Vitamin E-based redox-sensitive salinomycin prodrug-nanosystem with paclitaxel loaded for cancer targeted and combined chemotherapy

De-Sheng Liang,Jian Liu,Tao-Xing Peng,Hui Peng,Feng Guo,Hai-Jun Zhong

Cancer stem cells (CSCs) can resist conventional chemotherapy to lead to cancer recurrence. For complete eradication of cancers, an effective CSCs therapeutic strategy should be developed to combine with conventional chemotherapy. In this work, a novel vitamin E-based redox-sensitive salinomycin (SAL, an inhibitor for CSCs) prodrug nanoparticles (TS NPs) and hyaluronic acid (HA)-coated TS NPs (HTS NPs) were fabricated to deliver paclitaxel (PTX) for cancer-targeted and combined chemotherapy. Both TS and HTS prodrug NPs had mean diameter of about 200 nm with uniform size distribution, excellent drug loading capacity for PTX, and glutathione-triggered SAL and PTX release profiles. The HTS prodrug NPs had enhanced cellular uptake efficiency over TS NPs due to CD44 receptor-mediated endocytosis, hence exerting stronger potency of SAL upon CSCs-enriched mammospheres formation and G 0 /G 1 cell phase arresting. Cytotoxicity and 3D tumor spheroids assays demonstrated that both TS and HTS prodrug NPs themself can synergize with loaded PTX to maximize the chemotherapeutic effect. Obviously, the latter demonstrated a more potent anticancer efficacy due to improved intracellular drug delivery efficiency. These results suggested that the designed TS prodrug NPs, especially the coated HTS NPs can serve as an effective anti-CSCs strategy for cancer targeted and combination treatments.

Conjugation of vitamin E-TPGS and guar gum to carry borneol for enhancing blood–brain barrier permeability

Yude Liu,Lian Rao,Hongguan Zhang,Yanyou Cen,Kaili Cheng

Herb borneol is usually used in clinics for the treatment of central nervous system illness, for its ability of blood–brain barrier permeability, although its poor water solubility and poor bioavailability limit its clinical application to some degree. In this study, we developed a novel nanoparticle combining the benefits of vitamin E d-ɑ-tocopheryl poly(ethylene glycol) succinate (E-TPGS) (or TPGS) and guar gum to get TPGS-g-guar gum as a drug delivery system to carry borneol, which could improve the solubility of borneol and increase the drug-loading capacity efficiently. The results showed that TPGS-g-guar gum nanoparticles delivery system was suitable to carry borneol and release the drug effectively, and TPGS-g-guar gum/borneol nanoparticles would be a potential platform for improving the treatment of central nervous system illness and cerebrovascular disease.

Artificial Multi-Stimulus-Responsive E-Skin Based on an Ionic Film with a Counter-Ion Exchange Reagent

Baozhong Chen,Kangxin Shen,Yaping Li,Bo Huang,Huiming Su,Jintao Xu,Shuai Yang,Qi Zhou,Linfeng Lan,Junbiao Peng,Yong Cao

Sensing pressure and temperature are two important functions of human skin that integrate different types of tactile receptors. In this paper, a deformable artificial flexible multi-stimulus-responsive sensor is demonstrated that can distinguish mechanical pressure from temperature by measuring the impedance and the electrical phase at the same frequency without signal interference. The electrical phase, which is used for measuring the temperature, is totally independent of the pressure by controlling the surface micro-shapes and the ion content of the ionic film. By doping the counter-ion exchange reagent into the ionic liquid before pouring, the upper temperature measuring limit increases from 35 to 50 °C, which is higher than the human body temperature and the ambient temperature on Earth. The sensor shows high sensitivity to pressure (up to 0.495 kPa −1 ) and a wide temperature sensing range (−10 to 50 °C). A multimodal ion-electronic skin (IE M -skin) with an 8 × 8 multi-stimulus-responsive sensor array is fabricated and can successfully sense the distribution of temperature and pressure at the same time. Finally, the sensors are used for monitoring the touching motions of a robot-arm finger controlled by a remote interactive glove and successfully detect the touching states and the temperature changes of different objects.

