BACKGROUND The natural 1,8-dihydroxynaphthalene (DHN) melanin biosynthesis inhibitors (MBIs) are one of the promising approaches to the integrated management of plant diseases but have received scarce attention until now. Herein, to explore the natural DHN MBIs used in the control of northern corn leaf blight (NCLB), a library of 53 essential oil compounds was used to screen the MBIs against Exserohilum turcicum , the causal pathogen of NCLB, using tricyclazole as a reference compound. RESULTS The results of morphological change in the colony, thermogravimetric analysis, ultraviolet–visible spectroscopy, and transmission electron microscopy confirmed that guaiacol could effectively inhibit the melanin production at 50 μg/mL under in vitro conditions. The in vitro bioassay results indicated that this inhibition effect was concentration-dependent and the minimum inhibition concentration of guaiacol was 50 μg/mL. The in vivo experimental results demonstrated that guaiacol significantly inhibited appressorium formation and penetration on corn leaf sheaths at the concentration of 500 μg/mL. The pot experiment results revealed that there were no differences between guaiacol (500 μg/mL) and tricyclazole (100 μg/mL) in control efficacy. The enzymatic assay suggested that guaiacol might exert the activity through inhibiting DHN polymerization to form melanins, which was distinct from tricyclazole. CONCLUSIONS Taken together, this study screened out guaiacol as a natural MBI from 53 essential oil compounds and verified its effectiveness in the control of NCLB at 500 μg/mL. Above all, this research opened an avenue for exploring natural DHN MBIs in the integrated management of plant diseases. © 2022 Society of Chemical Industry.

BACKGROUND S -metolachlor (MET) was used to prevent weed infestation in sorghum fields, but inappropriate application could result in phytotoxicity on sorghum. Exogenous gibberellin A 3 (GA 3 ) has been applied for alleviating the phytotoxicity of MET. However, its detoxification mechanism is still not well known. RESULTS Leaf deformity of sorghum caused by 200 mg/L MET was alleviated by treating sorghum shoots with 800 mg/L GA 3 , and the injury recovery rate of growth index was over 73%. More importantly, GA 3 could not accelerate the metabolic rate of MET in sorghum. The result of phytohormone metabolomics showed that endogenous GA 3 content in sorghum decreased by 78.10% with MET treatment, while abscisic acid (ABA) content increased by 120.2%, resulting in 10.3-fold increase of ABA/GA 3 ratio. Content of ABA and GA 3 increased by 11.9- and 21.1-fold with MET and GA 3 treatment, respectively, leading to ABA/GA 3 ratio restoration. Moreover, MET inhibited the expression of genes encoding key enzymes related to GA synthesis including CPS1 , KO2 , KAO , GA20ox1D and ABA8ox gene related to ABA metabolism. The transcription levels of GA metabolism-related genes CYP714D1 and GA2ox were up-regulated by 11.2- and 7.2-fold, while ABA synthesis-related genes NCED and ZEP were up-regulated by 8.0- and 3.0-fold, respectively, with MET and GA 3 treatment. Conclusion In this study, exogenous GA 3 protecting sorghum shoots from MET phytotoxicity was due to supplement the MET-induced GA 3 deficiency by absorbing exogenous GA 3 , and restore homeostasis of ABA and GA 3 by promoting ABA synthesis, which provides novel insights for mechanism of GA 3 alleviating MET phytotoxicity. © 2022 Society of Chemical Industry.

