Mn2+-loaded carboxymethyl chitosan nanoparticle as a multifunctional adjuvant to efficiently enhance the performance of inactivated pseudorabies virus vaccines

Niling Xie,Jing Zhang,Yandong Cui,Bing Yang,Anan Guo,Yang Wu,Wei Sun,Daniel Scherman,Yingshuai Liu

Conventional adjuvants (such as Alhydrogel and oil-in-water emulsion) formulated vaccines normally induce an inadequate immune response and poor cellular immunity. In this work, a Mn 2+ -loaded carboxymethyl chitosan nanoparticle dispersed with sodium alginate (CMS adjuvant) has been designed and used as a novel nanoadjuvant in inactivated PRV vaccines to overcome above challenges. Comprehensive biocompatibility assessments confirmed the CMS adjuvant's favorable safety profile in both in vitro and in vivo models. CMS-adjuvated PRV vaccine successfully induced higher innate and adaptive immune response as compared with control adjuvants, including higher specific IgG antibody titres and IgG2a/IgG1 ratio, elevated Th1-/Th2-type cytokine secretion, and increased T cell activation and differentiation. The enhanced performance was attributed to three primary factors: (1) sustained antigen release at the injection site, (2) CMS-assisted highly efficient antigen uptake and presentation by dendric cells (DCs), and (3) Mn 2+ -induced activation of cyclic GMP-AMP synthase-stimulator of interferon gene (cGAS-STING) pathway, leading to increased type I interferons (IFNs) secretion and DCs maturation. In a mouse challenge experiment with a wild-type PRV, the CMS-adjuvanted vaccine demonstrated superior protective efficacy (90 % survival rate) to commercial Alum (40 %) and biphasic 201 (80 %) adjuvants. Thus, the CMS nanoparticles have a great potential to be a promising nanoadjuvant in various vaccines.

Oxygen vacancies mediated ferromagnetism with enhanced optical and dielectric properties in Gd3+ and Co2+/Mn2+ co-doped ZnO nanocrystals

Adil Murtaza,Xianghao Song,Saddaf Noreen,Wei Zhang,Wu Haoyi,Awais Ghani,Fazal Kabir,Wen-Liang Zuo,Kaili Li,Azhar Saeed,Sen Yang

Herein, diluted magnetic semiconductor (DMS) nanocrystals (NCs) of Gd x TM 0.07 Zn 0.93−x O (TM=Co, Mn) with Gd concentrations of 3 at% and 6 at% and a fixed TM concentration of 7 at% were synthesized using sol-gel technique, and their multifunctional properties were studied. X-ray diffraction (XRD) analysis confirmed a wurtzite hexagonal structure for all synthesized NCs with no detectable impurity peaks. Williamson–Hall (W–H) plots revealed a trend of decreasing crystallite size and increasing lattice strain with higher doping levels. Transmission electron microscopy (TEM) based morphological analysis showed irregularly shaped NCs with an average crystallite size of 32–40 nm. X-ray photoelectron spectroscopy (XPS) confirmed the 3 + oxidation state of Gd and 2 + states for Co/Mn, as well as the presence of oxygen defects. Ultraviolet–visible (UV–Vis) absorption spectroscopy indicated a reduction in the band gap for the doped samples. Photoluminescence (PL) spectra revealed the formation of defects and oxygen vacancies in the samples. Magnetic characterization demonstrated room-temperature ferromagnetic (RTFM) behavior in all doped NCs, which was further verified by electron paramagnetic resonance (EPR) spectroscopy. Combining with PL and XPS results, the observed ferromagnetism is attributed to interactions between dopant ions and trapped electrons, mediated by magnetic polarons. Dielectric measurements revealed an increase in both the dielectric constant and dielectric loss, suggesting enhanced dielectric polarization. The AC conductivity increased with frequency due to hopping charge carriers. These results show that oxygen vacancies created by Gd 3+ and Co 2+ /Mn 2+ co-doping in the host ZnO lattice have a significant impact on the multifunctional properties of ZnO nanocrystals.

