Cu-Fe bimetallic nanozyme@halloysite-sodium alginate composite hydrogel for the treatment of atopic dermatitis

Di Zhang,Luying Zeng,Xiangyu Chen,Shuiqing Zhou,Binghong Luo,Mingxian Liu

Atopic dermatitis is a typical chronic inflammatory disease with pathological characteristics of persistent immune activation and oxidative stress. The combined anti-inflammatory and antioxidant treatment can effectively block the inflammatory cascade while reducing oxidative damage. Halloysite nanotubes (HNTs) are the main component of the traditional Chinese medicine "Chishizhi", which shows the medicinal functions of astringency, hemostasis and astringency. However, the efficacy of HNTs alone in treating diseases is relatively weak, and its therapeutic effect can be improved by surface modification and drug loading. Herein, CuO-Fe3O4 nanoparticles were synthesized on the outer surfaces of HNTs by hydrothermal reaction. CuO-Fe3O4@HNTs has high SOD and CAT enzyme activities under neutral condition. Then, the nanozyme modified HNTs powder was prepared into sprayable hydrogels by introduction of sodium alginate (SA) and aloe vera extracts. Cell experiments confirmed that the hydrogel can promote HacaT cell proliferation within 0~200 μg/mL concentration. Through the mouse animal dermatitis model, CuO-Fe3O4@HNTs-SA composite hydrogel has a good therapeutic effect on atopic dermatitis. Comparing with the positive drug halcinonide solution, the CuO-Fe3O4 nanozymes incorporated hydrogel showed enhanced therapeutic effect, which shows promising prospects in the clinical treatment of atopic dermatitis.

Sodium alginate-based photothermal film: Preparation, antibacterial activity, and application for preservation of Kyoho Grapes

Zihan Hou,Yunxiao Xia,Yuan Sun,Ming Yang,Lingling Wang,Weizhuo Xu,Xu Zhao

The photothermal effect enhances the antibacterial activity by generating localized heat that disrupts microbial cells, leading to increased bacterial inactivation. The objective of this study was to develop and evaluate the antibacterial and photothermal applications of polydopamine-berberine nanoparticles (PDA-BBR-NPs), incorporating them into a sodium alginate matrix for food preservation. The self-assembled PDA-BBR-NPs were synthesized using a solution oxidation method and characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, and nanoparticle size analysis. The results demonstrated that PDA-BBR-NPs were uniform spheres with an average diameter of 177.1 nm. Drug loading and encapsulation efficiency tests showed that the optimal formulation had a drug loading capacity of 48.4% and an encapsulation efficiency of 84.7%. Antibacterial activity against E. coli and S. aureus was evaluated using the dilution coating plate method, showing an inhibition rate of over 98%. Photothermal testing under 808 nm near-infrared irradiation revealed a maximum photothermal conversion efficiency of 32.65%. To assess the practical application of the nanoparticles, they were incorporated into a sodium alginate-based coating for the preservation of Kyoho grapes. Key parameters such as weight loss, pH, titratable acidity, and soluble solids content were monitored over a 12-day storage period at room temperature. The PDA-BBR-NIR-treated group exhibited significantly reduced weight loss (6.08% lower than the control group) and a slower decrease in pH. The coating effectively inhibited microbial activity and extended the shelf life of the grapes by 3-6 days compared to the control group. These findings demonstrated the potential of PDA-BBR-NPs to enhance the preservation of fresh produce through their combined antibacterial and photothermal effects, offering a promising method for extending fruit shelf life.

Predator-prey interactions mediate the formation of Janus droplets: The role of sodium alginate droplets

Xiaolong Li,Changhu Xue,Zihao Wei

The asymmetric structure of Janus droplets endows them with unique functionalities, such as self-propulsion, compartmentalized reactions, and directional encapsulation, demonstrating broad application potential in the food industry. However, the fabrication of food-grade Janus droplets still faces the challenges of low efficiency and yield. Drawing inspiration from predator-prey relationships in nature, this study proposed a novel biomimetic strategy for constructing Janus droplets and regulating their yield through predator-prey interactions. The difference in the interfacial tension between zein and sodium alginate droplets conceptually rationalized the predator-prey interactions and the formation of Janus droplets in the mixed water-in-oil droplet system. The predator-prey interactions between the droplets were activated by external forces, driving the mixed droplet system toward the direction of free energy minimization. In this process, driven by the Marangoni effect arising from the interfacial tension gradient, zein droplets (the predator) captured sodium alginate droplets (the prey). Crucially, the yield of Janus droplets could be controlled by adjusting the physical parameters of sodium alginate droplets within the predator-prey framework. Specifically, regulating the number ratio and size ratio of sodium alginate to zein droplets, as well as the molecular weight of sodium alginate, led to an increase in Janus droplet proportion. This work provides a biomimetic approach for the efficient fabrication of food-grade Janus droplets, paving the way for their advanced applications in the food industry.

