MoO3/MIL-125-NH2 with boosted peroxidase-like activity for electrochemical staphylococcus aureus sensing via specific recognition of bacteriophages

Yajun Gao,Suhui Xu,Ge Guo,Yajie Li,Wenyuan Zhou,Huaxiang Li,Zhenquan Yang

Herein, novel nanozyme mimics MoO 3 /MIL-125-NH 2 were reported and conjugated with bacteriophages as a new electrochemical probe for high sensitivity and specific electrochemical detection of staphylococcus aureus . The excellent peroxidase-like activity of MoO 3 /MIL-125-NH 2 composites was attributed to the integration of MIL-125-NH 2 with MoO 3 , which can boost the generation of superoxide radicals (O• 2 – ) and thus promote the oxidation of TMB in the presence of H 2 O 2 . In this work, two bacteriophages named SapYZU04 and SapYZU10 were isolated from sewage samples by using staphylococcus aureus YZUsa12 as the host. In comparison, MoO 3 /MIL-125-NH 2 @SapYZU04 was selected as a recognition agent. The DPV current declined linearly with staphylococcus aureus YZUsa12 concentration in the range of 10 1 −10 8  CFU mL −1 , with a low detection limit of 16 CFU mL −1 (S/N = 3). 20 strains including 13 host strains and 7 non-host strains were used to evaluate the selectivity of the proposed sensor. Regardless of the differences in the degrees of lytic performance for phage SapYZU04, all selected host strains can be screened with merely the same DPV current. Host spectrum-oriented bacteriophage sensing is of great importance for the practical application of bacteriophage-based biosensors in the future.

NIR-II-amplify high-entropy MXene-based sonosensitizer as sonodynamic therapy promotes methicillin-resistant Staphylococcus aureus–Infected wound healing

Wei Song,Danyan Wang,Shuai Xiao,Xiaojun He,Wei Xiong,Jianliang Shen

The growing pace of two-dimensional (2D) transitional metal carbides and nitrides (MXenes) has become a spotlight for recent research. Similarly, the need to close the gap left by conventional antibiotics, fighting off bacterial infections, and drug resistance has become eminent. Especially, MXenes are widely used as excellent photothermal agents. However, there are limited reports on MXenes as sonosensitizer. Herein, a high-entropy MXene (HEM), TVNMC was fabricated for the generation of reactive oxide species (ROS) against Methicillin-resistance Staphylococcus aureus (MRSA). Interestingly the energy storage and conversion from the diverse composition of the TVNMC MXenes allowed the generation of ROS under US irradiation and even enhanced under Photothermal therapy. This combinatorial therapy undertook a near-total kill and eradication of MRSA with its biofilm. Moreover, TVNMC MXene simultaneously eliminated MRSA abscesses while promoting wound healing with negligible anomaly to murine tissues under sonodynamic and photothermal therapy, making the TVNMC MXene a promising sonosensitizer for possible biomedical usage. Overall, this system provides great potential for the use of high-entropy MXenes as a new type of sonosensitizer in anti-infection therapy.

In vivo photothermal inhibition of methicillin-resistant Staphylococcus aureus infection by in situ templated formulation of pathogen-targeting phototheranostics

Xujuan Guo,Bing Cao,Congyu Wang,Siyu Lu,Xianglong Hu

Bacterial infection has caused a serious threat to human public health. Methicillin-resistant Staphylococcus aureus (MRSA) is a representative drug-resistant bacterium, which is difficult to eradicate completely, resulting in high infection probability with severe mortality. Herein, pathogen-targeting phototheranostic nanoparticles, Van-OA@PPy, are developed for efficient elimination of MRSA infection. Van-OA@PPy nanoparticles are fabricated from the in situ templated formation of polypyrrole (PPy) in the presence of ferric ions (Fe3+) and a polymer template, hydrophilic poly(2-hydroxyethyl methacrylate-co-N,N-dimethyl acrylamide), P(HEMA-co-DMA). PPy nanoparticles are further coated with vancomycin conjugated oleic acid (Van-OA) to afford the resultant pathogen-targeting Van-OA@PPy. A high photothermal conversion efficiency of ∼49.4% is achieved. MRSA can be efficiently killed due to sufficient nanoparticle adhesion and fusion with MRSA, followed by photothermal therapy upon irradiation with an 808 nm laser. Remarkable membrane damage of MRSA is observed, which contributes greatly to the inhibition of MRSA infection. Furthermore, the nanoparticles have high stability and good biocompatibility without causing any detectable side effects. On the other hand, residual Fe3+ and PPy moieties in Van-OA@PPy endow the nanoparticles with magnetic resonance (MR) imaging and photoacoustic (PA) imaging potency, respectively. The current strategy has the potential to inspire further advances in precise diagnosis and efficient elimination of MRSA infection in biomedicine.