Enhanced removal of multiple metal ions (Fe3+, Mn2+, Ni2+, Cd2+, and Sr2+) from aqueous solutions using modified natural clinoptilolite

Tallat Munir,Ming Liu,Shiyang Bai,Jihong Sun

The effective removal of multiple metal ions from aqueous solutions in a single-treatment process was explored using modified natural clinoptilolite (NCP). Batch adsorption experiments were conducted to evaluate the impact of critical factors, such as adsorbent dosage, contact time, adsorbate concentration, pH, and temperature, on the removal efficiency. The modified NCP achieved maximum adsorption capacities of 81.4, 78.4, 75.2, 72.2, and 68.8 mg·g −1 for Cd 2+ , Fe 3+ , Ni 2+ , Mn 2+ , and Sr 2+ , with removal efficiencies of 95.0 %, 90.1 %, 90.0 %, 86.5 %, and 82.5 %, respectively, at pH 3.0 and 25 °C for Fe 3+ , and pH 7.5 and 85 °C for the others. Adsorption kinetics followed the pseudo-second-order model while intra-particle diffusion, analyzed via the Weber-Morris model indicated that pore diffusion was a dominant mechanism, particularly for Fe³⁺. The Langmuir isotherm provided the best fit to the equilibrium data. Thermodynamic analysis confirmed the viability of the adsorption process, with Fe 3+ exhibiting exothermic adsorption, while the uptake of Mn 2+ , Ni 2+ , Cd 2+ , and Sr 2+ was endothermic. Negative entropy changes indicated enhanced ordering of adsorbed metal ions via strong electrostatic interactions. The modified NCPs demonstrated excellent regeneration capability and stability under acidic, basic, and thermal conditions. These findings provide valuable insights for designing effective multi-ion adsorption systems with enhanced performance and reliability.

Anti-thermal quenching and self-reduction characteristics in Mn2+-doped LiScGeO4: Dual temperature effects on unique emission peak shifts

Yue Zhong,Tao Su,Jing Xie,Chao Wang,Wenming Wang,Yan Pan,Xiantao Wei,Yong Li

Mn-doped phosphors exhibiting self-reduction behavior have garnered significant attention in the study of luminescent materials due to the defects generated during the self-reduction process and their unique luminescent behavior. In this paper, LiScGeO 4 red phosphors activated by the self-reduction of Mn 4+ to Mn 2+ were synthesized by high-temperature solid-state reaction in an air atmosphere. The defects generated during the self-reduction process were leveraged to attain exceptional anti-thermal quenching characteristics, with luminescent intensity at a high temperature of 430 K being 1.23 times compared to that at room temperature of 300 K. In addition, a peculiar phenomenon occurred where the emission peak initially blueshifts and subsequently redshifts with the increasing temperature. This study includes an analysis of the Rietveld-refined lattice parameters and surface micromorphology of the samples. The theoretical energy gap of the perfect LiScGeO 4 matrix was calculated using density functional theory . Furthermore, the unique emission peak shift characteristics of Mn 2+ were elucidated through the interaction between Mn 2+ ions and the strength of the crystal field . Utilizing a defect model, the self-reduction mechanism and the anti-thermal quenching behavior were clarified. These findings will enhance the understanding of the effects of crystal defects and ion pair interactions on luminescent materials , and encourage further explorations of self-reducing phosphors to develop new high thermal stability phosphors for practical applications.

Orange LED and cell fluorescence imaging of Mn2+ doped CsPbCl3 water-soluble nanocrystals with improved stability and optimized luminescence

Ting Fan,Jian-tao Lü,Wen-qiang Deng,Jun-xian Mai,Jie Liang,Ting-ting Deng,Wen Fan,Qin-song Zhou,Qi-feng Lin