Injectable in situ dual-crosslinking hyaluronic acid and sodium alginate based hydrogels for drug release

Yufan Zhang,Xian Li,Nan Zhong,Yuanlin Huang,Kewen He,Xu Ye

A series of injectable in situ dual-crosslinking hydrogels (HA/ALG) based on oxidized sodium alginate (oxi-ALG) and hyaluronic acid modified with thiol and hydrazide (HA-SH/CDH) were prepared via hydrazone bonds and disulfide bonds. The chemical structures, morphologies, rheological properties, gelling time, swelling ratio, degradation rate and drug release behavior of hydrogels were investigated. HA/ALG hydrogels exhibited tunable gelling time, rheological properties, swelling ratio and degradation rate with varying precursor concentrations. The gelling time of HA/ALG hydrogels ranged from 157 s to 955 s, the values of yield stress of HA2/ALG2, HA3/ALG3 and HA4/ALG4 hydrogels were 1724, 4349 and 5306 Pa, and the degradation percentage of HA2/ALG2, HA3/ALG3 and HA4/ALG4 hydrogels were about 64%, 51% and 42% after incubating 35 days, respectively. Bovine serum albumin (BSA) was used as a model drug to investigate the drug controlled release properties, and the in vitro cumulative release percentage of BSA from HA2/ALG2, HA3/ALG3 and HA4/ALG4 drug-loaded hydrogels were about 79%, 72% and 69% after 20 days. The series of injectable in situ dual-crosslinking HA/ALG hydrogels could be an attractive candidate for drug delivery system, tissue engineering and regenerative medicine.

Double-network agar/sodium alginate hydrogel-based photothermal evaporator with layered structure for solar desalination

Lai Jinyin,Guo Zhengqiang,Liu Wangyu

Interface solar evaporators generate fresh water through solar evaporation, presenting significant potential to address global water scarcity. However, current evaporators struggle to balance sustainability, high evaporation rates, and robust mechanical properties, resulting in limited applications. In this study, agar/sodium alginate hydrogels were synthesized using a dual-network strategy, with the internal water state adjusted to optimize their properties. Additionally, the incorporation of nanocellulose significantly enhanced the mechanical properties of the evaporator (improved 65% compressive strength), thereby extending its service life in harsh environments. Moreover, nanocellulose acted as an efficient green dispersant for carbon nanotubes (CNTs), facilitating photothermal conversion and improving solar energy absorption. Notably, we employed a freeze–thaw process to induce lateral aggregation of sodium alginate, resulting in layered water channels that optimized water transport and evaporator performance. This unique design resulted in the development of evaporators that exhibited a high evaporation rate of 3.286 kg·m −2 ·h −1 in a 3.5 wt% NaCl solution, achieving a photothermal conversion efficiency of 98.65% under 1 kW·m −2 light intensity. Moreover, even in a 20 wt% NaCl solution, the evaporation rate remained at 3.02 kg·m⁻ 2 ·h⁻ 1 . This work combined highly hydrophilic natural materials with a specialized water delivery channel design to develop hydrogel-based solar evaporators with high evaporation rates and excellent mechanical properties, offering strong potential for long-term interfacial evaporation applications.

Calcium carbonate hollow microspheres encapsulated cellulose nanofiber/sodium alginate hydrogels as a sequential delivery system

Wenqian Bai,Haiying Chen,Junyao Li,Wenrong Cai,Yong Kong,Xiaoming Zuo

By using folic acid (FA) as the template, calcium carbonate hollow microspheres (CaCO 3 HMs) are prepared through the Ostwald ripening process, which can be utilized for the loading of doxorubicin hydrochloride (DOX). The DOX loaded CaCO 3 HMs (CaCO 3 /DOX) are co-encapsulated with ibuprofen (IBU) in the cellulose nanofiber (CNF)/sodium alginate (SA) hydrogels cross-linked by Ca 2+ . The loading efficiency of DOX in the CaCO 3 HMs is 95.7 %, and the loading efficiency of IBU in the hydrogels is 97.0 %. In the weakly alkaline environment (pH ~7.4) that is characteristic of intestinal fluids of human body, the CNF/SA hydrogels are swollen and the encapsulated IBU is first released for pain control, and the release rate of IBU can reach 57.8 %. In the weakly acidic environment (pH ~6.5) that is characteristic of colonic fluids of human body, the CaCO 3 HMs are decomposed to release the loaded DOX with a release rate of 50.1 %, which can be used for the treatment of colorectal cancer. The results of release kinetics indicate that the delivery of IBU is governed by first-order model and DOX by zero-order model. The developed sequential delivery system (SDS) can not only enable the release of DOX in colon of human body, but also simultaneously relieve the pain of patients during the chemotherapy of colon cancer.