Development of a Novel Probe-Based Dual-Detection platform System for Flexible Detection of Staphylococcus aureus across Transmission Pathways

Shengyang Wang,Wenjin Hu,Yuanrong Li,Jin Qi,Yi Luo,Yaling Liu

Immunodetection plays a crucial role in the prevention and treatment of Staphylococcus aureus infections. However, existing methods often suffer from limited performance, poor quantification, and applicability due to the use of low-sensitivity, nonspecific, and nontargeted probes. To address these challenges, we identified high-affinity, specific phage-displayed nanobody probes and established a dual-detection platform that enabled accurate and flexible screening of S. aureus across diverse settings. High-affinity phage-displayed nanobodies targeting S. aureus were selected to replace the conventional probes and were randomly paired with five monoclonal antibodies to demonstrate their universality. To further enhance detection sensitivity, we synthesized and optimized a core–shell magnetic–fluorescent nanolabel (Fe3O4@PS@TQD), which outperformed traditional colorimetric and fluorescent tags. By integrating these advanced components, we established a dual-detection platform capable of qualitative detection within 10 min and quantitative analysis within 30 min, achieving superior sensitivity and applicability compared with recently reported methods and commercial kits. The system exhibited excellent specificity, reproducibility, accuracy, and stability, with a detection limit as low as 5 CFU/mL after 4 h of pre-enrichment. In summary, this study presents a novel probe-driven dual-detection platform that meets the urgent need for rapid S. aureus on-site detection in complex environments.

Bacterial metabolism-triggered-chemiluminescence-based point-of-care testing platform for sensitive detection and photothermal inactivation of Staphylococcus aureus

Xinghui Yu,Yongqiang Ma,Siyuan Liu,Chunchun Qi,Weiqi Zhang,Wen Xiang,Zhaoxian Li,Kai Yang,Shaoxian Duan,Xinrao Du,Jian Yu,Yan Xie,Zicheng Wang,Wentao Jiang,Li Zhang,Xiaodong Lin

Post-operative pathogenic infections in liver transplantation seriously threaten human health. It is essential to develop novel methods for the highly sensitive and rapid detection of Staphylococcus aureus ( S. aureus ). Interestingly, the combination of the property of bacteria to secrete hydrogen peroxidase , bacterial metabolism-triggered-chemiluminescence (CL)-based bioassays can be as a candidate point-of-care testing (POCT) for the detection of S. aureus against the CL substrate Luminol and hydrogen peroxide without excitation light sources. Here, a CL-based strategy with stable and visualized CL intensity was fabricated according to a hybrid biomimetic enzyme of copper-Hemin metal-organic framework, which enhances the biological enzyme activity while improving the stability and sensitivity of the assay. By further integrating S. aureus -specific capture and one-step separation of the antibody-modified Fe 3 O 4 NPs (Fe 3 O 4 NPs@Ab), the portable device integrated smartphone enables CL-based POCT for specific detection of S. aureu s in the range of 10 1 –10 6  CFU/mL with a limit of detection as low as 1 CFU/mL. Specifically, S. aureus can be eliminated after detection with high antibacterial efficiency due to the excellent photothermal properties of Fe 3 O 4 NPs@Ab. The developed multifunctional platform has the advantages of simplicity of operation and low cost, indicating great potential in clinical applications.

Insights into the antibacterial activity and antibacterial mechanism of silver modified fullerene towards Staphylococcus aureus by multiple spectrometric examinations

Yin-Xu Pan,Qiu-Hui Xu,Hua-Ming Xiao,Chun-Ya Li

Clarifying the antibacterial mechanism of silver (Ag)-based materials is of great significance for the rational design, synthesis, and evaluation of antimicrobials. Herein, detailed description of the antibacterial mechanism of a synthesized silver deposited fullerene material (Ag(I)–C 60 ) towards Staphylococcus aureus was surveyed from the point of view of DNA damage by ultraviolet–visible spectroscopy (UV–vis), inductively coupled plasma mass spectrometry (ICP-MS), and liquid chromatography-mass spectrometry (LC-MS). The model material, Ag(I)–C 60 , was prepared by liquid-liquid interfacial precipitation method, and characterized by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), thermos-gravimetric analysis (TGA), and nitrogen adsorption/desorption analysis. Ultra-efficient bacteriostatic rate of Ag(I)–C 60 was found to be 88.98% under light irradiation for 20 min. UV–vis measurement of the composition changes of four DNA bases showed that they changed in the presence of Ag(I)–C 60 under light irradiation, suggesting Ag(I)–C 60 could destroy the cells and genetic material of Staphylococcus aureus and thereby inhibit its growth and reproduction. ICP-MS analysis demonstrated the releasing behavior of Ag + from Ag-based materials. Finally, the transformation pathway of G, A, C, and T were measured by LC-MS, demonstrating the conversion of Adenine ( m / z 136.06) to 8-OH-Ade ( m / z 174.04). These collective results suggested that Ag(I)–C 60 was a new ultra-efficient antibacterial by slowly releasing Ag + in water and producing a large amount of ROS under light.

