The amelioration of alcohol-induced liver and intestinal barrier injury by Lactobacillus rhamnosus Gorbach-Goldin (LGG) is dependent on Interleukin 22 (IL-22) expression

Yuli Ge,Huiling Sun,Lanman Xu,Weiping Zhang,Jiaojian Lv,Yongping Chen

Alcoholic liver disease (ALD) is a common clinical liver injury disease. Lactobacillus rhamnosus Gorbach-Goldin (LGG) has been revealed to alleviate alcohol-induced intestinal barrier and liver injury. However, the underlying mechanism of LGG treatment for ALD remains unclear. To clarify this aspect, a chronic plus binge ALD model was constructed using C57BL/6 mice in line with a chronic alcohol binge feeding protocol. Interleukin 22 (IL-22) level was determined by quantitative real-time polymerase-chain reaction and enzyme-linked immunosorbent assays. Effects of LGG in model or IL-22 knockdown in LGG-treated model on the liver injury and steatosis status, as well as intestinal barrier function were assessed by hematoxylin eosin (HE) staining. Levels of alanine aminotransferase (ALT), triglyceride (TG), and aspartate aminotransferase (AST) in serum were measured by the corresponding kit. Western blot analysis was conducted to detect protein expressions of intestinal tight junction protein 1 (ZO-1) and Claudin-1. Concretely, LGG elevated IL-22 level in liver tissues and serum, while inhibiting ALT, TG, and AST levels in alcohol-exposed mice. Moreover, LGG alleviated liver injury, steatosis, and intestinal barrier injury caused by alcohol, and enhanced ZO-1 and Claudin-1 expressions. Furthermore, IL-22 knockdown increased ALT, TG, and AST levels in serum, and aggravated liver injury, steatosis, and intestinal barrier injury. ZO-1 and Claudin-1 levels were downregulated by IL-22 silencing. Importantly, downregulation of IL-22 reversed the effect of LGG on the liver and intestinal barrier injury. To conclude, LGG protects against chronic alcohol-induced intestinal and liver injury via regulating the intestinal IL-22 signaling pathway.

A synbiotic formulation of Lactobacillus reuteri and inulin alleviates ASD-like behaviors in a mouse model: the mediating role of the gut–brain axis

Chuanchuan Wang,Weixuan Chen,Yishan Jiang,Xiao Xiao,Qianhui Zou,Jiarui Liang,Yu Zhao,Qianxu Wang,Tian Yuan,Rui Guo,Xuebo Liu,Zhigang Liu

Autism Spectrum Disorder (ASD), a complex neurodevelopmental disorder marked by social communication deficits and repetitive behaviors, may see symptom amelioration through gut microbiota modulation. This study investigates the effects of a synbiotic – specifically a probiotic amplified by prebiotic supplementation – on ASD-like mouse model's social deficiencies. This model was established via valproic acid injection into pregnant females. Post-weaning, male progeny received daily synbiotic treatment, a combination of Lactobacillus reuteri (L. reuteri) and inulin, for four weeks. Results indicated that the synbiotic rectified social impairments and attenuated inflammatory cytokine expressions in the brain. Moreover, synbiotic intervention protected gut barrier integrity and altered the gut microbiota composition, enhancing the butyrate-producing Bifidobacterium abundance. The synbiotic elevated metabolites such as butyrate and 3-hydroxybutyric acid (3-HB), alongside upregulated genes associated with 3-HB synthesis in the colon and liver, and brain receptors. Conclusively, the synbiotic combination of L. reuteri and inulin mitigated ASD-related social impairments, partially via their regulatory effect on the gut–brain axis.

Comparative Assessment of the Bioremedial Potentials of Potato Resistant Starch-Based Microencapsulated and Non-encapsulated Lactobacillus plantarum to Alleviate the Effects of Chronic Lead Toxicity

Muhammad Zafarullah,Ramzan Rabia,Zhang Shanshan,Hu Haijuan,Hameed Ahsan,Bakry Amr M.,Dong Yongzhen,Wang Lufeng,Pan Siyi