BACKGROUND Resistance to insect pests is an important self-defense characteristic of pepper plants. However, the resistance of different pepper cultivars to Spodoptera litura larvae, one of the main insect pest species on pepper, is not well understood. RESULTS Among seven pepper cultivars evaluated, cayenne pepper ‘FXBX’ showed the highest repellency to third instar S. litura larvae, Chao tian chili pepper ‘BLTY2’ showed the lowest repellency. Plant volatiles (1-hexene, hexanal, β -ionone, ( E , E )-2,6-nonadienal, and methyl salicylate) affected host selection by S. litura . Among these, 1-hexene, hexanal, and β -ionone at concentrations naturally-released by pepper leaves were found to repel S . litura . Interestingly, S. litura larvae fed on the larva-attracting pepper cultivar, (BLTY2) had an extended developmental period, which was about 13 days longer than larvae fed on FXBX. Besides, the survival rate of larvae fed on BLTY2 was 22.5 ± 0.0%, indicating that the leaves of BLTY2 can kill S. litura larvae. Correlation analysis showed that larval survival rate, emergence rate, female adult longevity, and pupal weight were positively correlated with the vitamin C, amino acids, protein, cellulose, and soluble sugar contents, but were negatively correlated with wax and flavonoids contents. CONCLUSION We identified two different modes of direct defense exhibited by pepper cultivars against S. litura . One involves the release of repellent volatiles to avoid been fed on (FXBX cultivar). The other involves the inhibition of the growth and development or the direct killing of S. litura larvae which feeds on it (BLTY2 cultivar). © 2022 Society of Chemical Industry.

BACKGROUND Research on selenium nanoparticles (SeNPs) in chemical defense and chemotherapy of plants has developed rapidly owing to their high microbial toxicity, environmental safety, and degradability. Pine wilt disease (PWD) threatens pine forests worldwide; however, it is difficult to kill the nematodes ( Bursaphelenchus xylophilus ) inside the tree that cause PWD using traditional pesticide formulations. SeNPs could be the key to controlling PWD. RESULTS In this study, approximately 50 nm SeNPs were prepared using a simple and green method, and chitosan was used to increase their biocompatibility and stability. The preparation and characterization results showed that the prepared SeNPs coated with chitosan (SeNPs@CS) were spherical and evenly dispersed. The bioassay results showed that SeNPs@CS had an LC 50 of 15.627 mg L −1 against B. xylophilus . In addition, the killing mechanism of SeNPs@CS against B. xylophilus was studied. Confocal microscopy and transmission electron microscopy demonstrated that B. xylophilus were killed by reactive oxygen species, and the penetration of nano-form materials to B. xylophilus was higher than that of non-nano-form materials. To verify the effective penetration of SeNPs in pine tissues, Cy5-labeled SeNPs@CS was observed inside pine needles and branches using frozen sections and confocal microscopy. In addition, the cytotoxicity of SeO 2 and SeNPs@CS was tested, and the results showed that the cytotoxicity of SeNPs@CS to MC3T3-E1 cells was reduced. CONCLUSION These results show that SeNPs are expected to be used as a new strategy for the control of PWD with oxidative damage and high penetration to B. xylophilus and effective target penetration and biosafety. © 2022 Society of Chemical Industry.

Background Paeonol is extracted and isolated as a rich and sustainable natural bioresource from the root bark of Paeonia suffruticosa , the derivatives of which exhibit numerous biological activities. It is well known that ester compounds play a very important role in pest control, such as organophosphorus, carbamate and pyrethroid pesticides. Results To discover biorational natural product-based pesticides, three series of (60) paeonol ester derivatives ( 7a–t , 8g , p , 9g , p , 10g–j , n–u , 11g , u , 12g , u , 13a–p , 14b , c , and 15b , c ) were prepared by structural modification of paeonol, and their structures were well characterized by proton nuclear magnetic resonance ( 1 H-NMR), carbon-13 nuclear magnetic resonance ( 13 C-NMR), high-resolution mass spectrometry (HRMS), and melting point. Furthermore, we assessed the compounds as insecticidal, nematicidal, and anti-oomycete agents against three serious agricultural pests, Mythimna separata , Heterodera glycines , and Phytophthora capsici . Among all tested compounds: (i) compound 8p showed more significant insecticidal activity than toosendanin, and the final mortality rates of 8p and toosendanin against M. separata (1 mg mL −1 ) were 70.4%, and 51.9%, respectively; (ii) compound 7a exhibited more promising nematicidal activity than paeonol, and the median lethal concentration (LC 50 ) values of 7a and 1 against H. glycines were 15.47 and 50.80 mg L −1 , respectively; (iii) compounds 7n and 13m displayed more significant anti-oomycete activity compared to zoxamide against Phytophthora capsici , and the median effective concentration (EC 50 ) values of 7n , 13m , and zoxamide were 23.72, 24.51, and 26.87 mg L −1 , respectively; and the protective effect of the compounds against Phytophthora capsici in vivo further confirmed the effectiveness of the agents. Conclusion This study suggested that the introduction of a nitro at the C5 or C3 position of paeonol could improve its bioactivity against M. separata , H. glycines , and Phytophthora capsici . © 2022 Society of Chemical Industry.

