Bacillus velezensis HY19 as a sustainable preservative in post-harvest citrus (Citrus reticulata Blanco L.) fruit management

Suping Li,Jingyu Hu,Shiqi Ning,Wei Li,Ru Jiang,Jianguo Huang,Yong Li

Citrus fruit post-harvest decay has caused economic loss for fruit-dependent industries worldwide in recent decades. However, bio-preservatives offer a promising and sustainable solution. In this study, culture experiments were carried out to assess the ability of Bacillus velezensis HY19, which we previously identified, to inhibit common pathogenic fungi in postharvest citrus fruit. HY19 was capable of producing extracellular siderophores and hydrolases (protease, phosphatase, and cellulase). The cells and metabolites (fermentation broth and volatiles) of the bacterium exhibited strong suppression against Penicillium italicum , P. digitatum , and Colletotrichum gloeosporioides . LC-MS analysis of B. velezensis HY19 fermentation broth (BFB) revealed 13 antimicrobial substances (simvastatin, cinnamic acid, cyclopentanone, etc.), 3 antifungal pesticide intermediates (M-cresol, 4-hydroxybenzaldehyde, and L-menthyl acetoacetate), and P-salicylic acid. GC-MS detected 18 antimicrobial substances (2-nonanol, 2-nonanone, benzoic acid, etc.) in HY19's volatile organic compounds (VOCs). Transcriptome analysis of P. digitatum exposed to BFB stress revealed that numerous genes involved in diverse biological processes, such as DNA replication and cell wall biosynthesis, were differentially expressed. Moreover, BFB inhibited the expression of multiple genes responsible for the biosynthesis of N-glycan and proteins in P. digitatum . BFB enhanced antioxidant enzyme activities in citrus peels, reduced electrolyte leakage and reactive oxygen species (ROS) accumulation, and induced the disease resistance in citrus fruit, leading to the decrease in the infection of pathogenic fungi on citrus fruit. During storage at low-temperature (4–8 °C), BFB inhibited the disease incidence in citrus fruit for up to 90 days and achieved over 60% control efficacy. Additionally, BFB had no impact on fruit quality, making it a lower risk option. Considering the benefits of sustainability and environmentally friendly options, it could be concluded that BFB was an effective alternative for controlling citrus diseases and had the potential to be used as a fruit preservative.

Chemical Composition of Cuticle and Barrier Properties to Transpiration in the Fruit of Clausena lansium (Lour.) Skeels

Huang Hua,Wang Ling,Qiu Diyang,Lu Yusheng

The plant cuticle, as a lipid membrane covering aerial plant surfaces, functions primarily against uncontrolled water loss. Herein, the cuticle chemical composition and the transpiration of wampee fruit (Clausena lansium (Lour.) Skeels) at the green, turning, and yellow stages in cultivars of “Jixin” and “Tianhuangpi” were comprehensively studied. The coverage of wax and cutin monomers per unit of fruit surface area at the green stage was lower in “Jixin” than in “Tianhuangpi” and increased gradually during development. Cutin monomers accumulated ranging from 22.5 μg cm−2 (green) to 52.5 μg cm−2 (turning) in “Jixin” and from 36.5 μg cm−2 (green) to 81.7 μg cm−2 (yellow) in “Tianhuangpi.” The total composition of waxes ranged between 6.0 μg cm−2 (green) and 11.1 μg cm−2 (turning) in “Jixin,” while they increased from 7.4 μg cm−2 (green) to 16.7 μg cm−2 (yellow) in “Tianhuangpi.” Cutin monomers were dominated by ω-, mid-dihydroxy fatty acids (over 40%), followed by multiple monomers of α,ω-dicarboxylic acids with or without added groups, α-monocarboxylic acids with or without ω- or mid-chain hydroxy or mid-epoxy groups, primary alcohols, and phenolics. The very-long-chain (VLC) aliphatic pattern of cuticular waxes was prominently composed of n-alkanes (ranging from 21.4% to 39.3% of total wax content), fatty acids, primary alcohols, and aldehydes. The cyclic waxes were dominated by triterpenoids (between 23.9 and 51.2%), sterols, and phenolics. Water loss in wampee fruit exhibited linear changes over time, indicating an overall monofunctional barrier to transpiration. Permeance for water in wampee fruit was higher at the green stage than at the yellow stage in both “Jixin” and “Tianhuangpi,” which showed a negative correlation with the changes of VLC n-alkanes. The results showed the cuticular chemicals, including cutin monomers and waxes, in wampee fruit and further indicated the potential contributions of the cuticular chemical composition to the physiological functions in fruits.

