Tryptophan (Trp) and tyrosine (Tyr) are the primary precursors of protein-like components in dissolved organic matter. Phenolic compounds are ubiquitous in aquatic environments and are considered the main electron donor in chromophoric dissolved organic matter (CDOM). Our results showed that Trp and Tyr (50 μM) enhanced the transformation of six monophenols (20 μM) with varying numbers of −CH3 and −OCH3 substituent groups by a factor of 1.0–1.8. The enhancement factor increased with the ratio of Trp (Tyr) to monophenols. In four different CDOM solutions (5 mg C/L, pH 8.0), a maximum enhancement factor of 3.2–6.7 was observed at a Trp/monophenol concentration ratio of 50. Conversely, monophenols greatly inhibited the transformation of Trp or Tyr. The enhancement factor decreased as the initial pH increased from 3.0 to 10.0. Additionally, the enhancement factor was not directly proportional to the oxidation potential of monophenol. We propose that the promotion effects are generated through the direct oxidation of monophenols by Trp (Tyr) radicals as well as through the reaction between Trp (Tyr) radicals and the one-electron reductant of CDOM.

An analytical methodology based on an O-[2-(methacryloyloxy)-ethylcarbamoyl]-10,11-dihydroquinidine (MQD)-silica hybrid monolithic column was developed for the enantioseparation of 9-fluorenylmethoxycarbonyl (FMOC) derivatized amino acids by nano-liquid chromatography. The mobile phase was optimized including the apparent pH, content of ACN, and concentration of the buffer to obtain a satisfactory enantioresolution performance. 27 FMOC derivatized amino acids including 19 protein and 8 non-protein amino acids were tested, and 19 out of them were enantiomerically discriminated obtaining baseline separation for 11 of them. Analytical characteristics of the method were evaluated for norvaline and tryptophan in terms of linearity, precision, accuracy, limits of detection (LOD) and quantitation (LOQ) showing good performance to be applied to the enantiomeric determination of these amino acids in dietary supplements. LOD and LOQ values were 9.3 and 31 μM for norvaline enantiomers and 7.5 and 25 μM for tryptophan enantiomers, respectively. The contents of d -norvaline and d -tryptophan were below their respective LODs in all the analyzed samples. Quantitation of l -tryptophan and l -norvaline showed good agreement with the labeled contents except for one sample which did not show presence of l -norvaline, contrary to the label indication.

The concurrent occurrence of summer heat stress and the non-breeding season has significantly impaired the semen quality of rams. Currently, there exists no straightforward and efficient method to address this issue. In this study, we demonstrate that two consecutive administrations of melatonin implants significantly mitigate the adverse effects of summer heat stress and the non-reproductive season on rams. Our findings indicate that implantation of exogenous melatonin enhances semen quality by improving sperm DNA integrity, mitochondrial integrity, and decreasing the proportion of abnormal sperm, as compared to control rams. This improvement is ascribed to the alleviation of oxidative stress and the optimized regulation of endocrine hormone levels in both serum and seminal plasma. Further exploration of the regulatory mechanism reveals that melatonin can also influence the tryptophan metabolism pathway. Additionally, our study revealed that certain indices and metabolites are strongly correlated with semen quality and can potentially serve as indicators for research aimed at improving semen quality. Notably, this is the first time that differences in tryptophan metabolites between serum and seminal plasma have been elucidated. All the above information suggests that melatonin implantation can protect sperm from heat stress by optimizing the blood and semen microenvironment.