Lead (Pb) is a well-recognized and potent heavy metal with non-biodegradable nature and can induce the oxidative stress, degenerative damages in tissues, and neural disorders. Certain lactic acid bacterial strains retain the potential to mitigate the lethal effects of Pb. The present work was carried out to assess the Pb bio-sorption and tolerance capabilities of Lactobacillus plantarum spp. Furthermore, potato resistant starch (PRS)-based microencapsulated and non-encapsulated L. plantarum KLDS 1.0344 was utilized for bioremediation against induced chronic Pb toxicity in mice. The experimental mice were divided into two main groups (Pb exposed and non-Pb exposed) and, each group was subsequently divided into three sub groups. The Pb exposed group was exposed to 100 mg/L Pb(NO3)2 via drinking water, and non-Pb exposed group was supplied with plain drinking water during 7 weeks prolonged in vivo study. The accumulation of Pb in blood, feces, renal, and hepatic tissues and its pathological damages were analyzed. The effect of Pb toxicity on the antioxidant enzyme capabilities in blood, serum, as well as, on levels of essential elements in tissues was also calculated. Moreover, KLDS 1.0344 displayed remarkable Pb binding capacity 72.34% and Pb tolerance (680 mg/L). Oral administration of both non- and PRS- encapsulated KLDS 1.0344 significantly provided protection against induced chronic Pb toxicity by increasing fecal Pb levels (445.65 ± 22.28 μg/g) and decreasing Pb in the blood up to 137.63 ± 2.43 μg/L, respectively. KLDS 1.0344 microencapsulated with PRS also relieved the renal and hepatic pathological damages and improved the antioxidant index by inhibiting changes in concentrations of glutathione peroxidase, glutathione, superoxide dismutase, malondialdehyde, and activated oxygen species, which were affected by the Pb exposure. Overall, our results suggested that L. plantarum KLDS 1.0344 either in free or encapsulated forms hold the potentiality to deliver a dietetic stratagem against Pb lethality.

Mangiferin-loaded Lactobacillus rhamnosus extracellular vesicles attenuate doxorubicin-induced mesenchymal stem cell senescence and preserve osteogenic and chondrogenic differentiation via multi-module protein cargo remodeling

Zhiyuan Li,Lei Jiang,Yuqi Li,Huifen Li,Shihan Zhang,Lijuan Han

Chemotherapy-induced mesenchymal stem cell (MSC) senescence impairs tissue repair and limits regenerative therapies. Probiotic-derived extracellular vesicles (EVs) have emerged as biocompatible nanocarriers, yet the molecular determinants of their therapeutic efficacy remain poorly defined.Culturing Lactobacillus rhamnosus PB LR76 with mangiferin (MGF) loads MGF into extracellular vesicles (EVs, 2.66% w/w) and remodels their proteome, generating multimodule anti senescence cargo. PB LR76 MGF EVs (50 μg/mL protein, 1.33 μg/mL MGF) significantly attenuated doxorubicin induced mesenchymal stem cell (MSC) senescence: they reduced ROS, DNA damage (γ-H2AX), and p53/p21 activation, while preserving mitochondrial membrane potential. In an eight group, three strain comparison (PB LR76, probiotic LGG, nonprobiotic ATCC 4356), only probiotic strain EVs preserved osteogenic and chondrogenic differentiation (Alizarin Red S/Alcian Blue; Runx2, ALP, COL1A1, OSX, ACAN, COL2A1, SOX9), confirming strain specificity. DIA Hyper quantitative proteomics (549 proteins, n = 3) identified 134 differentially abundant proteins enriched in PB LR76 MGF EVs, coordinated across four functional modules: redox/stress proteins (thioredoxin, NADH dehydrogenase FC = 30.8), six glycolytic moonlighting enzymes, DnaK GrpE/GroEL chaperones, and metabolic/ribosomal moonlighting proteins. PubMed cross referencing mapped 21 of 26 probiotic EV effector categories, 11 enriched in PB LR76 MGF EVs, to 10 human pathways, including TLR2/NF-κB, integrin/FAK survival, and NAD+/sirtuin dependent p53 deacetylation. These findings establish a proteome to phenotype framework for probiotic EV bioactivity and position PB LR76 MGF EVs as a food grade nanotherapeutic strategy against chemotherapy induced tissue damage.