Amelioration of chilling injury and enhancement of quality maintenance in cold-stored guava fruit by melatonin treatment

Hongbin Chen,Hetong Lin,Xuanjing Jiang,Mengshi Lin,Zhongqi Fan

The influence of melatonin treatment on the quality and chilling injury of guavas during storage at 4 ± 1 °C were evaluated. Compared with control group, fruit of guava cv. Xiguahong exposed to various concentrations (50, 100, 150, and 200 μmol/L) of melatonin showed a significantly lower fruit respiration rate, weight loss, cell membrane permeability, and chilling injury index, but a higher commercially acceptable fruit rate, higher peel L*, h ° value, and chlorophyll content. Melatonin treatment also delayed the decreases of fruit firmness, sucrose, total soluble sugar, vitamin C, titratable acidity, and total soluble solids. These data indicate that melatonin treatment could increase chilling tolerance and retain quality of cold-stored guavas. Among various concentrations of melatonin treatment, 100 μmol/L melatonin-treated guavas showed the preferable quality properties and lowest chilling injury index. Thus, melatonin may be a novel method of postharvest handling to enhance cold resistance and extend storage-life of cold-stored guava fruit.

Dispersive Micro-solid-Phase Extraction of Acaricides from Fruit Juice and Functional Food Using Cucurbituril as Sorbent

Gu Yu-Xin,Yan Tian-Ci,Yue Zi-Xuan,Li Min-Hui,Zheng Hui,Wang Shu-Ling,Cao Jun

A novel dispersive micro-solid-phase extraction (DMSPE) method is established for the determination of seven acaricides (clofentezine, benzomate, fenpyroximate, diafenthiuron, pyridaben, doramectin, and ivermectin) in fruit juice and functional food by ultra-high-performance liquid chromatography. In this method, these analytes were extracted from apple juice and curcuma wenyujin using cucurbituril used as the adsorbent, water as the extraction solvent, and methanol as the elution solvent. The primary parameters affecting the extraction efficiency were optimized by experiments for concentration of adsorbent, oscillation time, desorption solvent, and pH value and by response surface methodology. Under the optimal conditions, all target analytes had a good linear relationship, with correlation coefficients ranging from 0.9900 to 0.9940, the limits of detection were 0.42–5.16 ng/mL, the limits of quantification were 1.39–17.18 ng/mL, and the spiked recoveries of seven target compounds in real samples ranged from 70.04 to 94.95%. The intra-day precision RSD variations ranged from 0.13 to 0.70%, and the inter-day precision RSDs were between 0.38 and 1.4%, which had good reproducibility and repeatability. In general, DMSPE is a convenient, efficient, and environmentally friendly method for sample preparation, which has the potential for microextraction.

Ternary magnetic Fe3O4@C3N4@covalent organic framework for facile extraction and determination of organophosphorus pesticides in fruit

Yuhang Gao,Chenxi Zhao,Qiyue Tan,Mingkun Gao,Ge Chen,Rongqi Zhai,Xiaodong Huang,Xiaomin Xu,Guangyang Liu,Jing Wang,Yaowei Zhang,Donghui Xu

In this study, a magnetic composite material was prepared based on covalent organic framework (COFs) materials for the detection of trace organophosphorus pesticides . Fe 3 O 4 @C 3 N 4 @COFs were synthesized by a layer-by-layer assembly strategy and applied to the selective identification, detection and removal of profenofos pesticides using magnetic solid-phase extraction technique. After seven regeneration cycles, the composite exhibited excellent selectivity and affinity for profenofos, and eventually still achieved an extraction efficiency higher than 75%. Under optimized conditions, the proposed magnetic solid-phase extraction method coupled with high-performance liquid chromatography-tandem mass spectrometry showed a low limit of detection (1 × 10 −5 μg/mL) for profenofos. The developed method was successfully used to detect profenofos in spiked fruit samples with satisfactory recoveries (74.33% −116.59%) and relative standard deviations (0.27% −7.41%). This study highlights the potential of COFs-based magnetic composites in sample pretreatment for targeted analysis and is a novel pathway for efficient sample preparation in food safety analysis.

Efficacy and mechanistic insights of Kluyveromyces marxianus cell wall in enhancing pear fruit resistance against Penicillium expansum

Yalan Wu,Xin-ai Chen,Ting Yu,Fujie Yan

Postharvest fungal infestation poses a significant threat to fresh fruit, leading to substantial economic losses. This study investigated the efficacy and mechanism of yeast cell walls in inducing fruit resistance, with a focus on extracts from Kluyveromyces marxianus. Compared to the commonly referenced Saccharomyces cerevisiae cell wall, application of the K. marxianus cell wall provided superior protection in pear fruit against Penicillium expansum infection, markedly reducing disease incidence. This enhanced biocontrol activity is attributed to its distinct polysaccharide composition, characterized by 60.37% mannan, 38.34% glucan, and 1.29% chitin, and its unique structural properties. The treatment triggered a rapid wound oxidative burst, upregulated key antioxidant and defense-related enzymes, and promoted the accumulation of γ-aminobutyric acid in pear tissue. Transcriptomic analysis revealed that β-glucan and mannan components synergistically activated plant hormone signaling and plant–pathogen interaction pathways. Notably, mannan specifically modulated carbohydrate metabolism and MAPK-associated defense responses. These findings establish the K. marxianus cell wall as a potent and novel biocontrol agent, offering a sustainable strategy for managing postharvest decay through the targeted induction of fruit innate immunity.