Botanic polysaccharides can be metabolized by gut microbiota into short-chain fatty acids (SCFAs) to exert extensive bioactivities, yet targeted analysis of the effect of botanic polysaccharides on other gut microbial metabolites is scarcely seen. Tryptophan metabolites such as indole and indole derivatives play import roles in health and disease development. Using polysaccharides from Atractylodes macrocephala Koidz. (AMP) in treating ulcerative colitis as the example, we checked the effects of AMP on tryptophan metabolites. After examination of pharmacological effects of AMP, we established an ultra-performance liquid chromatography coupled with mass spectrometry/mass spectrometry (UPLC-MS/MS) method to simultaneously determinate the levels of 30 tryptophan metabolites and used the method to determine the levels of these metabolites in feces and plasma. The detection results showed that 12 metabolites in feces can be detected, and 17 metabolites can be detected in plasma samples. In addition, we found out that total levels of aryl hydrocarbon receptor ligands were decreased in colitis model whereas AMP treatment can increase the levels of total ligands in both feces and plasma. The results indicated that the therapeutical effect of AMP on colitis was associated with modulation of fecal and host tryptophan metabolism. This study provides new insight into the molecular mechanisms of polysaccharides that the beneficial effects of polysaccharides can be achieved by modulating microbial tryptophan metabolism in addition to SCFAs.

Background TRY-NAD metabolic network includes TRY (tryptophan), 5-HT (5-hydroxytryptamine), KYN (kynurenine), and NAD (nicotinamide adenine dinucleotide) pathway, which plays a significant role in neurological diseases and ageing. It is important to monitor these metabolites for studying the pathological anatomy of disease and treatment of responses evaluation. Although previous studies have reported quantitative methods for several metabolites in the network, the bottlenecks of simultaneously quantifying the whole metabolic network are their similar structures, diverse physico-chemical properties, and instability. Standardized protocols for the whole metabolic network are still missing, which hinders the in-depth study of TRY-NAD metabolic network in laboratory research and clinical screening. Results We developed a LC-MS/MS method for quantifying 28 metabolites in the TRY-NAD network simultaneously. Optimization was done for the mass spectral parameters, chromatographic conditions and sample pretreatment process. The developed method was fully validated in terms of standard curves, sensitivity, carryover, recovery, matrix effect, accuracy, precision, and stability. The pretreatment of 30 samples only takes 90 minutes, and the LC-MS/MS running time of one sample is only 13 minutes. With this method, we bring to light the chaos of global TRY-NAD metabolic network in sleep deprivation mice for the first time, including serum, clotted blood cells, hippocampus, cerebral cortex, and liver. NAD pathway levels in brain and blood decreased, whereas the opposite happened in the liver. The 5-HT pathway decreased and the concentration of KYN increased significantly in the brain. The concentration of many metabolites in KYN pathway (NAD + de novo synthesis pathway) increased in the liver. Significance This method is the first time to determine the metabolites of KYN, 5-HT and NAD pathway at the same time, and it is found that TRY-NAD metabolic network will be disordered after sleep deprivation. This work clarifies the importance of the pH of the extraction solution, the time and temperature control in pretreatment in standardized protocols building, and overcoming the problems of inconsistent sample pretreatment, separation, matrix effect interference and potential metabolite degradation. This method exhibits great prospects in providing more information on metabolic disturbances caused by sleep deprivation as well as neurological diseases and ageing.

In our previous study of 2,130 Chinese patients with coronary heart disease (CHD), we found that tryptophan (TRP) metabolites contributed to elevated risks of death. Many TRP-derived metabolites require the participation of intestinal bacteria to produce, and they play an important role in the pathogenesis of metabolic diseases such as CHD. So it is necessary to metabolize TRP into beneficial metabolites against CHD or prevent the production of harmful metabolites through external intervention. Indole-3-butyric acid (IBA) may be a key point of gut microbiota that causes TRP metabolism disorder and affects major adverse cardiovascular events in CHD. Therefore, this study aimed to develop a method based on in vitro culture bacteria to evaluate the effects of IBA on specific microbial metabolites quickly. We detected the concentrations of TRP and its metabolites in 11 bacterial strains isolated from feces using liquid chromatography–mass spectrometry, and selected Clostridium sporogenes as the model strain. Then, IBA was used in our model to explore its effect on TRP metabolism. Results demonstrated that the optimal culture conditions of C. sporogenes were as follows: initial pH, 6.8; culture temperature, 37°C; and inoculum amount, 2%. Furthermore, we found that IBA increases the production of TRP and 5-HIAA by intervening TRP metabolism, and inhibits the production of KYNA. This new bacteria-specific in vitro model provides a flexible, reproducible, and cost-effective tool for identifying harmful agents that can decrease the levels of beneficial TRP metabolites. It will be helpful for researchers when developing innovative strategies for studying gut microbiota.

