Mussels are nutrient-rich but perishable, resulting in substantial resource loss. A protease-producing strain (Bacillus velezensisZ-1,Mytilus edulis) isolated from marine sludge was used to hydrolyze mussels, producing Y-1, a hydrolysate with antioxidant activity. In this study, ultrafiltration, gel chromatography, and LC-MS/MS were employed to isolate and identify bioactive peptides from the hydrolysate. The results revealed that the hydrolysate exhibited antioxidant activity, pancreatic cholesterol esterase inhibitory activity, pancreatic lipase inhibitory activity, and α-glucosidase inhibitory activity. Molecular docking using AutoDock Tools 1.5.6 was performed to analyze the interactions of peptides with CD38 and Keap1, leading to the identification of five potentially bioactive peptides: VPPFY, IMLFP, LPFLF, FLPF, and FPRIM. These peptides formed hydrogen bonds and hydrophobic interactions with CD38 and Keap1, demonstrating strong DPPH radical scavenging and superoxide anion radical scavenging capacities. This study highlights the multifunctional bioactive potential of these peptides, offering insights into their therapeutic applications. The findings provide a novel approach for the effective utilization of mussel resources and highlight their potential application value in the development of functional foods.

Background The structural diversity of extracellular polymeric substances produced by microorganisms is attracting particular attention. Poly-gamma-glutamic acid (γ-PGA) is a widely studied extracellular polymeric substance from Bacillus species. The function of γ-PGA varies with its molecular weight (Mw). Results Herein, different endogenous promoters in Bacillus licheniformis were selected to regulate the expression levels of pgdS , resulting in the formation of γ-PGA with Mw values ranging from 1.61 × 10 3 to 2.03 × 10 4 kDa. The yields of γ-PGA and exopolysaccharides (EPS) both increased in the pgdS engineered strain with the lowest Mw and viscosity, in which the EPS content was almost tenfold higher than that of the wild-type strain. Subsequently, the compositions of EPS from the pgdS engineered strain also changed. Metabolomics and RT-qPCR further revealed that improving the transportation efficiency of EPS and the regulation of carbon flow of monosaccharide synthesis could affect the EPS yield. Conclusions Here, we present a novel insight that increased pgdS expression led to the degradation of γ-PGA Mw and changes in EPS composition, thereby stimulating EPS and γ-PGA production. The results indicated a close relationship between γ-PGA and EPS in B. licheniformis and provided an effective strategy for the controlled synthesis of extracellular polymeric substances.

AimsThis research aims to screen and identify a microbial strain capable of efficiently degrading waterborne polyurethane used in the biomedical field. Furthermore, the research seeks to enhance the strain’s degradation capability through ultraviolet mutagenesis and elucidate its enzymatic degradation mechanism. Methods and resultsWe successfully isolated the target strain from the soil sample and identified it as Bacillus sp. J-11. Under optimized culture conditions (substrate concentration of 0.4%, temperature of 30°C, and pH of 6.0), strain J-11 exhibited remarkable degradation capability towards biomedical-grade polyurethane emulsion (Baymedix® CD104), achieving a degradation efficiency of 78.2% within 4 days. Gel permeation chromatography (GPC) analysis demonstrated a progressive decrease in the molecular weight of the polyurethane film over time. Thermogravimetric analysis (TGA) further revealed a reduction in thermal stability, while extended depth field microscopy (EDFM) uncovered significant microscopic structural changes, including holes and collapse on the film surface. Additional degradation experiments conducted on commercial PU foam under co-carbon conditions showed that strain J-11 achieved a degradation rate of 37.6% within 30 days. Enzyme activity assays indicated that the hydrolytic and oxidative enzymes secreted by strain J-11 played a critical role in the degradation process.ConclusionsStrain J-11 exhibits highly efficient biodegradation potential through the secretion of hydrolases and oxidases, which break down the molecular chains of polyurethane. This study has elucidated its degradation pathway and provided a theoretical foundation for future mechanistic investigations and the management of biomedical material waste.