Mannose-cRGD Engineered Lactobacillus helveticus -Derived Exosomes Enhance Loaded Pterostilbene Bioavailability: From Dual Insights of Membrane Lipid Orderliness and Dual-Targeting Endocytosis Modulation

Qiu-Ya Ji,Ran Meng,Si-Jie Wu,Ran Feng,Bao Zhang

Abstract Exosomes derived from probiotics are emerging as innovative oral nanocarriers but face challenges in targeted delivery and stability. Here, Lactobacillus helveticus-derived exosomes (LHexo) were dual-functionalized with mannose and cRGD (McRLHexo), yielding improved membrane lipid ordering, denser PEG brush layer, and distinct “cloud-like” surface nanostructure. McRLHexo exhibited enhanced stability under UV (transmittance reduction > 98%), thermal, and digestive conditions (drug retention >86%), and increased pterostilbene (PTS) loading. Functionally, McRLHexo@PTS achieved efficient epithelial translocation via cRGD-integrin pathways and macrophage targeting via Man/cRGD dual-recognition. In DSS-induced colitis mice, McRLHexo showed optimal targeting accumulation and uptake in the colon. Importantly, McRLHexo@PTS markedly alleviated colitis, restoring body weight, colon length, and epithelial integrity while reducing inflammatory cytokines and oxidative stress and enhancing antioxidant defenses. This study establishes a dual-targeted probiotic exosome platform for oral therapy and provides insights into ligand-membrane engineering for precision intestinal delivery.

Lactobacillus paracasei N1115 alleviates hyperuricemia in mice: regulation of uric acid metabolism as well as its impact on gut microbiota and short-chain fatty acids

Hongyu Zhang,Da Wang,Dongning Li,Bingkun Bao,Qian Chen,Sunhaoran Wang,Shichao Han,Mengge Zhao

Hyperuricemia (HUA) is a worldwide metabolic disorder characterized by abnormally elevated serum uric acid (SUA) levels, and recent studies suggest that probiotics have potential in mitigating HUA. This study aimed to evaluate the efficacy of Lactobacillus paracasei N1115 in alleviating HUA in mice and explore its underlying mechanisms. The results demonstrated that both high and low-dose L. paracasei N1115 reduced SUA levels in vivo by 29.18 and 27.29%, respectively (p < 0.05), effectively mitigating HUA. Additionally, the probiotic protected renal function, mitigated tissue damages and inflammation. Mechanically, it effected uric acid (UA) metabolism by regulating the UA-production related enzymes xanthine oxidase (XOD), adenosine deaminase (ADA), and 5′-nucleotidase (5′-NT), as well as the urate transpoters urate transporter 1 (URAT1) and glucose transporter 9 (GLUT9), and organic anion transporter 3 (OAT3). Moreover, L. paracasei N1115 reshaped the gut microbiota and significantly increased the abundance of Bifidobacterium, while modulating renal metabolism and elevating butyric acid levels in gut. These findings suggest that L. paracasei may alleviate HUA by enhancing butyrate levels through a cross-feeding interaction with Bifidobacterium. Although further experiments are required to substantiate underlying mechanisms, this study provides a basis for HUA-targeting functional foods research.

Fabrication of WPI-EGCG covalent conjugates/gellan gum double network emulsion gels by duo-induction of GDL and CaCl2 for colon-controlled Lactobacillus Plantarum delivery

Xinsheng Qin,Qingli Bo,Panzhu Qin,Sufang Wang,Kaiyong Liu

In this study, we demonstrated the role of whey protein isolate (WPI)/(−)-epigallocatechin-3-gallate (EGCG) covalent conjugates/gellan   gum double network emulsion gels by duo-induction of glucono-δ-lactone (GDL) and CaCl 2 on the digestion viability of Lactobacillus Plantarum powders. We employed a novel one-step continuous cold gelation strategy to product double network emulsion gels. The gellan gum in continuous phase was first induced by CaCl 2 to form a gel in a short time. Besides, the GDL was improved covalent cross-linking between WPI-EGCG covalent conjugate Pickering particles or particle-coated oil droplets because of the slowly pH reduction. The first cross-linked gellan gum gels could be more closely integrated into the GDL-induced second gel networks with synergistic role, which promoted the formation of compact and homogeneous networks through hydrophobic interactions and disulfide bonds. Thus, the results provide a promising strategy for probiotic powders encapsulation of double network emulsion gels in oral applications of gel type foods.