Reliable methods for the determination of tryptophan and its metabolites are vital to the monitoring of biochemical states during the initiation and progression of cardiovascular disease. In the present study, a single-run liquid chromatography–tandem mass spectrometry (LC-MS/MS) method was developed for the simultaneous determination of tryptophan (Trp) and its metabolites, including kynurenine (Kyn), kynurenic acid (KA), xanthurenic acid (XA) and 5-hydroxytryptamine (5-HT), in human plasma. The plasma samples were prepared using a protein precipitation approach, and the chromatographic separation was performed by gradient elution on a C 18 column within a total analysis time of 3.5 min. The calibration ranges were 40–20,000 ng/mL for Trp, 4–2000 ng/mL for Kyn, 0.2–100 ng/mL for KA, 0.4–200 ng/mL for XA and 1–500 ng/mL for 5-HT, and the precision and accuracy were acceptable. The evaluation of recovery and internal standard-normalized matrix effect proved that the sample preparation approach was effective and the matrix effect could be negligible. The newly developed method was successfully applied to the analysis of plasma samples from healthy individuals and myocardial infarction patients. The findings suggested that the plasma concentrations of Trp, Kyn, 5-HT as well as the concentration ratios of Kyn/Trp and Trp/5-HT might serve as biomarkers for the monitoring of acute myocardial infarction.

In the past decade, carbon dots (CDs) have attracted considerable attention due to their excellent properties such as low toxicity, good biocompatibility, good fluorescence imaging, etc. Here, glutathione and l-tryptophan were used as carbon sources to hydrothermally synthesize CDs for sensitive and selective off/on detection of Fe3+ ions. The CDs are spherical nanoparticles with an average particle size of 3.8 nm and the presence of organic groups such as hydroxyl, carboxyl, sulfhydryl, and amino groups on their surface. The experiment results display that Fe3+ ions can be selectively and sensitively detected by quenching the fluorescence of CDs. Moreover, the fluorescence of the CDs+Fe3+ system can be restored after adding ascorbic acid. Thus, an off/on fluorescent probe for the determination of Fe3+ can be formed using the as-synthesized CDs solution. The CDs show a good linear range of 0–13.89 mM and a 0.0331 μM limit of detection for Fe3+, and the most probable mechanism concluded from ultraviolet–visible spectroscopy, electrospray ionization–mass spectrometry, and fluorescence spectrophotometry is a mixed static and dynamic quenching. Furthermore, the cytotoxicity experiment results show that CDs have low toxicity and can be used for intracellular imaging.

Dysregulation of the tryptophan (TRP) metabolic pathway is closely linked to the pathophysiology of neuropsychiatric disorders, such as depression. This study aimed to develop and validate a sensitive, rapid, and robust liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the simultaneous quantification of TRP and its metabolites, kynurenine (KYN) and kynurenic acid (KYNA), in human serum. Analytes were extracted from 100 µL of serum via simple protein precipitation with acetonitrile. Chromatographic separation was achieved on an Agilent ZORBAX HILIC Plus column (4.6 mm×100 mm, 3.5 µm) using isocratic elution with a mobile phase of methanol: acetonitrile containing 5 mM ammonium formate. Quantification was performed using an electrospray ionization source in positive ion multiple reaction monitoring (MRM) mode, with a total run time of only 2.0 min.The linear ranges were 1- 50 µg/mL for TRP, 0.1-5 µg/mL for KYN, and 1-50 ng/mL for KYNA, covering clinically relevant levels. A weighting factor of 1/x² provided the best fit for calibration curves (R² > 0.99). Extraction recoveries ranged from 88.23% to 99.39%, and mean internal standard-normalized matrix effects were 81%-100%. Accuracy and precision values met bioanalytical acceptance criteria. Stability assessments confirmed that samples were stable at -20°C and -80°C for 31 days and through three freeze-thaw cycles. This validated method was successfully applied to analyze serum samples from 103 adolescent patients with first-episode depression.

