Pyrrole-Doped Polydopamine-Pyrrole (PDA-nPY) Nanoparticles with Tunable Size and Improved NIR Absorption for Photothermal Therapy

Yuan He,Ziyang Li,Huiling Su,Yanan Sun,Wei Shi,Yunfeng Yi,Dongtao Ge,Zhongxiong Fan

Polydopamine (PDA) as a melanin-like biomimetic material with excellent biocompatibility, full spectrum light absorption capacity and antioxidation property has been extensively applied in the biomedical field. Based on the high reactivity of dopamine (DA), exploiting new strategies to fabricate novel PDA-based nano-biomaterials with controllable size and improved performance is valuable and desirable. Herein, we reported a facile way to synthesize pyrrole-doped polydopamine-pyrrole nanoparticles (PDA-nPY NPs) with tunable size and enhanced near-infrared (NIR) absorption capacity through self-oxidative polymerization of DA with PY in an alkaline ethanol/H2O/NH4OH solution. The PDA-nPY NPs maintain excellent biocompatibility and surface reactivity as PDA. By regulating the volume of added PY, PDA-150PY NPs with a smaller size (<100 nm) and four-fold higher absorption intensity at 808 nm than that of PDA can be successfully fabricated. In vitro and in vivo experiments effectively further demonstrate that PDA-150PY NPs can effectively inhibit tumor growth and completely thermally ablate a tumor. It is believed that these PY doped PDA-nPY NPs can be a potential photothermal (PT) agent in biomedical application.Keywords:polydopamine;pyrrole;self-oxidative polymerization;photothermal treatment;cancer therapy

A label-free fluorescent sensor for rapid and sensitive detection of ctDNA based on fluorescent PDA nanoparticles

Xiao Yang,Yang Huang,Siyi Yang,Miao Tang,Juan Liu,Jinhui Shen,Huanbao Fa,Danqun Huo,Changjun Hou,Mei Yang

Technological advances in the detection of circulating tumor DNA (ctDNA) have made new options available for diagnosis, classification, biological studies, and treatment selection. However, effective and practical methods for analyzing this emerging class of biomarkers are still lacking. In this work, a fluorescent biosensor was designed for the label-free detection of ctDNA (EGFR 19 del for non-small cell lung cancer, NSCLC). The biosensor was based on the fact that MnO2 nanosheets (MnO2 NSs) have stronger affinity towards single-stranded DNA (ssDNA), as compared with double-stranded DNA (dsDNA). As a high-performance nanoenzyme, MnO2 NSs could oxidize dopamine (DA) into fluorescent polydopamine nanoparticles (FL-PDA NPs), which could be used as a fluorescence signal. The probe ssDNA could be adsorbed on the surface of MnO2 NSs through π–π stacking, and the active site would be masked, causing a lower fluorescence. After the targets were recognized by probe ssDNA to form dsDNA, its affinity for MnO2 NSs decreased and the active site recovered, causing a restored fluorescence. It was verified that Mn ions, •OH radicals and electron transfer were the important factors in the catalytic oxidation of DA. Under the optimal experimental conditions, this biosensor exhibited a detection limit of 380 pM and a linear range of 25–125 nM, providing reliable readout in a short time (45 min). This sensor exhibited outstanding specificity, stability and reproducibility. In addition, this sensor was applied to the detection of ctDNA in serum samples and cell lysates. It is demonstrated that FL-PDA NPs can be used as a fluorescence signal for easy, rapid and label-free detection of ctDNA without any other amplification strategies, and the proposed strategy has great potential for biomarker detection in the field of liquid biopsy.

Preparation of AuNP-CQD/PDA/GO anode for MFC and its treatment of oily sewage from ships

Wang Tianshu,Shi Peibo,Wang Mingyu,Zhang Shaojun

Oily sewage discharged from ships has brought many harms to the marine environment, even endangered marine life and human life. As a new type of water treatment technology, microbial fuel cell (MFC) can efficiently treat pollutants and recover energy, which can be converted into electric energy. However, its large internal resistance restricts its development. In order to solve the problems of low power generation performance and poor biocompatibility of microbial fuel cell, a gold nanoparticle-carbon quantum dot/polydopamine/graphene oxide/bacterial cellulose (AuNP-CQD/PDA/GO/BC) electrode was prepared, and it was applied to the treatment of oily sewage from ships. Fourier transforms infrared spectroscopy, X-ray diffraction, scanning electron microscopy, gas chromatography-mass spectrometry, and contact angle measuring instrument were used to characterize the electrode. The results show that PDA bridges GO and AuNP-CQD particles through the electrostatic interaction/π-π bond/hydrogen bonding, respectively. This attracts a large number of microorganisms to attach to the surface of the porous anode material, which greatly improves the activity and quantity of microorganisms. Moreover, the maximum power density of AuNP-CQD/PDA/GO/BC electrode is 2624.91 mW/m 2 , which obviously improves the electrochemical performance of MFC. The oil content of the treated water is ≤ 15 mg/L, reaching the discharge of MARPOL 73/78 convention. Therefore, the proposed approach has paved new dimensions in not only the preparation of a new composite electrode materials but also its applications as effective degradation of ship oily sewage in MFC.

