Comparison of intestinal absorption of soybean protein isolate-, glutenin- and peanut protein isolate-bound Nε-(carboxymethyl) lysine after in vitro gastrointestinal digestion

Shuqi Hao,Mengyu Ye,Na Li,Zeyu Lu,Wei Quan,Huaide Xu,Mei Li

Advanced glycation end products (AGEs), a heterogeneous compound existed in processed foods, are related to chronic diseases when they are accumulated excessively in human organs. Protein-bound N ε -(carboxymethyl) lysine (CML) as a typical AGE, is widely determined to evaluate AGEs level in foods and in vivo . This study investigated the intestinal absorption of three protein-bound CML originated from main food raw materials (soybean, wheat and peanut). After in vitro gastrointestinal digestion, the three protein-bound CML digests were ultrafiltered and divided into four fractions: less than 1 kDa, between 1 and 3 kDa, between 3 and 5 kDa, greater than 5 kDa. Caco-2 cell monolayer model was further used to evaluate the intestinal absorption of these components. Results showed that the absorption rates of soybean protein isolate (SPI)-, glutenin (Glu)-, peanut protein isolate (PPI)-bound CML were 30.18%, 31.57% and 29.5%, respectively. The absorption rates of components with MW less than 5 kDa accounted for 19.91% (SPI-bound CML), 22.59% (Glu-bound CML), 23.64% (PPI-bound CML), respectively, and these samples were absorbed by paracellular route, transcytosis route and active route via PepT-1. Taken together, these findings demonstrated that all three protein-bound CML digests with different MW can be absorbed in diverse absorption pathways by Caco-2 cell monolayer model. This research provided a theoretical basis for scientific evaluation of digestion and absorption of AGEs in food.

Photosensitive poly(o-nitrobenzyloxycarbonyl-L-lysine)-b-PEO polypeptide copolymers: synthesis, multiple self-assembly behaviors, and the photo/pH-thermo-sensitive hydrogels

Pan Li,Jiacheng Zhang,Chang-Ming Dong

A photoresponsive poly(o-nitrobenzyloxycarbonyl-L-lysine)-b-PEO (PNBL-b-PEO) polypeptide copolymer was synthesized and its photosensitive cleavage and self-assembly were monitored by means of UV-vis spectroscopy, 1H NMR, DLS, and TEM. Three kinds of dual-sensitive (i.e., photo/pH-thermo) polypeptide normal and reverse micellar hydrogels were then fabricated by the physical cross-linking of the self-assembled micelles. Utilizing the inclusion complexation between α-cyclodextrin (α-CD) and PEO of the normal micelles of PNBL-b-PEO or polylysine-b-PEO, photo/pH-thermo-sensitive normal micellar hydrogels were fabricated; in contrast, the reverse micelles of polylysine-b-PEO@α-CD self-assembled at pH = 3 and then were transformed into a pH-thermo-sensitive reverse micellar hydrogel at pH = 10. The multiple self-assembly and hydrogel properties were thoroughly characterized by means of UV-vis spectroscopy, circular dichroism (CD), wide angle X-ray diffraction (WAXD), DLS, zeta potential, rheology, TEM, and SEM. Notably, these dual-sensitive polypeptide hydrogels with a higher mechanical modulus (10–105 kPa) were facilely constructed and the mechanical modulus can be further tuned by varying the UV-irradiation time, the amount of α-CD added, and the copolymer concentration. Consequently, this work provides a versatile platform for constructing polypeptide copolymer hydrogels with tunable mechanical and stimuli-sensitive properties.

Poly-l-lysine modified MOF nanoparticles with pH/ROS sensitive CIP release and CUR triggered photodynamic therapy against drug-resistant bacterial infection

Ya-li Xiang,De-yu Tang,Ling-ling Yan,Luo-lin Deng,Xing-han Wang,Xin-yang Liu,Qing-han Zhou