Modularly integrated synergistic SERS strategy for ultra-trace detection of imidacloprid in fresh fruit and vegetable juices

Xinru Yu,Hongbin Pu,Da-Wen Sun

A reliable IMIPCs–AMoO 2 /C SERS analytical method was established. • Validated results were acceptable for IMI in fresh fruit and vegetable juices. • The method displayed good linear correlation, low LOD, trueness and precision. • The mechanisms of a modularly integrated synergistic strategy were explored. • Rapid detection of trace levels of IMI in the sample was achieved (10 min). Imidacloprid (IMI), a first-generation neonicotinoid insecticide, is taken up by plants and distributed to edible parts owing to its systemic transport properties. This leads to its residues in fruit and vegetable juices, posing a potential food safety risk. Herein, we report a modular integrated synergistic SERS strategy for reliable, efficient and selective detection of IMI. An IMI-imprinted polymer-pesticide carb (IMIPCs) cartridge was designed for recognition, purification, and enrichment. The AMoO 2 /C SERS substrate, optimised through annealing, aligns its bandgap with the excitation wavelength, while IMI adsorption induces band bending and triggers charge-transfer resonance, enabling signal amplification. Under optimised conditions, the method exhibits excellent linearity from 0.500 ng/mL to 50.0 ng/mL, a low detection limit of 0.0904 μg/kg, and high accuracy in spiked typical juice matrices (tomato, celery, apple, and mixed juices), with recoveries of 92.7%–98.1% (RSD%, 0.332%–2.50%) and no significant difference compared with the HPLC method at the 95% confidence level. Application to twenty real samples collected from the market, including fruit and vegetable juice samples such as kiwi, kale, banana, and orange juices and their mixtures, demonstrated good selectivity, reliability, and reproducibility, with residue levels detected in the ppb range. Overall, this LEGO-inspired SERS method provides a simple, efficient and highly selective analytical strategy for monitoring IMI residues in fruit and vegetable juices, and can serve as a supplementary screening tool for routine food safety monitoring of such juice products. Its applicability to complex high-fat or protein-rich matrices, such as milk-based or nut-based beverages, remains to be further validated through subsequent research. Deep eutectic solvents (DESs) films based on gelatin as active packaging for moisture regulation in fruit preservation

Hongchen Wan,Zhiwei Zhu,Da-Wen Sun

To develop a novel active packaging for fruit preservation, two different deep eutectic solvents (DESs) comprising choline chloride, betaine and glycerol [ChCl:Gly (1:2) and Be:Gly (1:2)] were prepared and the corresponding DESs-based films (DES@Gel) using gelatin as polymer matrix were fabricated. DES@Gel showed smoother morphologies and better optical and mechanical properties as compared with Gel. Moisture sorption isotherm curves, the enhancement of water vapour permeability (WVP) and the excellent moisture absorption–desorption cyclist performance illustrated the moisture regulation hypothesis mechanism of DES@Gel. Furthermore, cherry tomato preservation experiment was carried out and the groups treated with DES@Gel showed better performances. The moisture regulation property of DES@Gel could broaden new avenues for active packaging.

p-Coumaric acid-loaded nanoliposomes: Optimization, characterization, antimicrobial properties and preservation effects on fresh pod pepper fruit

Peiye Liu,Jing Shen,Jiankang Cao,Weibo Jiang

Novel p -coumaric acid (pCA)-loaded nanoliposomes were prepared by the thin-film hydration method, assisted with ultrasonic treatment, and optimized by the response surface methodology. The characterization showed that the fabricated pCA-loaded liposomes were nanosized spherical vesicles (83.55 ± 0.34 nm), exhibiting favorable dispersibility and encapsulation efficiency (55.70 ± 0.10 %). Fourier transform infrared spectroscopy analysis indicated that pCA was encapsulated into phospholipid bilayer through hydrophobic interaction and hydrogen bonding. Tests of temperature stability and centrifugal stability suggested that pCA-loaded nanoliposomes were less sensitive to aggregation and fusion during storage. Incubation experiments revealed that pCA-loaded nanoliposomes had a good inhibitory effect on the expansion of disease area on fresh pod pepper fruit caused by Botrytis cinerea . Additionally, pCA-loaded nanoliposomes effectively extended shelf life of fresh pod peppers by reducing weight loss and naturally-infected decays. The findings presented a viable strategy for designing liposomal encapsulation technology to efficiently enhance antimicrobial activity of pCA in food preservation.