Fabricating a PDA-Liposome Dual-Film Coated Hollow Mesoporous Silica Nanoplatform for Chemo-Photothermal Synergistic Antitumor Therapy

Chuanyong Fan,Xiyu Wang,Yuwen Wang,Ziyue Xi,Yuxin Wang,Shuang Zhu,Miao Wang,Lu Xu

In this study, we synthesized hollow mesoporous silica nanoparticles (HMSNs) coated with polydopamine (PDA) and a D-α-tocopheryl polyethylene glycol 1000 succinate (TPGS)-modified hybrid lipid membrane (denoted as HMSNs-PDA@liposome-TPGS) to load doxorubicin (DOX), which achieved the integration of chemotherapy and photothermal therapy (PTT). Dynamic light scattering (DLS), transmission electron microscopy (TEM), N2adsorption/desorption, Fourier transform infrared spectrometry (FT-IR), and small-angle X-ray scattering (SAXS) were used to show the successful fabrication of the nanocarrier. Simultaneously, in vitro drug release experiments showed the pH/NIR-laser-triggered DOX release profiles, which could enhance the synergistic therapeutic anticancer effect. Hemolysis tests, non-specific protein adsorption tests, and in vivo pharmacokinetics studies exhibited that the HMSNs-PDA@liposome-TPGS had a prolonged blood circulation time and greater hemocompatibility compared with HMSNs-PDA. Cellular uptake experiments demonstrated that HMSNs-PDA@liposome-TPGS had a high cellular uptake efficiency. In vitro and in vivo antitumor efficiency evaluations showed that the HMSNs-PDA@liposome-TPGS + NIR group had a desirable inhibitory activity on tumor growth. In conclusion, HMSNs-PDA@liposome-TPGS successfully achieved the synergistic combination of chemotherapy and photothermal therapy, and is expected to become one of the candidates for the combination of photothermal therapy and chemotherapy antitumor strategies.Keywords:chemo-photothermal therapy;hollow mesoporous silica;liposome-TPGS;polydopamine;doxorubicin

Cavitation regulated sonochemical synthesis of flexible self-supported CuO@PDA/CC electrode for highly sensitive glucose sensor

Bo Shao,Fang Chen,Jianyuan Wang,Wei Zhai

Sonochemical synthesis is recognized as a promising high-efficiency method for fabricating transition metal oxides (TMOs) based flexible self-supported electrodes as nonenzymatic glucose sensor. However, it is still a challenge to obtain TMOs self-supported electrodes with well-controlled TMOs nanostructure and desirable interfacial properties via a sonochemical approach because of the uncontrollability of transient cavitation. In this study, CuO@polydopamine (PDA) nanoparticles (NPs) are in situ grown onto carbon cloth (CC) to fabricate CuO@PDA/CC self-supported electrode via a sonochemical process. The sonochemically synthesized PDA not only stabilized the transient cavitation intensity, but also regulated the growth of CuO due to the strong chelating ability between Cu 2+ and catechol groups from PDA. It is determined that the mean size of CuO@PDA NPs on the CuO@PDA/CC electrode is less than 100 nm with a narrow size distribution. Attributed to high electrochemical active surface area (4.61 mF cm −2 ) and low interface resistance (2.35 Ω), CuO@PDA/CC electrode displays excellent electrochemical performance as a glucose sensor with high sensitivity of 1.843 μA cm −2 mM −1 in a wide linear range up to 4.985 mM and excellent stability. This work provides a facile approach to fabricating the flexible self-supported electrodes with well-controlled nanostructure and desirable interfacial properties for high-performance electrochemical sensors via regulating the transient cavitation intensity in the sonochemical process.

Constructing a self-healing injectable SABA/Borax/PDA@AgNPs hydrogel for synergistic low-temperature photothermal antibacterial therapy

