A Red-Emission Fluorescent Probe with Large Stokes Shift for Detection of Viscosity in Living Cells and Tumor-Bearing Mice

Beilei Wang,Dezhi Yang,Xiaohong Zhong,Yuhui Liu,Yong Huang

Abnormal viscosity is closely related to the occurrence of many diseases, such as cancer. Therefore, real-time detection of changes in viscosity in living cells is of great importance. Fluorescent molecular rotors play a critical role in detecting changes in cellular viscosity. Developing red emission viscosity probes with large Stokes shifts and high sensitivity and specificity remains an urgent and important topic. Herein, a novel viscosity-sensitive fluorescent probe (TCF-VIS1) with a large stokes shift and red emission was prepared based on the 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) skeleton. Due to intramolecular rotation, the probe itself does not fluorescence at low viscosity. With the increase in viscosity, the rotation of TCF-VIS1 is limited, and its fluorescence is obviously enhanced. The probe has the advantages of simple preparation, large Stokes shift, good sensitivity and selectivity, and low cytotoxicity, which make it successfully used for viscosity detection in living cells. Moreover, TCF-VIS1 showed its potential for cancer diagnosis at the cell level and in tumor-bearing mice by detecting viscosity. Therefore, the probe is expected to enrich strategies for the detection of viscosity in biological systems and offer a potential tool for cancer diagnosis.

A smartphone-based portable electrochemical sensor enabled ultrasensitive detection of paclitaxel in serum and injection samples

Yang Ruo-Yu,Wang Jin-Hua,Yu Zi-Wei,Chen Yun-Ting,Wu Mei-Juan,Huang Pin-Fang,Liu Meng-Meng

A point-of-care testing platform, consisting of smartphone, miniature electrochemical workstation, and screen-printed carbon electrode (SPCE) modified by gold nanoparticle (AuNPs) and multi-walled carbon nanotubes (MWCNTs), is fabricated for the ultrasensitive detection of paclitaxel (PTX) in human serum and injection solution. To enhance conductivity of the sensing system, MWCNTs concentration and AuNPs electrodeposition time were optimized. The AuNPs/MWCNTs effectively increase the working electrode area of SPCE by a factor of 1.46, contributing to improved electrochemical performance. The steps of electrode surface modification and the characterization of AuNPs/MWCNTs/SPCE were investigated by differential pulse voltammetry, impedance spectroscopy, X-ray photoelectron spectroscopy, and scanning electron microscopy. The sensor shows good linearity between current response and PTX concentration in 0.2 M phosphate buffer at pH = 7.4 (0.05–10 μM, with a limit of detection (LOD) of 1.7 nM) and human serum (0.5–30 μM and a LOD of 3.6 nM). The recoveries range from 89.91 to 103.36% and 91.42 to 103.73% in human serum and injection solution, respectively, with satisfactory relative standard deviation. Moreover, the sensor has excellent stability during 8 weeks and exhibits outstanding specificity and reproducibility towards PTX detection, providing a possible option for PTX determination in practical application such as therapeutic drug monitoring and drug quality control. Asymmetric Cyclodextrin-Dimer-Involved Nanoassemblies by Selective Host–Guest Interactions: Concentration-Dependent Morphology Evolution and Light-Regulated Biomedical Applications

Qingjie Li,Jinhao Yan,Wenlong Jiang,Yunpeng Zhang,Peng Gao,Longxiang Tao,Jun Yin

Supramolecular assembly has attracted significant attention and has been applied to various applications. Herein, a β–γ-CD dimer was synthesized to complex different guest molecules, including single-strand polyethylene glycol (PEG)-modified C60 (PEG-C60), photothermal conversion reagent (IR780), and dexamethasone (Dexa), according to the complexation constant-dependent specific selectivity. Spherical or cylindrical nanoparticles, monolayer or bilayer vesicles, and bilayer fusion vesicles were discovered in succession if the concentration of PEG-C60 was varied. Moreover, if near-infrared light was employed to irradiate these nanoassemblies, the thermo-induced morphological evolution, subsequent cargo release, photothermal effect, and singlet oxygen (1O2) generation were successfully achieved. The in vitro cell experiments confirmed that these nanoparticles possessed excellent biocompatibility in a normal environment and achieved superior cytotoxicity by light regulation. Such proposed strategies for the construction of multilevel structures with different morphologies can open a new window to obtain various host–guest functional materials and achieve further use for disease treatment.

