Mesenchymal stem cells (MSCs) have favourable outcomes in the treatment of kidney diseases. Pre-B-cell leukaemia transcription factor 1 (PBX1) has been reported to be a regulator of self-renewal of stem cells. Whether PBX1 is beneficial to MSCs in the treatment of haemorrhagic shock (HS)-induced kidney damage is unknown. We overexpressed PBX1 in rat bone marrow-derived mesenchymal stem cells (rBMSCs) and human bone marrow-derived mesenchymal stem cells (hBMSCs) to treat rats with HS and hypoxia-treated human proximal tubule epithelial cells (HK-2), respectively. The results indicated that PBX1 enhanced the homing capacity of rBMSCs to kidney tissues and that treatment with rBMSCs overexpressing PBX1 improved the indicators of kidney function, alleviated structural damage to kidney tissues. Furthermore, administration with rBMSCs overexpressing PBX1 inhibited HS-induced NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome activation and the release of proinflammatory cytokines, and further attenuated apoptosis. We then determined whether NF-κB, an important factor in NLRP3 activation and the regulation of inflammation, participates in HS-induced kidney damage, and we found that rBMSCs overexpressing PBX1 inhibited NF-κB activation by decreasing the p-IκBα/IκBα and p-p65/p65 ratios and inhibiting the nuclear translocation and decreasing the DNA-binding capacity of NF-κB. hBMSCs overexpressing PBX1 also exhibited protective effects on HK-2 cells exposed to hypoxia, as shown by the increase in cell viability, the mitigation of apoptosis, the decrease in inflammation, and the inhibition of NF-κB and NLRP3 inflammasome activation. Our study demonstrates that MSCs overexpressing PBX1 ameliorates HS-induced kidney damage by inhibiting NF-κB pathway-mediated NLRP3 inflammasome activation and the inflammatory response.

Osteoarthritis (OA), the most common joint disease, is characterized by inflammation and cartilage degradation. Previous studies illustrated that Smad nuclear-interacting protein 1 (SNIP1) is an inhibitor of the TGF-β signal transduction pathway and SNIP1 has been reported as an anti-inflammatory factor. This study aimed to explore the role of SNIP1 in OA progression. In this study, the SNIP1 expression was evaluated in OA human and OA mice tissue and interleukin-1 beta (IL-1β)-induced chondrocytes . The Safranin-O (SO) staining and osteoarthritis research society international (OARSI) scoring system was used to evaluate cartilage injury. The gain- and loss-of-function studies for SNIP1 were performed in chondrocytes. The SNIP1 overexpression adenovirus was injected into mice by intra-articular injection. The SNIP1 expression was decreased in OA patients, OA mice, and IL-1β-stimulated chondrocytes. The cartilage injury of medial meniscus-induced OA (DMM-OA) mice at 8 weeks showed more severe than that at 4 weeks. The expression of SNIP1 was lower at 8 weeks than that at 4 weeks. In IL-1β-stimulated chondrocytes, SNIP1 overexpression reduced the expression of TNF-α and IL-6, alleviated ECM degradation, reduced the phosphorylation levels of p65 and IκBα , and decreased the p65 level in nuclear. Moreover, overexpression of SNIP1 alleviated cartilage injury in DMM-OA mice. In brief, our study suggested that SNIP1 alleviated OA and repressed inflammation by inhibiting the activation of NF-κB. This study might provide a new insight into OA treatment.

Bergenin (BG) is a polyphenolic substance which has therapeutic potential in the treatment of diabetic nephropathy (DN), a common complication of type II diabetes. However, the mechanisms underlying these effects remain unclear. We studied the protective effects and mechanisms of BG in DN mice, focusing on the TLR4/MyD88/NF-κB signalling pathway. C57BL/6J mice were used as experiments (n=60), and 10 animals were randomly selected as normal control. The DN model was developed by administering an intraperitoneal injection of streptozotocin (40 mg/kg BW for three days) and a high-fat diet (n=50). BG (20, 40, and 80 mg/kg BW, once a day) was administered orally for four weeks. After BG treatment, the food and water intake of DN mice decreased, blood glucose levels decreased, and insulin resistance reduced. As a result, serum LDL-C, TC, and TG levels decreased; HDL-C levels increased; SOD, CAT, and GSH-Px levels decreased; and MDA levels increased. BG administration reduced AST, ALT, BUN, and CRE levels and inflammatory factors (including TNF-α, MCP-1, IL-1β, and IL-6). Histopathology revealed a significant improvement in pathological damage to the liver, kidney, and spleen of mice treated with BG, and TLR4, MyD88, and NF-κB p65 were down-regulated at both mRNA and protein levels in the BG-treated group. Based on these results, BG therapeutic type II DN by hypoglycaemia, improving liver and kidney function, and anti-oxidative stress; reducing inflammation; and inhibiting the TLR4/MyD88/NF-κB signalling pathway. The results of this study suggest that BG can be used as an effective treatment for type II DN.

