Plant proteins are becoming increasingly popular as functional ingredients in foods because of their health, sustainability, and environmental benefits. The functional performance of plant proteins can often be improved by combining them with other food ingredients. For instance, they can be combined with polyphenols and metal ions to form surface active protein-polyphenol-iron complexes with biological activity. In this study, ternary complexes of pea protein isolate (PPI), epigallocatechin-3-gallate (EGCG) and iron (Fe 3+ ) was prepared using a simple molecular self-assembly method. The formation mechanism, characteristics, and functional properties of these complexes were then characterized. Our results showed that PPI, EGCG, and Fe 3+ could spontaneously combine to form nano-sheet networks held together by hydrogen bonds, phenolic hydroxyl coordination bonds, and electrostatic interactions. The addition of Fe 3+ increased the fraction of disordered secondary structures, reduced the thermal denaturation temperature, reduced the surface hydrophobicity, increased the particle size, and altered the surface potential of the proteins in a dose dependent manner. Optimizing the EGCG-to-Fe 3+ ratio in the complexes (1:2 w/w) led to the formation of protein composites with improved surface, antioxidant, and antibacterial activities. This study provided valuable insights into the interaction mechanisms of proteins, polyphenols, and metal ions in ternary complexes. These complexes may be used as multifunctional ingredients in plant-based foods because of their beneficial antioxidant, antimicrobial, nutritional, and emulsification properties.

In this study, a quantitative method based on fluorescein isothiocyanate (FITC)-labelledHizikia fusiformepolyphenol–polysaccharide complex (HPC) and its purified fractions (PC1, PC4) was used, and its pharmacokinetics and tissue distribution were investigated in mice. The results showed that the FITC-labelled method had good linearity (R2> 0.99), intra-day and inter-day precision (RSD, %) consistently lower than 15%, recovery (93.19–106.54%), and stability (RSD < 15%), which met the basic criteria for pharmacokinetic studies. The pharmacokinetic and tissue distribution results in mice after administration showed that all three sample groups could enter the blood circulation. and HPC-FITC had a longer half-life (T1/2: 26.92 ± 0.76 h) and mean retention time (MRT0–∞: 36.48 h) due to its larger molecular weight. The three groups of samples could be absorbed by the organism in a short time (0.5 h) mainly in the stomach and intestine; the samples could be detected in the urine after 2 h of administration indicating strong renal uptake, and faecal excretion reached its maximum at 12 h. The samples were also detected in the urine after 2 h of administration. This study provides some theoretical basis for the tissue distribution pattern of polyphenol–polysaccharide complex.

Polyphenol oxidase (PPO) is the culprit behind the browning of fruits and vegetables. Therefore, how to reduce the thermal deactivation temperature of PPO or use as few safe reagents as possible to inhibit enzymatic browning has practical significance. Mesoporous silica nanoparticles (MSNs) and multiwalled carbon nanotubes (MWCNTs) are stable and have high biosafety. In the present study, efficient PPO inhibitors were developed based on MSNs and MWCNTs. It is found that after modification with a very small amount of dodecyl trimethylammonium bromide (DTAB, ≥60 μg/mL), MSNs can significantly inhibit the activity of PPO although single MSNs and single DTAB show very limited effect on PPO activity. After modification with a very small amount of sodium dodecyl sulfate (SDS, 5.7–9.5 μg/mL), MWCNTs almost completely inactivate PPO. However, SDS@MSN and DTAB@MWCNT cannot decrease PPO activity significantly.

In this study, the curdlan-polyphenol complexes were constructed by a pH-driven method. The interaction between curdlan and various hydrophobic polyphenols (curcumin, quercetin, and chlorogenic acid) was investigated. Curdlan could self-assemble into particles for loading polyphenols through hydrogen bonding and hydrophobic interactions. The three polyphenols were embedded in curdlan in an amorphous state. The curdlan-curcumin complex showed the lowest viscoelasticity but exhibited the highest curcumin loading ability (34.04 ± 1.73 mg/g). However, the curdlan-chlorogenic acid complex emerged the opposite trend, indicating that the loading capacity was associated with the hydrophobicity of polyphenols. The antioxidant activity of curdlan significantly increased after combining with polyphenols, which could be maintained during in vitro simulated gastrointestinal digestion. In particular, the curdlan-quercetin complex exhibited the highest antioxidant activity and short-chain fatty acid concentration, which could influence gut microbiota composition by promoting the proliferation of Prevotella and inhibiting the growth of Escherichia_Shigella . In conclusion, the curdlan-polyphenol complexes prepared by an alcohol-free pH-driven method could effectively enhance the gastrointestinal stability of polyphenols as well as increase the antioxidant and prebiotic activities of curdlan, which could be applied as a functional ingredient to improve gut health.

