Preparation of galactosylated curcumol liposomes

Wenjie Li,Yan Lu,Yi Cheng,Hui Luo,Zhenbin Jia,Ning Li

Curcumol (Cur), which is the primary active anti-liver cancer component of Chinese traditional medicine Rhizoma zedoariae , was encapsulated in galactosylated liposomes to enhance its liver-targeting efficacy. In this study, we optimized the preparation methods, prescription, and process of galactosylated curcumol liposomes (Gal-Cur-L). The experimental results showed that the encapsulation efficiency, particle diameter, and stability coefficient of Gal-Cur-L obtained by the two-step emulsification method were improved compared with those of it obtained by other methods. Single-factor investigation showed that two factors, pH and the amount of triolein, contribute at least partly to the encapsulation efficiency and particle diameter of Gal-Cur-L. The best prescription and process were obtained through orthogonal test: the ratio of lecithin to cholesterol was 2:1, the ratio of drug to lipid was 1:2, and the amount of ether and Twain-80 were 4 ml and 1.6 g/L, respectively, 15 min for ultrasonic time, 100 W for ultrasonic power, and 20 min for homogeneous time. Using the best method, prescription, and process mentioned earlier, the basic test results of Gal-Cur-L were obtained, which demonstrated that our methods, prescription, and process of Gal-Cur-L were applicable. The encouraging results of this study provided references for the research of liver-targeting reagents of curcumol.

Synthesis of Biodegradable Porous Poly(δ-valerolactone-co-l-lactide) Monoliths from Star-Shaped Poly(l-lactide) Macroinitiators via Eutectic Polymerization

Jakkree Intranuwong,Arin Kim,Nutcha Nongyai,Seong Kyu Shin,Yong Ku Kwon

Porous polymeric biodegradable monoliths (PHIPEn, where n denotes the number of arms of the PLA macroinitiator) based on poly(δ-valerolactone-co-l-lactide) were prepared via bulk ring-opening polymerization of a δ-valerolactone/l-lactide (VL/LA) eutectic mixture (80/20) using various PLA macroinitiators. Linear (one- and two-arm) or star-shaped (four- and six-arm) PLA macroinitiators were employed to investigate the effect of initiator functionality on the morphology and properties of PHIPEns. The variation in initiator functionality strongly affected the pore morphology, cross-link density, and overall physical properties of the materials. Increasing the number of arms resulted in smaller pore size, higher cross-link density, greater crystallinity, and improved compressive strength and modulus. Thermal analysis revealed that PHIPEns prepared from star-shaped macroinitiators exhibited improved thermal stability and higher PVL crystallinity compared with those derived from linear macroinitiators. Hydrolytic degradation in phosphate-buffered saline at 37 °C showed that PHIPEns synthesized from star-shaped macroinitiators degraded more slowly, absorbed less water, and maintained a more stable pH than those derived from linear ones. These results demonstrate that combining star-shaped macroinitiators with a VL/LA eutectic mixture provides an effective strategy for designing biodegradable porous materials with tunable properties.

L-citrulline and L-arginine improve reproductive performance in primiparous and multiparous cows via distinct shared metabolic pathways and pyrimidine metabolism

Chen Fan,Changzheng Chen,Renping Liu,Muredili Maimaiti,Changgeng Li,Weijie Zao,Waike Lv,Guozhu Xu,Guodong Zhao