Exploring low-cost and high-performance oxygen evolution reaction (OER) catalysts has attracted great attention due to their crucial role in water splitting. Here, a bifunctional Cu5FeS4/Ni3S2@NF catalyst was in situ formed on a nickel (Ni) foam toward efficient photoassisted electrocatalytic (P-EC) OER, which displays an ultralow overpotential of 260 mV at 30 mA cm–2 in alkaline solution, outperforming most previously reported Ni-based catalysts. It also shows great potential in degradation of antibiotics as an alternative anode reaction to OER owing to the prompt transfer of photogenerated holes. The photocurrent test and transient photovoltage spectroscopy indicate that the synergistic coupling of charge extraction and sinking effects in Cu5FeS4 and Ni3S2 is critical for boosting the OER activity via photoassistance. Electrochemical active surface area and electrochemical impedance spectroscopy tests further prove that the photogenerated electromotive force can effectively compensate the overpotential of OER. This work not only provides a good guidance for integrating photocatalysis and electrocatalysis, but also indicates the key role of synergistic extraction and utilization of photogenerated charge carriers in P-EC.

A nanolipidcarrier (NLC) loaded homogalacturonan enriched pectin (citrus modified pectin, MCP4) hydrogel was designed as a novel colon inflammation site-specific oral delivery system for 6-gingerol (6G) (6G-NLC/MCP4 hydrogel) administration, and its colitis alleviation effect were investigated. 6G-NLC/MCP4 exhibited typical “cage-like” ultrastructure with 6G-NLC embedded in the hydrogel matrix as observed by cryoscanning electron microscope. And due to the homogalacturonan (HG) domain in MCP4 specifically combined with Galectin-3, which is overexpressed in the inflammatory region, the 6G-NLC/MCP4 hydrogel targeted to severe inflammatory region. Meanwhile, the prolonged-release characteristics of 6G-NLC provided sustained release of 6G in severe inflammatory regions. The matrix of hydrogel MCP4 and 6G achieved synergistic alleviation effects for colitis through NF-κB/NLRP3 axis. Specifically, 6G mainly regulated the NF-κB inflammatory pathway and inhibited the activity of NLRP3 protein, while MCP4 regulated the expression of Galectin-3 and peripheral clock gene Rev-Erbα/β to prevent the activation of inflammasome NLRP3.

Background Gastric cancer (GC) is a common cancer with a high incidence and mortality rate. Herein, the role of hsa_circ_0002019 (circ_0002019) in GC was investigated. Methods The molecular structure and stability of circ_0002019 were identified by RNase R, and Actinomycin D treatment. Molecular associations were verified by RIP. Proliferation, migration, and invasion were detected by CCK-8, EdU, and Transwell, respectively. The effect of circ_0002019 on tumor growth was analyzed in vivo. Results Circ_0002019 was elevated in GC tissues and cells. Circ_0002019 knockdown inhibited the proliferation, migration, and invasion. Mechanically, circ_0002019 activated NF-κB signaling by increasing TNFAIP6 mRNA stability by PTBP1. Activation of NF-κB signaling limited the antitumor effect of circ_0002019 silencing in GC. Circ_0002019 knockdown inhibited tumor growth in vivo by reducing TNFAIP6 expression. Conclusions Circ_0002019 accelerated the proliferation, migration, and invasion by regulating TNFAIP6/NF-κB pathway, suggesting circ_0002019 could be a key regulatory factor in GC progression.