Despite the use of surgical resection and chemotherapy in the clinical treatment of oral squamous cell carcinoma (OSCC), the 5-year survival rates of advanced patients are low. Therefore, more efficient strategies are urgently needed. Herein, a chemo/ferroptosis synergistic therapeutic system-DMEFe nanoparticles (NPs) is established for the treatment of OSCC. To create this system, the chemotherapeutic agent doxorubicin (DOX) was loaded into mesoporous silica nanoparticles and further coated with a pH-sensitive metal polyphenol (iron ion and epigallocatechin gallate). These nanoparticles displayed excellent pH-sensitive drug-control release properties, and the release ratio of DOX at pH 5.5 was twice as high than that at pH 7.4. Additionally, DMEF NPs were effectively taken up by the OSCC cell line SSC-25, which greatly impeded the proliferation of these cells. Notably, these nanoparticles increased the intracellular level of reactive oxygen species and effectively exhibited cytotoxity effects. The mechanistic results proved that DMEFe NPs regulated the expression of ferroptosis-related genes to induce ferroptosis of SSC-25 cells. Eventually, this chemo/ferroptosis therapeutic system exhibited remarkable antitumor effects and provided a novel strategy for the treatment of OSCC.

Diabetes-associated skin defects represent a significant global health challenge. While flap grafts have been a preferred treatment for soft-tissue injuries in diabetic patients, their survival is often compromised by impaired vascularization, infection, and the adverse diabetic pathological microenvironment. To address these limitations, a hybrid photo-crosslinkable hydrogel (HPC) integrated hemangioma stem cell-derived nanovesicle (HemV)-loaded dual-metal-polyphenol network (dMPN) (HemV@dMPN/HPC) is developed. HemVs, derived from highly vascularized infantile hemangioma tissues, play a key role in promoting cell proliferation and angiogenesis. The dMPN facilitates the gradual release of copper (Cu 2+ ) and magnesium ions (Mg 2+ ), stimulating angiogenesis and mitigating inflammation. The HPC further sustains ion release while preserving the therapeutic efficacy of HemVs. Moreover, both HPC and Cu 2+ act to confer antibacterial properties, further accelerating wound healing. This multifunctional HemV@dMPN/HPC platform offers a promising therapeutic strategy for treating large diabetic skin defects and can potentially improve flap graft survival.

Maintaining microbiota balance and enhancing the antioxidant performance of nanozyme-based probiotic systems are crucial for effective inflammatory bowel disease (IBD) therapy. Despite significant advancements, developing a green and safe coating technology that functionalizes probiotics with nanozymes while preserving the activity of both components remains a challenge. To address this, chitosan-modified epigallocatechin gallate (EGCG-CS, EC)is synthesized, leveraging the intrinsic adhesive and coordination properties of polyphenols to capture gold nanozymes (AuNPs), forming ECA complexes that enhance nanozyme activity. When coated onto Escherichia coli Nissle 1917 (EcN), the resulting ECA@EcN system effectively scavenged reactive oxygen species (ROS), improving probiotic viability and promoting colon accumulation. Mechanistically, ECA protected EcN by suppressing the activation of the Flagellar Assembly and Branched-Chain Amino Acid Synthesis pathways, ultimately alleviating inflammation and modulating intestinal microbial communities to relieve IBD symptoms. Given the biocompatibility of its components and the environmentally friendly assembly approach, this polyphenol-nanozyme-armored probiotic system represents a promising platform for IBD treatment.

Adhesives are widely used in everyday life; however, traditional polymer adhesives pose unavoidable hazards to the environment and living organisms due to their extensive use of toxic organic solvents, heavy reliance on fossil resources as synthetic raw materials, and lack of recyclability. Furthermore, they are incapable of maintaining a stable adhesion performance in harsh conditions. In this work, we present a facile yet powerful noncovalent bonding strategy for successfully constructing poly(glycidol-co-1,2-butylene oxide)/tannic acid supramolecular adhesives (PG-co-PBO/TA SAs) through hydrogen-bonding interactions between hyperbranched PG-co-PBO and natural TA. The SAs demonstrate an adhesion strength of 2.15 MPa at ambient temperature and maintain an adhesion strength of 1.53 MPa even at −196 °C. Moreover, it can be easily recycled through thermal treatment and retains stable adhesion strength even after undergoing multiple recycling cycles. The investigation of the structure–property relationship of SAs and molecular dynamics simulations has clarified that short alkyl chains of PG-co-PBO effectively protect the hydrogen-bonding networks from damage by repelling water at the adhesive interface. Consequently, SAs even maintain robust and long-lasting adhesion strength in acidic, alkaline, and seawater solutions. Importantly, unlike conventional adhesives, SAs do not employ organic solvents throughout the synthesis, bonding, and recycling processes. By designing functionalized polymers and utilizing polyphenolic compounds, this work provides a green and feasible strategy for the development of next-generation powerful supramolecular adhesives.