The challenge of drug resistance in bacteria caused by the over use of biotics is increasing during the therapy process, which has attracted great attentions of the clinicians and scientists around the world. Recently, photodynamic therapy (PDT) triggered by photosensitizer (PS) has become a promising treatment method because of its high efficacy, easy operation, and low side effect. Herein, the poly- l -lysine (PLL) modified metal-organic framework (MOF) nanoparticles, ZIF/PLL-CIP/CUR, were synthesized to allow both reactive oxygen species (ROS) responsive drug release and photodynamic effect for synergistic therapy against drug resistant bacterial infections. The PLL was modified on the shell of the zeolite imidazole framework (ZIF) by the ROS-responsive thioketal linker for controllable CIP release. CUR were encapsulated in ZIF as the photosensitizer for blue light mediated photodynamic effect to produce singlet oxygen ( 1 O 2 ) and superoxide anion radical ( O 2 − ) for efficient inhibition towards methicillin-resistant Staphylococcus aureus ( MRSA ). The charge conversion from negative charge (−4.6 mV) to positive charge (2.6 mV) was observed at pH 7.4 and pH 5.5, and 70.9 % CIP was found released at pH 5.5 in the presence of H 2 O 2 , which suggests the good biosafety at physiological pH and ROS-responsive drug release of the as-prepared nanoparticle in the bacterial microenvironment. The as-prepared nanoparticles could effectively kill MRSA and disrupt bacterial biofilm by combination of chemo- and photodynamic therapy. In mice model, the as-prepared nanoparticles exhibited excellent biosafety and synergistic effect with 98.81 % healing rate in treatment of MRSA infection, which is considered as a promising candidate in combating drug resistant bacterial infection.

Stability enhancement of pickering emulsions based on κ-carrageenan microgel: Synergistic effect of l-lysine and potassium ions at low ionic strength

Xuemin Wang,Chaofan Guo,Jing Liu,Xianchao Feng,Jiliang Zhang,Jianxin Cao,Xuejiao Wang,Tingting Feng

The study utilized the synergistic effect of l -lysine and K + to improve the stability of Pickering emulsions based on κ -carrageenan microgels. The gel strength of κ -carrageenan gels, the zeta potential and particle size revealed that the microgel particles prepared with l -lysine and K + had the potential to be Pickering particles. l -lysine formed electrostatic interactions and hydrogen bonds with κ -carrageenan. Structural modifications of microgel particles by l -lysine (0.05%–0.10%) and low K + strength (≤40 mmol/L) promoted the formation of a dense interface layer at the oil-water droplet surface to prevent the accumulation of oil droplets and further significantly enhance the appearance and centrifugal stability of Pickering emulsions. The microstructure and rheological analysis showed that K + at low ionic strength, in cooperation with l -lysine, formed a more tightly structured layer between the microgel particles wrapped on the surface of oil droplets, thereby enhancing the stability of emulsions. In conclusion, these Pickering emulsions with prolonged stability are expected to be suitable healthy animal fat substitutes for use in food applications.

Waterborne polyurethane synthesized for leather with excellent wear and hydrolysis resistance enabled by bio-based poly(trimethylene carbonate) and l-lysine diisocyanate

Mengchen Xue,Xiang Rao,Weihu Li,Jinghua Du,Wenhe Guo,Wangqin Zhou,Xiaoyu Dong,Le Wu,Guobing Zhang,Yunsheng Ding

A wide application of bio-based waterborne polyurethanes (BWPU) in the synthetic leather is the main direction in the future, however, it has been a great challenge for its development because of the poor mechanical properties and water resistance of BWPU. To address this issue, bio-based poly(trimethylene carbonate) diols (PTMC-OH) with a theoretical molecular weight of 2000 g/mol was prepared via ring-opening polymerization of trimethylene carbonate and 1,4-butanediol as an initiator. PTMC-OH, bio-based l -lysine diisocyanate as building blocks, and trimethylolpropane as a crosslinking agent were employed to prepare a series of linear and crosslinked bio-based waterborne polyurethanes (LBWPU and CBWPU). Remarkably, CBWPU-1 % film with crosslinked structure had high tensile strength and toughness shown as 41.5 MPa and 131 MJ·m −3 , respectively, while it also showed excellent hydrolysis resistance with the ignorable degradation in aqueous solution after 35 days since the weight loss of about 1.4 wt%. In addition, the synthetic leather coating made from CBWPU-1 % exhibited exceptional wear resistance, enduring 50,000 rubs without damages on the CBWPU coating. This work presents a facile and efficient method for the synthesis of BWPU and applying them to manufacture the bio-based synthetic leather.