Characterization of flavor substances in cooking and seasoned cooking brown seaweeds by GC-IMS and E-nose

Shan Jiang,Pengfei Jiang,Dingding Feng,Meiran Jin,Hang Qi

The flavor of algae was one of the key factors for consumer acceptance. The objective of this study was to investigate the characteristic volatile compounds in cooking and seasoned cooking edible brown seaweeds ( Undaria pinnatifida and Laminaria japonica ). The gas chromatography-ion mobility spectrometry (GC-IMS) and electronic nose (E-nose) analysis showed that baking resulted in significant difference in flavor of brown seaweeds. However, the overall effect of cooking was not as significant as that of the seasoning solution treatment. Additionally, brown seaweeds treated with the seasoning solution were more acceptable. Undaria pinnatifida was found to contain 72 volatile flavor compounds, while Laminaria japonica had a total of 70. This study proved the applicability of GC-IMS combined with E-nose technology to detect the changes of volatile components of brown seaweeds after processing, providing beneficial knowledge and basic theory for the deep processing of brown seaweeds.

A permeable, metal-like conductivity, stretchable, strain-insensitivity, self-assembled and rapidly formed Janus-structured e-skin

Shengxin Xiang,Xiao Wei,Lei Liu,Jianlong Hong,Shengshun Duan,Huiyun Zhang,Jinqiu Huang,Zhishui Chen,Zhiwei Zhao,Qiongfeng Shi,Jun Wu

Electrodes are transitioning from flexible to stretchable modality to accommodate more complex application scenarios. Here, inspired by the human skin, we report a rapidly formed Janus-structured stretchable (JSS) electrode. Wherein, the phase separation technology enables rapid film formation, and the gravity effect constructs the Janus structure. The Janus structure aggregates liquid metal (LM) on one side and thermoplastic polyurethane (TPU) on the other, providing a combination of highly stretchable and highly conductive nature at the same time. The JSS film has fast film forming time (15 s), metal-like conductivity (10 6 S/m), high stretchability (260 %), permeable (4077.41 g m −2 day −1 ), ultra-thin thickness (50–1000 μm), strain-insensitivity (stretching, folding, and rotation states), as well as excellent cuttable, transferable, recyclable, reconfigurable, and biocompatible property. Furthermore, the JSS films successfully achieve the effective acquisition of physiological, triboelectric, and resistive signals, when applied as standalone devices and integrated electrodes for electrophysiological signal recording, energy harvesting, and wearable sensing. With its prominent performance and broad adaptability, the proposed JSS film is expected to significantly advance the field of stretchable electrodes and further promote the practical applications of flexible and stretchable devices in industrial and electronic skin.

An inhibition mechanism of dihydromyricetin on tyrosinase and the joint effects of vitamins B6, D3 or E

Meihui Fan,Guowen Zhang,Junhui Pan,Deming Gong

Dihydromyricetin (DMY), a natural flavonoid, was found to effectively inhibit tyrosinase activity in a mixed-type manner with an IC50 value of (3.66 ± 0.14) × 10−5 mol L−1. DMY combined with the dietary vitamin D3 at lower concentrations exhibited a synergistic effect on the inhibition of tyrosinase. The formation of a DMY–tyrosinase complex led to fluorescence quenching and conformational changes of tyrosinase, which was driven mainly by hydrophobic interactions and hydrogen bonds. The molecular simulation further found that DMY inserted into the active pocket of tyrosinase interacted with amino acid residues Tyr78, His85, and Ala323, occupying the catalytic center of tyrosinase to hinder entrance of the substrate, leading to the inhibition of tyrosinase. This study may provide a scientific foundation for screening effective tyrosinase inhibitors.

Comparison of two kinds of docetaxel-vitamin E prodrugs: In vitro evaluation and in vivo antitumor activity