Nonhealing chronic wounds are intractable clinical complications of diabetes and are characterized by high protease activity, severe oxidative stress and sustained inflammatory response. In this case, the development of functional hydrogel dressings to modulate the immune microenvironment is a well-known strategy, where the precise stimuli-responsive and spatiotemporally controlled release of bioactive molecules remains a huge challenge. Herein, we developed responsive hydrogels with self-regulated bioactive molecule release based on the protease activity in diabetic wound sites, to serve as a smart immune microenvironment modulator for accelerating wound healing. The hydrogels were fabricated by grafting oxidized hyaluronic acid with epigallocatechin-3-gallate (EGCG) and gelatin methacryloyl (GelMA) under UV irradiation. Resveratrol nanoparticles were further loaded into the hydrogels before gelation to construct a polyphenol delivery system. The prepared hydrogels could achieve the on-demand release of polyphenol upon degradation by protease, as confirmed via degradation and polyphenol release experiments. The released polyphenol was demonstrated to have the capacity to effectively scavenge excessive free radicals, promote macrophage polarization, reduce proinflammatory factor (TNF-α) expression and augment anti-inflammatory factor (IL-10) expression in vitro. Additionally, in vivo rat wound healing model experiment results confirmed that these hydrogels promoted collagen deposition and granulation tissue regeneration, accelerating diabetic wound healing. Based on the protease-responsive degradation characteristic of the hydrogels and high protease activity in the diabetic wound microenvironment, hydrogels with exquisite polyphenol release controllability are promising candidates as dressings for diabetic wound management.

Tea polyphenol (TP) is used as a food additive to inhibit lipid peroxidation reactions and extend shelf life, but excessive intake can affect the human immune system. Therefore, developing effective TP detection is significant. Here, pyrene-based polyimide (Py-PI) was synthesized by one-step condensation copolymerization. This polyimide possessed excellent solubility and photoluminescence due to the introduction of pyrene-conjugated groups. It could be used as fluorescent probe for TP detection in solution. In addition, Py-PI fluorescent test paper with different morphology were prepared by electrospinning. As the concentration of the spinning solution was 15 wt%, the bead-like porous polymer electrospun membrane displayed excellent surface wettability which could make the water droplet completely penetrate into the sample within 6 min. The fluorescence enhancement phenomenon could be observed for 5 × 10 −6 mg/mL TP solution on Py-PI test paper. Furthermore, interference investigation of pH on test paper indicated that the sample exhibited a relatively stable fluorescence “turn-on” response in acidic conditions. This work provides a convenient and rapid detection of TP antioxidant in the future.

In this study, a novel dynamic polyphenol polymerization method for the preparation of nanofiltration membrane intercalations was proposed. In this method, TA, PEI and Al 3+ could successively form the first polyphenol-amine crosslink network and the second metal-phenol cross-linked network on the surface of the support layer. The dense crosslinked layer of TA-PEI-Al 3+ formed under dynamic conditions maintains high hydrophilicity compared to the crosslinked network formed under a single pH environment. Aluminum ions possess strong electronegativity since they have relatively high valence states and small ionic radius among transition metal ions. Therefore, this is the first time to incorporate Al 3+ into phenol-amine crosslinked networks to form the metal-phenol tri-complexes. The tri-complexes of TA and Al 3+ are more robust, promoting the coordination with more water molecules and forming more stable hydrophilic intermediate layers. When an interfacial polymerization reaction is performed on the intermediate layer, this hydrophilic intermediate layer can store aqueous-phase PIP and slow down the IP reaction rate through electrostatic interactions. This process helps to form a dense PA separation layer on the support layer surface. The prepared TFC-NF membranes can be used to separate dyes, antibiotics, and compound inorganic salts. The outstanding water permeance of the TFC-NF membrane was determined as 18.7 L/m 2 ·h·bar and the removal rate of mixed pollutants can reach 99 %. The firm intermediate layer enables the optimized modified membrane to maintain stable flux and high rejection rate in 72 h separation test. This modification method provides a useful reference for the preparation of high-performance nanofiltration membranes with plant polyphenols.

Global microbial spoilage highlights the pressing need for advanced packaging solutions. This study introduces a novel Cu-Chlorogenic acid nanoparticles (Cu-CGA NPs) packaging film system, which demonstrates exceptional photothermal antibacterial properties aimed at enhancing fruit preservation. The system utilizes a photoresponsive metal-polyphenol framework (Cu-CGA NPs) synthesized through a spontaneous chelation process under alkaline conditions, and seamlessly integrated with a carrageenan/gelatin (CG) film matrix. This integration capitalizes on the synergistic effects between the photothermal capabilities of Cu-CGA NPs and its inherent chemical antibacterial properties, resulting in the Cu-CGA 0.15 -CG film-where 0.15 denotes the proportion of Cu-CGA in the solution. This film demonstrates enhanced antibacterial efficacy, achieving 99% bacterial killing rates for both Escherichia coli and Staphylococcus aureus under brief near-infrared light (NIR) exposure. Additionally, the CGA component imparts significant antioxidant capacity to the Cu-CGA 0.15 -CG film, with a DPPH radical scavenging rate exceeding 60%. The inclusion of Cu-CGA NPs also enhanced the mechanical strength of Cu-CGA 0.15 -CG film to 25.8 MPa and improved the water contact angle (WCA) to 84.087°. These combined attributes result in a threefold extension of cherry tomato storage life, while maintaining product quality. Safety evaluations, including migration and cytotoxicity tests, confirm the suitability of the Cu-CGA 0.15 -CG film for food contact. Overall, the development of Cu-CGA NPs films represents a substantial advancement in food preservation technologies, offering robust protection for perishable items.
