Background To date, the rapid clearance from ocular surface has been a huge obstacle for using eye drops to treat glaucoma, since it has led to the short preocular residence time and low bioavailability. Methods The novel nanoparticles (NPs) were designed for topical ophthalmic controlled drug delivery system through intercalating the BH into the interlayer gallery of Na-montmorillonite (Na+Mt) and then further enchasing chitosan nanoparticles. The resulting nanoparticles had a positive charge (+29±0.18 mV) with an average diameter of 460±0.6 nm.Results In vitro study of drug release profiles suggested controlled release pattern. The irritation experiment analysis on both human immortalized cornea epithelial cell (iHCEC) and chorioallantoic membrane-trypan blue staining (CAM-TBS) showed good tolerance for ocular tissues. It was interestingly found that the nanoparticles could enter into iHCEC from the result of cellular uptake experiment measured by confocal layer scan microscopy (CLSM). Meanwhile, multilayered iHCEC was used to simulate the barrier of corneal epithelial cells for in vivo preocular retention capacity study, which suggested that BH-Mt/CS NPs could prolong the retention time in comparison with BH solution. The ocular pharmacokinetics studied by microdialysis sampling technique showed that AUC0−t and MRT0−t of BH-Mt/CS NPs were 1.99-fold and 1.75-fold higher than those of BH solution, indicating higher bioavailability. Moreover, the study of blood drug concentration, few researchers have reported, showed that low level drug could enter into blood, suggesting lower systematic side effect. Importantly, pharmacodynamics studies suggested that BH-Mt/CS NPs could make a significant decreased intraocular pressure on glaucomatous rabbits. Conclusion Inspired by these advance of montmorillonite/chitosan nanoparticles, we envision that the BH-Mt/CS NPs will be a potential carrier for BH, opening up the possible applications in glaucoma therapy.

Metabolic heterogeneity and the tumor immunosuppressive microenvironment (TIME) of triple-negative breast cancer (TNBC) hinder therapeutic effectiveness. Although emerging metabolic therapy and immunotherapy show promise, they are limited by off-target effects and immune escape. Here, a redox-activatable, sequentially-releasing nanoparticle (AMANC@M) for tumor-targeted delivery of anticancer agents and CRISPR/Cas9 has been developed. AMANC@M can reverse the TIME through dual metabolic inhibition, thereby enhancing TNBC therapy. AMANC@M demonstrates excellent biosafety and targets tumors precisely through biomimetic hybrid membrane-mediated homologous homing and the enhanced permeability and retention (EPR) effect. Once internalized into tumor cells, the CRISPR/Cas9 system (“energy nanolock”) is released through glutathione (GSH) cleavage and effectively knocks down the expression of lactate dehydrogenase A (LDHA) to suppress glycolysis. After peeling off of the gene editing shell, a newly synthesized targeted drug, CPI-Z2 (“nutrihijacker” and “energy nanolock”), is released in a controlled manner to block the mitochondrial tricarboxylic acid (TCA) cycle. Nitric oxide (NO) produced from loaded L-arginine enhances the efficiency of CPI-Z2 and reduces drug resistance. Combined with NO therapy, both blockades of nutrients and energy production transform the hypoxia and acidic TIME into an immunocompetent tumor microenvironment (TME) for tumor elimination. Furthermore, AMANC@M offers capabilities for photothermal (PT) therapy and provides clear imaging through PT, photoacoustic (PA), or computed tomography (CT) signals in tumor tissue. Thus, this study provides a new and promising sequentially stimuli-responsive targeting strategy for nanoparticle development, making it a potential treatment candidate for TNBC and other tumors.

An effective approach is presented for designing and tailoring diffusion-controlled systems for targeted release rate profiles. The diffusion-controlled system consists of PBA networks with precisely controlled crosslink densities via RAFT copolymerization of BA with ethylene glycol dimethacrylate, which gives a desired diffusivity. One-dimensional releases in the crosslinked PBA matrix (CPM) slabs are evaluated with a hydrophobic dye. Fick's second law is used to model the transient mass transfers in the CPMs with a mixed Newton-Tikhonov regularization method for determination of their diffusion coefficients. A two-layer CPM (CPM-2) with controlled crosslink densities is constructed. The dye release results from the CPM-2 agreed well with theoretical predictions.