Jing Wang,Peng Xue,Jiahua Zhou,Lin Li,Lu Xu,Yongjun Wang

To achieve optimal therapeutic index of docetaxel , we conjugated docetaxel (DTX) with vitamin E (VE) by ester bond with disulfide bond or thioether bond as spacer to produce DTX-ss-VE or DTX-s-VE. The two prodrugs were successfully loaded into liposomes . The physicochemical characterization and in vitro release profiles of the prodrug loaded liposomes were investigated. MTT assays showed that the cytotoxicity of DTX-ss-VE loaded liposomes on PC3 (IC 50   =   27.5   nM) and A549 cells (IC 50   =   63.4   nM) was comparable with the cytotoxicity of DTX-s-VE loaded liposomes (IC 50   =   99.2 and 159.5   nM, respectively). The pharmacokinetic studies demonstrated that DTX-ss-VE and DTX-s-VE loaded liposomes exhibited an extended DTX half-life (3.6   ±   1.2 and 10.0   ±   3.0   h, respectively) as compared to DTX solutions (2.1   ±   1.5   h) and an increased AUC (30487.3   ±   3791.6 and 20922.1   ±   5633.3   ng/L   ×   h, respectively) compared with DTX solutions (1779.3   ±   226.6   ng/L   ×   h). Finally, the in vivo anti-tumor studies showed that DTX-ss-VE loaded liposomes possessed similar antitumor activity compared with DTX solutions. Unexpectedly, not any tumor inhibition effect was observed in DTX-s-VE loaded liposomes group. In a conclusion, our studies suggested that simplex favorable properties of the anticancer prodrug can not ensure available good therapeutic index and the rational design of prodrug needs a comprehensive understanding of the in vitro and in vivo behavior of the prodrug.

Fabrication of TiO2–Bi2WO6 Binanosheet for Enhanced Solar Photocatalytic Disinfection of E. coli: Insights on the Mechanism

Yanan Jia,Sihui Zhan,Shuanglong Ma,Qixing Zhou

TiO2–Bi2WO6 binanosheet (TBWO), synthesized by a facile two-step hydrothermal method, was used as an effective visible-light-driven photocatalyst for the inactivation of E. coli and was characterized by TEM, SEM, XRD, FTIR, XPS, and BET. A series of TBWOs with different doping ratios of TiO2 loading from 10 to 55 wt % were synthesized. Among all of the TBWOs, 40% TBWO exhibited the best bacteria disinfection efficiency, and the quantity of viable bacteria could reach 10° with 40% TBWO (100 μg/mL) after being illuminated for 4 h. Furthermore, the confocal fluorescent-based cell live/dead test and the SEM technology were applied to verify the photocatalytically lethal effect toward E. coli and the rupture of bacterial membranes. The leak of bacterial contents, including the bacterial genome represented by relevant 16srDNA, and total protein were detected by PCR and bicinchoninic acid assay. In this work, the antibacterial mechanism was studied by employing photoelectrochemical techniques, electron spin resonance (ESR), and scavengers of different reactive species, revealing the pivotal roles of electron hole (h+) and electron (e–) in the photocatalytic process. In addition, the •O2– and •OH radicals were also detected in the TBWOs system by ESR. It was found that the adsorption of visible light and separation of photogenerated carriers within TiO2 have been largely promoted after being coupled with Bi2WO6, which should be responsible for the improved bactericidal effect.

Astragaloside IV alleviates E. coli-caused peritonitis via upregulation of neutrophil influx to the site of infection

Ping Huang,Xiaoyan Lu,Baohong Yuan,Tao Liu,Liangcheng Dai,Yunjun Liu,Hui Yin

Astragaloside IV (AS-IV), an active saponin purified from Astragali Radix , has been identified with broad biological and pharmacological activities . In the present study, we continue to explore the potential effect of AS-IV on antibacterial response using an acute E. coli peritoneal infection model. Our findings implied that administration of AS-IV decreases mortality in mice challenged by lethal E. coli infection. The protection of AS-IV was related to promotion of neutrophil extravasation into the peritoneum and bacterial clearance . Toll-like receptor (TLR) activation in neutrophils has been reported to reduce CXCR2 expression and subsequent neutrophil migration. Our data indicated that AS-IV prevented the reduction of CXCR2 expression and neutrophil migration induced by LPS, the activator for TLR4 . Moreover, we found that AS-IV blocks LPS-induced suppression of CXCR2 on neutrophils by inhibiting the expression of G protein-coupled receptor kinase-2 (GRK2), an agonist that regulates desensitization and internalization of chemokine receptors . Taken together, these data propose that AS-IV, through modulating GRK2-CXCR2 signal in neutrophils, offers an essential efficacy on host antibacterial immunity.