3D Printed Pedicle Screws with Microarc Oxidation Ceramic Interfaces Enhance Osteointegration and Orthopedic Fixation Feasibility

Zehao Chen,Binghao Wang,Chengliang Yang,Zhendong Lv,Yu Wei,Tianming Pan,Fuqing Xuan,Xingdie Zhou,Hao Chen,Hongxing Shen,Liqiang Wang,Yuhui Zhang

Effective osteointegration is of great importance for pedicle screws in spinal fusion surgeries. However, the lack of osteoinductive activity of current screws diminishes their feasibility for osteointegration and fixation, making screw loosening a common complication worldwide. In this study, Ti-6Al-4V pedicle screws with full through-hole design were fabricated via selective laser melting (SLM) 3D printing and then deposited with porous oxide coatings by microarc oxidation (MAO). The porous surface morphology of the oxide coating and the release of bioactive ions could effectively support cell adhesion, migration, vascularization, and osteogenesis in vitro. Furthermore, an in vivo goat model demonstrated the efficacy of modified screws in improving bone maturation and osseointegration, thus providing a promising method for feasible orthopedic internal fixation.

Antibiotic-free ocular sterilization while suppressing immune response to protect corneal transparency in infectious keratitis treatment

Yaou Peng,Shuaiyue Pang,Yanlin Zeng,Jiayi Wei,Jinda Lu,Yangfan Ruan,Xinyu Hong,Xiuhui He,Xiaoying Chu,Yishun Guo,Hanwen Guo,Siyuan Qian,Zipei Jiang,Zhengxuan Jiang,Bailiang Wang

Clinical guidelines for infectious keratitis treatment require that anti-inflammatory drugs can only be used after infection elimination, which causes irreversible inflammatory damage to the cornea. In this work, photodynamic metal organic frameworks (PCN-224) were used as drug carrier to load Pt NPs with catalase-like activity and anti-inflammatory drug (Dexamethasone, DXMS) for endogenous oxygen generation and reduced corneal damage, respectively. The photodynamic therapy (PDT) effect was greatly enhanced in bacteria elimination and bacterial biofilms removal through catalysis of overexpressed hydrogen peroxide (H 2 O 2 , ∼8.0 and 31.0 μM in bacterial solution and biofilms, respectively) into oxygen by Pt NPs. More importantly, the cationic liposome modified PCN-224@Pt@DXMS@Liposomes (PPDL NPs) greatly enhanced the adhesion to negatively charged ocular surface and penetration into corneal barrier and bacterial biofilms. Both in vitro cell viability test and in vivo eye irritation tests proved good biocompatibility of PPDL NPs under 660 nm laser irradiation. Furthermore, PDT of PPDL NPs in rapid bacteria killing was verified through infectious keratitis animal model. The superior bactericidal effect of antibacterial materials could largely replace the bactericidal effect of the immune system. It is worth mentioning that this simultaneous sterilization and anti-inflammation treatment mode is a new exploration against the clinical treatment guidelines.