Hao Zhu,Xuedan Cheng,Junqing Zhang,Qiang Wu,Chaoqun Liu,Jiahua Shi

Infections caused by bacteria are one of the biggest challenges humans face around the world. Photothermal therapy (PTT) has been regarded as a promising strategy in combating pathogenic infection, however the high temperatures (55–65 °C) required during a single PTT process can induce injury to healthy tissues nearby. Combination therapy could overcome this problem by reducing the photothermal temperature. Here, we developed a self-healing and injectable hydrogel to realize low-temperature PTT (LT-PTT, ≤45 °C) for antisepsis with high-efficiency. The hybrid hydrogel is prepared by incorporating borax into a mixture of 3-aminophenylboronic acid grafted sodium alginate and nano-silver decorated polydopamine nanoparticles. Our results showed that the SABA/Borax/PDA@AgNPs hydrogel possesses satisfactory mechanical properties and self-healing capacity, and as a result, it can repair itself after being damaged mechanically, retaining its integrality and recovering its initial functionalities. Furthermore, through utilizing the photothermal property of polydopamine nanoparticles and broad-spectrum antibacterial activity of nano-silver, the hybrid hydrogel achieves excellent LT-PTT for sterilization both in vitro as well as in an in vivo mice skin wound model with no distinct injury to normal tissues. Overall, our prepared hydrogel is expected to be an excellent candidate for treating bacterial infections.

Facile and simple fabrication of superhydrophobic and superoleophilic MS/PDA/DT sponge for efficient oil/water separation

Juxiang Yang,Yuan Jia,Beibei Li,Jiao Jiao

To overcome the problems of frequent leakage accidents during oil exploitation, a superhydrophobic and superoleophilic porous MS/PDA/DT sponge was successfully prepared via mild solvent evaporation method, and a polydopamine assisted surface coating of 1-dodecanethiol (DT) on a melamine sponge (MS) substrate. Surface structure and performance of the MS/PDA/DT sponge were characterized by Scanning Electron Microscope (SEM), Fourier transform infrared spectrometer (FTIR), and Video Optical Contact Angle (CA) metre. The results showed that the as-prepared MS/PDA/DT sponge has a high-water contact angle (WCA) of 147.2°, which is probably attributed to both the rough surface derived from in situ growth and the low surface energy due to grafting of hydrophobic 1-dodecanethiol. The durability of the as-constructed MS/PDA/DT sponge was studied by repeated abrasion tests. After 50 abrasion cycles, the superhydrophobicity of the MS/PDA/DT sponge good mechanical durability. The MS/PDA/DT sponge can effectively absorb oil with an absorption capacity of up to 24 times its weight. The superhydrophobic and superoleophilic MS/PDA/DT sponge has the potential as a promising adsorbent for oil/water separation.Highlights The MS/PDA/DT sponge was prepared via the mild solvent evaporation method. The contact angle of the MS/PDA/DT sponge was 147.2o.The adsorption capacity of the MS/PDA/DT sponge was 24 times their weight. The cost-efficient, environmentally friendly porous materials show high oil/water separation efficiency.

Jia Zheng,Zhaoxi Zhou,Lihong Zhu,Qihui Chen,Maochun Hong,Heqing Fu

A novel room temperature self-healing carbonyl iron powder/ polydopamine@ multi-walled carbon nanotubes (CIP/PDA@MWCNTs) composites as microwave absorber covering the whole X frequency band were prepared in this study. In order to improve the impedance matching and dispersion of CIP in composites, it was encapsulated by PDA to form a core-shell structure. Meanwhile, the polydopamine modified carbonyl iron powder (CIP/PDA) was mixed with MWCNTs to enhance the performance of microwave absorption , so that the the composites possessed dielectric and magnetic losses. Moreover, the composites have self-healing property. When the content of CIP/PDA@MWCNTs is 20 wt%, the minimum reflection loss of the self-healing composites is −30.69 dB, and the bandwidth of RL less than -10 dB can reach up to 9.44 GHz, covering the entire X. Therefore, this room temperature self-healing CIP/PDA@MWCNTs composite will become an ideal microwave absorbing material for novel function device.

Constructing a High-Density Thermally Conductive Network through Electrospinning–Hot-Pressing of BN@PDA/GO/PVDF Composites

Jiacheng Liang,Jinwei Luo,Jingxian Zhang,Yongqiang Xiong,Shaozao Tan

Polymer matrix composites with high thermal conductivity and elastic mechanical properties are promising materials for diverse practical applications. Herein, BN@PDA/GO/PVDF (PG) composite materials with a highly dense thermally conductive network were prepared by combining electrospinning and hot-pressing methods. Dopamine was used to modify hexagonal boron nitride (h-BN). The π–π interaction between the crystal plane structure of h-BN and the benzene ring in the polydopamine (PDA) structure can increase the surface adhesion of polyvinylidene fluoride (PVDF) fibers, thereby reducing the thermal resistance of the interface between the filler and polymer matrix. The contact per unit area was further improved by utilizing graphene oxide (GO) with a large specific surface area to clad the adhered fibers of BN@PDA. The thermal conductivity of PG60 was shown to be as high as 12.04 W/(m·K) owing to the increased density of the heat conduction path. It realizes the heat transfer from a point-to-point to a three-dimensional interconnected structure, implying that a synergistic effect is achieved. Moreover, when the mass ratio of BN@PDA to PVDF was 0.3 (P30), GO was added on the basis of P30. When the mass ratio of GO to BN@PDA was 1:8, PG30 was obtained. PG30 and P30 achieved tensile strengths of 26.65 and 23.38 MPa, respectively. This study provides an approach for designing a highly thermal conductive film with potential applications for the improvement of heat dissipation in electronic devices.