Ultrasonic engineering of zein-curcumin nanoparticles/sodium alginate composite films for active food packaging applications

Qiufang Liang,Panpan Cao,Haina Lu,Yunxia Du,Lixin Kang,Haile Ma,Xiaofeng Ren

Food spoilage poses a significant threat to human health and contributes to substantial global food waste. To address this issue, advancements in biopolymer-based packaging materials have garnered increasing research attention. A novel zein-curcumin nanoparticles/sodium alginate (U-ZcNPs/SA) composite films was fabricated by encapsulating curcumin (Cur) within zein nanoparticles via ultrasonic-assisted self-assembly, followed by integration into an SA matrix. Ultrasonic treatment induced structural reorganization of zein, enhancing electrostatic interactions and hydrogen bonding within the film matrix, which improved nanoparticle dispersion, mechanical properties (tensile strength: 40.51 N/mm 2 ; elongation at break: 72.23 %), and barrier performance (oxygen permeability: 2.44 g/hm 2 ). Characterization techniques (SEM, FTIR, XRD, TGA) confirmed superior interfacial compatibility, thermal stability, and amorphous structure in U-ZcNPs@SA films. Furthermore, the films demonstrated robust antibacterial activity against both Escherichia coli and Staphylococcus aureus , with inhibition zones measuring up to 11.09 cm and 10.78 cm, respectively. In the preservation experiment, the U-ZcNPs@SA film effectively extended the shelf life of grapes and strawberries by maintaining their visual quality. Notably, it significantly reduced weight loss in strawberries while suppressing their respiration rate and ethylene production. These results demonstrated that ultrasonic modification optimized the functional performance of biodegradable films, offering a sustainable strategy for active food packaging.

A sensitive humidity-responsive actuator based on dispersing graphene oxide into chitosan- sodium alginate nanofibers

Yonghao Chen,Meijie Qu,Chenshen Liu,Shuhui Li,Xincheng Wang,Daixuan Gong,Ping Tang,Yuezhen Bin

Humidity-driven actuators have attracted much attention from researchers due to their fast response and recovery speed, high sensitivity, and large bending deformation. Graphene oxide (GO), with abundant oxygen-containing functional groups and a large specific surface area, has been widely used in the field of flexible humidity-driven actuators. However, pure graphene oxide films have poor mechanical properties and are easily damaged in practical applications, and these limitations can be addressed by integrating GO with other materials. It has been reported that chitosan (CS) and sodium alginate (SA) can be assembled together by ultrasound to prepare CSSA nanofibers with high tensile strength and excellent hygroscopic properties. In this study, a uniform, flexible actuator based on a thin film was developed by combining graphene oxide with CSSA nanofibers. The obtained 32 μm CSSA10-GO1 film (the mass ratio of CSSA and GO is 10:1) exhibits excellent humidity-driven performance, with maximum deflection angle and driving force of 218° and 2.5 N at ΔRH = 45 %, and response and recovery times of 20.5 s and 201.8 s, respectively. In addition, the film has excellent cycle and solubility resistance stability, and the prepared film remains stable even after 50 humidity cycle tests and 7 days of immersion in water. In addition, the composite film was able to grip and lift heavy objects up to 70 times its own weight by simple design. With such excellent properties, the prepared composite film is expected to be used in soft robotics, artificial muscles and other applications.

Medium effects of polysaccharides: bidirectional regulation of invertase activity by sodium alginate and fenugreek gum

Lei Wang,Biao Wang,Pan-Pan Chen,Mi-Zhuan Li,Yan Cheng,Zhong-Xiu Chen

Sodium alginate enhanced invertase activity, whereas fenugreek gum suppressed it. Further investigation into the physicochemical properties of the medium revealed a bidirectional regulatory mechanism. Sodium alginate, characterized by a positive logarithm of the crowding factor (ln Γ), a smaller excluded volume (Vₑₓ), and improved hydration, interacted with invertase via enthalpy-driven hydrogen bonding. This interaction stabilized the α-helix and β-sheet structures of the enzyme, thereby increasing its activity. In contrast, fenugreek gum exhibited a negative ln Γ, a larger excluded volume, and lower hydration capacity. It perturbed the enzyme through entropy-driven hydrophobic interactions, leading to a reduction in β-sheet content and a consequent decrease in enzymatic activity. The overall influence of the medium—comprising viscosity, macromolecular crowding, and soft interactions—enables quantitative assessment of how polysaccharides affect invertase activity. This study offers valuable insights for the rational selection of polysaccharides in dietary formulations aimed at modulating digestive enzyme activity.

