Inflammatory arthritis primarily involves rheumatoid arthritis (RA) and inflammatory osteoarthritis (OA), both characterized by shared inflammation in the articular joints and cartilage degeneration. These conditions lead to joint pain, loss of function, and a decline in quality of life. Triamcinolone acetonide (TA), an intermediate-acting synthetic glucocorticoid, is widely used as an anti-inflammatory agent for the treatment of inflammatory arthritis. However, its clinical application faces challenges such as rapid clearance from the joint cavity following intra-articular (IA) administration, the risk of joint damage with repeated injections, and the invasive nature of the IA injection route. To address these drawbacks, we propose sucrose acetate isobutyrate (SAIB), a biocompatible matrix, as a promising strategy for sustained drug release via intramuscular (IM) injection. Building on this approach, we designed a gallic acid-modified triamcinolone acetonide derivative (TA-GA) and incorporated it into the SAIB matrix to create the TA-GA-SAIB depot. The modification of TA with gallic acid (GA) effectively mitigates the burst release commonly observed in SAIB formulations, attributed to hydrogen bonding interactions between TA-GA and the SAIB matrix. Benzyl alcohol (BA) was selected as the optimal solvent to achieve minimal burst release during the in vitro drug release study. The TA-GA-SAIB depot exhibited sustained in vivo drug release, enhanced anti-inflammatory effects, and significant improvement in cartilage damage, along with excellent safety and biocompatibility. The synergy between the SAIB matrix and TA-GA not only enables sustained drug release but also improves therapeutic efficacy, making it a promising system for the treatment of inflammatory arthritis via intramuscular administration.

Safe and effective Cu 2+ supplementation in local lesion is crucial for minimizing toxicity of DSF-based chemotherapy. Targeted delivery of Cu 2+ appears more promising. Intraperitoneal chemotherapy for peritoneal carcinoma (PC) establishes “face-to-face” contact between targeted nanocarriers and tumor tissue. Herein, this study developed a biodegradable, injectable thermosensitive hydrogel that coencapsulating DSF submicroemulsion (DSF-SE) and folate-modified liposome loading glycyrrhizic acid-Cu (FCDL). FCDL acted as ‘beneficial horse’ to target the tumor-localized folate receptor, thus liberating Cu 2+ in tumor nidus. The prepared FCDL and DSF-SE were found with uniform sizes (160.2 nm, 175.4 nm), low surface charge (−25.77 mV, −16.40 mV) and high encapsulation efficiency (97.93 %, 90.08 %). In vitro drug release profile of FCDL, DSF-SE and FCDL&DSF-SE@G followed a sustained release pattern. And the release behavior of Cu 2+ from FCDL was pH-related, i.e., Cu 2+ was released faster under acidic condition. When FCDL and DSF-SE were loaded into an PLGA-PEG-PLGA-based hydrogel system, FCDL&DSF-SE@G was formed to ensure separated delivery of Cu 2+ and DSF in space but synchronized release over time. The rheology experiment showed a satisfactory gelling temperature of 32.7 °C. In vitro cytotoxicity study demonstrated that FCDL&DSF-SE@G significantly lowered the IC 50 of free Cu 2+ /DSF, Cu 2+ /DSF hydrogel and non-targeted analogue by almost 70 %, 65 % and 32 %, respectively. Accordingly, in tumor-bearing mice, FCDL&DSF-SE@G augmented the tumor inhibition rates for the same formulations by 352 %, 145 % and 44 %, respectively. The main mechanism was attributed to higher uptake of FCDL and DSF-SE, resulting in increased Cu(DDTC) 2 formation, ROS production and cell apoptosis. In conclusion, this targeted nanotherapy approach with dual-nanocarriers loaded hydrogel system, with its focus on face-to-face contact between nanocarriers and tumor tissues in the peritoneal cavity, holds significant promise for intraperitoneal chemotherapy in PC.

Fungicides are among the most essential agrochemicals for controlling crop diseases. However, the increased consumption of fungicides has resulted in environmental pollution and adversely affected human health. The slow-release technology can improve the efficacy and reduce the required amount of fungicide. Here, we developed a low-swelling redox-responsive alginate/lignin network bearing covalently conjugated fungicide thymol-Cl. Natural reductants in the environment caused the selective cleavage of disulfide bonds and release of thymol-Cl and thiazolidin-2-one (a by-product), which both could inhibit the phytopathogenic fungus ( F. oxysporum ). At the same time, metal chelating groups in the gel endow strong interactions with heavy metal ions for improving soil remediation and preventing the adsorption of toxic metals by the plant roots. The fungicide-loaded carrier could alleviate the poisonous effect of fungi on maize seeds. Indeed, the secretion of natural reducing agents by fungi and maize led to the cleavage of disulfide bonds and the release of fungicides.