Development of High-Sensitivity Piezoresistive Sensors Based on Highly Breathable Spacer Fabric with TPU/PPy/PDA Coating

Xiujuan Wang,Xiaoyu Gao,Yu Wang,Xin Niu,Tanyu Wang,Yuanjun Liu,Fangxi Qi,Yaming Jiang,Hao Liu

In recent years, the research of flexible sensors has become a hot topic in the field of wearable technology, attracting the attention of many researchers. However, it is still a difficult challenge to prepare low-cost and high-performance flexible sensors by a simple process. Three-dimensional spacer fabric (SF) are the ideal substrate for flexible pressure sensors due to its good compression resilience and high permeability (5747.7 mm/s, approximately 10 times that of cotton). In this paper, Thermoplastic polyurethane/Polypyrrole/Polydopamine/Space Fabric (TPU/PPy/PDA/SF) composite fabrics were prepared in a simple in-situ polymerization method by sequentially coating polydopamine (PDA) and Polypyrrole (PPy) on the surface of SF, followed by spin-coating of different polymers (thermoplastic polyurethane (TPU), polydimethylsiloxane (PDMS) and Ecoflex) on the PPy/PDA/SF surface. The results showed that the TPU/PPy/PDA/SF pressure sensors prepared by spin-coating TPU at 900 rpm at a concentration of 0.3 mol of pyrrole monomer (py) and a polymerization time of 60 min have optimum sensing performance, a wide working range (0–10 kPa), high sensitivity (97.28 kPa−1), fast response (60 ms), good cycling stability (>500 cycles), and real-time motion monitoring of different parts of the body (e.g., arms and knees). The TPU/PPy/PDA/SF piezoresistive sensor with high sensitivity on a highly permeable spacer fabric base developed in this paper has promising applications in the field of health monitoring. Keywords:space fabric;Polypyrrole (PPy);piezoresistive sensors;thermoplastic polyurethane (TPU) coating

Preparation of black hollow TiO2 nanotube-coated PDA@Ag2S heterostructures for efficient photocatalytic reduction of Cr(VI)

Jun Jiang,Zhidong Chen,Peng Wang,Pei-Yang Gu,Jiadi Liu,Zhuzhu Zhang,Qingfeng Xu

At present, hexavalent chromium (Cr(VI)) has a serious impact on the water environment and human beings since it can be easily absorbed by the human body, lethal to aquatic organisms and carcinogenic to humans. Based on this, a black hollow TiO 2 nanotube supported polydopamine (PDA)@Ag 2 S composite photocatalytic material was prepared, which can be used for the efficient photocatalytic reduction of Cr(VI) in wastewater. Both black hollow TiO 2 and Ag 2 S have the shortcoming of undergoing ready recombination of photogenerated carriers. The introduction of PDA can not only improve the photogeneration of composite photocatalytic materials, but also broaden the absorption range in the visible-light region. A detailed mechanism for the TiO 2-x @PDA@Ag 2 S photocatalytic system has been proposed at the same time. This work provides a new methodology for the design and synthesis of sandwich organic-inorganic hybrid photocatalytic materials.

A GdW10@PDA-CAT Sensitizer with High-Z Effect and Self-Supplied Oxygen for Hypoxic-Tumor Radiotherapy

Lixia Chen,Yang Zhang,Xinming Zhang,Ruijuan Lv,Rongtian Sheng,Ruimeng Sun,Ting Du,Yuhan Li,Yanfei Qi

Anticancer treatment is largely affected by the hypoxic tumor microenvironment (TME), which causes the resistance of the tumor to radiotherapy. Combining radiosensitizer compounds and O2self-enriched moieties is an emerging strategy in hypoxic-tumor treatments. Herein, we engineered GdW10@PDA-CAT (K3Na4H2GdW10O36·2H2O, GdW10, polydopamine, PDA, catalase, CAT) composites as a radiosensitizer for the TME-manipulated enhancement of radiotherapy. In the composites, Gd (Z = 64) and W (Z = 74), as the high Z elements, make X-ray gather in tumor cells, thereby enhancing DNA damage induced by radiation. CAT can convert H2O2to O2and H2O to enhance the X-ray effect under hypoxic TME. CAT and PDA modification enhances the biocompatibility of the composites. Our results showed that GdW10@PDA-CAT composites increased the efficiency of radiotherapy in HT29 cells in culture. This polyoxometalates and O2self-supplement composites provide a promising radiosensitizer for the radiotherapy field.Keywords:radiosensitizer;polyoxometalates;catalase;O2self-supplement;radiotherapy