In this research, solubility of l-tryptophan in water, methanol, ethanol, acetone, isopropanol, n-butanol, acetonitrile, ethyl acetate, organic pure solvent, and in three binary mixed solvents (water + acetonitrile), (water + methanol), and (water + isopropanol) at the temperature range of T = 278.15–333.15 K was determined with the gravimetric method. The experimental results show that the solubility of l-tryptophan in the above solvents increased as temperature rose, and it achieved the highest dissolution efficiency in water. The improved Apelblat model, the Buchowski–Ksiazaczak λh model, the Redlich–Kister (CNIBS/R–K) model, and the Jouyban–Acree model were used to nonlinearly fit the experimental data of solubility, and the consistency was good. The physicochemical information of l-tryptophan provided in this report may be helpful for its extraction/separation, recrystallization, purification, and formulation development.

A glassy carbon electrode (GCE) was modified with poly(L-arginine) (P-Arg), reduced graphene oxide (rGO) and gold nanoparticle (AuNP) to obtain an electrode for simultaneous determination of dopamine (DA), serotonin (5-HT) and L-tryptophan (L-Trp) in the presence of ascorbic acid (AA). The modified GCE was prepared via subsequent ‘layer-by-layer’ deposition using an electrochemical technique. The surface morphology of the modified electrode was studied by scanning electron microscopy, and electrochemical characterizations were carried out via cyclic voltammetry and electrochemical impedance spectroscopy. The modified electrode showed excellent electrocatalytic activity toward DA, 5-HT and L-Trp at pH 7.0. Figures of merit for the differential pulse voltammetric reponse are as follows: (a) Response to DA is linear in two intervals, viz. 1.0–50 nM and 1.0–50 μM DA concentration range, the typical working voltage is 202 mV (vs. Ag/AgCl), and the detection limit is 1 nM (at an S/N ratio of 3). For 5-HT, the respective data are 10 to 500 nM and 1.0 to 10 μM, 381 mV, and 30 nM. For L-Trp, the respective data are 10–70 nM and 10–100 μM, 719 mV, and 0.1 μM. The modified GCE is fairly selective. It was successfully applied to the simultaneous determination of DA, 5-HT, and L-Trp in spiked urine samples, and high recovery rates were found.

Depression is a common mental disorder that substantially impairs patients’ daily life and work. To identify natural and safe preventive options, we investigated the preventive effect and underlying mechanism of theGanoderma lucidumandRosa roxburghiiTratt formula (GLRRTF) on depression. A total of 72 chemical components in GLRRTF were identified by UHPLC-ESI-Q-Exactive Plus Orbitrap-MS Analysis. GLRRTF (containing 400 mg/kg ofG. lucidumextract and 800 mg/kg ofR. roxburghiiextract per day), administered as a 1-week preventive intervention followed by 4 weeks of co-administration with chronic unpredictable mild stress, prevented the development of depression-like behaviors in male C57BL/6J mice and reduced neuronal damage in the hippocampus. Airflow-assisted desorption electrospray ionization mass spectrometry imaging and enzyme-linked immunosorbent assays showed that GLRRTF corrected abnormalities in neurotransmitter levels. The 16S rRNA sequencing indicated that GLRRTF restored dysbiosis of the gut microbiota. Metabolomic profiling revealed that GLRRTF increased the level of tryptophan and promoted tryptophan metabolism towards the 5-HT and indole pathways in feces and the brain. Western blot demonstrated that GLRRTF increased 5-HT production from tryptophan in the brain by regulating tryptophan hydroxylase 2 and DOPA decarboxylase. GLRRTF activated the PI3K/AKT pathway by regulating brain-derived neurotrophic factor and its receptor tropomyosin receptor kinase B. This research provides a comprehensive mechanistic understanding of GLRRTF’s preventive effect against depression, highlighting its potential as a novel, safe, and preventive functional food formulation.