BACKGROUND Laodelphax striatellus (Fallén) (Hemiptera: Delphacidae) is one of the most important insect pests of rice, and it has been able to develop strong resistance to many insecticides. Triflumezopyrim, a new type of mesoionic insecticide developed by Corteva Agriscience, showed high biological activity in controlling piercing-sucking insect pests such as planthopper and leafhopper. RESULTS In this study, we continuously selected a susceptible laboratory stain (Unsel) of L. striatellus for 16 generations by exposing it to triflumezopyrim in the laboratory. A 45.1-fold triflumezopyrim-resistant strain (Tri-sel) was established, in which cross-resistance to nitenpyram and acetamiprid was not detected. The realized heritability ( h 2 ) of the Tri-sel strain was estimated at 0.13. The mortalities of the testing F 1 (the offspring of a cross between Unsel and Tri-sel strains) suggested that the resistance of L. striatellus to triflumezopyrim was autosomal and incompletely dominant, as well as a polygenic inheritance. In addition, the results of synergist experiment showed that P450s potentially contributed to the triflumezopyrim resistance. The activities of detoxification enzymes in the Unsel and Tri-sel strains indicated that the activity of P450s in the Tri-sel strain was significantly increased, consistent with the results of synergist experiments. Furthermore, 12 P450 genes demonstrated significant up-regulation. CONCLUSIONS L. striatellus has a certain risk of resistance to triflumezopyrim after continuous selection. Triflumezopyrim resistance did not result in cross-resistance to neonicotinoid insecticides. The up-regulation of multiple P450 genes may mediate triflumezopyrim resistance in L. striatellus . © 2022 Society of Chemical Industry.

BACKGROUND Nanopesticides have been proved to be a powerful and promising tool to solve the issues in agriculture. The purpose of the present study was to develop ecofriendly nanopesticide systems by the strategy of comprehensive utilization of two natural biomass resources (bagasse and turpentine oil) because of their incomparable advantages. RESULTS In this research, a series of nanocellulose carriers ETOCN-1 – ETOCN-4 (ETOCN, esterified TEMPO-oxidized cellulose nanofibers) with different degrees of substitution were prepared and characterized by Fourier-transform infrared (FTIR), X-ray diffraction (XRD) and transmission electron microscopy (TEM). Then, 21 1,3,4-thiadiazole-amide compounds 8a – 8u containing gem -dimethylcyclopropane ring were designed, synthesized and characterized. A preliminary bioassay indicated that compound 8i (R = p -Br Ph) exhibited broad-spectrum antifungal activity against the tested fungi. Furthermore, drug-loading complexes 8i/ETOCN-1 – 8i/ETOCN-4 were fabricated by integration of nanocellulose-based carriers ETOCN-1 – ETOCN-4 with bioactive compound 8i , and the drug-loading capacities, microstructures and sustained-releasing performance of these complexes were also investigated. According to the observation of scanning electron microscopy (SEM) images of complex 8i/ETOCN-2 , the small-molecule drug and the carrier formed a well-distributed and compact complex, which led to the excellent drug-loading capacity and sustained-releasing performance in the ethanol/water (1:1, v/v ) system. CONCLUSIONS Complexes 8i/ETOCN-1 – 8i/ETOCN-4 deserved further study as the promising candidates for the development of nanopesticides. © 2022 Society of Chemical Industry.