NIR-II photo-amplified peroxidase-like activity using biocatalytic copper-based nanoplatform against subcutaneous methicillin-resistant Staphylococcus aureus infection

Danyan Wang,Xiaojun He,Qiang Wang,Pingping Li,Enoch Obeng,Dongyang Zheng,Jianliang Shen,Zi Jin

Methicillin-resistant Staphylococcus aureus (MRSA) has been associated with significant mortality and longer hospital stays. Artificial nanozymes not requiring antibiotics can generate enough reactive oxygen species (ROS) to kill bacteria. In this study, we synthesized hexagonal copper sulfide nanosheets (CuS NSs) using a modified one-pot synthesis. The CuS NSs demonstrated photothermal stability and superior photothermal conversion efficiency due to large surface area and existence in the second near-infrared (NIR-II) window (1270 nm), promoting the production of ROS. Upon NIR-II irradiation, CuS NSs displayed an excellent biocatalytic antibacterial role in vitro and accelerated abscess resolution, and enhanced the recovery of MRSA-infected wounds in vivo . Moreover, CuS NSs demonstrated negligible toxicity following an amplifying peroxidase-like activity through hyperthermia. Overall, CuS NSs provide a feasible strategy for using the synergistic therapeutic platform for effective therapy of deep-seated MRSA-infected wounds.

Photothermal and natural activity-based synergistic antibacterial effects of Ti3C2Tx MXene-loaded chitosan hydrogel against methicillin-resistant Staphylococcus aureus

Yuanhao Dong,Jianhong Liu,Yuan Chen,Tao Zhu,Yuanhong Li,Chaoliang Zhang,Xin Zeng,Qianming Chen,Qiang Peng

Methicillin-resistant Staphylococcus aureus (MRSA) has strong resistance to antibiotic therapy. In this regard, developing antibiotic-free antibacterial agents is of great significance to treat MRSA infections. Herein, we loaded Ti 3 C 2 T x MXene nanomaterial in the non-crosslinked chitosan (CS) hydrogel. The obtained MX-CS hydrogel is expected to not only adsorb MRSA cells via CS-MRSA interactions, but also gather the MXene-induced photothermal hyperthermia, achieving the efficient and intensive anti-MRSA photothermal therapy. As a result, under NIR irradiation (808 nm, 1.6 W/cm 2 , 5 min), MX-CS showed a greater photothermal effect than MXene alone did (30 μg/mL, 49.9 °C for MX-CS and 46.5 °C for MXene). Importantly, MRSA cells were rapidly adsorbed on MX-CS hydrogel (containing 30 μg/mL MXene) and completely inhibited (99.18 %) under NIR irradiation for 5 min. In contrast, MXene (30 μg/mL) and CS hydrogel alone only inhibited 64.52 % and 23.72 % MRSA, respectively, significantly lower than the inhibition caused by MX-CS ( P  < 0.001). Interestingly, when the hyperthermia was depleted by a 37 °C water bath, the bacterial inhibition rate of MX-CS significantly decreased to 24.65 %. In conclusion, MX-CS hydrogel has a remarkable synergistic anti-MRSA activity by gathering MRSA cells and MXene-induced hyperthermia, and may have great potentials in treating MRSA-infected diseases.

Background Iron oxide nanocomposites have received a great attention for their application in various fields like physics, medicine, biology, and material science etc., due to their unique properties, such as magnetism, electrical properties, small size, biocompatibility and low toxicity. Methods Fe 3 O 4 /Ag 3 PO 4 @WO 3 nanocomposites with different weight percent of Ag 3 PO 4 were successfully prepared through fabricated Ag 3 PO 4 /Fe 3 O 4 with WO 3 via in situ fabrication method, electrospinning involved precursor solution preparation and spinning to enhance photocatalyst performance under simulated sunlight for the degradation of methylene blue (MB) and antibacterial activity against Staphylococcus aureus ( S. aureus ). Results The photocatalytic degradation of methylene blue (MB) under simulated light irradiation indicated that the nanocomposite with 0.25 mg of Ag 3 PO 4 has the best activity. An additional advantage of these photocatalysts is magnetic recoverability, using external magnetic field and photocatalytic stability of the nanocomposites was evaluated for three cycles. In addition, using different scavengers, holes (h + ) and superoxide radical (O ·−) 2 radicals and hydroxide radical ( · OH) were identified the main oxidative species in the degradation reaction of methylene blue. Conclusions The results reveal that Fe 3 O 4 /Ag 3 PO 4 @WO 3 -0.25 nanocomposites have photocatalytic and antibacterial activity against S. aureus. The photocatalyst and mechanism based on the enhancement of electron transfer processes between Ag 3 PO 4 and WO 3 nanoparticles.

