activities increased progressively to 9.11 ± 0.17 U/mg and 12.19 ± 0.40 U/mg, respectively, which corroborated GGT's role in driving γ-GGDPs formation by catalyzing the transfer of γ-glutamyl groups to free amino acids (FAAs) generated by endogenous enzymes in Jinhua ham. Further validation via a ternary reaction system (comprising FAAs, γ-glutamyl donors, and GGT extracted from Jinhua ham) confirmed this GGT-catalyzed transfer reaction for γ-GGDPs biosynthesis. This study provides a theoretical foundation for the potential formation mechanisms of γ-GGDPs in Jinhua ham and regulation of Kokumi sensation in fermented foods.

Hepatocellular carcinoma (HCC) is one of the most prevalent malignant tumors worldwide. Both experimental and clinical studies confirm the anti-HCC effects of Yiguanjian ( YGJ ), though its material basis and pharmacological mechanisms remain unclear. This study integrated UHPLC-Q-TOF-MS with network pharmacology, molecular docking, and MDS to explore YGJ ’s potential anti-HCC mechanisms and active ingredients. In vitro analysis identified 96 chemical constituents in the YGJ extract, while network pharmacology revealed 57 potential targets and 51 bioactive compounds related to HCC treatment. The top nine key targets were AKT1, CTNNB1, EGFR, IL6, STAT3, BCL2, CASP3, MMP9, and TNF-α; the top seven active compounds included kaempferol, quercetin, luteolin, senkyunolide O, jasmolone, azaron, and dihydroartemisinin. These components may regulate HCC cell processes like gene expression, signal transduction, proliferation, apoptosis, and angiogenesis through pathways such as PI3K-Akt, TNF signaling pathway, and HIF-1 signaling pathway. Molecular docking and MDS showed that kaempferol, quercetin, luteolin, and dihydroartemisinin bind favorably to TNF-α, MMP9, BCL2, and IL6 proteins. Therefore, these proteins may serve as YGJ ’s therapeutic targets, while the compounds contribute to its pharmacological effects. Graphical This study utilized UHPLC-Q-TOF-MS integrated with network pharmacology, molecular docking, and molecular dynamics simulations (MDS) to explore the potential mechanisms and active ingredients of Yiguanjian ( YGJ ) for liver cancer treatment, thereby providing reference for identifying active substances and elucidating pharmacological mechanisms underlying YGJ ’s therapeutic effects against liver cancer.