AI-2/LuxS Quorum Sensing System Promotes Biofilm Formation of Lactobacillus rhamnosus GG and Enhances the Resistance to Enterotoxigenic Escherichia coli in Germ-Free Zebrafish

Zhaoxi Deng,Kangwei Hou,Teresa G. Valencak,Xin M. Luo,Jianxin Liu,Haifeng Wang

The LuxS enzyme plays a key role in both quorum sensing (QS) and the regulation of bacterial growth. It catalyzes the production of autoinducer-2 (AI-2) signaling molecule, which is a component of the methyl cycle and methionine metabolism. This study aimed at investigating the differences between the Lactobacillus rhamnosus GG (LGG) wild-type strain (WT) and its luxS mutant (ΔluxS) during biofilm formation and when resisting to inflammation caused by Enterotoxigenic Escherichia coli (ETEC) in germ-free zebrafish. Our results suggest that in the absence of luxS when LGG was knocked out, biofilm formation, extracellular polysaccharide secretion and adhesion were all compromised. Addition of synthetic AI-2 indeed rescued, at least partially, the deficiencies observed in the mutant strain. The colonizing and immunomodulatory function in WT versus ΔluxS mutants were further studied in a germ-free zebrafish model. The concentration of AI-2 signaling molecules decreased sharply in zebrafish infected with the ΔluxS. At the same time, compared with the ΔluxS, the wild-type strain could colonize the germ-free zebrafish more effectively. Our transcriptome results suggest that genes involved in immunity, signal transduction, and cell adhesion were downregulated in zebrafish infected with ΔluxS and WT. In the WT, the immune system of germ-free zebrafish was activated more effectively through the MAPK and NF-κB pathway, and its ability to fight the infection against ETEC was increased. Together, our results demonstrate that the AI-2/LuxS system plays an important role in biofilm formation to improve LGG and alleviate inflammation caused by ETEC in germ-free zebrafish.IMPORTANCE Lactobacillus rhamnosus GG is a widely used probiotic to improve host intestinal health, promote growth, reduce diarrhea, and modulate immunity. In recent years, the bacterial quorum sensing system has attracted much attention; however, there has not been much research on the effect of the LuxS/AI-2 quorum sensing system of Lactobacillus on bacteriostasis, microbial ecology balance, and immune regulation in intestine. In this study, we used germ-free zebrafish as an animal model to compare the differences between wild-type and luxS mutant strains. We showed how AI-2/LuxS QS affects the release of AI-2 and how QS regulates the colonization, EPS synthesis and biofilm formation of LGG. This study provides an idea for the targeted regulation of animal intestinal health with probiotics by controlling bacteria quorum sensing system.

Synergetic effects of Lactobacillus plantarum and Rhizopus oryzae on physicochemical, nutritional and antioxidant properties of whole-grain oats (Avena sativa L.) during solid-state fermentation

Han Wu,Hao-Nan Liu,Ai-Min Ma,Jian-Zhong Zhou,Xiu-Dong Xia

In this study, Lactobacillus plantarum strains, namely 70810, B1-6, and ZR were used to confirm the synergetic effects of co-inoculation with Lactobacillus and Rhizopus oryzae on whole-oat solid state fermentation (SSF). The results indicated that number of viable bacterial cells increased more quickly (10.72–11.23-fold higher) during co-inoculated SSF than that during pure Lactobacillus fermentation. Fungal ergosterol content reached slightly less during co-inoculation (137.04–155.12 mg/kg) than that during R. oryzae fermentation (193.64 mg/kg). However, compared to R. oryzae , R. oryzae combined with Lactobacillus produced more obvious hydrolysis of macromolecules and accelerated more water transformation from confined to free form. As for co-inoculated SSF of oat, the reducing sugar content reached the highest at 41.09–47.42 g/100g and then dropped, and free amino acid nitrogen content varied gradually until 40.66–41.67 mg/100g. Fermented oats (FOs) had higher total phenolic content than unfermented oat (UO) (2.56–2.72-fold, FO vs . UO), and their DPPH radical scavenging activity, ferric reducing capacity, and hydroxyl scavenging activity were increased 2.04-, 2.10-, and 1.99-fold at maximum (FO vs . UO), respectively. The Lactobacillus showed strain dependence during co-inoculated SSF, among which B1-6 displayed the best to enhance oat's overall quality.