Water-soluble Mn 2+ doped CsPbCl 3 nanocrystals coating by 6-aminocaproic acid (ACA) ligand were prepared by a simple ligand assisted reprecipitation method at room temperature. The CsPbCl 3 :Mn 2+ /ACA nanocrystals were almost cubic phase and had mean size of about 10 nm. Due to the coating and passivation of ACA, the CsPbCl 3 :Mn 2+ /ACA nanocrystals retained the initial fluorescence intensity of 20% after being dispersed in deionized water for 20 days. The orange LED device based on CsPbCl 3 :Mn 2+ /ACA nanocrystals were prepared with excellent luminescence properties. In the cytotoxicity test, even if the MCF-7 cells were cultured for 24 h at the concentration of CsPbCl 3 :Mn 2+ /ACA nanocrystals as high as 160 μg·ml −1 , the cell survival rate was still 70%. Confocal fluorescent cell imaging results showed that the nanocrystals had remarkable fluorescent cell imaging effect even after 24 h. The synthesized CsPbCl 3 :Mn 2+ /ACA nanocrystals have nice water solubility, good water stability, low cytotoxicity and highly luminescence properties, and have great application potential in the field of orange LED and fluorescent probes .

Chemiluminescence method based on the KIO4−K2CO3−Mn2+ reaction for rapid and sensitive determination of forchlorfenuron in dried fruit

Baisen Chen,Rongxia Tan,Yufei Hu,Gongke Li

Forchlorfenuron is a low-toxic phenylurea plant growth regulator. Excessive intake of forchlorfenuron can lead to metabolic disorders of the matrix and be harmful to human health. The chemiluminescence intensity of the KIO 4 –K 2 CO 3 –Mn 2+ reaction decreased in the presence of forchlorfenuron. Based on this result, a rapid and sensitive chemiluminescence method was established to determine forchlorfenuron by combining it with a batch injection static device. The injection speed, injection volume and reagent concentration of the forchlorfenuron–KIO 4 –K 2 CO 3 –Mn 2+ chemiluminescence reaction were optimized. Under these optimized conditions, the linear range of the method was 1.0–200.0 μg/L, and the limit of detection was 0.29 μg/L (S/N = 3). The chemiluminescence method for the determination of forchlorfenuron could be completed in 10 s. The method was applied to detect the residual forchlorfenuron in dried fruit samples, and the results are consistent with high-performance liquid chromatography-mass spectrometry. This method has the advantages of high sensitivity, rapid response, less reagent consumption, and convenient operation. It will provide a new perspective for chemiluminescence for the rapid and sensitive determination of forchlorfenuron in various complex samples.

Ce6/Mn2+-chelated polydopamine@black-TiO2 nanoprobes for enhanced synergistic phototherapy and magnetic resonance imaging in 4T1 breast cancer

Yang Gao,Luyun Zhang,Yanhong Liu,Sijia Sun,Zhibin Yin,Lili Zhang,Aiguo Li,Guangming Lu,Aiguo Wu,Leyong Zeng

Black titanium dioxide (TiO2) nanoparticles have attracted great attention due to their application in photothermal therapy (PTT). However, single-mode phototherapy has the risk of recurrence, and the high-dose laser usually imposed to improve the PTT performance can bring a potential threat to security. Here, polydopamine (PDA)-coated black TiO2 (b-P25@PDA) nanoparticles with a core–shell structure were synthesized for enhanced PTT; then, synergistic phototherapy nanoprobes (b-P25@PDA-Ce6 (Mn)) were constructed by coupling chlorin e6 (Ce6) and chelating Mn2+ for simultaneous photodynamic therapy (PDT)/PTT and magnetic resonance (MR) imaging, in which a low-dose laser was used and imaging-guided phototherapy with high efficiency and high safety was achieved. The prepared nanoprobes showed high photothermal conversion efficiency (32.12%), high reactive oxygen generation and excellent MR imaging. In the 4T1 tumor-bearing nude mouse model, the tumors completely disappeared under the combination of PDT/PTT with a low-dose laser but were only partially inhibited by single PDT and single PTT. The current work developed a multifunctional black TiO2-based nanoprobe for enhanced synergistic PDT/PTT and MR imaging, which will be important for the safe and efficient visualized theranostics of cancers.