Anti-fouling coating with ROS-Triggered On-Demand regulation of inflammation to favor tissue healing on vascular devices

Hui Yan,Lietao Wang,Haoshuang Wu,Yongqi An,Yumei Qin,Zhen Xiang,Huining Wan,Yanfei Tan,Li Yang,Fanjun Zhang,Qing Jiang,Rifang Luo,Yunbing Wang

Unhealthy vascular tissue healing after stenting is primarily triggered by endothelial dysfunction and excessive smooth muscle cell proliferation, which is induced and aggravated by local thrombosis and excessive inflammation. Excessive inflammatory response is closely related to elevated concentrations of reactive oxygen species (ROS). Here, an environment-friendly coating strategy termed “Anti-fouling plus” with inflammatory self-regulation ability was proposed. Upon this, membrane-mimicking copolymer MA(PCLA) served as the antifouling coating, effectively inhibiting coagulation and inflammation during the early stage of implantation. Furthermore, a ROS-responsive prodrug was drawn into the coating to promote tissue healing. A molecular prodrug of thioketal-bearing dexamethasone was engineered to be released in a controlled and responsive manner in response to externally high levels of ROS. The released dexamethasone exhibited highly effective anti-inflammation properties after implantation. The combination of ROS-responsive prodrug and membrane-mimicking antifouling interface strategy intelligently integrated inflammation responsiveness and regulation, which could mediate a gentle vascular microenvironment and thus promote vascular remodeling. In vivo stent implantation studies demonstrated that the proposed “Anti-fouling plus” coating effectively inhibited neointima hyperplasia, modulated the inflammatory response and promoted endothelialization, offering promising surface modification approaches for vascular stents.

3D-Printed In Situ Growth of Bilayer MOF Hydrogels for Accelerated Osteochondral Defect Repair

Kaiqi Qin,Xinyue Huang,Shengfeng Wang,Jiachen Liang,Zengjie Fan

Repairing osteochondral (OC) defect presents a significant challenge due to the intricate structural requirements and the unpredictable differentiation pathways of bone marrow mesenchymal stem cells (BMSCs). To address this challenge, a novel biomimetic OC hydrogel scaffold is developed that features a structure of soft and hard components. This scaffold incorporates bilayer metal–organic frameworks (MOFs), specifically ZIF-67 in the upper layer and ZIF-8 in the lower layer, achieved through an in situ printing process. This configuration enables the spatial and temporal modulation of BMSC differentiation by controlling the release of Co 2 ⁺ and Zn 2 ⁺. The results demonstrate that the bilayer MOF hydrogels significantly outperform hydrogels that either lack MOFs or contain a single type of MOF in enhancing repair outcomes in rabbit models of knee OC defects. The improved regenerative efficacy is attributed to the distinct chondrogenic and osteogenic differentiation cues provided by the bilayer MOFs, effectively guiding BMSCs toward enhanced tissue regeneration. This customizable biomimetic OC hydrogel scaffold not only opens new avenues for innovative therapeutic strategies but also holds great promise for widespread clinical applications.

Oppositely Charged Polyurethane Microspheres with Tunable Zeta Potentials as an Injectable Dual-Loaded System for Bone Repair

Yi Hou,Nan Jiang,Li Zhang,Yubao Li,Yuezhong Meng,Dongmei Han,Chen Chen,Yuan Yang,Songsong Zhu

To effectively repair irregular shaped bone defects by a minimally invasive procedure, the exploration of an injectable gel to fill the defect is desirable. Herein, positively and negatively charged polyurethane microspheres (PU-A and PU-B) with adjustable zeta potentials as well as the hydroxyapatite-loaded PU microsphere (PU-A/HA) and the dexamethasone-loaded PU microsphere (PU-B/Dex) were successfully prepared, and the oppositely charged microspheres could self-assemble into injectable gels with 3D structures by a mutually electrostatic attraction. The self-assembly PU-A/HA+PU-B/Dex gel exhibited a much higher elastic modulus (about 0.20 MPa) and excellent shear-thinning and self-recovery behaviors, which would allow the gel to be injected through a fine syringe to fill the irregular defect. The in vitro and in vivo experiments demonstrated that the coexistence of HA and Dex in PU-A/HA+PU-B/Dex gel had a synergistic effect on cell differentiation and accelerating new bone formation, displaying a good prospect as an injectable gel for bone repair in minimally invasive surgery.