In this study, a novel slow-release material using recycled waste foamed polystyrene (WFPS) as the carrier was developed for the degradation of aniline-contaminated groundwater. Sodium persulfate (SPS) and zero-valent iron (ZVI) were embedded in WFPS, enabling the controlled and sustained release of reactive species. Systematic investigations were conducted to optimize the material’s composition and evaluate its performance under various conditions, including pH, initial aniline concentration, and the presence of common groundwater anions. The results revealed that the slow-release material effectively enhanced aniline degradation, achieving a maximum removal rate of 93.45% under flowing conditions. The degradation pathway was analyzed using GC-MS, identifying intermediates such as benzoquinone, hydroquinone, and dodecane, with eventual mineralization into CO2and H2O. The material demonstrated robust performance, offering an efficient, cost-effective, and environmentally sustainable approach for in situ groundwater remediation.

Chitosan-based pluronic thermo-sensitive hydrogels represent a class of biomaterials characterized by their favorable biocompatibility and exceptional thermo-sensitive properties. The purpose of this study was to investigate their potential as carriers for embedding α-Mangostin (α-M), a bioactive compound derived from mangosteen peel, with the aim of inhibiting methicillin-resistant Staphylococcus aureus (MRSA). During the synthesis process, chitosan (CS), tannic acid (TA), pluronic F-127 (F127), and pluronic F-68 (F68) were combined to form the thermo-sensitive hydrogel, into which α-M was physically incorporated through crosslinking (designated as CTFF@α-M). We are very interested in the results obtained during the research process. The results of fluorescence imaging in small animals showed that CTFF extended the residence time of α-M in the body from 3 to 9 days, improved the bioavailability of α-M, and achieved the goal of sustained drug release. In vitro antibacterial experiments showed that CTFF@α-M (30 mg/mL) had an inhibition rate of 79.69% against MRSA, which was higher than CTFF@amoxicillin (AM) (5 mg/mL) increased by 71.68%. This study is expected to provide new insights into the integration of biomaterials and natural therapy, and emphasize the therapeutic potential of thermosensitive hydrogels and α-M in antibacterial treatment, especially in the fight against MRSA infection. Graphic In this study, a CTPP hydrogel was prepared using a physical cross-linking method, and α-M was encapsulated to create the CTPP@α-M drug-loaded hydrogel. Various characterization techniques, including SEM, FT-IR, and XRD, were employed to confirm its physicochemical properties. The hydrogel exhibited excellent water solubility, swelling capacity, sustained release characteristics, and biocompatibility, with the ability to transform into a semi-solid gel at 37 °C. In vitro experiments demonstrated that the CTPP@α-M hydrogel showed a powerful inhibitory effect on MRSA, exhibiting strong resistance to antibiotic resistance. Infection experiments using a mouse model confirmed that CTPP@α-M not only accelerated wound healing but also significantly reduced bacterial counts at the wound site. Moreover, the CTPP@α-M group significantly decreased the expression of pro-inflammatory cytokines while increasing the levels of anti-inflammatory factors, indicating its promising anti-inflammatory properties. Collectively, these data suggest that CTPP@α-M is a highly effective antibacterial thermo-sensitive hydrogel with excellent performance.

Water is essential for crop growth, particularly during the seed germination stage. This study proposes a novel strategy utilizing a natural bio-based hydrogel to enhance soil water retention and improve the hydration capacity for seeds. A hydrogel (CMC-Na/HACC/FeCl 3 ), synthesized from sodium carboxymethyl cellulose (CMC Na) and chitosan quaternary ammonium salt (HACC) via ferric chloride (FeCl 3 ), has the advantages of high stability and low cytotoxicity. Upon water absorption, the swelling degree of CMC-Na/HACC/FeCl 3 exceeds 20 g/g, providing sufficient moisture for plant growth. The experimental results showed that CMC-Na/HACC/FeCl 3 could significantly increase the seed germination rate compared with the traditional soil, and the germination rate could be up to 75 %, and the germination time, the length of germ and the length of the embryonic axis were significantly improved. In addition, in this study, the plant extracted insecticide matrine (MT) was added into the hydrogel system to form CMC-Na/HACC/FeCl 3 /MT. The results of the study showed that MT could be gradually released from the hydrogel, and the release time could last for 21 days. The hydrogel provided moisture to the seeds while creating a barrier against pest invasion and showed significant antibacterial effects. Both CMC-Na/HACC/FeCl 3 and CMC-Na/HACC/FeCl 3 /MT are anticipated to be valuable in agricultural production due to their excellent properties, which contribute to improving soil composition and minimizing resource waste and environmental pollution.