Chemical composition of the cuticular membrane in guava fruit (Psidium guajava L.) affects barrier property to transpiration

Hua Huang,Qiaoqiao Lian,Ling Wang,Youxia Shan,Fengjun Li,Sui Kiat Chang,Yueming Jiang

The cuticular membrane covering almost all aerial plant organs has a primary function in limiting uncontrolled water loss . The guava fruits were collected and this work was done to study the potential contribution of cuticular chemical composition to fruit transpiration after harvest. The detailed cuticular chemical composition, based on gas chromatography together with mass spectrometry , and the transpiration rate determined gravimetrically in guava fruit were characterized in the present study. The predominant wax mixtures were fatty acids and primary alcohols with homologous series of C 16 –C 33 , as well as various pentacyclic triterpenoids with abundant amounts of ursolic acid, maslinic acid and uvaol. The most prominent cutin compounds were C 16 and C 18 ‒type monomers dominated by 9(10),16-diOH-hexadecanoic acid and 9,10-epoxy-ω–OH–octadecanoic acid, respectively. Relatively high water permeability with a value of 5.1 × 10 −4  m s −1 was detected for guava fruit. The lower efficiency of the cuticle as barrier to transpiration in guava fruit, as compared to that of other reported fruits, leaves, and petals, was seemingly related to the relatively short average chain-length of acyclic compounds in wax mixtures. These findings provide useful insights linking the chemical composition of the cuticular membrane that covers plant organs to putative physiological roles.

Size-modulated optical property of gold nanorods for sensitive and colorimetric detection of thiourea in fruit juice

Bing Qiang Zou,Hong Zhi Zhang,Zhu Fu,Tianrong Zhan,Jian Wang

As an important sulfur compound , thiourea (TU) has caused great concern because of its wide application as well as its serious toxicity and hazard to the environment. Thus, it is necessary to develop a sensitive and selective method for TU analysis. In this work, gold nanorods (AuNRs) acted as an optical probe to realize the sensitive and colorimetric detection of TU. In HCl medium, Fe 3+ at low concentration was difficult to oxide Au 0 to form Au + because of the high redox potential or the positive Gibbs free energy change . However, this process was possible when TU was present since the association constant between Au + and TU is great enough to bind with TU to form a stable complex to further promote the etching of AuNRs, resulting in the lower aspect ratio of AuNRs with the blue shift and intensity decrease in extinction spectra, accompanied by the divisive colors of AuNRs solution or colorful dark-field light scattering imaging of single AuNR. The blue-shift of AuNRs longitudinal plasmon resonance absorption (LPRA) band was proportional to the concentration of TU in the range of 1–250 nM and the limit of detection (3σ/k) was as low as 0.4 nM. In addition, the colorimetric method was proven with high selectivity in the presence of potential interfering compounds, which was successfully applied to the detection of TU in fruit juice samples. This proposed colorimetric method provides a simple, sensitive yet selective measurement tool for TU sensing, which may offer new opportunities in the development of colorimetric sensors for food safety in the future.

Alleviation of pericarp browning in harvested litchi fruit by synephrine hydrochloride in relation to membrane lipids metabolism

Mingyang He,Zhixing Ge,Min Hong,Qu Hongxia,Xuewu Duan,Yun Ze,Taotao Li,Yueming Jiang

This study investigated the effect of synephrine hydrochloride (Syn-HCl) on litchi pericarp browning and the potential regulating mechanism. Results showed that 0.5 g L −1 Syn-HCl significantly inhibited the development of pericarp browning. Compared to control, Syn-HCl treatment reduced malondialdehyde and H 2 O 2 content and inhibited activities of polyphenol oxidase (PPO) and peroxidase (POD). Meanwhile, Syn-HCl-treated litchi fruit exhibited higher amounts of USFA (unsaturated fatty acids) as well as lower amounts of SFA (saturated fatty acids) through regulating activities of membrane lipids-degrading enzymes: lipase and lipoxygenase. Additionally, higher ratio of USFA to SFA and double bond index were shown in Syn-HCl-treated fruit. Further analysis showed that key genes involved in fatty acid synthesis were also regulated by Syn-HCl treatment. Taken together, these findings indicated that Syn-HCl alleviated litchi pericarp browning resulting from regulation of fatty acid metabolism and maintenance of membrane integrity, which inhibited the enzymatic browning caused by PPO and POD.