BACKGROUND The fall armyworm, Spodoptera frugiperda (J. E. Smith), is a polyphagous moth species that is spreading all around the globe. It uses ( Z )-9-tetradecenyl acetate (Z9-14:Ac) and ( Z )-7-dodecenyl acetate ( Z 7-12:Ac) (100:3.9) as essential sex pheromone components. However, our understanding of the molecular basis of pheromone detection of S. frugiperda is still incomplete. RESULTS Herein, we identified six PRs , i.e. SfruOR6 , 11 , 13 , 16 , 56 , and 62 , by transcriptome sequencing. Subsequently, we heterologously expressed them in Drosophila OR67d neurons and determined their response spectra with a large panel of sex pheromones and analogs. Among them, SfruOR13-expressing neurons strongly respond to the major sex pheromone component Z9-14:Ac, but also comparably to ( Z , E )-9,12-tetradecadienyl acetate (Z9,E12-14:Ac) and weakly to ( Z )-9-dodecenyl acetate (Z9-12:Ac). Both SfruOR56 and SfruOR62 are specifically tuned to the minor sex pheromone component Z7-12:Ac with varying intensities and sensitivities. In addition, SfruOR6 is activated only by Z9,E12-14:Ac, and SfruOR16 by both ( Z )-9-tetradecenol (Z9-14:OH) and ( Z )-9-tetradecenal (Z9-14:Ald). However, the OR67d neurons expressing SfruOR11 remain silent to all compounds tested, a phenomenon commonly found in the OR11 clade of Noctuidae species. Next, using single sensillum recording, we characterized four sensilla types on the antennae of males, namely A, B, C and D types that are tuned to the ligands of PRs, thereby confirming that S. frugiperda uses both SfruOR56 and SfruOR62 to detect Z7-12:Ac. Finally, using wind tunnel assay, we demonstrate that both Z9,E12-14:Ac and Z9-14:OH act as antagonists to the sex pheromone. CONCLUSION We have deorphanized five PRs and characterized four types of sensilla responsible for the detection of pheromone compounds, providing insights into the peripheral encoding of sex pheromones in S. frugiperda .

BACKGROUND A controlled-release formulation based on mesoporous silica nanoparticles (MSNs) provides an effective way for reducing pesticide use and protecting the ecological environment. In this study, MSNs loaded with pyraclostrobin (PYR@MSNs) were prepared using a one-pot method. RESULTS The characteristics of PYR@MSNs were systematically investigated, including morphology, loading content, ultraviolet (UV) resistance, release behavior, control effects against pathogens, and safety to nontarget organisms. The results show that the prepared PYR@MSNs presented characteristics of regular spherical shapes, uniform particle size (200 nm), high drug loading (38.9%), and enhanced UV resistance. Compared with traditional formulation, PYR@MSNs exhibited improved control effects against Fusarium graminearum , an extended control period, and lower toxicity to zebrafish, earthworms and BEAS-2B cells. CONCLUSIONS This research will facilitate the development of efficient and safe pesticide delivery systems. The PYR@MSNs has showed its potential as a new controlled-release formulation with increased efficacy and is expected to benefit the sustainable development of agriculture. © 2022 Society of Chemical Industry.

BACKGROUND Insect pheromone synergists have been widely used to produce potent pheromone products for environment-friendly pest control. Codlemone (Cod) and ( Z )-8-dodecenol (Dod) are two major Grapholita molesta pheromone synergists, with Cod having greater synergism and affinity for G. molesta pheromone binding protein 2 (GmolPBP2). Uncovering structural information key to the different binding affinity of Cod and Dod to GmolPBP2 would gain insights into what causes their synergy activity discrepancy. RESULTS Binding modes of the two synergists in the binding pocket of GmolPBP2 were analyzed and compared by molecular dynamics-based approaches. Although Cod and Dod were stabilized in a similar hydrophobic pocket, their interaction details with GmolPBP2 were divergent due to the extra double bond (C10═C11) in Cod. The C10═C11 improved the hydrophobic interactions of Cod with around residues. Such hydrophobic interaction improvement was also reflected in the raised importance of Phe11 in the GmolPBP2-Cod interaction. Not only that, the increased hydrophobic forces introduced by the C10═C11 changed the CH2–OH orientation in the GmolPBP2-Cod complex, which improved the H-bond interaction. Electrostatic complementarity analysis further indicated the positive role of C10═C11 in optimizing GmolPBP2-Cod interaction. CONCLUSION The C10═C11 is thought to contribute greatly to Cod's stronger synergy as a group key to the higher GmolPBP2-affinity, based on which the improvement directions for Cod and Dod were addressed as well. Our findings will aid in the development and optimization of more effective pheromone synergists, resulting in more effective pheromone-based pest management.