Probiotics, prebiotics, and synbiotics can alleviate metabolic syndrome by altering the composition of the gut microbiota. Live combined Enterococcus faecium and Bacillus subtilis has been indicated to promote growth and reduce inflammation in animal models. However, the modulatory effects of live combined B. subtilis R-179 and E. faecium R-026 (LCBE) on human microbiota remain unclear. The current study examined the growth of these two strains in the presence of various oligosaccharides and assessed the effects of this probiotic mixture on human and murine gut microbiota in vitro and in vivo. Oligosaccharides improved the growth of E. faecium R-026 and B. subtilis R-179 as well as increased their production of short-chain fatty acids. E. faecium R-026 or B. subtilis R-179 co-incubated with Bifidobacterium and Clostridium significantly increased the number of the anaerobic bacteria Bifidobacterium longum and Clostridium butyricum by in vitro fermentation. Moreover, LCBE significantly reduced plasma cholesterol levels in mouse models of hyperlipidemia. LCBE combined with galacto-oligosaccharides led to a significant decrease in the Firmicutes/Bacteroidetes ratio and a significant increase in the relative abundance of Akkermansia and Bifidobacteria after treating mice with LCBE (0.23 g/day) for eight weeks. Furthermore, in vitro fermentation also showed that both the single strains and the two-strain mixture modulated human gut microbiota, resulting in increased Lactobacillus and Bifidobacteria, and decreased Escherichia-Shigella. Overall, these results suggest that LCBE can improve host health by reducing the level of cholesterol in mouse models by modifying the composition of the gut microbiota.

A novel electrochemical biosensor was developed for detecting short DNA oligonucleotide of Bacillus thuringiensis (Bt) transgenic sequence based on Ag nanoparticle aggregates. To fabricate this DNA biosensor, the thiol-modified capture DNA (cDNA) was first anchored on gold (Au) electrode, and then the target DNA (tDNA) was hybridized with the immobilized cDNA. Subsequently, the probe DNA (pDNA) functionalized by biotinylated Ag nanoparticle was associated with the fixed tDNA, and the single Ag nanoparticle label was obtained (cited as SAg label). Finally, dissociative biotinylated Ag nanoparticle was bound to the resultant biotinylated SAg label assembled on Au electrode by virtue of bridge molecule streptavidin (SA) through biotin–SA specific interaction, which could lead to in situ aggregate of Ag nanoparticles on Au electrode and induce a novel tag including multiple Ag nanoparticles (cited as MAg tag). The novel tag exhibited excellent electroactive property in the solid-state Ag/AgCl process and was successfully applied to Bt transgenic sequence assay. A detection limit of 10 fM was achieved, which was improved by three orders of magnitude as compared to the SAg label. Furthermore, this novel DNA biosensor demonstrated a good selectivity towards tDNA.

Post-harvest diseases of raspberries affect their storage and subsequently the ability to provide consumers with fresh products. The antifungal activity of microbial volatile organic compounds (MVOCs) against both pre and postharvest diseases has previously been demonstrated, but the control of raspberry postharvest diseases caused by Botrytis cinerea and Rhizopus stolonifer using MVOCs has not been extensively studied. We have thus investigated the effects of Bacillus siamensis G-3 volatile organic compounds (VOCs) on controlling the diseases caused by B. cinerea and R. stolonifer . Bioassays of the VOCs of B. siamensis G-3 exhibited significant control effects (89.4%) on the two pathogens. The VOCs composition was studied with solid phase micro extraction and GC/mass spectrometry (SPME-GC-MS), and 22 VOCs, different from the controls, were identified. Among these compounds, only 2, 6-di- tert -butyl-4-methylphenol (BHT), and 2,4-di- tert -butylphenol (2,4-DTBP) had significant control effects on the pathogens (* P < 0.05), 52.38 and 93.33%, respectively, compared with the controls in vivo . With the 2,4-DTBP treatments, the raspberries could be stored for 20 d at 0 °C, with the disease rate maintained at ∼10%. In addition, we have reported for the first time, that 2,4-DTBP may be a better preservative than BHT and could be used as a bio-fumigant for controlling raspberry postharvest diseases caused by B. cinerea and R. stolonifer in vivo .