Emodin alleviates severe acute pancreatitis-associated acute lung injury by decreasing pre-B-cell colony-enhancing factor expression and promoting polymorphonuclear neutrophil apoptosis

Hongzhang Cui,Shu Li,Caiming Xu,Jingwen Zhang,Zhongwei Sun,Hailong Chen

The present study aimed to evaluate the protective effects of emodin on severe acute pancreatitis (SAP)‑associated acute lung injury (ALI), and investigated the possible mechanism involved. SAP was induced in Sprague‑Dawley rats by retrograde infusion of 5% sodium taurocholate (1 ml/kg), after which, rats were divided into various groups and were administered emodin, FK866 [a competitive inhibitor of pre‑B‑cell colony‑enhancing factor (PBEF)] or dexamethasone (DEX). DEX was used as a positive control. Subsequently, PBEF expression was detected in polymorphonuclear neutrophils (PMNs) isolated from rat peripheral blood by reverse transcription‑quantitative polymerase chain reaction and western blotting. In addition, histological alterations, apoptosis in lung/pancreatic tissues, apoptosis of peripheral blood PMNs and alterations in the expression of apoptosis‑associated proteins were examined by hematoxylin and eosin staining, terminal deoxynucleotidyl‑transferase‑mediated dUTP nick end labeling assay, Annexin V/propidium iodide (PI) assay and western blotting, respectively. Serum amylase activity and wet/dry (W/D) weight ratios were also measured. An in vitro study was also conducted, in which PMNs were obtained from normal Sprague‑Dawley rats and were incubated with emodin, FK866 or DEX in the presence of lipopolysaccharide (LPS). Apoptosis of PMNs and the expression levels of apoptosis‑associated proteins were examined in cultured PMNs in vitro by Annexin V/PI assay and western blotting, respectively. The results demonstrated that emodin, FK866 and DEX significantly downregulated PBEF expression in peripheral blood PMNs. In addition, emodin, FK866 and DEX reduced serum amylase activity, decreased lung and pancreas W/D weight ratios, alleviated lung and pancreatic injuries, and promoted PMN apoptosis by regulating the expression of apoptosis‑associated proteins: Fas, Fas ligand, B‑cell lymphoma (Bcl)‑2‑associated X protein, cleaved caspase‑3 and Bcl‑extra‑large. In addition, the in vitro study demonstrated that emodin, FK866 and DEX significantly reversed the LPS‑induced decrease of apoptosis in PMNs by regulating the expression of apoptosis‑associated proteins. In conclusion, the present study demonstrated that emodin may protect against SAP‑associated ALI by decreasing PBEF expression, and promoting PMN apoptosis via the mitochondrial and death receptor apoptotic pathways.

Platelet membrane-cloaked biomimetic nanoparticles for targeted acute lung injury therapy

Peihong Lin,Rui Gao,Wenjing Yang,Zhengyu Fang,Zhouru Wang,Mengdie Yu,Lihua Xu,Zhen Ma,Jie Fang,Wenying Yu

Acute lung injury (ALI) is a medical condition characterized by significant morbidity and elevated mortality rates; however, to date, there are no clinically approved pharmacological interventions that are both safe and effective for its treatment. In the pathophysiology of ALI, a robust inflammatory response is a critical factor. Dexamethasone (Dex), a potent glucocorticoid, is commonly employed in clinical settings to manage inflammatory conditions. However, the frequent or high-dose administration of corticosteroids can result in significant adverse effects and long-term complications. In this study, we have developed a biomimetic anti-inflammatory nanosystem, designated PM-LPs@Dex, aimed at treating ALI. This system leverages the inherent affinity of platelets for sites of inflammation, alongside the advantageous drug encapsulation properties of liposomes (LPs). By harnessing the suitable physicochemical characteristics of LPs and the distinctive biological functions of platelet membranes (PM), PM-LPs@Dex is capable of stable and sustained drug release in vitro. Experimental results regarding cellular uptake and biodistribution reveal that PM-LPs@Dex is preferentially internalized by inflammatory cells and exhibits enhanced accumulation in inflamed lung tissue compared to LPs@Dex. Pharmacokinetic studies displayed that PM-LPs@Dex showed prolonged circulation time in blood. Additionally, pharmacodynamic assessments demonstrate that PM-LPs@Dex significantly mitigates the severity of ALI, as evidenced by reductions in pulmonary edema, tissue pathology, bronchoalveolar lavage cell counts, protein concentration, and levels of inflammatory cytokines. Notably, PM-LPs@Dex also exhibits favorable biocompatibility. This research is anticipated to contribute novel strategies for the safe and effective targeted management of inflammatory diseases.