Background To investigate the impact of L -arginine ( L -Arg) and L -citrulline ( L -Cit) on cow reproductive performance. Methods 90 healthy, age-matched Simmental cows were randomly assigned to three groups: control, L -Arg, and L -Cit, with 30 cows per group. Estrus synchronization was performed, and various parameters were assessed, including estrus rate, conception rate, and early embryo apoptosis, following intraperitoneal administration of L -Arg and L -Cit. Additionally, reproductive hormones, serum antioxidant levels, and plasma metabolites were measured. Results Results indicated that both the estrus and conception rates in the L -Arg and L -Cit groups were significantly higher than those in the control group ( p < 0.05). Furthermore, serum LH levels and VEGF concentrations on day 1 and day 8 were elevated in the L -Arg and L -Cit groups compared to controls ( p < 0.05). Estrus rate and serum NO concentration in the L -Cit group were notably higher than those in the control group ( p < 0.05). At day 8 and day 21, estrus rate, conception rate, P levels, and serum VEGF concentrations were significantly greater in both the L -Arg and L -Cit groups compared to controls ( p < 0.05). Additionally, serum NO concentration was significantly higher in the L -Cit group than in the control group ( p < 0.05). The activities of T-AOC and GSH-Px in serum were significantly increased in both primiparous and multiparous cows in the L -Arg and L -Cit groups relative to the control group ( p < 0.05). Metabolomic analysis revealed that in primiparous cows, the protein digestion and absorption, ABC transporters, and AMPK signaling pathways were significantly enriched. In multiparous cows, pyrimidine metabolism was notably enriched. Conclusion Under the experimental conditions, L-Arg modulated the production of Dimethyl sulfone, 1,5-pentanediamine, Bassanolide, and Phenylalanine in primiparous cows through the protein digestion and absorption and ABC transporters pathways, while L -Cit influenced the same metabolites via the ABC transporters pathway. Both L -Arg and L -Cit promoted increased NO, GSH-Px, T-AOC, and VEGF levels, enhancing early embryo implantation. Additionally, L -Arg and L -Cit synergistically influenced pyrimidine metabolism, regulating the production of N-myristoylphosphorylcholine, N-oleoyl-d-erythro-sphingosylphosphorylcholine, 1-stearoyl-2-docosahexaenoyl-sn-glycero-3-phosphocholine, and 1-docosahexaenoyl- 2-stearoyl-sn-glycero-3-phosphocholine. This modulation resulted in increased NO, GSH-Px, T-AOC, and VEGF in multiparous cows, ultimately supporting early embryo implantation.

A UPLC–MS/MS method for determination of endogenous l-carnitine and acetyl-l-carnitine in serum of patients with depression

Li-Juan Nie,Jun Liang,Feng Shan,Ya-Yun Xu,Chun-Yu Yan,Xuan Zhou,Zhuo-Yu Cheng,Qing-Rong Xia

A simple, rapid, and selective ultra-performance liquid chromatography–tandem mass spectrometry method for determination of l -carnitine (LC) and acetyl- l- carnitine (ALC) in human serum was developed. Acetyl- l- carnitine-d 3 (ALC-d 3 ) was selected as internal standard (IS). After protein precipitation with acetonitrile–water (1 mL, 2:1, v/v), the analytes and IS were separated on a 2.5-μm XSelect HSS T3 C 18 column by gradient elution with methanol–water (containing 0.01% ammonia water) as the mobile phase at a flow rate of 0.2 mL/min. Analytes were detected with multiple reaction monitoring using a positive scan mode with electrospray ionization. Good linearity ( R 2 > 0.999) was observed in the concentration range for LC and ALC. The inter- and intra-day values of relative error were −10.4% to 10.0% with CVs less than 9.84%. The average recoveries of the two analytes were 91.29%–98.23%. Blood samples containing LC and ALC were stable under various storage conditions. Normal, haemolytic, and hyperlipidaemic serum had no significant effect on the quantification of LC and ALC. This method was successfully applied to study the concentrations of endogenous LC and ALC in the serum of patients with first-episode depression.