The synthesis of furan-based platform chemicals from abundant and renewable biomass-based hexoses plays an important role in the development and utilization of biomass energy. The electrochemical 5-hydroxymethylfurfural oxidation reaction (HMFOR) represents a promising route for synthesizing the 2,5-furandicarboxylic acid (FDCA) product which is a high value-added biomass-based monomer. Interface engineering is an effective strategy to adjust the electronic structure, optimize the adsorption of intermediates, and expose more active sites, thus attracting extensive attention for designing efficient HMFOR electrocatalysts. Herein, a NiO/CeO2@NF heterostructure with an abundant interface is designed for boosting the HMFOR performance under alkaline conditions. At 1.475 V vs. RHE, the conversion of HMF is nearly 100%, the selectivity of FDCA is 99.0%, and the faradaic efficiency is as high as 98.96%. The NiO/CeO2@NF electrocatalyst also exhibits robust stability for HMFOR for 10 cycles. When coupled with the cathode hydrogen evolution reaction (HER) in alkaline medium, the yields of FDCA and hydrogen production are 197.92 and 600 μmol cm−2 h−1, respectively. The NiO/CeO2@NF catalyst is also suitable for the electrocatalytic oxidation of other biomass-derived platform compounds. The abundant interface between NiO and CeO2, which can regulate the electronic properties of Ce and Ni atoms, improve the oxidation state of Ni species, regulate intermediate adsorption, and promote electron/charge transfer, makes the most contribution to high HMFOR performance. This work will provide a simple route for the design of heterostructured materials and reveal the application prospect of interface engineering for promoting the upgrading of biomass derivatives.

Reverse osmosis is an effective seawater desalination way extending the available sources of fresh water, but challenged by the gypsum scaling. Herein, D (+)-glucosamine (DGA) was rationally selected and utilized to prepare a novel polyesteramide (PEA) NF membrane that can reject SO 4 2− but release Ca 2+ . Then it was employed in the pretreatment of seawater reverse osmosis to spatially isolate SO 4 2− and Ca 2+ , thus removing the “foundation” for forming gypsum. Compared to the referenced NF270, the optimized sample denoted as DGA-PEA-NF has a looser structure with stronger negative charge, as well as a higher resistance to both organic fouling and chlorine attack. During the pretreatment of seawater, both the water permeability and SO4 2− /Ca 2+ selectivity of the DGA-PEA-NF membrane were much higher than those of NF270; the seawater permeance and SO 4 2− /Ca 2+ selectivity of DGA-PEA-NF were 10.25 L m −2 ·h −1 ·bar −1 and 7.62, respectively, while those of NF270 were only 5.11 L m −2 ·h −1 ·bar −1 and 2.08. Consequently, the DGA-PEA-NF membrane effectively reduced the gypsum scaling both in NF and RO stages rather than just transferring the fouling problem forwards to NF stage. The implementation of this research is expected to provide guidance for a targeted fabrication of NF membranes, as well as support for the sustainable development of seawater desalination.

Currently, most nanofiltration membranes exhibit satisfactory separation performance on the anions or cations, and few membranes could concurrently separate anion and cation . Therefore, preparing nanofiltration membranes with excellent separation performance for both anionic and cationic ions remains a challenge. Herein, we have developed a series of nanofiltration membranes using conventional interfacial polymerization with different ratios of trimesoyl chloride (TMC) and piperazine (PIP) concentrations. In the interface polymerization process, the amine groups carried by PIP imparted a positive charge, while the hydrolysis of acyl chloride groups resulted in the generation of carboxylic acid groups that introduced a negative charge. The mix-charged nanofiltration membrane could be fabricated by varying PIP/TMC concentration ratios. The optimized membrane exhibited high rejection rates for bivalent ions (Na 2 SO 4 rejection of 96.6 %, MgCl 2 rejection of 93.0 %), and its selectivity for Mg 2+ /Li + mixed solution was improved significantly. This work provides a novel approach for the development of nanofiltration membranes capable of effectively separating mono-/divalent ions.