Revisiting the Enhanced Red Upconversion Emission from a Single β-NaYF4:Yb/Er Microcrystal By Doping with Mn2+ Ions

Yuan Maohui,Wang Rui,Zhang Chaofan,Yang Zining,Yang Xu,Han Kai,Ye Jingfeng,Wang Hongyan,Xu Xiaojun

The presence of manganese ions (Mn 2+ ) in Yb/Er-co-doped nanomaterials results in suppressing green (545 nm) and enhancing red (650 nm) upconversion (UC) emission, which can achieve single-red-band emission to enable applications in bioimaging and drug delivery. Here, we revisit the tunable multicolor UC emission in a single Mn 2+ -doped β-NaYF 4 :Yb/Er microcrystal which is synthesized by a simple one-pot hydrothermal method. Excited by a 980 nm continuous wave (CW) laser, the color of the single β-NaYF 4 :Yb/Er/Mn microrod can be tuned from green to red as the doping Mn 2+ ions increase from 0 to 30 mol%. Notably, under a relatively high excitation intensity, a newly emerged emission band at 560 nm ( 2 H 9/2  →  4 I 13/2 ) becomes significant and further exceeds the traditional green (545 nm) emission. Therefore, the red-to-green (R/G) emission intensity ratio is subdivided into traditional (650 to 545 nm) and new (650 to 560 nm) R/G ones. As the doped Mn 2+ ions increase, these two R/G ratios are in lockstep with the same tunable trends at low excitation intensity, but the tunable regions become different at high excitation intensity. Moreover, we demonstrate that the energy transfer (ET) between Mn 2+ and Er 3+ contributes to the adjustment of R/G ratio and leads to tunable multicolor of the single microrod. The spectroscopic properties and tunable color from the single microrod can be potentially utilized in color display and micro-optoelectronic devices.

Tunable dual emission in Mn2+-doped CsPbX3 (X = Cl, Br) quantum dots for high efficiency white light-emitting diodes

Weiwei Chen,Xiaosheng Tang,Zhigang Zang,Yue Shi,Zhaoqi Yang,Juan Du

Doping of Mn2+ into semiconductor nanocrystals has been demonstrated to endow them with novel electronic, optical and magnetic functionalities. In this paper, Mn-doped CsPbX3 (X = Br, Cl) quantum dots (QDs) were synthesized at room temperature via a facile strategy by introducing dimethyl sulfoxide (DMSO)-MnBr2/PbX2 composite as a precursor. The excitonic emission spectra of the as-obtained Mn-doped CsPbX3 QDs can be tuned from 517 nm to 418 nm by adjusting the ratio of PbBr2/PbCl2 precisely, and the luminescence mechanism of the doped QDs is discussed in detail. Moreover, the highest photoluminescence quantum yield of the Mn2+ emission achieves 36.7%, which is comparable with QDs prepared by the conventional hot-injection method. Depending on the ratios of PbPb2/PbCl2, the energy transfer rate from the band-edge to Mn2+ excited state is in the range of 0.006–20.42 × 107 s−1. Furthermore, white light-emitting diodes (LEDs) were successfully fabricated by combining the as-prepared Mn-doped CsPbX3 QDs with commercial UV GaN chips, and the high luminous efficiency of the as-prepared white LEDs was developed to 55.9 lm W−1. This work strongly supports the fact that Mn-doped CsPbX3 QDs are promising materials for application in lighting and displaying fields.

Zhihua Wang,Yafang Zhang,Bing Zhang,Xiaoquan Lu

A newly fluorescence probe for detection of Serotonin based on a Mn-doped ZnS quantum dots@silica nanoparticle@molecularly imprinted polymers (QDs@SiO 2 @MIPs) was successfully created. QDs@SiO 2 @MIPs were characterized by scanning electron microscope, transmission electron microscope, Fourier transform infrared spectroscopy, UV spectroscopy , X-ray powder diffraction, and fluorescence spectroscopy. A complex was produced between the amino group of QDs@SiO 2 @MIPs and the hydroxyl group of 5-HT when 5-HT rebinding, the energy of the QDs would be transferred to the complex, which led to its fluorescence quenching . The composite material has a high selectivity with an imprinting factor of 5.96. The linear regression equation is F 0 /F = 0.0036Cq-0.0512 , with a correlation coefficient of 0.9928 in a liner range from 50 to 500 ng/mL. The limit of detection(LOD) was 0.69 ng/mL. The probe exhibited simple preparation process, high sensitivity, good selectivity, low detection limit, short analysis time and could be applied to detect 5-HT in human serum successfully.