A lignin-based polyurethane coating with self-healing ability was developed. • Dynamic disulfide bonds reduced coating porosity and improved toughness. • Nutrient-release period of controlled-release fertilizers was extended by 20 days. • The coating maintained release performance even after damage via self-repair. • Molecular simulations revealed optimal self-healing at a modifier content of 4.37%. Lignin is the second most abundant biomass resource in the world; it contains abundant active groups and has great application potential as a coating in controlled-release fertilizers (CRFs). However, lignin-based coatings are porous and vulnerable to damage. Herein, dynamic disulfide bonds were introduced into a lignin-based polyurethane network, increasing the interpenetrating crosslinking density of the network and reducing its porosity, which improved the damage resistance and self-healing performance of the coating. Specifically, the porosity of the lignin-based coating was reduced by 10.98% and the controlled-release period of encapsulated fertilizer was extended by 20 days (from 32 to 52 days) after disulfide-bond introduction. After the wear test, the lignin-based coating without disulfide bonds was almost completely damaged and lost its controlled-release capability, whereas the lignin-based coating with disulfide bonds remained intact, showing release behavior comparable to that of the unworn one. Additionally, cut lignin-based coating with disulfide bonds showed ∼12 days longer nutrient release than cut lignin-based CRFs. Finally, correlation between the self-healing modifier content, interfacial adhesion energy, and self-healing efficiency was investigated using molecular simulations. At a modifier content of 4.37%, the interfacial adhesion energy between the coating interfaces was 209.25 kcal/mol, and the repair efficiency reached the maximum value (44.16%). This study delivers useful theoretical insights into the high-value utilization of lignin and the creation of novel bio-based CRFs. Halloysite nanotubes (HNTs) loaded with the corrosion inhibitor benzotriazole (BTA) have been utilized to enhance the corrosion resistance of epoxy coatings. To improve BTA loading capacity, the HNTs were thermally treated and modified with Zn 2+ ions. Furthermore, to control the release of BTA, Zn 2+ ions were also used to form Zn–BTA complexes at the tube ends, acting as end stoppers. Thermogravimetric analysis showed that the combination of heat treatment and Zn 2+ modification significantly increased the BTA loading capacity of HNTs, reaching up to 15.5 wt%. UV–Vis spectroscopy demonstrated that, under neutral conditions, the additive with Zn 2+ end stoppers exhibited a much slower BTA release rate compared to the non-end-stoppered system, while the release rate significantly increased in acidic environments. The corrosion protection performance of epoxy coatings containing the BTA-loaded, Zn 2+ -modified HNTs was evaluated using electrochemical methods and salt spray testing. Results indicated that a 3 wt% additive loading provided the best corrosion resistance, significantly outperforming conventional epoxy coatings. Additionally, salt spray tests on commercial zinc-rich epoxy coatings with scribed defects revealed that the inclusion of the BTA-loaded, Zn 2+ -modified HNTs imparted active corrosion inhibition. These findings suggest that the prepared benzotriazole-loaded zinc ion-modified HNTs are a promising candidate for incorporation into epoxy coatings to improve the anticorrosion performance of carbon steel.

Electrostatics governs the formation of supported lipid bilayers on silica hollow nanospheres • Lipid coating passivates the particle surface and enables non-adsorptive encapsulation of hydrophilic cargo • The mesoporous silica shell acts as a secondary diffusion barrier, exhibiting hydrated-ionic-radius-dependent salt release Hollow silica nanospheres (SHNs) coated with a supported lipid bilayer (SLB) combine the high loading capacity of a hollow interior with the biocompatibility and tunable surface properties of a lipid membrane, making them promising for controlled delivery applications. Here, we systematically investigate the formation of SLBs on SHNs of varying diameters using neutral, positively charged, and negatively charged liposomes. We demonstrate that SLB formation requires sufficient ionic strength to overcome electrostatic repulsion between the negatively charged silica surface and liposomes of like charge. In contrast, positively charged liposomes form SLBs irrespective of salt concentration due to electrostatic attraction. Using an aggregation-based titration method, we quantify surface coverage and reveal that smaller SHNs are slightly more favorable for surface bilayer assembly. Notably, the lipid coating effectively passivates the silica surface, imparting exceptional colloidal stability. Beyond surface functionalization, we explore the encapsulation and release properties using alkali metal chlorides as non-adsorbing model cargos. The hollow cavity is readily accessible for loading, and the mesoporous silica shell acts as a secondary diffusion barrier, leading to release kinetics that scale with the hydrated radius of the cation. These findings establish design principles for lipid-coated SHNs as tunable carriers for hydrophilic substances with controlled release capabilities. In this work, a castor oil-based
polyurethane-based controlled-release
urea modified with dehydroabietic acid-grafted chitosan was prepared,
overcoming the poor hydrophobicity of traditional coatings. Dehydroabietic
acid-grafted chitosan (HNCSDA) was synthesized and incorporated into
castor oil-based polyurethane to fabricate modified coated fertilizers.
The chemical structure, micromorphology, thermal stability, hydrophobicity,
nitrogen release, phytotoxicity and antifungal performance of the
coatings were systematically characterized. HNCSDA improved coating
compactness and continuity, reduced structural defects and formed
an effective diffusion barrier. Increasing the HNCSDA dosage from
0% to 20% increased the hydrophobicity and thermal stability of the
coating; the water contact angle increased from 80.6 ° to 116.7 ° ,
and the release longevity increased by 15.54%. In the absence of maize
phytotoxicity and effectiveness against Botrytis cinerea and Rhizoctonia solani , the modified coatings increase
seedling growth, offering a feasible route to eco-friendly biobased
slow-release antifungal fertilizers for sustainable agriculture.