Construction and Characterization of Emulsions Stabilized by Whey Protein Isolate-Naringin-Sodium Alginate Ternary Complex

Si Chen,Mengmeng Wei,Guoyang Liu,Benguo Liu

This study constructed ternary complexes of whey protein isolate (WPI), naringin (NAR), and sodium alginate (SA) and their stabilized emulsions, and characterized their physicochemical properties. The results confirmed interactions between NAR and SA with WPI, which induced quenching of WPI intrinsic fluorescence; formation of the ternary complexes could effectively inhibit WPI flocculation near its isoelectric point. Compared with the corresponding binary complexes, the WPI-NAR-SA ternary complexes developed more stable emulsions with smaller droplet sizes and higher absolute ζ-potential values, exhibiting superior storage stability that increased with SA concentration. After storage at 4 °C for 20 days, the emulsion system exhibited a peroxide value remaining below 4.7 μg/mL; after 9 days of UV irradiation, lutein retention reached 82.49%. Both protective effects increased with rising SA concentration. This study provides a strategy for the development of a novel functional emulsion delivery system.

Sodium alginate and naloxone loaded macrophage-derived nanovesicles for the treatment of spinal cord injury

Xiaoyao Liu,Xue Jiang,Qi Yu,Wenwen Shen,He Tian,Xifan Mei,Chao Wu

Spinal cord injury (SCI) causes Ca 2+ overload, which can lead to inflammation and neuronal apoptosis. In this study, we prepared a nanovesicle derived from macrophage membrane (MVs), which encapsulated sodium alginate (SA) and naloxone (NAL) to inhibit inflammation and protect neurons by reducing the free Ca 2+ concentration at the SCI site. Based on the transmission electron microscopy (TEM) image, the encapsulated sample (NAL–SA–MVs) had a particle size of approximately 134 ± 11 nm and exhibited a sustained release effect. The encapsulation rate of NAL and SA was 82.07% ± 3.27% and 72.13% ± 2.61% in NAL–SA–MVs, respectively. Targeting tests showed that the NAL–SA–MVs could accumulate in large quantities and enhance the concentration of SA and NAL at the lesion sites. In vivo and in vitro studies indicated that the NAL–SA–MVs could decrease the concentration of free Ca 2+ , which should further alleviate the inflammatory response and neuronal apoptosis. Anti-inflammation results demonstrated that the NAL–SA–MVs could reduce the pro-inflammation factors (iNOS, TNF-α, IL-1β, IL-6) and increase the expression of anti-inflammation factors (IL-10) at the cell and animal level. Concurrently, fluorescence, flow cytometry and western blot characterization showed that the apoptotic condition of the neurons was significantly inhibited. In addition, the motor function of C57 mice were significantly improved after NAL–SA–MVs treatment. In conclusion, it is suggested that the NAL–SA–MVs has tremendous potential in the treatment of SCI.

Encapsulation and characterisation of grape seed proanthocyanidin extract using sodium alginate and different cellulose derivatives

Kangliang Sheng,Guanghui Zhang,Xiaowei Kong,Jingmin Wang,Wanmeng Mu,Yongzhong Wang

Summary Grape seed proanthocyanidin extract (GSPE) has health benefits. However, these phenolic compounds undergo degradation reactions and have undesirable sensorial characteristics. GSPE was encapsulated using sodium alginate (SA), SA-methyl cellulose (MC) and SA-hydroxypropyl methylcellulose (HPMC) for promoting controlled release, pH stability, temperature and storage period tolerance of GSPE. The microcapsules were characterised using scanning electron microscopy (SEM) and Fourier transform infra-red (FTIR) analysis. The SA-MC and SA-HPMC microcapsules appeared to have a more compact surface than SA-alone microcapsules. FTIR analysis indicated successful immobilisation of GSPE into the polymeric microcapsules. Moreover, the SA-MC and SA-HPMC microcapsules showed higher thermal stability. The microcapsules showed a relatively higher amount of released GSPE at a pH above six than at a lower pH. The SA-MC and SA-HPMC microcapsules could be used to retain more GSPE content in the gastric phase and to release it in the intestinal phase for possible absorption. Furthermore, after 28 days of storage at 25 °C, the GSPE retention rate of the microcapsules was still higher than 80%. GSPE encapsulated in SA-MC and SA-HPMC microcapsules results in lesser degradation and can be absorbed more effectively. This method has potential for the delivery of colon-specific materials while exhibiting a sustained-release characteristic.