Raspberry ketone is a primary aroma component of the red raspberry. The glycosylation of this compound is a potential approach used to improve its pharmaceutical properties. In this work, raspberry ketone glycosides are produced in bacteria for the first time. Bacillus licheniformis PI15, an organic solvent-tolerant glycosyltransferase-producing strain, was isolated from chemically polluted soil. The cloning and heterologous expression of a glycosyltransferase, which was designated PI-GT1, in Escherichia coli BL21 resulted in the expression of an active and soluble protein that accounted for 15% of the total cell protein content. Purified PI-GT1 was highly active and stable over a broad pH range (6.0–10.0) and showed excellent pH stability. PI-GT1 maintained almost 60% of its maximal activity after 3 H of incubation at 20–40 °C and demonstrated optimal activity at 30 °C. Additionally, PI-GT1 displayed high stability and activity in the presence of hydrophilic solvents with log P ≤ −0.2 and retained more than 80% of its activity after 3 H of treatment. Supplementation with 10% DMSO markedly improved the glycosylation of raspberry ketone, resulting in a value 26 times higher than that in aqueous solution. The organic solvent-tolerant PI-GT1 may have potential uses in industrial chemical and pharmaceutical synthesis applications.

Along with banning antibiotics, necrotic enteritis (NE), especially subclinical enteritis (SNE), poses a significant threat to the chicken industry; however, probiotics are a potentially promising intervention. We aimed to investigate the beneficial effects of Bacillus subtilis DSM29784 (BS) on the treatment of Clostridium perfringens (CP)-induced SNE in broilers. A total of 360 1-day-old broiler chicks were divided into three treatment groups, namely control (Ctr), SNE, and BS treatment (BST) groups, all of which were fed with a basal died for 21days, and then from day 22 onward, only the BST group had a BS supplemented diet (1×10 9 colony-forming units BS/kg). On day 15, all chicks, except the Ctr group, were challenged with a 20-fold dose coccidiosis vaccine and 1ml CP (2×10 8 ) on days 18–21 for SNE induction. Beneficial effects were observed on growth performance in BST compared to SNE broilers. BST treatment alleviated intestinal lesions and increased the villus height/crypt depth ratio. Further, BST broilers showed increased maltase activity in the duodenum compared with SNE chicks, and a significantly decreased caspase-3 protein expression in the jejunum mucosa. Moreover, an increased abundance of Ruminococcaceae and Bifidobacterium beneficial gut bacteria and an altered gut metabolome were observed. Taken together, we demonstrate that the manipulation of microbial gut composition using probiotics may be a promising prevention strategy for SNE by improving the composition and metabolism of the intestinal microbiota, intestinal structure, and reducing inflammation and apoptosis. Hence, BS potentially has active ingredients that may be used as antibiotic substitutes and effectively reduces the economic losses caused by SNE. The findings of this study provide a scientific foundation for BS application in broiler feed in the future.

To mitigate plastic pollution and reduce reliance on fossil fuels, the development of high-performance polymeric coatings from renewable resources to mitigate plastic pollution and reduce reliance on fossil fuels remains a critical challenge. Among them, substituting petroleum-based isocyanates with bio-based 1,5-pentanediisocyanate (PDI) not only significantly reduces the carbon footprint but also provides a key green feedstock for developing high-performance biodegradable polyurethanes. In this study, bio-based poly (lactic acid) polyol (PLA) and PDI were utilized as the raw materials. By incorporating polytetramethylene glycol (PTMEG) into the soft segment of waterborne polyurethane (WPU), a series of PLA/PDI bio-based waterborne polyurethanes (PP-WPU-Ts) were synthesized. The results demonstrate that PP-WPU-3 (n PLA : n PTMEG = 5:5) achieved a high bio-based content of 53.1 wt% along with a balanced combination of mechanical properties, including a tensile strength of 4.02 MPa, an elongation at break of 1118.6%, and a toughness of 26.7 MJ/m 3 . Additionally, PP-WPU-3 exhibited outstanding wear and yellowing resistance, reaching 80,000 abrasion cycles and a grade 1 yellowing resistance (ΔE = 2.8). Most importantly, the material exhibited controllable degradation across various environments: rapid disintegration in alkaline media (65% weight loss in 6 h), efficient biodegradation in soil (54.3% in 120 days), and synergistic mineralization by Bacillus sp. (19.3% in 15 days), offering a promising end-of-life solution. The optimized PP-WPU-3 possesses balanced mechanical properties while also exhibiting excellent yellowing resistance and degradation capabilities. These characteristics validate its potential as an eco-friendly alternative for organic coatings.