Polypeptides Micelles Composed of Methoxy-Poly(Ethylene Glycol)-Poly(l-Glutamic Acid)-Poly(l-Phenylalanine) Triblock Polymer for Sustained Drug Delivery

Xingzheng Liu,Rongrong Fan,Boting Lu,Yuan Le

Methoxy-poly(ethylene glycol)-poly(l-glutamic acid)-poly(l-phenylalanine) triblock polymers with different architecture were synthesized as drug carrier to obtain sustained and controlled release by tuning the composition. These triblock polymers were prepared by ring opening polymerization and poly(ethylene glycol) was used as an initiator. Polymerization was confirmed by1H NMR, FT-IR and gel penetration chromatography. The polymers can self-assemble to form micelles in aqueous medium and their critical micelle concentrations values were examined. The micelles were spherical shape with size of 50–100 nm and especially can arranged in a regular manner. Sorafenib was selected as the model drug and the drug loading performance was dependent on the composition of the block copolymer. In vitro drug release indicated that the polymers can realize controlled and sustained drug release. Furthermore, in vitro cytotoxicity assay showed that the polymers were biocompatible and the drug-loaded micelles can increase toxicity towards tumor cells. Confocal fluorescence microscopy assays illustrated that the micelles can be uptaken quickly and release drug persistently to inhibit tumor cell growth.Keywords:methoxy-poly(ethylene glycol);polypeptide;methoxy-poly(ethylene glycol)-poly(l-glutamic acid)-poly(l-phenylalanine);micelles;drug delivery

Selective recognition of d-tryptophan from d/l-tryptophan mixtures in the presence of Cu(II) by electropolymerized l-lysine film

Fang Wang,Wencheng Gong,Lili Wang,Zilin Chen

Selective recognition of d -tryptophan ( d -Trp) in the presence of Cu(II) was investigated at poly- l -lysine (p-l-Lys) film using electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV). p-l-Lys film was immobilized on a glassy carbon electrode (GCE) by cyclic voltammetry between 0.0 and 1.9 V in 20 mM phosphate buffer solution (pH 8.6). After the p-l-Lys/GCE electrode was incubated with d -Trp solution containing Cu(II) ions, obvious enhancement of electron transfer resistance and decrease of voltammetric current could be observed. If d -Trp was replaced by l -tryptophan (l-Trp), there was no apparent resistance and current changes. Moreover, no resistance and current changes could be found in the absence of Cu(II). It may be due to the formation of Cu complex with l -lysine and d -tryptophan. Finally, this method was successfully applied to monitoring enantiomeric composition of the d -Trp and l-Trp mixtures.

A completely bio-based corn starch/sodium carboxymethyl cellulose film with antibacterial and antioxidant performance modified by a natural alcohol ester of amino acid and curcumin

Qian Jiang,Lijia Liu,Xuan Yuan,Huan Li,Yudan Wang,Junqing Li

To address the ecological pollution caused by synthetic antimicrobial agents in conventional food packaging films, we have successfully developed a fully bio-based and sustainable food packaging film that exhibits exceptional antibacterial and antioxidant properties. In this study, we designed and synthesized two novel natural alcohol esters of amino acid (NAEAAs)—L-Trp-Men and L-Trp-Bor—through esterification reactions involving L -tryptophan ( L -Trp) and L -Menthol or Borneol, respectively. A multifunctional composite film system was constructed using natural corn starch (CS) and sodium carboxymethyl cellulose (CMC) as the matrix materials, with NAEAAs serving as the antimicrobial active components and curcumin (Cur) as the antioxidant. Experimental results demonstrated that the CS/CMC/Men2/Cur2 composite film, containing 2 wt% additives, exhibited nearly complete (≈100 %) broad-spectrum antibacterial efficiency against both Escherichia coli and Staphylococcus aureus , alongside a DPPH radical scavenging rate of 45.69 ± 1.39 %. Cytotoxicity tests confirmed the excellent biocompatibility of NAEAAs, which hydrolyze into amino acids and natural alcohols, thereby minimizing environmental contamination. These fully natural material-based films present a novel approach for developing green, pollution-free, and renewable bio-based food packaging materials.