Effects of amino acids methionine, lysine, and taurine on feed intake and mRNA expression of appetite-related neuropeptides in layer-type chicks

Elham Ghashghaei,Minghui Wang,Ahmed Mijiyawa,Hai Lin

Amino acids in the brain modulate eating behavior and energy balance. The aim of the present study was to investigate the effects of DL-methionine (DL-Met), L-lysine monohydrochloride (L-Lys-HCl), and taurine (Tau) on feed intake and the mRNA expression levels of appetite-related hypothalamic neuropeptides in chicks. DL-Met, L-Lys-HCl, or Tau was intracerebroventricularly (ICV) administered to 5-day-old layer-type chicks, and the feed intake was recorded until 1 h post-injection. Quantitative PCR was performed to determine the hypothalamic mRNA expression levels of neuropeptide Y ( NPY ) and its receptors, agouti-related protein ( AgRP ), pro-opiomelanocortin ( POMC ), cocaine- and amphetamine-regulated transcript ( CART ), and melanocortin receptors. Our results indicated that injection time and treatment have a significant effect on food intake. Food intake decreased significantly at 30 and 60 minutes after ICV administration of DL-Met (1.678 μmol), L-Lys-HCl (6.856 μmol), and Tau (5.006 μmol) compared to the control group ( P > 0.05). On the contrary, a high dose of each amino acid did not alter feed intake ( P > 0.05). The result showed that ICV injection of DL-Met (1.678 μmol) significantly decreased the mRNA expressions of NPY , NPY4R , and NPY6R ( P < 0.05), but had no significant effect on the mRNA expressions of NPY1R, NPY5R, and AgRP in the hypothalamus ( P > 0.05). Similarly, ICV injection of L-Lys-HCl (6.856 μmol) and Tau (5.006 μmol) significantly decreased mRNA levels of NPY, NPY4R, and NPY6R ( P < 0.05), without affecting the mRNA expressions of NPY1R, NPY5R, and AgRP ( P > 0.05). DL-Met had no effect on anorexigenic gene expression ( P > 0.05). L-Lys-HCl upregulated MC1R ( P < 0.05), and Tau upregulated POMC and MC3R ( P < 0.05), and both had no influence on CART, MC4R , or MC5R ( P > 0.05). The result suggests that both orexigenic and anorexigenic neuropeptide genes are involved in the effects of DL-Met, L-Lys-HCl, and Tau. The effect of DL-Met, L-Lys-HCl, and Tau acts in a dose-dependent manner in the hypothalamus to influence feed intake.

Injectable supramolecular hydrogel formed from α-cyclodextrin and PEGylated arginine-functionalized poly(l-lysine) dendron for sustained MMP-9 shRNA plasmid delivery

Qianming Lin,Yumeng Yang,Qian Hu,Zhong Guo,Tao Liu,Jiake Xu,Jianping Wu,Thomas Brett Kirk,Dong Ma,Wei Xue

Hydrogels have attracted much attention in cancer therapy and tissue engineering due to their sustained gene delivery ability. To obtain an injectable and high-efficiency gene delivery hydrogel, methoxypolyethylene glycol (MPEG) was used to conjugate with the arginine-functionalized poly( l -lysine) dendron (PLLD-Arg) by click reaction, and then the synthesized MPEG-PLLD-Arg interacted with α-cyclodextrin (α-CD) to form the supramolecular hydrogel by the host-guest interaction. The gelation dynamics, hydrogel strength and shear viscosity could be modulated by α-CD content in the hydrogel. MPEG-PLLD-Arg was confirmed to bind and deliver gene effectively, and its gene transfection efficiency was significantly higher than PEI-25k under its optimized condition. After gelation, MMP-9 shRNA plasmid (pMMP-9) could be encapsulated into the hydrogel matrix in situ and be released from the hydrogels sustainedly, as the release rate was dependent on α-CD content. The released MPEG-PLLD-Arg/pMMP-9 complex still showed better transfection efficiency than PEI-25k and induced sustained tumor cell apoptosis. Also, in vivo assays indicated that this pMMP-9-loaded supramolecular hydrogel could result in the sustained tumor growth inhibition meanwhile showed good biocompatibility. As an injectable, sustained and high-efficiency gene delivery system , this supramolecular hydrogel is a promising candidate for long-term gene therapy. Statement of Significance To realize the sustained gene delivery for gene therapy, a supramolecular hydrogel with high-efficiency gene delivery ability was prepared through the host-guest interaction between α-cyclodextrin and PEGylated arginine-functionalized poly( l -lysine) dendron. The obtained hydrogel was injectable and biocompatible with adjustable physicochemical property. More importantly, the hydrogel showed the high-efficiency and sustained gene transfection to our used cells, better than PEI-25k. The supramolecular hydrogel resulted in the sustained tumor growth inhibition meanwhile keep good biocompatibility. As an injectable, sustained and high-efficiency gene delivery system, this supramolecular hydrogel is a promising candidate in long-term gene therapy and tissue engineering.