The preservative potential of Octopus scraps peptides−Zinc chelate against Staphylococcus aureus: Its fabrication, antibacterial activity and action mode

Zheng Fang,Liangzong Xu,Yanlan Lin,Xixi Cai,Shaoyun Wang

A new type of zinc-chelating peptide was fabricated from Octopus scraps protein hydrolysate and its antibacterial activity against Staphylococcus aureus was characterized. Results of Fluorescence spectra and Fourier Transform Infrared Spectrometer (FT-IR) demonstrated that the amino nitrogen atoms and the oxygen atoms belonging to the carboxylate groups were the primary chelating sites for zinc ions. The zinc-peptide chelate exerted remarkably antimicrobial ability towards S. aureus , whose minimum inhibitory concentration (MIC) was 1.56 mg/mL. To investigate the antibacterial mechanism, changes of bacterial morphology, cell constituents, membrane potential, intracellular reactive oxygen species (ROS) level and intracellular enzyme activity were measured. Results revealed that Zn-peptide exerted its bactericidal activity by damaging bacterial cell membranes and cell walls, increasing the cell permeability which resulted in structural lesions and release of cell components. Besides, Zn-peptide induced S. aureus cell death via ROS triggered pathway. These findings further promote the development of Zn-peptide complexes, suggesting that Zn-peptide chelate has potential as natural bacteriostatic agent in food processing industry.

Green synthesis characterization and antimicrobial activity against Staphylococcus aureus of silver nanoparticles using extracts of neem, onion and tomato

Kishore Chand,M. Ishaque Abro,Umair Aftab,Ahmer Hussain Shah,Muhammad Nazim Lakhan,Dianxue Cao,Ghazanfar Mehdi,Abdalla Mohamed Ali Mohamed

Recently, it has been shown that silver nanoparticles (AgNPs) exhibit great potential for different applications, including food storage, cosmetic products, electronic components, biosensor materials, cryogenics, dental materials and especially for drug-delivery activities. In this study, we synthesized AgNPs with neem extract (NE) alone and mixed plant extracts of neem, onion and tomato (NOT) as a combined reducing and stabilizing agent by a green synthesis method at different pHs. The synthesized products were characterized by ultraviolet-visible spectroscopy (UV-vis), X-ray diffraction (XRD), dynamic light scattering (DLS), atomic force microscopy (AFM), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The antibacterial effects of the synthesized products were studied by the Kirby disk diffusion method. It was confirmed that the AgNPs work effectively as a drug material against Gram-positive bacteria Staphylococcus aureus in nutrient agar. In addition, it was seen that the reducing and stabilizing agent NOT could work effectively with six medicines with a different nature at the maximum addition of 15 μg. However, the synthesized product with NE alone only worked for four of the medicines. Therefore, it was evident that the AgNPs synthesized with NOT extract were more susceptible to the Gram-positive bacteria Staphylococcus aureus. We believe that this new route for synthesizing AgNPs with NOT extract could be more beneficial in comparison to NE alone for improved antibacterial properties in drug-delivery applications.

Ultrasensitive hairpin mediated upconversion fluorescence biosensor for Staphylococcus aureus detection in foods and waters exploiting g–C3N4–assisted catalysis

Yi Xu,Waqas Ahmad,Md Mehedi Hassan,Huanhuan Li,Qin Ouyang,Quansheng Chen

A novel g-C 3 N 4 nanosheets (g-C 3 N 4 NSs)-assisted upconversion fluorescent aptasensor was proposed for Staphylococcus aureus ( S. aureus ) detection by adopting hybridization chain reaction (HCR) as a sensitizer. Two hairpin (H1 and H2) structured DNA probes were engineered predicated on the partial complementary sequence (cDNA) of S. aureus aptamer and modified on the exterior of the upconversion nanoparticles (UCNPs), respectively. The presence of S. aureus initiated the HCR system and activated H1 and H2 probes to form a double-helix away from the g-C 3 N 4 NSs vicinity. This led to the decrease in peroxidase-like activity (PA) of the g-C 3 N 4 NSs and corresponding fluorescence recovery proportional to the concentration of S. aureus (10-10 6  cfu mL −1 ). The method was applied to real food samples with acceptable recoveries (91.1–101.6%) and further validated by traditional plate counting method (p > 0.05).