Preparation and recrystallization of amorphous L-glutamic acid

Yuhang Liu,Yuchen Zheng,Ziming Li,Zhihai Wu,Xiong Xiao,Yongming Liu,Wangchuan Xiao,Fenghua Chen,Rongrong Xue

Amino acids, as essential biological molecules and industrial raw materials, exhibit polymorphic behavior that significantly impacts their physicochemical properties and applications. Non-classical crystallization pathways involving amorphous intermediates are commonly observed in the crystallization process of amino acids including L-glutamic acid (Glu). An effective research approach in the field of non-classical crystallization is to synthesize metastable intermediates to study the crystallization mechanisms. Herein, amorphous Glu was successfully synthesized using CaCl2 as a stabilizer through ball milling (1 : 1 Glu–CaCl2 molar ratio), spray drying (1 : 1 and 2 : 1 ratios) and aqueous evaporation (up to 5 : 1 ratio). The metastable amorphous Glu with high Glu content (87 wt% for the 5 : 1 ratio) was prepared via evaporation for the first time. The amorphous nature of the samples was confirmed by powder X-ray diffraction and low-frequency Raman spectroscopy. IR, mid-frequency Raman, and 13C solid-state NMR spectroscopy confirmed the main existence form of Glu neutral zwitterions in the amorphous Glu, while IR and mid-frequency Raman spectroscopy revealed that the structure of the amorphous Glu resembles Glu aqueous solution. Solubility and pH measurements indirectly showed that Glu are coordinated with Ca2+. Suspension recrystallization demonstrated that α polymorph is the primary recrystallization product of the amorphous Glu. The discovery of metastable amorphous intermediates would provide new insights into the non-classical crystallization of amino acids.

Electrochemiluminescent Enantiodiscrimination on Chiral Engineered l-His-ZIF8

Huimin Wang,Guangxin Wang,Ruoxi Liu,Jin Yong Lee,Shufeng Liu,Yingqi Tang,Jing Yu

The development of highly efficient chiral recognition systems by simple and cost-effective means is critical to advance the precise application of chiral molecules. Herein, a novel electrochemiluminescence (ECL) enantioselective recognition platform was constructed based on l-histidine-functionalized zeolitic imidazolate frameworks (l-His-ZIF8). Detailed characterization verified the successful preparation of the chiral l-His-ZIF8 material. It was then applied to the chiral discrimination of six amino acids: cysteine (Cys), tryptophan (Trp), phenylalanine (Phe), glutamic acid (Glu), histidine (His), and proline (Pro). Among them, the system employing Ru(bpy)32+ as the luminophore demonstrated the most efficient chiral recognition of d-Cys over l-Cys, as evidenced by a distinct ECL signal ratio (ECLd-Cys/ECLl-Cys) of 3.3 at 1 mM. A detection limit of approximately 0.193 mM was achieved for Cys enantiomers. Conversely, an inverse chiral recognition was observed for the tryptophan (Trp)/sodium tetraphenylborate (TPB) system, resulting in a stronger ECL response for l-Trp over that for d-Trp (ECLl-Trp/ECLd-Trp = 2.4 at 1 mM). Theoretical calculations indicate that the excellent ECL enantioselectivity stems from the preferential binding of the “host” l-His-ZIF8 with specific “guest” enantiomers (d-Cys or l-Trp) via stronger intermolecular interactions. This preferential binding leads to the formation of more stable diastereoisomers that facilitate electrooxidation of more “guests” to generate a stronger luminescent signal than its isomer. This study establishes a novel strategy for electrochemiluminescent enantiomer discrimination using chiral site-engineered metal–organic frameworks (MOFs) and opens new avenues for the future of chiral sensing.

Prokinetic Activity of Mulberry Fruit, Morus alba L.