SA@BC@CTD hydrogel microspheres were successfully prepared. • High drug loading (16.15%) and encapsulation efficiency (91.68%) were achieved. • The formulation exhibited environmentally responsive controlled release. • Long-term pest control effectiveness was maintained for up to 60 days. • Reduced and delayed acute toxicity to earthworms compared with conventional CTD was observed. Sugarcane ( Saccharum officinarum L.) constitutes a major sugar crop and renewable bioenergy feedstock that plays a vital role in shaping the global agricultural economy and energy systems. However, sugarcane production is significantly threatened by pest infestations, which reduce yield and long-term sustainability levels. Although chemical pesticides are widely employed for pest control, their excessive use has caused significant environmental concerns and potential risks to human and ecological health. To address these challenges, nanobiochar (BC)-based clothianidin (CTD) composite hydrogel microspheres (SA@BC@CTD) were developed via the use of sodium alginate (SA) as the matrix and BC as a functional carrier via Ca²⁺ crosslinking. The release behavior, insecticidal performance, crop safety, and environmental risks of the prepared microspheres were comprehensively characterized through laboratory, pot, and field experiments. Compared with SA@CTD, SA@BC@CTD exhibited a stable three-dimensional porous structure, with a drug loading content of 16.15 ± 0.53% and an encapsulation efficiency of 91.68 ± 1.72%, representing increases of 87.40% and 16.70%, respectively. CTD release was significantly affected by pH and ionic strength, with optimal controlled-release behavior under neutral conditions (pH of 7) and high ionic strength (1 mol/L NaCl), with the duration of sustained release exceeding 96 h. The results of indoor bioassays demonstrated that compared with SA@CTD, SA@BC@CTD exhibited significantly increased concentration-dependent insecticidal activity against Spodoptera frugiperda Smith (third-instar larvae) and Chilo sacchariphagus Bojer (second-instar larvae). The results of pot experiments demonstrated that the application of SA@BC@CTD significantly enhanced sugarcane growth (plant height: 134.05 ± 24.17 cm; fresh weight: 431.35 ± 46.63 g at 60 d) while maintaining high insecticidal effectiveness (mortality rates of 35.12 ± 3.26% for S. frugiperda and 37.64 ± 3.52% for C. sacchariphagus at 60 d). This effect could be attributed to the sustained release of CTD and its enhanced translocation to aerial tissues, as indicated by a leaf transfer coefficient of 0.33 ± 0.02 at 60 d. Field trials confirmed that the use of SA@BC@CTD provided long-term control of dead sugarcane heart (81.59 ± 4.22%) at 60 d, thereby significantly outperforming conventional CTD granules. Moreover, acute toxicity tests using Eisenia fetida Savigny showed reduced toxicity compared with conventional CTD. In conclusion, this study provides a feasible strategy for sustained-release pesticide formulations with prolonged efficacy, enabling reduced pesticide use and sustainable sugarcane cultivation. In this study, the preparation of hydrogel beads through electrostatic interactions between Ι-carrageenan (IC) and ε-polylysine (ε-PL) using a dropwise method is reported. As the concentration of ε-PL increased, the degree of swelling of the hydrogel beads decreased. FTIR and XRD analyses revealed shifts in the absorption peaks, suggesting the formation of hydrogen bonding, electrostatic interactions in the hydrogel beads. TGA showed that the mass loss of the IC/ε-PL hydrogel beads was significantly lower compared to that of pure ε-PL. These results indicate that the IC component has a protective stabilizing effect on the hydrogel structure. ε-PL concentration of 2% yielded the highest encapsulation efficiency of curcumin (Cur) in the hydrogel beads. In vitro release studies showed that the hydrogel beads sustained the release of Cur and the release kinetics followed the first-order and Hixson-Crowell models. Collectively, these findings demonstrate that these hydrogel beads have the potential to be effective carriers of hydrophobic substances. Graphical