Hemorrhagic shock (HS) is defined as a reduction in tissue oxygenation and organ dysfunction due to severe blood loss. Lung injury is a frequent complication of HS. Baicalin, isolated from Radix Scutellariae , has been reported to profile the antitumor, anti-oxidative, anti-inflammatory, and antibacterial roles in various pathological processes. Nevertheless, the effects of baicalin on HS-induced lung injury are unclear. This study aims to examine the therapeutic effects of baicalin on lung injury. We first established the lung injury rat models by withdrawing blood in the femoral artery followed by resuscitation. A pathological analysis showed that HS-administrated rats presented severe capillary leakage and pulmonary edema, while baicalin therapy alleviated the symptoms. Baicalin therapy reduced the number of macrophages and neutrophils in bronchoalveolar lavage fluid and decreased the expression and activity of myeloperoxidase (neutrophile infiltration marker) in the lung tissues of HS rats, indicating that baicalin alleviated HS-induced infiltration of inflammatory cells. The secretion of inflammatory cytokines, including interleukin (IL)-1 β , IL-6, IL-18, and tumor necrosis factor α (TNF- α ), as well as the activation of the nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing-3 (NLRP3) inflammasome, were inhibited by baicalin administration. Furthermore, we found that the NF- κ B pathway, a canonical pro-inflammatory pathway, was also blocked after treatment with baicalin in HS-evoked rats, as indicated by the decreased expression of p65 and p65 phosphorylation in the lung tissues. In summary, we infer that baicalin may exert a protective role in HS-induced lung injury by suppressing the activation of NLRP3 inflammasome via the NF- κ B pathway.

The investigation of earth-abundant, durable, and efficient bifunctional electrocatalysts for overall water splitting was critical to future renewable energy systems. Herein, flower-like Mn-doped Cu 7.2 S 4 @NiS 2 @NiS was successfully assembled on 3D nickel foam (NF) substrate via a facile two-step hydrothermal‑sulfuration method (denote as flower-like Mn 2 -CNN/NF). Benefited from unique 3D configuration with firm contact between nanoflowers and Ni foam, flower-like Mn 2 -CNN/NF electrode demonstrated a low overpotential of 252 mV to achieve a current density of 50 mA cm −2 and 24 h-long stability toward oxygen evolution reaction (OER) in 1 M KOH. For hydrogen evolution reaction (HER), a stable current density of 10 mA cm −2 at only an overpotential of 87 mV and a low Tafel slope of 43.7 mV dec −1 were observed. Furthermore, when assembled as electrolyzers on both cathode and anode for stable water splitting, a low cell voltage of 1.514 V was needed for flower-like Mn 2 -CNN/NF to afford the current density of 10 mA cm −2 and distinguished durability for over 46 h was observed, which presented a wide range of possibilities for the development of earth-abundant and high efficient electrode pairs.

Hepatocellular carcinoma (HCC) has become a global public health problem. Therefore, the development of novel and effective therapeutic agents for the treatment of HCC is considered an emergency. Avicularin, a bio‑active flavonoid from plants, has been reported to exhibit diverse pharmacological properties. The aim of the present study was to investigate the role of avicularin in HCC and the underlying mechanism of action. Huh7 cells were treated with avicularin in a concentration‑dependent manner, and the cell proliferation was examined using a 3‑(4, 5‑dimethylthiazol‑2‑yl)‑2, 5‑diphenyltetrazolium bromide assay kit. The cell migration and invasion abilities were detected using wounding‑healing assays and Transwell assays. Flow cytometric analysis was performed to investigate the cell cycle distribution and cell apoptosis. The activity of nuclear factor (NF)‑κB (p65), cyclooxygenase‑2 (COX‑2) and peroxisome proliferator‑activated receptor γ (PPAR‑γ) were measured by reverse transcription‑quantitative polymerase chain reaction and western blot analyses, respectively. The results indicated that avicularin treatment markedly decreased cell proliferation concentration‑dependently in HCC, and inhibited cell migration and invasion in Huh7 cells. It was also found that the treatment of avicularin markedly inhibited the G0/G1‑phase cells and decreased the accumulation of S‑phase cells in the cell cycle and induced cell apoptosis. In addition, it was confirmed that the anticancer efficacy of avicularin in HCC was dependent on the regulation of NF‑κB (p65), COX‑2 and PPAR‑γ activities. In conclusion, the findings suggested that avicularin serves an antineoplastic role in HCC and may provide a potential therapeutic strategy for the treatment of HCC.