Tae Sik Sung,Seung-Bum Ryoo,Chang-Hyun Lee,Seon-Min Choi,Joo-Won Nam,Hyun-Bok Kim,Ji young Lee,Jung-Dae Lim,Kyu Joo Park,Hyun-Tai Lee

The fruit ofMorus albaL. (MAF) has been consumed as a food worldwide. MAF has also been widely used in traditional medicine for thousands of years in East Asia, and its diverse bioactivities have been reported in numerous publications. However, no prokinetic activity has been reported for MAF or its components. In the present study, therefore, we investigated the effects of MAF on gastrointestinal motor function by measuring the intestinal transit rate (ITR) of Evans blue in mice in vivo. The ITR values accelerated by MAF were significantly higher than those accelerated by cisapride or metoclopramide, suggesting that MAF has potential as a new prokinetic agent to replace cisapride and metoclopramide. We also investigated the effects of MAF on myogenic and neurogenic contractions in human intestinal smooth muscles by measuring spontaneous contractions of smooth muscle strips, smooth muscle contractions induced by neural stimulation, and migrating motor complexes from intestinal segments in the human ileum and sigmoid colon in situ. MAF increased both myogenic and neurogenic contractions to enhance ileal and colonic motility in the human intestine. Taken together, these results indicate that MAF enhanced intestinal motility by increasing both myogenic and neurogenic contractions, thereby accelerating the ITR.Keywords:intestinal transit;Morus alba;mulberry fruit;myogenic contraction;neurogenic contraction;prokinetic;smooth muscle

Phytochemical profiles and bioactivities of Parnassia palustris L.

Liang Liu,Jia Chen,Xuejian Chang,Juan Qin,Hanpeng Lai,Xianwen Zhang

Introduction The whole grass Parnassia palustris L. has long been used as Mongolian medicine in China. However, few data are available on its phytochemical profile and bioactivity . This study aims to investigate several secondary metabolite profiles and bioactivities of P. palustris extract and its fractions. Methods A spectrophotometric method was used to measure the total phenol, total phenolic acid, total flavonoid , and total terpenoid contents of the extract and different extraction sites. 2,2-Diphenyl-1-picrylhydrazyl(DPPH)/2,2′-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) assays were performed to evaluate in vitro antioxidant activities . α-Glycosidase/α-amylase/acetylcholinesterase (AChE)/butyrylcholinesterase (BChE)-inhibitory activities were determined using spectrophotometric methods. A Cell Counting Kit-8 (CCK-8) assay was carried out to test tumor cytotoxicity. Results The n -butanol fraction (NBF) had the highest total phenol/flavonoid/phenolic acid contents, and the petroleum ether fraction (PEF) had the highest total terpenoid content. The IC 50 values of DPPH and ABTS radicals scavenged by the 95% ethanol crude extract of the whole grasses of P. palustris (EEP), ethyl acetate fraction (EAF), NBF, and aqueous fraction (AF) were in the range of 15.33 ± 1.24 to 87.33 ± 3.86 μg/mL and 75.33 ± 2.87 to 815.67 ± 7.76 μg/mL, respectively. The NBF showed the best scavenging effects, while the PEF did not show activities in either assay. EEP, PEF, EAF, and AF did not show inhibitory activities against AChE, except for NBF (IC 50  = 484.00 ± 9.93 μg/mL). The IC 50 values of EEP, NBF, and AF in the BChE-inhibitory assay were in the range of 220.24 ± 8.56 to 413.93 ± 2.03 μg/mL, with NBF exhibiting the best BChE-inhibitory activity. Additionally, neither PEF nor EAF showed inhibitory effects against BChE. None of the test samples were found to have inhibitory activities against α-glucosidase and α-amylase. EEP, PEF, EAF, and NBF exhibited cytotoxicity against the HepG2 cell line, with IC 50 values in the range of 92.93 ± 7.61 to 377.93 ± 13.43 μg/mL. EAF showed the best tumor cytotoxicity, while AF had no cytotoxicity. In general, NBF was found to have potent in vitro antioxidant activity and dual AChE/BChE inhibitory activity for the first time. The EAF exhibited potent tumor cytotoxic activity against the HepG2 cell line. The results of Pearson's correlation analysis showed that flavonoids and phenols may be responsible for the BChE-inhibitory activity of P. palustris . Furthermore, phenols may contribute to its antioxidant activity . Conclusion NBF has the potential for the development of new antioxidants and anti-Alzheimer's disease (AD) drugs . The present report will be helpful for better understanding and applying this traditional medicinal plant in the future.