Fusarium wilt, caused by the soil-borne fungusFusarium oxysporumf. sp.lycopersici(Fol), is a devastating disease that severely constrains tomato production worldwide. In this study, an endophytic bacterium, designated ZN-S18, was isolated from healthy tomato stems and identified asBacillus amyloliquefaciensbased on phylogenetic analyses of the 16S rRNA andrpoBgenes. Dual culture assays demonstrated that ZN-S18 exhibited strong antagonistic activity against Fol and significantly inhibited fungal mycelial growth. The cell-free culture filtrate of ZN-S18 markedly suppressed Fol spore germination and mycelial growth, and the inhibitory effect on spore germination was irreversible. LC-MS analyses confirmed that ZN-S18 produces the cyclic lipopeptides surfactin and iturin A. In planta assays further demonstrated that pretreatment with ZN-S18, its culture filtrate, or the combination of surfactin and iturin A significantly reduced Fusarium wilt in tomato seedlings. Moreover, the culture filtrate disrupted fungal cell wall integrity, as indicated by the increased sensitivity of Fol to Congo red stress. Notably, the combined application of surfactin and iturin A exhibited significantly stronger antifungal activity than either compound alone under Congo red-containing conditions, suggesting a synergistic effect on fungal cell wall integrity. Collectively, these findings indicate that the endophytic strainB. amyloliquefaciensZN-S18 is a promising biocontrol agent against tomato Fusarium wilt, primarily through the synergistic action of surfactin and iturin A. This study provides an environmentally friendly alternative to chemical fungicides and establishes a foundation for future investigations into the molecular mechanisms underlying lipopeptide-mediated antagonism.

The widespread presence of the environmental contaminant acrylamide (AA) in thermally processed foods has been linked to serious environmental and health challenges. Although the formation of AA can be fundamentally inhibited by L-asparaginase (L-ASNase), its removal efficiency is highly dependent on substrate affinity. However, the structural mechanism by which the affinity of this L-ASNase is regulated remains poorly understood. Herein, a rational design strategy of active site loop (ASL) engineering was initially employed to improve the substrate affinity of L-ASNase from Bacillus licheniformis . As a result, two single mutants (K18S and L24Y) both exhibited approximately 50% increased substrate affinities with unaffected catalytic activities. Moreover, the combined double mutant K18S/L24Y achieved a significantly lower K m value of 1.49±0.25 mM. Subsequently, structural analysis revealed that mutants K18S and L24Y obtained disordered active site loops (ASLs) compared with the β-hairpin structure in wild type by reducing the hydrogen bonds and changing the hydrophobic interactions, displaying the altered interaction forces between amino acids on the active site for substrate binding. Building on these structural observations, molecular dynamics simulations were further performed to uncover the underlying physical mechanism. It was revealed by the simulations that the mutations lead to reduced dynamic fluctuations and the achievement of an energetically neutral state in the ASL region, thereby converting substrate binding from a "high-resistance" mode to a "zero-resistance" mode and consequently enhancing affinity. Additionally, mutant K18S held the same α-helix orientation linked to ASL as the wild type for maintaining catalytic activity. To our knowledge, this study fills a gap in the structural mechanism of substrate affinity for L-ASNases and offers a guideline for engineering L-ASNases from different origins for environmental contaminant applications.

A previously uncharacterized GH18 Chitinase from Bacillus haynesii (BhGH18) was heterologously expressed
and characterized. BhGH18 displayed maximal activity at pH 7.5 and
40 ° C, and activity was enhanced by Mg 2+ /Zn 2+ but inhibited by Cu 2+ /Fe 2+ /Ni 2+ . After further purification, the enzyme exhibited markedly improved
kinetics toward pNP-(GlcNAc) 2 ( K m = 0.103 mM, V max = 0.217 μmol/min).
Importantly, on the natural substrate colloidal chitin, BhGH18 gave
an apparent K m of 3.51 mg/mL and V max of 1.69 μmol/min (for colloidal chitin).
The enzyme displayed an endohydrolytic preference, converting ultrasonically
pretreated chitin predominantly into chitobiose (DP2). Ultrasonication
at 500 W increased the hydrolysis yield nearly 6-fold (23.86% conversion).
Molecular docking identified Trp157, Glu197, Tyr198 and Arg333 as
key substrate-binding residues. This work demonstrates an integrated
ultrasound-enzyme strategy using BhGH18 for sustainable production
of DP2-enriched chitin oligosaccharides.