BACKGROUND The seasonal long-distance migration of pests amongst different seasonal habitats may encounter different degrees of insecticide selection pressure, but knowledge of the evolution of resistance and the underlying mechanisms remains sparse. Here, we show that the development of resistance of the oriental armyworm Mythimna separata (Walker), a notorious agricultural pest that migrates between northern and northeast China seasonally across the Bohai Gulf, is influenced by seasonal migration. RESULTS There are two conspicuous migrations of M. separata , the northeastwards population in the second half of May and the returning population in the second half of August, between northern and northeast China per year, and the abundance values of migrants from April to October are significantly different. The resistance levels of seasonal migratory populations to λ-cyhalothrin vary in different months, and the resistance levels and estimated frequency of the resistance allele of the first northeastward population (second half of May, May.-2) in spring are significantly higher than those of the returning populations in autumn. Moreover, resistance decline was observed in migrating population Jul.-2, with the resistance levels in the F 1 progeny higher than those in their F 2 progeny. Synergism tests indicate that cytochrome P450 monooxygenases (P450s) play a major role in resistance. Enzymatic assays show that P450 activity is significantly correlated with resistance levels in migratory populations, indicating that increased P450 activity is the main mechanism of resistance. The expression levels and correlation analysis of the relative expression of P450s with resistance levels show that three ( CYP9A144 , CYP9G40 , and CYP6B79 ) out of 23 genes from CYP6 and CYP9 subfamilies are potentially involved in resistance to λ-cyhalothrin. CONCLUSION Our results show that the resistance of M. separata to λ-cyhalothrin is unstable and likely to be metabolically driven by enhanced P450 activity mediated by the overexpression of multiple P450 genes. Notably, the development of resistance is probably affected by seasonal migration.

BACKGROUND Chinese sprangletop ( Leptochloa chinensis (L.) Nees) is an invasive grass weed severely infesting rice fields across China. In October 2020, a suspected resistant Leptochloa chinensis population HFFD3 that survived the acetyl-CoA carboxylase (ACCase)-inhibiting herbicide cyhalofop-butyl applied at its field-recommended rate was collected from a rice field in Feidong County, Anhui Province, China. This study aimed to determine the resistance profile of HFFD3 to ACCase inhibitors and to investigate its mechanisms of resistance to cyhalofop-butyl. RESULTS Single-dose testing confirmed that HFFD3 had evolved resistance to cyhalofop-butyl. Two loci encoding plastidic ACCase were each amplified from the susceptible (S) and resistant (R, HFFD3) individual plants. Target gene sequencing and derived cleaved amplified polymorphic sequence assay revealed all the R plants carried a Trp-2027-Leu substitution in their ACCase1,2 copies. Dose–response bioassays revealed that HFFD3 was highly resistant to cyhalofop-butyl and exhibited cross-resistance to metamifop, fenoxaprop- P -ethyl, quizalofop- P -ethyl, and clethodim. Pre-treatment with piperonyl butoxide and 4-chloro-7-nitrobenzoxadiazole considerably reversed the resistance of the R plants to cyhalofop-butyl, by 23% and 43%, respectively. Liquid chromatography–tandem mass spectrometry analysis suggested the metabolic rates of cyhalofop-butyl were significantly faster in the R than in the S plants. CONCLUSION This study confirmed the first case of an arable weed species featuring cross-resistance to ACCase-inhibiting herbicides due to a novel Trp-2027-Leu mutation of ACCase . Target gene mutation and cytochrome P450s- and glutathione- S -transferases-involved enhanced metabolism may have simultaneously participated in the resistance of HFFD3 population to cyhalofop-butyl.