Dual-modality immune nano-activator harnessing Mn2⁺ and quercetin to potentiate the cGAS-STING pathway for advanced cancer metalloimmunotherapy

Ma Shanshan,Zhang Xuequan,Zhu Xiaoqi,Yan Kangning,Wang Qin,Lei Lei,Li Jiasheng,Guo Jing,Tang Weizhong,Liu Junjie,Cao Jun,Wang Duo,Luo Tao

Manganese ions (Mn 2+ ) have emerged as promising activators of the cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway. However, their clinical application was hindered by low bioavailability and limited immune activation pathways, which impaired their ability to trigger robust immune responses and achieve significant antitumor effects. To address these challenges, we developed a dual-modality immune nano-activator by coordinating manganese ions with quercetin. This strategy was designed to enhance the cGAS-STING pathway activation and elicit the immunogenic cell death, thereby strengthening the antitumor immune response. The engineered nano-activator demonstrated superior tumor-targeting ability and efficient cellular internalization. Upon exposure to near-infrared irradiation, the system harnessed photothermal effects to induce apoptosis in tumor cells while simultaneously accelerating the release of manganese ions and quercetin. The released manganese ions facilitated the generation of reactive oxygen species, which in conjunction with quercetin-induced apoptosis, amplified photothermal-induced DNA damage. This DNA damage further promoted the release of cytosolic DNA, which in turn activated the cGAS-STING pathway, thereby intensifying immune activation. Notably, the nano-activator also triggered immunogenic cell death, which synergized with the cGAS-STING activation to promote dendritic cell maturation and activate antigen-specific T-cell, significantly enhancing the immune response against the tumor. Both in vitro and in vivo studies confirmed that this nano-activator effectively inhibited tumor growth, with particularly pronounced effects when combined with anti-CTLA-4 antibodies. Fabrication of CsPb1−xMnxBr3−2xCl2x (x = 0–0.5) quantum dots for near UV photodetector application

Ba-Duc Tran,Phuong-Nam Tran,Xuan-Thanh Bui,Duy-Hung Nguyen,Thanh-Tung Duong

In this study, CsPb 1– x Mn x Br 3–2 x Cl 2 x ( x = 0–0.5) nanoparticles were synthesized directly in toluene solvents at high temperature. This approach results in small-size nanoparticles, which can be used in photoelectric components without adding a filtration process to eliminate high-temperature solvents such as octadecene. The high content Mn 2+ -incorporated CsPb(Cl/Br) 3 host is observed by strongly wideband emission at 592 nm of manganese d-states spin and orbital forbidden transition. After infiltration into mesoporous TiO 2 , the Mn 2+ ion receives energy from the CsPb(Cl/Br) 3 host (irradiated 405 nm light-emitting diode source) and effectively transfers to the TiO 2 scaffold layer. As a result, a metal/semiconductor/metal planar structure photodetector with m-TiO 2 /CsPb 1– x Mn x Br 3–2 x Cl 2 x ( x = 0–0.5) composite showed several figures of merit compared to bare m-TiO 2 and m-TiO 2 /CsPbBr 3 such as on/off ratio of 10 4 times, responsivity of 1.67 A·W −1 , and detectivity of 4.42 × 10 9 Jones. The key factors contributing to the growth of the on/off ratio include the decreasing dark current and enhancing exciton energy and transportation due to the infiltration of CsPb 1– x Mn x Br 3–2 x Cl 2 x ( x = 0–0.5) perovskite quantum dots into mesopores of the m-TiO 2 scaffold layer.