(E)-2-Hexenal-loaded ethyl cellulose/agarose aerogel: Preparation, characterization and application in fresh rice noodles preservation

Haiyan Li,Zekun Jin,Nengguo Tao

This study aimed to develop food-grade ethyl cellulose/agarose aerogels for the controlled release of plant essential oils to extend the shelf life of fresh rice noodles. The antibacterial efficacy of ten unsaturated aldehyde essential oils was evaluated against primary spoilage bacteria, including Leuconostoc lactis , Lactococcus lactis , Acinetobacter johnsonii , Exiguobacterium indicum , and Enterobacter sichuanensis . Based on its superior antimicrobial performance, (E)-2-hexenal was selected for encapsulation within ethyl cellulose/agarose aerogels to achieve controlled release and extend the shelf life of fresh rice noodles. Increasing the ethyl cellulose concentration resulted in a more uniform pore size distribution, enhanced mechanical properties and hydrophobicity, and improved controlled release of (E)-2-hexenal in the aerogels. The optimized aerogel, loaded with 0.20 μL/mL of (E)-2-hexenal, exhibited significant antimicrobial activity, effectively inhibiting bacterial growth for 72 h. Moreover, the aerogel formulation effectively preserved the quality of fresh rice noodles by mitigating changes in total plate count, pH, water content, water activity, cooking properties, color, and texture, ultimately extending the product's shelf life by 4 days at 25 °C. These findings highlight the potential of ethyl cellulose/agarose aerogels loaded with (E)-2-hexenal as an innovative preservation strategy for fresh rice noodles, providing a promising method for enhancing food quality and longevity.

Cellulose nanofiber-mediated manifold dynamic synergy enabling adhesive and photo-detachable hydrogel for self-powered E-skin

Zhang Lei,Chen Lu,Wang Siheng,Wang Shanshan,Wang Dan,Yu Le,Xu Xu,Liu He,Chen Chaoji

Self-powered skin attachable and detachable electronics are under intense development to enable the internet of everything and everyone in new and useful ways. Existing on-demand separation strategies rely on complicated pretreatments and physical properties of the adherends, achieving detachable-on-demand in a facile, rapid, and universal way remains challenging. To overcome this challenge, an ingenious cellulose nanofiber-mediated manifold dynamic synergy strategy is developed to construct a supramolecular hydrogel with both reversible tough adhesion and easy photodetachment. The cellulose nanofiber-reinforced network and the coordination between Fe ions and polymer chains endow the dynamic reconfiguration of supramolecular networks and the adhesion behavior of the hydrogel. This strategy enables the simple and rapid fabrication of strong yet reversible hydrogels with tunable toughness ((Value max -Value min )/Value max of up to 86%), on-demand adhesion energy ((Value max -Value min )/Value max of up to 93%), and stable conductivity up to 12 mS cm −1 . We further extend this strategy to fabricate different cellulose nanofiber/Fe 3+ -based hydrogels from various biomacromolecules and petroleum polymers, and shed light on exploration of fundamental dynamic supramolecular network reconfiguration. Simultaneously, we prepare an adhesive-detachable triboelectric nanogenerator as a human-machine interface for a self-powered wireless monitoring system based on this strategy, which can acquire the real-time, self-powered monitoring, and wireless whole-body movement signal, opening up possibilities for diversifying potential applications in electronic skins and intelligent devices.

Perceptual interactions of key aroma compounds in Fu brick tea: Odor threshold, sensory, and E-nose analysis

Xuexue Zheng,He Xie,Wenjie Fu,Chao Wang,Tengfei Hu,Xingchang Ou,Jian'an Huang,Zhonghua Liu,Qin Li

While extensive research has focused on the flavor of Fu brick tea (FBT), the perceptual interactions among its aromatic components remain unclear. To address this, twenty representative aroma compounds were selected from FBT to determine their thresholds and evaluate their interactions by analyzing threshold changes before and after mixing. The results revealed discrepancies between the determined thresholds and literature values, particularly for α -terpineol, methyl heptenone, and geranylacetone, which demonstrated ratios of 73.54, 6.62, and 6.41, respectively. Mixtures with similar aromas or structures exhibited synergy, whereas those with differing aromas or structures displayed masking effects. Additionally, the odor compound ( E )-nerolidol was added to the tea infusion at a concentration below its threshold level to investigate its impact on the overall aroma perception of FBT through sensory analysis and electronic nose assessments. The findings indicated that sub-threshold compounds could significantly influence the overall aroma profile of FBT.