In orthopedic treatment, long-term nonunion of bone defects has always been a significant issue. As bone graft materials, tetra-armed polyethylene glycol (Tetra-PEG) hydrogel scaffolds are anticipated to address this issue due to their extracellular matrix (ECM)-like structure. However, poor osteoinductivity and cell adhesion after implantation restricted its clinical application in bone tissue engineering. Herein, graphene oxide (GO) loaded with icariin (ICA) was loaded into the Tetra-PEG hydrogel semi-interpenetrating network composite gel by in situ composite approach, and silk fibroin (SF) was incorporated to create a new organic–inorganic composite hydrogel scaffold (PNSG-ICA) to investigate the controlled drug release characteristics and osteogenic properties. The experiments of this study demonstrate that PNSG-ICA scaffolds can effectively encapsulate and release ICA over a period of 12 days. The scaffolds exhibited interconnected porous microarchitectures, satisfactory mechanical properties, appropriate degradation characteristics, and excellent biocompatibility. In vitro analyses indicated that ICA-doped hydrogels significantly enhanced the osteogenic differentiation of rat bone mesenchymal stem cells (rBMSCs), as demonstrated by alkaline phosphatase and alizarin red staining, as well as qRT-PCR assessments. Moreover, in vivo investigations revealed that the ICA sustained-release system significantly enhanced the regeneration of bone defects mediated by rBMSCs in situ, as evidenced by mouse calvarial bone defect models. In summary, we envision that the prolonged release of ICA from PNSG-ICA scaffolds may become a new strategy for the clinical treatment of bone defects.

Compromised cellular function and bacterial infection are major obstacles to effective wound healing. Simultaneous bacterial elimination and cellular function restoration represent a promising strategy for treating infected wounds. Here, a multifunctional hydrogel (QMPZN) composed of methacrylate anhydride (MA)-grafted quaternary ammonium chitosan (QCS-MA), polyvinyl alcohol (PVA), zeolitic imidazolate framework-8 (ZIF-8), and nicotinamide mononucleotide (NMN, a precursor of NAD + involved in cellular metabolism and repair) was developed to achieve combined antibacterial and regenerative effects. The hydrogel rapidly forms in situ under UV light via methacrylate double bond cross-linking, enabling application to irregular wound surfaces. PVA enhances its mechanical strength and tissue adhesion through hydrogen bonding. Benefiting from quaternary ammonium groups and the sustained Zn 2+ release from ZIF-8, QMPZN exhibits strong and durable antibacterial activity. Meanwhile, the controlled release of NMN helps restore cellular function and promote tissue regeneration. In vivo studies showed that QMPZN effectively inhibited bacterial infection and accelerated epithelial regeneration and collagen deposition in infected wounds. These results suggest that QMPZN hydrogel is expected to be a therapeutic platform for the treatment of infected wounds.

Microneedle technology has emerged as a potential new means of painless drug delivery for diabetic patients. Maintaining blood glucose levels (BGLs) within the normal range for a long time is important for people with diabetes. However, it is a new challenge to realize long-term therapy due to insufficient drug loading. In this study, a polymeric porous microneedle array with continuous micropores was prepared by combining the microtemplate method and pore-causing agent method. By changing the ratio of a pore-causing agent, the fluid uptake capacity and mechanical strength were adjusted to balance the trade-off between rapid imbibition and the mechanical properties of puncture. A special drug loading chamber was developed to realize a large package (135.69 mg) of metformin-loaded porous microneedle system. Experiments in diabetic rat models in vivo showed that the prepared porous microneedles can effectively puncture the skin and maintain stable BGLs up to 7 h × 5 cycles over five experimental cycles (108 h) without the risk of hypoglycemia. These results suggested that the high-dose polymeric porous microneedle-mediated metformin delivery system had the advantages of rapid response and durable release, and was a potential candidate for sustained-release transdermal drug delivery in patients with diabetes mellitus and even chronic conditions.