Coaxial electrospun PVA/PCL nanofibers with dual release of tea polyphenols and ε-poly (L-lysine) as antioxidant and antibacterial wound dressing materials

Xingzi Lan,Yurong Liu,Yaqi Wang,Feng Tian,Xiaomin Miao,Han Wang,Yadong Tang

Preparing wound dressing with dual-delivery of antioxidant and antibacterial agents is highly desirable in clinical wound treatment. Herein, a series of coaxial nanofiber membranes loaded with antioxidant tea polyphenols (TP) in the core and antibacterial ε-poly (L-lysine) (ε-PL) in the shell layer were successfully fabricated by coaxial electrospinning. The physicochemical characterizations by transmission electron microscopy , inverted fluorescence microscopy and fourier transform infrared spectroscopy confirmed the formation of core-shell structure. The results of in vitro drug release indicated that ε-PL exhibited a fast release profile while TP released in a sustained manner, which is favorable to the achievement of quick bacteria inhibition in the initial phase as well as long-term antioxidant activity during wound healing. The antioxidant activity of coaxial nanofibers was found to be increased with the increment of TP content and incubation time . The antibacterial assays against Escherichia coli and Staphylococcus aureus demonstrated that the incorporation of ε-PL in the coaxial nanofibers led to strong antibacterial activity . Additionally, all the coaxial nanofibers possessed good cytocompatibility. Therefore, the prepared coaxial nanofibers simultaneously incorporated with ε-PL and TP are promising as potential wound dressing materials.

Poly(l-diaminopropionic acid), a novel non-proteinic amino acid oligomer co-produced with poly(ε-l-lysine) by Streptomyces albulus PD-1

Xia Jun,Xu Hong,Feng Xiaohai,Xu Zhaoxian,Chi Bo

Poly(ε- l -lysine) (ε-PL) producer strain Streptomyces albulus PD-1 secreted a novel polymeric substance into its culture broth along with ε-PL. The polymeric substance was purified to homogeneity and identified. Matrix-assisted laser desorption ionization-time of flight mass spectrometry and nuclear magnetic resonance spectroscopy as well as other analytical techniques revealed that the substance was poly( l -diaminopropionic acid) (PDAP). PDAP is an l -α,β-diaminopropionic acid oligomer linking between amino and carboxylic acid functional groups. The molecular weight of PDAP ranged from 500 to 1500 Da, and no co-polymers composed of l -diaminopropionic acid and l -lysine were present in the culture broth. Compared with ε-PL, PDAP exhibited stronger inhibitory activities against yeasts but weaker activities against bacteria. ε-PL and PDAP co-production was also investigated. Both ε-PL and PDAP were synthesized during the stationary phase of growth, and the final ε-PL and PDAP concentration reached 21.7 and 4.8 g L -1 , respectively, in fed-batch fermentation. Citric acid feeding resulted in a maximum ε-PL concentration of 26.1 g L -1 and a decrease in the final concentration of PDAP to 3.8 g L -1 . No studies on ε-PL and PDAP co-production in Streptomyces albulus have been reported previously, and inhibition of by-products such as PDAP is potentially useful in ε-PL production.

Synthesis and properties of non-cytotoxic poly(l-malic acid acetate-co-l-lysine ester)s with thermo-sensitivity as hydrophobic drug nanocarrier