Nutrition and flavor analysis of Spirulina through co-fermentation with Lactobacillus acidophilus and Kluyveromyces marxianus and its effect on attenuating metabolic associated fatty liver disease

Jinhui Yu,Chao Liu,Mingjie Wang,Yuanyuan Liu,Liyuan Ran,Zichao Yu,Deyuan Ma,Gao Chen,Fuwen Han,Shengbo You,Yuping Bi,Yingjie Wu

Spirulina is widely known as the “Super Nutritious Food of the Future” due to its exceptional nutritional properties. In this article, we investigated the effects of fermentation using a combination of Lactobacillus acidophilus KDB-03 and Kluyveromyces marxianus GDMCC 2.119 on Spirulina. Fermentation significantly increased the total flavonoid and phenol levels in Spirulina, and enhanced the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging capacity and ferric-reducing antioxidant power. Untargeted ultra-performance liquid chromatography-electrospray tandem mass spectrometry-based metabolomics and gas chromatography-ion mobility spectrometry analysis revealed alterations in the metabolites and flavor components of fermented Spirulina (FS). Furthermore, a mice experiment demonstrated that FS could effectively alleviate metabolism-associated fatty liver disease (MAFLD) by regulating liver metabolism, intestinal flora, and facilitating communication between the liver and gut. These findings suggest that fermentation could be a innovative approach to enhance the diversity of Spirulina products, and potentially be used as a strategy for the prevention and treatment of MAFLD.

Characterization of a novel beta-cypermethrin-degrading strain of Lactobacillus pentosus 3-27 and its effects on bioremediation and the bacterial community of contaminated alfalfa silage

Fang Liu,Jie Bai,Wenkang Huang,Fuhou Li,Wencan Ke,Yixin Zhang,Dongmei Xie,Bo Zhang,Xusheng Guo

In this study, a novel beta-cypermethrin (beta-cyp)-degrading strain Lactobacillus pentosus 3–27 (LP3–27) was screened from beta-cyp-contaminated silage. The strain could degrade 96% of beta-cyp (50 mg/L) in MSM medium after 4 d of culture, while the strain lost its degradation ability when the beta-cyp concentration reached 250 mg/L. The effects of LP 3–27 on fermentation, bacterial community, and bioremediation of contaminated alfalfa silage at two dry matter (DM) contents were studied. The results showed that inoculation with LP3–27 not only degraded beta-cyp, but also improved the fermentation quality of alfalfa silage after 60 d of ensiling. Meanwhile, L. pentosus dominated the bacterial community during ensiling in LP3–27 inoculated silages, whereas Pediococcus acidilactici was the dominant species in the control silage. LP3–27 inoculation also simplified the bacterial interaction networks of ensiled alfalfa. Beta-cyp degradation was positively correlated with L. pentosus in LP- inoculated silages, which confirmed the function of beta-cyp degradation by L. pentosus . In addition, higher beta-cyp degradation was observed in silage with 35% versus 43% DM. In summary, strain LP3–27 could be used as a candidate inoculum for bioremediation of beta-cyp-contaminated silage and to produce safe silage for animal production.

W/O/W emulsions with gelled aqueous and oil phases: Construction, characterization, and their effect on enhancing the stress resistance of Lactobacillus plantarum and Streptococcus thermophilus

Yejun Zhong,Hailin Zheng,Ruiyun Chen,Wei Liu,Shan Sun,Liqiang Zou,Er Sheng Gong

Dual-phase gelation in W/O/W emulsions enables robust, tunable probiotic delivery. • Oil-phase gelation using PGPR and wax reduces droplet size and raises viscosity. • Aqueous-phase gelation using CaCO₃-pectin/WPI network increases bound water. • Dual-phase gelation yields highest mechanical strength and probiotic survival. To overcome the poor stability and low probiotic survival associated with traditional delivery systems, this study systematically developed and compared water-in-oil-in-water (W/O/W) emulsions with different phase gelation strategies. Three emulsion types were constructed: oil-phase (O-W/O/W), external aqueous-phase (A-W/O/W), and dual-phase gelled (B-W/O/W) emulsions. The results revealed that dual-phase gelation (B-W/O/W) exhibited a superior synergistic effect compared to single-phase gelation. Both aqueous- and oil-phase gelation increased apparent viscosity and restricted internal water mobility, but their combination provided the most robust physical barrier. Specifically, oil-phase gelation reduced particle size by immobilizing free water, while aqueous gelation formed a dense protective network. This synergistic protection significantly enhanced probiotic viability during pasteurization, in vitro digestion, and long-term storage. This study demonstrates that dual-phase gelation is a highly effective and novel strategy for the development of high-performance delivery vehicles, providing a theoretical basis for the commercial application of sensitive probiotics in functional foods.