This study aimed to prepare novel chitosan (CS)-based aerogels for efficient loading of hydrophilic chlorogenic acid (CGA) and to facilitate its sustained release in vivo. The mechanical properties and water permeability of the CS-based aerogels were enhanced by incorporating sodium alginate. Melonal sodium alginate CS (MSCS) was synthesised by crosslinking melonal with CS aerogel via a Schiff base reaction. MSCS possesses a three-dimensional porous structure, featuring abundant micropores and mesoporous distributions. Due to the addition of melonal, MSCS demonstrates environmental stability and moisture resistance, with a surface contact angle of 121°. Adsorption capacity tests revealed that MSCS exhibits high adsorption capacity for CGA, reaching 769.4 mg/g, with an adsorption efficiency of 76.94 %. Molecular dynamics simulation results indicate that the adsorption process is exothermic, with increased adsorption capacity at lower temperatures, and is a combination of physical and chemical adsorption. Through quantum chemistry calculations, we identified the adsorption sites of CGA on MSCS and the tendency of MSCS to donate electrons. The in vitro sustained-release experiments demonstrated continuous release of CGA for 48 h in both artificial gastric juice and intestinal fluid, with release rates of 34.26 % and 34.96 %, respectively.

Myocardial infarction (MI) induces cardiomyocyte necrosis, inflammation, fibrosis, and ventricular remodeling, leading to heart failure. To address this, we developed an intelligent cardiac patch, SMM@Gel, composed of a reactive oxygen species (ROS)-responsive PVA-TSPBA hydrogel matrix reinforced via solvent exchange and salting-out technology, loaded with mannose-functionalized, danshensu sodium-loaded hollow mesoporous polydopamine nanoparticles (Sa@mPDA-Man). This design makes sustained drug release and ROS scavenging come true. In vitro, studies demonstrated SMM@Gel promoted endothelial tube formation (24 ± 3 nodes vs. 6 ± 2 in controls) and M2 macrophage polarization (CD206+ cells) while reducing inflammation (iNOS downregulation). In vivo, experiments in MI rats revealed SMM@Gel preserved left ventricular ejection fraction (LVEF: 52.3 % ± 4.1 % vs. 38.5 % ± 3.2 % in the saline group), normalized ventricular dimensions (EDV/ESV) and enhanced wall thickness. Histological analysis showed reduced infarct size (18.7 % ± 2.5 % vs. 32.1 % ± 3.8 %), decreased inflammation, and improved neovascularization. RNA-seq identified pathways linked to angiogenesis, inflammation resolution, and extracellular matrix remodeling. These findings highlight SMM@Gel's potential as a dual-action therapy for MI, combining ROS scavenging, anti-inflammatory effects, and angiogenic promotion to mitigate post-MI remodeling and preserve cardiac function.

As gastric ulcers remain a significant clinical challenge, this study presents the development of a flexible microneedle system based on cerium/zinc-doped bioactive glass (CeZnBG) and a PCL/PLA matrix for targeted ulcer therapy. The design leverages the synergistic antioxidant and antibacterial properties of CeZnBG, combined with the localized drug delivery capability of microneedles, to enhance ulcer healing. CeZnBG was synthesized via high-temperature sintering at various temperatures (950 °C, 1050 °C, 1150 °C, and 1250 °C), and subsequently incorporated into PCL/PLA microneedles (PCMN) at doping concentrations of 0.5 %, 1.5 %, and 2.5 % to form composite materials. The structural and physicochemical characteristics of the CeZnBG-PCMN, were investigated using X-ray diffraction (XRD), scanning electron microscopy (SEM), Energy Dispersive X-ray Spectroscopy (EDS), mechanical properties, differential scanning calorimetry (DSC), and X-ray photoelectron spectroscopy (XPS). In vitro assays, including ROS scavenging and cytocompatibility, were used to assess the bioactivity of the composites. The results demonstrated that PCMN containing 1.5 % CeZnBG maintained excellent cytocompatibility (with relative cell viability exceeding 80 %) while exhibiting notable antibacterial performance, indicating promising potential for targeted gastric ulcer repair.