Ying Yue,Zhihui Fan,Chang Zheng,Lan Ding,Huiying Wang,Miao Zhang,Kuilin Deng

In this investigation, a group of poly( l -malic acid acetate- co - l -lysine ester)s (PMALs) with excellent thermo-sensitivity and non-cytotoxicity were prepared by an optimized synthetic route from natural l -malic acid and l -lysine. The structure and properties of PMALs including monomers were systematically characterized by FTIR, 1 H NMR, UV, gel permeation chromatograph, scanning electron microscope, contact angle measurement, cell counting kit assess (CCK-8), and confocal laser scanning microscopy (CLSM). Three PMALs show a reversible lower critical solution temperature of 8–36 °C depending on their chemical structure. The contact angle measurement revealed a considerable discrepancy in the hydrophilicity/hydrophobicity of PMALs and further influence on their thermo-sensitivity. The viability of HeLa cells exposed to 0.2–100 μg/mL PMALs solution was found to be in a range 80–103% after 24, 48, and 72 h of incubation, indicating no cytotoxicity. Moreover, a spherical nanocarrier with core-shell structure was facilely fabricated via the thermo-sensitivity of PMALs and hydrophobicity of drug. CLSM observations manifested that the hydrophobic-curcumin-enwrapped nanocarriers can clearly internalize into the cellular inside. The sustained release of curcumin from nanocarriers in vitro provided a possibility of depressing fast hydrolytic degradation at physiological pH or other side-effects. © 2017 Wiley Periodicals, Inc. Improving the Quality of Low-Salt Beef Myofibrillar Protein Gels with L-Lysine and Konjac Glucomannan: Water Retention, Texture, and Protein Structural Changes

Xiuyun Guo,Jinsheng Yang,Chao Fu,Jiangpeng Yao,Zhikun Yang,Xiangren Meng

Reducing sodium in meat products is nutritionally desirable, but salt reduction often weakens myofibrillar protein gelation and reduces texture quality and water retention. This study investigated the effects of L-lysine (Lys) and konjac glucomannan (KGM) on the physicochemical properties, gel characteristics, and structural changes of beef myofibrillar protein (MP) gels under low-salt conditions. The results indicated that reducing NaCl from 0.6 to 0.2 M decreased water-holding capacity (WHC), increased cooking loss, and produced a loose gel network. Compared with the 0.2 M NaCl group, the combined Lys-KGM treatment increased WHC from 28.53% to 56.70% and reduced cooking loss to 22.19% (p< 0.05). Texture analysis showed that Lys-KGM increased hardness and springiness by 69.40% and 80.43%, respectively (p< 0.05). LF-NMR indicated a higher proportion of immobilized water and reduced water mobility in the combined treatment. Lys increased reactive sulfhydryl content and surface hydrophobicity, whereas KGM reduced surface hydrophobicity and enhanced water immobilization. Lys-KGM slightly but significantly decreased α-helix and increased β-sheet contents (p< 0.05), accompanied by changes in the relative contributions of intermolecular forces and a more continuous gel network. Molecular docking and molecular dynamics simulations provided supporting evidence for different interaction patterns between myosin and Lys/KGM. These results suggested that Lys and KGM might help maintain the quality of low-salt meat protein gels and provide a formulation basis for reduced-sodium meat products.

Classification of pulmonary inflammation stages and assessment of multicomponent drug intervention based on spatiotemporal imaging variations utilizing RhB-conjugated poly-L-lysine nanoparticles

Man Zhang,Shanshan Zhai,He Gao,Tong Sun,Kaixin Liu,Wenshuang Wang,Yuanyuan Hou,Gang Bai

Acute pulmonary inflammation, a major cause of morbidity and mortality, involves complex immune responses and increased vascular permeability. Conventional imaging techniques for assessing pneumonia and monitoring vascular leakage have limitations in spatial resolution and practicality. Rhodamine B-conjugated poly-L-lysine (RhB-PLL) self-assembled nanoparticles (NPs) were synthesized for targeting mitochondria and sensing vascular permeability. Dynamic spatiotemporal imaging analysis stratifies the progression of acute pneumonia into four distinct stages. Six phosphatidylcholine metabolites, exhibiting stage-dependent variations, were proposed as potential biomarkers to support RhB-PLL detection. Subsequently, Fuzheng Jiedu decoction (FZJD) was used as a model to validate the rationale of syndrome-specific treatment for pneumonia. Three simplified prescriptions derived from FZJD showed unique advantages at different stages. Xiaochaihu decoction (XCH) relieved inflammation and oxidative stress in mild to moderate cases; Sanren decoction (SR) reduced vascular leakage and prevented pulmonary edema in moderate to severe cases; Maxingshigan decoction (MXSG) improved mitochondrial function, prevented microcirculatory disorders in severe to critical cases. The combined prescription, FZJD, mitigated disease progression in mild to moderate cases and demonstrated the benefits of overall intervention. This conclusion was further substantiated by metabolomics cluster analysis. Real-time RhB-PLL multimodal imaging enables precise visualization and detection of inflammation progress, facilitates therapy assessment, and enhances the understanding of multicomponent drug combinations.