Organo-layered double hydroxides (OLDHs) are widely used as slow-release carriers for drugs but are rarely used in controlled drug release. To develop slow-release substrates of pretilachlor with excellent controlled release properties, OLDH hybrids were prepared by intercalating different masses of sodium dodecyl sulfate (SDS) into MgAl-LDHs. The underlying structural characteristics were analyzed by X-ray diffraction (XRD), Fourier Transform Infrared Spectroscopy (FT-IR), scanning electron microscopy (SEM), and contact angle measurements. Also, the adsorption capacity and release properties of OLDHs were explored by batch adsorption isotherms, adsorption kinetics, and release kinetics to reveal the structural effects of OLDHs on the release of pretilachlor. The results showed that the release of pretilachlor from OLDHs was dominated by Fickian diffusion and was closely related to the adsorption capacity and permeability of the matrix. The intercalation of SDS enhanced the hydrophobicity of LDHs, resulting in a higher adsorption capacity for pretilachlor and a lower permeability. Furthermore, as the SDS loading amount increased from 0.5 to 4.0 g, the adsorption capacity of the OLDHs was further enhanced (kF value increased from 2.52 to 12.83), while the permeability (d001/foc value) reversed, decreasing from 0.21 to 0.09. The release kinetics demonstrated that the release rate of pretilachlor from OLDHs was significantly lower compared to pure LDHs and showed a continuous decrease with increasing SDS loading amounts. The release time of 50% (T50) pretilachlor from OLDHs with SDS loading amounts of 25%, 50%, 100%, and 200% of the anion exchange capacity was 4.78, 8.28, 25.4, and 45.34 times longer than the release time from pure LDHs. This displayed a strong curvilinear relationship with the adsorption rate constant kF and permeability d001/foc and a good linear relationship with the added amount of SDS. These findings highlight the potential of OLDHs in preparing pretilachlor controlled release formulations.

Mesoporous silica microspheres (MSMs) possess poor biocompatibility . This study focuses on integrating MSMs with polymers to obtain hybrid materials with superior performance compared to the individual components and responsive release in specific environments. The synthesized MSMs were aminated, and subsequently, soybean hull polysaccharide (SHPs) was modified onto MSMs-NH 2 to produce MSMs-NH 2 @SHPs nanoparticles . The encapsulation rate, loading rate of curcumin (Cur), and in vitro release behavior were investigated. Results indicated that the encapsulation efficiency of Cur by MSMs-NH 2 @SHPs nanoparticles reached 75.58%, 6.95 times that of MSMs-NH 2 with a load capacity of 35.12%. It is noteworthy that these nanoparticles exhibit pH-responsive release capacity in vitro. The cumulative release rate of the three nanoparticles at pH 5.0 was higher than that at pH 7.4. MSMs-NH 2 @SHPs had a cumulative release rate of 56.55% at pH 7.4, increasing to 76.21% at pH 5.0. In vitro experiments have shown that MSMs-based nanoparticles have high delivery efficiency and can achieve pH-sensitive drug release, with a high release rate in a slightly acidic acid, highlighting the potential for controlled release of Cur.

Fish oil faces challenges like oxidation, low bioavailability, and poor sensory properties. While microencapsulation improves stability, traditional methods suffer from poor monodispersity, core damage, and limited wall material control. In this study, the plug-and-play microfluidic system overcame these limitations by precisely controlling emulsion formation and bypassing charge-matching requirements. Based on microfluidic technology, a plug-and-play microfluidic chip (single-channel and three-parallel channel) was used to control preparation (coefficient of variation < 5 %, shell thickness from 163 to 592 μm, core number from 1 to 5) and scaled-up production of fish oil microcapsules. A multi-dimensional wall material screening platform was developed via ionotropic crosslinking, using sodium alginate as the network skeleton and integrating gum arabic (GA) for emulsifying stability and low-methoxyl pectin (LM) for pH responsiveness. The results showed that the ternary composite wall material system (TCWMS) exhibited lower peroxide value (POV) and Malondialdehyde (MDA) values, and strong hydrogen bonding interactions. The multi-dimensional wall material screening platform and plug-and-play microfluidic chip were built to enable stable, controllable industrial preparation of monodisperse oily microcapsules.