Preparation of NIR responsive resveratrol@Au nanocages using for regulating oxidative stress microenvironment of periodontitis

Mofei Han,Kaiqi Tang,Zhenhua Chen

Periodontitis, as a chronic inflammatory oral disease, is the primary cause of tooth loss. The oxidative stress microenvironment of periodontitis causes damage of both osteoblasts’ mitochondrion and periodontal tissue. Natural extracts with antioxidant properties, including resveratrol (RSV), have been reported in regulating oxidative stress microenvironment, however, the controlled release of them in periodontal tissue is still a challenge. Herein, a smart photothermal-responsive gold nanocages hydrogel (RSV-Au@H) was constructed for the treatment of periodontitis by controlling the release of resveratrol. Under the near-infrared (NIR) light irradiation for 5 min, RSV-Au@H could be heated to 45.8 ℃ due to the good photothermal effect of Au nanocages encapsulated in the hydrogel. This NIR stimulated photothermal effect of RSV-Au@H could control the release of RSV. Molecular biology indicated that RSV-Au@H could scavenge ROS with 42 % efficiency. Transmission electron microscopy (TEM) showed a significant improvement in osteoblasts mitochondrial morphology. Micro-CT scanning revealed that RSV-Au@H reduced the distance between cemento-enamel junction and alveolar bone crest (CEJ-ABC) by up to 55 % and increased bone volume fraction by about 48 %. These results suggested that this multifunctional nanoplatform with NIR controlled release of RSV might be a new therapeutic strategy for periodontitis.

Preparation of Rutin–Whey Protein Pickering Emulsion and Its Effect on Zebrafish Skeletal Muscle Movement Ability

Yiting Zhang,Wenyun Xiong,Yijing Ren,Jian Huang,Xiaoying Wang,Ou Wang,Shengbao Cai

Nutritional supplementation enriched with protein and antioxidants has been demonstrated to effectively strengthen skeletal muscle function and mitigate the risk of sarcopenia. Dietary protein has also been a common carrier to establish bioactive delivery system. Therefore, in this study, a Pickering emulsion delivery system for rutin was constructed with whey protein, and its structural characteristics, bioaccessibility, and molecular interactions were investigated. In the in vivo study, zebrafish (n= 10 in each group), which have a high genetic homology to humans, were treated with dexamethasone to induce sarcopenia symptoms and were administered with rutin, whey protein and the Pickering emulsion, respectively, for muscle movement ability evaluation, and zebrafish treated with or without dexamethasone was used as the model and the control groups, respectively. Results showed that the Pickering emulsion was homogeneous in particle size with a rutin encapsulation rate of 71.16 ± 0.15% and loading efficiency of 44.48 ± 0.11%. Rutin in the Pickering emulsion exhibited a significantly higher bioaccessibility than the free form. The interaction forces between rutin and the two components of whey proteins (α-LA and β-LG) were mainly van der Waals forces and hydrogen bonds. After treatment for 96 h, the zebrafish in Picking emulsion groups showed a significantly increased high-speed movement time and frequency, an increased level of ATP, prolonged peripheral motor nerve length, and normalized muscular histological structure compared with those of the model group (p< 0.05). The results of this study developed a new strategy for rutin utilization and provide scientific evidence for sarcopenia prevention with a food-derived resource.
